The objectives of the present study were to assess how the stability of the emulsion recovered from aqueous extraction processing of soybeans was affected by characteristics of the starting material and extraction and demulsification conditions. Adding endopeptidase Protex 6L during enzyme-assisted aqueous extraction processing (EAEP) of extruded soybean flakes was vital to obtaining emulsions that were easily demulsified with enzymes. Adding salt (up to 1.5 mM NaCl or MgCl2) during extraction and storing extruded flakes before extraction at 4 and 30 °C for up to 3 months did not affect the stabilities of emulsions recovered from EAEP of soy flour, flakes and extruded flakes. After demulsification, highest free oil yield was obtained with EAEP of extruded flakes, followed by flour and then flakes. The same protease used for the extraction step was used to demulsify the EAEP cream emulsion from extruded full-fat soy flakes at concentrations ranging from 0.03% to 2.50% w/w, incubation times ranging from 2 to 90 min, and temperatures of 25, 50 or 65 °C. Highest free oil recoveries were achieved at high enzyme concentrations, mild temperatures, and short incubation times. Both the nature of enzyme (i.e., protease and phospholipase), added alone or as a cocktail, concentration of enzymes (0.5% vs. 2.5%) and incubation time (1 vs. 3 h), use during the extraction step, and nature of enzyme added for demulsifying affected free oil yield. The free oil recovered from EAEP of extruded flakes contained less phosphorus compared with conventional hexane-extracted oil. The present study identified conditions rendering the emulsion less stable, which is critical to increasing free oil yield recovered during EAEP of soybeans, an environmentally friendly alternative processing method to hexane extraction.  相似文献

Microwave-assisted ammonium hydroxide (NH4OH) followed by phosphoric acid (H3PO4) treatments were used to release monomeric sugars from Miscanthus sinensis grown in Cha-Chueng-Sao province, Thailand. Treatment with 1.0% (w/v) NH4OH, 15:1 liquid-to-solid ratio (LSR) at 120 °C temperature for 15 min liberated 2.9 g of monomeric sugars per 100 g of dried biomass, whereas the corresponding yield for a treatment with 1.78% v/v H3PO4, 15:1 LSR at 140 °C for 30 min was 62.3 g/100 g. The two-stage pretreatment, treatment with NH4OH at 120 °C temperature for 15 min followed by treatment with H3PO4 at 140 °C for 30 min, impressively provided the highest total monomeric sugar yield of 71.6 g/100 g dried biomass.  相似文献

Native wheat straw (WS) was pretreated with various concentrations of H2SO4 and NaOH followed by secondary treatments with ethylene diamine (EDA) and NH4OH prior to enzymatic saccharification. Conversion of the cellulosic component to sugar varied with the chemical modification steps. Treatment solely with alkali yield 51–75% conversion, depending on temperature. Acid treatment at elevated tempeatures showed a substantial decrease in the hemicellulose component, whereas EDA-treated WS (acid pretreated) showed a 69–75% decrease in the lignin component. Acid-pretreated EDA-treated straw yielded a 98% conversion rate, followed by 83% for alkali–NH4OH treated straws. In other experiments, WS was pretreated with varying concentration of H2SO4 or NaOh followed by NH4OH treatment prior to enzymatic hydrolysis. Pretreatment of straw with 2% NaOH for 4 h coupled to enzymatic hydrolysis yield a 76% conversion of the cellulosic component. Acid–base combination pretreatment yielded only 43% conversions. A reactor column was subsequently used to measure modification–saccharification–fermentation for wheat straw conversion on a larger scale. Thirty percent conversions of wheat straw cellulosics to sugar were observed with subsequent fermentation to alcohol. The crude cellulase preparation yielded considerable quantities of xylose in addition to the glucose. Saccharified materials were fermented directly with actively proliferating proliferating yeast cells without concentration of the sugars.  相似文献

Water hyacinth was pretreated, under variable conditions, with NaOH, alkaline H2O2, peracetic acid and sodium chlorite. Combined pretreatments included sodium chlorite with each of NaOH, alkaline H2O2 and peracetic acid. Combined pretreatment with 0.1% NaClO2 for 1 h at 100 °C and peracetic acid at 100 °C for 15 min afforded the most promising sample. The recovered lignin, cellulose and hemicellulose of this sample was 2.56%, 96.69%, and 81.38%, respectively. The same sample, by cellulase hydrolysis showed the highest cellulose conversion (80.8%) and 90% saccharification using 200 FPU/g substrate. Some ambient factors affecting saccharification of pretreated water hyacinth were investigated. Enzymic saccharification after 6 h was about 50% of that at 48 h, indicating a slow hydrolysis rate by time. Addition of 8% glucose at the beginning of the enzymic hydrolysis decreased the saccharification to about its half while addition of 8% ethanol brought about complete inhibition of the enzyme. Addition of cellobiase to the reaction mixture increased cellulose conversion and saccharification by 10%.  相似文献

Seventeen Cyathus stercoreus isolates were tested for their ability to treat rice straw for improved enzymatic saccharification. These isolates showed a negative correlation between cellulase and xylanase activity and enzymatic saccharification yields. Incubation of rice straw pretreated at 60 °C for 15 min with strain C. stercoreus TY-2 for 25 days resulted in an enzymatic saccharification yield of 57% as compared to a yield of 11% for the same straw in the absence of the fungus. These findings highlight the potential of this isolate for biological pretreatment of rice straw under conditions of low energy input.  相似文献

Sodium hydroxide (NaOH) and lime (Ca(OH)2) were innovatively used together in this study to improve the cost-effectiveness of alkaline pretreatment of switchgrass at ambient temperature. Based on the sugar production in enzymatic hydrolysis, the best pretreatment conditions were determined as: residence time of 6 h, NaOH loading of 0.10 g/g raw biomass, NaOH addition at the beginning, Ca(OH)2 loading of 0.02 g/g raw biomass, and biomass wash intensity of 100 ml water/g raw biomass, at which the glucose and xylose yields were respectively 59.4% and 57.3% of the theoretical yields. The sugar yield of the biomass pretreated using the combination of 0.10 g NaOH/g raw biomass and 0.02 g Ca(OH)2/g raw biomass was found comparable with that of the biomass pretreated using 0.20 g NaOH/g raw biomass at the same conditions, while the chemical expense was remarkably reduced due to the low cost of lime and the reduced loading of NaOH.  相似文献

This aim of this study was to investigate the effect of ammonium hydroxide (NH4OH) and sodium hydroxide (NaOH) pre-treatment on the digestibility of sugarcane bagasse (SCB) by hemicellulase action. It was found that pre-treatment of SCB with NH4OH removed a larger percentage of the SCB lignin and effectively increased SCB digestibility 13.13 fold. The greatest amount of reducing sugar (1194.88 μmol/min) and largest degree of synergy (2.85) was obtained using a combination of two enzymes (25% ManA and 75% XynA) with NH4OH pre-treated SCB. In this study, NH4OH therefore appeared to be a more effective pre-treatment step for subsequent hydrolysis by hemicellulases.  相似文献

In this study, the pretreatment of cassava residues by thermal-dilute sulfuric acid (TDSA) hydrolysis was investigated by means of a statistically designed set of experiments. A three-factor central composite design (CCD) was employed to identify the optimum pretreatment condition of cassava residues for methane production. The individual and interactive effects of temperature, H2SO4 concentration and reaction time on increase of methane yield (IMY) were evaluated by applying response surface methodology (RSM). After optimization, the resulting optimum pretreatment condition was 157.84 °C, utilizing 2.99% (w/w TS) H2SO4 for 20.15 min, where the maximum methane yield (248 mL/g VS) was 56.96% higher than the control (158 mL/g VS), which was very close to the predict value 56.53%. These results indicate the model obtained through RSM analysis is suit to predict the optimum pretreatment condition and there is great potential of using TDSA pretreatment of cassava residues to enhance methane yield.  相似文献

Catalytic conversion of un-pretreated Jatropha oil with high-acid value (13.8 mg KOH/g) to biodiesel was studied in ionic liquids (ILs) with metal chlorides. Several commercial ILs were used to catalyze the esterification of oleic acid. It was found that 1-butyl-3-methylimidazolium tosylate ([BMIm][CH3SO3]; a Brønsted acidic IL) had the highest catalytic activity with 93% esterification rate for oleic acid at 140 °C but only 12% biodiesel yield at 120 °C. When FeCl3 was added to [BMIm][CH3SO3], a maximum biodiesel yield of 99.7% was achieved at 120 °C. Because metal ions in ILs supplied Lewis acidic sites, and more of the sites could be provided by trivalent metallic ions than those of bivalent ones. It was also found that the catalytic activity with bivalent metallic ions increased with atomic radius. Mixture of [BMIm][CH3SO3] and FeCl3 was easily separated from products for reuse to avoid producing pollutants.  相似文献

This study aimed to determine the effect of moisture content of three different feedstocks on overall ethanol yield. Switchgrass and sugarcane bagasse from two sources were either soaked in water (∼80% moisture) or left dry (∼12% moisture), and half each of these were impregnated with 3% w/w SO2 and all were steam pretreated. The twelve resulting substrates were compared based on overall sugar recovery after pretreatment, cellulose conversion following enzymatic hydrolysis, and ethanol yield following simultaneous saccharification and fermentation. The overall ethanol yield after simultaneous saccharification and fermentation of hexoses was 18-28% higher in samples that were soaked prior to SO2 addition than in SO2-catalyzed samples that were not soaked. In samples that were uncatalyzed, soaking made little difference, indicating that the positive effect of increased moisture content may be related to increased permeability of the biomass to SO2.  相似文献

The aim of this work is to study the catalyst prepared by glucose-starch mixture. Assessment experiments showed that solid acid behaved the highest esterification activity when glucose and corn powder were mixed at ratio of 1:1, carbonized at 400 °C for 75 min and sulfonated with concentrated H2SO4 (98%) at 150 °C for 5 h. The catalyst was characterized by acid activity measurement, XPS, TEM and FT-IR. The results indicated that solid acid composed of CS0.073O0.541 has both Lewis acid sites and Broˇnsted acid sites caused by SO3H and COOH. The conversions of oleic acid esterification and triolein transesterification are 96% and 60%, respectively. Catalyst for biodiesel production from waste cottonseed oil containing high free fatty acid (FFA 55.2 wt.%) afforded the methyl ester yield of about 90% after 12 h. The catalyst deactivated gradually after recycles usage, but it could be regenerated by H2SO4 treatment.  相似文献

Supercritical carbon dioxide extraction (SC-CO2) of oil from desilked silkworm pupae was performed. Response surface methodology (RSM) was applied to optimize the parameters of SC-CO2 extraction. The effects of independent variables, including pressure, temperature, CO2 flow rate, and extraction time, on the yield of oil were investigated. The statistical analysis showed that the pressure, extraction time, and the quadratics of pressure, extraction time, and CO2 flow rate, as well as the interactions between pressure and temperature, and temperature and flow rate, showed significant effects on oil yield. The optimal extraction condition for oil yield within the experimental range of the variables researched was at 324.5 bar, 39.6 °C, 131.2 min, and 19.3 L/h. At this condition, the yield of oil was predicted to be 29.73%. The obtained silkworm pupal oil contained more than 68% total unsaturated fatty acids, and alpha-linolenic acid (ALA) accounted for 27.99% in the total oil.  相似文献

Oil palm trunks are a possible lignocellulosic source for ethanol production. Low enzymatic digestibility of this type of material (11.9% of the theoretical glucose yield) makes pretreatment necessary. An enzymatic digestibility of 95.4% with insoluble solids recovery of 49.8% was achieved after soaking shredded oil palm trunks in ammonia under optimum conditions (80 °C, 1:12 solid-to-liquid ratio, 8 h and 7% (w/w) ammonia solution). Treatment with 60 FPU of commercial cellulase (Accellerase 1000) per gram of glucan and fermentation with Saccharomyces cerevisiae D5A resulted in an ethanol concentration of 13.3 g/L and an ethanol yield of 78.3% (based on the theoretical maximum) after 96 h. These results indicate that oil palm trunks are a biomass feedstock that can be used for bioethanol production.  相似文献

Regenerated films were successfully prepared from cellulose/NaOH/urea solution by coagulating with water at temperature from 25 to 45 °C. The results of solid 13C NMR, wide angle X-ray diffraction, scanning electron microscopy (SEM) and tensile testing revealed that the cellulose films possessed homogeneous structure and cellulose II crystalline, similar to that prepared previously by coagulating with 5 wt% H2SO4. By changing the coagulation temperature from 25 to 45 °C, tensile strength of the films was in the range of 85-139 MPa. Interestingly, the RC35 film coagulated at 35 °C exhibited the highest tensile strength (σb = 139 MPa). The inclusion complex associated with cellulose, NaOH and urea hydrates in the cellulose solution were broken by adding water (non-solvent), leading to the self-association of cellulose to regenerate through rearrangement of the hydrogen bonds. This work provided low-cost and “green” pathway to prepare cellulose films, which is important in industry.  相似文献

In this work, the potential of Acacia dealbata as raw material for ethanol production was evaluated, as well as its composition with regard to cellulose, hemicelluloses, lignin, extractives and ash. The tree samples were subjected to several dilute acid pretreatments using a combined severity parameter ranging from 0.7 to 3.7. The highest ethanol concentration obtained was 10.31 g ethanol/L within 24 h by using a separate hydrolysis and fermentation of the water insoluble fraction after pretreatment at 180 °C with 0.8% of sulfuric acid for 15 min. With simultaneous saccharification and fermentation, results obtained for the washed solids of water insoluble fraction were better than those obtained with the whole slurry.  相似文献

The effect of chemical pretreatments using NaOH, H2O2, and Ca(OH)2 on Empty Palm Fruit Bunches (EPFB) to degrade EPFB lignin before pyrolyis was investigated. Spectrophotometer analysis proved consecutive addition of NaOH and H2O2 decomposed almost 100% of EPFB lignin compared to 44% for the Ca(OH)2, H2O2 system while NaOH and Ca(OH)2 used exclusively could not alter lignin much. Next, the pretreated EPFB was catalytically pyrolyzed. Experimental results indicated the phenolic yields over Al-MCM-41 and HZSM-5 catalysts were 90 wt% and 80 wt%, respectively compared to 67 wt% yield for the untreated sample under the same set of conditions. Meanwhile, the experiments with HY zeolite yielded 70 wt% phenols.  相似文献

The present study describes production of bio-ethanol from fresh red alga, Kappaphycus alvarezii. It was crushed to expel sap - a biofertilizer - while residual biomass was saccharified at 100 °C in 0.9 N H2SO4. The hydrolysate was repeatedly treated with additional granules to achieve desired reducing sugar concentration. The best yields for saccharification, inclusive of sugar loss in residue, were 26.2% and 30.6% (w/w) at laboratory (250 g) and bench (16 kg) scales, respectively. The hydrolysate was neutralized with lime and the filtrate was desalted by electrodialysis. Saccharomyces cerevisiae (NCIM 3523) was used for ethanol production from this non-traditional bio-resource. Fermentation at laboratory and bench scales converted ca. 80% of reducing sugar into ethanol in near quantitative selectivity. A petrol vehicle was successfully run with E10 gasoline made from the seaweed-based ethanol. Co-production of ethanol and bio-fertilizer from this seaweed may emerge as a promising alternative to land-based bio-ethanol.  相似文献

A dual modification procedure composed of cross-linking and protein coating with K2SO4 was employed to modify Geotrichum sp. lipase for catalyzing biodiesel production from waste cooking oil. Compared to single modification of protein coating with K2SO4, the dual modification of cross-linking and lipase coating improved catalytic properties in terms of thermostable stability, organic solvent tolerance, pH stability and operational stability in biodiesel production process, although biodiesel yield and initial reaction rate for CLPCMCs were not improved. After five successive batch reactions, CLPCMCs could still maintain 80% of relative biodiesel yield. CLPCMCs retained 64% of relative biodiesel yield after incubation in a pH range of 4-6 for 4 h, and 85% of relative biodiesel yield after incubation in a range of 45-50 °C for 4 h. CLPCMCs still maintained 83% of relative biodiesel yield after both treated in polar organic solvent and non-polar organic solvent for 4 h.  相似文献

Spent mushroom substrate (SMS) was treated with dilute sulfuric acid followed by cellulase and xylanase treatment to produce hydrolysates that could be used as the basis for media for the production of value added products. A L9 (34) orthogonal experiment was performed to optimize the acid treatment process. Pretreatment with 6% (w/w) dilute sulfuric acid at 120 °C for 120 min provided the highest reducing sugar yield of 267.57 g/kg SMS. No furfural was detected in the hydrolysates. Exposure to 20 PFU of cellulase and 200 XU of xylanase per gram of pretreated SMS at 40 °C resulted in the release of 79.85 g/kg or reducing sugars per kg acid pretreated SMS. The dilute sulfuric acid could be recycled to process fresh SMS four times. SMS hydrolysates neutralized with ammonium hydroxide, sodium hydroxide, or calcium hydroxide could be used as the carbon source for cultivation of Lactococcus lactis subsp. lactis W28 and a cell density of 2.9 × 1011 CFU/mL could be obtained. The results provide a foundation for the development of value-added products based on SMS.  相似文献

Effects of different physical pretreatments on water hyacinth for dilute acid hydrolysis process (121 ± 3 °C, 5% H2SO4, 60 min) were comparatively investigated. Untreated sample had produced 24.69 mg sugar/g dry matter. Steaming (121 ± 3 °C) and boiling (100 ± 3 °C) for 30 min had provided 35.9% and 52.4% higher sugar yield than untreated sample, respectively. The highest sugar yield (132.96 mg sugar/g dry matter) in ultrasonication was obtained at 20 min irradiation using 100% power. The highest sugar production (155.13 mg sugar/g dry matter) was obtained from pulverized samples. Hydrolysis time was reduced when using samples pretreated by drying, mechanical comminution and ultrasonication. In most methods, prolonging the pretreatment period was ineffective and led to sugar degradations. Morphology inspection and thermal analysis had provided evidences of structure disruption that led to higher sugar recovery in hydrolysis process.  相似文献
