Summary The present work demonstrates that a normal stage-XIII hypoblast, if submitted to an X-ray dose of 6,000 rads (sufficient to stop cell division), retains the capacity to induce axis formation on the chick epiblast. Examination of the process of hypoblast formation indicates, in agreement with previous findings, that a hypoblastic layer can form in the absence of cell division from an irradiated stage-X blastoderm. Furthermore, it is observed that this hypoblastic layer, formed in the absence of cell division, can induce the development of an embryonic axis when combined with a normal stage-XIII epiblast.  相似文献

The signals which induce vertebrate neural tissue and pattern it along the anterior-posterior (A-P) axis have been proposed to emanate from Spemann's organizer, which in mammals is a structure termed the node. However, mouse embryos mutant for HNF3 beta lack a morphological node and node derivatives yet undergo neural induction. Gene expression domains occur at their normal A-P axial positions along the mutant neural tubes in an apparently normal temporal manner, including the most anterior and posterior markers. This neural patterning occurs in the absence of expression of known organizer genes, including the neural inducers chordin and noggin. Other potential signaling centers in gastrulating mutant embryos appear to express their normal constellation of putative secreted factors, consistent with the possibility that neural-inducing and -patterning signals emanate from elsewhere or at an earlier time. Nevertheless, we find that the node and the anterior primitive streak, from which the node derives, are direct sources of neural-inducing signals, as judged by expression of the early midbrain marker Engrailed, in explant-recombination experiments. Similar experiments showed the neural-inducing activity in HNF3 beta mutants to be diffusely distributed. Our results indicate that the mammalian organizer is capable of neural induction and patterning of the neural plate, but that maintenance of an organizer-like signaling center is not necessary for either process.  相似文献

In order to elucidate the functional role of cercomer in larvae of the monocercus type their transplantation from the specific host Chironomus obtusidens to Gammarus lacustris was conducted. At early stages after the transplantation proceeds an increase in the functional activity of the tegument of follicles of the cercomer followed by their complete destruction in 3 or 4 days. On the surface of the exocyst membrane an adhesion of the host's haemocytes occurs, which becomes more distinct in 3-4 days when the process acquires a character of local encapsulation. Within the same period, in the places of haemocytes aggregation, a local resorption of the exocyst external membrane takes place. Later intensification of the host response to transplant is associated with the destruction of follicles of the cercomer. In one case the occurrence of follicles of the cercomer in the cavity of cysticercoid was observed that is caused by the microbe affection of the latter. In the zone of contact of the tegument of scolex and neck with follicles of the cercomer an increased secretion (the microapocrine type) of the tegument, disturbance of the microvillous tegument of the cercomer's follicles and their destruction are observed. Incompatibility of the tegument of definitive departments and cercomer, which arises during differentiation of larvae, is supposed to affect the formation of scolex invagination in the evolution of larvae of Hymenolepis.  相似文献

Synopsis Different types of ontogenies in fishes-indirect and direct- are correlated with different nutrient availability and feeding during early life history. A comprehensive life-history model, developed earlier, facilitates the understanding of decisive events in the life of an organism. Embryos with insufficient endogenous food supply (yolk) to build a definitive phenotype directly need the transient form of a nutrient-gathering larva. They represent an indirect development. In contrast, a large endogenous supply of nutrients enables the definitive adult phenotype to develop directly, avoiding an intervening larva and the cost of metamorphosis. The larger and more advanced an individual at the onset of exogenous feeding, the better are its chances to survive. This can be achieved by heterochronies related to feeding. Different types of feeding during the early ontogeny of fishes — endogenous, exogenous, absorptive, and a combination of all (mixed) — are demonstrated and integrated into the life-history model.  相似文献

The purpose of this article is to describe briefly the methods by which the intra-mitochondrial volume may be measured both in vitro and in situ, to summarise the mechanisms thought to regulate the mitochondrial volume and then to review in more detail the evidence that changes in the intra-mitochondrial volume play an important part in the regulation of liver mitochondrial metabolism by glucogenic hormones such as glucagon, adrenaline and vasopressin. It will be shown that these hormones cause an increase in matrix volume sufficient to produce significant activation of fatty acid oxidation, respiration and ATP production, pyruvate carboxylation, citrulline synthesis and glutamine hydrolysis. These are all processes activated by such hormones in vivo. I will go on to demonstrate that the increase in matrix volume is brought about by an increase in mitochondrial [PPi]. This is able to stimulate K+ entry into the matrix, perhaps through an interaction with the adenine nucleotide translocase. The rise in matrix [PPi] is a consequence of an increase in cytosolic and hence mitochondrial [Ca2+] which inhibits mitochondrial pyrophosphatase. In the final section of the review I provide evidence that changes in mitochondrial volume may be important in the responses of a variety of tissues to hormones and other stimuli. I write as a metabolist with a working knowledge of bioenergetics rather than the converse, and this will certainly be reflected in the approach taken. If I cause offence to any dedicated experts in the field of bioenergetic by my ignorance or lack of understanding of their studies I can only offer my apologies and ask to be corrected.  相似文献

This study addresses changes in health which were consequential to the Neolithic transition in the southern Levant, judged on the basis of the study of specific and nonspecific stress indicators, trauma, and degenerative joint disease in 200 Natufian (hunter‐gatherer) skeletons (10,500–8300 BC) and 205 Neolithic (agricultural) skeletons (8300–5500 BC) from the southern Levant. The comparison of the health profiles of pre‐Neolithic (Natufian) and Neolithic populations reveals a higher prevalence of lesions indicative of infectious diseases among the Neolithic population, and an overall reduction in the prevalence of skull trauma among males. No change over time was observed in the prevalence of degenerative joint disease. These results indicate that in the southern Levant the Neolithic transition did not simply lead to an overall deterioration in health but rather resulted in a complex health profile which was shaped by 1) an increase exposure to disease agents, 2) changes in diet, 3) population aggregation in larger and denser settlements, 4) changes in activity patterns and the division of labor, and possibly 5) a higher resistant immunological system and response capacity to environmental aggressions (mainly infections). Effects of novel or relevant (a single exposure to experimental chamber) and irrelevant (20 exposures to experimental chamber) stimuli on the levels of serotonin (5-HT) and its metabolite 5-hydroxyindoleacetic acid (5-HIAA) in the left and right hippocamp and amygdala were studied in male and female rats. It was found that hemispheric specificity of 5-HT metabolism in hippocampus and amygdala depended on sex and novelty of information. In male rats, the hippocampal level of 5-HT in response to the novel stimulus increased in the left hemisphere, and the 5-HIAA hippocampal level increased bilaterally in response to irrelevant stimulus. In females, an increase in 5-HT and/or 5-HIAA levels was observed only in the left hippocampus in response both to relevant and irrelevant stimuli. In the amygdala, a hemispheric asymmetry of the 5-HT involvement, due to right-hemispheric changes in 5-HT metabolism, was observed only in male rats. In females, an increase in 5-HT level was found in the left and right amygdalas in response to irrelevant stimulus. These data suggest that serotonergic neurotransmitter mechanisms are an important factor which determines hemispheric and sex differences in selective attention.  相似文献

Studies on the rate of adverse events in hospitalized patients seldom examine temporal patterns. This study presents evidence of both weekly and annual cycles. The study is based on a large and diverse data set, with nearly 5 yrs of data from a voluntary staff-incident reporting system of a large public health care provider in rural southeastern Australia. The data of 63 health care facilities were included, ranging from large non-metropolitan hospitals to small community and aged health care facilities. Poisson regression incorporating an observation-driven autoregressive effect using the GLARMA framework was used to explain daily error counts with respect to long-term trend and weekly and annual effects, with procedural volume as an offset. The annual pattern was modeled using a first-order sinusoidal effect. The rate of errors reported demonstrated an increasing annual trend of 13.4% (95% confidence interval [CI] 10.6% to 16.3%); however, this trend was only significant for errors of minor or no harm to the patient. A strong "weekend effect" was observed. The incident rate ratio for the weekend versus weekdays was 2.74 (95% CI 2.55 to 2.93). The weekly pattern was consistent for incidents of all levels of severity, but it was more pronounced for less severe incidents. There was an annual cycle in the rate of incidents, the number of incidents peaking in October, on the 282 nd day of the year (spring in Australia), with an incident rate ratio 1.09 (95% CI 1.05 to 1.14) compared to the annual mean. There was no so-called "killing season" or "July effect," as the peak in incident rate was not related to the commencement of work by new medical school graduates. The major finding of this study is the rate of adverse events is greater on weekends and during spring. The annual pattern appears to be unrelated to the commencement of new graduates and potentially results from seasonal variation in the case mix of patients or the health of the medical workforce that alters health care performance. These mechanisms will need to be elucidated with further research.  相似文献

Carotenoids are widely distributed natural pigments which are in an increasing demand by the market, due to their applications in the human food, animal feed, cosmetics, and pharmaceutical industries. Although more than 600 carotenoids have been identified in nature, only a few are industrially important (β-carotene, astaxanthin, lutein or lycopene). To date chemical processes manufacture most of the carotenoid production, but the interest for carotenoids of biological origin is growing since there is an increased public concern over the safety of artificial food colorants. Although much interest and effort has been devoted to the use of biological sources for industrially important carotenoids, only the production of biological β-carotene and astaxanthin has been reported. Among fungi, several Mucorales strains, particularlyBlakeslea trispora, have been used to develop fermentation process for the production of β-carotene on almost competitive cost-price levels. Similarly, the basidiomycetous yeastXanthophyllomyces dendrorhous (the perfect state ofPhaffia rhodozyma), has been proposed as a promising source of astaxanthin. This paper focuses on recent findings on the fungal pathways for carotenoid production, especially the structure and function of the genes involved in the biosynthesis of carotenoids in the Mucorales. An outlook of the possibilities of an increased industrial production of carotenoids, based on metabolic engineering of fungi for carotenoid content and composition, is also discussed.  相似文献

Abstract

When migrant status and citizenship are defined by means of state categories, the language of inclusion and exclusion is key to an understanding of their contemporary shape and historical transformation. This introductory article provides an overview of some of the most relevant concepts in the discourse analysis of in- and exclusion, specifically with a view to the functioning of nation-state categories. It discusses forms of discursive problematization, such as defining, claiming, legitimizing, expanding, sensationalization and suggestion, and it connects these to the discursive drawing of boundaries discussed by the authors contributing to this issue. They focus on discursive constructions of ‘illegality’, race, class, gender, immigrant integration and transnationalism. We argue that, as state categorizations continuously differ, both the historical analysis of their genesis, functioning and transformation and the contemporary analysis of their effectuation in practices are crucial to an understanding of in- and exclusion.  相似文献

This study examined the potential for "cryoimmunology" to increase the destruction of the Dunning AT-1 prostate tumor after cryosurgery. Two possible mechanisms explaining the cryoimmunologic response were studied. The first was that an antitumor antibody is produced after cryosurgery. The second was that freezing induces an immunostimulatory signal that creates a T-cell response to the tumor. Six groups of animals (three experimental groups and three control groups) were treated once per week for 4 weeks with different therapies designed to investigate these mechanisms. Three types of immune response were measured: (1) the anti-AT-1 tumor immune titer (Ab response) by serum ELISA, (2) the effect on secondary tumor growth after challenge with live AT-1 cells (size and weight of the secondary tumor over time), and (3) the nature of the immunologic infiltrate into the secondary tumors by immunoperoxidase stain. ELISA showed that immune titers were present in the experimental groups after therapy, but the presence of an immune titer did not have a significant effect on tumor propagation. Histology showed the immunologic infiltrate was similar in all groups. These results showed that an immune response to AT-1 tumor was measurable by serum antibody, but it did not significantly limit secondary tumor growth or affect tumor histology. This suggests that the growth of AT-1 tumors is not inhibited by a cryoimmunological response. Thus, the effect of in vivo cryosurgery in the AT-1 tumor system would likely be limited to cellular and vascular changes.  相似文献

The laws of thermodynamics are employed as an analytical framework within which results about society's metabolism may be rigorously deduced in energetic and material terms. We demonstrate that the occurrence of waste is an unavoidable necessity in the industrial production of desired goods. Although waste is thus an essential qualitative element of industrial production, the quantitative extent to which waste occurs may vary within certain limits according to the degree of thermodynamic (in) efficiency with which these processes are operated. We discuss the question of which proportion of the amount of waste currently generated is due to thermodynamic necessity and which proportion is due to thermodynamic inefficiency.  相似文献

In order to study the relationships between the claustrum and motor cortex neurons both of area 4 and area 6 in the cat, an experiment was performed in which the effect of claustral stimulation on pyramidal tract neurons was investigated. Single shock electrical activation of the claustrum caused an inhibition of the spontaneous activity of the neurons, or an inhibition preceded by short activation, or no effect. These evidences lead to the conclusion that the claustrum exerts a role in movement organization.  相似文献

The Hodgkin-Huxley (HH) model is the basis for numerous neural models. There are two negative feedback processes in the HH model that regulate rhythmic spiking. The first is an outward current with an activation variable n that has an opposite influence to the excitatory inward current and therefore provides subtractive negative feedback. The other is the inactivation of an inward current with an inactivation variable h that reduces the amount of positive feedback and therefore provides divisive feedback. Rhythmic spiking can be obtained with either negative feedback process, so we ask what is gained by having two feedback processes. We also ask how the different negative feedback processes contribute to spiking. We show that having two negative feedback processes makes the HH model more robust to changes in applied currents and conductance densities than models that possess only one negative feedback variable. We also show that the contributions made by the subtractive and divisive feedback variables are not static, but depend on time scales and conductance values. In particular, they contribute differently to the dynamics in Type I versus Type II neurons.  相似文献

A major problem in tissue engineering is the availability of a sufficient number of cells with the appropriate phenotype for delivery to damaged or diseased cartilage and bone; the challenge is to amplify cell numbers and maintain the appropriate phenotype for tissue repair and restoration of function. The microcarrier bioreactor culture system offers an attractive method for cell amplification and enhancement of phenotype expression. Besides serving as substrates for the propagation of anchorage-dependent cells, microcarriers can also be used to deliver the expanded undifferentiated or differentiated cells to the site of the defect. The present article provides an overview of the microcarrier culture system, its utility as an in vitro research tool and its potential applications in tissue engineering, particularly in the repair of cartilage and bone.  相似文献

Organ complexes of bats (Nyctalis noctula), consisting of oesophagus, stomach and initial part of the small intestine have been subjected to histological and morphometrical investigation. As demonstrates the histological investigation of the preparations, the denticulate line of the mucous membrane of the oesophageal-gastric passage is displaced towards the stomach. In the cardia a well manifested internal sphincter has been revealed. In the place, where the stomach passes into the small intestine, on a small area of the submucosal tela of the intestine, there is an accumulation of secretory parts of the duodenal glands. They form a glandular torulus, which narrows the transitional zone lumen. A peculiarity of the bat pylorus is absence of an anatomical sphincter in it.  相似文献

To date humans, chimpanzees, and orangutans are the only species which have been shown capable of recognizing themselves in mirrors. Several species of macaques have now been provided with years of continuous exposure to mirrors, but they still persist in reacting to their reflection as if they were seeing other monkeys. Even gibbons (apes) and gorillas (great apes) seem incapable of learning that their behavior is the source of the behavior depicted in the image. Most primates, therefore, appear to lack a cognitive category for processing mirrored information about themselves. The implications of these data for traditional views of consciousness are considered briefly, and a recent attempt to develop an operant analog to self-recognition is critically evaluated. Finally, an attempt is made to show that self-awareness, consciousness, and mind are not mutually exclusive cognitive categories and that the emergence of self-awareness may be equivalent to the emergence of mind. Several indices of “mind” which can be applied to nonhuman species are discussed in the context of an attempt to develop a comparative psychology of mind.  相似文献

TonB protein appears to couple the electrochemical potential of the cytoplasmic membrane to active transport across the essentially unenergized outer membrane of gram-negative bacteria. ExbB protein has been identified as an auxiliary protein in this process. In this paper we show that ExbD protein, encoded by an adjacent gene in the exb cluster at 65', was also required for TonB-dependent energy transduction and, like ExbB, was required for the stability of TonB. The phenotypes of exbB exbD+ strains were essentially indistinguishable from the phenotypes of exbB+ exbD strains. Mutations in either gene resulted in the degradation of TonB protein and in decreased, but not entirely absent, sensitivities to colicins B and Ia and to bacteriophage phi 80. Evidence that the absence of ExbB or ExbD differentially affected the half-lives of newly synthesized and steady-state TonB was obtained. In the absence of ExbB or ExbD, newly synthesized TonB was degraded with a half-life of 5 to 10 min, while the half-life of TonB under steady-state conditions was significantly longer, approximately 30 min. These results were consistent with the idea that ExbB and ExbD play roles in the assembly of TonB into an energy-transducing complex. While interaction between TonB and ExbD was suggested by the effect of ExbD on TonB stability, interaction of ExbD with TonB was detected by neither in vivo cross-linking assays nor genetic tests for competition. Assays of a chromosomally encoded exbD::phoA fusion showed that exbB and exbD were transcribed as an operon, such that ExbD-PhoA levels in an exbB::Tn10 strain were reduced to 4% of the levels observed in an exbB+ strain under iron-limiting conditions. Residual ExbD-PhoA expression in an exbB::Tn10 strain was not iron regulated and may have originated from within the Tn10 element in exbB.  相似文献

THERE is overwhelming evidence to connect cyclic 3′,5′-adenos-ine monophosphate (cyclic AMP) with hormone action in various tissues1. For example, the toad bladder responds to neurohypophyseal hormones (ADH) with an increase in water permeability2 and with an increase in the mucosal to serosal transport of sodium3. ADH also causes an increase in cyclic AMP levels in the tissue4 and the actions of the hormone can be mimicked by addition of the cyclic nucleotide2,5. Orloff and Handler5 have suggested that cyclic AMP directly affects the permeability of the mucosal face of the epithelial cells to water and to sodium and that there are two separate adenyl cyclase systems responsible for controlling the permeability to water and to sodium6,7.  相似文献
