The aim of the present study was to investigate the protective role of grape seed extract (containing proanthocyandin) against the adverse effects of plant growth regulators (GA3 (gibberellic acid) and IAA (indoleacetic acid)). The present data showed that the administration of either GA3 and IAA caused undesirable changes in both hepatic and testicular structure. This was evidenced by a disturbed hepatic strands, pyknotic nuclei, central vein with collapsed endothelium, dilatation in bile sinusoids, congested blood vessel, binucleatd hepatocytes, lymphocytic infiltration, vacuolation, giant hepatic cells, increased Kupffer cells and karyoryxis. Additionally, it was shown that degenerative changes in the testis, spermatogenic arrest, moderate tubular necrosis, Leydig cell degeneration and reduction in the number and size of the seminiferous tubules with some spermatogonia detached from the basement membrane. Concerning flow cytometric study of the liver a significant decrease in G0/1 % and a significant increase in S phase %, G2/M  %, P53 % and apoptosis % (sub G1) were detected. However, in testis the data recorded a significant decrease in the percentage of mature sperm (percentage of haploid cells) and a significant increase in the percentage of spermatide, diploid cells, P53 and of apoptotic cells. On the other hand, a distinct recovery of the mentioned hepatic and testicular histopathological and cytogenetic disorders was observed when proanthocyanidin was supplemented to rats administered either of the plant growth hormones (GA3 and IAA).  相似文献

In the present study, the influence of subchronic effects of two plant growth regulators (PGRs) [Abcisic acid (ABA) and Gibberellic acid (GA3)] on antioxidant defense systems [reduced glutathione (GSH), glutathione reductase (GR), superoxide dismutase (SOD), glutathione-S-transferase (GST) and catalase (CAT)] and lipid peroxidation level (malondialdehyde = MDA) in various tissues of the rat were investigated during treatment as a drinking water model. 75 ppm of ABA and GA3 in drinking water were continuously administered orally to rats (Sprague-Dawley albino) ad libitum for 50 days. The PGRs treatments caused different effects on the antioxidant defense systems and MDA content of dosed rats compared to controls. The lipid peroxidation end product MDA significantly increased in the lungs, heart and kidney of rats treated with GA3 without significant change in the spleen. ABA caused also a significant increase in MDA content in the spleen, lungs, heart and kidney. The GSH levels were significantly depleted in the spleen, lungs and stomach of rats treated with ABA without any change in the tissues of rats treated with GA3 except the kidney where it increased. Antioxidant enzyme activities such as SOD significantly increased in the lungs and stomach and decreased in the spleen and heart tissues of rats treated with GA3. Meanwhile, SOD significantly decreased in the spleen, heart and kidney and increased in the lungs of rats treated with ABA. While CAT activity significantly decreased in the lungs of rats treated with GA3, a significant increase occurred in the heart of rats treated with both PGRs. On the other hand, the ancillary enzyme GR activity in the tissues were either significantly depleted or not changed with PGRs treatment. The drug metabolizing enzyme GST activity significantly decreased in the lungs of rats treated with ABA but increased in the stomach of rats treated with both PGRs.

As a conclusion, the rats resisted oxidative stress via the antioxidant mechanism. But the antioxidant mechanism could not prevent the increases in lipid peroxidation in rat's tissues. This data, along with changes, suggests that PGRs produced substantial systemic organ toxicity in the spleen, lungs, stomach, heart and kidney during a 50-day period of subchronic exposure.  相似文献

During the entire period of internode growth of Merremia emarginata contents of gibberellic acid (GA3), phenyl-acetic acid (PAA), indole-3-acetic acid (IAA, free and conjugated) and abscisic acid (ABA, free and conjugated) were estimated by ELISA using polyclonal antibodies raised against each hormones. At the time of internode elongation free auxin content was low and increased with the decrease in the rate of elongation. In contrast, conjugated IAA showed declining trend where free IAA content was remarkably high, suggesting thereby that conjugated IAA might have mobilized during the later phase of internode development. The endogenous GA3 contents were high as compared to other hormones; however, no significant role of GA3 was discernible in elongation growth. Dog rose (Rosa canina L.) plants in the bloom stages of flowering were sprayed by indole-3-acetic acid (IAA) in concentrations of 0.06 and 0.60 mM and gibberellic acid (GA3) in concentrations of 0.60 and 1.50 mM. Ascorbic acid, total sugar, reducing sugar and carotenoid contents gradually increased, while the protein content remained unchanged and the content of phenolic substances decreased during hypanthium development. Ascorbic acid, total sugar, reducing sugar and carotenoid contents increased in hypanthium sprayed by GA3 and IAA. However, IAA and GA3 applications (except low concentrations) decreased contents of phenolic substances. IAA and GA applications might be a good way to produce the high quality hypanthium in R. canina.  相似文献

The effect of indole-3-acetic acid (IAA) and gibberellic acid (GA3) applications on parthenocarpic fruit set in Rosa canina and Diospyros lotus was investigated. GA3 induced parthenocarpic fruit set in both plants, but IAA only in D. lotus. Maturation of seedless fruits was earlier than the seeded fruits. GA3 caused a decrease in the fresh mass and size of both fruits. IAA induced an increase in the fresh mass and size in parthenocarpic fruit of D. lotus. Abscisic acid (ABA) inhibits the production of ethylene induced by water stress in excised wheat leaves and counteracts the stimulatory effect of 6-benzyladenine (BA) on this process. The stimulatory effect of BA and the inhibitory effect of ABA were equally pronounced whether external or endogenous ethylene levels were determined. When leaves were sprayed or floated on solutions of BA, indole-3-acetic acid (IAA), gibberellic acid (GA3), or ABA, the relative activities of these growth regulators on stress-induced ethylene at 10-4 mol l-1 were BA>IAA >GA3>controls>ABA. In non-stressed leaves, however, where the levels of ethylene produced were 2–20 times smaller, the relative activities were IAA >BA>GA3>controls>ABA. The effects of BA and ABA spray treatment on water stress induced ethylene were closely similar whether the solutions were applied 2 or 18 h prior to the initiation of water stress. Summary The effect of 3-indoleacetic acid (IAA), 6-furfurylaminopurine (kinetin), and gibberellic acid (GA3) on germination of the orchid Comparettia falcata was evaluated in a factorial experiment (4×4×4) with Murashige and Skoog (1962) basal medium. It was established that seeds of this orchid could be maintained under aseptic conditions as long as the necessary nutrients and appropriate concentrations of growth regulators were provided. Of the three growth regulators used, IAA significantly decreased seed germination of Comparettia falcata. There was a synergistic effect in the kinetin:GA3 combination that produced a positive response in both percentage seed germination and development of seedlings. This study describes a single medium-based protocol able to achieve more than 160000 seedlings within 21 wk, starting from a single capsule, sufficient for both large-scale propagation and in vitro conservation of this threatened orchid.  相似文献

The hypothesis that auxin and gibberellic acid (GA3) control the differentiation of primary phloem fibers is confirmed for the stem of Coleus blumei Benth. Indoleacetic acid (IAA) alone sufficed to cause the differentiation of a few primary phloem fibers. In long term experiments auxin induced a considerable number of fibers in mature internodes. GA3 by itself did not exert any effect on fiber differentiation. Combinatiosn of IAA with GA3 completely replaced the role of the leaves in primary phloem fiber differentiation qualitatively and quantitatively. Although the combined effect of the two growth hormones diminished considerably with increasing distance from the source of induction, auxin with GA3 or IAA alone induced fibers in a few internodes below the application site. When various combinations of both hormones were applied, high concentrations of IAA stimulated rapid differentiation of fibers with thick secondary walls, while high levels of GA3 resulted in long fibers with thin walls. The size of the primary phloem fibers correlated with the dimensions of the differentiating internode, thereby providing evidence that both growth regulators figure in the control of stem extension. High IAA/low GA3 concentrations have an inhibitory effect on internode elongation, whereas low IAA/high GA3 concentrations promote maximal stem elongation.  相似文献

The effects of 5-bromo-2-deoxyuridine (BUdR, thymidine analogue), AMO-1618 (2-isopropyl-4-dimethylamino-5-methylphenyl-1-piperidine carboxylate methyl chloride), a growth retardant, and p-chlorophenoxyisobutyric acid (PCIB, an antiauxin) on growth (dry weight increase) and fiber development in unfertilized cotton (Gossypium hirsutum L.) ovules grown in vitro have been studied. BUdR (5 M) causes about 70% inhibition of fiber production, with little effect on ovule growth, if applied during the first 6 d of culture in the presence of GA3 and IAA. AMO-1618, when used with GA3 alone, causes only a small reduction in both dry weight and fiber production, but when used with IAA alone reduces both fiber production and dry weight, the effect on the latter being predominant. In the presence of both IAA and GA3, AMO-1618 causes a small decrease in fiber production but a major decrease in dry weight. PCIB completely inhibits fiber growth but has little effect on dry weight, especially when GA3 is present. The hypothesis that auxin (IAA) and gibberellic acid (GA3) control the formation of lignin is confirmed for the primary phloem fibers and for the secondary xylem in the stem of Coleus blumel Benth. Indoleacetic acid alone, or a combination of high IAA/low GA3 (w/w), induced short phloem fibers with thick secondary walls, that contained lignin rich in syringyl units (high ratio of syringyl/guaiacyl). On the other hand, a combination of high GA3/low IAA (w/w), which promoted the differentiation of long phloem fibers with thin walls, decreased the relative content of the syringyl units (low syringyl/guaiacyl ratio). In the secondary xylem, these hormonal treatments yielded only slight changes in the noncondensed monomeric guaiacyl units, confirming the relative stability of the guaiacyl lignification pattern in this tissue. In the xylem, indoleacetic acid alone, or a combination of high IAA/low GA3 induced lignin poor in syringyl units (low syringyl/guaiacyl ratio). A combination of high GA3/low IAA promoted a relatively slight increase in syringyl yield, indicating greater responsiveness of the syringyl lignification pattern to growth regulators. Gibberellic acid (GA3) is an endogenous plant growth regulator used worldwide in agriculture. The objective of this study was to investigate the effects of GA3 on the kidney function of adult rats and their pups. Female Wistar rats were given daily 200 ppm GA3 in drinking water from the 14th day of pregnancy until day 14 after delivery. GA3 induced nephrotoxicity, as evidenced by a reduction in the 24-h urine volume and an increase in plasma creatinine, urea and uric acid levels. Nephrotoxicity was objectified by a significant increase of malondialdehyde level and a decrease of antioxidant enzyme activities like catalase, superoxide dismutase, glutathione peroxidase and glutathione content in kidneys of suckling pups and their mothers. Kidney histological studies confirmed biochemical parameters. We concluded that the exposure of rats to GA3 induced oxidative stress and histopathological changes in kidneys of suckling rats and their mothers during late pregnancy and early postnatal periods.  相似文献

Indole-3-acetic acid (IAA) and gibberellic acid (GA3) are essential for the growth and development of plants. In the present study, the ameliorative potential of these phytohormones on growth, protein content, and antioxidant enzymes was investigated in in vitro-grown Solanum tuberosum L. cultivars ‘Cardinal’ and ‘Desiree’ under salt stress. A 4 × 3 factorial combination of 0, 40, 60, or 80 mM NaCl with 0, 7, or 14 μM IAA, or 0, 14, or 21 μM GA3, were added to Murashige and Skoog (MS) basal medium, followed by inoculation of nodal explants or callus cultures. The data for root and shoot number and length, number of nodes and leaves, fresh weight of plants, increase or decrease in fresh weight of callus cultures, total soluble protein, and superoxide dismutase (SOD) and peroxidase (POD) activities were recorded after 30 d. The growth of both callus cultures and nodal explants subjected to NaCl stress was substantially reduced compared with the control. Both IAA and GA3 successfully alleviated the harmful effects of salt stress on all of the growth parameters studied. Salt stress resulted in decreased protein content, which increased when the media also contained phytohormones. The activities of SOD and POD were increased with either IAA or GA3 under NaCl stress. Therefore, the exogenous application of both IAA and GA3 not only played a positive role in terms of in vitro potato growth but also significantly affected the biochemical parameters tested.

Growth regulators, indole-3-acetic acid (IAA), gibberellic acid (GA3), and kinetin (KIN), were used in different concentrations to stimulate the initiation and further development of the fruiting bodies of the fungusLentinus tigrinus. Vegetative mycelium of the fungus was cultivated on cellulose cylinders soaked with a synthetic nutrient solution or with a 3% malt extract. When the mycelium covered the surface of the cylinders, further cultivation was carried out in graduated concentrations of the growth regulators mentioned above. The number of developed fruiting bodies showed that the optimum IAA and GA3 concentrations were in both media 300 p.p.m. The optimum concentration of kinetin was 400 p.p.m. The individual growth regulators influenced characteristically also the shape of the fruiting bodies and changed in them the natural level of endogenous growth regulators. The addition of IAA into the medium raised the level of endogenous auxins in the cap. The presence of gibberellic acid and of kinetic in the medium resulted in an increase in the level of these regulators in the fruiting bodies.  相似文献

Salicylic acid (SA) can alleviate damage to rice plants induced by heat stress, but its role in preventing spikelet degeneration under high-temperature stress has not been documented. Rice plants pretreated with SA (0–50 mmol L?1) were subjected to heat stress at 40?°C at the pollen mother cell meiosis stage for 10 days. The results indicated that there were no significant differences in grain yields and yield components among rice plants that were exogenously sprayed with SA (0–50 mmol L?1) under natural conditions. Under heat stress, the grain yield, spikelet number per panicle and setting rate in response to SA treatments were higher than under the control (0 mmol L?1 SA or NON-SA) treatment, especially with 1 and 10 mmol L?1 SA. A higher grain yield, spikelet number per panicle and setting rate were recorded in these two SA treatments compared with the NON-SA treatment. During this process, soluble sugars, proline, phytohormones including ABA, GA3, BRs, IAA, ZR and JA, and antioxidant enzymes, such as superoxide dismutase, peroxidase (POD), catalase (CAT) and ascorbate peroxidase (APX), were induced by SA. Moreover, soluble sugars, IAA, POD and APX in the spikelets with SA treatments were not only higher than with the NON-SA treatment but the changing patterns were also similar to that of the spikelet number per panicle under natural conditions and heat stress. Therefore, our results suggest that sugars, antioxidant enzymes and IAA might mediate SA to prevent spikelet degeneration caused by heat stress.  相似文献

The effects of indole-3-acetic acid (IAA), abscisic acid (ABA), gibberellic acid (GA3) and kinetin on the hydrolytic activity of proton pumps (adenosine triphosphatase, H+-ATPase, pyrophosphatase, H+-PPase) of tonoplasts isolated from stored red beet (Beta vulgaris L. cv. Bordo) roots were studied. Results suggest that the phytohormones can regulate the hydrolytic activities of H+-ATPase and H+-PPase of the vacuolar membrane. Each of the proton pumps of the tonoplast has its own regulators in spite of similar localization and functions. IAA and kinetin seem to be regulators of the hydrolytic activity for H+-PPase whereas for H+-ATPase it may be GA3. In order to evolve a quick method for smooth and optimum germination for Withania somnifera- a medicinally efficacious multipurpose plant, present investigation was carried to study the effect of physico-chemical treatments, storage, temperature, photoperiod and growth regulators (GA3, IAA, IBA, 2–4 D and BA) on germinability. The most effective treatment is GA3 at 150 μg/ml concentration at 25 °C. The optimal temperature for germination is 25 °C and continuous light favored germination showing that photoperiod has a significant role. The seedlings derived from seeds performed well when grown in a glasshouse. The data have implications for conservation and cultivation of the species studied.  相似文献

The contents of several photosynthetic metabolites — 3-phosphoglyceric acid (3-PGA), pyruvate, nicotinamide adenine dinucleotide phosphate (NADP) and adenosine triphosphate (ATP) — were determined in leaves of cotton plants (Gossypium hirsutum L. cv. H-777) subjected to waterlogging at vegetative stage, and/or drought at the reproductive stage. In controls, soil moisture contents was kept at field capacity. One day prior to stress, the plant shoots were sprayed with 5 M aqueous solution of indole-3-acetic acid (IAA), gibberellic acid (GA3), benzylaminopurine (BAP), abscisic acid, and ethrel. In control plants, various growth regulators reduced contents of 3-PGA and ATP while increased contents of NADP and pyruvate. During waterlogging IAA promoted 3-PGA content, and BAP enhanced pyruvate content. During drought, GA3 enhanced ATP and 3-PGA contents, while IAA enhanced pyruvate content.  相似文献

In the present report, we described the effects of indole-3-acetic acid (IAA), zeatin (ZT), gibberellin (GA3), and abscisic acid (ABA) on in vitro pollen germination and pollen tube growth in Torenia fournieri L. The results showed that IAA and GA3 stimulated in vitro pollen tube growth, ABA inhibited pollen tube growth, and ZT had no significant effect on the process. The stimulating effect of exogenous IAA was particularly distinct, and led to synchronous growth of straighter and more slender pollen tubes compared with the controls. However, no significant changes were found in the germination of the treated pollen. The auxin efflux inhibitor, 10 μM 1-N-naphthylphthalamic acid (NPA), was also found to stimulate pollen tube growth. We measured the content of hormones (free IAA, ZT, GA3, and ABA) in the stigmas and styles before and after pollination. The hormone contents of stigmas measured 0.5 h after pollination (0.5 HAP) showed that ABA content decreased, whereas the content of IAA, ZT, or GA3 did not change significantly. The hormone level in pollinated styles (4 HAP) when pollen tubes had grown into the middle part of style was characterized by an increase in free IAA and GA3 and a decrease in ABA, which was in agreement with the results that IAA and GA3 promoted but ABA inhibited pollen tube growth in vitro. Furthermore, the change of IAA level in styles was most notable, which was accordant to the fact that auxin stimulated significantly pollen tube growth in vitro. Using immunoenzyme and immunogold labeling techniques and an anti-IAA monoclonal antibody, we confirmed that free IAA was present throughout style tissues, and distributed in the nucleus and cytoplasm of style cells. All these results suggested that hormones, especially IAA, play important roles in pollen tube growth of T. fournieri. The antagonistic effects of some growth regulators [i.e. indol-3-yl-acetic acid (IAA), gibberellic acid (GA3) or kinetin] on stress imposed by sea water on leaf area, pigment and photosynthetic activity in leaves of broad bean plants at different stages of development were investigated. Seed priming with GA3 alleviated either partially or completely the effects induced by the two levels of sea water (10 and 25 %) used on leaf area at all experimental stages. However, IAA, GA3 and kinetin inhibited leaf growth by themselves in almost all measurements. Seed pretreatment with kinetin alleviated the inhibition of pigment production in sea water-irrigated plants. Furthermore, GA3 or kinetin nullified the deleterious effects imposed by irrigation with sea water particularly the high level (25 %) on photosynthetic14CO2 fixation.  相似文献

Satsuma [Citrus unshiu (Mak) Marc.] and Clementine [Citrus reticulata (Hort.) Ex. Tanaka, cv. Oroval] are two related species of seedless mandarins which differ in their tendency to set parthenocarpic fruits. Satsuma fruits naturally set parthenocarpically whereas Clementine mandarins show very low ability to set fruit in the absence of cross-pollination. The endogenous levels of gibberellins (GAs) and free and conjugated indole-acetic acid (IAA) and abscisic acid (ABA) throughout early stages of fruit development were investigated in seedless cultivars of both species. Analyses performed by full-scan combined gas chromatography-mass spectrometry (GC-MS) of extracts from ovaries at anthesis demonstrated the presence of GA19, GA20, GA29, GA1, GA8, GA3 and iso-GA3 in Satsuma mandarin, whereas only GA29, GA3 and trace levels of GA8 were detected in Clementine. At this developmental stage GA-like substances, as estimated by bioassay, reached their highest levels in Satsuma, while Clementine mandarins contained relatively lower levels. In both species the highest levels of free IAA were found at petal-fall stage at which time free ABA levels also peaked. Developing fruits of Clementine had higher amounts of both free IAA and ABA. In Satsuma, levels of conjugated IAA remained low throughout reproductive development whereas in Clementine they increased as the free form declined. In contrast, conjugated ABA was at low levels in Clementine but reached higher concentrations in Satsuma. These results suggest that in these mandarins the potential for setting parthenocarpic fruits is mainly influenced by the hormonal status of the fruit during the later stages of cell division and early stages of cell enlargement. Thus, the condition of low ability to set parthenocarpic fruits appears to be associated with lower levels of active GAs, lower capability to catabolize ABA to conjugated ABA and higher ability to conjugate IAA during this period.  相似文献
