Inoculum production of Valdensinia heterodoxa Peyr. (isolate PFC 3027), a foliar pathogen of salal (Gaultheria shallon Pursh.), was evaluated on various solid substrates. Incubation on salal leaf pieces resulted in the greatest number of discharged conidia, which is deemed a suitable indicator for sporulation and inoculum dispersal of V. heterodoxa. Subsequently, V. heterodoxa inoculum of two isolates (PFC 2761 and PFC 3027) was produced on salal leaf pieces using four different growth periods (0, 4, 8, or 12 days) and three different cold storage treatments (0, 4, or 8 weeks) following harvest of colonised leaf pieces. Overall, the maximum number of discharged conidia was obtained with a growth period of 4 or 8 days, whereas the effect of a cold storage treatment was inconclusive between the fungal isolates and various growth periods. Conidia discharge from salal leaf piece inoculum of V. heterodoxa (PFC 2761 and PFC 3027) applied on pot soil level resulted in 8-41% leaf damage of intact salal plants. Results from this study indicate the potential of salal plant tissue as both a growth substrate and a suitable delivery technique for V. heterodoxa to control salal.  相似文献

Valdensinia heterodoxa (Sclerotiniacae) is a potential fungal bioherbicide for control of salal (Gaultheria shallon). The effect of culture media, substrates and relative humidity (RH) on growth, sporulation and conidial discharge of V. heterodoxa was determined for two isolates PFC2761 and PFC3027 in vitro. Culture media significantly affected the growth, sporulation, and conidial discharge of V. heterodoxa. Of eight agar media used, colony radial growth was optimal on salal oatmeal agar and salal potato dextrose agar for isolates PFC2761 and PFC3027, respectively; whereas sporulation was at an optimum on salal oatmeal agar for both isolates. Of the eight liquid media tested, mycelial production was highest on wheat bran–salal–potato dextrose broth. Growth on solid substrates greatly stimulated sporulation and conidial discharge of V. heterodoxa. Of the 12 solid substrates used, the greatest numbers of discharged conidia were observed from wheat bran and wheat bran–salal within 14 d of sporulation. Sporulation on solid substrates continued for 42 d. RH significantly affected the sporulation and conidial discharge for both isolates across all solid substrates tested. No conidia were produced or discharged below 93 % RH on wheat bran–salal and millet. With an increase of the RH from 93 to 97 %, sporulation and the number of discharged conidia increased significantly for both isolates on wheat bran–salal, but not on millet.  相似文献

On agar media the maximum conidia production of Exserohilum monoceras occurred on V-8 juice agar (VA) or centrifuged V-8 juice agar, whereas the optimal radial mycelial growth occurred on Czapek-Dox agar. The optimal temperatures for radial mycelial growth and conidia production were 28 and 27°C respectively. Light prohibited E. monoceras conidia production. The best sporulation occurred under continuous dark conditions. Echinochloa leaf decoction significantly increased conidia production on potato dextrose agar (PDA) and VA, and significantly increased germ tube length on PDA, lima bean agar and VA, but did not affect conidia germination. No conidia were produced in liquid media. Of 22 agricultural-based products evaluated as solid substrates, the most abundant sporulation (1.8 × 106 conidia g-1 of dry weight) occurred on corn leaves. The conidia production of E. monoceras on corn leaves was affected by incubation period, moisture content and substrate quantity. There were no differences in germination rate, germ tube length and virulence of conidia produced on agar media or corn leaves.  相似文献

The effect of temperature regimes on conidia germination, radial growth and virulence of Beauveriabassiana and Metarhizium anisopliae against the spotted stalk borer Chilo partellus was investigated with Ethiopian isolates. Conidia germination, radial growth and sporulation of all isolates were retarded at 15 and 35°C. A suitable temperature range for the isolates was between 20 to 30°C. Conidia germination was less tolerant of low temperature (15°C) than radial growth. Radial growth and sporulation reacted differently to temperature. At both 25 and 30°C, all isolates induced 100% mortality to C. partellus larvae in six days. The LT50 decreased for the isolates with increasing temperature.  相似文献

Selected cultural and environmental variables were investigated for their influence on the efficacy of Phoma herbarum and Phoma exigua to cause disease on dandelion (Taraxacum officinale) under growth room conditions. In both species, mycelial fragments caused significantly greater disease severity on dandelion than spore suspensions. Mycelial age was not an important factor in disease severity caused by P. herbarum, with all cultures causing high disease ratings. However, younger cultures of P. exigua caused the greatest disease severity on dandelion, but significantly less than that caused by P. herbarum. The initial pH of the growth medium (potato dextrose broth) did not affect disease severity caused by either Phoma species. Increasing concentrations of mycelia of P. herbarum were applied to dandelions that were then exposed to various leaf wetness durations. Disease severity increased with increasing leaf wetness duration. For dandelions exposed to no leaf wetness duration, the greater the mycelial concentration, the greater the disease rating. However, for dandelions exposed to all leaf wetness durations, all concentrations of mycelia caused similar disease ratings. As P. herbarum caused high disease ratings on dandelion, it therefore warrants further investigation as a potential bioherbicide for the control of this weed.  相似文献

Paecilomyces lilacinus and in particular the commercial strain 251 has been intensively tested for biological control of plant parasitic nematodes. Since this species has been mentioned in a number of reports concerning infection of humans, the human health risk for Paecilomyces lilacinus strain 251 was investigated. The effects of time, temperature and growth medium on radial colony growth and germination were determined. Additionally, exposure to 36°C and its effect on germ-tube extension and on survival of conidia was evaluated. Radial growth was significantly affected by temperature, growth medium and their interaction. Optimum temperatures were between 24 and 30°C, but no growth was found at 36°C. Germination rate was significantly influenced by time, medium, temperature and their interactions. The optimum temperature range for germination was between 28 and 30°C. Formulated conidia were capable of germinating at 36°C. However, studies on germ-tube extension conducted at 36°C showed a delay in development for 28-49 h and no further germ-tube extension was found after exposure for 80-95 h. Slopes of survival curves were significantly influenced by the type of conidia tested. In general, conidia did not survive exposure to 36°C for 168 h. These experiments indicate the temperature conditions where the strain is likely to be active and provide supporting data for full environmental and health risk assessments of biocontrol fungi.  相似文献

In shake flask and fermentor studies, various media components and culture inocula were tested to improve P. fumosoroseus spore production rates, yield and stability. To evaluate inoculum potential and inoculum scale-up for fermentor studies, conidia and liquid culture-produced spores of various strains of P. fumosoroseus were compared as inoculum. Inoculation of liquid cultures with blastospores at concentrations of at least 1×106 spores mL-1 resulted in the rapid production of high concentrations of blastospores (∼1×109 spores mL-1, 48 h fermentation time) for all strains tested. The rapid germination rate of blastospores (90% after 6 h incubation) compared to conidia (>90% after 16 h incubation) and the use of higher inoculum rates reduced the fermentation time from 96 to 48 h for maximal spore yields. A comparison of various complex nitrogen sources showed that liquid media supplemented with acid hydrolyzed casein or yeast extract supported the production of high concentrations of blastospores that were significantly more desiccation-tolerant (79-82% survival after drying) when compared to blastospores produced in media supplemented with other nitrogen sources (12-50% survival after drying). For rapid spore production, requirements for trace metals and vitamin supplementation were dependent on the type of hydrolyzed casein used in the medium. Fermentor studies with two strains of P. fumosoroseus showed that high concentrations (1.3-1.8×109 spores mL-1) of desiccation-tolerant blastospores could be produced in 48-h fermentations. These studies have demonstrated that the infective spores of various strains of the fungal bioinsecticide Paecilomyces fumosoroseus can be rapidly produced using deep-tank, liquid culture fermentation techniques.  相似文献

Tween 70 at 0.1% provided the best conditions for germination of Aschersonia placenta. Optimum germination and growth of the germ tube occurred over a temperature range of 25–30°C and a pH range of 5.0–6.0. A temperature of 30°C resulted in the longest germ tube at 45 μm. Apparently, temperature and pH did not affect the type of germination, with polar germination being consistently recorded for in excess of 60% of conidia. In general, growth and sporulation seemed to be much better in semi‐solid than in liquid media. Amongst several plant media tested, pumpkin consistently gave the most mycelial growth and sporulation. The ability of A. placenta to sporulate on the surface of liquid culture has increased the possibilty of its mass production for the purpose of formulating a microbial pesticide against the indigenous scale insects of tropical fruits such as durian and guava in Malaysia.  相似文献

Solar radiation, particularly its UV wavelengths, greatly influences conidium survival and this study looked at the impact of radiation and its interactions with temperature on Plectosporium alismatis and Colletotrichum orbiculare, two fungi that are potential mycoherbistats. UV radiation, rather than temperature, was found to be the primary cause of conidium mortality; however, there were interactions between these factors leading to the enhancement of the lethal effects of UVB on conidium germination at high temperatures. C. orbiculare was more sensitive than P. alismatis with conidium germination being halved by UVB doses of 1.47 and 13.1 kJ m-2, respectively, for the two pathogens. Conidium mortality was dose-dependent and for P. alismatis exposed to a dose of 3.7 kJ m-2 reciprocity was observed. However, for C. orbiculare equivalent doses were not reciprocal as higher doses for short periods were more lethal than lower doses of longer duration. Low UVB doses only caused delays in conidium germination, whereas higher doses killed conidia and caused delays in the germination of any survivors. Radiation also affected appressorium formation. Appressorium formation was stimulated by UVA and was dose dependent with P. alismatis requiring a higher dose than C. orbiculare to initiate formation. Microcycle conidiation by P. alismatis was observed following exposure to sunlight. This knowledge of how conidia of these potential mycoherbistats react to climate suggests that rapid conidium germination and appressorium formation could be achieved by manipulation of the time at which they are applied in the field. Conidia could be applied so that they receive sufficient UVA to stimulate appressorium formation but without receiving a dose that would significantly affect conidium germination. However, for this, additional protection from UVB may be needed.  相似文献

光在调控真菌的多种生理过程中发挥着重要作用。为探究光照对新蚜虫疠霉Pandora neoaphidis产孢的影响,本文研究了不同波长光源(蓝光、绿光、白光、红光、黄光)和无光黑暗条件对新蚜虫疠霉分生孢子弹射能力的影响,通过cDNA末端的快速扩增(RACE)对新蚜虫疠霉的蓝光受体蛋白基因pnwc-1进行克隆并对其进行生物信息学分析,利用qRT-PCR对蓝光光源不同照射时长下pnwc-1的表达量进行了定量分析。结果表明,蓝色光源(波长460–465nm)照射后的新蚜虫疠霉菌丝产生的分生孢子数量显著高于其他波长光源,排序为:蓝光>绿光>白光>红光>黄光>无光。另外,分析克隆获得的全长为2 423bp的pnwc-1基因发现,其编码的蛋白具有蓝光受体蛋白典型的保守结构域,同源比对结果显示新蚜虫疠霉与接合菌门真菌归为一类但相对独立。qRT-PCR的定量分析结果表明随着照射时长增加,蓝光处理能显著提高pnwc-1的表达量,而且pnwc-1的相对表达量与累积产孢量存在正相关(R2=0.9798)。本研究为后续蓝光及其受体基因功能的深入研究提供了实验基础,并促进以新蚜虫疠霉为代表的虫霉目真菌在害虫生物防治中的应用。  相似文献

A method was developed for applying strains of Actinoplanes spp. that are hyper-parasites of oospores of Pythium ultimum to soil for reducing Pythium damping-off of plants. The method is based on the augmentation of soil with sporangia of a strain of Actinoplanes spp. borne on clay granules. In vitro sporulation of strains K30, W57, W257 and 25844 was: (1) greater for most strains on dilute Czapek-Dox agar than on four other agar media; (2) inhibited by continuous exposure to fluorescent light of intensity 4-150 μEm-2s-1, but not by exposure to 1 μEm-2s-1 or darkness; (3) greater at 20-307deg;C than at 10°C;and (4) greater at pH 6-7 than at pH 5 or 8. On solid carriers treated with dilute Czapek-Dox broth (pH 7) and incubated in the dark at 30°C for 3 weeks, strains sporulated poorly or not at all on vermiculite, perlite and rice hulls, but sporulated abundantly (107-109 colony-forming units (CFU) g-1 of granules) on montmorillonite clay granules. When strains 25844, W57 and W257 were applied as granules (4 107 - 4 × 108 CFU g-1) at 5% (w/w) to field plots infested with 750-1000 oospores of P. ultimum g-1 of soil, only strain 25844 consistently increased emergence and reduced root rot of table beets 8- 1 at 24-28 days after planting compared with controls. Strain 25844 (108 CFU g-1 of granules) at 1% (w/w) also increased the emergence of bush beans at 28 days after planting in P. ultimum-infested plots, but lower rates were ineffective. The inoculum viability of strain 25844 on clay granules declined 100-fold during 2 months of storage at 5-35°C, but thereafter remained stable for another 4 months. Strain 25844 on 6-month-old granules retained a high degree of hyper-parasitic activity toward oospores of P. ultimum. Augmentation of field soil with sporangia of Actinoplanes spp. is a valid approach to the biological control of pythium damping-off.  相似文献

Laboratory experiments were conducted to assess effects of nutrients on germination of Verticillium lecanii (=Lecanicillium sp.) conidia and infection of the greenhouse whitefly, Trialeurodes vaporariorum. Suspensions of V. lecanii conidia were prepared in four nutrient solutions: 2% glucose, 2% sucrose, 2% maltose, and 2% peptone. Suspensions in de-mineralized water served as the control. At 23°C the germination rate was highest in the 2% glucose solution, followed by sucrose, maltose, demineralized water, and peptone, respectively. Germ tube growth was greatest at 23°C in the 2% glucose solution after 10 h incubation. Results of the bioassays indicated that the nutrients influenced whitefly infection. Infection levels were highest for conidial suspensions (1×106 conidia/mL) prepared in 2% glucose, and were significantly greater than for peptone, demineralized water and maltose. Infection levels at 1×108 conidia/mL were not significantly different from each other for all materials tested. The potential use of nutrients in a spray formulation as a means of enhancing field efficacy are discussed.  相似文献

Effects of sixteen chemical pesticides on conidial germination of C. thromboides and P. nouryi were investigated at two concentrations (R and 0.2R, where R=the lowest manufacture's recommended concentration for field use). Of the fungicides tested, propamocarb hydrochloride did not significantly affect conidial germination of C. thromboides, but at R it completely inhibited conidial germination of P. nouryi. At 0.2R of procymidone, iprodione, haloxyfop-methyl and fenoxaprop-p-ethyl, conidia of C. thromboides had some germination after 24 h, although they entirely restricted conidial germination of the fungus at the concentration of R. Tribenuron-methyl, one herbicide, had no adverse effect on conidial germination of C. thromboides and P. nouryi after 6 h at the two concentrations, whereas the other seven herbicides were strongly antagonistic to P. nouryi, except for carfentrazone-ethyl at 0.2R.  相似文献

Growth of Coniothyrium minitans on potato dextrose broth was compared with that on an inexpensive molasses-yeast liquid medium at 18-22°C in static culture. Biomass and conidial production were, in general, similar, although the rate of biomass production was quicker and conidial production was slightly greater per unit volume of medium in the molasses-yeast medium. Air-dried biomass from molasses-yeast liquid culture containing mycelia, pycnidia and conidia of C. minitans was mixed (12%, w/w) with kaolin to give a kaolin-biomass dust. The ability of C. minitans to survive and subsequently infect and reduce the viability of sclerotia of Sclerotinia sclerotiorum from this kaolin-biomass dust was found to be little affected by storage for 48 weeks between 4 and 15°C but was decreased by higher storage temperatures. The kaolin-biomass dust preparation did not differ from a standard maizemeal-perlite inoculum of C. minitans in its ability to infect sclerotia of S. sclerotiorum or reduce their viability or carpogenic germination in glasshouse and field pot bioassays. Further, when either inoculum was applied once to glasshouse soil naturally infested with S. sclerotiorum prior to planting three successive crops of lettuce, the pattern of disease control, reduction of sclerotial numbers/ plot, infection of sclerotia, reduction of sclerotial viability and survival in soil were similar for both inocula. The potential for the commercial development of liquid-culture-produced inocula of C. minitans is discussed.  相似文献

This paper presents results on the effect of light, temperature and substrate during spore formation on the germinability of conidia in Colletotrichum falcatum. Light seems to have no effect on the germination of conidia unless the cultures were exposed to a high intensity of light during sporulation, in which case the spores showed a reduced germination and an increased appressoria formation. Conidia produced at temperatures higher than the optimum showed better germination and less appressoria formation than the spores produced at the temperature optimum for the growth and sporulation of the fungus. A similar increase in germination was also observed in conidia obtained from inoculated sugarcane leaves as compared to those produced on culture media. The light type virulent isolates of C. falcatum showed greater sensitivity to all these treatments than the dark type weakly pathogenic isolates.  相似文献

Plectosphaerella cucumerina was identified as a potential bioherbicide for controlling Cirsium arvense in Canada and New Zealand. The current study evaluated production conditions using two isolates (one from each country) to determine whether the yield and shelf life of inoculum are suitable for mass production. Mycelial growth and sporulation in culture both increased from 15°C to 25°C and declined at higher temperatures with no mycelial growth at 37°C. The Canadian isolate produced fewer conidia than a New Zealand isolate. Potato dextrose-based liquid media with moderate to high concentrations of carbohydrates (25%, 50%, and 100%) maximised conidia production and these base media produced conidia with the highest germination rate (>80%) both at harvest and after 4 weeks stored at 4°C in 2.5% glycerol, 40% milk glycerol or after air drying. However, after 10-week storage, the conidia failed to germinate. Sporulation occurred during growth on all solid substrates tested (rice, rolled barley, and triticale), but conidial germination was highest on rice and barley, both before and after air drying. By contrast to conidia, 90% of mycelia-infested barley grains were viable after 3 years of storage at room temperature, although viability was lost by this time on the other substrates. This study has shown that the nutritional base is an important determinant of sporulation and shelf life for P. cucumerina. Although the yield of conidia in liquid medium was adequate to justify further development of P. cucumerina as a bioherbicide, improvement in its shelf life, or alternate formulation types that extend the shelf life, must be made for commercial efficiency.  相似文献

The effects of drying methods (freeze-, spray-, and fluid bed-drying) on viability of Penicillium frequentans conidia were compared. Viability, estimated by germination of fluid bed- and freeze-dried conidia, was similar to that of fresh conidia. Skimmed milk alone, or in combination with other protectants, was added to conidia before freeze-drying. After the freeze-drying process, all protectants used, except glycerol improved conidial viability. Freeze-dried P. frequentans conidia did not maintain viability after 30 days of storage at room temperature, while conidia dried by fluid bed-drying showed 28% viability following 180 days after drying. This work also demonstrated a relationship between conidial viability after 1 year of storage at room temperature, moisture content after fluid bed-drying and initial weight of sample. Conidial moisture contents must be reduced to 5-15% for optimal storage at room temperature. P. frequentans conidia dried by fluid bed-drying were as effective as fresh conidia in controlling brown rot of peaches.  相似文献

Ten Beauveria bassiana isolates were characterized using the following parameters: virulence, morphological measurements, germination in solid and liquid media, in vitro growth and physiological changes in liquid media. There were significant differences in mortality of second-instar larvae of the diamondback moth (DBM), Plutella xylostella (Lepidoptera: Plutellidae) and the Colorado potato beetle (CPB), Leptinotarsa decemlineata (Coleoptera, Chrysomelidae) according to isolate used (DBM; 14.51-53.37%, CPB; 25.05-74.07%). The most virulent isolates to DBM and CPB were KCF107 (soil origin) and KCF106 (Chilo suppressalis origin), respectively. Several morphological and physiological characteristics of isolates were evaluated for a possible link to virulence against both insect species. Special mean diameter (2.45-3.58 µm) and area of conidia (5.72-10.25 µm2) were significantly different among isolates. Time for 50% of conidia to germinate (GT50) varied from 11 to 21 h depending on isolate. There was no correlation to virulence of size of conidia, GT50, red pigment production and mycelial growth. Acidity of 7-day-old broth cultures of isolates was positively correlated with virulence. Iranian isolates KCF106 and KCF107 were the most promising and virulent isolates but field testing is needed to further evaluate their potential for use in DBM and CPB management.  相似文献

Studies were conducted on agar media to characterize the factors for the optimization of sporulation of Alternaria alternata f. sp. sphenocleae , a fungal pathogen being evaluated as a biological control agent for Sphenoclea zeylanica (gooseweed). A. alternata f. sp. sphenocleae conidiation was affected by nutrition, temperature, light conditions, and moisture. On all agar media tested, except for half-strength potato dextrose agar (½ PDA) and V-8 juice agar (VJA), exposure to different light conditions did not have any significant effect on conidia production. However, when comparing ½ PDA and VJA, sporulation under constant near-ultraviolet (NUV) light at 28 o C increased markedly on VJA, but decreased substantially on ½ PDA. This trend, however, was opposite under dark conditions since ½ PDA produced the greatest number of conidia whereas a 75% reduction in conidia production occurred on VJA in the dark. On all the standard agar media evaluated, the most virulent conidia were obtained on ½ PDA at 28 o C under constant NUV incubated for 4 weeks. Sporulation of A. alternata f. sp. sphenocleae using the sporulation medium (S-medium) technique was rapid. Conidia were produced within 24 h and continuous sporulation was still observed until 120 h. The best primary agar media for conidia production were PDA, ½ PDA and VJA, while water agar was the poorest. Conidia production was optimized with the addition of 20 g l -1 of calcium carbonate (CaCO 3 ) and the addition of 2 ml of sterile distilled water on the medium. The most virulent conidia were produced when the primary agar was ½ PDA, the CaCO 3 concentration was 20 g l -1 , and the cultures were incubated at 18 o C in the dark. Conidiophore induction occurred on nutrient rich media and was stimulated by NUV, while formation of conidia proceeded in darkness after nutrients were depleted under warm dry or cool moist conditions. Culture media, growth conditions, and CaCO 3 affected the inoculum potential of A. alternata f. sp. sphenocleae conidia.  相似文献

Species of Penicillium and Trichoderma were found to dominate the rhizosphere of established tea bushes in a detailed study conducted from various tea growing locations in India. Penicillium erythromellis, P. janthinellum, P. raistrickii, Trichoderma pseudokoningii and T. koningii were found to be closely associated with tea roots. While seasonal fluctuation was observed in the case of Penicillium spp., the population of Trichoderma spp. showed less variation during the year. Both species were sensitive to low temperatures. In general, fungi associated with the tea rhizosphere were found to prefer a mesophillic temperature range (15 °C to 35 °C). The dominant species of Penicillium and Trichoderma also exhibited tolerance to lower temperatures, i.e., 5 to 10 °C on agar plates. Most fungi were able to grow in a wide range of pH (4 to 12). Lowering of soil pH in the rhizosphere of tea bushes was positively correlated with the age of the bush and may have affected the development of a specific microbial community in the rhizosphere.

The populations of Penicillium and Trichoderma species were inversely correlated with the populations of two most dominant rhizosphere bacteria, Bacillus subtilis and B. mycoides. Both Bacillus species have been shown to have antagonistic activity against these two fungi under in vitro conditions. The present study demonstrates the existence of a similar antagonism under in situ conditions in the rhizosphere of established tea bushes.  相似文献
