Feeding and oviposition preferences of Monochamus galloprovincialis (Olivier) (Coleoptera: Cerambycidae: Monochamini), the vector of the pine wilt nematode (PWN), Bursaphelenchus xylophilus (Steiner and Bührer) Nickle (Nematoda: Aphelenchoididae), in Portugal, was studied in three laboratory experiments: an adult feeding preference trial between branches of five pines, a no‐choice oviposition experiment on seven conifer bolts, and an oviposition choice trial between maritime pine, Pinus pinaster Aiton, and four other pines, P. pinea L., P. sylvestris L., P. halepensis Miller, or P. radiata D. Don. Scots pine (P. sylvestris) was the pine with the largest bark feeding area, while P. radiata was the least chosen to feed upon. Female M. galloprovincialis laid eggs on P. sylvestris, P. halepensis, P. pinaster, P. radiata, P. pinea, and Pseudotsuga menziesii (Mirbel) Franco, but larvae successfully completed development only on the first four pines. Beetles emerging from maritime pine bolts were slightly bigger than the others. No preference for oviposition was detected when beetles were given the choice between P. pinaster and P. sylvestris or P. halepensis. Inversely, greater oviposition occurred on P. pinaster when compared with P. pinea and P. radiata. Although PWN currently affects only P. pinaster in Portugal, the results on host suitability for M. galloprovincialis suggest that P. sylvestris and P. halepensis are adequate hosts for the insect's feeding and oviposition, and thus may also be PWN hosts in Europe. The systematically high feeding and oviposition attraction for P. sylvestris exhibited by the beetles means that the eventual spread of the PWN to areas where both M. galloprovincialis and P. sylvestris occur may have a significant impact on this widespread and economically important pine, considered to be highly susceptible to B. xylophilus.  相似文献

Heteroblastic development in pine seedlings includes extreme morphological changes with still unclear adaptive and evolutionary significance. In particular, Mediterranean and Eurasian pines (section Pinus) living in the Mediterranean basin seem to follow quite distinct developmental trajectories at the seedling stage. Aiming to confirm this ontogenetic differentiation we performed a nursery experiment with seedlings of five Mediterranean pines (Pinus pinaster, P. brutia, P. halepensis, P. pinea and P. canariensis) and three Eurasian pines (P. sylvestris, P. uncinata, and P. nigra subsp. salzmannii), also including P. radiata as an outgroup. After destructive analyses at two harvest times (9 and 32 weeks), we found sharp differentiation between Mediterranean and Eurasian pines in a combination of traits linked to shoot heteroblasty. In particular, Mediterranean pines showed a marked delay in the proportion of adult needles to total needles in the shoot compared to Eurasian species, especially at the second harvest. However, two Mediterranean pines, P. pinaster and P. brutia showed a slightly higher proportion of secondary needles, and a higher rate of budset at a more advanced stage (68 weeks) compared to the other three Mediterranean species. Meaningfully, the outgroup P. radiata was the only species combining a high proportion of adult foliage since the first harvest with a delayed formation of the first terminal bud. We discussed the adaptive significance of these findings at the light of species’ climatic niches and life histories.  相似文献

Essential oils (EOs) from Schinus molle, Helichrysum gymnocephalum, Cedrelopsis grevei and Melaleuca viridiflora, four aromatic and medicinal plants, are commonly used in folk medicine. EOs were characterized by gas chromatography/mass spectrometry (GC/MS) and quantified by gas chromatography‐flame ionization detection (GC‐FID); then evaluated for their behavioral effects on adults of the green pea aphid Acyrthosiphon pisum (Harris ) using a Perspex four‐armed olfactometer in order to test the compatibility of their use as phytoinsecticides to control this insect pest. Our results showed that the EOs from the leaves of Smolle, Mviridiflora and Cgrevei did not change aphids’ behavior. However, Smolle fruits EO seemed to be attractive while Hgymnocephalum leaves EO exhibited repellency towards aphids at a dose of 10 μl. The major compounds in Smolle fruits EO were 6‐epi‐shyobunol (16.22%) and d ‐limonene (15.35%). While, in Hgymnocephalum leaves EO, 1,8‐cineole was the main compound (47.4%). The difference in aphids’ responses to these two EOs could be attributed to the differences in their compositions. The pine shoot beetle Tomicus destruens (Wollaston) (Coleoptera: Curculionidae: Scolytinae) is one of the main pests of Mediterranean forests, where it is oligophagous on Mediterranean pines. However, possible global warming may make the insect move to higher latitudes and altitudes, allowing it to attack new pine species. In this respect, the aim of the present article was to assess both the acceptance and performance of T. destruens offered host and non‐host pine species. A no‐choice breeding experiment was set up under laboratory conditions, using logs of three Mediterranean (Pinus pinea L., Pinus pinaster Miller, and Pinus halepensis Aiton) and two continental (Pinus nigra Arnold and Pinus sylvestris L.) pine species. Log debarking at the end of adult emergence assessed parent fecundity, egg, and larval mortality. The quality of callow adults emerging from each tested pine was evaluated on the basis of their longevity on a semiartificial diet. Tomicus destruens colonised all tested pine species, but did not reproduce in Scots pine, taking about 79 days to complete development with no differences among pines tested. The best breeding performance, evaluated as female fecundity and adult production, was observed in P. halepensis, and the lowest in P. pinaster. On average, adults emerging from P. pinea survived longer (83 days) than from other pines, and adult longevity was the lowest in males emerging from P. nigra. Austrian pine, which under natural conditions is usually not a host of T. destruens, allowed insect development and adult production similar to P. pinea and P. pinaster.  相似文献

The hydraulic properties of Pinus pinea, Pinus halepensis and Tetraclinis articulata were studied in a coastal dune area from Eastern Spain. The measured variables include vulnerability to xylem embolism (vulnerability curves), hydraulic conductivity and carbon isotopic discrimination in leaves. Leaf water potentials were also monitored in the three studied populations during an extremely dry period. Our results showed that roots had always wider vessels and higher hydraulic conductivity than branches. Roots were also more vulnerable to xylem embolism and operated closer to their hydraulic limit (i.e., with narrower safety margins). Although it was not quantified, extensive root mortality was observed in the two pines during the study period, in agreement with the high values of xylem embolism (> 75%) predicted from vulnerability curves and the water potentials measured in the field. T. articulata was much more resistant to embolism than P. pinea and P. halepensis. Since T. articulata experienced also lower water potentials, safety margins from hydraulic failure were only slightly wider in this species than in the pines. Combining species and tissues, high resistance to xylem embolism was associated with low hydraulic conductivity and with high wood density. Both relationships imply a cost of having a resistant xylem. The study outlined very different water-use strategies for T. articulata and the pines. Whereas T. articulata had a conservative strategy that relied on the low vulnerability of its conducting system to drought-induced xylem embolism, the two pines showed regulatory mechanisms at different levels (i.e., embolism, root demography) that constrained the absorption of water when it became scarce. Esteya vermicola (Ophiostomataceae) is the first reported endoparasitic fungus of the pinewood nematode (PWN), Bursaphelenchus xylophilus (Nematoda: Aphelenchoidoidea). It has high in vitro infectivity. In this study, the nematocidal effect of E. vermicola in logs was investigated and evaluated. Two months after inoculation of pine wilt-killed Pinus densiflora logs with E. vermicola conidia suspensions of 3 × 108 and 3 × 106 ml−1, the density of nematodes decreased by approximately 79% and 47%, respectively. When the fungus was sprayed on to four-year-old pine seedlings one month before PWN inoculation, the survival index of seedlings reached 0.67 compared with only 0.067 for control seedlings without fungal spraying. These results suggest that conidia spraying of Evermicola can, to some extent, protect pine trees from wilt disease. Moreover, infected nematodes and hyphae of Evermicola were observed in the treated wood sections.  相似文献

 Although Pinus pinea L. is an important forest species in the Mediterranean region, few reports exist on its ectomycorrhizal associates. Sixty isolates, obtained from fungal sporocarps collected in mixed forests of P. pinea in Catalonia (northeastern Spain), were tested for ectomycorrhiza formation on containerized P. pinea seedlings when applied as mycelial inoculum produced in peat-vermiculite. A total of 17 isolates, in 8 genera (Amanita, Hebeloma, Laccaria, Lactarius, Pisolithus, Rhizopogon, Scleroderma and Suillus), formed ectomycorrhizas and the percentages of mycorrhizal short roots varied among isolates and species from 13% to 89%. Some of these fungi are cited for the first time in association with P. pinea. The results indicate further fungal candidates for controlled inoculation of P. pinea seedlings in the nursery. This work provides a tool whereby the needle remains of native, south‐western European Pinus spp. can be easily identified from species‐specific epidermal features. To construct this tool, the needles of P. uncinata, P. sylvestris, P. nigra, P. pinaster, P. pinea and P. halepensis were gathered across the Northern Hemisphere range of each taxon and compared with non‐indigenous trees growing in two South Australian Botanic Gardens. Three needles from each of these species were taken from three adult trees growing at three different localities. Light microscopy was used to observe the key epidermal and stomatal features of the needles. To improve interpretation, additional scanning electron microscopy samples were prepared. Epidermal features, including variation in the diameter of the epistomatal chamber aperture (pore), are described. A taxonomic key based on the size, shape and arrangement of the subsidiary cells of the stomatal complexes was constructed. This key enables the identification of pine needle fragments at the species level (except those belonging to the group P. gr. nigrauncinata). Despite their overlapping range, pore size was helpful in distinguishing between P. nigra and P. uncinata and between three groups of species. Isolated stomata were also observed. Cluster and discriminant analyses of stomatal variables described in earlier studies were performed. Overlap in guard cell variables hampers species‐level identification of isolated stomata. Microsatellite transfer across coniferous species is a valued methodology because de novo development for each species is costly and there are many species with only a limited commodity value. Cross-species amplification of orthologous microsatellite regions provides valuable information on mutational and evolutionary processes affecting these loci. We tested 19 nuclear microsatellite markers from Pinus taeda L. (subsection Australes) and three from P. sylvestris L. (subsection Pinus) on seven Eurasian hard pine species (P. uncinata Ram., P. sylvestris L., P. nigra Arn., P. pinaster Ait., P. halepensis Mill., P. pinea L. and P. canariensis Sm.). Transfer rates to species in subsection Pinus (36–59%) were slightly higher than those to subsections Pineae and Pinaster (32–45%). Half of the trans-specific microsatellites were found to be polymorphic over evolutionary times of approximately 100 million years (ten million generations). Sequencing of three trans-specific microsatellites showed conserved repeat and flanking regions. Both a decrease in the number of perfect repeats in the non-focal species and a polarity for mutation, the latter defined as a higher substitution rate in the flanking sequence regions close to the repeat motifs, were observed in the trans-specific microsatellites. The transfer of microsatellites among hard pine species proved to be useful for obtaining highly polymorphic markers in a wide range of species, thereby providing new tools for population and quantitative genetic studies.Communicated by D.B. Seeds ofPinus halepensis andPinus pinaster subsp.pinaster were subjected to four heat treatments (90, 110, 150 and 200 °C) for 1 and 5 min to investigate their tolerances to high temperatures resulting from fires. Another group of seeds of each species were not heated and considered as a control. Information on responses of seeds to high temperatures would help to determine whether or not the species are pyrophytes. Heating for 1 min at all temperatures did not affect the average germination ofPinus halepensis seeds whereas that ofP. pinaster subsp.pinaster seeds was decreased significantly when they were heated to 200 ° C. HoweverP. pinaster seeds were more resistant than that ofP. halepensis when heat treatment was 150 °C for 5 min. Data obtained also indicated that, in general,P. halepensis andP. pinaster subsp.pinaster seed germination is not favoured by a temperature increase than can be reached in soils during a fire. Thus, the species are not pyrophytes but colonizers of burnt sites.  相似文献

Chemical analysis, antimicrobial activity and cytotoxic effects of essential oils (EOs) from leaves of Piper aduncum var. ossanum from two localities Bauta (EO‐B) and Ceiba (EO‐C), Artemisa Province, Cuba, were determined. EOs were obtained by hydrodistillation and analyzed by gas chromatography/mass spectrometry. EO‐B demonstrated higher activity against Saureus and Lamazonensis; while a lower cytotoxicity on mammalian cells was observed. Both EOs displayed the same activity against Plasmodium falciparum, Trypanosoma cruzi, Trypanosoma brucei, and Leishmania infantum. Both EOs were inactive against Escherichia coli and Candida albicans.  相似文献

Sphaeropsis shoot blight, caused by Diplodia pinea and Diplodia scrobiculata, damage conifers throughout the world. In France, the first disease outbreaks were reported during the 1990s. The factors associated with the pathogen presence in stands and the relationship between pathogen and disease distributions were analysed in order to understand the Sphaeropsis emergence. Eighty‐two stands of Pinus nigra, Pinus sylvestris, Pinus pinaster and Pinus radiata were visited. Cones were collected on the ground to assess the pathogen frequency. Diplodia spp were isolated and determined by a species‐specific PCR test. The role of potential explaining factors of D. pinea prevalence on cones was analysed by logistic regression. D. pinea was the dominant species in visited stands. The main factors influencing the pathogen presence selected in the models were host species (the pathogen being less frequent on P. pinaster than on P. nigra and P. sylvestris cones), winter temperature and summer rain, which were both positively correlated with cone colonization. The climate became more favourable to D. pinea presence within the last 15 years compared with the previous 30‐year period. By contrast, future climatic changes over the next 40 years should have far less impact on the pathogen presence.  相似文献

Aerial parts of Artemisia arborescens were collected from different sites of the Mediterranean area (southwestern Algeria and southern Italy) and the chemical composition of their essential oil (EO) extracted by hydrodistillation was studied by both gas chromatography (GC) equipped with an enantioselective capillary column and GC/mass spectrometry (GC/MS). The EOs obtained were tested against several Listeria monocytogenes strains. Using GC and GC/MS, 41 compounds were identified, accounting for 96.0 – 98.8% of the total EO. All EOs showed a similar terpene profile, which was rich in chamazulene, β‐thujone, and camphor. However, the concentration of such compounds varied among the EOs. Aarborescens EO inhibited up to 83.3% of the Lmonocytogenes strains, but the inhibitory spectrum varied among the EOs, with those from Algeria showing a higher inhibition degree than the Italian EOs. Such effect likely depended on the ketone (β‐thujone + camphor) content of the EO. The differences in the EO composition support the hypothesis that Aarborescens has at least two different chemotypes: a β‐thujone and a chamazulene type. The EO inhibitory spectrum indicates the Aarborescens EO as a valuable option in the control of the food‐borne pathogens.  相似文献

This study examined seasonal variations in the content of water-soluble phenolics extracted from litter and Ah horizon of forest soil under Pinus laricio Poiret trees. After having identified and quantified different phenolics, the effects of seasonal phenolic extracts, single phenolic acids and synthetic phenol mixtures on seed germination of Pinus laricio Poiret, Pinus pinaster Aiton and Pinus halepensis Mill. were evaluated, and the activity of phenolics on the main enzymes involved in the glyoxylate cycle was tested. The results indicated a seasonal variation of phenol content in soil, with largest concentrations of water-soluble phenolic acids in autumn and smallest concentrations in summer. Addition of phenol extracts to germination medium reduced seed germination showing phytotoxic effects which differed, depending on the species and the fractions tested. Phenols extracted from litter and Ah horizon in autumn and winter, vanillic acid, and synthetic mixtures reproducing phenols extracted from soil in winter inhibited seed germination of Pinus laricio, pinaster and halepensis more than other treatments. A greater inhibitory effect was observed on seed germination of Pinus pinaster and halepensis compared to Pinus laricio. Several studies have reported glacial refugia and migration pathways for different pine species in the Iberian Peninsula, all of them based on a single‐species approach. In this paper, chloroplast microsatellites (cpSSRs) are used to compare population genetic structure and diversity estimates for interspecific pairs of populations located along a cline from southwestern (latitude 36°32′ N, longitude 5°17′ W) to northeastern Spain (latitude 42°14′ N, longitude 2°47′ E) in two widely distributed Mediterranean pines, Pinus halepensis Mill. and Pinus pinaster Ait. Some cpSSRs were shared between species, facilitating comparison of levels of gene diversity at the species level and inferences about within and among species differentiation. P. pinaster showed a much higher number of variants (29) and haplotypes (69) than P. halepensis (20 and 21, respectively). Moreover, genetic diversity estimates for interspecific pairs of populations along the cline were negatively correlated. Three main causes may explain the differences between species in the present‐day distribution of genetic diversity: (1) the distribution of genetic variability before the Quaternary glaciations, with an earlier presence of P. pinaster in the Iberian Peninsula and a late spread of P. halepensis from eastern and central Europe, (2) the location of the Holocene glacial refugia and the migration pathways from these refugia to the present‐day range (from northeast to southwest in P. halepensis and from southwest to northeast and northwest in P. pinaster) and (3) the interactions between species during the postglacial spread.  相似文献

Pinus sylvestris L., Abies alba Mill. and Fagus sylvatica L.—the significant forest forming tree species in Europe are important for palaeoecological interpretations based on the results of pollen analysis of fossil deposits. The potential pollen loading for Pinus sylvestris, Abies alba and Fagus sylvatica was modelled using simulated and actual vegetation maps, measured fall-speed values and pollen productivity estimates from the literature. The influx of fir pollen drops sharply with distance from the pollen source due to the high fall speed and moderate pollen productivity. The vast majority of Abies alba pollen is deposited within less than 50 m of the sampling site and a major proportion within 100 m. For beech the corresponding numbers would be 300 and 1,800 m, and for pine 1,000 and 4,500 m. The observed mean pollen accumulation rate (PAR) values for Pinus and Fagus were ca. 5,800 and 1,100 grains cm−2 year−1, respectively. In the case of Abies, the mean annual PAR for the whole region is ca. 700 grains cm−2 year−1. In SE Poland the regional signal is represented by PARs of Abies alba <200 grains cm−2 year−1 and of Fagus sylvatica <500 grains cm−2 year−1. The local presence/absence threshold values for Abies alba, Fagus sylvatica and Pinus sylvestris are >1,000 grains cm−2 year−1, >2,000 grains cm−2 year−1 and >3,500 grains cm−2 year−1 respectively.  相似文献

To initiate somatic embryogenesis in Pinus sylvestris and Pinus pinaster, immature seeds were collected from June to August and the developmental stage of the zygotic embryos was determined. Four developmental stages were distinguished and the response of the zygotic embryos at each of the four developmental stages was compared intra- and inter-species. For this study, modified Litvay's medium (LM), with or without growth regulators, was chosen. Somatic embryogenesis was initiated and maintained on both media but the two species displayed different propensities. In P. sylvestris, the highest initiation frequency was obtained with intact megagametophytes containing embryos at the four-cell stage to the stage of cleavage polyembryony (up to 22 and 9%, respectively). The culture medium had no significant effect on the initiation and proliferation of embryogenic cultures. In P. pinaster, however, the best response occurred from excised zygotic embryos at the stage prior to elongation of cotyledon primordia (up to 40% explants responded), on medium with growth regulators. Another characteristic distinguishing the two species in culture was that in some embryogenic cell lines of P. sylvestris, somatic embryos matured spontaneously when initiated and maintained on medium without growth regulators. Some of these embryos developed into plantlets on the same medium at the frequency of 40%. Therefore, in P. sylvestris all the stages of somatic embryogenesis were achieved on the medium without growth regulators. However, in both species, maturation of a large number of somatic embryos was greatly improved on medium containing high concentration of gellan gum (Gelrite 10 g l?1) and abscisic acid (60 μM). Cotyledonary somatic embryos subsequently germinated (72 and 80% for P. sylvestris and P. pinaster, respectively) and developed into plantlets (48 and 29%, for P. sylvestris and P. pinaster, respectively). The diamondback moth (DBM), Plutella xylostella (L.), is a globally destructive pest of cruciferous vegetables. Excessive use of synthetic pesticides to control this species results in negative effects on the environment, human health, and nontargeted organisms. The essential oils (EOs) derived from plants may be developed as effective alternatives to conventional pesticides. In this study, thirteen EOs were extracted by hydrodistillation, respectively. Their chemical compositions were identified by gas chromatography-mass spectrometry (GC–MS). Furthermore, the biological activities of EOs such as toxicity, antifeedant activity, and growth inhibition effect, toward DBM larvae were investigated. Against DBM second-instar larvae, the most toxic EO was Pelargonium graveolens (LC50 = 0.36 μg/μl) after 72 hr of exposure, followed by Polygonum hydropiper (LC50 = 0.53 μg/μl). The Ocimum basilicum EO exhibited the highest antifeedant effect to third-instar larvae at at all set concentrations. At 15 μg/μl, the EOs of Acorus calamus, O. basilicum, and P. graveolens completely inhibited the feeding activities of larvae (100%). The Ruta graveolens EO showed the lowest relative growth rate and the highest growth inhibition rate towards third-instar larvae at diverse concentrations. And the EOs of P. hydropiper, A. calamus, and O. basilicum showed promising growth inhibition activities. Overall, the five EOs (P. graveolens, O. basilicum, R. graveolens, P. hydropiper, and A. calamus) showed moderate to high bioactivity, whereas eight EOs were found to be less active against DBM larvae. These results indicate that the five tested EOs are promising to be developed as novel botanical insecticides to control DBM population.  相似文献

This study aimed to characterize the chemical composition of Aloysia polystachia, Acantholippia seriphioides, Schinus molle, Solidago chilensis, Lippia turbinata, Minthostachys mollis, Buddleja globosa, and Baccharis latifolia essential oils (EOs), and to evaluate their antibacterial activities and their capacity to provoke membrane disruption in Paenibacillus larvae, the bacteria that causes the American Foulbrood (AFB) disease on honey bee larvae. The relationship between the composition of the EOs and these activities on Plarvae was also analyzed. Monoterpenes were the most abundant compounds in all EOs. All EOs showed antimicrobial activity against Plarvae and disrupted the cell wall and cytoplasmic membrane of Plarvae provoking the leakage of cytoplasmic constituents (with the exception of Blatifolia EO). While, the EOs’ antimicrobial activity was correlated most strongly to the content of pulegone, carvone, (Z)‐β‐ocimene, δ‐cadinene, camphene, terpinen‐4‐ol, elemol, β‐pinene, β‐elemene, γ‐cadinene, α‐terpineol, and bornyl acetate; the volatiles that better explained the membrane disruption were carvone, limonene, cis‐carvone oxide, pentadecane, trans‐carvyl acetate, trans‐carvone oxide, trans‐limonene oxide, artemisia ketone, trans‐carveol, thymol, and γ‐terpinene (positively correlated) and biciclogermacrene, δ‐2‐carene, verbenol, α‐pinene, and α‐thujene (negatively correlated). The studied EOs are proposed as natural alternative means of control for the AFB disease.  相似文献
