Evidence is presented for the existence of discrete, naturalcuticular pores concomitant with anticlinally-oriented transcuticularcanals found in the mature leaf surfaces of 26 out of 37 taxaamong 19 families. This investigation is an extension of earlierobservations made on 32 other taxa among 14 families. Dewaxed,chemically isolated, adaxial and abaxial cuticular membranesin conjunction with transverse leaf sections were examined usingordinary staining techniques. The ubiquitous pores occur randomlywith no evidence of clustering. Pore and canal diameters averageapprox. 1 µm. Canal lengths are directly related to cuticlethickness. No correlations were found between cuticle thicknessesand either pore frequencies or pore and canal diameters. Evidenceis provided by light microscopy photomicrographs. Developmental studies were made on the dewaxed thin-sectionedapple fruit cuticles of 10 Malus sylvestris Mill, cultivarsfrom 4 weeks before anthesis through fruit maturation and harvest.Cuticular development appears to correlate well with the generalgrowth of the fruit. However, no correlation exists betweenfruit size and cuticle thickness. Cuticular pores were evidentas early as 1 week before anthesis and transcuticular canalsbecame evident by 1 week following anthesis. Dewaxed thin-sectioned,as well as isolated, mature fruit cuticles of 16 cultivars andfour crab-apples consistently revealed the distinct presenceof ubiquitous pores and canals. Evidence is provided by lightmicroscopy and scanning electron microscopy (SEM). Measurementswere taken of cuticle thicknesses during development and ofcuticular pore dimensions, and calculations were made of poreand canal numbers. Fruit size alone is not directly indicativeof total pore numbers per surface area. Canal lengths are directlyrelated to the developmental thickness of the cuticle. No correlationwas found between the thickness of the mature cuticle and eitherthe number of pores present or the pore diameters. Dewaxed thin-sectioned and dewaxed isolated mature fruit cuticlesrevealed the unequivocal presence in situ of visibly discrete,ubiquitous, cuticular pores or orifices concomitant with anticlinally-orientedtranscuticular canals in 51 varieties of fruit among 20 plantfamilies. More than 66 per cent of the fruit cuticles have poresand/or canals. No correlation exists between either fruit sizeor pore size and cuticle thickness. The cuticular conductance (gc) of the astomatous adaxial surfaceof Fagus sylvatica L. leaves was determined under varying vapourpressure deficits at a constant temperature of 20 °C. Cuticularconductance was determined from the weight loss of detachedleaves after both the stomatous abaxial surface and the cutend of the petiole had been sealed using low melting-point paraffinwax. Cuticular conductance was found to decrease as the watervapour pressure was increased in steps. No response was observedwhen vapour pressure deficit was decreased from an initiallyhigh value. It is concluded that these results are consistentwith the hypothesis that cuticular conductance is influencedby the water content of the cuticle and that the apparent hysteresisis likely to be a result of the long time-constant for the processof cuticle rehydration in intact leaves. Cuticular conductance of adaxial (astomatous) and abaxial (stomatous)surfaces ofFagus sylvatica L. leaves was measured under varyingvapour pressure deficits (D). Conductance was determined fromgravimetric measurements of water flux made using a leaf discenvelope specially designed to maintain leaf relative watercontent and minimize reduction in cuticular hydration. The adaxialsurface provided a determination of ‘true’ cuticularconductance (gc) and transpiration (Ec). The abaxial surfacewas used to estimate minimum leaf surface conductance (gMINsur)and transpiration (EMINsur). In experiment I, leaf discs wereplaced under one of a range of water vapour pressure deficits(0.4-2.0 kPa). Both gc and gMINsur decreased approximately 2-foldwith an increase in D between 0.4-2.0 kPa. The decrease in gcwas linear, but gMINsur declined more steeply at D between 0.4-0.95kPa than at D between 0.95-2.0 kPa. In experiment II, leaf discswere exposed to a stepwise change in D. After a period of acclimationto D of 0.95 kPa, responses of gc and gMINsur to an increaseor decrease in D were recorded. The response time of gc to increasingor decreasing D were similar (<60 min). By contrast, gMINsurresponded more slowly to increasing than to decreasing D. Thesesignificant responses of gc and gMINsur to increasing and decreasingD are discussed in relation to hydration state of the cuticleand current knowledge of cuticle structure. In order to determine the authenticity of the multiple epidermallayers in the pomaceous fruit of Medlar or Mespil (Mespilusgermanica L.), developmental studies were made from 2 weeksbefore anthesis through harvest and storage. Various histochemicaltechniques were used for the determination of epidermal cellwall structures and cell contents, and for the isolation andstudy of the cuticular membranes. The multiseriate epidermisis derived from successive tangential divisions of the initiallyuniseriate epidermis commencing about 3 weeks post-anthesis.The divisions occur simultaneously around the fruit circumferenceand result in the formation of a normally four- to five-layeredepidermis at fruit maturity. As each epidermal layer is derivedit concomitantly develops a distinct, flanged cuticle that remainspersistent until sloughing of the layer occurs. Evidence providedby light and scanning electron microscopy indicates the presenceof discrete ubiquitous pores and non-anastomosing, anticlinallyoriented canals in the cuticular membranes. Measurements weremade of individual cuticular membrane thicknesses during development,and of cuticular pore dimensions in the mature fruit; cuticularpore numbers were calculated. In flowers of Hoya carnosa R. Br. the nocturnal emission of fragrance occurs according to an endogenous circadian rhythmicity. This was demonstrated by the continuation of rhythmical emission for two to three cycles under conditions of permanent illumination. The free-running period was approx. 29 h under these conditions. The inversion of light/dark cycles caused the entrainment of the next peak of emission by approx. 12 h whether the inversion was started with a light period or a with dark period prolonged for 12 h. When only the flower used for fragrance analysis was subjected to an inverted photoperiod, the rest of the plant remaining under the original light/dark cycle, the synchronization of fragrance emission occurred according to the Zeitgeber perceived by the individual flower.  相似文献

Changes in nuclear DNA content and cell size of adaxial andabaxial epidermal pavement cells were investigated using brightlight-induced leaf expansion of Phaseolus vulgaris plants. Inprimary leaves of bean plants grown under high (sunlight) ormoderate (ML; photon flux density, 163 µmol m–2s–1) light, most adaxial epidermal pavement cells hada nucleus with the 4C amount of DNA, whereas most abaxial pavementcells had a 2C nucleus. In contrast, plants grown under lowintensity white light (LL; 15 µmol m–2 s–1)for 13 d, when cell proliferation of epidermal pavement cellshad already finished, had a 2C nuclear DNA content in most adaxialpavement cells. When these LL-grown plants were transferredto ML, the increase in irradiance raised the frequency of 4Cnuclei in adaxial but not in abaxial pavement cells within 4d. On the other hand, the size of abaxial pavement cells increasedby 53% within 4 d of transfer to ML and remained unchanged thereafter,whereas adaxial pavement cells continuously enlarged for 12d. This suggests that the increase in adaxial cell size after4 d is supported by the nuclear DNA doubling. The differentresponses between adaxial and abaxial epidermal cells were notinduced by the different light intensity at both surfaces. Itwas shown that adaxial epidermal cells have a different propertythan abaxial ones. Structure and function of the inflorescence of cv. 'Hass' and'Fuerte' avocado (Persea americana Mill.) were examined by scanningelectron microcopy (SEM) and by porometry. Sepals and petalscould not be distinguished by their position in the flower,by visual gross morphology or by microscopic surface structureand were hence designated as tepals. These tepals were arrangedin two whorls of three, followed by two whorls of three outerand three inner stamens, each opposite a tepal. The most conspicuousfeature of tepals, developing leaves and peduncles was the densecover of hair which were most frequent on the adaxial tepalsurface (925-1200 trichomes mm-2), followed by their abaxialsurface (625-1000 mm-2) and peduncles (375-655 mm-2). Stomatawere absent from the adaxial surfaces of both tepal and leaves,as well as peduncles. On the tepals, abaxial stomata appearedfunctional, small (8-9 x 11-13 µm) and scarce with 2·8-3·4stomata mm-2, i.e. very low relative to avocado leaves (350-510stomata mm-2) or young fruit (50-75 stomata mm-2. However, flowersincluding tepals transpired 1·2-1·3 mmol underfield conditions in Southern California (1·6-2 kPa),i.e. in excess of leaves (0·7-1·1 mmol) and peduncles(0·6-0·8 mmolH2O m-2 s-1). Two common tallgrass prairie species, Andropogon gerardii, thedominant C4 grass in this North American grassland, and Salviapitcheri, a C3 forb, were exposed to ambient and elevated (twiceambient) CO2 within open-top chambers throughout the 1993 growingseason. After full canopy development, stomatal density on abaxialand adaxial surfaces, guard cell length and specific leaf mass(SLM; mg cm-2) were determined for plants in the chambers aswell as in adjacent unchambered plots. Record high rainfallamounts during the 1993 growing season minimized water stressin these plants (leaf xylem pressure potential was usually >-1·5 MPa in A. gerardii) and also minimized differencesin water status among treatments. In A. gerardii, stomatal densitywas significantly higher (190 ± 7 mm-2; mean ±s.e.) in plants grown outside of the chambers compared to plantsthat developed inside the ambient CO2 chambers (161 ±5 mm-2). Thus, there was a significant 'chamber effect' on stomataldensity. At elevated levels of CO2, stomatal density was evenlower (P < 0·05; 121 ± 5 mm-2). Most stomatawere on abaxial leaf surfaces in this grass, but the ratio ofadaxial to abaxial stomatal density was greater at elevatedlevels of CO2. In S. pitcheri, stomatal density was also significantlylower when plants were grown in the open-top chambers (235 ±10 mm-2 outside vs. 140 ± 6 mm-2 in the ambient CO2 chamber).However, stomatal density was greater at elevated CO2 (218 ±12 mm-2) compared to plants from the ambient CO2 chamber. Theratio of stomata on adaxial vs. abaxial surfaces did not varysignificantly in this herb. Guard cell lengths were not significantlyaffected by growth in the chambers or by elevated CO2 for eitherspecies. Growth within the chambers resulted in lower SLM inS. pitcheri, but CO2 concentration had no effect. In A. gerardii,SLM was lower at elevated CO2. Stomata and Structure of Tetraploid Apple Leaves cultured in Vitro   总被引:2,自引:0,他引:2  
Leaves of anther-derived tetraploid apple (Malus pumila Mill.)shoots were examined by low-temperature scanning electron microscopy(LT-SEM). Leaves were serrate and wide with an undulating adaxialsurface due to convex epidermal cells, apparently without crystallineepicuticular wax. Stomata were absent from the adaxial surface,except for the marginal teeth which exhibited 40-60 stomataper leaf; they probably originated from residual mitotic activity.One third of abaxial stomata was occluded by the residual cuticleof the mother guard cell across the stomatal pore which rupturedwhen the stomata became functional. The stomatal index was 7·2(± 1·6) with 60-75 stomata mm-2, i.e. abaxialstomata of tetraploid leaves expanded in vitro were less frequentthan those in triploid leaves either cultured in vitro (475-575stomata mm-2) or grown on the tree (320-390 stomata mm-2) wherethe stomatal index was 21 (± 4). Freeze-fracture transsectionsshowed that the tetraploid in vitro leaves were composed ofa layer of adaxial epidermal cells, followed by a single layerof palisade cells and four to five layers of spongy mesophyllcells and the abaxial layer of epidermal cells, in contrastto juvenile seedling-grown apple leaves in which the two layersof palisade cells comprised the majority (52-60%) of the leafvolume. The same morphological features, such as wide and lesspointed leaves, reduced stomatal density and stomatal index,and increased stomatal size that were previously reported fortree-grown tetraploid leaves were also expressed in vitro. Foliar cuticle of a new species of Ruflorinia, R. orlandoi Carrizo et Del Fueyo, from the Lower Cretaceous Springhill Formation in Santa Cruz province, Argentina, is herein described with light and electronic (scanning and transmission) microscopy. The fronds of R. orlandoi nov. sp. are bipinnate and imparipinnate with a conspicuous main rachis bearing alternate to sub-opposite pinnae. Cuticular striations and hairs are present in both epidermises whereas stomata are only on the abaxial cuticle. Stomata have several papillate subsidiary cells and the guard cells are sunken. Ultrastructurally the adaxial and abaxial cuticles are composed of three and two cuticular layers, respectively. The cuticular characters here described are interpreted as a response to a possible adaptation to a wet climate as well as to the fluctuating environment produced by the gradual transition from fluvial to an estuarine and marine setting that characterizes the Springhill Formation. The discovery of this new species allows expanding the previous knowledge of the paleofloristic composition of the Springhill Formation with the addition of the first pteridospermous megafossil described for this formation. Moreover, the record of Ruflorinia orlandoi nov. sp. in the Springhill Formation highlights the importance of the Caytoniales during the Cretaceous in Patagonia.  相似文献

The cuticle micromorphology of the leaves of the monospecific genus Saxegothaea (Podocarpaceae) was studied by scanning electron microscopy. The external and internal features of the adaxial and abaxial surfaces were characterized. The leaves are hypostomatic. The external adaxial cuticle is rugose with irregular ridges and shallow trench‐like structures that do not correspond to any feature of the inner cuticle surface. The external abaxial cuticle has densely crowded stomata arranged in two bands. The stomata are sunken with pronounced, interrupted Florin rings. Stomatal plugs were not observed. Internally, the adaxial epidermal cells are usually rectangular to square; the abaxial epidermal cells are mainly restricted to the midrib and margins and narrowly rectangular; any among the stomata are irregularly shaped. The stomata are nearly all in direct contact. They show unusual features, including an extra pair of cuticular flanges between the guard cell flanges and those of the lateral subsidiary cells, and ‘bridges’ of lateral subsidiary cell tissue extending polewards above the polar extensions to unite with those at their tips. Neither of these features has been reported previously in Podocarpaceae. The results are discussed in the light of recent phylogenetic studies. The cuticular waxes on the leaves of Prunus laurocerasus are arranged in distinct layers differing in triterpenoid concentrations (Jetter et al., Plant Cell Environ 23:619–628, 2000). In addition to this transversal gradient, the lateral distribution of cuticular triterpenoids must be investigated to fully describe the spatial distribution of wax components on the leaf surfaces. In the present investigation, near infrared (NIR) Raman microspectroscopy, coherent anti-Stokes Raman scattering (CARS) microscopy, and third harmonic generation (THG) spectroscopy were employed to map the triterpenoid distribution in isolated cuticles from adaxial and abaxial sides of P. laurocerasus leaves. The relative concentrations of ursolic acid and oleanolic acid were calculated by treating the cuticle spectra as linear combinations of reference spectra from the major compounds found in the wax. Raman maps of the adaxial cuticle showed that the triterpenoids accumulate to relatively high concentrations over the periclinal regions of the pavement cells, while the very long chain aliphatic wax constituents are distributed fairly evenly across the entire adaxial cuticle. In the analysis of the abaxial cuticles, the triterpenoids were found to accumulate in greater amounts over the guard cells relative to the pavement cells. The very long chain aliphatic compounds accumulated in the cuticle above the anticlinal cell walls of the pavement cells, and were found at low concentrations above the periclinals and the guard cells.  相似文献

Cuticular transport properties of intact leaves, isolated cuticularmembranes and reconstituted cuticular waxes of the three treespecies Prunus laurocerasus L., Ginkgo biloba L. and Juglansregia L. were measured using six different 14C-labelled compounds,benzoic acid, salicylic acid, 2,4-dichlorophenoxy acid, metribuzin,4-nitrophenol, and atrazine. For the same compound and the samespecies, the permeance of the intact leaf and the isolated cuticlewas equal. This provides strong evidence demonstrating thattransport properties of cuticles are not altered during isolation.Additionally, diffusion coefficients of the 14C-labelled compoundsin isolated and subsequently reconstituted cuticular wax ofthe three tree species were measured. Permeances of intact leavesand isolated cuticles could be predicted from diffusion coefficients,wax/water partition coefficients and the thickness of the transport-limitingwax layer with a mean deviation of about 1.7. This providesevidence that transport properties of recrystallized cuticularwaxes do indeed reflect barrier properties of isolated cuticularmembranes and intact leaves with in situ waxes. Thus, it canbe concluded that the investigation of cuticular permeabilityusing the three independent experimental systems of differentcomplexity give comparable results. Finally, it was observedthat permeances and diffusion coefficients measured with P.laurocerasus were always significantly lower than those measuredwith G. biloba and J. regia. This is interpreted as an ecologicaladaptation of the respective species. The evergreen speciesP. laurocerasus must be more adapted to environmental stresssuch as drought and frost injury compared to the two deciduousspecies G. biloba and J. regia. The cheirolepidiaceous conifer Pseudofrenelopsis dalatzensis from the Lower Cretaceous Dalazi Formation, Jilin Province, Northeast China is studied in detail based on abundant material. The gross morphology of vegetative shoots shows a greater variation than previously known. Additional information of microstructure is that the adaxial cuticle from the free part of leaves is also papillate as the abaxial cuticle. The excellent preservation of cuticles allows for the first time in this family to four distinct types of cuticle in ultrastructural level: subsidiary cells, guard cells, epidermal cells with and without papillae. Based on 30 measurements the statistic approach on each type of cell cuticle, i.e. not only the total thickness of the cuticle but also details and proportions of all different layers. A key is provided using a combination of 7 significant ultrastructural characters. These results seem to be very promising for the identity of each taxon within the Cheirolepidiaceae family. The cuticular ultrastructure of this family is characterized by having a cuticle proper A composed of A1 wavy polylamellate layer and A2 granular layer, above the cuticular layer B with B1 fibrillate and B2 granular layers. The importance of ultrastructural characters at different levels of taxonomy and palaeoecology is also discussed.  相似文献

Cuticular water permeabilities of adaxial and abaxial leaf surfaces and their dependence on relative air humidity (RH) applied in long-term and short-term regimes have been analysed for Hedera helix, native in a temperate climate, and Zamioculcas zamiifolia, native in subtropical regions. The water permeability of cuticular membranes (CM) isolated from the adaxial (astomatous) and abaxial (stomatous) leaf sides was measured using a method which allowed the separation of water diffusion through the remnants of the original stomatal pores from water diffusion through the solid cuticle. The long-term effects of low (20-40%) or high (60-80%) RH applied during plant growth and leaf ontogeny ('growth RH') and the short-term effects of applying 2% or 100% RH while measuring permeability ('measurement RH') were investigated. With both species, water permeability of the solid stomatous CM was significantly higher than the permeability of the astomatous CM. Adaxial cuticles of plants grown in humid air were more permeable to water than those from dry air. The adaxial CM of the drought-tolerant H. helix was more permeable and more sensitive to growth RH than the adaxial CM of Z. zamiifolia, a species avoiding water stress. However, permeability of the solid abaxial CM was similar in both species and independent of growth RH. The lack of a humidity response in the abaxial CM is attributed to a higher degree of cuticular hydration resulting from stomatal transpiration. The ecophysiological significance of higher permeability of the solid stomatous CM compared to the astomatous CM is discussed.  相似文献
