We recently reported the detection of methanol emissions from leaves (R. MacDonald, R. Fall [1993] Atmos Environ 27A: 1709-1713). This could represent a substantial flux of methanol to the atmosphere. Leaf methanol production and emission have not been investigated in detail, in part because of difficulties in sampling and analyzing methanol. In this study we used an enzymatic method to convert methanol to a fluorescent product and verified that leaves from several species emit methanol. Methanol was emitted almost exclusively from the abaxial surfaces of hypostomatous leaves but from both surfaces of amphistomatous leaves, suggesting that methanol exits leaves via stomates. The role of stomatal conductance was verified in experiments in which stomates were induced to close, resulting in reduced methanol. Free methanol was detected in bean leaf extracts, ranging from 26.8 [mu]g g-1 fresh weight in young leaves to 10.0 [mu]g g-1 fresh weight in older leaves. Methanol emission was related to leaf development, generally declining with increasing leaf age after leaf expansion; this is consistent with volatilization from a cellular pool that declines in older leaves. It is possible that leaf emission could be a major source of methanol found in the atmosphere of forests.

Midday depression of net photosynthesis and transpiration in the Mediterranean sclerophylls Arbutus unedo L. and Quercus suber L. occurs with a depression of mesophyll photosynthetic activity as indicated by calculated carboxylation efficiency (CE) and constant diurnal calculated leaf intercellular partial pressure of CO2 (Ci). This work examines the hypothesis that this midday depression can be explained by the distribution of patches of either wide-open or closed stomata on the leaf surface, independent of a coupling mechanism between stomata and mesophyll that results in a midday depression of photosynthetic activity of the mesophyll. Pressure infiltration of four liquids differing in their surface tension was used as a method to show the occurrence of stomatal patchiness and to determine the status of stomatal aperture within the patches. Liquids were selected such that the threshold leaf conductance necessary for infiltration through the stomatal pores covered the expected diurnal range of calculated leaf conductance (g) for these species. Infiltration experiments were carried out with leaves of potted plants under simulated Mediterranean summer conditions in a growth chamber. For all four liquids, leaves of both species were found to be fully infiltratable in the morning and in the late afternoon while during the periods leading up to and away from midday the leaves showed a pronounced patchy distribution of infiltratable and non-infiltratable areas. Similar linear relationships between the amount of liquid infiltrated and g (measured by porometry prior to detachment and infiltration) for all liquids clearly revealed the existence of pneumatically isolated patches containing only wide-open or closed stomata. The good correspondence between the midday depression of CE, calculated under the assumption of no stomatal patchiness, and the diurnal changes in non-infiltratable leaf area strongly indicates that the apparent reduction in mesophyll activity results from assuming no stomatal patchiness. It is suggested that simultaneous responses of stomata and mesophyll activity reported for other species may also be attributed to the occurrence of stomatal patchiness. In Quercus coccifera L., where the lack of constant diurnal calculated Ci and major depression of measured CE at noontime indicates different stomatal behavior, non-linear and dissimilar relationships between g and the infiltratable quantities of the four liquids were found. This indicates a wide distribution of stomatal aperture on the leaf surface rather than only wide-open or closed stomata.Dedicated to Professor Otto L. Summary Pressure infiltration of water into a leaf via the stomatal pores can be used to quickly determine whether all stomata are open, or as recently described for several mesophytic and xerophytic species, whether there is a non-homogeneous distribution of stomatal opening (stomatal patchiness) on the leaf surface. Information about this phenomenon is important since the commonly used algorithms for calculation of leaf conductance from water vapor exchange measurements imply homogeneously open stomata, which in the occurrence of stomatal patchiness will lead to erroneous results. Infiltration experiments in a growth chamber with leaves of the Mediterranean evergreen shrub Arbutus unedo, carried out under simulated Mediterranean summer day conditions, where the species typically exhibits a strong midday stomatal closure, revealed a temporary occurrence of stomatal patchiness during the phase of stomatal closure in the late morning and during the stomatal reopening in the afternoon. Leaves were, however, found to be fully (i.e. homogeneously) infiltratable in the morning and in the evening. At midday during maximum stomatal closure, leaves were almost non-infiltratable. During the day, the infiltrated amount of water was found to be linearly correlated with porometer measurements of leaf conductance of the same leaves, carried out with the attached leaves immediately before infiltration.

Plant leaves emit a broad spectrum of organic compounds that typically play multiple roles in plant protection. Furthermore, most of these compounds actively participate in tropospheric chemistry. There has been rapid progress in understanding how the emission of volatiles is regulated, mostly focusing on the biochemical controls over compound production. However, physicochemical characteristics such as low volatility or diffusion can also control the emissions and interact with physiological limitations. In particular, non-specific leaf storage of less volatile compounds smooths the emission responses to fluctuating environmental conditions, and diffusion through stomata leads to conspicuous emission bursts after stomatal opening and modifications of diurnal emission time courses. Because natural conditions always fluctuate, both physiological and physicochemical controls exert a major influence over plant volatile emissions.

Diurnal pattern of acetaldehyde emission by flooded poplar trees
The emission of the tropospheric trace gas acetaldehyde was determined in leaves of 4-month-old poplar trees ( Populus tremula × P. alba ) grown under controlled environmental conditions in a greenhouse. Using a dynamic cuvette system together with a high sensitivity laser-based photoacoustic detection unit, rates of acetaldehyde emission were measured with the high time resolution of about 15 min. Submergence of the roots resulted in the emission of acetaldehyde by the leaves. The emission increased linearly before reaching more or less steady-state values (ca 350 nmol m−2 min−1; ca 470 ng g−1 dry weight min−1) after approximately 6 h. Prolonged flooding of poplar trees resulted in a clear diurnal rhythm of acetaldehyde emission. The emission rates decreased when the light was switched off in the evening and peaked in the morning after the light was turned on again. This pattern significantly correlated with diurnal rhythms of stomatal conductance, photosynthesis, transpiration and with the concentrations of ethanol, the assumed precursor of acetaldehyde, in the xylem sap of flooded poplar trees. It may be concluded that under conditions of diminished stomatal conductance, acetaldehyde emission declines because its diffusive flux is reduced. Alternatively, reduced transpiration may decrease ethanol transport from the roots to the shoots and appreciable amounts of the acetaldehyde precursor ethanol are lacking in the leaves. The present results support the view that acetaldehyde emitted by the leaves of plants is derived from ethanol produced by alcoholic fermentation in submerged roots and transported to the leaves with the transpiration stream.

Responses of abaxial and adaxial stomata of Populus trichocarpa Torr. & Gray. × P. deltoides Bartr. (ex Marsh.) cv. Unal to incident light, sudden darkening and leaf excision in the light and in the dark were studied on 5-year-old trees in the field using diffusion porometry. Stomatal closure in the dark was found to be incomplete in most cases studies. Stomata closed after leaf excision in the dark within 90 min. Stomatal closure after darkening of an entire tree or an entire branch (white the rest of the tree was in the light) was slower, and complete stomatal closure was noticed only for adaxial stomata after 3 h. Adaxial stomata were more reactive and sensitive than abaxial stomata to sudden darkening and leaf excision in the light and the dark. In all treatments, stomatal response was more responsive in mature leaves than in young, still expanding leaves.

It is generally accepted that various physiological, morphological and gene expression phenomena are under the control of a circadian clock, and that this time keeping mechanism is universally present. Although such endogenously regulated phenomena have first been documented in plants more than 250 years ago and much work has been accumulated particularly in the past 70 years, it was not obvious from the literature whether such time keeping mechanisms exist in gymnosperms. Two prominent parameters were investigated in several gymnosperm species which have been demonstrated to be under the control of a circadian clock in many plants: (i) leaf movement and (ii) stomata movement. In young plants of Pinus sylvestris, Picea abies, Taxus baccata, Araucaria angustifolia, Araucaria heterophylla and Ginkgo biloba leaf oscillations could be recorded for about 5 days. However, compared to angiosperm plants, the amplitude was small. The period length under free running conditions (constant temperature and continuous light) was characteristic for the species. Stomatal movement was observed in Ginkgo biloba leaves by electron microscopy. Stomata were open at noon and closed at midnight under normal day/night conditions (LD) as well as under constant light conditions (LL), indicating that stomatal aperture is under circadian control in the gymnosperm Ginkgo biloba. Online recordings of stomata conductance however, exhibited diurnal but not circadian oscillations of net CO2-exchange in G. biloba leaves. Our results show that a circadian clock controls leaf and stomatal movements in gymnosperm species indicating that endogenous time keeping mechanisms are present.

Circadian Stomatal Rhythms in Epidermal Peels from Vicia faba
Circadian rhythms in stomatal aperture and in stomatal conductance have been observed previously. Here we investigate circadian rhythms in apertures that persist in functionally isolated guard cells in epidermal peels of Vicia faba, and we compare these rhythms with rhythms in stomatal conductance in attached leaves. Functionally isolated guard cells kept in constant light display a rhythmic change in aperture superimposed on a continuous opening trend. The rhythm free-runs with a period of about 22 hours and is temperature compensated between 20 and 30°C. Functionally isolated guard cell pairs are therefore capable of sustaining a true circadian rhythm without interaction with mesophyll cells. Stomatal conductance in whole leaves displays a more robust rhythm, also temperature-compensated, and with a period similar to that observed for the rhythm in stomatal aperture in epidermal peels. When analyzed individually, some stomata in epidermal peels showed a robust rhythm for several days while others showed little rhythmicity or damped out rapidly. Rhythmic periods may vary between individual stomata, and this may lead to desynchronization within the population.

Summary In Australia, diurnal courses of leaf conductance and transpiration of hemiparasitic mistletoes (Loranthaceae) and their hosts were measured using steady-state porometers under conditions of partial drought and high evaporative demand. The sites spanned a diversity of climatic regions ranging from the subtropical arid zone with winter rainfall, through the subtropical arid zone with summer rainfall to the tropical summer rainfall zone. With one exception (Acacia farnesiana with deciduous leaves), the hosts were trees or shrubs with evergreen, sclerophyllous leaves or phyllodes. The measurements confirm previous observations that mistletoes transpire at higher rates than their hosts. For adult leaves from all of the 18 different host/mistletoe pairs investigated, the daily average leaf conductances were higher in the parasites than in their hosts. The ratios ranged from 1.5 to 7.9. In the most extreme case,Amyema maidenii had a daily rate of water loss 8.9 times higher than its hostAcacia cowleana. Hoever, the parasites did not exhibit unlimited transpiration. Despite high water loss rates, leaf conductance showed large and consistent changes during the course of the day, indicating definite stomatal regulation. The typical diurnal pattern of conductance in both mistletoes and hosts consisted of an early morning peak followed by a continuous decrease throughout the remainder of the day. There was no abrupt decrease in leaf conductance of the parasites that might be interpreted as a threshold response with respect to internal water potential. In most cases, the continuous stomatal closure occurred without substantial changes in leaf water potential over a time span of several hours. The decrease in leaf conductance was correlated with an increase in leaf-to-air water vapor difference, which was associated with increasing leaf temperatures. It seems probable that external humidity plays a major role in the stomatal response. Diurnal courses of leaf conductance of the host/parasite pairs usually showed similar general patterns, even when the absolute rates were quite different. Thus, mistletoes not only control their water loss by stomatal action but this regulation seems to occur in coordination with the stomatal response of their hosts. The integrated mistletoe/host system must also endure severe drought conditions. Controlled water use is necessary for long-term survival of the host. Assuming stomatal behavior in the host is well adapted to ensure its existence, then similar performance in the mistletoe would promote survival of both host and parasite.

Uptake of CO2 by the leaf is associated with loss of water. Control of stomatal aperture by volume changes of guard cell pairs optimizes the efficiency of water use. Under water stress, the protein kinase OPEN STOMATA 1 (OST1) activates the guard‐cell anion release channel SLOW ANION CHANNEL‐ASSOCIATED 1 (SLAC1), and thereby triggers stomatal closure. Plants with mutated OST1 and SLAC1 are defective in guard‐cell turgor regulation. To study the effect of stomatal movement on leaf turgor using intact leaves of Arabidopsis, we used a new pressure probe to monitor transpiration and turgor pressure simultaneously and non‐invasively. This probe permits routine easy access to parameters related to water status and stomatal conductance under physiological conditions using the model plant Arabidopsis thaliana. Long‐term leaf turgor pressure recordings over several weeks showed a drop in turgor during the day and recovery at night. Thus pressure changes directly correlated with the degree of plant transpiration. Leaf turgor of wild‐type plants responded to CO2, light, humidity, ozone and abscisic acid (ABA) in a guard cell‐specific manner. Pressure probe measurements of mutants lacking OST1 and SLAC1 function indicated impairment in stomatal responses to light and humidity. In contrast to wild‐type plants, leaves from well‐watered ost1 plants exposed to a dry atmosphere wilted after light‐induced stomatal opening. Experiments with open stomata mutants indicated that the hydraulic conductance of leaf stomata is higher than that of the root–shoot continuum. Thus leaf turgor appears to rely to a large extent on the anion channel activity of autonomously regulated stomatal guard cells.

In the spring of 1987, entire shoots were removed from Quercus rubra L. saplings in two southwestern Wisconsin forest openings. Shoots possessed newly expanding leaves at the time of coppicing. All coppiced individuals sprouted from dormant stem buds near the root collar. Leaf gas exchange and water potential were monitored on these sprouts and on untreated (control) Q. rubra saplings throughout several clear warm days during the 1987 growing season. Daily maxima and averages for sprout leaf photosynthesis and stomatal conductance generally exceeded those of controls. On average, treatment differences in daily maximum photosynthetic rate were modest (11–14%) and were attributed primarily to a 30–38% enhancement of sprout leaf stomatal conductance. Relative differences in daily average photosynthetic rate (29–39%) were substantially larger than those in daily maximum photosynthesis, owing to the fact that sprouts and controls exhibited distinct diurnal gas exchange patterns. Photosynthetic rate and stomatal conductance of control leaves typically declined during the day following a mid-morning maximum. Sprout leaves, on the other hand, tended to maintain gas exchange rates nearer to their morning maxima throughout the day. This difference in diurnal gas exchange pattern was associated with an apparent differential leaf sensitivity to leaf-to-air vapor pressure gradient (VPG). The relative decline in sprout leaf gas exchange rates with increasing VPG was less than that of controls. Treatment differences in gas exchange did not appear to be related to leaf water potential or tissue water relations, but sprouts had a higher soil-to-leaf hydraulic conductivity than controls.

The coordination of veins and stomata during leaf acclimation to sun and shade can be facilitated by differential epidermal cell expansion so large leaves with low vein and stomatal densities grow in shade, effectively balancing liquid‐ and vapour‐phase conductances. As the difference in vapour pressure between leaf and atmosphere (VPD) determines transpiration at any given stomatal density, we predict that plants grown under high VPD will modify the balance between veins and stomata to accommodate greater maximum transpiration. Thus, we examined the developmental responses of these traits to contrasting VPD in a woody angiosperm (Toona ciliata M. Roem.) and tested whether the relationship between them was altered. High VPD leaves were one‐third the size of low VPD leaves with only marginally greater vein and stomatal density. Transpirational homeostasis was thus maintained by reducing stomatal conductance. VPD acclimation changed leaf size by modifying cell number. Hence, plasticity in vein and stomatal density appears to be generated by plasticity in cell size rather than cell number. Thus, VPD affects cell number and leaf size without changing the relationship between liquid‐ and vapour‐phase conductances. This results in inefficient acclimation to VPD as stomata remain partially closed under high VPD.

Stomatal control of transpiration from a developing sugarcane canopy
Abstract. Stomatal conductance of single leaves and transpiration from an entire sugarcane (Saccharum spp. hybrid) canopy were measured simultaneously using independent techniques. Stomatal and environmental controls of transpiration were assessed at three stages of canopy development, corresponding to leaf area indices (L) of 2.2, 3.6 and 5.6. Leaf and canopy boundary layers impeded transport of transpired water vapour away from the canopy, causing humidity around the leaves to find its own value through local equilibration rather than a value determined by the humidity of the bulk air mass above the canopy. This tended to uncouple transpiration from direct stomatal control, so that transpiration predicted from measurement of stomatal conductance and leaf-to-air vapour pressure differences was increasingly overestimated as the reference point for ambient vapour pressure measurement was moved farther from the leaf and into the bulk air. The partitioning of control between net radiation and stomata was expressed as a dimensionless decoupling coefficent ranging from zero to 1.0. When the stomatal aperture was near its maximum this coefficient was approximately 0.9, indicating that small reductions in stomatal aperture would have had little effect on canopy transpiration. Maximum rates of transpiration were, however, limited by large adjustments in maximum stomatal conductance during canopy development. The product of maximum stomatal conductance and L. a potential total canopy conductance in the absence of boundary layer effects, remained constant as L increased. Similarly, maximum canopy conductance, derived from independent micrometeorological measurements, also remained constant over this period. Calculations indicated that combined leaf and canopy boundary layer conductance decreased with increasing L such that the ratio of boundary layer conductance to maximum stomatal conductance remained nearly constant at approximately 0.5. These observations indicated that stomata adjusted to maintain both transpiration and the degree of stomatal control of transpiration constant as canopy development proceeded.

Aspects of leaf anatomical and physiological development were investigated in the broad-leaved evergreen species, Eucalyptus regnans F.Muell. Newly emergent leaves were tagged in the field and measured for stomatal conductance while a subset was collected every 14 days for the measurement of stomata and cuticle over a 113-day period. Cuticle thickness increased during leaf expansion, the increase following a sigmoid curve. Stomatal frequency (no. mm−2) decreased from 56 to 113 days after leaf emergence. The frequency of both immature and intermediate developmental stages of stomata also decreased over this time, but the total number of stomata per leaf remained relatively constant. Stomatal conductance (g s) of young expanding leaves increased during expansion, and was significantly linearly correlated with stomatal frequency (excluding immature stomata), and with cuticle thickness. The progressive increase in g s in young developing leaves was contrary to the observed changes in structural characteristics (increased cuticle thickness and decreased stomatal frequency). This increase in g s with development may be related to the progressive increase in number of mature stomata with larger apertures and, therefore, a higher total pore area in fully expanded leaves.

Temperature of leaves or canopies can be used as an indicator of stomatal aperture, which is considered a sensitive response to soil water deficit. In this paper we analyse the robustness and sensitivity of thermal imaging for detecting changes in stomatal conductance and leaf water status in a range of plant species. Thermal imaging successfully distinguished between irrigated and non-irrigated plants of a variety of species under greenhouse or controlled chamber conditions, with strong correlations between thermal indices and stomatal conductance measured by porometry. Our results also highlighted issues that need to be addressed in order to be confident of always detecting drought stress using this technique. These include variability in leaf angles and the limited reliability of 'wet' and 'dry' leaves to represent leaves with stomata fully open or stomata fully closed. These results should assist the design of protocols for application in crop production or ecosystem monitoring.

Isoprene emission in relation to stomatal distribution and conductance was determined for the hypostomatous species, aspen and white oak, and the amphistomatous species, cottonwood. For aspen and oak, isoprene emission from the adaxial (nonstomatal) surface was <2% of that from the abaxial (stomatal) surface, even when stomata were closed by addition of abscisic acid (ABA). When treated with ABA, the total flux rate of isoprene emission from leaves of these two hypostomatous species was unchanged, despite decreases in stomatal conductance of over 90%. The lack of control over isoprene emission rate by stomatal conductance, despite the apparent movement of isoprene through the stomatal pores, was due to increases in the intercellular isoprene concentration that compensated for the decreased stomatal conductance and restored the equilibrium between the isoprene synthesis rate and emission rate. This relationship was demonstrated by (a) an experiment in which the decrease in the internal isoprene pool following the imposition of darkness took longer in the presence of ABA than in its absence, and (b) direct measurements of the internal isoprene concentration through vacuum extraction, which revealed substantially higher values in the presence of ABA than in its absence. In the amphistomatous species, cottonwood, isoprene was emitted from both surfaces and addition of ABA caused an increase in isoprene emission from one surface coupled with a decrease from the other surface. The specific surface exhibiting an increase varied among leaves, with some leaves exhibiting an increase from the adaxial surface and other leaves from the abaxial surface. We interpret this as indicating nonuniform stomatal closure with concomitant emission of isoprene at the greatest rate from the surface with the highest stomatal conductance. We also observed an increase in the total isoprene emission rate from cottonwood leaves following treatment with ABA. We interpret this as indicating a stimulation of isoprene synthesis in response to ABA or stomatal closure, with unknown cause.
