Environmental temperature is one of the critical factors affecting fish development. The aim of this study was to examine the impact of three different rearing temperatures (16, 19 and 22°C) throughout the endogenous feeding phase of the Siberian sturgeon Acipenser baerii. This was performed by assessing (a) larval survival and growth; (b) immunofluorescence localization and expression of genes involved in muscle development and growth – myog and Igf1; and (c) stress status through the expression of thermal stress genes – Hsp70, Hsp90α and Hsp90β – and whole body cortisol. Overall survival rate and larval weight did not differ significantly across temperatures. Larvae subjected to 22°C showed faster absorption of the yolk-sac than larvae subjected to 19 or 16°C. Both at schooling and at the end of the trial, larvae reared at 16°C showed significantly lower levels of cortisol than those reared at 19 or 22°C. IGF-1 immunopositivity was particularly evident in red muscle at schooling stage in all temperatures. The expression of all Hsps as well as the myog and Igf1 genes was statistically higher in larvae reared at 16°C but limited to the schooling stage. Cortisol levels were higher in larvae at 22°C, probably because of the higher metabolism demand rather than a stress response. The observed apparent incongruity between Hsps gene expression and cortisol levels could be due to the lack of a mature system. Further studies are necessary, especially regarding the exogenous feeding phase, in order to better understand if this species is actually sensitive to thermal stress. 相似文献

The Chinese sturgeon, Acipenser sinensis, is an anadromous protected species that presently only spawns in the Yangtze River. Using laboratory experiments, we examined the behavioral preference of young Chinese sturgeon to physical habitat (water depth, illumination intensity, substrate color, and cover) and monitored their downstream migration. Hatchling free embryos were photopositive, preferred open habitat, and immediately upon hatching, swam far above the bottom using swim-up and drift. Downstream migration peaked on days 0–1, decreased about 50% or more during days 2–7, and ceased by day 8. Days 0–1 migrants were active both day and night, but days 2–7 migrants were most active during the day. After ceasing migration, days 8–11 embryos were photonegative, preferred dark substrate and sought cover. Free embryos developed into larvae and began feeding on day 12, when another shift in behavior occurred–larvae returned to photopositive behavior and preferred white substrate. The selective factor favoring migration of free embryos upon hatching and swimming far above the bottom may be avoidance of benthic predatory fishes. Free embryos, which must rely on yolk energy for activity and growth, only used 19 cumulative temperature degree-days for peak migration compared to 234 degree-days for growth to first feeding larvae, a 1:12 ratio of cumulative temperature units. This ratio suggests that sturgeon species with large migratory embryos, like Chinese sturgeon, which require a high level of energy to swim during migration, may migrate only a short time to conserve most yolk energy for growth. 相似文献

In this study, we investigated the effects of different stocking densities water quality and blood parameters during the transportation of the Siberian sturgeon (Acipenser baerii). Experiments were carried out in high-density polyethylene tank with three different stocking densities (50, 100 and 150 kg/m3) for 20 h. Ammonium nitrogen (NH4-N) and nitrite nitrogen (NO2-N) were measured in the water samples. The erythrocyte, leucocyte, haematocrit (HCT), haemoglobin (Hb), cortisol, and sodium ion (Na+) were measured in blood samples. Ammonium nitrogen and nitrite nitrogen concentrations in the water have increased in parallel with the stocking density depending on the time. In the highest stocking density, the maximum levels of NH4-N and NO2-N at the 16th h reached 2.64 mg/L and 4.74 mg/L, respectively. Erythrocyte, leucocyte, HCT, and Hb values did not differ significantly between the experimental groups (p > .05). The results showed that fish could be transported safely for 20 h in 15℃ water temperature at 50 kg/m3 stocking density; however, stocking density of 100 kg/m3 and over could threaten fish welfare and health as from 16th h. 相似文献

The head is considered the major novelty of the vertebrates and directly linked to their evolutionary success. Its form and development as well as its function, for example in feeding, is of major interest for evolutionary biologists. In this study, we describe the skeletal development of the cranium and pectoral girdle in Siberian (Acipenser baerii ) and Russian sturgeon (A. gueldenstaedtii ), two species that are commonly farmed in aquaculture and increasingly important in developmental studies. This study comprises the development of the neuro‐, viscero‐ and dermatocranium and the dermal and chondral components of the pectoral girdle, from first condensation of chondrocytes in prehatchlings to the early juvenile stage and reveals a clear pattern in formation. The otic capsules, the parachordal cartilages, and the trabeculae cranii are the first centers of chondrification, at 8.4mm TL. These are followed by the mandibular, then the hyoid, and later the branchial arches. Teeth form early on the dentary, dermopalatine, and palatopterygoid, and then appear later in the buccal cavity as dorsal and ventral toothplates. With ongoing chondrification in the neurocranium a capsule around the brain and a strong rostrum are formed. Dermal ossifications start to form before closure of the dorsal neurocranial fenestrae. Perichondral ossification of cartilage bones occurs much later in ontogeny. Our results contribute data bearing on the homology of elements such as the lateral rostral canal bone that we regard homologous to the antorbital of other actinopterygians based on its sequence of formation, position and form. We further raise doubts on the homology of the posterior ceratobranchial among Actinopteri based on the formation of the hyoid arch elements. We also investigate the basibranchials and the closely associated unidentified gill‐arch elements and show that they are not homologous. J. Ontogenetic behavior of Hudson River Atlantic sturgeon and Connecticut River shortnose sturgeon early life intervals were similar during laboratory observations. After hatching, free embryos were photonegative and sought cover. When embryos developed into larvae, fish left cover, were photopositive, and initiated downstream migration. Free embryos may remain at the spawning site instead of migrating downstream because the risk of predation at spawning sites is low. The two species are sympatric, but not closely related, so the similarities in innate behaviors suggest common adaptations, not phylogenetic relationship. Atlantic sturgeon migrated downstream for 12 days (peak, first 6 days), shortnose sturgeon migrated for 3 days, and year-0 juveniles of both species did not resume downstream migration. Short or long migrations of larvae may reflect different styles related to the total migratory distance from spawning sites to juvenile rearing areas. Atlantic sturgeon need to move a short distance to reach rearing areas and they had a long 1-step migration of 6–12 days. In contrast, shortnose sturgeon need to move a long distance to reach all rearing areas. This may be accomplished by a 2-step migration, of which the brief migration of larvae is only the first step. Early migrant Atlantic sturgeon were nocturnal, while late migrants were diurnal, and shortnose sturgeon were diurnal. These diel differences may also be adaptations for long (Atlantic sturgeon) or short (shortnose sturgeon) migrations. Cultured shortnose sturgeon, and possibly Atlantic sturgeon, have a dominance hierarchy with large fish dominant when competing for limited foraging space. Social behavior may be more important in the life history of wild sturgeons than is generally recognized. 相似文献

Historically, shortnose sturgeon inhabited most major rivers on the Atlantic coast of North America south of the Saint John River, Canada. Today, only 16 populations may remain. Major anthropogenic impacts on shortnose sturgeon are blockage of spawning runs by dams, harvest of adults (bycatch and poaching), dredging of fresh/saltwater riverine reaches, regulation of river flows, and pollution. The pattern of anadromy (adult use of salt water) varies with latitude. The pattern may reflect bioenergetic adaptations to latitudinal differences between fresh and salt water habitats for thermal and foraging suitability. The greater adult abundance in northern and north-central populations likely reflects a historical difference with southern populations that is currently accentuated by increased anthropogenic impacts on southern populations. Adult abundance is less than the minimum estimated viable population abundance of 1000 adults for 5 of 11 surveyed populations, and all natural southern populations. Across the latitudinal range, spawning adults typically travel to about river km 200 or farther upstream. Dams built downstream of spawning reaches block spawning runs, and can divide amphidromous populations into up- and downstream segments. Conservation efforts should correct environmental and harvest impacts, not stock cultured fish into wild populations. 相似文献

Eleven of 34 sturgeons caught in the River Volga classified morphologically as Acipenser gueldenstaedtii were identified as Acipenser baerii from sequence analysis of the mitochondrial cytochrome- b gene. The Caspian Sea and its tributaries including the Volga are not native habitats of A. baerii . No A. baerii haplotype was observed in A. gueldenstaedtii from the Sea of Azov or the South Caspian Sea. Genetic contamination of A. gueldenstaedtii with A. baerii or A. baerii hybrids has occurred in the Volga. Crosses and backcrosses of these specimens with native A. gueldenstaedtii resulted in the loss of the morphological diagnostic A. baerii features. These findings are of special concern for conservation and management programmes, as well as for specimen identification for caviar trading control. 相似文献

The objectives of this study were to compare the physicochemical properties of coelomic fluid (CF) in three sturgeon species, sterlet sturgeon Acipenser ruthenus (age 5–8 years), Siberian sturgeon Acipenser baerii (age 15–20 years), and Russian sturgeon Acipenser gueldenstaedtii (age 13–18 years). For the study, CF was collected by plastic pipette from the eggs of mature female sterlet sturgeon (N = 10), Siberian sturgeon (N = 7) and Russian sturgeon (N = 4); osmolality, pH, ionic composition (Na+, K+, Ca2+, Mg2+, Cl?), glucose, cholesterol and total protein of the CF were then analyzed. Mean concentration of K+ (4.4–6.11 mmol/L), Cl? (89.8–98.0 mmol/L), Ca2+ (0.87–0.96 mmol/L), Mg2+ (0.57–0.67 mmol/L), cholesterol (0.13–0.16 mmol/L), total protein (2.41–3.57 g/L), pH (7.92–7.98), and osmolality (190–213 mOsm/kg) of CF were not significantly different among these three species. However, the mean concentration of Na+ was significantly lower in sterlet sturgeon (104.6 ± 7.7 mmol/L; p < .05) than in the Siberian (126.4 ± 6.2) and Russian (123.0 ± 5.9) sturgeon. For these three species, Na+, K+, Cl?, and Mg2+ were the dominating ions; several significant correlations were observed among these ions and other physiochemical properties of CF. This is the first report on the ionic and biochemical composition of the CF of sterlet, Siberian and Russian sturgeon, which can be used as a reference point for further development of artificial media for the short‐term storage of unfertilized sturgeon eggs as well as for the standardization of the fertilization protocol for these species in controlled reproduction. 相似文献

Ovaries of Acipenser baerii are of an alimentary type and probably are meroistic. They contain ovarian nests, individual follicles, inner germinal ovarian epithelium, and fat tissue. Nests comprise cystoblasts, germline cysts, numerous early previtellogenic oocytes, and somatic cells. Cysts are composed of cystocytes, which are connected by intercellular bridges and are in the pachytene stage of the first meiotic prophase. They contain bivalents, finely granular, medium electron dense material, and nucleoli in the nucleoplasm. Many cystocytes degenerate. Oocytes differ in size and structure. Most oocytes are in the pachytene and early diplotene stages and are referred to as the PACH oocytes. Oocytes in more advanced diplotene stage are referred to as the DIP oocytes. Nuclei in the PACH oocytes contain bivalents and irregularly shaped accumulation of DNA (DNA‐body), most probably corresponding to the rDNA‐body. The DNA‐body is composed of loose, fine granular material, and comprises multiple nucleoli. At peripheries, it is fragmented into blocks that remain in contact with the inner nuclear membrane. In the ooplasm, there is the rough endoplasmic reticulum, Golgi complexes, free ribosomes, complexes of mitochondria with cement, fine fibrillar material containing granules, and lipid droplets. The organelles and material of nuclear origin form a distinct accumulation (a granular ooplasm) in the vicinity of the nucleus. Some of the PACH oocytes are surrounded by flat somatic cells. There are lampbrush chromosomes and multiple nucleoli present (early diplotene stage) in the nucleoplasm. These PACH oocytes and neighboring somatic cells have initiated the formation of ovarian follicles. The remaining PACH oocytes transform to the DIP oocytes. The DIP oocytes contain lampbrush chromosomes and a DNA‐body is absent in nuclei. Contrary to the opinion of Kynard et al. (2002), the Siberian sturgeon, Acipenser baerii, does not belong to the Ponto-Caspian species. It inhabits Siberian rivers and Lake Baikal. Acipenser baerii is a typical potamodromous species and the comparison of the behavior of its embryos and larvae with those of the anadromous Russian sturgeon, Acipenser gueldenstaedtii, should be done with understanding that these species have different life histories. The present study was carried out with two sturgeon species, Siberian sturgeon (Acipenser baerii) and sterlet (A. ruthenus) to compare spermatological parameters to better understand inter‐species differences. Significant differences between morphometric features were observed such as acrosome length, acrosome width, head length, midpiece width and flagellar length, while midpiece length did not reveal such differences. The sterlet has a shorter spermatozoon than the Siberian sturgeon. Ultrastructural parameters vary significantly in terms of length of the nucleus, diameter of the endonuclear canals (EC), size of posterolateral projections (PP) and diameter of flagellum. Mean values for density of spermatozoa in the semen, seminal plasma pH, osmolality (mOsmol kg?1), along with Ca2+, Na+, K+, Cl? ions concentrations (mm ) were determined to be 0.61 ± 0.37 × 109, 8.16 ± 0.18, 77.20 ± 52.28, 0.24 ± 0.06, 31.39 ± 10.21, 3.51 ± 1.10, 14.00 ± 4.30 in A. baerii and 0.41 ± 0.32 × 109, 8.13 ± 0.19, 50.74 ± 6.27, 0.16 ± 0.11, 20.11 ± 3.78, 1.26 ± 0.54, 6.11 ± 0.60 in A. ruthenus, respectively. Significant differences were observed in Na+, K+ and Cl? concentrations in the seminal plasma as well as in sperm velocity. The percentage of motile spermatozoa did not show any significant difference between the two species. Comparing the results of this study with published literature data on sturgeon spermatozoa reveals that morphological and ultrastructural parameters of spermatozoa together with some parameters of the seminal fluid and spermatozoa velocity can be used in comparative spermatology to better understand inter‐species differences. The observed biochemical and physiological differences should be also considered for the development of methods for controlled reproduction and for sperm cryopreservation techniques. 相似文献

The fatty acid metabolism in fish is influenced by various factors, including fish species, water temperature, water environment and diet supply. The aim of present work is to investigate the fatty acid composition of yolk‐stage Siberian sturgeon larvae reared at three different temperatures. Fertilized Siberian sturgeon eggs were transferred to the Lodi Aquaculture Research Center of the University of Milan, divided in three aquaria, each containing three incubators and incubated at 16°C. After hatching the temperature was switched to 16, 19 and 22°C. Larvae sampling was performed at the end of yolk sac reabsorption. No feed was dispensed during the trial. Eggs and larvae were weighed and fatty acid profile was determined by GC‐FID analysis after lipid extraction by chloroform/methanol mixture and fatty acid transesterification by methanolic hydrogen chloride. The fertilized eggs had a weight of 23.27 mg and a lipid content of 2.67 mg/egg. At hatching, the weight was 12.2 (0.17 SD) mg and lipid content 1.9 (0.6 SD) mg/larva. At the end of the trial, larvae mean weight was 33.6 (3.6 SD), 34.7 (1.8 SD) and 36.9 (1.1 SD) mg, while lipid content was 2.0 (0.3 SD), 2.1 (0.3 SD) and 2.0 (0.2 SD) mg for larvae reared at 16, 19 and 22°C respectively, without statistically significant difference. Larvae subjected to the highest water temperature showed a faster yolk‐sac absorption. No differences were found across temperatures regarding survival rates and regarding ontogenic development. The fatty acid composition of larvae was affected by the temperature. Larvae reared at 16°C had the lowest amount of saturated fatty acids, mainly due to a lower palmitic acid content, that was offset by a higher level of linolenic and linoleic acid, if compared with larvae reared at 19°C and 22°C. The study suggests that at a lower temperature sturgeon spare unsaturated fatty acid consuming preferably saturated fatty acids, increasing our knowledge of the fatty acid metabolism in this species. 相似文献

BACKGROUND INFORMATION: Available data concerning the sperm morphology of teleost fishes demonstrate wide variation. In the present study, the spermatozoa of Siberian sturgeon (Acipenser baerii Brandt, 1869), a chondrostean fish, was investigated. In contrast with teleost fish, chondrostean spermatozoa have a head with a distinct acrosome, whereas other structures, such as a midpiece and a single flagellum, are present in spermatozoa of most species. RESULTS: The average length of the head including the acrosome and the midpiece was 7.01+/-0.83 microm. Ten posterolateral projections derived from the acrosome were present on a subacrosomal region, with mean lengths of 0.94+/-0.15 microm and widths of 0.93+/-0.11 microm. The nucleus consisted of electrodense homogeneous nuclear chromatin. Three intertwining endonuclear canals, bound by membranes, traversed the nucleus longitudinally from the acrosomal end to the basal nuclear fossa region. There were between three and six mitochondria, two types of centrioles (proximal and distal) in the midpiece and two vacuoles composed of lipid droplets. The flagellum (44.75+/-4.93 microm in length), originating from the centriolar apparatus, had a typical 9+2 eukaryotic flagellar organization. In addition, there was an extracellular cytoplasm canal between the cytoplasmic sheath and the flagellum. CONCLUSIONS: A principal components analysis explained the individual morphological variation fairly well. Of the total accumulated variance, 41.45% was accounted for by parameters related to the head and midpiece of the sperm and the length of the flagellum. Comparing the present study with previous studies of morphology of sturgeon spermatozoa, there were large inter- or intra-specific differences that could be valuable taxonomically. 相似文献

Receptor-interacting serine/threonine-protein kinase 2 (RIPK2) is an adaptor protein of the pattern recognition receptors NOD1 and NOD2 involved in regulating inflammatory response and resisting pathogenic microbial infection. In this study, Acipenser baerii RIPK2 (AbRIPK2) was identified. The open reading frame of AbRIPK2 was 1815 bp encoding 604 amino acids. AbRIPK2 possessed the typical N-terminal kinase domain (KD) and C-terminal caspase recruitment domain (CARD). The phylogenetic tree analysis revealed that AbRIPK2 shared a relatively high identity with bony fish. Real-time fluorescence quantitative PCR (qRT-PCR) results indicated that AbRIPK2 was highly expressed in the gill, followed by muscle, liver and heart. AbRIPK2 was significantly induced in the spleen and valvular intestine after Streptococcus iniae and Aeromonas hydrophila infection. AbRIPK2 was significantly upregulated after peptidoglycan (PGN) treatment in the splenic leukocytes. This study indicated that AbRIPK2 played a potential role in resisting the pathogenic infection of Siberian sturgeon by responding to bacteria. 相似文献

Synopsis We studied Sacramento River white sturgeon, Acipenser transmontanus, in the laboratory to develop a conceptual model of ontogenetic behavior and provide insight into probable behavior of wild
sturgeon. After hatching, free embryos initiated a low intensity, brief downstream dispersal during which fish swam near the
bottom and were photonegative. The weak, short dispersal style and behavior of white sturgeon free embryos contrasts greatly
with the intense, long dispersal style and behavior (photopositive and swimming far above the bottom) of dispersing free embryos
of other sturgeon species. If spawned eggs are concentrated within a few kilometers downstream of a spawning site, the adaptive
significance of the free embryo dispersal is likely to move fish away from the egg deposition site to avoid predation and

sturgeon populations may be a mis-match between the innate fish dispersal and post-dispersal rearing habitat, which is now

Nocturnal behavior is likely a response to predation, which occurs during both activities. An intense black-tail body color
