An instrument package carrying pH and oxygen electrodes and a thermistor was floated across a reef flat at different times of the day and night. The data collected were used to obtain profiles of primary production, respiration, and calcification or solution of reef rock across the transect area. Average rates for these processes across the transect area, and rates at particular points along the transect, were related to light intensity and light response curves were constructed for productivity and calcification. Productivity tended to saturate at high light intensities and the amplitude and shape of the response curve changed with distance from the reef crest. Calcification showed no such saturation. Rates of calcification, and the dependence of calcification on light intensity, increased with distance from the reef crest. Average reef flat gross productivity and net calcification, based on the light response curves were 8.8 g C · m ?2 · day?1 and 9.4 g CaCO3 · m?2 · day?1 (3.5 kg · m?2· yr?1), and the production: consumption ratio was 1.16:1. These results were obtained in late summer, and are for a cloudless day. The results suggest that the degree of stability in the reef surface, and the amount of disturbance experienced by different reef flat communities, probably exert major controls on reef flat community metabolism.  相似文献

Community metabolism and air-sea carbon dioxide (CO2) fluxes were investigated in July 1992 on a fringing reef at Moorea (French Polynesia). The benthic community was dominated by macroalgae (85% substratum cover) and comprised of Phaeophyceae Padina tenuis (Bory), Turbinaria ornata (Turner) J. Agardh, and Hydroclathrus clathratus Bory (Howe); Chlorophyta Halimeda incrassata f. ovata J. Agardh (Howe); and Ventricaria ventricosa J. Agardh (Olsen et West), as well as several Rhodophyta (Actinotrichia fragilis Forskál (Børgesen) and several species of encrusting coralline algae). Algal biomass was 171 g dry weight· m?2. Community gross production (Pg), respiration (R), and net calcification (G) were measured in an open-top enclosure. Pg and R were respectively 248 and 240 mmol Co2·m?2·d?1, and there was a slight net dissolution of CaCO3 (0.8 mmol · m?2·d?1). This site was a sink for atmospheric CO2 (10 ± 4 mmol CO2·m?2·d?1), and the analysis of data from the literature suggests that this is a general feature of algal-dominated reefs. Measurement of air-sea CO2 fluxes in open water close to the enclosure demonstrated that changes in small-scale hydrodynamics can lead to misleading conclusions. Net CO2 evasion to the atmosphere was measured on the fringing reef due to changes in the current pattern that drove water from the barrier reef (a C02 source) to the study site.  相似文献

There is an interest in developing approaches to “ecosystem-based” management for coral reefs. One aspect of ecosystem performance is to monitor carbon metabolism of whole communities. In an effort to explore robust techniques to monitor the metabolism of fringing reefs, especially considering the possible effects of ocean acidification, a yearlong study of the carbonate chemistry of a nearshore fringing reef in Hawaii was conducted. Diurnal changes in seawater carbonate chemistry were measured once a week in an algal-dominated and a coral-dominated reef flat on the Waimanalo fringing reef, Hawaii, from April of 2010 until May of 2011. Calculated rates of gross primary production (GPP) and net community calcification (G) were similar to previous estimates of community metabolism for other coral reefs (GPP 971 mmol C m?2 d?1; G 186 mmol CaCO3 m?2 d?1) and indicated that this reef was balanced in terms of organic metabolism, exhibited net calcification, and was a net source of CO2 to the atmosphere. Average slopes of total alkalinity versus dissolved inorganic carbon (TA–DIC slope) for the coral-dominated reef flat exhibited a greater calcification-to-net photosynthesis ratio than for the algal-dominated reef flat (coral slope vs. algal slope). Over the course of the time series, TA–DIC slopes remained significantly different between sites and were not correlated with diurnal averages in reef-water residence time or solar irradiance. These characteristic slopes for each reef flat reflect the relationship between carbon and carbonate community metabolism and can be used as a tool to monitor ecosystem function in response to ocean acidification.  相似文献

Two axenic, in vitro liquid suspension cultures were established for Agardhiella subulata (C. Agardh) Kraft et Wynne, and their growth characteristics were compared. This study illustrated how reliable routes for the development of suspension cultures of macrophytic red algae of terete thallus morphology can be achieved for biotechnology applications. Undifferentiated filament clumps of 2–8 mm diameter were established by induction of callus-like tissue from thallus explants, and lightly branched microplantlets of 2–10 mm length were established by regeneration of filament clumps. The filament clumps were susceptible to regeneration. Adventitious shoot formation was reliably induced from 40% to 70% of the filament clumps by gentle mixing at 100 rev min?1 on an orbital shaker. The specific growth rate of the microplantlets was higher than the filament clumps in nonagitated well plate culture (4%–6% per day for microplantlets vs. 2%–3% per day for filament clumps) at 24° C and 8–36 μmol photons·m?2·s?1 irradiance (10:14 h LD cycle) when grown on ASP12 artificial seawater medium at pH 8.6–8.9 with 20%–25% per day medium replacement. Oxygen evolution rate vs. irradiance measurements showed that relative to the filament clumps, microplantlets had a higher maximum specific oxygen evolution rate (Po,max= 0.181 ± 0.035 vs. 0.130 ± 0.023 mmol O2·g?1 dry cell mass·h?1), but comparable respiration rate (Qo= 0.040 ± 0.013 vs. 0.033 ± 0.017 mmol O2·g?1 dry cell mass·h?1), compensation point (Ic= 3.8 ± 2.4 vs. 5.7 ± 1.2 μmol photons·m?2·s?1), and light intensity at 63.2% of saturation (Ik= 17.5 ± 3.9 vs. 14.9 ± 2.6 μmol photons·m?2·s?1). The microplantlet culture was more suitable for suspension culture development than the filament clump culture because it was morphologically stable and exhibited higher growth rates.  相似文献

Optimum light, temperature, and pH conditions for growth, photosynthetic, and respiratory activities of Peridinium cinctum fa. westii (Lemm.) Lef were investigated by using axenic clones in batch cultures. The results are discussed and compared with data from Lake Kinneret (Israel) where it produces heavy blooms in spring. Highest biomass development and growth rates occurred at ca. 23° C and ≥50 μE· m?2·s1 of fluorescent light with energy peaks at 440–575 and 665 nm. Photosynthetic oxygen release was more efficient in filtered light of blue (BG 12) and red (RG 2) than in green (VG 9) qualities. Photosynthetic oxygen production occurred at temperatures ranging from 5° to 32° C in white fluorescent light from 10 to 105 μE·m?2·s?1 with a gross maximum value of 1500 × 10?12 g·cell?1·h?1 at the highest irradiance. The average respiration amounted to ca. 12% of the gross production and reached a maximum value of ca. 270·10?12 g·cell?1·h?1 at 31° C. A comparison of photosynthetic and respiratory Q10-values showed that in the upper temperature range the increase in gross production was only a third of the corresponding increase in respiration, although the gross production was at maximum. Short intermittent periods of dark (>7 min) before high light exposures from a halogen lamp greatly increased oxygen production. Depending on the physiological status of the alga, light saturation values were reached at 500–1000 μE·m?2·s?1 of halogen light with compensation points at 20–40 μE·m?2·s?1 and Ik-values at 100–200 μE·m?2·s?1. The corresponding values in fluorescent light in which it was cultured and adapted, were 25 to 75% lower indicating the ability of the alga to efficiently utilize varying light conditions, if the adaptation time is sufficient. Carbon fixation was most efficient at ca. pH 7, but the growth rates and biomass development were highest at pH 8.3.  相似文献

Measurements of bimodal oxygen uptake have been made in a freshwater air-breathing fish,Notopterus chitala at 29.0±1(S.D.)°C. xhe mean oxygen uptake from continuously flowing water without any access to air, was found to be 3.58±0.37 (S.E.) ml O2 · h?1 and 56.84+4.29 (S.E.) ml O2 · kg?1 · h?1 for a fish weighing 66.92 + 11.27 (S.E.) g body weight. In still water with access to air, the mean oxygen uptake through the gills were recorded to be 2.49 ± 0.31 (S.E.) ml O2 · h?1 and 38.78 ± 1.92 (S.E.) ml O2 · kg?1 · h?1 and through the accessory respiratory organs (swim-bladder) 6.04±0.87 (S.E.) ml O2 · h?1 and 92.32±2.91 (S.E.) ml O2 · kg?1 · h?1 for a fish averaging 66.92±11.27 (S.E.) g. Out of the total oxygen uptake (131.10 ml O2 · kg?1 · h?1), about 70% was obtained through the aerial route and the remainder 30% through the gills.  相似文献

We used microsensors to characterize physicochemical microenvironments and photosynthesis occurring immediately after water saturation in two desert soil crusts from southeastern Utah, which were formed by the cyanobacteria Microcoleus vaginatus Gomont, Nostoc spp., and Scytonema sp. The light fields within the crusts presented steep vertical gradients in magnitude and spectral composition. Near-surface light-trapping zones were formed due to the scattering nature of the sand particles, but strong light attenuation resulted in euphotic zones only ca. 1 mm deep, which were progressively enriched in longer wavelengths with depth. Rates of gross photosynthesis (3.4–9.4 mmol O2·m?2·h?1) and dark respiration (0.81–3.1 mmol O?2·m?2·h?1) occurring within 1 to several mm from the surface were high enough to drive the formation of marked oxygen microenvironments that ranged from oxygen supersaturation to anoxia. The photosynthetic activity also resulted in localized pH values in excess of 10, 2–3 units above the soil pH. Differences in metabolic parameters and community structure between two types of crusts were consistent with a successional pattern, which could be partially explained on the basis of the microenvironments. We discuss the significance of high metabolic rates and the formation of microenvironments for the ecology of desert crusts, as well as the advantages and limitations of microsensor-based methods for crust investigation.  相似文献

Experimental studies have shown that coral calcification rates are dependent on light, nutrients, food availability, temperature, and seawater aragonite saturation (Ω arag), but the relative importance of each parameter in natural settings remains uncertain. In this study, we applied Calcein fluorescent dyes as time indicators within the skeleton of coral colonies (n = 3) of Porites astreoides and Diploria strigosa at three study sites distributed across the northern Bermuda coral reef platform. We evaluated the correlation between seasonal average growth rates based on coral density and extension rates with average temperature, light, and seawater Ω arag in an effort to decipher the relative importance of each parameter. The results show significant seasonal differences among coral calcification rates ranging from summer maximums of 243 ± 58 and 274 ± 57 mmol CaCO3 m?2 d?1 to winter minimums of 135 ± 39 and 101 ± 34 mmol CaCO3 m?2 d?1 for P. astreoides and D. strigosa, respectively. We also placed small coral colonies (n = 10) in transparent chambers and measured the instantaneous rate of calcification under light and dark treatments at the same study sites. The results showed that the skeletal growth of D. strigosa and P. astreoides, whether hourly or seasonal, was highly sensitive to Ω arag. We believe this high sensitivity, however, is misleading, due to covariance between light and Ω arag, with the former being the strongest driver of calcification variability. For the seasonal data, we assessed the impact that the observed seasonal differences in temperature (4.0 °C), light (5.1 mol photons m?2 d?1), and Ω arag (0.16 units) would have on coral growth rates based on established relationships derived from laboratory studies and found that they could account for approximately 44, 52, and 5 %, respectively, of the observed seasonal change of 81 ± 14 mmol CaCO3 m?2 d?1. Using short-term light and dark incubations, we show how the covariance of light and Ω arag can lead to the false conclusion that calcification is more sensitive to Ω arag than it really is.  相似文献

Aims and methods

To evaluate the seasonal and spatial variations of methane (CH4) emissions and understand the controlling factors, we measured CH4 fluxes and their environmental variables for the first time by a static chamber technique in high Suaeda salsa marsh (HSM), middle S. salsa marsh (MSM), low S. salsa marsh (LSM) and bare flat (BF) in the northern Yellow River estuary throughout a year.

Results

CH4 emissions from coastal marsh varied throughout different times of the day and significant differences were observed in some sampling periods (p?<?0.05). Over all sampling periods, CH4 fluxes averaged between ?0.392 mgCH4 m?2?h?1 and 0.495 mgCH4 m?2?h?1, and emissions occurred during spring (0.008 mgCH4 m?2?h?1) and autumn (0.068 mgCH4 m?2?h?1) while sinks were observed during summer (?0.110 mgCH4 m?2?h?1) and winter (?0.009 mgCH4 m?2?h?1). CH4 fluxes from the four marshes were not significantly different (p?>?0.05), and emissions occurred in LSM (0.026 mgCH4 m?2?h?1) and BF (0.055 mgCH4 m?2?h?1) while sinks were observed in HSM (?0.035 mgCH4 m?2?h?1) and MSM (?0.022 mgCH4 m?2?h?1). The annual average CH4 flux from the intertidal zone was 0.002 mgCH4 m?2?h?1, indicating that coastal marsh acted as a weak CH4 source. Temporal variations of CH4 emission were related to the interactions of abiotic factors (temperatures, soil moisture and salinity) and the variations of limited C and mineral N in sediments, while spatial variations were mainly affected by the vegetation composition at spatial scale.

Conclusions

This study observed a large spatial variation of CH4 fluxes across the coastal marsh of the Yellow River estuary (CV?=?7856.25 %), suggesting that the need to increase the spatial replicates at fine scales before the regional CH4 budget was evaluated precisely. With increasing exogenous nitrogen loading to the Yellow River estuary, the magnitude of CH4 emission might be enhanced, which should also be paid more attentions as the annual CH4 inventory was assessed accurately.  相似文献

The rate of emergence of micropredatory gnathiid isopods from the benthos, the proportion of emerging gnathiids potentially eaten by Labroides dimidiatus, and the volume of blood that gnathiids potentially remove from fishes (using gnathiid gut volume) were determined. The abundance (mean ±s.e .) of emerging gnathiids was 41·7 ± 6·9 m?2 day?1 and 4552 ± 2632 reef?1 day?1 (reefs 91–125 m2). The abundance of emerging gnathiids per fish on the reef was 4·9 ± 0·8 day?1; but excluding the rarely infested pomacentrid fishes, it was 20·9 ± 3·8 day?1. The abundance of emerging gnathiids per patch reef was 66 ± 17% of the number of gnathiids that all adult L. dimidiatus per reef eat daily while engaged in cleaning behaviour. If all infesting gnathiids subsequently fed on fish blood, their total gut volume per reef area would be 17·4 ± 5·6 mm3 m?2 day?1; and per fish on the reefs, it would be 2·3 ± 0·5 mm?3 fish?1 day?1 and 10·3 ± 3·1 mm3 fish?1 day?1 (excluding pomacentrids). The total gut volume of gnathiids infesting caged (137 mm standard length, LS) and removed from wild (100–150 mm LS) Hemigymnus melapterus by L. dimidiatus was 26·4 ± 24·6 mm3 day?1 and 53·0 ± 9·6 mm3 day?1, respectively. Using H. melapterus (137 mm LS, 83 g) as a model, gnathiids had the potential to remove, 0·07, 0·32, 0·82 and 1·63% of the total blood volume per day of each fish, excluding pomacentrids, caged H. melapterus and wild H. melapterus, respectively. In contrast, emerging gnathiids had the potential of removing 155% of the total blood volume of Acanthochromis polyacanthus (10·7 mm LS, 0·038 g) juveniles. That L. dimidiatus eat more gnathiids per reef daily than were sampled with emergence traps suggests that cleaner fishes are an important source of mortality for gnathiids. Although the proportion of the total blood volume of fishes potentially removed by blood‐feeding gnathiids on a daily basis appeared to be low for fishes weighing 83 g, the cumulative effects of repeated infections on the health of such fish remains unknown; attacks on small juvenile fishes, may result in possibly lethal levels of blood loss.  相似文献

The kinetics of the light-driven Cl? uptake pump of Synechococcus R-2 (PCC 7942) were investigated. The kinetics of Cl? uptake were measured in BG-11 medium (pHo, 7·5; [K+]o, 0·35 mol m?3; [Na+]o, 18 mol m?3; [Cl?]o, 0·508 mol m?3) or modified media based on the above. Net36Cl? fluxes (?Cl?o,i) followed Michaelis-Menten kinetics and were stimulated by Na+ [18 mol m?3 Na+ BG-11 ?Cl?max= 3·29±0·60 (49) nmol m?2 s?1 versus Na+-free BG-11 ?Cl?max= 1·02±0·13 (54) nmol m?2 s?1] but the Km was not significantly different in the presence or absence of Na+ at pHo 10; the Km was lower, but not affected by the presence or absence of Na+ [Km = 22·3±3·54 (20) mmol m?3]. Na+ is a non-competitive activator of net ?Cl?o,i. High [K+]o (18 mol m?3) did not stimulate net ?Cl?o,i or change the Km in Na+-free medium. High [K+]o (18 mol m?3) added to Na+ BG-11 medium decreased net ?Cl?o,i [18 mol m?3K+ BG-11; ?Cl?max= 2·50±0·32 (20) nmol m?2 s?1 versus BG-11 medium; ?Cl?max= 3·35±0·56 (20) nmol m?2 s?1] but did not affect the Km 55·8±8·100 (40) mmol m?3]. Na+-stimulation of net ?Cl?o,i followed Michaelis-Menten kinetics up to 2–5 mol m?3 [Na+]o but higher concentrations were inhibitory. The Km for Na+-stimulation of net ?Cl?o,i [K1/2(Na+)] was different at 47 mmol m?3 [Cl?]o (K1/2[Na+] = 123±27 (37) mmol m?3]. Li+ was only about one-third as effective as Na+ in stimulating Cl? uptake but the activation constant was similar [K1/2(Li+) = 88±46 (16) mmol m?3]. Br? was a competitive inhibitor of Cl? uptake. The inhibition constant (Ki) was not significantly different in the presence and absence of Na+. The overall Ki was 297±23 (45) mmol m?3. The discrimination ratio of Cl? over Br? (δCl?/δBr?) was 6·38±0·92 (df = 147). Synechococcus has a single Na+-stimulated Cl? pump because the Km of the Cl? transporter and its discrimination between Cl? and Br? are not significantly different in the presence and absence of Na+. Accurate measurement of coral reef community metabolism is a necessity for process monitoring and in situ experimentation on coral reef health. Traditional methodologies used for these measurements are effective but limited by location and scale constraints. We present field trial results for a new benthic chamber system called the Submersible Habitat for Analyzing Reef Quality (SHARQ). This large, portable incubation system enables in situ measurement and experimentation on community-scale metabolism. Rates of photosynthesis, respiration, and calcification were measured using the SHARQ for a variety of coral reef substrate types on the reef flat of South Molokai, Hawaii, and in Biscayne National Park, Florida. Values for daily gross production, 24-h respiration, and net calcification ranged from 0.26 to 6.45 g O2 m–2 day–1, 1.96 to 8.10 g O2 m–2 24 h–1, and 0.02 to 2.0 g CaCO3 m–2 day–1, respectively, for all substrate types. Field trials indicate that the SHARQ incubation chamber is an effective tool for in situ isolation of a water mass over a variety of benthic substrate types for process monitoring, experimentation, and other applications.  相似文献

Here, we report the first‐ever measurements of light CO2 respiration rate (CRR) by seaweeds. We measured the influence of temperature (15–25°C) and light (irradiance from 60 to 670 μmol · m?2 · s?1) on the light CCR of two subtropical seaweed species, and measured the CRR of seven different seaweed species under the same light (150 μmol · m?2 · s?1) and temperature (25°C). There was little effect of irradiance on light CRR, but there was an effect of temperature. Across the seven species light CRR was similar to OCR (oxygen consumption rate in the dark), with the exception of a single species. The outlier species was a coralline alga, and the higher light CRR was probably driven by calcification. CRR could be estimated from OCR, as well as carbon photosynthetic rates from oxygen photosynthetic rates, which suggests that previous studies have probably provided good estimations of gross photosynthesis for seaweeds.  相似文献

Chlamydomonas reinhardtü Dangeard, adenine or guanine can be used as the sole nitrogen source for growth by means of an inducible system which is repressed by ammonia. Cells grown on either adenine or guanine were able to take up both purines, although the adenine uptake rate was always about 40% of the guanine uptake rate. Both adenine and guanine were taken up by an inducible system(s) exhibiting hyperbolic kinetics with identical apparent A, values of 3-2 mmol m?3 for adenine and 3-2mmol m?3 for guanine. Adenine and guanine utilization depended on pH, with similar optimal pH values of 7·3 and 7·4, respectively. Adenine and guanine each acted as a competitive inhibitor of the other's uptake, and their utilization was also inhibited by hypoxanthine, xanthine and urate. Inhibition of adenine uptake by guanine and hypoxanthine was competitive, with A′, values of 5·5 and 1. 6 mmol m?3 respectively. Guanine uptake was also inhibited competitively by adenine (K1= 1·3mmol m?3) and hypoxanthine (K1= 3. 3 mmol m?3). Utilization of both adenine and guanine was inhibited by cyanide, azide, 3-(3,4-dichlorophenyl)-1,1-dimethyl urea, 2,4-dinitrophenol and carbonylcyanide m-chlorophenylhydrazone, and was also sensitive to p-hydroxymercuribenzoate and N-ethyl-maleimide. On the basis of these results, taken together, the possibility that adenine and guanine are translocated into Chlamydomonas by a common system is discussed.  相似文献

Rates of net photosynthesis and dark respiration were determined under submersed and emerged conditions for Hesperophycus harveyanus S. & G. and Pelvetia fastigiata f. gracilis (Decne.) S. & G. Both species exhibited submersed photosynthesis-light relationships and dark respiration rates similar to those established for other closely related intertidal, fucoids. Maximal net photosynthesis of H. harveyanus (0.21 mmol O2 g dry wt.-1· h-1; 0.18 mmol CO2 g dry wt.-1· h-1) was similar to that of P. fastigiata f. gracilis (0.17 mmol. O2 g dry wt.-1· h-1; 0.14 mmol CO2 g dry wt. -1· h-1). Light saturation occurred between 150 and 250 μE · m-2· s-1 for H. harveyanus and between 75 and 150 μE · m-2· s-1 for P. fastigiata f. gracilis; photon flux densities required for compensation were 6.4 and 9.2 μE · m-2· s-1, respectively. Photoinhibition was not observed for either species. The light-saturated, submersed net photosynthetic performances of both species varied significantly with temperature. Greatest photosynthetic rates were obtained at 23° C for H. harveyanus and at 18° C for P. fastigiata f. gracilis. Under emersed conditions, the maximal net photosynthetic rate and the photon flux densities required for saturation were greater for H. harveyanus (0.08 mmol CO2 g dry wt.-1· h-1; 260 to 700 μE · m-2· s-1) than for P. fastigiata f. gracilis (0.02 mmol CO2g dry wt.-1· h-1; 72 to 125 μE · m-2· s-1). However, for both species, emersed photosynthetic rates were much lower (14–44%) than those obtained under submersed conditions. Desiccation negatively influenced emersed photosynthesis, of both species, but H. harveyanus thalli contained more water when fully hydrated and lost water more slowly during dehydration, thus suggesting greater photosynthetic potential during field conditions of emersion.  相似文献

Three photosynthetic parameters of 7 species of marine diatoms were studied using Na214CO3 at 5–8 C using log phase axenic cultures. The cell volumes of the different species varied from 70 μm3 to 40 × 105μm3. The present experiment is consistent with the interpretation that the initial slope α (mg C · [mg chl a]?1· h?1· w?1· m2) of photosynthesis vs. light curves is controlled by self-shading of chlorophyll a in the cell. Pm, the rate of photosynthesis at light saturation (mg C · [mg cell, C]?1· h?1) and R, the intercept at zero light intensity (mg C · [mg cell C]?1· H?1) are both dependent on the ratio of surface area to volume of cell.  相似文献

In spring 2005, monthly sampling was carried out at a sublittoral site near Tautra Island. Microphytobenthic identification, abundance (ABU), and biomass (BIOM), were performed by microscopic analyses. Bacillariophyceae accounted for 67% of the total ABU, and phytoflagellates constituted 30%. The diatom floristic list consisted of 38 genera and 94 species. Intact light‐harvesting pigments chl a, chl c, and fucoxanthin and their derivatives were identified and quantified by HPLC. Photoprotective carotenoids were also observed (only as diadinoxanthin; no diatoxanthin was detected). Average fucoxanthin content was 4.57 ± 0.45 μg fucoxanthin · g sediment dry mass?1, while the mean chl a concentration was 2.48 ± 0.15 μg · g?1 dry mass. Both the high fucoxanthin:chl a ratio (considering nondegraded forms) and low amounts of photoprotective carotenoids indicated that the benthic microalgal community was adapted to low light. Microphytobenthic primary production was estimated in situ (MPPs, from 0.15 to 1.28 mg C · m?2 · h?1) and in the laboratory (MPPp, from 6.79 to 34.70 mg C · m?2 · h?1 under light saturation) as 14C assimilation; in April it was additionally estimated from O2‐microelectrode studies (MPPO2) along with the community respiration. MPPO2 and the community respiration equaled 22.9 ± 7.0 and 7.4 ± 1.8 mg C · m?2 · h?1, respectively. A doubling of BIOM from April to June in parallel with a decreasing photosynthetic activity per unit chl a led us to suggest that the microphytobenthic community was sustained by heterotrophic metabolism during this period.  相似文献

Carbon distribution and kinetics of Candida shehatae were studied in fed-batch fermentation with xylose or glucose (separately) as the carbon source in mineral medium. The fermentations were carried out in two phases, an aerobic phase dedicated to growth followed by an oxygen limitation phase dedicated to ethanol production. Oxygen limitation was quantified with an average specific oxygen uptake rate (OUR) varying between 0.30 and 2.48 mmolO2 g dry cell weight (DCW)?1 h?1, the maximum value before the aerobic shift. The relations among respiration, growth, ethanol production and polyol production were investigated. It appeared that ethanol was produced to provide energy, and polyols (arabitol, ribitol, glycerol and xylitol) were produced to reoxidize NADH from assimilatory reactions and from the co-factor imbalance of the two-first enzymatic steps of xylose uptake. Hence, to manage carbon flux to ethanol production, oxygen limitation was a major controlled parameter; an oxygen limitation corresponding to an average specific OUR of 1.19 mmolO2 g DCW?1 h?1 allowed maximization of the ethanol yield over xylose (0.327 g g?1), the average productivity (2.2 g l?1 h?1) and the ethanol final titer (48.81 g l?1). For glucose fermentation, the ethanol yield over glucose was the highest (0.411 g g?1) when the specific OUR was low, corresponding to an average specific OUR of 0.30 mmolO2 g DCW?1 h?1, whereas the average ethanol productivity and ethanol final titer reached the maximum values of 1.81 g l?1 h?1 and 54.19 g l?1 when the specific OUR was the highest.  相似文献

Photosynthesis-irradiance relationships were determined in the field for five species of littoral and shallow sublittoral marine benthic green algae (Chlorophyta) of differing morphologies. Each species exhibited a linear increase in photosynthetic rate with increasing irradiance up to a maximum light-saturated value. Full sunlight (1405 to 1956 μE·m?2·s?1) inhibited photosynthesis of all species except the thick, optically dense, Codium fragile (Sur.) Har. Compensation irradiances ranged from 6.1 μE·m?2·s?1 for Enteromorpha intestinalis (L.) Link to 11.4 μE·m?2·s?1 for Ulva lobata (Kütz) S. & G. and did not reveal a consistent relationship to seaweed morphology. Saturation irradiances were determined statistically (Ik) and visually from graphical plots. with the latter technique resulting in values three to eight times higher and different comparative rankings of species than the former. Ik saturation irradiances were highest for Chaetomorpha linum (Müll.) Kütz. (81.9 μE·m?2·s?1) and lowest for Codium fragile (49.6 μE·m?2·s?1) and did not reveal a relationship with seaweed morphology. Regression equations describing light-limited photosynthetic rates and the relative magnitudes of the maximal net photosynthetic responses both strongly suggested a relationship with seaweed morphology. Highest net photosynthetic rates were obtained for the thin, sheet-like algae Ulva lobata (9.2 mg C·g dry wt?1·h?1), U. rigida C. Ag. (6.5 mg C·g dry wt?1·h?1) and the tubular form, Enteromorpha intestinalis (7.3 mg C·g dry wt?1·h?1), while lowest rates occurred for Codium fragile (0.9 mg C·g dry wt?1·h?1). Similarly, steepest light-limited slopes were found for the algae of simpler morphology, while the most gradual slope was determined for Codium fragile, the alga with greatest thallus complexity.  相似文献

The net carbon uptake rate and net production rate of mycosporine‐like amino acids (MAAs) were measured in phytoplankton from 2 different melt ponds (MPs; closed and open type pond) in the western Arctic Ocean using a 13C stable isotope tracer technique. The Research Vessel Araon visited ice‐covered western‐central basins situated at 82°N and 173°E in the summer of 2012, when Arctic sea ice declined to a record minimum. The average net carbon uptake rate of the phytoplankton in polycarbonate (PC) bottles in the closed MP was 3.24 mg C · m?3 · h?1 (SD = ±1.12 mg C · m?3 · h?1), while that in the open MP was 1.3 mg C · m?3 · h?1 (SD = ±0.05 mg C · m?3 · h?1). The net production rate of total MAAs in incubated PC bottles was highest (1.44 (SD = ±0.24) ng C · L?1 · h?1) in the open MP and lowest (0.05 (SD = ±0.003) ng C · L?1 · h?1) in the closed MP. The net production rate of shinorine and palythine in incubated PC bottles at the open MP presented significantly high values 0.76 (SD = ±0.12) ng C · L?1 · h?1and 0.53 (SD = ±0.06) ng C · L?1 · h?1. Our results showed that high net production rate of MAAs in the open MP was enhanced by a combination of osmotic and UVR stress and that in situ net production rates of individual MAA can be determined using 13C tracer in MPs in Arctic sea ice.  相似文献
