Introduction Fungal spores constitute an important fraction of bioaerosols in the atmosphere. Objectives To analyse the content of Alternaria and Cladosporium spores in the atmosphere of Beja and the effect of meteorological conditions on their concentrations. Methodology The daily and hourly data of Alternaria and Cladosporium fungal spores concentration in the atmosphere of Beja were monitored from April 12, 2012 to July 30, 2014, based on the Portuguese Aerobiology Network methodology. The influence of meteorological conditions on the studied types of fungal spore concentrations was assessed through Spearman’s correlation analysis. Results During the study period, 20,741 Alternaria spores and 320,862 Cladosporium spores were counted. In 2013, there were 5,822 Alternaria spores and 123,864 Cladosporium spores. The absolute maximum concentrations of Alternaria and Cladosporium spores were recorded on November 8, 2013, with 211 and 1301 spores/m3, respectively. Temperature, insolation and wind direction parameters showed a positive correlation with Alternaria and Cladosporium spore levels, while relative humidity and precipitation presented a negative correlation, which is statistically significant. Wind speed only showed a statistically significant positive correlation in terms of Alternaria spore levels. Conclusion Alternaria and Cladosporium spores are present in the atmospheric air of Beja throughout the year, with the highest concentration period occurring during spring and autumn. There was a clear effect of meteorological parameters on airborne concentrations of these fungal spores.  相似文献

Over the period 2002–2014, air temperature significantly increased regionally for Bratislava. However, no significant shifts have been observed in other meteorological parameters examined. The aim of this study was to analyse the effect of significant temperature trends on timing, duration and intensity of Alternaria, Epicoccum and Stemphylium spore seasons. Aerobiological monitoring was conducted using a Burkard 7-day volumetric spore trap. Mann–Kendall tau test was used to determine trends in spore seasons characteristics, whereas Spearman’s correlation coefficients were calculated to establish the relationships between temperature and spore season time series. Spore seasons of analysed taxa changed throughout the years of study. Alternaria spore season now starts earlier, ends later and lasts longer. Start, end and peak dates as well as duration of Alternaria spore seasons were significantly correlated with recorded increases in winter temperatures. Despite significant lengthening of Alternaria spore seasons, the lack of rising trend in its spore season index has been registered. This phenomenon could be partly explained by the reduction in the source vegetation due to drop of agricultural land use areas in Bratislava. In contrast, the intensity of Stemphylium spore seasons significantly increased during the study period and was correlated with recorded increases in summer–autumn temperatures. Based on the results of this study, it could be concluded that changes in selected fungal spore season patterns in Bratislava (earlier start date, later end date and longer duration of Alternaria spore seasons and higher Stemphylium spore season indexes) might be caused by the recorded local change in air temperature.  相似文献

This study determined the relationship between airborne concentration of Cladosporium spp. spores and wind speed and direction using real data (local wind measured by weather station) and modelled data (air mass flow computed with the aid of HYbrid Single Particle Lagrangian Trajectory model). Air samples containing fungal conidia were taken at an urban site (Worcester, UK) for a period of five consecutive years using a spore trap of the Hirst design. A threshold of ≥6000 s m?3 (double the clinical value) was applied in order to select high spore concentration days, when airborne transport of conidia at a regional scale was more likely to occur. Collected data were then examined using geospatial and statistical tools, including circular statistics. Obtained results showed that the greatest numbers of spore concentrations were detected in July and August, when C. herbarum, C. cladosporioides and C. macrocarpum sporulate. The circular correlation test was found to be more sensitive than Spearman’s rank test. The dominance of either local wind or the air mass on Cladosporium spore distributions varied between examined months. Source areas of this pathogen had an origin within the UK territory. Very high daily mean concentrations of Cladosporium spores were observed when daily mean local wind speed was v s ≤ 2.5 m s?1 indicating warm days with a light breeze.  相似文献

A study was made of the link between time of day, weather variables and the hourly content of certain fungal spores in the atmosphere of the city of Szczecin, Poland, in 2004–2007. Sampling was carried out with a Lanzoni 7-day-recording spore trap. The spores analysed belonged to the taxa Alternaria and Cladosporium. These spores were selected both for their allergenic capacity and for their high level presence in the atmosphere, particularly during summer. Spearman correlation coefficients between spore concentrations, meteorological parameters and time of day showed different indices depending on the taxon being analysed. Relative humidity (RH), air temperature, air pressure and clouds most strongly and significantly influenced the concentration of Alternaria spores. Cladosporium spores correlated less strongly and significantly than Alternaria. Multivariate regression tree analysis revealed that, at air pressures lower than 1,011 hPa the concentration of Alternaria spores was low. Under higher air pressure spore concentrations were higher, particularly when RH was lower than 36.5%. In the case of Cladosporium, under higher air pressure (>1,008 hPa), the spores analysed were more abundant, particularly after 0330 hours. In artificial neural networks, RH, air pressure and air temperature were the most important variables in the model for Alternaria spore concentration. For Cladosporium, clouds, time of day, air pressure, wind speed and dew point temperature were highly significant factors influencing spore concentration. The maximum abundance of Cladosporium spores in air fell between 1200 and 1700 hours.  相似文献

The chickweed (Stellaria media L.) pro-SmAMP2 gene encodes the hevein-like peptides that have in vitro antimicrobial activity against certain harmful microorganisms. These peptides play an important role in protecting the chickweed plants from infection, and the pro-SmAMP2 gene was previously used to protect transgenic tobacco and Arabidopsis plants from phytopathogens. In this study, the pro-SmAMP2 gene under control of viral CaMV35S promoter or under control of its own pro-SmAMP2 promoter was transformed into cultivated potato plants of two cultivars, differing in the resistance to Alternaria: Yubiley Zhukova (resistant) and Skoroplodny (susceptible). With the help of quantitative real-time PCR, it was demonstrated that transgenic potato plants expressed the pro-SmAMP2 gene under control of both promoters at the level comparable to or exceeding the level of the potato actin gene. Assessment of the immune status of the transformants demonstrated that expression of antimicrobial peptide pro-SmAMP2 gene was able to increase the resistance to a complex of Alternaria sp. and Fusarium sp. phytopathogens only in potato plants of the Yubiley Zhukova cultivar. The possible role of the pro-SmAMP2 products in protecting potatoes from Alternaria sp. and Fusarium sp. is discussed.  相似文献

We explored the temporal and spatial variations in airborne Alternaria spore quantitative and phenological features in Europe using 23 sites with annual time series between 3 and 15 years. The study covers seven countries and four of the main biogeographical regions in Europe. The observations were obtained with Hirst-type spore traps providing time series with daily records. Site locations extend from Spain in the south to Denmark in the north and from England in the West to Poland in the East. The study is therefore the largest assessment ever carried out for Europe concerning Alternaria. Aerobiological data were investigated for temporal and spatial patterns in their start and peak season dates and their spore indices. Moreover, the effects of climate were checked using meteorological data for the same period, using a crop growth model. We found that local climate, vegetation patterns and management of landscape are governing parameters for the overall spore concentration, while the annual variations caused by weather are of secondary importance but should not be neglected. The start of the Alternaria spore season varies by several months in Europe, but the peak of the season is more synchronised in central-northern Europe in the middle of the summer, while many southern sites have peak dates either earlier or later than northern Europe. The use of a crop growth model to explain the start and peak of season suggests that such methods could be useful to describe Alternaria seasonality in areas with no available observations.  相似文献

During the spring and summer of 2014 and 2015, wheat and barley fields in the Iranian provinces of Golestan and Alborz showed a high incidence of symptoms of black (sooty) head mold of wheat and barley. The isolation results revealed that Alternaria was associated with these symptoms. One hundred and forty isolates were collected and morphologically characterized based on the development of conidial chains with primary, secondary, and tertiary branching patterns, consistent with the three-dimensional sporulation complexity of members of Alternaria in sections Infectoriae and Pseudoalternaria. Subsequently, 16 Alternaria isolates exhibiting high morphological diversity were characterized based on extensive morphological and molecular comparisons. Phylogenetic analyses of three loci [ITS, glyceraldehyde 3-phosphate dehydrogenase (gpd), and plasma membrane ATPase (ATPase)] revealed that 15 isolates belonged to section Infectoriae but could not be assigned to phylogenetic species and one isolate represents a new species, Alternaria kordkuyana sp. nov., in section Pseudoalternaria. Morphological assessments revealed a high degree of variation among section Infectoriae isolates and that A. kordkuyana has significant morphological differences as compared to the three other species currently described in section Pseudoalternaria.  相似文献

Fungal spores are normal components of external environments. They have been reported to be associated with human, animal and plant diseases causing primary and opportunistic infections. Cladosporium is commonly the most frequently isolated genus from air samples, and its species are considered mainly saprophytic, but there is a wide variety of taxa that can cause adverse effects on human and animal health and also on plants. In this work, we aimed to record Cladosporium spores frequency of isolation in northern Argentinean Patagonia and to perform a molecular characterization based on actin gene complemented with EF1α and ITS genes. The ability of the pathogens to cause disease in pip fruits of Packham’s Triumph and Abate Fetel pears and red delicious apples was determined. Results confirmed Cladosporium spores as the main genera isolated from air samples, and the molecular characterization revealed the existence of 11 species in this region grouped in C. cladosporioides and C. herbarum complexes. Pathogenicity tests revealed that Cladosporium sp. cause disease on fruit. Abate Fetel was the most susceptible to infection. These results compose the first study in Argentina in respect of identification at species level of airborne Cladosporium spores, and furthermore, it is the first report in northern Patagonia and the high valley of Río Negro productive region. This knowledge can help farmers to take preventive measures in order to avoid biological damage.  相似文献

Here we evaluate the origins and relationships of Mexican and Central American Diplazium hybrids derived from crosses involving either D. plantaginifolium or D. ternatum. Based on study of live plants and herbarium specimens, we distinguish D. ×verapax from the similar D. riedelianum and present evidence that the former is a sterile hybrid derived from a cross between D. plantaginifolium and D. werckleanum. We also describe new hybrids, D. ×torresianum and D. ×subternatum from Mexico and northern Central America. Both involve D. ternatum as one parent. Diplazium. cristatum is the other putative parent of D. ×torresianum, and D. plantaginifolium is the second parent of D. ×subternatum. We also designate lectotypes for D. cordovense and D. dissimile.  相似文献

Wild relatives of Brassica are a rich reservoir of genes that are invaluable for the improvement of cultivated species. Sinapis alba is a close relative of crop Brassicas that possesses several desirable traits such as tolerance to Alternaria black spot disease, heat stress, insect pests and nematodes. This study is aimed at developing and characterizing hybrids between Brassica juncea and S. alba with the ultimate goal of transferring genes for tolerance to Alternaria brassicae and heat stress, the traits that are lacking in cultivated Brassica. We generated three hybrids between B. juncea and S. alba through protoplast fusion. The hybridity was confirmed through cytology and molecular markers. While two of the hybrids were symmetric, the third one was asymmetric and had greater resemblance to B. juncea. Hybrids showed some characteristic features of the parents and were fully male and female fertile and also set seeds upon back crossing with the parent species. In vitro leaf assay and field inoculation studies revealed that the hybrids are highly resistant to A. brassicae. Besides, hybrids set seeds at temperature of >?38 °C when parents failed to produce seeds indicating that hybrids possess heat tolerance. These stable hybrids provide a reliable genetic resource for transfer of genes from S. alba into cultivated Brassica species.  相似文献

Pathogenicity of Candida albicans is associated with its capacity switch from yeast-like to hyphal growth. The hyphal form is capable to penetrate the epithelial surfaces and to damage the host tissues. Therefore, many investigations have focused on mechanisms that control the morphological transitions of C. albicans. Recently, certain studies have showed that non-albicans Candida species can reduce the capacity of C. albicans to form biofilms and to develop candidiasis in animal models. Then, the objective of this study was to evaluate the effects of Candida krusei and Candida glabrata on the morphogenesis of C. albicans. Firstly, the capacity of reference and clinical strains of C. albicans in forming hyphae was tested in vitro. After that, the expression of HWP1 (hyphal wall protein 1) gene was determined by quantitative real-time PCR (polymerase chain reaction) assay. For both reference and clinical strains, a significant inhibition of the hyphae formation was observed when C. albicans was incubated in the presence of C. krusei or C. glabrata compared to the control group composed only by C. albicans. In addition, the culture mixed of C. albicans-C. krusei or C. albicans-C. glabrata reduced significantly the expression of HWP1 gene of C. albicans in relation to single cultures of this specie. In both filamentation and gene expression assays, C. krusei showed the higher inhibitory activity on the morphogenesis of C. albicans compared to C. glabrata. C. krusei and C. glabrata are capable to reduce the filamentation of C. albicans and consequently decrease the expression of the HWP1 gene.  相似文献

Abstract

While DE-ETIOLATED 1 (DET1) is well known as a negative regulator of light development, here we describe how det1 mutants also exhibit altered responses to salt and osmotic stress, specifically salt and mannitol resistant germination. LONG HYPOCOTYL 5 (HY5) positively regulates both light and abscisic acid (ABA) signalling. We found that hy5 suppressed the det1 salt and mannitol resistant germination phenotype, thus, det1 stress resistant germination requires HY5. We then queried publically available microarray datasets to identify genes downstream of HY5 that were differentially expressed in det1 mutants. Our analysis revealed that ABA regulated genes, including ABA RESPONSIVE ELEMENT BINDING FACTOR 3 (ABF3), are downregulated in det1 seedlings. We found that ABF3 is induced by salt in wildtype seeds, while homologues ABF4 and ABF1 are repressed, and all three genes are underexpressed in det1 seeds. We then investigated the role of ABF3, ABF4, and ABF1 in det1 phenotypes. Double mutant analysis showed that abf3, abf4, and abf1 all suppress the det1 salt/osmotic stress resistant germination phenotype. In addition, abf1 suppressed det1 rapid water loss and open stomata phenotypes. Thus interactions between ABF genes contribute to det1 salt/osmotic stress response phenotypes.

Fungal spores are among the most commonly encountered airborne biological particles, and it is widely proved that they represent a potential source of allergens involved in rhinitis and asthma. A change in temperature may influence the colonisation and growth of fungi directly through the physiology of individual organisms, or indirectly through physiological effects on their host plants or substrates and any competitors or enemies. In order to detect and monitor the evolution of the spore counts, air sampling was carried out using standard equipment (Hirst-type volumetric traps) and an identical method in several stations across France. Cladosporium has been here emphasised because of its very large contribution to the total fungal spectrum. Moreover, this taxon is of particular clinical importance because it possesses a high allergenic potential. The data from the oldest traps (Aix-en-Provence, Bordeaux, Lyon, Paris and Toulouse) were analysed on an annual base. Located at different latitudes and in different climatic areas, these five cities showed fundamentally different trends for the concentrations of Cladosporium spores: downward trend at the southernmost locations and upward trend at the other locations, whereas temperature was everywhere continuously rising over the study period. However, longer data sets are needed to be able to draw more definitive conclusions about quantitative trends in airborne fungal spore concentrations.  相似文献

Present investigation was undertaken to study the dynamics of relationships between atmospheric fungal spores and meteorological factors in western Romania. The airborne spore sampling was carried out by employing volumetric sampling. A total of nine meteorological parameters were selected for this investigation. During 2008–2010, it was found the same pattern of behaviour in the atmosphere for selected spore types (Alternaria, Cladosporium, Pithomyces, Epicoccum and Torula). The spores occurred in the air throughout the whole year, but maximum concentrations were reached in summer. Cladosporium and Alternaria peak levels were observed in June. Epicoccum peak value was found in September. The relationships between airborne spore concentrations and environmental factors were assessed using the analysis of Spearman’s rank correlations and multiple linear regressions. Spearman’s rank correlation analysis revealed that maximum, minimum and mean temperature, and number of sunshine hours were strongly (p < 0.01) and directly proportional to the concentration of all analysed fungal spores. Negative and significant correlations were with daily mean relative humidity. The variance explained percentage by regression analyses varied between 30.6 and 39.6 % for Alternaria and Cladosporium airborne spores. Statistical methods used in this study are complementary and confirmed stable dependence of Alternaria and Cladosporium spore concentrations on meteorological factors. The climate change parameters either increased temperatures, changed precipitation regimes or a combination of both affected allergenic fungal spore concentrations in western Romania. This study demonstrates the need for investigations throughout the year, from month to month, regarding the correct interpretation of airborne spore relationships with meteorological parameters.  相似文献

Abstract

Cedrela species are highly valued because of the quality of their timber. Understanding the behaviour of each different Cedrela species and their ecology is of importance to ensuring that forest harvesting and management do not endanger the survival of natural populations. These species grow in a wide range of environmental gradients and different types of forests in Bolivia. This study used dendrochronological methods to analyse growth–precipitation relationships of two Cedrela species coming from three locations with different environmental conditions: dry Chiquitano (Concepción), Chiquitano transitional Amazonian (Guarayos), and Bolivian-Tucuman montane forests (Postrervalle). The rainy season in all locations runs from October to April and the dry season runs from May to September. Twelve Cedrela fissilis specimens were sampled from dry Chiquitano, 11 Cedrela fissilis specimens from Chiquitano transitional Amazonian, and 30 Cedrela angustifolia specimens from Bolivian-Tucuman montane forests. The samples were crossdated and exhibited a common signal between trees from three sites, despite tree rings from the Chiquitano transitional Amazonian forest being narrower and displaying blurred bands of parenchyma in the boundaries. Significant inter-series correlation was found for the C. fissilis species series from dry Chiquitano with r = 0.261 (p < 0.01) and Chiquitano transitional Amazonian forests with r = 0.284 (p < 0.01), and for Cedrela angustifolia from Bolivian-Tucuman montane forests with r = 0.374 (p < 0.01). Mean annual growth was 2.07, 1.92, and 2.82 mm year?1 at the three sites, respectively. Cedrela species from dry Chiquitano and Bolivian-Tucuman montane forests were sensitive to precipitation from October to April of the current growth year (wettest season) and to low temperatures from May to July of the current growth year (driest season). Samples from Chiquitano transitional Amazonian were more sensitive to precipitation during late rainy season (March, April, and May of the current growth year) and high temperatures during the rainy months (November–December). Growth differences between sites and species in response to climate variations and local conditions should be taken into account and handled with different forest management guidelines.

Wheat is the most important cereal produced in Iran. A mycological survey was carried out for the first time, on the stored wheat samples in Tehran, East Azarbayejan and Mazandaran provinces in 2007. Exogenous and endogenous fungi, were isolated by the method of flotation with Malachite green agar (MGA 0.25) and Freeze blotter techniques respectively. In this study, 46 species belonging to 23 different genera were isolated.Cladosporium spp. (57.1–89.2%) andAlternaria spp. (82.4–100%) species were the predominant fungal species identified as endogenous mycoflora. The predominant exogenous fungi werePenicillium spp. (78.4–92.8%) andAspergillus spp. (71.4–85.7%) species.Fusarium proliferatum was the most prevalent species ofFusarium isolates.Aspergillus niger (39.4%) andAspergillus flavus (36.7%) were the predominantAspergillus species identified as exogenous mycoflora.Aspergillus flavus (26.6%) was the predominantAspergillus species identified as endogenous mycoflora. Flotation method with MGA 0.25 recommended for isolating of hyaline fungi from wheat cereals. In this study one isolate fromFusarium species was isolated on the basis of morphology and ribosomal internal transcribed spacer classified asFusarium langsethiae but on the basis of partial translation elongation factor-1alpha gene grouped withFusarium sporotrichioides. To our knowledge, this is the first report aboutF. cf.langsethiae in Iran and Asia.  相似文献

For the enhancement of lipase stability in organic solvent containing reaction, live immobilization method, using Bacillus subtilis spore as a display vehicle was attempted. Bacillus subtilis coat protein cotE was used as an anchoring motif for the display of lipA and lipB of Bacillus subtilis. Using this motif, lipolytic enzyme Lipase A and Lipase B were functionally displayed on the surface of Bacillus subtilis spore. Purified spore displaying CotE-LipB fusion protein showed higher lipolytic activity compared to that of CotE-LipA fusion protein. The surface localization of Lipase B was verified with flow cytometry and protease accessibility experiment. Spore displayed lipase retained its activity against acetone and benzene which completely deactivated free soluble lipase in the same reaction condition.  相似文献

A complex study on the adaptation of cn and vn mutants and the allozymes of alcoholdehydrogenase (ADH) was carried out in initially pure lines, and their panmixia populations during exchange of the mutant genotype with that of wild-type flies (C-S) and D) through saturating crossings. The relative adaptation of the genotypes was estimated by their effect on reproductive efficiency in the experimentally obtained population. Fecundity, lifespan, and the resistance of the studied genotypes to hyperthermia were investigated individually. It was shown that the high level of adaptation of the cn mutants and the low level of adaptation of the vg mutants was correlated with the presence of different ADH allozymes. In the studied population, the F-allozyme of ADH accompanied the vg mutation, while the S-allozyme of the enzyme was detected in cn mutants. Saturating crossings of C-S(Adh Svg(Adh F) and D(Adh F) × cn(Adh S), along with the parallel determination of the allele composition of the Adh locus, demonstrated that the complete substitution of the F-allozyme of ADH in the vg mutants by the S-allozyme in D flies, as well as the substitution of the S-allozyme of ADH in the cn mutants by the F-allozyme in D flies was realized only after the 15th–20th backcrosses. These results favor the coadaptation of cn and vg marker genes with alleles of the Adh locus and indicate the important role of the latter in the adaptation of genotypes. In the studied population, selection acted primarily against the vg mutants, which were inferior to the cn mutants, and heterozygote genotypes in indices of the main adaptation components.  相似文献
