Salinity-induced Malate Accumulation in Chara   总被引:3,自引:0,他引:3  
Ion absorption by Chara corallina from solutions containingpredominantly KC1 or RbCl at up to 100 mol m–3 resultedin accumulation of salts and turgor regulation. Turgor regulationdid not occur in solutions containing Na+ or Li+salts. Duringion absorption from various salts of K+ and Rb+ vacuolar cationconcentration exceeded Cl concentration. This differencewas shown to be balanced by the synthesis and accumulation ofmalate. Vacuolar malate concentration reached 48 mol m3,with accumulation occurring at rates of up to 0.45 mol m–3h–1. Malate accumulation was inhibited by low externalpH and was dependent upon external HCO3 concentration.The synthesis of malic acid and its subsequent dissociationimposed a severe acid load on the cell. Biophysical regulationof cellular pH was achieved by a H+efflux at a rate of about40 nmol m–2 s–1from the cell. The results presentedargue against cytoplasmic Cl, HCO3 or pH regulatingmalate accumulation in Chara and it is suggested that malatetransport across the tonoplast may regulate malate accumulation. This paper describes experiments designed to investigate theeffects of increases in external osmotic pressure on the electrophysiologicalbehaviour of the plasmalemma in cells of the brackish-wateralga, Chara inflata. The electrical conductance of the plasmalemmaof these cells of, due to the diffusion of ions, consists mainlyof K+, Cl and leak components. The addition of sorbitolat concentrations in the range 40–280 mol m–3 tothe external solution bathing the cells, progressively and reversiblydepolarized the cell membrane and increased the total membraneconductance, chiefly due to increases in each of the separateionic conductances. At concentrations higher than about 280mol m–3 when the cells became plasmolysed, the effectsof sorbitol on the electrical properties of the cell ceasedto be fully reversible. When the membrane electrical potentialdifference is stepped in a negative direction with a voltage-clamp,the resulting inward current has voltage-dependent componentsconsisting of an inactivating K+ current, an activating Clcurrent and a constant leak current. The addition of sorbitolto the external solution modified these currents by increasingtheir magnitude, by increasing the half-time of the inactivationof the K+ current, and by decreasing the half-time of activationof the Cl current. Following 20 d of exposure to 75 or 150 mol m–3 NaCl Sorghumbicolor (L.) Moench plants become capable of growing in mediumcontaining 300 mol m–3 NaCl. Control plants, which havenot been pretreated, or plants pretreated for less than 20 ddie within 2 weeks when exposed to 300 mol m–3 NaCl. Weconsider this induction of a capacity to survive in and toleratea high NaCl concentration as an adaptation to salinity. We suggestthat adaptation to salinity is more than osmotic adjustmentand that it takes longer to develop than osmotic adjustment.Concomitantly with the appearance of the ability to grow inhigh salinity, adaptation also comprises the development ofa capacity to regulate internal Na+ and Cl concentrations,even when external salinity is high. Shoot mean relative growthrates are similar for both control plants and for adapted plantsgrowing in 300 mol m–3 NaCl, although their shoot Na+and Cl concentrations are quite different. Based on thesedata, we propose that adaptation of Sorghum to high salinityresults from a modulation of genome expression occurring duringextended exposure to non-lethal NaCl concentrations. Ammonium and methylammonium ions greatly increase the rate ofCl transport in Chara corallian. This effect is dependenton the pH of the bathing solution. The amine-stimulated Clinflux is small at pH 5·5, increases to a maximum atpH 6·5–7·5, and decreases again as the pHis raised to 8·5. Increased Cl influx is accompaniedby an increase in cytoplasmic pH, as calculated from the distributionof DMO. When the external pH lies between 5·5 and 7·3,cytoplasmic pH in the absence of amine is 7·65–7·70,with an increase of 0·15–0·25 in the presenceof amine. As external pH is increased above 7·3, cytoplasmicpH also increases, with progessively less effect of amine. Although the relationship between Cl influx and cytoplasmicpH is not simple, the results provide evidence in accord withthe hypothesis that Cl transport in Chara involves H+—Clsymport, or the equivalent OH—Cl antiport.The possible role of cytoplasmic pH as a factor involved inthe regulation of membrane transport in Chara is discussed.  相似文献

Bowling, D. J. F. 1987. Measurement of the apoplastic activityof K+ and Cl in the leaf epidermis of Commelina communisin relation to stomatal activity.–J. exp. Bot. 38: 1351–1355. Ionic activity of K+ and Cl in the apoplast of the lowerepidermis of the leaf of Commelina communis was measured usingion selective micro-electrodes. Large gradients across the stomatalcomplex were observed which were related to stomatal aperture.On stomatal closure the activity of K+ and Cl in theapoplast of the guard cell rose from 3·0 mol m–3to 100 mol m–3 and 33 mol m–3 respectively. In springwater (25.5 mol m–3 Cl, 20.4 mol m–3Na+, 0.14 mol m–3 K+) Enteromorpha intestinalis couldnot survive for more than a few weeks unless provided with 0.5mol m–3 K+ in the medium or alternatively exposed to seawaterfor 1 day per week. Maintenance of a cytoplasmic K+ level ofabout 200 mol m–3 is critical for the maintenance of normalmetabolic activity. Net gains of intracellular K+ occurred whenthe plants were transferred from low-salinity to seawater; converselylarge net losses occurred when plants were transferred fromseawater to springwater. These two processes were not simplythe reverse of one another; net gain of K+ involved a largeincrease in the tracer flux both into and out of the cell butnet loss of K+ virtually halted the tracer flux into the cell.Any injury incurred by rapid salinity changes was short-lived;plants were rapidly able to adjust intracellular [K1.K+). K+(orto some extent Rb+) was found to be necessary in the effluxmedium for 42K+ exchange to occur. The osmotic concentrationof the medium was also important but extracellular Na+ and Clconcentrationswere not critical. K+ influx and efflux in both springwaterand seawater were largely independent of light and were sensitivein varying degrees to a range of common metabolic inhibitorsand uncouplers. The results are best explained by the presenceof an active K+ influx, generated by an ATP-dependent K+ pumpat the plasmalemma. Use of permeabilised cells of Chara corallina provides a uniqueopportunity to study the electrical characteristics of the tonoplastwhilst being able to control ionic conditions on the outsideof the membrane. Current-voltage (I/V) analysis over wide voltagespans, and admittance measurements at 5 Hz showed that manypermeabilised cells had a similar conductance and capacitanceto the tonoplast of intact cells. Cells developed two regionsof negative-slope conductance upon addition of external Cl,which suggests the existence of potential-dependent Clchannels in the Chara tonoplast. The pH of the cytoplasm of Chara corallina cells has been measuredwith the weak acid 5,5-dimethyloxazolidine-2,4-dione (DM0).Over an external pH range 4·5–9·5 the resultsfit the regression equation pHcytoplasm=6·28+0·22pHout. Using measured values of the electric potential difference acrossthe plasmalemma we have calculated the electrochemical potentialdifference across this membrane for H+ and Cl. Thesedata are used to test the hypothesis that the inward transportof Cl is coupled to the inthix of H+ or, which comesto the same thing, efflux of OH. One-for-one couplingwill not give net Cl uptake from solutions with pH greaterthan about 7·2, unless the cytoplasmic Cl concentrationis lower than 10 mM, or the pH just outside the membrane islower than that in the bulk solution. It is shown that net Cluptake proceeds from solutions with pH up to 9. The alternative possibility is that Cl transport is broughtabout by co-transport of two H+ for each Cl; this isnot ruled out by the results reported. Such a mechanism mightbe detectable by its electrogenic effect: although such effectshave not been detected, it is shown that they would be smallunder most conditions. The early time-course of turgor adjustment following a hyper-or hypo-osomotic shock was examined in the brackish-water charophyteLamprothamnium papulosum. The response to a reduction in turgorwas a five to seven-fold stimulation of the influxes of Cl,K+ and Na+. The distribution of radioactive tracers in the cellsuggested that the ionic composition of the cytoplasm was strictlycontrolled during turgor adjustment. Metabolic activity wasrelatively unaffected by the loss of turgor. high fluxes throughthe cytoplasm, and a cytoplasmic K concentration possiblyas high as 280 mol m–3. Osmotic adjustment to a lower salinity was achieved by largeincreases in the passive effluxes of K+ and Cl ratherthan by decreases in their influxes. The membrane remained hyperpolarized during hyperosmotic adjustmentbut depolarized after a hypo-osmotic change. A method has been developed to measure the cell volume of theunicellular green alga Dunaliella parva 19/9 using Li+ measurementsonly. Concentrations of internal solutes can also be calculatedif they are assayed in the same samples as Li+. We found thatD. parva cells grown in 0.4 kmol m–3 NaCl have an averageaqueous cell volume of 65.1 ?2.9 µm3, a K+ concentrationof 126?6 mol m–3, a Na+ concentration of 11?11 mol m–3and a glycerol concentration of 615?27 mol m–3 (n= 12).Algae grown in 1.5 kmol m–3 NaCl have an average aqueouscell volume of 131 ?7.5 µm3, a K+ concentration of 109?4mol m–3, a Na+ concentration of 10?39 mol m–3 anda glycerol concentration of 1 425?59 mol m–3 (n = 12).These results indicate that D. parva cells adapted to high salinitieshave larger cell volumes than those adapted to lower salinities.However, there is no evidence for a significant difference ininternal Na+ concentration, despite the almost 4-fold differencein the concentration of external NaCl. The intracellular glycerolconcentration alone accounts for 65% and 54%, respectively,of the osmotic balance in low and high salt grown cells. A Cl channel and two types of K+ channel have been observed,by the use of the patch-clamp technique, in the membrane surroundingcytoplasmic droplets from Chara corallina. Measurements on cell-attachedpatches showed that the channel selective for Cl hada chord conductance of 21 pS at the resting membrane p.d. (mean= 11 mV, n = 19) and when open, passed an outward current of1.4 pA (n = 24 patches) at the resting p.d., with reversal ofthe direction of current at –54 mV (130 mol m–3Cl in the external solution). The Cl concentrationin the cytoplasmic droplet calculated from the reversal p.d.was 15 mol m–3. The channel strongly rectified outwardcurrent flow, but this rectification disappeared with symmetricalCl concentrations across detached patches of membrane.It is concluded that rectification observed in cell-attachedpatches is primarily due to asymmetric Cl– concentrationsrather than an asymmetry in energy barriers to Cl permeationin the channel or any voltage-dependent kinetics of the channel.The channel was rarely observed in detached patches despitebeing commonly observed in cell-attached patches. However, theabsence of Ca2+ at the cytoplasmic face of the membrane allowedobservation of the channel in detached patches for brief periods,during which ion substitution experiments revealed a permeabilitysequence of aspartate (76:33:1). A 100 pS K+ channel previously described by Luhring (1986) wasfrequently observed, in some instances simultaneously, witha channel having a conductance of 60 pS and displaying outwardrectification. This rectification was due to the channel remainingopen almost continuously for positive membrane potential differences(p.d.) and remaining shut almost continuously for negative p.d.'s.The 60 pS channel, like the 100 pS K+ channel, reversed currentflow at the resting p.d., suggesting that it was also permeableto K+. Ammonia (pKa 9.25) and methylamine (pKa, 10.65) increase cytoplasmicpH and stimulate Cl influx in Chara corallina, theseeffects being associated with influx of the amine cations ona specific porter. The weak base imidazole (pKa 6.96) has similareffects but diffuses passively into the cell both as an unionizedbase and as a cation. When the external pH is greater than 6.0influx of the unionized species predominates. Imidazole accumulates to high concentrations in the vacuole,where it is protonated. Cytoplasmic pH and vacuolar pH riseby only 0.2–0.3 units, suggesting a large balancing protoninflux across the plasma membrane. Balance of electric chargeis partially maintained by net efflux of K+ and net influx ofCl. Calculation of vacuolar concentrations of imidazole(from (14C] imidazole uptake, assuming that there is no metabolism)plus K+ and Na+ indicates an excess of cations over inorganicanions (Cl). However, although the osmotic potentialof the cells increases, also indicating increased solute concentrations,the increase is less than that predicted by the calculated ionicconcentrations. This discrepancy remains to be resolved. Becausethe osmotic potential also increases when imidazole is absorbedfrom Cl-free solutions it is likely that maintenanceof charge-balance can also involve synthesis and vacuolar storageof organic or amino acids. The effects of different NaCl concentrations on the growth andnitrogen fixation activity of white lupin (Lupinus albus [L.])was studied over a 6 d period. Plant growth parameters, photosynthesisand shoot respiration were unaffected by NaCl concentrationsup to 150 mol m–3. However, nitrogenase activity decreasedwith increased NaCl concentration up to 100 mol m–3, whilstthe O2 diffusion resistance increased with 100 mol m–3NaCl, but showed no further change when 150 mol m–3 NaClwas applied for 6 d. Increases in NaCl concentration decreasednodular starch content while increasing sucrose content, suggestingan osmotic regulation. These changes were associated with a77% decrease in sucrose synthase activity. The effect on theO2 diffusion resistance was paralleled by changes in glycoproteincontent of the nodules, as determined by immunogold localizationand ELISA. X-ray microanalysis studies of nodules showed that,following a 6 d exposure to 150 mol m–3 NaCl, Na+ ionswere largely excluded from the infected zone, whilst only lowlevels of Cl- ions penetrated into this region. Na+ entry intoroots and leaves was also at a low level. Leghaemoglobin contentdecreased with saline stress, as did superoxide dismutase; whichdecreased by 36% following exposure to 100 mol m–3 saltfor 6 d. These results are discussed in relation to the relativesalt tolerance of the Multolupa/ISLU-16 symbiosis. Salt Tolerance in the Succulent, Coastal Halophyte, Sarcocornia natalensis   总被引:2,自引:0,他引:2  
The effects of 0, 50, 100, 200, 300, 400 and 500 mol m–3NaCl on growth and ion accumulation in the succulent, coastalhalophyte Sarcocornia natalensis (Bunge ex Ung.-Sternb.) A.J. Scott were investigated. Increase in salinity from 0 to 300 mol m–3 NaCl stimulatedproduction of fresh, dry, and organic dry mass, increased succulenceand shifted resource allocation from roots to shoots. Growthwas optimal at 300 mol m–3 and decreased with furtherincrease in salinity. Water contributed to a large proportion of the increase in freshmass. Inorganic ions, especially Na+ and Cl– contributedsubstantially to the dry mass. At 300 mol m–3 NaCl inorganicions contributed to 37% of total dry mass and NaCl concentrationin the shoots was 482 mol m–3. Expressed sap osmotic potentialsdecreased from –2.10 to –3.95 MPa as salinity increasedfrom 0 to 300 mol m–3 NaCl. Massive accumulation of inorganicions, especially Na+ and Cl, accounted for 86% of theosmotic adjustment at 300 mol m–3 NaCl. Salinity treatments decreased the concentrations of K+ in shoots.Plant Na+ :K+ ratios increased steadily with salinity and reacheda maximum of 16.6 at 400 mol m3 NaCl. It is suggested that the exceptional salt tolerance of S. natalensisis achieved by massive inorganic ion accumulation which providessufficient solutes for osmoregulation, increased water fluxand turgor-induced growth. The contribution of membrane transport to regulation of cytoplasmicpH in Chara corallina has been measured during proton-loadingby uptake of butyric acid. In the short-term (i.e. up to 20min) uptake of butyric acid is not affected by removal of externalK+, Na+ or Cl but over longer periods uptake is decreased(by 20–50% in different experiments) in the absence ofexternal Na+ or, sometimes, K+. Influxes of both Na+ and K+increase temporarily after addition of butyrate, Na+ immediatelyand K+ after a lag. Effects on Cl influx are small butCl efflux increases enormously after a short lag. Anapproximate comparison of internal butyrate with changes inthe concentration of K+, Na+, and Cl suggests that initially(i.e. for a few min) cytoplasmic pH is determined by bufferingand possibly by some decarboxylation of organic acids (biochemicalpH regulation), and that biophysical pH regulation involvingefflux of H+ balanced by influxes of K+, Na+ and especiallyefflux of Cl progressively becomes dominant. When butyric acid is washed out of the cells, cytoplasmic pHis restored completely or partially (depending on the butyrateconcentration used) and this is independent of the presenceor absence of external Cl. Where Cl is present,its influx is relatively small. It is suggested that cytoplasmicpH is then controlled biochemically, involving the synthesisof an (unidentified) organic acid and the accumulation of acidicanions in place of butyurate lost from the cell. During thesecond application of butyrate, net Cl efflux is small:it is suggested that control of cytoplasmic pH then involvesdecarboxylation of the organic acid anions. The questions of the source of Cl lost from the cell(cytoplasm or vacuole) and of possible cytoplasmic swellingassociated with the accumulation of butyrate are discussed. Na+, K+ and Cl- in Xylem Sap Flowing to Shoots of NaCl-Treated Barley   总被引:7,自引:0,他引:7  
Munns, R. 1985. Na+, K+ and Cl in xylem sap flowing toshoots of NaCl-treated barley.—J. exp. Bot. 36: 1032–1042. Na+, Cl and K+ concentrations were measured in xylemsap obtained by applying pressure to the roots of decapitatedbarley plants grown at external [NaCl] of 0, 25, 50, 100, 150and 200 mol m–3. For any given NaCl treatment, ion concentrationsin the xylem sap were hyperbolically related to the flux ofwater. Ion concentrations in sap collected at very low volumefluxes (without applied pressure) were 5–10 times higherthan in sap collected at moderate fluxes (under pressure). Fora given moderate volume flux, Na+ concentration in the xylemsap, [Na+]x, was only 4.0 mol m–3 at external [NaCl] of25–150 mol m–3, and increased to 7.0 mol m–3at 200 mol m–3. [Cl-]x showed a similar pattern. Thisshows there would be little difference in the rate of uptaketo the shoot of plants at 25–150 mol m–3 externalNaCl and indicates little change even at 200 mol m-3 NaCl becausetranspiration rates would be much lower. Thus the reduced growthof the shoot of plants at high NaCl concentrations is not dueto higher uptake rates of Na+ or Cl. The fluxes of Na+, Cl and K increased non-linearlywith increasing volume flux indicating little movement of saltin the apoplast. The flux of K+ increased even when [K+]x wasgreater than external [K+], indicating that membrane transportprocesses modify the K+ concentration in the transpiration streamas it flows through the root system. The electrical conductance of the plasmalemma of cells of Charainflata, due to the diffusion of ions, consists predominantlyof K+, Cl and leak components. When the membrane electricalpotential difference is stepped in a negative direction witha voltage-clamp, the resulting inward current has componentsIK, ICl and IL (leak). During such voltage-clamp steps IK isinactivated, and Ic activated with voltage-dependent half-times.Increases in the external NaCl concentration reduce the magnitudeof IK and increase the magnitude of Ic, but reduce the half-timeof inactivation or activation. The NaCl-induced changes in Ikand ICl and their kinetics were more pronounced at pH0 =6.5than at pH0 =9.5. When the concentration of external CaCl2 wasincreased, Ik, ICl and the half-time of inactivation, (T1/2),of Ik were all reduced. The half-time of activation of ICl wasincreased. The NaCI-induced changes could result from increases in bothexternal ion concentration and osmotic pressure. Previous experimentshave shown that an increase in external osmotic pressure alonealters the properties of the conductances. Allen, S. and Smith, J A. C. 1986. Ammonium nutrition in Ricinuscommunis: its effect on plantgrowth and the chemical compositionof the whole plant, xylem and phloem saps.—J. exp. Bot.37: 1599–1610. The growth and chemical composition of Ricinus communis cultivatedhydroponically on 12 mol m – 3 NO3-N were comparedwith plants raised on a range of NH4+-N concentrations. At NH4+-Nconcentrations between 0·5 and 4·0 mol m–3,fresh- and dry-weight yields of 62-d-old plants were not significantlydifferent from those of the NO3-N controls. Growth wasreduced at 0·2 mol m–3 NH4+-N and was associatedwith increased root. shoot and C: organic N ratios, suggestingthat the plants were N-limited. At 8·0 mol m–3NH4+-N, growth was greatly restricted and the plants exhibitedsymptoms of severe ‘NH4+ toxicity’. Plants growingon NH4+-N showed marked acidification of the rooting medium,this effect being greatest on media supporting the highest growthrates. Shoot carboxylate content per unit dry weight was lower at mostNH4+-N concentrations than in the NO3-N controls, althoughit increased at the lowest NH4+-N levels. Root carboxylate contentwas comparable on the two N sources, but also increased substantiallyat the lowest NH4+-N levels. N source had little effect on inorganic-cationcontent at the whole-plant level, while NO3 and carboxylatewere replaced by Cl as the dominant anion in the NH4+-N plants.This was reflected in the ionic composition of the xylem andleaf-cell saps, the latter containing about 100 mol m–3Cl in plants on 8·0 mol m–3 NH4+. Xylem-saporganic-N concentration increased more than threefold with NH4+-N(with glutamine being the dominant compound irrespective ofN source) while in leaf-cell sap it increased more than 12-foldon NH4+-N media (with arginine becoming the dominant species).In the phloem, N source had little or no effect on inorganic-cation,sucrose or organic-N concentrations or sap pH, but sap fromNH4+-N plants contained high levels of Cl and serine. Collectively, the results suggested that the toxic effects ofhigh NH4+ concentrations were not the result of medium acidification,reduced inorganic-cation or carboxylate levels, or restrictedcarbohydrate availability, as is commonly supposed. Rather,NH4+ toxicity in R. communis is probably the result of changesin protein N turnover and impairment of the photorespiratoryN cycle. Bean plants (Phaseolus vulgaris L. cv. Fardenlosa Shiny) werelabelled with carbon-11 via their first trifoliate leaves when3-weeks-old and the transient inhibitions of translocation causedby the application of ammonium chloride solutions (10 mol m–3)to a peeled region of stem were studied. At pH 6·5 theammonium was without effect. At pH 11·0 even a briefapplication inhibited translocation for many minutes, whilelonger applications inhibited translocation for considerablylonger. Solutions of 10 mol m–3 sodium chloride were withouteffect at either pH. At pH 6·5 ammonium chloride solution contains predominantlyammonium ions (NH4+) and at pH 11·0 predominantly dissolvedammonia gas (NH3). Hence we conclude that phloem transport withinbean stems is inhibited by dissolved ammonia gas but not ammoniumions.
