The effects of irradiance on the biochemical composition of the prymnesiophyte microalga, Isochrysis sp. (Parke; clone T-ISO), a popular species for mariculture, were examined. Cultures were grown under a 12:12 h light: dark (L:D) regime at five irradiances ranging from 50 to 1000 μE·m 2·s?1 and harvested at late-logarithmic phase for analysis of biochemical composition. Gross composition varied aver the range of irradiances. The highest levels of protein were present in cells from cultures grown at 100 and 250 μE·m 3·s1, and minimum levels of carbohydrate and lipid occurred at 50 μE·m?2·s?1. Because the cell dry weight was reduced at lower irradiances, different trends were evident when results were expressed as percentage of dry weights. Protein percentages were highest at Wand 100 μE·m?2·s?1 and carbohydrate at 100 μE·m?2·s?1. The composition of amino acids did not differ over the range of irradiances. Glutamate and aspartate were always present in high proportions (9.0–13.5%); histidine. methionine, tryptophan, cystine, and hydroxy-proline were minor constituents (0.0–2.6%). Glucose was the predominant sugar in all cultures, ranging from 23.0% (50 μE·m?2·s?1) to 45.0% (100 μE·m?2·s?1) of total polysaccharide. No correlation was found between the proportion of any of the sugars and irradiance. The proportions of the lipid class components and fatty acids showed little change with irradiance. The main fatty acids were 14:0, 16:0, 16:1(n-7), 18:1(n-9), 18:3(n-3). 18:4(n-3), 18:5(n-3), and 22:6(n-3). Proportions of 22: 6(n-3) increased, whereas l8:3(n-3). 18:3(n-6). and 18:4(n-3) decreased, with increasing irradiance. Pigment concentrations were highest in cultures grown at 50 μE·m?2·s?1, except for fucoxanthin and diadinoxanthin (100 μE·m?2·s?1). The concentrations of accessory pigments correlated with chlorophyll a, which decreased in concentration with increasing irradiance. On the basts of biochemical composition, an irradiance of 100 μE·m?1·s?1 (12:12 h L:D cycle)for the culture of Isochrysis sp. (clone T-ISO) may provide optimal nutritional value for maricultured animals, although feeding trials are now necessary to substantiate this.  相似文献

Gametophytes of Macrocystis pyrifera (L.) C. Ag. were cultured under a series of quantum irradiances in three photoperiod regimes. The quantum irradiances in each photoperiod were adjusted to provide equal daily irradiation dosages between photoperiods which allowed a critical examination of the interactions between quantum irradiance and quantum dose in determining gametophyte fertility. The lowest quantum irradiance which stimulated gametogenesis in more than 50% of the female gametophytes was 5 μE·m?2·s?1. The saturating irradiance was ca. 10 μE·m?2·s?1 at photoperiods of 12 h or greater. In terms of daily quantum dose, the lowest dose at which greater than 50% gametogenesis occurred was 0.2 E·m?2·d?1. However, this critical quantum dose was higher (0.4 E·m?2·d?1) when instantaneous irradiances were less than 5 μE·m?2·s?1. The saturation quantum dose was also affected by the rate at which the quantum dose was received and varied from 0.4 to 0.8 E·m?2·d?1. Gametophytes in all three photoperiods reached 100% fertility at quantum irradiances above 5 μE·m?2·s?1. Photoperiod effects were small and could be accounted for by quantum dosage effects.  相似文献

Ulothrix zonata (Weber and Mohr) Kütz. is an unbranched filamentous green alga found in rocky littoral areas of many northern lakes. Field observations of its seasonal and spatial distribution indicated that it should have a low temperature and a high irradiance optimum for net photosynthesis, and at temperatures above 10°C it should show an increasingly unfavorable energy balance. Measurements of net photosynthesis and respiration were made at 56 combinations of light and temperature. Optimum conditions were 5°C and 1100 μE·m?2·s?1 at which net photosynthesis was 16.8 mg O2·g?1·h?1. As temperature increased above 5° C optimum irradiance decreased to 125 μE·m?2·s?1 at 30°C. Respiration rates increased with both temperature and prior irradiance. Light-enhanced respiration rates were significantly greater than dark respiration rates following irradiance exposures of 125 μE·m?2·s?1 or greater. Polynomials were fitted to the data to generate response surfaces. Polynomial equations represent statistical models which can accurately predict photosynthesis and respiration for inclusion in ecosystem models.  相似文献

Laboratory streams were used in a 42-day experiment designed to investigate how the spatial and temporal distribution of lotic periphyton created by current flow over cobble-size substrates is a affected by irradiance. The streams contained 22.5 × 22.5 × 4 cm substrate blocks and were exposed to either 385, 90 or 20 μE·m?2·s?1. We monitored periphyton succession in fast current regimes on top of blocks and in slower current regimes on surfaces recessed between blocks. The absolute differences in AFDW algal biomass between top and recessed substrates were significantly affected by irradiance and time. At the end of the experiment, biomass in streams exposed to 385 μE·m?2·s?1. was approximately 2 and 8 times greater than in streams exposed to 90 and 20 μE·m?2·s?1, respectively. Differences in biomass were greater between irradiance levels than between top and recessed substrates within an irradiance level. Irradiance also had a greater effect than current regime on the taxonomic composition of assemblages. Oscillatoria agardhii Gomont and Navicula minima Grun. characterized assemblages at 20 μE·m?2·s?1, whereas Fragilaria vaucheriae (Kütz.), Nitzschia oregona Sov., Navicula arvensis Hust. and Stigeoclonium tenue (Ag.) Kütz. were more abundant at the two higher irradiances. Detrended correspondence analysis indicated that the rate of succession was relatively high for assemblages at high irradiance and in the slow current regimes between blocks. The results suggested that in natural streams, periphyton patches produced by large differences in irradiance should have a greater effect on periphyton heterogeneity than substrate-induced patches. Moreover, the heterogeneity of algal patches produced by hydrologic differences over a substrate is constrained by irradiance level.  相似文献

Optimum light, temperature, and pH conditions for growth, photosynthetic, and respiratory activities of Peridinium cinctum fa. westii (Lemm.) Lef were investigated by using axenic clones in batch cultures. The results are discussed and compared with data from Lake Kinneret (Israel) where it produces heavy blooms in spring. Highest biomass development and growth rates occurred at ca. 23° C and ≥50 μE· m?2·s1 of fluorescent light with energy peaks at 440–575 and 665 nm. Photosynthetic oxygen release was more efficient in filtered light of blue (BG 12) and red (RG 2) than in green (VG 9) qualities. Photosynthetic oxygen production occurred at temperatures ranging from 5° to 32° C in white fluorescent light from 10 to 105 μE·m?2·s?1 with a gross maximum value of 1500 × 10?12 g·cell?1·h?1 at the highest irradiance. The average respiration amounted to ca. 12% of the gross production and reached a maximum value of ca. 270·10?12 g·cell?1·h?1 at 31° C. A comparison of photosynthetic and respiratory Q10-values showed that in the upper temperature range the increase in gross production was only a third of the corresponding increase in respiration, although the gross production was at maximum. Short intermittent periods of dark (>7 min) before high light exposures from a halogen lamp greatly increased oxygen production. Depending on the physiological status of the alga, light saturation values were reached at 500–1000 μE·m?2·s?1 of halogen light with compensation points at 20–40 μE·m?2·s?1 and Ik-values at 100–200 μE·m?2·s?1. The corresponding values in fluorescent light in which it was cultured and adapted, were 25 to 75% lower indicating the ability of the alga to efficiently utilize varying light conditions, if the adaptation time is sufficient. Carbon fixation was most efficient at ca. pH 7, but the growth rates and biomass development were highest at pH 8.3.  相似文献

Photosynthetic response to high light was determined for Bull kelp, Nereocystis luetkeana (K. Mertens) Postels and Ruprecht in order to understand how this species is affected by short‐term fluctuations in irradiance. Exposure of N. luetkeana blades to high intensity photosynthetically active radiation (1000 µmol photons m?2 s–1) caused increased non‐photochemical quenching of fluorescence and higher de‐epoxidation ratios for xanthophyll pigments indicating that energy‐quenching xanthophylls were used to protect blades against photoinhibition. Despite initiation of these photoprotective mechanisms, maximum photochemical efficiency of photosystem II (Fv/Fm) decreased 40% in response to a 60 min exposure to 1000 µmol photons m?2 s–1 photosynthetically active radiation indicating that photoinhibition had occurred. Light‐saturated rates of oxygen evolution were not changed significantly by the high light treatment. Recovery of maximum photochemical efficiency of photosystem II to within 8% of initial values occurred after a 300‐min dim light period. Younger sections of the blades were slightly more susceptible to high light damage than older sections. Middle sections of the blades were more prone to light‐induced damage at water temperatures of 7°C or 18°C, as compared to 13°C. Exposure to biologically effective ultraviolet‐B radiation (UV‐Bbe) (up to 4.5 kJ m–2 day–1) in photoinhibitory light conditions did not significantly affect light‐induced damage to photosystem II.  相似文献

A dense community of shade adapted microalgae dominated by the diatom Trachyneis aspera is associated with a siliceous sponge spicule mat in McMurdo Sound, Antarctica. Diatoms at a depth of 20 to 30 m were found attached to spicule surfaces and in the interstitial water between spicules. Ambient irradiance was less than 0.6 μE · m?2· s?1 due to light attenuation by surface snow, sea ice, ice algae, and the water column. Photosynthesis-irradiance relationships determined by the uptake of NaH14CO3 revealed that benthic diatoms beneath annual sea ice were light-saturated at only 11 μE·m?2·s?1, putting them among the most shade adapted microalgae reported. Unlike most shade adapted microalgae, however, they were not photoinhibited even at irradiances of 300 μE·m?2·s?1. Although in situ primary production by benthic diatoms was low, it may provide a source of fixed carbon to the abundant benthic invertebrates when phytoplankton or ice algal carbon is unavailable.  相似文献

Under 30-min high irradiance (1500μmol m^-2 s^-1), the roles of the xanthophyll cycle and D1 protein turnover were investigated through chlorophyll fluorescence parameters in morning glory (Ipomoea setosa) leaves, which were dipped into water, dithiothreitol (DTT) and lincomycin (LM), respectively. During the stress, both the xanthophyll cycle and D1 protein turnover could protect PSI from photoinhibition. In DTT leaves, non-photochemical quenching (NPQ) was inhibited greatly and the oxidation level of P700 (P700^+) was the lowest one. However, the maximal photochemical efficiency of PSII (Fv/Fm) in DTT leaves was higher than that of LM leaves and was lower than that of control leaves. These results suggested that PSI was more sensitive to the loss of the xanthophyll cycle than PSII under high irradiance. In LM leaves, NPQ was partly inhibited, Fv/Fm was the lowest one among three treatments under high irradiance and P700^+ was at a similar level as that of control leaves. These results implied that inactivation of PSII reaction centers could protect PSI from further photoinhibition. Additionally, the lowest of the number of active reaction centers to one inactive reaction center for a PSII cross-section (RC/CSo), maximal trapping rate in a PSll cross-section (TRo/CSo), electron transport in a PSll cross-section (ETo/CSo) and the highest of 1-qP in LM leaves further indicated that severe photoinhibition of PSII in LM leaves was mainly induced by inactivation of PSII reaction centers, which limited electrons transporting to PSh However, relative to the LM leaves the higher level of RC/CSo, TRo/CSo, Fv/Fm and the lower level of 1-qP in DTT leaves indicated that PSI photoinhibition was mainly induced by the electron accumulation at the PSI acceptor side, which induced the decrease of P700^+ under high irradiance.  相似文献

Sea ice microalgae in McMurdo Sound, Antarctica were examined for photosynthesis-irradiance relationships and for the extent and time course of their photoadaptation to a reduction in in situ irradiance. Algae were collected from the bottom centimeter of coarse-grained congelation ice in an area free of natural snow cover. Photosynthetic rate was determined in short term (1 h) incubations at ?2° C over a range of irradiance from 0 to 286 μE·m?2·s?1. Assimilation numbers were consistently below 0.1 mg C·mg chl a?1·h?1. The Ik's3 averaged only 7 μE·m?2·s?1, and photosynthesis was inhibited at irradiances above 25 μE·m?2·s?1. Photosynthetic parameters of the ice algal community were examined over a nine day period following the addition of 4 cm of surface snow while a control area remained snow-free. A reduction of 40% in PmB relative to the control occurred after two days of snow cover; α, β, Ik, and Im were not significantly altered. Low assimilation numbers and constant standing crop size, however, suggested that the algal bloom may have already reached stationary growth phase, possibly minimizing their photoadaptive response.  相似文献

The photosynthesis-irradiance dependence of natural phytoplankton assemblages from surface waters of Vineyard Sound, Massachusetts, was investigated over a several month period during late winter —early spring, 1982, when water temperatures were ? 0.5 to 8.5°C. Maximal photosynthetic rates not only were consistently observed between 7–15% I0, but were substantially higher than previously reported rates for cold-water assemblages, averaging 10–20 μg C · μg Chl a?1 · h?1. At higher light intensities photo-inhibition was severe and developed within minutes to tens of minutes of the start of the experiment. Several lines of evidence, however, suggest that photoinhibition in situ may be much less that that measured in incubations of > 30 min duration; residence time of phytoplankton at surface light intensities is sufficiently short, and adaptation of photosynthetic capacity appears to have occurred to approximately the mean depth of the water column. These results further highlight the importance of establishing the time-dependent photosynthetic responses of phytoplankton and the relationship to the physical mixing regime in estimating primary productivity.  相似文献

An inverse linear relationship between chlorophyll fluorescence yield (R) and light intensity was recorded in the near-surface waters of six lakes (New Zealand, England) of greatly different trophic status and phytoplankton species composition. This surface depression of R values could be removed by incubation of samples in dim light or darkness and was not observed in situ below a threshold irradiance (146 μEin ·m?2·s?1 for Lake Taupo, New Zealand). The time course of chlorophyll fluorescence depression and recovery in response to light treatment was measured in samples from Lake Windermere (England). Fluorescence exponentially decreased upon exposure to bright light and the response was 100% (5 m samples) or 83% (dim light-adapted 0 m samples) complete within 2 min. An increase in R values in the dim light occurred after a lag of 60 s and the rate of increase decreased exponentially with time. Full recovery took 15 min or more. Deep (6.5 m) populations from Lake Windermere exhibited large, time-dependent variations in chlorophyll fluorescence over the first 25 s of exposure to 450 nm light, whereas surface populations did not. These data were interpreted in terms of decreased spillover from PSII to PSI with increasing depth, to a minimum at the threshold light intensity below which cells are in light state 1.  相似文献

Synechococcus sp. (PTCC 6021), a cyanobacterium species, was cultivated in an internally illuminated photobioreactor. The reactor was designed to achieve a monoseptic cultivation of the species. The goal was to study the growth–irradiance behavior of Synechococcus sp. (PTCC 6021). To accomplish this, different initial light irradiances were implemented inside the photobioreactor and the growth of the cells was monitored. It was observed that cell growth increased with higher light intensity until the photoinhibition occurrence at light irradiance higher than 250?μE?m?2?s?1. The maximum OD600, maximum growth rate, and biomass productivity increased, and hence the extinction coefficient decreased, with the increase in light irradiance before photoinhibition. The maximum optical density (OD600) of 5.91 was obtained with irradiance below 250?μE?m?2?s?1 during a growth period of 80 days. The modified Monod function could model the growth–irradiance of cells with satisfactory agreement with the experimental data. The comparison of growth–irradiance of the studied species with other photosynthetic organisms showed the same trend as for cyanobacteria with photoinhibition.  相似文献

Photosynthesis-irradiance relationships were determined in the field for five species of littoral and shallow sublittoral marine benthic green algae (Chlorophyta) of differing morphologies. Each species exhibited a linear increase in photosynthetic rate with increasing irradiance up to a maximum light-saturated value. Full sunlight (1405 to 1956 μE·m?2·s?1) inhibited photosynthesis of all species except the thick, optically dense, Codium fragile (Sur.) Har. Compensation irradiances ranged from 6.1 μE·m?2·s?1 for Enteromorpha intestinalis (L.) Link to 11.4 μE·m?2·s?1 for Ulva lobata (Kütz) S. & G. and did not reveal a consistent relationship to seaweed morphology. Saturation irradiances were determined statistically (Ik) and visually from graphical plots. with the latter technique resulting in values three to eight times higher and different comparative rankings of species than the former. Ik saturation irradiances were highest for Chaetomorpha linum (Müll.) Kütz. (81.9 μE·m?2·s?1) and lowest for Codium fragile (49.6 μE·m?2·s?1) and did not reveal a relationship with seaweed morphology. Regression equations describing light-limited photosynthetic rates and the relative magnitudes of the maximal net photosynthetic responses both strongly suggested a relationship with seaweed morphology. Highest net photosynthetic rates were obtained for the thin, sheet-like algae Ulva lobata (9.2 mg C·g dry wt?1·h?1), U. rigida C. Ag. (6.5 mg C·g dry wt?1·h?1) and the tubular form, Enteromorpha intestinalis (7.3 mg C·g dry wt?1·h?1), while lowest rates occurred for Codium fragile (0.9 mg C·g dry wt?1·h?1). Similarly, steepest light-limited slopes were found for the algae of simpler morphology, while the most gradual slope was determined for Codium fragile, the alga with greatest thallus complexity.

Ice algae are a case study in photo-autotrophic growth and metabolism under chronically low temperature and irradiance. We measured the allocation of 14C-labelled photosynthate among major classes of intracellular carbon (low molecular weight compounds, or LMW; lipid; protein; and polysaccharide) and found light-dependent changes in allocation broadly similar to photo-adaptive responses known in phytoplankton at higher temperatures; average relative allocation to protein varied inversely (10–37%) and allocation to lipids and polysaccharides directly (10–23%, and 16–21%, respectively) with the sub-inhibiting irradiance levels we employed (3.5–33.0 μE·M−2·s−1). Unlike many observations at higher temperatures, ours indicated (on average) a large and light-insensitive allocation to LMW (ca. 40%) and a greater light-sensitivity in lipid than in polysaccharide allocation. At the higher incubation irradiances, resembling in situ levels typical of areas with little (0–5 cm) snow cover, allocation to protein was often low (10–13%) compared to many observations of nutrient-sufficient or light-limited phytoplankton. Allocation to protein increased substantially (to ca. 40%) during a period of intensified tidal mixing, and assimilation numbers also attained a maximum at about the same time. Suck dynamics show that the ice algae are not constrained to their often protein-poor allocation by the constantly low ambient temperature. Rather, they display marked shifts in metabolism consistent with major changes in light and inorganic nutrient supply, driven in part by the physical process of tidal mixing.  相似文献

In many lakes in the northern United States and Canada the filamentous green alga Ulothrix zonata (Weber and Mohr) Kütz grows abundantly in early spring in shallow waters. Asexual reproduction occurs by formation of quadriflagellate zoospores which disrupt, the integrity of the cells upon release causing the filament to disintegrate. Study of the effects of 100 different combinations of irradiance, temperature and photoperiod revealed that zoospore formation is favored by high temperatures near 20°C, high light levels of 520 μE·m?2·s?1 and photoperiods of either short day (8:16 h light-dark) or long day cycles (16:8 h light-dark). Zoospore formation is minimal under conditions of low temperature (5°C), low irradiance (32.5 μE·m?2·s?1) and neutral day-lengths (12:12 h light-dark). These observations explain the decline in U. zonata biomass when water temperatures rise above 10° C. The combined effect of rising water temperatures and increasing daylengths causes progressively more filaments to switch from vegetable growth to zoospore production resulting in an increasing loss of biomass.  相似文献

The maximum quantum yield of photosystem II was estimated from variable chlorophyll a fluorescence in samples of phytoplankton collected from the Taihu Lake in China to determine the responses of different phytoplankters to irradiance and vertical mixing. Meteorological and environmental variables were also monitored synchronously. The maximum quantum yield of three phytoplankton groups: cyanobacteria, chlorophytes, and diatoms/dinoflagellates, showed a similar diurnal change pattern. F v/F m decreased with a significant depth-dependent variation as irradiance increased during the morning and increased as irradiance declined in the afternoon. Furthermore, the rates of F v/F m depression were dependent upon the photon flux density, whereas the rates of recovery of F v/F m were dependent upon the historical photon density. Moreover, photoinhibition affected the instantaneous growth rates of phytoplankton. Although at noon cyanobacteria had a higher photoinhibition value (up to 41%) than chlorophytes (32%) and diatoms/dinoflagellates (34%) at the surface, no significant difference in diurnal growth rates among the three phytoplankton groups were observed indicating that cyanobacteria could photoacclimate better than chlorophytes and diatoms/dinoflagellates. In addition, cyanobacteria had a higher nonphotochemical quenching value than chlorophytes and diatoms/dinoflagellates at the surface at noon, which indicated that cyanobacteria were better at dissipating excess energy. The ratios of enclosed bottle samples F v/F m to free lake samples F v/F m showed different responses for the three phytoplankton groups to irradiance and vertical mixing when wind speed was approximately constant at about 3.0 m s−1. When wind speed was lower than 3.0 m s−1, cyanobacteria accumulated mainly at the surface and 0.3 m, because of their positive buoyancy, where diurnal growth rates of phytoplankton were relatively higher than those at 0.6 m and 0.9 m. Chlorophytes were homogenized completely by vertical mixing, while diatoms/dinoflagellates avoided active high irradiance by moving downward at noon, and then upward again when irradiance decreased. These results explain the dominance of cyanobacteria in Taihu Lake. Handling editor: L. Naselli-Flores  相似文献

The effect of different irradiance levels on the interactions between the algal host and its virus was investigated for two marine phytoplankton, Phaeocystis globosa Scherff. and Micromonas pusilla (Butcher) Manton et Parke. The algal cultures were acclimated at 25, 100, and 250 μmol photons · m?2 · s?1 (LL, ML, and HL, respectively), after which they were infected with a lytic virus (PgV‐07T and MpV‐02T) and monitored under the appropriate irradiance and in darkness. The effect of irradiance levels on the host–virus interactions differed for the two algal host–virus systems examined. For P. globosa, the LL‐acclimated cultures showed a 4 h prolonged latent period (11–16 h), which may be related to the subsaturated growth observed at this irradiance. The burst size was reduced by 50% at LL and HL compared to ML (525 PgV · cell?1). The fraction of infectious viruses, however, remained unchanged. Viral replication was prevented when the LL P. globosa cultures were kept in darkness (up to 48 h) but recovered when placed back into the light. PgV‐07T still replicated in the dark for the ML‐ and HL‐acclimated cultures, but viral yield was reduced by 50%–85%. For M. pusilla, the burst size (285–360 MpV · cell?1), the infectivity, and the latent period of MpV‐02T (7–11 h) remained unaffected by the incident light. Conversely, darkness not only inhibited MpV replication but also resulted in substantial cell lysis of the noninfected cultures. Our study implies that irradiance level is an important factor controlling algal host–virus interactions and hence the dynamics of phytoplankton populations.  相似文献

Photosynthetic parameters were determined in disks from leaves of C. arabica cv. Red Catuaí and C. canephora cv. Kouillou grown in the field. Kouillou showed a relatively higher irradiance requirement for saturating photosynthesis, lower chlorophyll (Chl) content, and higher Chl a/b ratio than Catuaí. Photoinhibition of photosynthesis under bright irradiance was manifested by decreases in maximum photochemical efficiency (evaluated by the variable to maximum fluorescence ratio, Fv/Fm), as a consequence of an increased initial and a quenched maximum fluorescence. Restoration of Fv/Fm following photoinhibition in low irradiance was faster in Kouillou than in Catuaí. Chloramphenicol both accelerated photoinhibition (mainly in Kouillou) and blocked its recovery for at least 190 min in either cultivar. Photosynthetic oxygen evolution under photoinhibitory conditions was decreased by chloramphenicol; in control leaf disks this decrease was only observed in C. arabica, but with a rapid recovery within 90 min of low irradiance exposure. In both coffee cultivars, the depressed photochemical efficiency of photosystem 2 was not accompanied by a concomitant lowering in oxygen evolution during reversal from photoinhibition.  相似文献
