急性髓系白血病(acute myeloid leukemia,AML)是一类造血干细胞的恶性克隆性疾病,目前的诊断方法不利于疾病的早期发现,且诊断结果重复性较差。已有大量研究显示,细胞外microRNA(miRNA或miR)富集在外泌体(exosome)中,且受其表面膜的保护而具有很好的稳定性,是理想的分子标志物。目前,多种实体肿瘤均已检测到肿瘤特异性外泌体miRNA(exosomal miRNA)。然而,在AML患者中未见此外泌体miRNA报道。本研究探讨急性髓系白血病血浆外泌体miRNA表达谱差异及新miRNA序列。采用solexa高通量测序技术对7例AML患者(AML组)及7例健康对照者(对照组)血浆外泌体miRNAs进行测序,利用Mireap预测软件进行新miRNAs分析,通过edger差异分析软件筛选组间差异miRNA,获得211个已知的差异表达miRNAs以及2个新miRNAs,选择4个差异表达的miRNAs:miR-155-5p、miR-335-5p、miR-451a及xxx-m0038 5p(新miRNA),在两组(各23例)的血浆外泌体样本中,进行实时荧光定量PCR(qRT-PCR)验证,验证结果与测序结果一致。对差异表达的外泌体miRNA进行靶基因预测及其GO(Gene Ontology)和信号通路富集分析,发现靶基因聚集的生物学功能多数参与生物进展过程的调控。靶基因主要富集在FoxO、MAPK、Hippo信号通路以及HTLV-I感染等。结果显示,AML患者与健康对照者的血浆外泌体miRNA存在着差异性表达。差异性表达的miRNA特异性很高,对进一步阐明AML白血病发生与发展的分子机制、研发新的无创诊断方法、新的诊断标记物和有效治疗AML的方法具有十分重要和深远的意义。 相似文献

目的:研究急性髓系白血病免疫表型特征以及遗传学特征。方法:选取2011年1月到2014年5月我院收治的急性髓系白血病患者169例,采用流式细胞术和相关的单克隆抗体来分析所有患者的骨髓免疫表型,采用染G染色体显带技术分析患者的核型,根据淋系抗原(lym Ag)的表达将患者分为lym Ag+组和lym Ag-组。结果:抗原CD13、CD33、CD117以及MPO等髓系抗原最常在急性髓系白血病患者中表达,其中CD117在M3型病例中表达为85.7%(24/28),而CD34、HLA-DR双阴性、较强的自发荧光、CD13、CD33和MPO对M3型的诊断也具有一定的价值;其中47.9%(81/169)的患者伴随着淋系抗原表达,以CD7和CD56为主;60.4%(102/169)的患者伴随着核型异常;而伴随着t(8:21)的M2患者中的CD15、CD19和CD56的表达显著增强,而t(15:17)均发生于M3型患者中;而lym Ag+组患者CD34的阳性患者为77.8%(63/81)显著高于lym Ag-组的47.7%(42/88),两组比较差异具有统计学意义(P0.05)。结论:免疫表型对急性髓系白血病的诊断具有重要的意义,且免疫表型和异常核型存在密切的联系。 相似文献

急性髓系白血病(acute myeloid leukemia,AML)是异质性造血干细胞恶性克隆性疾病,主要表现为外周血、骨髓以及其他组织中的原始细胞克隆性扩增。近年来,AML发病率呈逐年增加的趋势,对人类健康产生巨大的威胁。在不同种族及不同性别中,AML发病率及死亡率存在显著差异。AML是一种复杂疾病,与遗传突变及异常表观遗传修饰密切相关。DNA甲基化是重要的表观遗传修饰,AML相关基因的异常DNA甲基化修饰对疾病的发生发展极其重要。该文对AML相关基因的异常甲基化修饰进行了系统的作用机制分析,并对重要基因进行了功能分类。该文总结的具有异常DNA甲基化修饰的基因,有望辅助预测AML的治疗及预后效果,并为设计个体化AML治疗方案提供全新的思路。 相似文献

目的 探讨RNAi技术治疗急性髓系白血病(acute myeloid leukemia,AML)对p38信号通路的影响。方法 以NC(HL-60细胞)和HK(含siRNA-FLT3的阴性对照的HL-60细胞)、F1(实验组,含siRNA-FLT3的HL-60细胞,本实验室已实验证明对FLT3有干扰作用)三种细胞株为研究对象,分为药物组(信号通道抑制作用组)和对照组(未加信号通道抑制剂)。通过RT-PCR和Western blot检测对照组和药物组p38 mRNA水平和蛋白表达的变化,MTT和FCM分别测定细胞活性和细胞凋亡率的变化。结果 药物组、对照组组内比较:以NC做参照,F1能诱导p38 mRNA水平和蛋白表达下降、从而抑制细胞的增殖,促进细胞凋亡,差异有统计学意义(均P 0.05);与对照组相比,药物组p38 mRNA水平和蛋白表达下降,细胞活性下降,细胞凋亡率增加,差异有统计学意义(均P<0.05);药物组的F1与药物组的NC、药物组的F1与对照组的F1分别比较,差异有统计学意义(均P s<0.05)。结论 p38信号通路是RNAi技术干扰HL-60细胞FLT3基因表达引起细胞凋亡的途径之一,SB203580(P38 MAPK抑制剂)对其有协同增效作用。 相似文献

目的:探索缓解与未缓解急性髓系白血病干细胞表面抗原表达差异,为判定化疗疗效及其预后提供依据。方法:按照急性白血病诊断标准,根据患者入院时骨髓白血病细胞数量多少分成临床缓解与未缓解两组,以流式细胞仪分别检测骨髓中白血病干细胞表面相关抗原表达情况,比较二者之间差异。其中经标准化疗方案治疗结束后,通过复查骨髓象判定疗效并比较化疗前后白血病干细胞表面相关抗原表达变化。结果:与缓解的急性髓系白血病患者骨髓白血病干细胞相关抗原表达值相比,未缓解的患者骨髓白血病干细胞表面相关抗原表达明显升高,差异具有统计学意义(P〈0.05,0.001);未缓解的患者经标准方案化疗后骨髓虽然已经获得完全缓解,但依然具有白血病干细胞表面抗原高表达,提示这部分患者依然有复发的可能性。结论:急性髓系白血病患者的白血病干细胞相关抗原表达值升高是急性白血病复发难治的根源之一。 相似文献

通过数据挖掘和生物信息学分析手段,探讨施旺膜蛋白相互作用因子1(schwannomin interacting protein 1,SCHIP1)基因在急性髓系白血病患者中的表达情况及其临床意义。首先,对Oncomine数据库中收录的所有急性髓系白血病(acute myloid leukaemia,AML)数据集进行荟萃分析,筛选出目标基因SCHIP1并进一步分析其在AML病人中的表达变化。随后,从GEO数据库中下载含生存信息的AML数据集源文件,分析SCHIP1表达对疾病的预后作用。另外,利用TCGA数据库对SCHIP1的表达情况进行亚组分析及与FLT3基因突变、PML/RARα融合基因和RAS活化等高危因素进行相关性分析。最后,利用GEPIA2工具验证SCHIP1的表达情况、预后意义及与FLT3、PML基因表达的相关性。结果发现Oncomine数据库中收录了44个AML数据集,总计共3534个样本数据。其中5个数据集共1188个样本包含“Cancer vs.Normal”的mRNA表达数据,对其进行荟萃分析显示SCHIP1位于显著高表达分子的第17位。生存分析显示,SCHIP1表达量与AML患者总体生存率呈负相关。亚组分析显示SCHIP1在M0/M1/M2中较M3/M6中表达更高,但与年龄、性别和种族无关。另外,相关性研究分析显示SCHIP1与FLT3基因突变弱相关,但与PML/RARα融合基因和RAS活化等高危因素无显著相关性。这些结果表明SCHIP1在急性髓系白血病中高表达,且其高表达与患者的生存预后呈显著负相关。因此,SCHIP1可作为疾病的预后生物标志物,并有望成为AML的精准治疗靶点。 相似文献

The focus of this study was to investigate the expression status of Circ-vimentin (VIM) and further analyze its pathogenesis and clinical significance in acute myeloid leukemia (AML) patients. Real-time quantitative polymerase chain reaction was carried on Circ-VIM in 113 AML patients and 42 healthy controls. Circ-VIM was significantly upregulated in AML compared with control and was positively correlated with white blood cells (WBC) count. Receiver operating characteristic curve analysis indicated that the performance of Circ-VIM expression could serve as a promising biomarker for differentiating AML patients from controls. Significant correlations of Circ-VIM expression were found with WBC and French–American–British classifications. Survival analyses further showed that over-expressed Circ-VIM were associated with markedly shorter overall survival (OS) and leukemia-free survival (LFS) in whole-cohort AML, nonacute promyelocytic leukemia AML and cytogenetically normal-AML patients. Multivariate analysis also disclosed that Circ-VIM over-expression was an independent poor prognostic factor for OS and LFS in AML patients. Remarkably, Pearson correlation analysis evidenced that the expression of Circ-VIM was positively correlated with VIM expression in all AML patients. Mounting data have shown that long non-coding RNAs (lncRNAs) widely participate in tumour initiation, development, progression and glycolysis in a variety of tumours. However, the clinical prognosis and molecular mechanisms of TMEM161B-AS1 in oesophageal squamous cell carcinoma (ESCC) remain still unknown. Here, TMEM161B-AS1 and HIF1AN were significantly lower in ESCC tissues than in normal samples, and their low expressions were both related to TNM stage, lymph node metastasis and poor prognosis of ESCC patients. Functionally, TMEM161B-AS1 overexpression or miR-23a-3p depletion suppressed the proliferation, invasion and glycolysis as well as reduced glucose consumption and lactate production in ESCC cells. Mechanistically, TMEM161B-AS1 manipulated HIF1AN expression by competitively sponging miR-23a-3p in ESCC cells. MiR-23a-3p mimic and HIF1AN siRNA partly reversed cell phenotypes mediated by TMEM161B-AS1 in ESCC cells. Collectively, TMEM161B-AS1, miR-23a-3p and HIF1AN may be tightly involved in ESCC development and progression as well as patients’ prognosis, and TMEM161B-AS1/miR-23a-3p/HIF1AN signal axis may be a promising target for the treatment of ESCC patients. 相似文献

IL-23 is an IL-12 cytokine family member with pleiotropic functions that regulates tumour growth in various cancer types, exhibiting both anti-tumorigenic and pro-tumorigenic properties. Preclinical studies have shown a potential anti-leukemic action on childhood B-ALL cells. The study involved 65 children with acute leukemia [59 patients with acute lymphoblastic leukemia (ALL) and 6 patients with acute myeloid leukemia (AML)] and 27 healthy controls. Using an enzyme-linked immunosorbent assay, we aimed to determine the IL-23 levels in the peripheral blood (PB) and bone marrow (BM) of patients at diagnosis and at the end of the induction therapy (EIT). PB IL-23 levels were lower in leukemia patients compared to the healthy controls. In all acute leukemia patients, IL-23 levels were significantly lower at diagnosis both in PB (P = .015) and in BM (P = .037) compared to the PB and BM concentrations at the EIT. The same pattern was present in both subgroups of ALL and AML patients. The high leukemic burden at diagnosis was related with lower IL-23 levels, which were increased with the disease remission. Considering the anti-leukemic potential of this cytokine, the elevation of the IL-23 concentration at the disease remission indicates a beneficial role of IL-23 in paediatric acute leukemia. 相似文献

MicroRNAs are key regulators of many biological processes, including cell differentiation. These small RNAs exert their function assembled in the RNA-induced silencing complexes (RISCs), where members of Argonaute (Ago) family of proteins provide a unique platform for target recognition and gene silencing. Here, by using myeloid cell lines and primary blasts, we show that Ago2 has a key role in human monocytic cell fate determination and in LPS-induced inflammatory response of 1,25-dihydroxyvitamin D3 (D3)-treated myeloid cells. The silencing of Ago2 impairs the D3-dependent miR-17-5p/20a/106a, miR-125b and miR-155 downregulation, the accumulation of their translational targets AML1, VDR and C/EBPβ and monocytic cell differentiation. Moreover, we show that Ago2 is recruited on miR-155 host gene promoter and on the upstream region of an overlapping antisense lncRNA, determining their epigenetic silencing, and miR-155 downregulation. These findings highlight Ago2 as a new factor in myeloid cell fate determination in acute myeloid leukemia cells. 相似文献
