Refixation of xylem sap CO2 in Populus deltoides   总被引:1,自引:0,他引:1  
Vascular plants have respiring tissues which are perfused by the transpiration stream, allowing solubilization of respiratory CO2 in the xylem sap. The transpiration stream could provide a conduit for the internal delivery of respiratory CO2 to leaves. Trees have large amounts of respiring tissues in the root systems and stems, and may have elevated levels of CO2 in the xylem sap which could be delivered to and refixed by the leaves. Xylem sap from the shoots of three Populus deltoides trees had mean dissolved inorganic carbon concentrations (CO2+H2CO3+HCO?3) ranging from 0. 5 to 0. 9 mM. When excised leaves were allowed to transpire 1 mM[14C]NaHCO3, 99. 6% of the label was fixed in the light. Seventy-seven percent of the label was fixed in major veins and the remainder was fixed in the minor veins. Autoradiography confirmed that label was confined to the vasculature. In the dark, approximately 80% of the transpired label escaped the leaf, the remainder was fixed in the major veins, slightly elevating dark respiration measurements. This indicates that the vascular tissue in P. deltoides leaves is supplied with a carbon source distinct from the atmospheric source fixed by interveinal lamina. However, the contribution of CO2 delivered to the leaves in the transpiration stream and fixed in the veins was only 0. 5% of atmospheric CO2 uptake. In the light 90% of the label was found in sugar, starch and protein, a pattern similar to that found for atmospheric uptake of[14C]CO2. Compared with leaves labelled in the light, leaves labelled in the dark had more label in organic acid, amino acid and protein and less label in sugar and starch. After a 5-s pulse the majority of the label fed to petioles in both the light and the dark was found in malate. The majority of the label was found in malate at 120 s in the dark; only 2% of the label was found in phosphorylated compounds at 120 s. The proportion of label found in phosphorylated compounds increased from 17% at 5 s to 80% at 120 s in the light. This suggests that CO2 delivered to leaves in the light via the transpiration stream is fixed in the veins, a small portion through dark fixation into malate, the remainder by C-3 photosynthesis.  相似文献

Partitioning of exogenously supplied U-14C-saccharose into primary metabolic pool as sugars, amino acids, and organic acids was analyzed and simultaneous utilization for production of alkaloid by leaf, stem, and root in twigs and rooted plants of Catharanthus roseus grown in hydroponic culture medium was determined. Twigs revealed comparable distribution of total 14C label in leaf and stem. Stems contained significantly higher 14C label in sugar fraction and in alkaloids [47 kBq kg−1(DM)] than leaf. In rooted plants, label in 14C in metabolic fractions in root such as ethanol-soluble, ethanol-insoluble, and chloroform-soluble fractions and in components such as sugars, amino acids, and organic acids were significantly higher than in stems and leaves. This was related with significantly higher content of 14C in alkaloids in stems and leaves. 14C contents in sugars, amino acids, and organic acids increased from leaf to stem and roots. Roots are the major accumulators of metabolites accompanied by higher biosynthetic utilization for alkaloid accumulation.  相似文献

Incubation of intact pea plants (Pisum sativum), or detached shoots, in continuous light caused a substantial increase (up to 4-fold in 2 days) in levels of homoserine. Amino acids supplied to leaves in the transpiration stream enhanced the accumulation, with glutamate, aspartate, and asparagine causing similar enhancement. Aminooxyacetate (AOA), a transamination inhibitor, at 1 millimolar prevented the accumulation. 14C-labeling experiments showed that succinate was a good source of carbon for homoserine synthesis; carbon from aspartate or asparagine was also incorporated into homoserine. For each precursor, the transfer of label was prevented by AOA. The keto acid analog of homoserine was rapidly transaminated in leaves to give homoserine. The results suggest that accumulating homoserine is synthesised by transamination rather than being derived from aspartate via the aspartate kinase/homoserine dehydrogenase pathway. The latter pathway was shown to be operating in the chloroplasts, and was sensitive to threonine (but was not inhibited by AOA), suggesting that this path has a role in synthesis of aspartate-derived amino acids but is not involved in the accumulation of excess homoserine in the pea.  相似文献

The net partitioning of current photosynthate among vegetative organs of nodulated alfalfa (Medicago sativa L.) was investigated by determining radiolabel distribution from upper and lower source leaves, at different times of day, and during successively longer chase periods. Photosynthate was exported sooner and more completely by a fully expanded lower than by a fully expanded upper source leaf. The radiolabel pulse from the lower source leaf peaked in the main stem within 1.5 hours, in the crown and nodules after 3 hours, in the unexpanded leaves and apex of the main stem after 6 hours, and in the shoots growing from leaf axils on the main stem after 24 hours. The results suggest that the crown, apex, axillary shoots, and nodules both imported and mobilized photosynthate originating at a lower source leaf, while roots and shoots growing from the crown showed net accumulation. The pulse from the upper source leaf was initially rapidly exported by the main stem and imported by the root between 1.5 and 3 hours, but there was no net change of label content of these organs during the ensuing 21 hours. Rapidly growing organs had the highest concentrations, and the largest organs had the highest content of radiolabel. These results provide new information about the accumulation and circulation of photosynthate within the alfalfa plant.  相似文献

* Proposed mechanisms of embolism recovery are controversial for plants that are transpiring while undergoing cycles of dehydration and rehydration. * Here, water stress was imposed on grapevines (Vitis vinifera), and the course of embolism recovery, leaf water potential (Psi(leaf)), transpiration (E) and abscisic acid (ABA) concentration followed during the rehydration process. * As expected, Psi(leaf) and E decreased upon water stress, whereas xylem embolism and leaf ABA concentration increased. Upon rehydration, Psi(leaf) recovered in 5 h, whereas E fully recovered only after an additional 48 h. The ABA content of recovering leaves was higher than in droughted controls, both on the day of rewatering and the day after, suggesting that ABA accumulated in roots during drought was delivered to the rehydrated leaves. In recovering plants, xylem embolism in petioles, shoots, and roots decreased during the 24 h following rehydration. * A model is proposed to describe plant recovery after rehydration based on three main points: embolism repair occurs progressively in shoots and further in roots and in petioles, following an almost full recovery of Psi(leaf); hydraulic conductance recovers during diurnal transpiring hours, when formation and repair of embolisms occurs in all plant organs; an ABA residual signal in rehydrated leaves hinders stomatal opening even when water relations have recovered, suggesting that an ABA-induced transpiration control promotes gradual embolism repair in rehydrated grapevines.  相似文献

Studies were conducted to determine whether protein phosphorylation may be a mechanism for regulation of spinach (Spinacia oleracea L.) leaf sucrose-phosphate synthase (SPS), shown previously to be light-dark regulated by some type of covalent modification. Radioactive phosphate was incorporated into the 120-kDa subunit of SPS during labeling of excised leaves with [32P]Pi, as shown by immunoprecipitation and denaturing gel electrophoresis of the enzyme. Conditions which activated the enzyme (illumination of leaves or mannose treatment of leaf discs in darkness) reduced the incorporation of radiolabel into SPS in the in vivo system. The partially purified SPS protein could also be phosphorylated in vitro using [gamma-32P]ATP. In the in vitro system, the incorporation of radiolabel into the 120-kDa subunit of SPS was dependent on time and magnesium concentration, and was closely paralleled by inactivation of the enzyme. These results provide the first evidence to establish protein phosphorylation as a mechanism for the covalent regulation of SPS activity.  相似文献

Structural and hydraulic correlates of heterophylly in Ginkgo biloba   总被引:1,自引:0,他引:1  
This study investigates the functional significance of heterophylly in Ginkgo biloba, where leaves borne on short shoots are ontogenetically distinct from those on long shoots. Short shoots are compact, with minimal internodal elongation; their leaves are supplied with water through mature branches. Long shoots extend the canopy and have significant internodal elongation; their expanding leaves receive water from a shoot that is itself maturing. Morphology, stomatal traits, hydraulic architecture, Huber values, water transport efficiency, in situ gas exchange and laboratory-based steady-state hydraulic conductance were examined for each leaf type. Both structure and physiology differed markedly between the two leaf types. Short-shoot leaves were thinner and had higher vein density, lower stomatal pore index, smaller bundle sheath extensions and lower hydraulic conductance than long-shoot leaves. Long shoots had lower xylem area:leaf area ratios than short shoots during leaf expansion, but this ratio was reversed at shoot maturity. Long-shoot leaves had higher rates of photosynthesis, stomatal conductance and transpiration than short-shoot leaves. We propose that structural differences between the two G. biloba leaf types reflect greater hydraulic limitation of long-shoot leaves during expansion. In turn, differences in physiological performance of short- and long-shoot leaves correspond to their distinct ontogeny and architecture.  相似文献

Effects of the length: width ratio of a leaf blade and petiole length on shoot light capture were studied with computer simulation. Both a larger length: width ratio and longer petiole contributed to larger light capture per unit leaf area due to a reduced aggregation of leaf area around the stem. Other conditions being equal, shoots with narrow leaves and no petioles and those with wide leaves with petioles showed similar light capture as long as the mean distance of the leaf blade from the stem was the same. In shoots with a short internode and/or distichous phyllotaxis, however, narrow leaves contributed more to avoiding mutual shading than wide leaves with petioles. The predominance of light coming from a higher angular altitude also favored narrow leaves. The possible consequences of these results in the adaptive geometry of plant architecture are discussed.  相似文献

Several lines of evidence indicate that glucose and fructose are essentially absent in mobile phloem sap. However, this paradigm has been called into question, especially but not entirely, with respect to species in the Ranunculaceae and Papaveraceae. In the experiments in question, phloem sap was obtained by detaching leaves and placing the cut ends of the petioles in an EDTA solution. More hexose than sucrose was detected. In the present study, these results were confirmed for four species. However, almost identical results were obtained when the leaf blades were removed and only petiole stubs were immersed. This suggests that the sugars in the EDTA solution represent compounds extracted from the petioles, rather than sugars in transit in the phloem. In further experiments, the leaf blades were exposed to (14)CO(2) and, following a chase period, radiolabelled sugars in the petioles and EDTA exudate were identified. Almost all the radiolabel was in the form of [(14)C]sucrose, with little radiolabelled hexose. The data support the long-held contention that sucrose is a ubiquitous transport sugar, but hexoses are essentially absent in the phloem stream.  相似文献

Morphology, water relations, and xylem anatomy of high-light (sun)- and low-light (shade)-grown Vitis vinifera L. shoots were studied to determine the effects of shading on the hydraulic conductance of the pathway for water flow from the roots to the leaves. Shade shoots developed leaf area ratios (leaf area: plant dry weight) that were nearly threefold greater than sun shoots. Water-potential gradients (·m–1) in the shoot xylem accounted for most of the ·m–1 between soil and shoot apex at low and high transpiration rates in both sun and shade shoots, but the gradients were two- to fourfold greater in shade-grown plants. Low light reduced xylem conduit number in petioles, but had an additional slight effect on conduit diameter in internodes. The hydraulic conductance per unit length (Kh) and the specific hydraulic conductivity (ks, i.e. Kh per xylem cross-sectional area) of internodes, leaf petioles, and leaf laminae at different developmental stages leaf plastochron index was calculated from measurements of water potential and water flow in intact plants, from flow through excised organs, and from vessel and tracheid lumen diameters according to Hagen-Poiseuille's equation. For all methods and conductance parameters, the propensity to transport water to sink leaves was severalfold greater in internodes than in petioles. The Kh and ks increased logarithmically until growth ceased, independent of treatment and measurement method, and increased further in pressurized-flow experiments and Hagen-Poiseuille predictions. However, the increase was less in shade internodes than in sun internodes. Mature internodes of shade-grown plants had a two- to fourfold reduced Kh and significantly lower ks than sun internodes. Except very early in development, leaf lamina conductance and ks from shade-grown plants was also reduced. The strong reduction in Kh with only a slight reduction in leaf area (17% of sun shoots) in the shade shoots indicated a decoupling of water-transport capacity from the transpirational surface supplied by that capacity. Trees of Sassafras albidum (Nuttall) Nees. display leaves that fall into discrete categories of form. Unlobed leaves, with undissected margins, predominate at proximal and distal nodes of shoots, while lobed leaves are most common at intermediate nodes. In this study we investigated the hypothesis that shoots of sassafras respond to environmental changes over the growing season by generating predictable nodal patterns for leaf morphology and physiology. We recorded leaf shape category and measured leaf surface area, node number, chlorophyll content, nitrogen content, and photosynthesis in four trees. Over 1,000 measurements of photosynthesis were made in situ, using the LI-Cor LI 6200 portable photosynthesis system. The two trees growing under heavy shade had few lobed leaves (2.0% and 18.8%) and often had negative carbon balance, with positive net photosynthesis occurring during sun flecks. The two trees growing in more open conditions had many more lobed leaves (56.3% and 61.0%) and higher, less variable net photosynthetic rates. As indicated by chlorophyll and nitrogen contents, the highest leaf photosynthetic rates occurred at intermediate nodes (nodes six to nine), and this was shifted distally along the shoot during the growing season. Leaves at intermediate nodes also had the largest surface areas and the greatest frequency of lobing. In comparative experiments with models, lobing was shown to enhance free-convectional heat loss relative to unlobed leaves of the same surface area. Due to their large surface area, these leaves also have the highest rates of whole leaf photosynthesis. Under conditions of equivalent photosynthetically active radiation, lobed leaves had higher rates of net photosynthesis than did unlobed leaves. We conclude that shoots of sassafras produce groups of leaves with predictably different morphological and physiological specializations.  相似文献

Diurnal pattern of acetaldehyde emission by flooded poplar trees   总被引:7,自引:0,他引:7  
The emission of the tropospheric trace gas acetaldehyde was determined in leaves of 4-month-old poplar trees ( Populus tremula × P. alba ) grown under controlled environmental conditions in a greenhouse. Using a dynamic cuvette system together with a high sensitivity laser-based photoacoustic detection unit, rates of acetaldehyde emission were measured with the high time resolution of about 15 min. Submergence of the roots resulted in the emission of acetaldehyde by the leaves. The emission increased linearly before reaching more or less steady-state values (ca 350 nmol m−2 min−1; ca 470 ng g−1 dry weight min−1) after approximately 6 h. Prolonged flooding of poplar trees resulted in a clear diurnal rhythm of acetaldehyde emission. The emission rates decreased when the light was switched off in the evening and peaked in the morning after the light was turned on again. This pattern significantly correlated with diurnal rhythms of stomatal conductance, photosynthesis, transpiration and with the concentrations of ethanol, the assumed precursor of acetaldehyde, in the xylem sap of flooded poplar trees. It may be concluded that under conditions of diminished stomatal conductance, acetaldehyde emission declines because its diffusive flux is reduced. Alternatively, reduced transpiration may decrease ethanol transport from the roots to the shoots and appreciable amounts of the acetaldehyde precursor ethanol are lacking in the leaves. The present results support the view that acetaldehyde emitted by the leaves of plants is derived from ethanol produced by alcoholic fermentation in submerged roots and transported to the leaves with the transpiration stream.  相似文献

Effects of kinetin on transpiration rate and abscisic acid content were determined. Leaves from 9-day-old wheat plants (Triticum aestivum L. cv. Weibull's Starke II) were used. —Transpiration rate decreased in excised leaves put in water, but it was maintained at a higher rate when kinetin was supplied. When excised leaves were water stressed by air-drying for 1 h, addition of kinetin resulted in a considerable stimulation of transpiration rate. The effect reached its maximum after 15 h and this level remained relatively unchanged for at least 10 h. Intact seedlings which were stressed before leaf excision, showed only a slight stimulation of kinetin on transpiration rate. — Abscisic acid content slowly increased up to three-fold in 2 days in excised leaves put in water. In excised and water-stressed leaves the abscisic acid content was reduced during the first 24 h and then increased. As the leaves were fully turgid, the increase could not have been caused by water stress. However, both in stressed and unstressed leaves kinetin addition reduced the increase in abscisic acid content. — It is suggested that the stimulation by kinetin on transpiration rate in excised and water stressed leaves was mainly due to the combined effect of (1) a reduction in the activity of endogenous cytokinins, (2) kinetin acting as a ‘substitute’ for the inactivated cytokinins but exerting a stronger effect on transpiration than the endogenous cytokinins, and (3) the ‘extra’ reduction in abscisic acid content caused by the kinetin treatment. Furthermore, the results indicate that changes in cytokinins might be partly responsible for the aftereffect on transpiration.  相似文献

The effect of nitrogenous nutrients on endogeneous cytokinins and senescence of tobacco leaves was investigated. Ammonium nitrate was the most effective in retarding senescence and its activity was attributed principally to NH4+ ions. Repeated applications or a continuous supply of ammonium nitrate was required for maximal retardation of tobacco leaf senescence. Ammonium nitrate solution supplied via the petioles reduced the senescence retarding effect of dihydrozeatin applied directly to the laminae of detached tobacco leaves. Ammonium nitrate also elevated the endogenous levels of cytokinins (especially zeatin and dihydrozeatin) particularly in growing tobacco leaves excised from near the apex of the plant. Ammonium nitrate induced retardation of leaf senescence may be mediated at least partly by its effect on foliar cytokinin content.  相似文献

Cytokinin-like activity in extracts of leaf laminae, petioles, shoots, roots and flowers of young plants of the water hyacinth, Eichhornia crassipes S. was analyzed following Sephadex LH-20 column chromatography using the soybean callus bioassay. In all plant parts analyzed, two prominent peaks of cytokinin activity having elution volumes similar to zeatin and zeatin riboside were detected. Putative cytokinin gluco-side-like activity was detected only in leaves and flowers. The cytokinin complements of the leaves and the roots were qualitatively different. It would appear that cytokinins supplied by the roots are metabolized in the leaves or certain cytokinins are synthesized in the leaves themselves. The possible significance and distribution of cytokinins in different plant parts in relation to roots is discussed.  相似文献

The capacity of tomato leaf tissues to accumulate proline in response to a salt shock (150 mM NaCl) applied to excised shoots, leaves, leaflets or leaf discs was determined and compared to that of whole plants grown at the same salinity. The associated changes in free amino acids, Na+, K+ and Cl- contents were also investigated. In excised organs treated for 80 h, up to 200 mumol g-1 DW of proline were accumulated, whereas the amount of proline in leaf discs did not exceed a value ten-fold lower. In the whole plants subjected to salinity the Na+, Cl- and K+ contents remained low in comparison to that observed in excised organs. Proline and other amino acids increased more slowly in whole plants than in excised shoots. The contribution of roots and vascular tissues to the control of Na+ and Cl- accumulation and to the regulation of proline metabolism are discussed.  相似文献

The leaves of trees emit significant amounts of acetaldehyde which is synthesized there by the oxidation of ethanol. In the present study, we examined plant internal and environmental factors controlling the emission of acetaldehyde by the leaves of young poplar ( Populus tremula × P. alba ) trees. The enzymes possibly involved in the oxidation of ethanol in the leaves of trees are catalase (CAT; EC 1.11.1.6) and alcohol dehydrogenase (ADH; EC 1.1.1.1), both expressed constitutively in the leaves of poplars. Inhibition of ADH in excised leaves caused a significant decrease of acetaldehyde emission accompanied by an increased ethanol emission. Since inhibition of CAT by aminotriazole did not affect acetaldehyde and ethanol emission, it is concluded that the oxidation of ethanol in the leaves is mediated by ADH rather than by CAT. Further studies indicated that aldehyde dehydrogenase (ALDH; EC 1.2.1.5) seems to be responsible for the oxidation of acetaldehyde. The present results demonstrate that acetaldehyde emission is clearly dependent on its production in the leaves as controlled by the delivery of ethanol to the leaves via the transpiration stream. Environmental factors that control stomatal conductance seem to be of less importance for acetaldehyde emission by the leaves.  相似文献

Procedures are described for producing axillary shoots fromseedling apices and adventitious shoots from petioles and leaf-derivedcallus of sugar beet cultivars. The rate of adventitious shootregeneration from petioles was influenced by temperature, BAPconcentration of the medium, and the time in culture of theseedling apices from which the petioles were excised. Petiolesectioning confirmed that adventitious shoots originated inthe sub-epidermal parenchyma. Two distinct types of callus wereproduced from leaf explants, but only white friable callus wascapable of shoot development. This callus developed from browntissue and was composed of thin-walled cells with dense cytoplasmand prominent nuclei. Green compact callus with thick-walledlignified cells developed from green tissue, but did not produceshoots. Successful seed sterilization and shoot regenerationfrom petiole explants and callus was cultivar-dependent. Adventitiousshoots were rooted and successfully transplanted to pottingcompost under glasshouse conditions. With tomato spotted wilt virus in petunia leaf strips, N-6 benzyladenine (BA) was as effective as kinetin in decreasing numbers of local lesions, a result which could not be attributed to an effect on the virus per se. Benzimidazole, adenine and ammonium nitrate were without effect. Benzyladenine was more effective than kinetin when supplied through the petioles of excised whole leaves. Local lesions and infectivity of TSWV in detached leaves of Nicotiana rustica were decreased by supplying BA before and after inoculation. Lesions and infectivity were also decreased in attached leaves when BA was applied 9 days before inoculation. BA supplied to attached leaves after inoculation increased infectivity. Supplying BA to the lower leaves of tomato plants before inoculating with TSWV decreased infectivity of unsprayed, systemically infected tip leaves taken as inoculum; BA supplied after inoculation increased infectivity. Local lesions caused by lucerne mosaic virus in excised leaves of Phaseolus vulgaris were decreased in number by supplying BA. The effects of pre- and post-inoculation sprays of BA are considered in relation to cell metabolism. Since pretreating leaves with kinins did not prevent infection, it is suggested that those which move freely through plants without adverse effects on normal growth may prove of value in increasing the tolerance of plants to virus infection.  相似文献

Seasonal changes in photosynthesis of apple trees (Malus domestica Borkh.) were monitored to examine the effect of source-sink interactions on photosynthesis and photorespiration. Elevated photosynthetic rates were observed during two periods of the growing season and correlated with the fruiting process. The first period of increased photosynthetic rates was during the bloom period, when spur leaves on flowering shoots exhibited up to 25% higher photosynthetic rates than vegetative spur leaves on a leaf area basis. CO2 assimilation rates were also higher in fruiting trees than nonfruiting trees during the period of rapid fruit growth from July to September. Photorespiration, dark respiration, leaf resistance, and transpiration exhibited no seasonal changes which correlated to the presence or absence of fruit. These data represent the first comprehensive examination of the effects of flowering/fruit formation on photosynthesis and photorespiration in perennial plants.  相似文献
