The aim of this study was to assess the grazing, social and comfort behaviour of the indigenous purebred Ankole cattle breed and crossbred (Holstein × Ankole) animals under typical management conditions in south western Uganda. Twelve focal animals in each of four groups (two groups per genotype) were observed regarding their grazing, social and comfort behaviour on pasture.No significant differences in grazing behaviour patterns (eating, walking, standing) were found between the genotypes. Resting occurred only very rarely in both genotypes. Walking distances of Ankole and Ankole × Holstein crosses were also similar. There was no difference in the occurrence of agonistic interactions between the two genotypes. However, Ankole cattle engaged in more non-agonistic social interactions than their crossbred counterparts. Individual distances were lower in Ankole heifers and more herd mates were found within a radius of 5 m around the Ankole animals. The most important comfort behaviour pattern in both genotypes was self-licking, which occurred to similar frequency in Ankole and crossbred heifer groups. Crossbred animals scratched themselves and rubbed on objects more often than Ankole heifers.Although Ankole cattle and their Holstein crosses did not differ in grazing, distances walked and agonistic behaviours, the significant differences between the two genotypes in herd cohesion and comfort behaviour may pose challenges on the management of crossbred animals under extensive open grazing conditions as present in south western Uganda. Thus, apart from (re)productive performance traits, behavioural traits of both genotypes may also be taken into account for breeding decisions and management under current production conditions.  相似文献

The effect on arachidonate metabolism of two compounds (BW755C and benoxaprofen) which have been reported to inhibit 5′ lipoxygenase in leukocytes has been evaluated in human polymorphonuclear leukocytes (PMN) stimulated with the calcium ionophore A23187 and serum-treated zymosan (STZ). The syntheses of leukotriene B4 (LTB4) and thromboxane B2 (TXB2) from endogenous substrate were determined by specific radioimmunoassays as indicators of 5′ lipoxygenase and cyclo-oxygenase activity in the PMN respectively. Benoxaprofen inhibited the synthesis of leukotriene B4 by human PMN stimulated with the calcium ionophore A23187, but it was approximately 5 times less potent than BW755C. However, benoxaprofen (IC50 1.6 × 10−4M) was approximately 100 times less potent than BW755C (IC50 1.7 × 10−6M) at inhibiting leukotriene B4 synthesis induced by serum-treated zymosan. Both drugs inhibited thromboxane synthesis by leukocytes stimulated with A23187 or serum-treated zymosan at similar concentrations (approximately 5 × 10−6M). The data obtained using STZ as stimulus are consistent with previous studies and indicate that benoxaprofen is a relatively selective inhibitor of cylco-oxygenase. However, this selectivity was far less apparent when A23187 was used as a stimulus to release the eicosanoids which suggests that this inhibition could be via an indirect mechanism and therefore A23187 should be used with caution as a stimulus of 5′ lipoxygenase for evaluating inhibitors of eicosanoid synthesis.  相似文献

Studies were undertaken to identify cell surface markers specific for different phases of the cell cycle. Antisera were prepared in rabbits against membrane protein preparations from synchronized BW 5147 cells, an AKR mouse T-lymphoma cell line, in the G1, S, G2 or M phases of the cell cycle. These antisera were used to precipitate radioiodinated surface proteins from synchronized cells in the different phases. The immunoprecipitates were quantitatively analyzed by sodiumdodecylsulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Cells in S phase had significantly higher concentrations of proteins weighing 70 × 103 and 165 × 103 D than cells in G1 or G2 phase. The other major labeled surface components did not vary. These results were confirmed by quantitative absorption of the antisera with synchronized cells. Comparative analysis of the antisera showed that the 165 × 103 D peak contained at least two antigens, one recognized by both a-G1 and a-S and the other by a-G1 only. Though cells in S phase had large quantities of the 70 × 103 D protein, intact and SDS-solubilized membrane preparations from S phase could not elicit in rabbits any antibody against that protein. These antisera did, however, have good antibody titers to the other major protein peaks and the antisera developed against cells in G1, G2 or M had good anti-70 × 103 activity. The results suggest a qualitative molecular change in the 70 × 103 protein during S phase.  相似文献

Prolactin plays an important regulatory function in mammary gland development, milk secretion, and expression of milk protein genes. Hence the PRL gene is a potential quantitative trait locus and genetic marker of production traits in dairy cattle. We analysed the sequence of the PRL gene to investigate whether mutations in this sequence might be responsible for quantitative variations in milk yield and composition. Using SSCP and direct sequencing, we detected six single-nucleotide polymorphisms within a 294-bp prolactin gene fragment involving exon 4. All detected mutations were silent with respect to the amino acid sequence of the protein. PCR-RFLP genotyping of SNP 8398 R (RsaI) was used to assess allele frequencies in 186 Black-and-White cows (0.113 and 0.887 for A and G, respectively) and in 138 Jersey cows (0.706 and 0.294 for A and G, respectively). Black-and-White cows with genotype AG showed the highest milk yield, while cows with genotype GG showed the highest fat content.  相似文献

Low pasture allowance during gestation affects ewes’ BW at parturition, the bond with their lamb, lamb development, and thus also may affect their responses to weaning. The objectives were to determine if native pasture allowance from before conception until late pregnancy affects ewe–lamb behaviours at lambing, ewes’ milk yield, lambs’ BW, and the behavioural and physiological changes of ewes and lambs at weaning. From 23 days before conception until 122 days of pregnancy, 24 ewes grazed on two different native pasture allowances: high (10 to 12 kg of dry matter (DM)/100 kg of BW per day; HPA treatment; n=12) or low (5 to 8 kg of DM/100 kg of BW per day; LPA treatment; n=12). Thereafter, all ewes grazed on Festuca arundinacea and received rice bran and crude glycerine. Ewes’ body condition score (BCS) and BW were recorded during pregnancy and postpartum periods. Milk yield was determined on days 32, 41 and 54 after lambing. Lambs’ BW was recorded from birth until 72 days after lambing. Latency from parturition until the ewe licked her lamb, maternal behaviour score (a test that evaluates maternal attachment to the lamb) and latency for lamb to stand up and suckle were determined. The behaviour of the lambs and ewes was recorded before and after weaning (at 65 days). The ewes’ serum total protein, albumin and globulin concentrations were measured before and after weaning. The HPA ewes presented greater BW (P<0.005) and BCS (P<0.005) than the LPA ewes during pregnancy and postpartum (P<0.04), and had a greater milk yield than the LPA ewes (P<0.03). Treatments did not influence any behaviour at lambing, lambs’ BW, neither the ewes’ behavioural and physiological changes at weaning. HPA lambs paced and vocalized more than LPA lambs (P<0.0001). The variation of albumin concentration before and after weaning was greater in the HPA lambs than in the LPA lambs (P<0.0001). In conclusion, although ewes’ BW, BCS and milk production were affected by pasture allowance until late pregnancy, this did not affect the behaviours that lead to the establishment of the mother–young bond, nor the ewes’ behavioural responses at weaning. Lambs reared by ewes that grazed on low pasture allowance during pregnancy presented fewer behavioural changes and a lower decrease of albumin concentration after weaning. Lambs’ BW was not affected by the feeding received by their mothers.  相似文献

Accurate estimates of milk production or milk intake are difficult, as all methods interfere to some degree with the natural behaviour of the dam and her young, and potentially alter milk yield itself. The present study compared milk yield obtained by the “oxytocin” method, udder dimensions (UD), the isotope dilution method, and live weight change of the lamb, in an attempt to select the most accurate and convenient way of measuring milk production in non-dairy sheep. In addition, the study investigated which of the three milk-estimation techniques was an accurate predictor of growth rates of lambs. Thirty-seven singleton-bearing and rearing ewes were milked once a week, for seven consecutive weeks, using the “oxytocin” method. Prior to each afternoon milking, the external dimensions of the ewe's udder were measured. Lambs were weighed weekly for the first seven weeks of life and live weight change was calculated. The deuterium oxide (D2O) dilution technique was used to estimate milk intake of the lambs and performed at approximately 7 days post-partum and finishing on approximately day 14. Pearson's correlation coefficients and multiple regression coefficients among techniques were calculated. The UD-models at d7 (R2 = 0.35), d35 (R2 = 0.36) and d42 (R2 = 0.34), were the best models explaining variation in milk yield (concordance correlation coefficient (CCC) = 0.49; 0.53; 0.51; for d7, d35 and d42, respectively). The lamb live weight-change model explained the variation in milk yield best at d28 (R2 = 0.32; CCC = 0.49), at d35 (R2 = 0.22; CCC = 0.36) and at d42 (R2 = 0.28; CCC = 0.44). At d14, the intake of milk by lambs as measured by the D2O technique, did not explain the variation in milk yield. In conclusion, udder dimensions, lamb live weight change and lamb milk intake are relatively poor estimators of the milk yield of singleton-rearing ewes obtained by the “oxytocin” method. Additionally, udder dimensions, milk yield and lamb milk intake do not give an accurate prediction of growth rates of singleton lambs. These results emphasize that there is a difference between ewe milk production potential and lamb milk intake, which need to be considered when estimating milk production in non-dairy animals.  相似文献

Ten evolutionary conservative sequences with high identity level to homological sequences in other mammal species were revealed in 5'-flanking region of casein's genes cluster. Five novel SNPs located inside of the evolutionary conservative regions were identified. The binding sites were revealed to be present in one allelic variant of four detected SNPs. So these SNPs were considered as rSNPs. Significant differences of allelic frequencies were revealed between beef cow's group and dairy cow's group in two rSNPs (NCE4, NCE7, p<0.001). Different alleles of those two rSNPs were shown to be associated with some milk performance traits in Black-and-White Holstein dairy cows. Significant difference of protein percentage has been found between cows with G/G and A/A genotypes (P<0.05) and A/G and A/A genotypes (P<0.05) for NCE4 polymorphism. The groups of animals with genotypes G/G and A/G for NCE7 polymorphism were significantly different in milk yield at the first lactation (kg) (P<0.01), milk fat yield (kg) (P<0.05) and milk protein yield (kg) (P<0.01). For the last trait the difference was significant also between cows with genotypes G/G and A/A for rSNP NCE7 (P<0.05).  相似文献

The substitution of lysine for alanine (K232A) in the acyl-CoA:diacylglycerol acyltransferase, which is encoded by the DGAT1 gene, was tested for the significance for breeding evaluation of bulls of the holsteinized Black-and-White breed. The breeding value was estimated by the DYDC (daughter yield deviation to contemporaries) method with modification. The frequency of allele 232K in the bulls examined was 0.28, lower than in Holstein bulls (0.4–0.6). The greatest effect of the A232K substitution was observed for the percent fat milk yield (1.4σ) and milk yield (0.76σ), and a lower effect was established for the milk protein yield (0.47σ) and percent protein milk yield (0.44σ). In the case of milk fat yield, the effect was nonsignificant. A method was proposed for converting the data on fat yield in order to obtain significant results in this case as well. The effect of the A232K substitution was estimated at 154 kg for milk yield, 2.8 kg for milk protein yield, 0.079% for percent milk fat, and 0.015% for percent milk protein yield. The results are discussed in the context of multiple pleiotropic effects of the K232A substitution in the DGAT1 gene. It is proposed that the K232A substitution of the DGAT1 gene may be used as a golden standard in comparisons of the effect on milk production traits for the total gene set. This approach will allow a meta-analysis of the gene effects in spite of the different dairy cattle breeds and methods used to analyze their breeding value. In view of more than 30-year experience of using sperm of Holstein bulls, including those breeds in North America, it was noted that the effect of the A232K substitution on milk production traits agreed well with the data reported for the North American commercial population of Holstein cattle.  相似文献

Oviduct segments from infundibulum, magnum, uterus, uterovaginal junction and vagina of actively laying hens at preoviposition time were tested for their contractile reaction to prostaglandin E1 by or methods. Maximum stimulatory response was observed from the muscular strips of the proximal oviduct segment (infundibulum) and a complete relaxation was recorded from the distal part (vagina) at molar concentrations of 1.4 × 10−7, 3.4 × 10−7 and 7.0 × 10−7. The uterine strips reacted with a stimulatory response at higher concentrations (1.4 × 10−6 and 2.8 × 10−6 moles), but lacked any significant change at lower concentrations. The uterovaginal muscular strips showed a mild but prolonged inhibitory response, while the magnum responded with a significant increase in the luminal pressure when tested . It is concluded that PGE1 exerts a stimulatory effect on the uterus to initiate transport of the egg to subsequent segments (uterovaginal junction and vagina), which relax under PGE1 influence and allow passage of the egg by pressure differences.  相似文献

The aim of this study was to determine how cows with different genetic merit behave and perform when grazing biodiverse and heterogeneous mountain pastures with different slopes. Three groups of 12 cows in late lactation, each composed of four Holstein, four Montbéliarde and four Valdostana Red Pied cows, breeds of increasing presumed robustness and decreasing milk yield (MY) potential. Cows grazed without concentrate either on a low-diversity flat pasture or on two species-rich mountainous pastures having slopes of either 7° or 22°. Milk yield, BW and grazing behaviour were monitored two times in the first and once in the second grazing cycle. Cows of different breeds had similar behaviour on all pastures. The Montbéliarde cows performed close to their production potential; Holstein and Valdostana cows produced less milk than anticipated. No breed difference in terms of BW loss was found. The Valdostana cows exhibited the least selective behaviour with respect to plant species and plant growth stage. Still, all cows searched for the most palatable vegetation regardless of pasture diversity. On the steep pasture, cows optimised the trade-off between ingesting and saving energy to obtain feed. They remained longer at the lowest zone and selected forbs, whereas cows on the flatter pasture went to the upper zone to select grasses. The present study gave no evidence for a superior short-term adaptation to harsh grazing conditions through an optimised feeding behaviour of the Valdostana breed compared to Montbéliarde and Holstein cows.  相似文献

Forty-six lactating Egyptian Nubian (Zaraibi) does were randomly assigned to four groups to investigate the effects of two energy levels at 100% (low) and 125% (high) of NRC recommended standards with rations containing two roughage ratios (20 and 40%) using a 2 × 2 factorial design. Milk yield was measured weekly after 7 days of parturition, for a 32 week period. Composite samples of milk were analyzed for fat, solids-not-fat (SNF), protein and ash contents. Rumen fluid samples were collected at 0, 2, 4 and 8 h after morning feeding to determine total and molar proportions of volatile fatty acids (VFA). Average daily milk yield was 660 g within the test period. Increasing dietary energy level was without significant effects on milk yield and fat percent, while increasing ratio of roughage significantly (P < 0.01) increased percentage of milk fat. This was concomitant with marked increases in VFA concentration in the rumen fluid and molar ratios of acetic acid rather than propionic and butyric acids. This response was more pronounced in does on high energy than in those on the low energy rations. Increasing dietary energy level resulted in significant increases in SNF (P < 0.05) and protein (P < 0.01) contents of milk, while increasing roughage ratio was without significant effect on this parameter. Neither dietary energy level nor roughage ratio affected milk ash.  相似文献

Inversion of aggressive behaviour into a submissive one led to immunosuppression in C57BL/6J mice as well as in those mice which did not change their behaviour, whereas inversion of submissive behaviour into aggressive one resulted in immunostimulation. A possibility to influence immune response changing the brain neurochemical pattern by reversing behaviour under conditions of a social conflict, is discussed.  相似文献

In this study, the hydraulic conductivity (Lp), Me2SO permeability ( Me2SO), and the reflection coefficients (ς) and their activation energies were determined for Metaphase II (MII) mouse oocytes by exposing them to 1.5 M Me2SO at temperatures of 30, 20, 10, 3, 0, and −3°C. These data were then used to calculate the intracellular concentration of Me2SO at given temperatures. Individual oocytes were immobilized using a holding pipette in 5 μl of an isosmotic PBS solution and perfused with precooled or prewarmed 1.5 M Me2SO solutions. Oocyte images were video recorded. The cell volume changes were calculated from the measurement of the diameter of the oocytes, assuming a spherical shape. The initial volume of the oocytes in the isoosmotic solution was considered 100%, and relative changes in the volume of the oocytes after exposure to the Me2SO were plotted against time. Mean (means ± SEM) Lpvalues in the presence of Me2SO ( Me2SOp) at 30, 20, 10, 3, 0, and −3°C were determined to be 1.07 ± 0.03, 0.40 ± 0.02, 0.18 ± 0.01, 7.60 × 10−2± 0.60 × 10−2, 5.29 × 10−2± 0.40 × 10−2, and 3.69 × 10−2± 0.30 × 10−2μm/min/atm, respectively. The Me2SOvalues were 3.69 × 10−3± 0.3 × 10−3, 1.07 × 10−3± 0.1 × 10−3, 2.75 × 10−4± 0.15 × 10−4, 7.83 × 10−5± 0.50 × 10−5, 5.24 × 10−5± 0.50 × 10−5, and 3.69 × 10−5± 0.40 × 10−5cm/min, respectively. The ς values were 0.70 ± 0.03, 0.77 ± 0.04, 0.81 ± 0.06, 0.91 ± 0.05, 0.97 ± 0.03, and 1 ± 0.04, respectively. The estimated activation energies (Ea) for Me2SOp, Me2SO, and ς were 16.39, 23.24, and −1.75 Kcal/mol, respectively. These data may provide the fundamental basis for the development of more optimal cryopreservation protocols for MII mouse oocytes.  相似文献

A substitution of thyrosine for phenylalanine (F297Y) in the transmembrane domain of the growth hormone receptor (GHR) was tested for significance for breeding evaluation of bulls of the holstenized Black-and-White breed. The breeding value was estimated by the method of daughter yield deviation to contemporaries with modification. The frequency of genotype FF in the bulls examined was 0.37, lower than in Holstein bulls (0.67). The F297Y substitution exerted the greatest effect on the milk fat content (1.5??) and milk yield (0.8 ??) and a lower effect on the milk fat yield (0.6 ??), milk protein yield (0.5 ??), and milk protein content (0.6 ??). The GHR4.2 single nucleotide substitution (SNP) in the promoter of the GHR gene did not affect the milk production traits. A substitution of asparagine for serine (S18N) in the transmembrane domain of the prolactin receptor (PRLR) was also examined, but it did not significantly affect the milk production traits. The results are discussed in the context of the hypothesis that multiplicity of causal mutations of a particular gene is common and should be taken into account in the genetics of quantitative traits.  相似文献

Our objective was to determine using electron microscopy how nonagouti (a), lethal yellow (Ay), and albino (c2J) genes affect the program of mouse hairbulb melanosome differentiation; 1,921 hairbulb melanosomes from four genotypes (a/a C/C = B,Ay/a C/C = Y, a/a c2J/c2J = BA, and Ay/a c2J/c2J = YA) were scored for developmental stage, length, and width. Qualitative and quantitative electron microscopy revealed the following. An albino locus-induced diminution of melanosome size suggests that the albino locus is involved in structural features of melanosomes not directly related to the synthesis and deployment of tyrosinase. Ratio data on melanosome length-to-width confirm that the agouti locus determines melanosome shape, either spherical or elliptical; melanization is not required for melanosomes to achieve their agouti-locus-determined shapes. YA (Ay/a c2J/c2J) melanosomes, characterized by poorly organized matrices, absence of active tyrosinase, unusually large membrane invaginations, and significantly smaller dimensions than those of BA (a/a c2J/c2J), showed additive effects of both Ay and c2J alleles. These data suggest that the albino locus plays a structural as well as functional (tyrosinase) role in the differentiation of mouse hairbulb melanosomes. The agouti locus, even in the absence of melanization, directs melanosome shape either via synthesis and deployment of agouti-locus-encoded matrix proteins or by other structural factors. The additive effects of Ay and c2J alleles in compound YA mutants document the importance of specific interactions both functional and structural between agouti and albino loci.  相似文献

Obese Ay/a females of 120 days or older, when compared to age-matched a/a controls (strain C57BL/6J), exhibited abnormal oestrous cyclicity characterized by reduced frequencies of true oestrous-stage smears, decreased mating success to proven a/a males, lowered uterine weights, and depressed ovulation rates. Exogenous gonadotrophins (PMSG/hCG) partly restored ovulation in obese Ay/a females to near control levels, demonstrating the sensitivity of Ay/a ovarian tissues to FSH and LH, at least at superovulatory levels. Concentrations of endogenous gonadotrophins and/or sensitivity of ovarian target cells to gonadotrophins may therefore be impaired in obese Ay/a females. Aberrant copulatory behaviour, reduced uterine weights, and depressed conception rates strongly suggest ovarian steroid deficiencies, perhaps secondary effects of reduced endogenous gonadotrophin activity. As in other obese rodent syndromes e.g. ob/ob, db/db, and fa/fa), a possible fundamental Ay-induced hypothalamic lesion is consistent with our data.  相似文献

With the aim of testing the hypothesis that regrouping decreases milk production in French Alpine goats that were lactating, a study was done using two groups (n = 8, 7). During their third month after parturition, four goats from each group were exchanged (first regrouping) between pens and left for 2 weeks, then the same two subgroups of four goats were taken back to their original pen for another 2 weeks (second regrouping). In the third regrouping, the two groups were all placed in the one pen. Milk production and social behaviour were measured daily before and after each regrouping. All regroupings led to an increase in aggressive behaviours that last by 1–2 days. Mean daily milk production decreased after first (2.82 ± 0.2 kg versus 2.53 ± 0.2 kg; P < 0.05) but not after second and third regrouping. It is concluded that aggressive behaviour increases after all regroupings, whereas milk production decreases only after the first regrouping, suggesting an important capacity of adaptation to a novel and stressful managements in the French Alpine goat. The study highlights the importance of considering effects of common practices in herd managements on social behaviour and production.  相似文献

This study examined the relationships between the attitude and the behaviour of the stockperson towards cows and the behavioural response to humans and the milk production of cows at 31 commercial dairy farms over one lactation. The attitude of the stockperson was measured on the basis of the stockperson's opinion of the cow's behaviour and how the stockperson evaluated his own behaviour. The behaviour of the stockperson was measured by recording the nature and frequency of the tactile interactions and some visual and auditory interactions directed towards the cows. The behavioural response of cows to humans was assessed by observing their approach behaviour to an unfamiliar experimenter in a standard test and production records were collected for the entire lactation at each farm. Correlation and regression analyses using farm averages were used to examine relationships between human and cow variables. Several cow behaviour variables, indicative of fear of humans, were moderately (P<0.05) to highly (P<0.01) correlated with milk yield and composition and regression analysis indicated that fear of humans accounted for 19% of the variation in milk yield between farms. The results suggest that at farms where milk yield was low, cows showed less approach to the experimenter in the standard fear test than at farms where milk yield was higher. A composite attitude score, based on the responses of stockpeople to questions about patting and talking to cows, ease of movement of cows and cows recognising unfamiliar stockpeople, was moderately (P<0.05) to highly (P<0.01) correlated with the behaviour of the stockperson. While a number of stockperson behaviour variables were correlated (P<0.05) with milk yield, the former variables were generally poorly correlated with cow behaviour. Therefore these preliminary findings provide evidence that, as seen in the pig industry, sequential relationships may exist between the attitude and behaviour of the stockperson and the behaviour and productivity of commercial dairy cows. Research is required to further examine these relationships because of the possible implications on cow productivity and welfare.  相似文献

This study determined impacts of a commercial blend of microencapsulated essential oils (EO), fed at increasing dose levels to high yielding primiparous dairy cows, on milk yield and composition. Eight pregnant Holstein heifers, selected to have their parturition within a period of 30 d, were kept in tie stalls equipped for individual feeding, and with free access to water, from the 7th month of gestation. At an average of 40 d postpartum, cows were assigned to one of four dietary EO levels in a replicated 4 × 4 Latin square design. The experimental diets (i.e., corn silage, fescue hay and a compound feed, 6.5, 4.4 and 8.7 kg DM/d respectively) differed only in the addition level of a microencapsulated EO supplement (RumaXol Feed) mixed at different levels into the compound feed (fed at 1 kg/cow day) in order to provide 0, 0.32, 0.64 and 0.96 g/d of the EO mixture. The experiment had four experimental periods of 21 d, with the last 5 d used for collection of faeces and urine, for milk yield and composition recording and measurement of cow biometric data. The dietary EO supplementation had no effect on dry matter (DM) intake, water consumptions or faecal DM, while urine density was (P=0.01) lower at the intermediate EO dosages. Digestibility and biometric measures of cows were not modified by EO feeding. The protein content of milk tended (P=0.06) to be higher at the intermediate EO dosages, as did (P=0.05) the milk energy concentration.
