Sexually mature gilts were actively immunized against gonadotropin-releasing hormone (GnRH) by conjugating GnRH to bovine serum albumin, emulsifying the conjugate in Freund's adjuvant, and giving the emulsion as a primary immunization at Week 0 and as booster immunizations at Weeks 10 and 14. Antibody titers were evident by 2 wk after primary immunization and increased markedly in response to booster immunizations. Active immunization against GnRH caused gonadotropins to decline to nondetectable levels, gonadal steroids to decline to basal levels, and the gilts to become acyclic. Prolactin concentrations in peripheral circulation were unaffected by immunization against GnRH. The endocrine status of the hypothalamic-pituitary-ovarian axis was examined by giving GnRH and two agonists to GnRH and by ovariectomy. An i.v. injection of 100 micrograms GnRH caused release of luteinizing hormone (LH) and follicle-stimulating hormone (FSH) in control animals, but not in gilts immunized against GnRH. In contrast, administration of 5 micrograms D-(Ala6, des-Gly-NH2(10] ethylamide or 5 micrograms D-(Ser-t-But6, des-Gly-NH2(10] ethylamide resulted in immediate release of LH and FSH in both control and GnRH-immunized gilts. Circulating concentrations of LH and FSH increased after ovariectomy in the controls, but remained at nondetectable levels in gilts immunized against GnRH. Prolactin concentrations did not change in response to ovariectomy. We conclude that cyclic gilts can be actively immunized against GnRH and that this causes cessation of estrous cycles and inhibits secretion of LH, FSH, and gonadal steroids.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Immunocontraception has become an increasingly valuable tool in the population management of captive exotic ungulates. Although porcine zona pellucida vaccine (PZP) was used successfully in other cervids, a previous study with fallow deer (Cervus dama) suggested that the vaccine did not work in this species. In the current study, PZP was tested in two captive herds of fallow deer. Antibody titers were monitored over a 3‐year period to evaluate three different adjuvant protocols, and the vaccine was applied to an entire herd to determine the impact on fawning rates. In a semi‐free‐ranging herd, antibody titers rose from preimmunization levels of 2.6% of positive control serum to 56.5% 4 weeks after initial inoculations, to 65.1% at 1 year, and to 81.3% at 2 years, after a single annual booster was applied. Fawn production in this herd was reduced significantly over 3 years. The adjuvant protocol of Freund's Modified Adjuvant® (FMA) for the initial inoculation followed by a booster with Freund's Incomplete Adjuvant® (FIA), and the protocol of FMA for the initial inoculation followed in 3 weeks by a booster with FMA both produced significantly higher antibody titers than the 3× FIA (3 weeks apart) protocol after year 1. The FMA+FMA protocol produced significantly higher titers than the 3× FIA protocol at year 2, but was not different from the titers produced by the FMA+FIA protocol at year 2. There is a public perception that the white‐tailed deer Odocoileus virginianus (Artiodactyla: Cervidae) is the main reservoir supporting the maintenance and spread of the causative agent of Lyme disease, Borrelia burgdorferi. This study examines the pathogen prevalence rate of Borrelia in adult Ixodes scapularis (Ixodida: Ixodidae), the black‐legged tick, collected from white‐tailed deer and compares it with pathogen prevalence rates in adult ticks gathered by dragging vegetation in two contiguous counties west of the Hudson Valley in upstate New York. In both Broome and Chenango Counties, attached and unattached ticks harvested from white‐tailed deer had significantly lower prevalences of B. burgdorferi than those collected from vegetation. No attached ticks on deer (n = 148) in either county, and only 2.4 and 7.3% of unattached ticks (n = 389) in Broome and Chenango Counties, respectively, were harbouring the pathogen. This contrasts with the finding that 40.8% of ticks in Broome County and 46.8% of ticks in Chenango County collected from vegetation harboured the pathogen. These data suggest that a mechanism in white‐tailed deer may aid in clearing the pathogen from attached deer ticks, although white‐tailed deer do contribute to the spatial distribution of deer tick populations and also serve as deadend host breeding sites for ticks.  相似文献

Quantifying the relative influence of multiple mechanisms driving recent range expansion of non‐native species is essential for predicting future changes and for informing adaptation and management plans to protect native species. White‐tailed deer (Odocoileus virginianus) have been expanding their range into the North American boreal forest over the last half of the 20th century. This has already altered predator–prey dynamics in Alberta, Canada, where the distribution likely reaches the northern extent of its continuous range. Although current white‐tailed deer distribution is explained by both climate and human land use, the influence each factor had on the observed range expansion would depend on the spatial and temporal pattern of these changes. Our objective was to quantify the relative importance of land use and climate change as drivers of white‐tailed deer range expansion and to predict decadal changes in white‐tailed deer distribution in northern Alberta for the first half of the 21st century. An existing species distribution model was used to predict past decadal distributions of white‐tailed deer which were validated using independent data. The effects of climate and land use change were isolated by comparing predictions under theoretical “no‐change between decades” scenarios, for each factor, to predictions under observed climate and land use change. Climate changes led to more than 88%, by area, of the increases in probability of white‐tailed deer presence across all decades. The distribution is predicted to extend 100 km further north across the northeastern Alberta boreal forest as climate continues to change over the first half of the 21st century.  相似文献

Conservation and management of species require basic knowledge on their geographic distribution and abundance. Here, we propose a novel approach, based on the theory of the ecological niche, to model the spatial patterns of the white‐tailed deer Odocoileus virginianus population density in two regions of central Mexico (Balsas Basin and Tehuacán‐Cuicatlán Valley). We used an ecological niche model to generate binary geographic distribution maps of the white‐tailed deer in each region based on occurrence data and a set of environmental variables. Then, the centroid of the distributions was calculated in ecological space (niche centroid) and the multidimensional Euclidian ecological distance of each pixel to the niche centroid was estimated. Finally, for each region the distance to the niche centroid (DNC) was regressed against 14 independent occurrence points in each site containing white‐tailed deer density information to determine the function describing the DNC‐density relationship, which was used to generate maps describing the distribution of white‐tailed deer density. Our results indicated an inverse DNC‐density relationship in both regions (Balsas Basin: r2 = 0.90 and Tehuacán‐Cuicatlán: r2 = 0.76) that was validated via bootstrapping resulting in a predicting capacity of near 62% for Balsas Basin and 65% for Tehuacán‐Cuicatlán Valley. Our results suggest that the distance to the niche centroid method is a robust, science‐based correlative approach that resulted useful to predict the population density of the white‐tailed deer in a spatially explicit fashion. The proposed approach is suitable for predicting the distribution of density for white‐tailed deer for which occurrence data with accompanying density information exists, but relative abundance can also be estimated when no abundance data are available.  相似文献

Endozoochory is a prominent form of seed dispersal in tropical dry forests. Most extant megafauna that perform such seed dispersal are ungulates, which can also be seed predators. White‐tailed deer (Odocoileus virginianus) is one of the last extant megafauna of Neotropical dry forests, but whether it serves as a legitimate seed disperser is poorly understood. We studied seed dispersal patterns and germination after white‐tailed deer gut passage in a tropical dry forest in southwest Ecuador. Over 23 mo, we recorded ca 2000 seeds of 11 species in 385 fecal samples. Most seeds belonged to four species of Fabaceae: Chloroleucon mangense, Senna mollissima, Piptadenia flava, and Caesalpinia glabrata. Seeds from eight of the 11 species dispersed by white‐tailed deer germinated under controlled conditions. Ingestion did not affect germination of C. mangense and S. mollissima, whereas C. glabrata showed reduced germination. Nevertheless, the removal of fruit pulp resulting from ingestion by white‐tailed deer could have a deinhibition effect on germination due to seed release. Thus, white‐tailed deer play an important role as legitimate seed dispersers of woody species formerly considered autochorous. Our results suggest that more research is needed to fully understand the ecological and evolutionary effects of the remaining extant megafauna on plant regeneration dynamics in the dry Neotropics.  相似文献

A heterozygosity–fitness correlations (HFCs) may reflect inbreeding depression, but the extent to which they do so is debated. HFCs are particularly likely to occur after demographic disturbances such as population bottleneck or admixture. We here study HFC in an introduced and isolated ungulate population of white‐tailed deer Odocoileus virginianus in Finland founded in 1934 by four individuals. A total of 422 ≥ 1‐year‐old white‐tailed deer were collected in the 2012 hunting season in southern Finland and genotyped for 14 microsatellite loci. We find significant identity disequilibrium as estimated by g2. Heterozygosity was positively associated with size‐ and age‐corrected body mass, but not with jaw size or (in males) antler score. Because of the relatively high identity disequilibrium, heterozygosity of the marker panel explained 51% of variation in inbreeding. Inbreeding explained approximately 4% of the variation in body mass and is thus a minor, although significant source of variation in body mass in this population. The study of HFC is attractive for game‐ and conservation‐oriented wildlife management because it presents an affordable and readily used approach for genetic monitoring that allowing identification of fitness costs associated with genetic substructuring in what may seem like a homogeneous population.  相似文献

Immunization against gonadotrophin releasing hormone (GnRH) was studied as an alternative for the commonly used surgical castration in stallions. Two GnRH vaccines comprising non-mineral oil adjuvants were evaluated for their potential to induce high antibody titers directed against GnRH and subsequent effects on reproductive characteristics. Twelve sexually mature male hemicastrated Shetland ponies were assigned to three groups. Group 1 and 2 were injected with 1mg peptide equivalent of G6k-GnRH-tandem-dimer conjugated to ovalbumin (OVA) in CoVaccine HT adjuvant (GnRH/CoVaccine) and in Carbopol (GnRH/Carbopol), respectively, and group 3 was injected with CoVaccine HT adjuvant without antigen (controls). After immunization no adverse effects were observed with respect to the injections sites or general health. Two weeks after the second vaccination antibody titers against GnRH increased rapidly in all animals of the GnRH/CoVaccine group, at the same time reducing serum testosterone levels maximally for the further duration of the experiment. In the GnRH/Carbopol group antibody responses and effects on testosterone levels were intermediate in two stallions and not apparent in the remaining stallions of this group. Semen evaluation showed that from 2 weeks after the second immunization onwards, sperm motility was affected in all stallions treated with GnRH/CoVaccine and one stallion treated with GnRH/Carbopol. Seven weeks after the second immunization, no semen could be collected from two stallions, one of each group, due to suppressed libido. Histological examination of the testes, 15 weeks after the initial immunization, demonstrated reduction in seminiferous tubuli diameters in all stallions of the GnRH/CoVaccine group and one stallion of the GnRH/Carbopol group. Furthermore, spermatogenesis was extremely disorganized in these stallions, as indicated by absence of the lumen in the seminiferous tubules, the absence of spermatozoa and spermatids in the tubular cross-sections and the impossibility to determine the stage of the tubular cross-sections. Testis size was also substantially reduced in three out of four stallions treated with GnRH/CoVaccine. The results demonstrate that two immunizations with G6k-GnRH-tandem-dimer-OVA conjugate in a suitable adjuvant such as CoVaccine HT caused a rapid and complete reduction of serum testosterone levels in sexually mature stallions, subsequently leading to reduced sperm motility and affected testis function, while no adverse reactions were observed after immunizations.  相似文献

Juvenile survival is a highly variable life‐history trait that is critical to population growth. Antipredator tactics, including an animal's use of its physical and social environment, are critical to juvenile survival. Here, we tested the hypothesis that habitat and social characteristics influence coyote (Canis latrans) predation on white‐tailed deer (Odocoileus virginianus) and mule deer (O. hemionus) fawns in similar ways during the neonatal period. This would contrast to winter when the habitat and social characteristics that provide the most safety for each species differ. We monitored seven cohorts of white‐tailed deer and mule deer fawns at a grassland study site in Alberta, Canada. We used logistic regression and a model selection procedure to determine how habitat characteristics, climatic conditions, and female density influenced fawn survival during the first 8 weeks of life. Fawn survival improved after springs with productive vegetation (high integrated Normalized Difference Vegetation Index values). Fawns that used steeper terrain were more likely to survive. Fawns of both species had improved survival in years with higher densities of mule deer females, but not with higher densities of white‐tailed deer females, as predicted if they benefit from protection by mule deer. Our results suggest that topographical variation is a critical resource for neonates of many ungulate species, even species like white‐tailed deer that use more gentle terrain when older. Further, our results raise the possibility that neonatal white‐tailed fawns may benefit from associating with mule deer females, which may contribute to the expansion of white‐tailed deer into areas occupied by mule deer.  相似文献

An important assumption in observational studies is that sampled individuals are representative of some larger study population. Yet, this assumption is often unrealistic. Notable examples include online public‐opinion polls, publication biases associated with statistically significant results, and in ecology, telemetry studies with significant habitat‐induced probabilities of missed locations. This problem can be overcome by modeling selection probabilities simultaneously with other predictor–response relationships or by weighting observations by inverse selection probabilities. We illustrate the problem and a solution when modeling mixed migration strategies of northern white‐tailed deer (Odocoileus virginianus). Captures occur on winter yards where deer migrate in response to changing environmental conditions. Yet, not all deer migrate in all years, and captures during mild years are more likely to target deer that migrate every year (i.e., obligate migrators). Characterizing deer as conditional or obligate migrators is also challenging unless deer are observed for many years and under a variety of winter conditions. We developed a hidden Markov model where the probability of capture depends on each individual's migration strategy (conditional versus obligate migrator), a partially latent variable that depends on winter severity in the year of capture. In a 15‐year study, involving 168 white‐tailed deer, the estimated probability of migrating for conditional migrators increased nonlinearly with an index of winter severity. We estimated a higher proportion of obligates in the study cohort than in the population, except during a span of 3 years surrounding back‐to‐back severe winters. These results support the hypothesis that selection biases occur as a result of capturing deer on winter yards, with the magnitude of bias depending on the severity of winter weather. Hidden Markov models offer an attractive framework for addressing selection biases due to their ability to incorporate latent variables and model direct and indirect links between state variables and capture probabilities.  相似文献

Five lighthorse mares were actively immunized against gonadotropin releasing hormone (GnRH) to determine the relative importance of this hypothalamic hormone in the secretion of luteinizing hormone (LH) and follicle-stimulating hormone (FSH). Five mares immunized against the conjugation protein served as controls. Mares were initially immunized in November and received secondary immunizations 4 wk later, and then at 6-wk intervals until ovariectomy in June. All mares immunized against GnRH exhibited an increase (p less than 0.01) in the binding of tritiated GnRH by plasma, an indication that antibodies against this hormone had been elicited. Concentrations of LH, FSH and progesterone in weekly blood samples were lower (p less than 0.05) in GnRH-immunized mares than in controls after approximately 4 mo of immunization. However, the LH concentrations were affected to a greater degree than were FSH concentrations. All five control mares exhibited normal cycles of estrus and diestrus in spring, whereas no GnRH-immunized mare exhibited cyclic displays of estrus up to ovariectomy. All mares were injected intravenously with a GnRH analog (which cross-reacted less than 0.1% with the anti-GnRH antibodies) in May, after all control mares had displayed normal estrous cycles, to characterize the response of LH and FSH in these mares; two days later, the mares were injected with GnRH. The LH response to the analog, which was assessed by net area under the curve, was lower (p less than 0.01) by approximately 99% in mares immunized against GnRH than in control mares. Female white‐tailed deer (Odocoileus virginianus) are thought to choose between two behavioral strategies to maximize the quality of potential mates: sit and wait, characterized by concentrating activity within a restricted area, and excursive behavior, characterized by increased activity and excursions outside the home range. As movement patterns may influence conception, our goal was to examine the patterns of female white‐tailed deer movements to evaluate which breeding strategy was employed. We equipped 36 female white‐tailed deer with GPS collars from August 2013 to December 2015. We found that movement rate and probability of activity were greatest near the peak of the breeding season, and we observed increases in both metrics during the 40 days prior to estimated conception. Peak size of home range and core area occurred in the days surrounding conception. We found that 11 deer performed an excursion, ranging from 43 days before until 36 days after conception, with the peak probability of being outside of an individual home range occurring 1 day prior to conception. Our results suggest that female white‐tailed deer may attempt to maximize the quality of their mates by advertising availability for breeding through excursive behaviors just prior to entering estrus.  相似文献

Mature female guinea pigs that had been observed to undergo three consecutive periods of estrus at approximately 16-day intervals were immunized with either 100 μg gonadotropin releasing hormone (GnRH) conjugated to 100 μg bovine serum albumin (BSA) or 100 μg BSA alone during diestrus (day 5–10) of the fourth cycle. Booster immunizations were administered 32 days after the first injection. Animals were bled by cardiac puncture at the time of first injection and at 16, 32, 48 and 64 days. Animals were necropsied at 64 days after first treatment.Daily observation indicated that vaginal manifestation of estrus was not apparent after a period equal to one estrous cycle in seven of ten GnRH immunized guinea pigs and after two cycles in the remaining three GnRH immunized guinea pigs. Estrous cycles persisted in BSA treated females throughout the experiment.Serum luteinizing hormone (LH) declined significantly by 32 days after the first immunization against GnRH and remained lower than both pretreatment values and levels in control animals at the same bleeding times throughout the experiment. Serum progesterone levels were significantly lower in the GnRH immunized group than in the control group at 48 and 64 days.At necropsy the weight of the ovaries of GnRH immunized guinea pigs was significantly lower than that of controls. Corpora lutea and antral follicles were present in both GnRH treated and control females. The presence of serum progesterone levels and of antral follicles in the GnRH immunized females suggests that a low level of gonadotropic support may have persisted to 64 days after initiation of treatment.Results indicate that immunization against GnRH can reduce LH and progesterone levels and induce cessation of estrous cycles in the guinea pig.  相似文献

Culicoides spp. biting midges (Diptera: Ceratopogonidae) are vectors of pathogens that have a significant economic impact on the livestock industry. White‐tailed deer (Odocoileus virginianus), a farmed species in the U.S.A., are susceptible to two Culicoides spp. borne orbiviruses: bluetongue virus and epizootic haemorrhagic disease virus. Elucidating host–vector interactions is an integral step in studying disease transmission. This study investigated the host range of Culicoides spp. present on a big game preserve in Florida on which a variety of Cervidae and Bovidae freely roam. Culicoides were captured with Centers for Disease Control and Prevention (CDC) miniature light traps run twice weekly on the preserve for 18 consecutive months (July 2015–December 2016). Host preference was quantified through forage ratios, based upon PCR‐based bloodmeal analysis of Culicoides spp. and overall animal relative abundance on the preserve. Culicoides stellifer preferentially fed on Cervus spp. and fallow deer (Dama dama) and displayed a relative avoidance of Bovidae and white‐tailed deer. Culicoides debilipalpis preferred white‐tailed deer and avoided all Bovidae. Culicoides pallidicornis and Culicoides biguttatus showed preferences for white‐tailed deer and Père David's deer (Elaphurus davidianus), respectively. These results add to current knowledge of preferred hosts of Florida Culicoides spp. and have implications for the spread of orbiviruses. Copyright © 2018 John Wiley & Sons, Ltd.  相似文献

Questing adult blacklegged tick (Ixodes scapularis Say) abundance declined markedly three years after the 1999 removal of white‐tailed deer (Odocoileus virginianus Zimmermann) from Monhegan Island, ME. Since 2000, subadult ticks have not been found on Norway rats (Rattus norvegicus Berkenhout); questing nymphs have not been found since 2002. This suggested I. scapularis was reintroduced annually via bird importation of subadult ticks, but unable to complete its two‐year life cycle on the island due to lack of deer. To investigate this, we used uncertainty analysis to estimate 1) questing adult ticks/ha that would result from avian importation of nymphs, and 2) questing adult ticks/ha on Monhegan Island, using bird capture and tick burden data from Appledore Island, ME, flagged tick data from Monhegan Island, and ten uncertain parameters. During the deer‐fed period (1990–2001), estimated tick density on Monhegan Island was 18 times greater than that of imported ticks. During the post‐deer‐fed period (2002–2008), Monhegan Island tick density was equivalent to imported tick density. This supported the premise that all I. scapularis ticks on Monhegan Island have been bird‐derived since 2002.  相似文献

Aim of the present study was to characterize the pituitary GnRH receptor in white sturgeon, Acipenser transmontanus, using a superagonist analog of mammalian GnRH [D‐Ala6, des‐Gly10–Pro9]‐ethylamide and to investigate the possible effect of estradiol‐17β treatment on the concentration and affinity of the GnRH receptor in immature white sturgeon. The binding of 125I‐GnRH‐A to sturgeon pituitary receptors was rapid and saturable at 4°C and 20°C. However, maximal binding at 20°C was almost two‐fold greater than the highest binding noted at 4°C. Specific binding of radioligand was directly related to the amount of tissue included in the assay system over the range of 5–20 mg fresh tissue equivalents per ml. The binding capacity of 125I‐GnRH‐A with sturgeon pituitary tissue was much greater than radiolabeled GnRH. Administration of E2 to immature sturgeon caused an almost two‐fold increase in GnRH‐A binding capacity (E2 treated: Bmax = 2.87 fmoles 3 mg?1 FTE; control: Bmax = 1.70 fmoles 3 mg?1), and did not affect GnRH‐A binding affinity (E2 treated: Ka = 0.13 × 1011 m ?1; control: Ka = 0.15 × 1011 m ?1). Overall, the study provides evidence that the GnRH analog is effective for characterizing the GnRH receptor in white sturgeon; however, more experimentation is necessary to determine whether E2 administration to immature white sturgeon can increase the GnRH receptor capacity.  相似文献

Abstract

Changes in the circulating plasma volume were monitored for twelve consecutive months in five white‐tailed deer (Odocoileus virginianus) maintained outdoors in a 2.4‐ha enclosure in northern New York stale. The mean annual relative plasma volume of the two male white‐tailed deer (43.6 ± 1.0 ml/kg) was not significantly different from that of three females (45.7 ± 0.7 ml/kg). Mean annual absolute plasma volume, however, was significantly higher in males (2588 ± 90.7 ml) than in females (2092 ± 79.1 ml), mainly because of the greater body weights of males in late summer and early fall. Both sexes had a marked seasonality in the level of relative and absolute plasma volume regulation. Relative plasma volumes were lowest in February‐March and highest in May‐June and again in October, while absolute plasma volumes were lowest in late winter to early spring, when body weights in the annual cycle were lowest, and volumes were highest in mid‐fall, at the annual peak of body condition. Males had a greater excursion about their annual mean relative plasma volumes (‐16 to 13%) and absolute plasma volumes (‐23 to 32%) than did females (respectively, ‐11 to 9% and ‐13 to 19%). Variations in the level of plasma volume regulated are related to the annual cycle of environmental conditions and changes in body and physiological condition of white‐tailed deer.  相似文献

在1997~2000年间,甘肃兴隆山麝场的圈养马麝发情交配的时间节律保持相对年间恒定,1996年麝场进行了大规模的圈群间动物调整,导致该年的马麝圈群进入发情较迟,发情持续时间也较其他年份长。总体上,圈养马麝的发情持续时间达3个月,而66%的的发情交配发生于1个月内(从11月21日~12月21日),发情季节(75%的发情完成累计时间)长36天。雌性圈养马麝发情的时间格局受圈养环境的季节性因子影响。饲养人员的饲养风格及各个饲养区内雌性马麝的社会行为的调节效应,各饲养区动物的发情时间格局有显著差异。年龄可影响马麝的发情交配,5.5岁龄以上的马麝发情定时更早。上一年度的繁殖成功与否和当年动物发情迟早无显著相关。野捕圈养麝群和其F1代圈群间的发情时间格局无显著差异。有较多发情周期的个体并不比发情周期仅1~2次的个体更早进入发情。  相似文献

Climate change is expected to create novel environments in which extant species cannot persist, therefore leading to the loss of them and their associated ecological functions within the ecosystem. However, animals may employ behavioral mechanisms in response to warming that could allow them to maintain their functional roles in an ecosystem despite changed temperatures. Specifically, animals may shift their activity in space or time to make use of thermal heterogeneity on the landscape. However, few studies consider the role of behavioral plasticity and spatial or temporal heterogeneity in mitigating the effects of climate change. We conducted experiments to evaluate the potential importance of behavior in mediating the net effects of warming on white‐tailed deer (Odocoileus virginianus). We used shade structures to manipulate the thermal environment around feeding stations to monitor deer feeding activity and measure total consumption. In individual experiments where deer only had access to unshaded feeders, deer fed less during the day but compensated by increasing feeding during times when temperature was lower. In group experiments where deer had access to both shaded and unshaded feeders, deer often fed during the day but disproportionally preferred the cooler, shaded feeders. Our results suggest that deer can capitalize on temporal and spatial heterogeneity in the thermal environment to meet nutritional and thermal requirements, demonstrating the importance of behavioral plasticity when predicting the net effects of climate change.  相似文献
