We have developed 11 microsatellite markers that are specific to Chattonella antiqua, C. marina, and C. ovata, the red tide‐forming harmful phytoplanktons. The 11 loci were amplified in the three species. The number of alleles per locus ranged from 5 to 16. The three species shared most microsatellite regions, although the genetic differences in specific loci were detected among them. These markers of the Chattonella species will be beneficial for biogeographical, detailed taxonomic, studies.  相似文献

The polyamines caldopentamine and homocaldopentamine were detected in axenic strains of Chattonella antiqua and Heterosigma akashiwo ( Raphidophyceae ), respectively, as well as spermidine, the most abundant polyamine in both phytoplankton species. Trace amounts of putrescine, diaminopropane and norspermine were also detected in both species. Spermine was detected only from C. antiqua . These long linear polyamines are characteristic components of thermophilic bacteria. The detection from two species of Raphidophyceae indicates that the occurrence of long linear polyamines is not restricted to thermophilic microorganisms.  相似文献

These loci were highly variable, with between 2 and 10 alleles per locus, and estimated gene diversity ranging from 0.08 to 0.82. During a bloom of Heterosigma akashiwo (Raphidophyceae) in northernHiroshima Bay, Japan, in June 1993, the proportion of H.akashiwocells containing virus-like particles (VLPs) was monitored bytransmission electron microscopy. Until 3 days before the terminationof the red tide, no VLP-containing cells were detected, andthe proportion of VLP-containing cells was <1% on the last2 days of the red tide. However, the sample collected on thelast day, continuously incubated for 26 h at 22°C, revealeda high frequency (11.5%) of VLP-containing cells. These findingssuggest that viral mortality occurs in quite a short time andmay play an important role in regulating the disintegrationof H.akashiwo red tide.  相似文献

Outbreaks of paralytic shellfish poisoning caused by the toxic dinoflagellate Alexandrium minutum (Dinophyceae) are a worldwide concern from both the economic and human health points of view. For population genetic studies of A. minutum distribution and dispersal, highly polymorphic genetic markers are of great value. We isolated 12 polymorphic microsatellites from this cosmopolitan, toxic dinoflagellate species. The outbreak of paralytic shellfish poisoning caused by the toxic dinoflagellate Alexandrium catenella (Dinophyceae) is a worldwide concern from the economic and food hygiene point of view. To assess the dispersal mechanism, a population genetic study using highly polymorphic genetic markers is one of the crucial approaches. We isolated 12 polymorphic microsatellites from this species. These loci provide one class of highly variable genetic marker as the number of alleles ranged from three to 12 and the estimate of gene diversity was from 0.248 to 0.897 across the 12 microsatellites; we consider that these loci have a potential to detail the genetic structure and gene flow among A. catenella populations.  相似文献

Temporal fluctuations of algicidal micro-organisms against the red tide causing raphidophycean flagellates Chattonella antiqua (Hada) Ono and Heterosigma akashiwo (Hada) Hada ex Hara et Chihara were investigated using the microplate most probable number (MPN) method in northern Hiroshima Bay and Harima-Nada, the Seto Inland Sea, in 1992 and 1993. In Har-ima-Nada, both flagellates appeared at low levels (< 1 cell mL?1), and killer micro-organisms against the two flagellates (C-killer for C. antiqua and H-killer for H. akashiwo) also appeared at low densities (< 2 mL?1). In northern Hiroshima Bay, C. antiqua cells were scarce (< 1 cell mL?1), and C-killers occurred at a low level (≤ 3.4 mL?1). Conversely, red tides of H. akashiwo occurred there in June of both years. The dynamics of H-killers revealed a close relationship with that of H. akashiwo populations. H-killers followed the increase of H. akashiwo cells, reached a maximum level after the beginning of decline of H. akashiwo, maintained a high level for at least 1 week after the crash of bloom, and then decreased. C-killers consistently remained at low densities during the period of H. akashiwo red tides in both 1992 and 1993. Hence, algicidal micro-organisms specifically associated with the occurrence and crash of H. akashiwo red tides, and presumably contributed to the rapid termination of the red tides in the coastal seas such as northern Hiroshima Bay.  相似文献

Seven microsatellite loci were isolated and characterized from the red‐capped robin Petroica goodenovii, using nonradioactive polymerase chain reaction (PCR)‐based techniques to screen an enriched genomic library. Five loci showed no evidence of null alleles and were variable [mean heterozygosity (HE) = 0.440, mean number of alleles = 8]. Cross‐amplification using primers for microsatellites in Phylloscopus occipitalis and Emberiza schoeniclus yielded another two polymorphic loci. The combined set of five red‐capped robin and two cross‐amplified loci are suitable for paternity assignment (exclusion probability for seven unlinked loci = 0.9760).  相似文献

These loci provide one class of highly variable genetic marker as the number of alleles ranged from three to 12 and the observed and expected heterozygosities ranged from 0.130 to 0.783 and from 0.163 to 0.862, respectively. To aid in understanding the structure, dispersal and genetic dynamics of their populations, we developed microsatellite and amplified fragment length polymorphism (AFLP) markers for the sea palm, Postelsia palmaeformis (Laminariales) for samples taken from nine sites in the area of Cape Flattery, Washington State, USA. We identified two AFLP primers that yielded 798 variable fragments and five microsatellite markers with three to seven alleles each. We also report patterns of allelic variation for four previously identified microsatellite markers in this species and several new alleles.  相似文献

Todarodes pacificus is an important species for commercial fisheries, but its reproductive biology has been little understood. We isolated 11 polymorphic microsatellite loci from this species. These loci provided highly polymorphic microsatellite markers with four to 26 alleles per locus. The observed and expected heterozygosities ranged from 0.150 to 1.000, and from 0.660 to 0.974, respectively. These markers will be useful for research about the mating systems and genetic diversity of T. pacificus.  相似文献

Black locust (Robinia pseudoacacia) is an economically and ecologically important tree species in the world. We isolated seven polymorphic microsatellite loci from R. pseudoacacia using a dual‐suppression‐PCR technique. These loci provided microsatellite markers with high polymorphism ranging from three to 12 alleles per locus and expected heterozygosity between 0.538 and 0.944. The markers are now available for more detailed investigation of population genetic structure and pollen and seed dispersal.  相似文献

Betula platyphylla var. japonica is a typical pioneer tree species in the secondary succession in northern Japan. We describe the cloning and characterization of 13 polymorphic, codominant microsatellite loci isolated from this species. These polymorphic loci had 2–8 alleles per locus and a range of expected heterozygosities from 0.050 to 0.808.  相似文献

The grey‐crowned babbler (Pomatostomus temporalis) is a cooperative breeding bird species in which nonbreeding helpers of both sexes care for the young of breeding individuals. To measure the genetic relatedness between breeders and their offspring and helpers, we developed nine microsatellite markers. Most of the loci were highly polymorphic. These loci will be useful in understanding the evolution and maintenance of cooperative breeding and helping behaviour in this species.  相似文献
