The morphology, morphogenesis and molecular phylogeny of the oxytrichid ciliate, Monomicrocaryon euglenivorum euglenivorum (Kahl, 1932) Foissner, 2016, isolated from freshwater in a seaside park, Guangzhou, China, were investigated. Monomicrocaryon euglenivorum euglenivorum can be recognized as follows: caudal cirri in midline of body; dorsal kinety 1 without a one-kinetid-wide gap; transverse cirri acicular or rod-shaped with a fringed distal end; right marginal row commences at level of buccal vertex or anterior to buccal vertex. The main events during binary fission are as follows: (1) the proter retains the parental adoral zone of membranelles entirely; (2) frontoventral-transverse cirral anlagen I–VI are segmented in the ordinary pattern 1:3:3:3:4:4 from left to right, which form three frontal, four frontoventral, one buccal, three postoral ventral, two pretransverse ventral and five transverse cirri, respectively; (3) dorsal morphogenesis is in the typical Oxytricha-pattern, but fragmentation of dorsal kinety 3 is indistinct; and (4) three caudal cirri are formed, one at the posterior end of each of dorsal kineties 1, 2 and 4. Phylogenetic analyses based on SSU rDNA sequences showed that M. euglenivorum euglenivorum clustered with Kleinstyla dorsicirrata and Heterourosomoida lanceolata rather than with its congener M. elegans. The genus Monomicrocaryon is not monophyletic in this study; however, its monophyly is not rejected by the AU test.  相似文献

The morphology, morphogenesis and molecular phylogeny of a hypotrichous ciliate, Lamtostyla granulifera sinensis subsp. nov., isolated from northern China, were investigated. This population appeared highly similar in morphology to L. granulifera Foissner, 1997. However, on detailed investigation some non-overlapping features were identified, i.e., the body shape and the arrangement of the cortical granules. These differences suggested the separation at subspecies level. Furthermore, the morphogenesis of the new subspecies is described, which is characterized by: (1) the posterior part of the parental adoral zone of membranelles is renewed; (2) the amphisiellid median cirral row is formed from two anlagen; and (3) the frontoventral-transverse cirral anlagen II to VI generate one transverse cirrus each. Phylogenetic analyses based on SSU rDNA sequence data show that Lamtostyla species are scattered in different clades. The monophyly of the genus Lamtostyla is also rejected by the AU test in this study.  相似文献

In this study, we investigated the morphology and molecular phylogeny of four marine or brackish spirotrichean ciliates found in China, namely: Caryotricha sinica sp. nov., Prodiscocephalus orientalis sp. nov., P. cf. borrori, and Certesia quadrinucleata. Caryotricha sinica is characterized by its small size, seven cirral rows extending posteriorly to about 65% of the cell length, and four transverse cirri. Prodiscocephalus orientalis differs from its congeners mainly by the number of cirri in the “head” region and on the ventral side. The SSU rDNA sequence of P. cf. borrori differs from that of other population of P. borrori by ca. 40 bp. Consequently, the nominal species P. borrori is considered to be a species-complex. New data are provided for Certesia quadrinucleata. The Chinese population of C. quadrinucleata, for example, has fewer left marginal cirri than the other populations for which such data are available. Phylogenetic analyses based on SSU rDNA sequence data show that the genus Caryotricha is monophyletic. All typical “discocephalids” with a discoid “head” form a strongly supported clade that is sister to the unstable uronychiids + pseudoamphisiellids clade within the Euplotia. The genus Certesia forms a sister group to the Euplotes clade, also within the Euplotia assemblage.  相似文献

The morphology and molecular phylogeny of a new metopid ciliate, Urostomides spinosus nov. spec., discovered in a freshwater ditch in Qingdao, China, were investigated using live observation, morphometry and protargol staining as well as molecular phylogenetic methods. Diagnostic features of the new species include a broadly obpyriform body carrying three posterior spines, eight somatic kineties, five preoral dome kineties with specialized row 3, adoral zone composed of about 28 membranelles, making a 270° turn around body axis. Phylogenetic analyses of the SSU rDNA sequence revealed that the genus Urostomides is monophyletic, but its interspecific relationships remained unresolved. Moreover, a closer relationship of the new species with the morphologically similar Urostomides campanula was not supported by the molecular data.  相似文献

The morphology and morphogenesis of a new saline soil hypotrichous ciliate, Urosoma quadrinucleatum n. sp., collected from northwestern China, were studied based on live observations and protargol stained specimens. The new species is characterized as follows: size in vivo 90–130 × 20–30 μm; body outline elongate-elliptical with both ends broadly rounded; four macronuclear nodules; cortical granules present; paroral in front of endoral; usually 16 frontal-ventral-transverse cirri. Urosoma quadrinucleatum n. sp. has almost the same morphogenetic characteristics as its congeners U. gigantea and U. salmastra, and differs from other three congeners whose morphogenesis is known in the formation of the frontal-ventral-transverse cirral anlagen as well as the development of marginal and dorsal kineties anlagen. The sequence differences among U. quadrinucleatum n. sp. and other Urosoma species further support the validity of the present organism as novel species. Further, U. quadrinucleatum n. sp. clusters with U. salmastra in the phylogenetic analyses based on small subunit ribosomal RNA gene (SSU rDNA) sequence data.  相似文献

The living morphology, infraciliature, morphogenesis and phylogenetic position of a new hypotrichous ciliate, Heterourosomoida sinica n. sp., discovered in saline soil in Northwest of China, were investigated. Heterourosomoida sinica n. sp. is characterized as follows: body size about 70–95 × 20–30 μm in vivo, ellipsoid or elongate; cortical granules dark green, 0.5 μm across, arranged around dorsal cilia and in short irregular rows on ventral side; consistently 18 frontoventral-transverse cirri; left and right marginal rows composed of 17–24 and 13–21 cirri, respectively; two macronuclear nodules and one or two micronuclei. The detailed morphogenesis of a species of Heterourosomoida is reported for the first time. The ontogenesis on dorsal side is basically similar to that in Urosomoida-species and the process on ventral side is characterized by: 18 frontoventral-transverse cirri develop from six anlagen with the anlagen V and VI formed in the primary mode. Phylogenetic analyses based on SSU rDNA sequence data reveal that Heterourosomoida sinica n. sp. clusters with Kleinstyla dorsicirrata and forms a clade with the type species H. lanceolata.  相似文献

The morphology, morphogenesis and molecular phylogeny of a new saline soil hypotrich ciliate, Uroleptoides salina nov. spec., discovered from China, was investigated. The new species is characterized as follows: body 150–215 × 40–50 μm in vivo, slender and highly flexible; usually four ellipsoidal macronuclear nodules; contractile vacuole absent; cortical granules absent; endosymbiotic algae present; amphisiellid median cirral row consists of 14–25 cirri and terminates about 47% down length of body; usually three buccal cirri and 3–13 cirri left of anterior portion of amphisiellid median cirral row; 3–5 transverse cirri. Morphogenesis during binary fission is characterized by: (1) the parental adoral zone of membranelles is retained completely, parental paroral contributes to the formation of the undulating membranes anlage for the proter; (2) the oral primordium of the opisthe is formed apokinetally; and (3) the amphisiellid median cirral row is formed from two anlagen. Phylogenetic analyses based on SSU rDNA sequence data show that Uroleptoides salina nov. spec. has a close relationship with its morphologically similar species, U. longiseries, U. magnigranulosus, Orthamphisiella breviseries, and Parabistichella variabilis.  相似文献

The morphogenesis and molecular phylogeny of two soil hypotrich ciliates, Australocirrus australis and A. aspoecki, collected from Xi’an, Shaanxi Province, China, were investigated using protargol preparation. The main features of morphogenesis of A. australis are as follows: (1) the parental adoral zone of membranelles is retained completely in the proter; (2) parental cirri and the undulating membranes are involved in the formation of six primordial streaks; (3) multiple fragmentation of the third dorsal kinety anlage; (4) more than three dorsomarginal kineties are formed. An early divider of A. aspoecki was also found, and its morphogenetic events resemble those of other Australocirrus species. Furthermore, the SSU rDNA sequence of the Shaanxi populations of A. australis and A. aspoecki was provided. The molecular phylogeny indicates that A. aspoecki clusters together with A. rubrus and two populations of A. australis with full support, which are separated from the clade comprising the two populations of A. shii.  相似文献

A new ciliate species of the genus Loxophyllum Dujardin, 1841, Loxophyllum paludosum sp. n., is described from a mangrove wetland near Daya Bay in Guangdong Province, southern China, based on morphological and molecular analyses. The new species can be distinguished from its congeners by a combination of the following characters: (1) 12–14 right kineties and 4–6 left kineties; (2) two macronuclear nodules and one micronucleus; (3) a single contractile vacuole located terminally; (4) extrusomes bar-shaped, evenly spaced along entire ventral margin, and clustered to form 5–7 warts along dorsal margin; and (5) presence of three ridges on the left side of cell. The new species is divergent from its congeners from 0.4% to 6.7% (5–104 nucleotide sites) based on the small subunit (SSU) rRNA gene sequence data. The validity of the new species is also supported by molecular phylogenetic trees inferred from SSU rRNA gene sequences.  相似文献

The live morphology, infraciliature and morphogenesis of a new urostylid ciliate, Trichototaxis marina n. sp., collected from coastal water in Qingdao, China, were studied based on the observations of live and silver stained specimens. The new species is characterised as follows: body very flexible and contractile, slight to brick-reddish in colour due to irregularly-shaped, brick-red pigments; ca. 70 adoral membranelles; about 17 frontal cirri arranged in a bicorona; average 67 midventral pairs, the right base of each pair being conspicuously larger than the left base; five to seven transverse cirri; constantly two frontoterminal, one buccal and two pretransverse ventral cirri; two or three left marginal rows; right and innermost left marginal rows with 56–92 and 66–106 cirri, respectively; six bipolar dorsal kineties; more than 100 macronuclear nodules. The characteristic morphogenetic feature in T. marina is the development of the left marginal rows, that is, only one left marginal row is newly built the other one or two being retained from the parental cell. Phylogenetic analyses based on small subunit ribosomal gene sequence data reveal a close relationship of T. marina with members of family Pseudokeronopsidae.  相似文献

Four epibiotic sessilid peritrichs, i.e., Zoothamnium wilberti n. sp., Baikalonis microdiscus n. sp., Epistylis anastatica (Linnaeus, 1767) Ehrenberg, 1830, and Rhabdostyla commensalis Möbius, 1888, were isolated from one syllid polychaete and three crustacean hosts in Qingdao, China. For each species, specimens were observed both in vivo and following silver staining. Their SSU rDNA was also sequenced for phylogenetic analyses. Zoothamnium wilberti n. sp. is characterized by the appearance of its colony, which is up to 350 μm high, and usually has fewer than 16 zooids, and the dichotomously branched stalk with transverse wrinkles, the conspicuously conical peristomial disc, and infundibular polykinety 3 comprising three isometric ciliary rows. Baikalonis microdiscus n. sp. can be recognized by its barrel-shaped zooid, small peristomial disc, smooth and short stalk, and its unusual infundibular polykinety 3 comprising a long inner row and a short outer row. Two poorly known species, i.e., Epistylis anastatica and Rhabdostyla commensalis, are redescribed and redefined. Phylogenetic analyses reveal that: (i) R. commensalis is closely related to the family Astylozoidae rather than to the morphologically similar Epistylididae; (ii) B. microdiscus n. sp. is sister to the family Scyphidiidae; (iii) E. anastatica groups with vorticellids and ophrydiids, which further supports the polyphyly of the genus Epistylis; and (iv) Z. wilberti n. sp. is nested within the Zoothamniidae, as expected.  相似文献

This paper investigates the morphology, morphogenesis and SSU rRNA gene-based phylogeny of Apoholosticha sinica n. g., n. sp., isolated from mangrove wetland in Shenzhen, southern China. The new genus Apoholosticha is characterized by its bipartite adoral zone, clearly differentiated frontal cirri arranged in a bicorona, midventral complex composed of midventral pairs only, one marginal cirral row on each side, presence of frontoterminal and transverse cirri, and the lack of a buccal cirrus and caudal cirri. The type species, Apoholosticha sinica n. sp. is diagnosed by the elongated body shape and two kinds of cortical granules. Its main morphogenetic features are similar to that of Pseudokeronopsis except for (1) no buccal cirrus is formed and (2) its macronuclear nodules fuse into a single mass during cell division. Phylogenetic analyses for the new taxon indicate that Apoholosticha n. g. is most closely related to Nothoholosticha and Heterokeronopsis, and falls into the family Pseudokeronopsidae within the core Urostylida clade. In addition, a species that had been misidentified in previous literature is here recognized and assigned to the new genus as Apoholosticha sepetibensis (Wanick and Silva-Neto, 2004) n. comb. (basionym: Pseudokeronopsis sepetibensis Wanick and Silva-Neto, 2004).  相似文献

In a study to investigate ciliate diversity, we discovered a new soil ciliate. Holostichides terrae nov. spec. was examined and identified based on observations of living cells and stained specimens. In addition, the nuclear SSU rRNA gene along with morphology was analyzed to infer its phylogenetic position. The new species closely resembles H. dumonti, but can be distinguished by the morphology of the pharynx (with rod-shaped structure vs. lacking) and the number of frontoterminal cirri (invariably two vs. usually more than two). Molecular analyses indicate that the genus Holostichides is not monophyletic, and H. terrae is closely related with the genera Birojimia and Hemicycliostyla, both of which have a pharynx with rod-shaped structures, as also seen in H. terrae.  相似文献

Two marine urostylid ciliates, Caudiholosticha marina sp. nov. and Nothoholosticha flava sp. nov., isolated from intertidal sediment in the Yellow Sea, are investigated using morphological and small subunit rDNA phylogenetic analyses. Caudiholosticha marina is 210?310 μm × 40?55 μm in vivo, and has 10?20 macronuclear nodules, 23?37 midventral cirral pairs extending to 5?8 transverse cirri, and two caudal cirri. It differs from congeners by its marine habitat, larger size, macronuclear arrangement pattern and high number of midventral pairs. Molecular phylogenetic analyses indicate a polyphyly of Caudiholosticha. Nothoholosticha flava is yellow to brownish and 240?320 μm × 40?60 μm sized, and has a bipartite adoral zone, six frontal cirri in atypical bicorona, usually four frontoterminal, one buccal and 5?7 transverse cirri and 28?54 midventral pairs. Phylogenetic analyses allocate N. flava as sister of N. fasciola, type of the genus. The two Nothoholosticha species differ distinctly by the presence/absence of frontoterminal cirri, a feature often used to define genera in the Hypotrichia. However, the SSU rDNA sequence similarity between these two species is 99.3%, which weakens the justification for separating the new isolate at genus level. The taxonomic significance of frontoterminal cirri is discussed based on morphological and molecular data.  相似文献

In a study on ciliate diversity, we discovered the new hypotrich species, Gonostomum jangbogoensis n. sp., in freshwater from Terra Nova Bay, Victoria Land, southeast Antarctica. We describe its morphology and morphogenesis using standard methods, and the SSU rRNA gene phylogeny is provided as well. Morphology of Gonostomum jangbogoensis n. sp. is characterized as follows: slender to elongated body shape; grayish under low magnification; cortical granules present; 32–41 adoral membranelles; 3 enlarged frontal cirri; 1 buccal cirrus; 2 frontoterminal cirri; 3 or 4 frontoventral cirral pairs, 2 pretransverse cirri, 6–7 transverse cirri; 13–19 left and 18–26 right marginal cirri; 17–23 paroral kinetids; 3 dorsal kineties; 3 caudal cirri; 2 macronuclear nodules with 1–3 micronuclei. The morphogenesis of the new species confirms that it has at least seven frontal-ventral-transverse cirral anlagen, which is also reported in Gonostomum sp. 1 sensu Shin from Korea. Even though these two populations occur very far from each other, the morphometric data prove that this character state, the seven cirral anlagen, is a stable feature across these populations and might be an apomorphy. The phylogenetic analyses show that the genus Gonostomum is non-monophyletic and that the new species is a sister to G. bromelicola.  相似文献

The morphology and infraciliature of two pleurostomatid ciliates, Amphileptus aeschtae nov. spec. and Amphileptus eigneri nov. spec., collected from shrimp-culturing ponds of the Bohai Sea, China, were investigated using live observations and the protargol impregnation method. The new species A. aeschtae is identified by the presence of 200-300 macronuclear nodules and several contractile vacuoles along the ventral side of the cell. A. eigneri differs from its congeners, inter alia, by the marine habitat, the presence of many contractile vacuoles along the ventral margin and the number of somatic kineties on both sides. Loxophyllum ozakii Shigematsu, 1953 is transferred to Kentrophyllum.  相似文献

The morphology and infraciliature of two pleurostomatid ciliates, Loxophyllum choii nov. spec. and L. shini nov. spec., collected from inshore waters and a shrimp-culturing pond near Qingdao (Tsingtao), China, were investigated using live observations and the protargol-impregnation method. L. choii is distinguished from its congeners by the presence of conspicuous warts and prominent spines along the dorsal margin. L. shini is identified by a combination of characters including the small body size, the presence of two macronuclear nodules and the evenly distributed extrusomes whose tips protrude from the surface along both margins.  相似文献

A new hypotrichous ciliate, Schmidtiella ultrahalophila gen. nov., spec. nov., was isolated from a solar saltern on the island of Sal, Cape Verde. The possession of only one short dorsal kinety clearly distinguishes S. ultrahalophila from other known hypotrichous genera and species. Further diagnostic characters include: a flexible and slender body, an average size of 85 × 15 μm in vivo; a bipartite adoral zone with two hypertrophied frontal adoral membranelles and nine to twelve ventral adoral membranelles; three frontal, one parabuccal, two frontoventral, two or three postoral ventral, and two or three frontoterminal cirri; and marginal cirral rows variable in number, usually one on each side. Ontogenetic data indicate the following: the frontal‐ventral cirri originate from six or five anlagen; the proter inherits the parental adoral zone; the frontal and ventral cirri originate from five or six anlagen; and the marginal cirral rows and the dorsal kinety tend to originate intrakinetally. Additional marginal rows are rarely derived from de novo anlagen. Based on its morphology, morphogenesis and its SSU rRNA phylogenetic placement, the new species should be assigned to the order Sporadotrichida Fauré‐Fremiet, 1961. Due to low taxon sampling, however, its exact position in this order remains enigmatic.  相似文献
