蛋鸡品系RAPD变异及其与杂种优势关系的分析   总被引:36,自引:5,他引:31  
为探讨RAPD标记在家鸡杂种优势预测中的应用价值,分析了力康自壳蛋鸡纯系的RAPD变异及其与杂种优势的关系.5个纯系内和系间显示出低水平的变异.根据品系间遗传距离DS用UPGMA法进行聚类分析,品系a、e、d聚为一类,而h、l聚为另一类.简单线性相关分析表明,父系相似系数与F1代的60周龄产蛋数存在正相关而与蛋重的杂种优势率存在负相关;母系相似系数与产蛋数的杂种优势率存在高度负相关.但品系间遗传距离与各性状及其杂种优势率之间没有显著的相关关系.  相似文献

Summary The nature and extent of heterosis for grain yield and its component characters was estimated in a seven-parent F1-diallel cross in rice under the cold stress conditions of Palampur, representing sub-temperate climate of the mid hills of Himachal Pradesh. The highest heterotic effects for grain yield were observed in Himalaya 1/Phul Patas 72 and China 988/Himdhan hybrids, which gave 60.36 and 32.48% heterosis, respectively, over mid parent, 38.76 and 26.86% heterosis, respectively, over high parent and 32.30 and 26.86% heterosis, respectively, over the best variety, Himdhan. Heterosis for grain yield in these crosses was due to an increase in tiller number, panicle length, spikelets/panicle and 1,000-grain weight. Large number of crosses exhibited significant heterosis for high spikelet sterility and the majority of them had significant heterosis for late flowering, taking more than 120 days to flower, which resulted in lack of heterosis for grain yield in such crosses due to cold stress at the reproductive stage.Part of the thesis submitted by the senior author to Himachal Pradesh Agricultural University, Palampur (H.P.) The production of pigs transgenic for human decay accelerating factor (hDAF) as potential donors for clinical organ xenotransplantation was reported several years ago. For this purpose it is required that high levels of hDAF are expressed at relevant sites in transplantable organs. Currently, homozygous lines have been produced as well as lines from crosses between heterozygous animals from different founder lines, termed jigsaw pigs. The purpose of the jigsaw crosses is to combine the desirable hDAF protein expression patterns found in different founder lines. Initial selection of the jigsaw pigs is based on the inheritance of the hDAF integration sites from both lines. Litters with potential homozygous transgenics and jigsaw transgenics were analysed by fluorescence in situ hybridization (FISH) and slot blot analysis. Results show that both slot blot analysis and FISH are suitable to distinguish between pigs that are heterozygous and homozygous for hDAF. However, FISH has the advantage of producing results more rapidly. For the identification of jigsaw pigs FISH analysis was required since slot blot analysis lacked the required accuracy. On basis of these results, FISH analysis was made part of the routine screening programme for hDAF transgenic pigs  相似文献

Summary Inheritance of thiocyanate and (+)S-methyl-L-cysteine sulphoxide (SMCO) contents were studied in a diallel cross involving five varieties of forage rape. Although both additive and dominance components were significant for the two characters, the greater mean squares of the additive gene effect indicated its greater importance. Non-allelic gene interaction was detected for thiocyanate only. Narrow sense heritability was relatively high for both characters. The general and specific combining ability effects and heterosis of the individual crosses, identified the potential of the cross Nevin × Akela for production of varieties with low thiocyanate content. Thiocyanate and SMCO contents were positively correlated, and they were not correlated with dry matter yield and its components.  相似文献

Summary We have carried out two equivalent selection experiments to increase and decrease heat shock resistance of Drosophila subobscura adults, using an indirect selection method that avoids excessive consanguinity. Heat shock was 33±0.5 °C at saturation humidity. Control lines showed a rapid change of the physiological trait as a consequence of laboratory culture conditions, expressed as a decrease both in heat shock resistance and in the initial population variability for heat shock resistance. Thus, this reduction of variability seems to consist in the loss of those combinations of genes that confer high resistance to heat shock. After eight generations of selection, the selected lines were differentiated from their respective control lines, and the selection response obtained was similar in resistant and sensitive lines. Differences in survival of progeny of reciprocal crosses between selected lines suggest that inheritance of heat resistance may depend in part on the origin of egg cytoplasm.  相似文献

Summary Three different diallel crosses were studied in Cicer arietinum; two of size 6×6, one within each of the two botanical groups macrosperma and microsperma of the cultivated subspecies, and one of 9×9 involving lines covering most of the morphological variation of chickpea. Barriers to crossability present neither a botanical nor a geographical pattern, being probably a direct consequence of interactions between genotypes. The genetic systems of twelve quantitative characters were analysed. Full dominance in a negative sense (small values dominant) is shown by leaflet length, width and shape index, rachis length, leaflet density on the rachis and pod length. Full dominance in a positive sense is shown by seeds per pod. Overdominance (in a positive sense) is evident for pods, seeds and yield per plant. Weak reciprocal differences were manifested by pod length, and pods, seeds and yield per plant. The system controlling number of leaflets per leaf is not clear. Dominance of primitive over selected characters seems to be the rule. As far as the environmental effects have permitted the analysis, no differences in genetic systems were observed between botanical groups.  相似文献

Identification and mapping of cleistogamy genes in barley   总被引:1,自引:0,他引:1  
Cleistogamy is a closed type of flowering with ensured self-pollination and an important trait to study evolutionary development in flower organs, reproduction systems, gene flow, and disease control. Still, very limited information is available about the genetic control and regulatory mechanism of this trait in barley. In this work, from the eight crosses between cleistogamous and chasmogamous accessions, five crosses generated chasmogamous F1 plants and their F2 plants segregated as 3 chasmogamous:1 cleistogamous, whereas three crosses generated cleistogamous F1 plants, and their F2 plants segregated as 1 chasmogamous:3 cleistogamous. Although a single gene was responsible for the control of cleistogamy in these two groups of crosses, the direction of dominance was opposite, suggesting two genes, cly1 and Cly2, for the genetic control of cleistogamy in barley. Epistatic type of gene interaction between the two loci was detected. In the analysis of 99 recombinant inbred lines of Azumamugi × Kanto Nakate Gold and doubled haploid lines of Harrington × Mikamo Golden, where in both crosses F1 was chasmogamous, the cly1 locus has been mapped on chromosome 2HL. Using the analysis of the F2 population of Misato Golden and Satsuki Nijo where F1 was cleistogamous, the Cly2 locus was mapped in the same region of chromosome 2HL. Because the cly1 and Cly2 loci were mapped in the same region in these three different mapping populations, it was concluded that the expression of cleistogamy is under the control of two tightly linked genes or different alleles of the same gene.  相似文献

Summary Nucleocytoplasmic determination of male sterility in Thymus vulgaris L. has been assumed in all papers attempting to explain the remarkably high frequencies of male steriles found in natural populations of this species. This paper provides strong evidence that both nuclear and cytoplasmic genes are involved in the determination of male sterility of this species, giving a complex inheritance. Interpopulation and intrapopulation crosses have shown that the ratio of females versus hermaphrodites among offsprings varied widely from 10 to 11. Furthermore, interpopulation crosses consistently yielded a higher frequency of females than intrapopulation crosses. Nucleocytoplasmic inheritance was demonstrated by an absence of male fertiles in backcrosses and asymmetrical segregation in reciprocal crosses. Molecular analysis of the mitochondrial DNA of some of the parents used in crosses suggested the involvement of different cytoplasms in the inheritance of male steriliy.  相似文献

Summary Muskmelon yellowing disease was one of the most serious problems affecting muskmelon crops along the south-east coast of Spain throughout the 1980s. The causal agent of this disease is a virus that we call muskmelon yellows virus (MYV); MYV is transmitted by the greenhouse whitefly Trialeurodes vaporariorum Westwood. It has proven impossible to find sources of resistance to MYV within a wide collection of Spanish muskmelon landraces and exotic varieties. However, Nagata Kin Makuwa and PI 161375, lines of Asiatic origin, show tolerance to this disease. These two lines were studied together with two others (Galia and Piel de Sapo type) that are very susceptible to MYV. The crosses between them (susceptible x tolerant) and the segregant generations derived from these crosses were also investigated. The studies were carried out in two different places and years. The expression of tolerance is influenced by the environment. A parabolic type relationship exists between the average value of percentage of tolerant plants and their variation. This allowed us to quantify the expected response in the segregant generations. The results observed in these generations agreed with a simple genetic control of tolerance. This tolerance, combined with protective measures which delay the infection, can contribute notably to mitigating the effects of MYV.  相似文献

Summary Genetic diversity among ten varieties of spring wheat used as parents in a diallel cross was assessed through multivariate analysis (D2-statistics) and then related to heterosis and SCA effects of their hybrids. The parents fell into three groups. Group I contained the varieties, Nobre, Girua and Carazinho; group II contained Sonalika, Lyallpur and Pitic 62 and group III contained Indus 66, Balaka, Sonora 64rs and MSl. The varieties of group I were good general combiners, while the varieties of group III were poor combiners. Significant heterotic and SCA effects for yield and yield components were observed in the hybrids of the parents belonging to different groups but not in the same group. Genetic divergence between the parents had a positive relationship with heterosis and SCA effects of the hybrids.  相似文献

The presence of heterosis has been observed in many species at both phenotypic and gene levels. Strangely, the genetic basis of heterosis was and still is largely unknown. In this study, we extended and simplified some formulas that we reported previously. The foundation of our model was based on partitioning the F 1 phenotypic variance of the cross between two pure lines into additive, dominance and epistasis components, which lead to the estimation of effective factors, crossheritability in the broad and narrow sense and heterotic power. In the model, we assume that all polygenes controlling a quantitative trait have an equal genetic effect and are independent of each other. By extension of the heritability to a cross population, new features appear. The word crossheritability acquires the status of a new genetic parameter that suffices to deal with the problem of crossbreeding and clarifies the picture of heterosis. Lastly, an example of employing the proposed method in analyzing the crossing data from Drosophila melanogaster is given to illustrate its application.  相似文献

Summary Studies on callus growth and shoots/cotyledon, using seven different genotypes of pigeonpea and their hybrid progenies, revealed continuous variation for these traits. Hence, the type of gene action influencing in vitro cell proliferation and differentiation has been investigated in a diallel analysis of seven pigeonpea genotypes. Highly significant average heterosis was recorded for callus growth and seed yield/plant. In general, the F1 hybrids which showed heterosis for callus growth also exceeded their better parent for yield/ plant. Combining ability analysis revealed both additive and non-additive gene effects for callus growth, while number of shoots/cotyledon was mostly governed by non-additive gene effects. The genotype, ICP 7035, was the best general combiner for callus growth and shoot forming capacity of cotyledons. Two cross combinations, 7186×6974 and 7035×T-21, showed maximum SCA effects for callus growth and shoots/cotyledon. Callus dry weight was positively correlated with seed yield/plant and seedling weight. The strong positive association of callus growth with seed yield indicates the possibility of using this system for mass screening and selection of superior hybrids.  相似文献

Restriction fragment analysis was used to examine the inheritance of lentil mitochondrial DNA (mtDNA) in F1 and F5 progeny from intrasubspecific (Lens culinaris ssp. culinaris) crosses and in F1 progeny from intersubspecific (Lens culinaris ssp. orientalis x L. culinaris ssp. culinaris) crosses. Southern blots of digested parental and progeny DNA were hybridized to heterologous maize mtDNA probes specific to coxI and atp6 genes. Two restriction fragment polymorphisms separated L.c. ssp. culinaris Laird and Eston from L.c. ssp. culinaris ILL5588, and one restriction fragment polymorphism distinguished L.c. ssp. culinaris Laird and Eston from L.c. ssp. orientalis LO4. Twelve of 13 f1 progeny and all F5 progeny from the intrasubspecific crosses, and all F1 progeny from intersubspecific crosses had only maternal mtDNA restriction fragments. One f1 plant from an Eston x ILL5588 cross inherited mtDNA fragments from both parents. Nuclear DNA inheritance was biparental in all F1 progeny.NRCC No. 38451  相似文献

Summary The effects of gamma irradiation and EMS seed treatment on the genetic properties of pure line and hybrid wheat populations were measured. Two hundred lines were derived from each mutagenic treatment of each genetic source and, together with their control materials, were assayed for heading date, plant height, spike length, and kernel weight in a replicated field experiment.Both mutagens induced significant genetic variability for the four traits in either Giza 150 or Sonora 64, two pure cultivars, but neither was effective in increasing the genetic variation in their hybrid background. The relative magnitude of induced variation compared with that from hybridization depended on the particular mutagen and attribute and averaged less than 50% of that from hybridization. Heritabilities and expected genetic gains were not much lower in mutagenic populations than in hybrid populations. The relative distributions of the variance components among families and within families, and the estimates of the genetic correlations in the various populations, indicated that induced mutations were somewhat similar in nature to the variation released from hybridization. The induced variation was not accompanied by any shift in the population mean and, in most cases, variation was equally distributed around the population mean.  相似文献

Pepper (Capsicum chinense Jacq.) has been reported to be an important reservoir of resistance genes to tomato spotted wilt virus (TSWV). The genes for TSWV resistance present in three C. chinense lines (PI 152225, PI 159236 and Panca) were investigated for allelism. All resistant lines were crossed with each other. Parents, F1, backcrosses and F2 populations (including reciprocals) developed from those crosses were mechanically inoculated with a highly virulent TSWV isolate. Susceptible C. annuum cv Magda was used to check inoculum virulence. Fifty plants of the F1 hybrids; Magda x PI 152225, Magda x PI 159236, and Magda x 'Panca, were also inoculated with the TSWV isolate. The resistance response in all C. chinense sources was associated with a localized, hypersensitive-like reaction that was phenotypically expressed as a prompt formation of large local lesions accompanied by premature leaf abscission. All F1 generations presented a final score of resistant; indicating that the expression of resistance to TSWV is conditioned by a dominant gene regardless of the source. The absence of segregation for resistance to TSWV that was observed in all generations of the crosses between C. chinense lines indicated that either a tightly linked group of genes exists or that the resistance is governed by the same single major gene (probably the already described Tsw gene). Previous reports have indicated that the Tsw gene is not effective against tospovirus members of serogroup II, i.e. tomato chlorotic spot virus (TCSV) and groundnut ring spot virus (GRSV). In the assay described here, all of the C. chinense lines showed, after mechanical inoculation, an identical susceptibility response to the TCSV and GRSV isolates.  相似文献

Summary Restriction fragment length polymorphisms of chloroplast (ct), mitochondrial (mt) and nuclear DNA were investigated using eight cultivars of Oryza sativa and two cultivars of O. glaberrima. Relative variability in the nuclear and cytoplasmic genomes was estimated by a common measure, genetic distance. Based on the average genetic distances among ten cultivars for each genome, the evolutionary variabilities of the mitochondrial and nuclear genomes were found to be almost the same, whereas the variability of the chloroplast genome was less than half that of the other two genomes. Cluster analyses on ct and mt DNA variations revealed that chloroplast and mitochondrial genomes were conservative within a taxon and that their differentiations were well-paralleled with respect to each other. For nuclear DNA variation, an array of different degrees of differentiation was observed in O. sativa, in contrast with little variation in O. glaberrima. As a whole, differentiation between O. sativa and O. glaberrima was clearly observed in all three genomes. In O. sativa, no notable difference was found between the cultivars Japonica and Javanica, whereas a large differentiation was noticed between Japonica (including Javanica) and Indica. In all three genomes, the average genetic distances within Indica were much larger than those within Japonica (including Javanica), and almost similar between Japonica (including Javanica) and Indica. These facts indicate that differentiation in O. sativa was due mainly to Indica.  相似文献

Summary Dodecaploid plants of Japanese persimmon (Diospyros kaki L.) were obtained by colchicine treatment of protoplasts. Callus protoplasts of Jiro (2n=90, x=15) were cultured in modified KM8p medium with 0.1% colchicine for 3–9 days. After colchicine treatment, they were cultured using agarose bead culture. Microcalli were recovered from the protoplasts after 3 months. Flow cytometric measurement showed that nine of 31 callus lines obtained from 6 days of colchicine treatment had twice the nuclear DNA content as non-treated controls. Plantlets were regenerated from the calli with twice the nuclear DNA content. Microscopic observation of root tip cells showed that their somatic chromosome number was 2n=180 (x=15). Compared with Jiro, dodecaploid plants had longer stomatal guard cells and lower stomatal densities, consistent with increased ploidy.  相似文献

We have developed a polymerase chain reaction (PCR)-based assay that could effectively reduce the time period required to screen and select for Gall Midgeresistant rice lines under field conditions. The primers for the assay were designed on the basis of sequence information of two phenotype specific random amplified polymorphic DNA fragments which were found to be tightly linked to Gall Midge biotype-1 resistance gene (Gm2). The two RAPD fragments, F81700 in the susceptible parent ARC6650 and F10600 in the resistant parent Phalguna, were identified after screening 5450 loci using 520 random primers on genomic DNAs of ARC6650 and Phalguna. These primers, when used in a multiplexed PCR, amplified specifically a 1.7-kb and 0.6-kb fragment in the susceptible and resistant parents, respectively. When this assay was performed on genomic DNAs of 44 recombinant inbred lines derived from ARC6650 x Phalguna and 5 lines derived from other crosses where one of the parents was Phalguna, ARC6650 or their derivatives, the primers amplified a 1.7-kb fragment in all of the susceptible lines or a 0.6-kb fragment in all of the resistant ones. These markers can be of potential use in the marker-aided selection of Gall Midge biotype-1 resistant phenotypes. As screening for resistance can now be conducted independent of the availability of insects, the breeding of resistant varieties can be hastened.  相似文献

Success of seed development following sexual crosses is primarily dependent on proper endosperm function and development. The failure to produce triploids, or triploid block in 4x×2x crosses served as the impetus for numerous studies of embryo and endosperm to attempt to explain cross failure. Early explanations were based upon a concept of a 232 ploidy balance between maternal tissue, endosperm, and embryo. Subsequent studies done with maize demonstrated that normal endosperm development in intraspecific maize crosses is dependent solely on having a 21 maternal to paternal genome dosage in the endosperm. These results have been modified and extended to solanaceous species in the form of an endosperm dosage system in which empirically determined factors must bear the same 21 relationship for crosses to succeed. Crossing behavior of these species suggest that the system is polygenically controlled and regulates both interspecific and intraspecific crosses. Endosperm dosage systems explain many aspects of species evolution, but the system appears to have originated as an ancient means of ensuring diploid fidelity.  相似文献

Origin and diversity of North American hard spring wheats   总被引:7,自引:0,他引:7  
Genetic diversity is an important safeguard against crop vulnerability to biotic and abiotic stresses. Coefficient-of-parentage (COP) values of 248 North American hard spring wheat (Triticum aestivum L. em. Thell) cultivars released from 1901 to 1991 were used to estimate the genetic similarity of cultivars. COP values were used: to (1) quantify germ plasm sources and their contributions to the North American hard spring wheat gene pool; (2) measure changes in genetic diversity through time; and (3) identify major groupings of related cultivars. Landraces and local cultivars that contributed to the formation of the gene pool were: spring wheat (64%), winter wheat (16%), T. turgidum var. durum L. (10%), and T. turgidum var. emmer L. (8%). Fife, Hard Red Calcutta, and Turkey Red accounted for 18%, 13%, and 8%, respectively, of the hard spring wheat origins. Era and Butte in the US, and Neepawa and HY 320 in Canada, were the most commonly used named parents of cultivars released from 1981 to 1991. Both Canada and US had the greatest level of similarity among new cultivar releases in the 1930s (Canada: r=0.39, US: r=0.34). Genetic similarity in the US declined to r=0.14 in the 1940s and remained relatively constant thereafter. Similarity among released Canadian cultivars remained relatively high until the 1970s when the introduction of new market classes resulted in a 50% reduction in genetic similarity to approximately the same level of similarity found in the US. Cluster analysis was used to group cultivars released after 1941 into 13 clusters of similar genotypes.
