SUMMARY 1. Grazing and photosynthetic contributions to the carbon balance of planktonic, mixotrophic cryptophytes in Lakes Fryxell and Hoare in the Taylor Valley, Antarctica were measured during November and December 2000.
2. The cryptophytes never became entirely photosynthetic, although carbon derived from grazing decreased in December. Individual grazing rates ranged between 5.28 and 10.08 bacteria cell−1 day−1 in Lake Fryxell and 0.36–11.76 bacteria cell−1 day−1 in Lake Hoare. Grazing rates varied temporally and with depth in the water column. In Lake Fryxell, which is a meromictic lake, highest grazing occurred just above the chemocline. Individual photosynthetic rates ranged from 0.23 to 1.35 pg C cell−1 h−1 in Lake Fryxell and 0.074 to 1.08 pg C cell−1 h−1 in Lake Hoare.
3. Carbon acquisition by the cryptophyte community gained through grazing ranged between 8 and 31% during November in Lake Fryxell, dropping to between 2 and 24% in December. In Lake Hoare grazing contributed 12–21% of the community carbon budget in November and 1–28% in December. Around 4% of the carbon acquired from grazing and photosynthesis was remineralised through respiration.
4. Mixotrophy is probably a major survival strategy for cryptophytes in the extreme lakes of the Dry Valleys, because perennial ice-cover severely limits light penetration to the water column, whereas these phytoflagellates are not normally mixotrophic in lower latitude lakes. The evidence suggests that mixotrophy may be a mechanism for supplementing the carbon budget, as well as a means of acquiring nutrients for growth.  相似文献

1. The ingestion rates of planktonic, mixotrophic cryptophytes in two perennially ice-covered Antarctic lakes in the McMurdo Dry Valleys, were investigated during the summer of 1997–1998.
2. In Lake Fryxell, which is meromictic, ingestion rates increased with depth in November and were highest in a cryptophyte maximum close to the chemocline. In Lake Hoare, which is unstratified and freshwater, there was no significant difference in ingestion rates with depth. In both lakes, the highest ingestion rates occurred in early summer, decreasing in December and January. Ingestion rates varied between 0.2 bacteria cell−1 h−1 and 3.6 bacteria cell−1 h−1.
3. During November, mixotrophic cryptophytes removed up to 13% of bacterial biomass day−1 and had a greater grazing impact than heterotrophic nanoflagellates (HNAN). As summer progressed, the grazing impact of cryptophytes and HNAN became similar.
4. The maximum depth of cryptophytes in Lake Fryxell was predated by a population of the ciliate Plagiocampa. Plagiocampa had an ingestion rate of 0.13–0.19 cryptophytes cell−1 h−1. The grazing impact on the cryptophyte community was insignificant. However, the ciliate appeared to be indulging in temporary mixotrophy, sequestering the cryptophytes for a number of weeks before digesting them.
5. It is suggested that mixotrophy is an important survival strategy in the extreme lake ecosystems of the McMurdo Dry Valleys.  相似文献

Abstract: The rates of ingestion of bacteria and of accumulation of bacterial biomass by hungry Pteridomonas danica and Paraphysomonas imperforata were measured using dual radioactive-labelled bacteria in experiments lasting 4–8 h. Pteridomonas continuously consumed 4–5 bacteria h−1 throughout experiments lasting 8 h, irrespective of bacterial concentration above a threshold of about 5 × 105 bacteria ml−1, and continued to catch bacteria even below this density. The clearance rate of about 1 nl cell−1 h−1 at higher bacterial concentrations increased three or four times as bacterial numbers fell. Paraphysomonas cells, with only half the biomass of Pteridomonas , ingested up to 10 bacteria h−1 at high bacterial concentrations, and gradually reduced the feeding rate, effectively ceasing to feed at 106 bacteria ml−1; their initial clearance rate of 1–2.5 nl cell−1 h−1 subsequently fell as low as 0.1 nl cell−1 h−1. Estimation of feeding rate by extrapolation from short-term experiments on such flagellates requires extreme caution. These flagellates, starved to levels typical of the natural environment, accumulated ingested bacterial biomass at an efficiency of between 16 and 21%, indicating that in nature they would recycle 80% or more of the nutrients contained in their food.  相似文献

Viral activity in two contrasting lake ecosystems   总被引:3,自引:0,他引:3
For aquatic systems, especially freshwaters, there is little data on the long-term (i.e., >6-month period) and depth-related variability of viruses. In this study, we examined virus-induced mortality of heterotrophic bacteria over a 10-month period and throughout the water column in two lakes of the French Massif Central, the oligomesotrophic Lake Pavin and the eutrophic Lake Aydat. Concurrently, we estimated nonviral mortality through heterotrophic nanoflagellate and ciliate bacterivory. Overall, viral infection parameters were much less variable than bacterial production. We found that the frequency of visibly infected cells (FVIC), estimated using transmission electron microscopy, peaked in both lakes at the end of spring (May to June) and in early autumn (September to October). FVIC values were significantly higher in Lake Pavin (mean [M] = 1.6%) than in Lake Aydat (M = 1.1%), whereas the opposite trend was observed for burst sizes, which averaged 25.7 and 30.2 virus particles bacterium(-1), respectively. We detected no significant depth-related differences in FVIC or burst size. We found that in both lakes the removal of bacterial production by flagellate grazing (M(Pavin) = 37.7%, M(Aydat) = 18.5%) was nearly always more than the production removed by viral lysis (M(Pavin) = 16.2%, M(Aydat) = 19%) or ciliate grazing (M(Pavin) = 2.7%, M(Aydat) = 8.8%). However, at specific times and locations, viral lysis prevailed over protistan grazing, for example, in the anoxic hypolimnion of Lake Aydat. In addition, viral mortality represented a relatively constant mortality source in a bacterial community showing large variations in growth rate and subject to large variations in loss rates from grazers. Finally, although viruses did not represent the main agent of bacterial mortality, our data seem to show that their relative importance was higher in the less productive system.  相似文献

In contrast to the water column, the fate of bacterial production in freshwater sediments is still a matter of debate. Thus, the importance of virus-induced lysis and protozoan grazing of bacteria was investigated for the first time simultaneously in a silty sediment layer of a mesotrophic oxbow lake. Microcosms were installed in the laboratory in order to study the dynamics of these processes over 15 days. All microbial and physicochemical parameters showed acceptable resemblance to field data observed during a concomitant in situ study, and similar conclusions can be drawn with respect to the quantitative impact of viruses and protozoa on the bacterial compartment. Viral decay rates ranged from undetectable to 0.078 h−1 (average, 0.033 h−1), and the control of bacterial production from below the detection limit to 36% (average, 12%). The contribution of virus-induced lysis of bacteria to the dissolved organic matter pool as well as to benthic bacterial nutrition was low. Ingestion rates of protozoan grazers ranged from undetectable to 24.7 bacteria per heterotrophic nanoflagellate (HNF) per hour (average, 4.8 bacteria HNF−1 h−1) and from undetectable to 73.3 bacteria per ciliate per hour (average, 11.2 bacteria ciliate−1 h−1). Heterotrophic nanoflagellate and ciliates together cropped up to 5% (average, 1%) of bacterial production. The viral impact on bacteria prevailed over protozoan grazing by a factor of 2.5–19.9 (average, 9.5). In sum, these factors together removed up to 36% (average, 12%) of bacterial production. The high number of correlations between viral and protozoan parameters is discussed in view of a possible relationship between virus removal and the presence of protozoan grazers.  相似文献

1. The seasonal and vertical distribution of the abundance of virus‐like particles (VLPs), together with the abundances of other microbial organisms (bacteria, unpigmented and pigmented nanoflagellates and ciliates), were determined in an oligomesotrophic lake (Pavin, France) and in a eutrophic lake (Aydat, France) between March and December 2000. 2. The abundance of the viral plankton and those of other microbial taxa were significantly higher in the more productive system. The same was for the virus‐to‐bacteria quotient (VBQ), which averaged seven in Lake Pavin and nine in Lake Aydat. 3. The abundance of viruses increased during the period of thermal stratification in both lakes, with the highest values being recorded at the end of summer/early autumn in the epi‐ and the metalimnion. The seasonal pattern of abundance of viruses in both lakes in the surface layer was similar, indicating that the dynamics of viruses may be controlled by environmental factors such as light conditions. 4. There was no correlation between the abundance of viruses and protists. We found correlations between viruses and heterotrophic bacteria in the whole water column in Lake Pavin, but only in the dark bottom waters in Lake Aydat. 5. Overall, the empirical findings in this study lead us to speculate that the weaker correlation between bacteria and viruses in Lake Aydat than in Lake Pavin, as well as the higher VBQ in the former, is a consequence of the increasing relative abundance of non‐bacteriophage VLPs along the trophic gradient of aquatic systems.  相似文献

Abstract: Plankton community structure and fluxes of carbon for bacteria (production and bacterivory) were investigated in the urban, hypertrophic Lake Rodó (Uruguay) using a short time interval for sampling (5–15 d) during one year. The lake sustains a high phytoplankton biomass (up to 335 μg l−1 chlorophyll a ) always dominated by the filamentous cyanobacteria Planktothrix agardhii . The zooplankton community was numerically dominated by rotifers and ciliates; cladocerans were rare during most of the year. The rotifer abundance was very high (up to 105 individual l−1), the bacterivorous Anuraeopsis fissa being the most abundant species. Predation rates of heterotrophic nanoflagellates (HNF) on bacteria (range: 31–130 bacteria HNF−1 h−1) were higher than those reported in the literature for field studies. A carbon budget showed that HNF can consume on average 91 and 76% of the bacterial carbon production in summer and winter, respectively. Bacterial turnover times are the lowest reported until now from field conditions (5 to 42 h). Consequently, bacterial carbon production was extremely high (72 to 1071 μg C l−1 d−1). Bacterial production was positively correlated to bacterial abundance but the relationship was significantly improved by the inclusion of temperature (82% variability explained). My results support the general trend for increased bacterial production with increasing trophic status, and suggest a lower energy transfer efficiency to higher trophic levels in hypertrophic lakes due to the many trophic interactions involved.  相似文献

1. The seasonal development of heterotrophic nanoflagellates (HNF), bacteria, rotiferans and crustacean zooplankton was studied in the epilimnion of Lake Pavin, an oligomesotrophic lake in the Massif Central of France.
2. HNF abundance varied from 0.1 to 2.5 × 103 mL–1. Free-living HNF reached their highest density in spring when the copepod Acanthodiaptomus denticornis dominated the metazooplankton. They were present in low numbers when rotifers and cladocerans were numerous.
3. Attached HNF, consisting of bicoecids and choanoflagellates, were fixed to large diatoms and to the colonial cyanobacterium Anabaena flos-aquae . The abundance of attached HNF was significantly correlated to bacterial abundance, which fluctuated between 1.1 and 2.7 × 106 mL–1. Highest abundance of these epiphytic protists was recorded when free-living heterotrophic nanoflagellates declined.
4. The comparison of the dynamics of heterotrophic nanoflagellates, bacteria, and the impact of zooplankton grazing suggested that prey abundance, the presence of suitable attachment sites and limited competition from the free-living forms were the main factors controlling the development of the epiphytic flagellate protists. In contrast, the low abundance of free-living forms during the period of rotiferan and cladoceran development suggests the prevalence of a top-down control by predation of the metazoopankton.  相似文献

SUMMARY. 1. Heterotrophic bacterioplankton growth and production rates were estimated in a tropical lake by various methods. Mean growth rates, determined by tritiated thymidine incoporation into DNA, frequency of dividing cells and increase in cell density varied between 0.013 and 0.014 (with a range of 0.006–0.026) h−1 corresponding to bacterial production of 1.16–1.22 (0.34–3.63) mg C m−3 h−1.
2. Heterotrophic bacterial production estimated from oxygen and inorganic carbon consumption in the dark were compared with these values. The oxygen method gave similar results, while values from dark carbon uptake were as much as 2.5 times higher.
3. Although the different estimates of rates of bacterial production showed different patterns, the existence of spatial (vertical) and temporal (diel and seasonal) variation was demonstrated. Bacterial production was 13–41% of the net primary production and 10–30% of gross primary production.
4. Bacterial grazing mortality rate was estimated from size-fractionation and metablic inhibitor experiments. Average grazing rates were between 0.34 and 3.77 mg C m−3 h−1 corresponding to 76–120% of the mean bacterial production rate. Organisms 1–12 μm in size, possibly mainly ciliates. were implicated as important bacterial grazers.  相似文献

We explored the abundance and infection rates of viruses on a time series scale in the euphotic zone of the humic mesotrophic Lake Vassivière (Massif Central, France) and compared them to nonhumic lakes of contrasting trophy (i.e., the oligomesotrophic Lake Pavin and the eutrophic Lake Aydat) located in the same geographical region and sampled during the same period. In Lake Vassivière, the abundances of virus-like particles (range, 1.7 × 10(10) to 2.6 × 10(10) liter(-1)) were significantly (P < 0.001) lower than in Lakes Pavin and Aydat. The percentage of virus-infected prokaryotic cells (mean, 18.0%) was significantly higher (P < 0.001) in Vassivière than in Pavin (mean, 11.5%) and Aydat (mean, 9.7%). In Vassivière, the abundance of prokaryotes was a good predictor (r = 0.78, P < 0.001) of the number of virus-like particles, while the potential grazing rate from heterotrophic nanoflagellates was positively correlated to the viral infection rate (r = 0.75, P < 0.001; n = 20), indicating the prevalence of cycling interactions among viruses, prokaryotes, and grazers, which is in agreement with past experiments. The absence of correlation between chlorophyll a concentrations (Chl) and viral parameters suggested that the resources for the lytic activity of viruses in Vassivière were mainly under allochthonous control, through host activity. Indeed, compilation of data obtained from several nonhumic lakes in the French Massif Central revealed that Chl was positively correlated to the abundance of virus-like particles at concentrations above 0.5 μg Chl liter(-1) and negatively at concentrations below 0.5 μg Chl liter(-1), suggesting that phytoplankton-derived resources could force prokaryotic growth to attain a certain threshold level when the host availability is sufficient to boost the proliferation of viruses. Therefore, based on the high level of lytic infection rates in Lake Vassivière, we conclude that viruses are key agents for prokaryotic mortality and could influence the food web dynamics in humic lakes, which may ultimately depend on the internal cycling of resources and, perhaps, mainly on the allochthonous inputs and the associated humic substances.  相似文献

Abstract A method proposed for estimating the rate of bacterial mortality in aquatic environments consists in following the disappearance of radioactive tracer from the macromolecular fraction of 3H-thymidine labeled natural assemblages of bacteria. The data presented in this paper offer a further technical validation of this procedure. Application of the method to North and Mediterranean Sea, estuaries, rivers and lakes yields first order mortality constant in the range 0.008–0.06 h−1. Mortality due to grazing by protozoans retained by 2 μm filtration range from 20 to 90% of the mortality detected by the method.  相似文献

Abstract Bradyrhizobium japonicum and Shewanella putrefaciens were unable to oxidize hydrogen at atmospheric concentrations (0.55 ppmv), neither in suspension nor when added to sterile soil. The K m-value of S. putrefaciens for H2 (39 ppmv in gas phase, 0.22 μM in aqueous phase), using Fe(III) as electron acceptor, showed a 4–5-fold higher affinity for H2 than that of B. japonicum (1200 ppmv; 0.84 μM) or other hydrogen-oxidizing bacteria. However, the V max (4.54 fmol H2 h−1 cell −1) and threshold (> 0.5 ppmv; 0.35 nM) of S. putrefaciens and the V max (7.19 fmol H2 h−1 cell−1) and threshold (> 0.5 ppmv; 0.35 nM) of B. japonicum were in the same order of magnitude as data for Knallgas bacteria from relevant literature. To enable hydrogen oxidation in soil the soil-samples with S. putrefaciens even had to be supplemented with Fe(III). Fresh soil, on the other hand, oxidized hydrogen very efficiently below atmospheric mixing ratios, demonstrating that there must be other oxidation activities in soil.  相似文献

Abstract The fluorescent antibody (FA) technique was employed to estimate the filtration rates of Daphnia cucullata on the Alcaligenes eutrophus JMP134 strain introduced into lake water. We investigated the effect of animal density and food concentration on the filtration rates in 0.2-μm filtered and autoclaved lake water. The individual filtration rates were 0.08–1.12 ml animal−1 h−1. Filtration rates decreased with increasing grazer density and with increasing food concentrations. The grazing impact on indigenous and introduced bacteria was compared in experiments with 2-μm filtered lake water. Filtration rates of D. cucullata on A. eutrophus were greater than on indigenous bacteria. These differences were explained by size-selective grazing, implying that this mechanism is important in the elimination of introduced bacteria from lake water when the introduced bacteria are in the larger size-range of the indigeneuos bacterial flora.  相似文献

Water samples were collected along transects from the shore to the centre of two French lakes: the deep, volcanic, oligomesotrophic and low allochthonic-impacted Lake Pavin, and the productive and higher allochthonic-impacted Lake Aydat. The biodiversity was analysed using two approaches: the classical approach consisting of cloning/sequencing of the 18S, ITS1, 5.8S, ITS2 and partial 28S region using primers designed for fungus sequences, and the pyrosequencing of 18S rRNA hypervariable V2, V3 and V5 regions using two primer sets (one universal for eukaryotes and one for fungi). The classical approach yielded 146 (Lake Pavin) and 143 (Lake Aydat) sequences, corresponding to 46 and 63 operational taxonomic units (OTUs) respectively. Fungi represented half of the OTUs identified in Lake Pavin and 30% in Lake Aydat, and were dominated by sequences from Chytridiomycota found throughout Lake Pavin but mostly in the central pelagic zone of Lake Aydat. The pyrosequencing approach yielded 42,064 (Pavin) and 61,371 (Aydat) reads, of which 12-15% and 9-19% reads were assigned to fungi in Lakes Pavin and Aydat respectively. Chytridiomycota members were also dominant among these reads, with OTUs displaying up to > 33-fold overrepresentation in the centre compared with the riparian areas of Lake Aydat. Besides fungi, both approaches revealed other major eukaryote groups, with the highest diversity in the central areas of lakes. One of the major findings of our study was that the two lakes displayed contrasting spatial distributions, homogenous for Lake Pavin and heterogeneous for Lake Aydat, which may be related to their peculiarities. This study represents the first unveiling of microbial eukaryote and fungus diversity assessed with two complementary molecular methods, and is considered a major milestone towards understanding the dynamics and ecology of fungi in freshwater lake ecosystems, which are directly link to the abundance and distribution of taxa.  相似文献

SUMMARY. Dissolved ATP, defined as ATP which passes through 0.2 μm filters, was found in fresh water. During the spring diatom bloom in two eutrophic Danish lakes, concentrations of dissolved ATP varied between 0.1 and 3.8 μgl−1, constituting 14–76% of the total ATP (particulate plus dissolved ATP). The kinetics of the light emission obtained from mixing firefly enzyme with dissolved ATP demonstrated that the major proportion of the dissolved ATP was in fact ATP. Despite some variations, the seasonal changes in dissolved ATP paralleled the changes in the increasing phytoplankton population during the rise of the diatom blooms. The dissolved ATP increased after the diatom peak, indicating that release of ATP from the phytoplankton due to mortality may be a major source of dissolved ATP.
Consumption of dissolved ATP was evaluated in uptake experiments using 3H-ATP. Rates of uptake of 3H-ATP by micro-organisms (diameter 0.2–0.6 μm) proved to be close to the rates for 3H-D-glucose uptake. The variations in 3H-ATP uptake during the diatom blooms showed non-systematic changes and ranged between 1.0 and 15.8% h−1 (mean = 4.9% h−1) of the quantity added. Turnover rates for dissolved ATP varied between 12 and 730 ng l−1 h−1 (mean = 175 ng l−1). These rather high rates of turnover suggest that dissolved ATP is an important compound in the metabolism of freshwater bacteria.  相似文献

Catch and effort data for the period 1973–1990 demonstrate a dramatic decline of lungfish in the Tanzanian waters of Lake Victoria. Bottom trawl catches in the Mwanza Gulf showed a decline in catch rates from 67.5 kg h−1 in 1973 to 5.5 kg h−1 in 1986. Trawling of commercial vessels in the Speke Gulf revealed a decline in lungfish catches from 1.3 kg h−1 in 1986 to 0.07 kg h−1 in 1990. The development of anoxia in the deeper waters of Lake Victoria, the algal blooms, and the decline of water transparency, all associated with eutrophication, are not likely to have contributed to the decreased catch rate. However, the lungfish decline may reflect the interaction of overexploitation by the fishery and a low level of Nile perch predation that restricts lungfish to wetland refugia. We suggest that this may have been reinforced over the past few decades by large-scale conversion of wetlands to agricultural land and harvesting of nest-guarding male lungfish leading to decreased recruitment of young.  相似文献

Methanogenesis and methanotrophy within a Sphagnum peatland   总被引:4,自引:0,他引:4
Abstract: Methane production and consumption activities were examined in a Massachusetts peatland. Peat from depths of 5–35 cm incubated under anaerobic conditions, produced an average of 2 nmol CH4 g−1 h−1 with highest rates for peat fractions between 25–30 cm depth. Extracted microbial nucleic acids showed the strongest relative hybridization with a 16S rRNA oligonucleotide probe specific for Archaea with samples from the 25–30 cm depth. In aerobic laboratory incubations, the peat consumed methane with a maximum velocity of 67 nmol CH4 g−1 h−1 and a K s of 1.6 μM. Methane consumption activity was concentrated 4–9 cm below the peat surface, which corresponds to the aerobic, partially decomposed region in this peatland. Phospholipid fatty acid analysis of peat fractions demonstrated an abundance of methanotrophic bacteria within the region of methane consumption activity. Increases in temperature up to 30°C produced an increase in methane consumption rates for shallow samples, but not for samples taken from depths greater than 9 cm. Nitrogen fixation experiments were carried out using 15N2 uptake in order to avoid problems associated with inhibition of methanotrophy. These experiments demonstrated that methane in peat samples did not stimulate nitrogen fixation activity, nor could activity be correlated with the presence of methanotrophic bacteria in peat fractions.  相似文献

Twenty-four ammocoete larvae of the sea lamprey Petromyzon marinus were tested individually for 3-day periods in electronic shuttleboxes (Ichthyotrons) with natural lighting through windows, to determine their diel activity pattern in terms of hourly percentages of the 24-h total. Mean hourly activity was slightly greater during the day (4–7 % h−1) than at night (3·7% h−1). Peak hourly activity (7·2% h−1) occurred during the dawn crepuscular period as light levels were increasing, while the minimum hourly activity (1·6 % h-l) occurred during the dusk crepuscular period while light levels were decreasing. The observed activity pattern is atypical of other aquatic animals similarly tested, and might be explained as an extraptically mediated photoinetic response.  相似文献

Abstract Bacteria showing rapid growth on a nitrogenfree medium and acetylene-reducing activity were isolated from maize roots collected from agricultural soils in Spain. The isolates were Gram-negative motile rods and were identified as Azotobacter chroococcum . Acetylene-reducing activity and microbial counts were determined on root segments from 7- and 30-day-old plants. Rates obtained were in the range of 0.0053–0.848 nmol C2H2· g−1· h−1. Root populations were 1.4–6.0 × 104 micro-organisms · g−1. These results showed that there was an association between A. chroococcum strains and roots of maize planted in some Spanish soils.  相似文献
