High purity monoacylglycerol (MAG) containing pinolenic acid was synthesized via stepwise esterification of glycerol and fatty acids from pine nut oil using a cold active lipase from Penicillium camembertii as a biocatalyst. Effects of temperature, molar ratio, water content, enzyme loading, and vacuum on the synthesis of MAG by lipase‐catalyzed esterification of glycerol and fatty acid from pine nut oil were investigated. Diacylglycerol (DAG) as well as MAG increased significantly when temperature was increased from 20 to 40°C. At a molar ratio of 1:1, MAG content decreased because of the significant increase in DAG content. Water has a profound influence on both MAG and DAG content through the entire course of reaction. The reaction rate increased significantly as enzyme loading increased up to 600 units. Vacuum was an effective method to reduce DAG content. The optimum temperature, molar ratio, water content, enzyme loading, vacuum, and reaction time were 20°C, 1:5 (fatty acid to glycerol), 2%, 600 units, 5 torr, and 24 h, respectively. MAG content further increased via lipase‐catalyzed second step esterification at subzero temperature. Sesamin is a specific inhibitor of Δ5 desaturation, the conversion from dihomo-γ-linolenic acid (20: 3, n-6) to arachidonic acid (AA, 20: 4, n-6). Previously, we reported that sesamin inhibited Δ5 desaturation of n-6 fatty acids in rat hepatocytes but not that of n-3 fatty acids, from 20: 4 (n-3) to eicosapentaenoic acid (EPA, 20: 5, n-3). In this study, we investigated the interaction of sesamin and EPA on Δ5 desaturation of both series and the n-6/n-3 fatty acids ratio by measuring actural fatty acid contents in vivo. Rats were fed three types of dietary oils; 1) linoleic acid (LA, 18: 2, n-6): linolenic acid (LLA, 18: 3, n-3) = 3: 1, n-6/n-3 ratio of 3: 1 (LA group), 2) LA: LLA =1: 3, n-6/n-3 ratio of 1: 3 (LLA group), 3) LA: LLA: EPA =1: 0.5: 3, n-6/n-3 ratio of 1: 3.5 (EPA group) with or without sesamin (0.5% w/w) for 4 weeks. In all groups, sesamin administration increased the content of dihomo-γ-linolenic acid (20: 3, n-6) in the liver and decreased the Δ5 desaturation index of n-6 fatty acid, the ratio of 20: 4/20: 3 (n-6). On the contrary, the Δ5 desaturation index of n-3 fatty acid, the ratio of 20: 5 + 22: 5 + 22: 6/20: 4 (n-3), was increased by the administration of sesamin. These results suggest that sesamin inhibits the A5 desaturation of n-6 fatty acid, but not that of n-3 fatty acid in rat livers. Sesamin administration decreased incorporation of EPA (n-3) and simultaneously increased the AA (n-6) content in the liver. The n-6/n-3 ratio in the liver was increased by administering sesamin under n-3 rich conditions, i.e., the LLA and EPA groups.  相似文献

Pathways of lipid resynthesis in the intestine of fish are relatively unknown. Various reports have suggested the existence of both sn-1,3-specific (pancreatic) and non-specific (bile salt-activated) lipase activity operating on dietary triacylglycerol (TAG) in the intestinal lumen of fish during digestion. Thus, sn-2-monoacylglycerol (2-MAG) and glycerol, respective hydrolytic products of each lipase, are absorbed and utilised for glycerolipid synthesis in enterocytes via two alternative routes: monoacylglycerol (MAG) and glycerol-3-phosphate (G3P) pathways. Despite different precursors, both pathways converge at the production of sn-1,2-diacylglycerol (1,2-DAG) where TAG or phosphatidylcholine (PC) synthesis can occur. To elucidate the relative activities of MAG and G3P pathways in Atlantic salmon enterocytes, intestinal segments were mounted in Ussing chambers where equimolar mixtures of sn-2-oleoyl-[1,2,3-(3)H]glycerol (2-MAG) and [(14)C(U)]glycerol, plus unlabelled 16:0 and 18:2n-6 as exogenous fatty acid sources, were delivered in bile salt-containing Ringer solution to the mucosa. The MAG pathway predominated, over the G3P pathway, synthesizing ca. 95% of total TAG and ca. 80% of total PC after a 3 h incubation period at 10 degrees C. Further, the 1,2-DAG branch point into TAG or PC was polarised towards TAG synthesis (6:1) via the MAG pathway but more evenly distributed between TAG and PC (1:1) via the G3P pathway. Effect of long-chain saturated, monounsaturated and polyunsaturated fatty acids on the synthesized TAG/PC ratio was assessed by individually exchanging 16:0, 18:1n-9 or 18:2n-6, for 16:0+18:2n-6, in mucosal solutions. TAG synthesis was influenced considerably more than PC synthesis, via either pathway, by exogenous fatty acids utilised. 18:1n-9 significantly stimulated TAG synthesis via the MAG pathway yielding a TAG/PC ratio of 12:1. Alternatively, 18:2n-6 stimulated TAG synthesis the most via the G3P pathway (TAG/PC=4:1). 16:0 significantly attenuated TAG synthesis via either pathway. Micellar fatty acid species also significantly affected intestinal active transport mechanisms as shown by decreasing transepithelial potential (TEP) and short-circuit current (SSC) with increasing fatty acid unsaturation. The epithelial integrity was, however, not compromised after 3 h of exposure to any of the fatty acids. The implications of these findings on dietary fatty acid composition and enterocytic lipid droplet accumulation are discussed.  相似文献

The Δ9-elongase isolated from Thraustochytrium aureum, which contains a high level of polyunsaturated fatty acids (PUFAs), was demonstrated to be associated with the synthesis of C20 PUFAs. The TaELO gene contains a 825 bp ORF that encodes a protein of 274 amino acids that shares a high similarity with other PUFA elongases. The expression of the TaELO gene in Pichia pastoris resulted in the elongation of linoleic acid (LA, C18:2; n-6) and α-linolenic acid (ALA, C18:3; n-3) to eicosadienoic acid (EDA, C20:2; n-6) and eicosatrienoic acid (ETrA, C20:3; n-3), respectively. The endogenous conversion rate of LA and ALA to EDA and ETrA was 32.68 and 38.57%, respectively. In addition, TaELO was also able to synthesize eicosenoic acid (C20:1; n-9) from oleic acid (OA, C18:1; n-9), even though the conversion level was low (2.81%). Furthermore, TaELO was able to carry out the 6Δ-elongation of γ-linolenic acid (GLA, C18:3; n-6) to dihomo-γ-linolenic acid (DGLA, C20:3; n-6) and Δ5-elongation of eicosapentaenoic acid (EPA, C20:5; n-3) to docosapentaenoic acid (DPA, C22:5; n-3). The conversion rate of GLA to DGLA and EPA to DPA were 93 and 28.36%, respectively. The TaELO protein was confirmed to have multifunctional activities, such as Δ9, Δ6, and Δ5-elongations as well as the elongation of monounsaturated fatty acid.  相似文献

Abstract

The fatty acid composition, moisture, and total lipid of the eggs from the swimming crab, Portunus pelagicus, at three different embryonic stages (within 24 h, during the eye placode stage and the final heart beat stage), were measured. Results showed that the moisture and lipid content significantly increased and decreased (p < 0.05), respectively, as the stages progressed. The most prevalent fatty acids that were initially deposited included C16:0, C18:1n-9, and C18:0, while the most consumed fatty acids were C22:5n-6, C22:5n-3, and C20:1n-7. Among the major fatty acid groups, polyunsaturated fatty acids (PUFA) and long-chain PUFA (LC-PUFA) were consumed more than saturated fatty acids and significantly more (p < 0.05) than monounsaturated fatty acids (p < 0.05). Meanwhile, n-3 PUFA was deposited in significantly higher amounts (p < 0.05) than n-6 PUFA, but both were consumed at similar amounts at 43.4% and 41.3%, respectively. The relatively low amount of C20:5n-3 and C22:6n-3 consumption may indicate these fatty acids were conserved, while the essential fatty acids C18:3n-3 and C18:3n-6 were consumed at high amounts. These findings may have implications for broodstock nutrition in order to formulate a well-balanced diet.  相似文献

Zooxanthella nutricula is a photosynthetic dinoflagellate symbiont of polycystine radiolarians. As such, it is hypothesized to provide fixed organic carbon, including in the form of acylglycerolipids and sterols, to its non-photosynthetic host. We have previously characterized the sterols of Z. nutricula that may be transferred to its host and, in the present study, have turned our attention to three classes of fatty acid-containing lipids, chloroplast-associated galactolipids, betaine lipids, which are non-phosphorylated phospholipid analogs present in many eukaryotes, and triglycerides. Zooxanthella nutricula was observed using positive-ion electrospray/mass spectrometry (ESI/MS) and ESI/MS/MS to produce the galactolipids mono- and digalactosyldiacylglycerol (MGDG and DGDG, respectively) enriched in octadecapentaenoic (18:5(n-3)) and octadecatetraenoic (18:4(n-3)) acid to place it within a group of peridinin-containing dinoflagellates in a C18/C18 (sn-1/sn-2 fatty acid regiochemistry) cluster, as opposed to another cluster with C20/C18 MGDG and DGDG, where the C20 fatty acid is eicosapentaenoic acid (20:5(n-3)) and the C18 fatty acid is either 18:5(n-3) or 18:4(n-3). Zooxanthella nutricula was also observed to produce 38:10 (total number of fatty acid carbons:total number of double bonds), 38:6, and 44:7 diacylglycerylcarboxyhydroxymethylcholine (DGCC) as the sole type of betaine lipid. Although it is more difficult to determine which fatty acids are present in the sn-1 and sn-2 positions on the glycerol backbone of DGCC using ESI/MS/MS, gas chromatography/mass spectrometry (GC/MS)-based examination indicated the putatively DGCC-associated polyunsaturated fatty acid (PUFA) docosahexaenoic acid (22:6(n-3)). Coupled with the C18 PUFAs of MGDG and DGDG, and fatty acids associated with triglycerides (also examined via GC/MS), Z. nutricula could serve as a rich source of PUFAs for its radiolarian host. These data demonstrate that Z. nutricula produces a similar set of PUFA-containing lipids as Symbiodinium microadriaticum, a photosynthetic dinoflagellate symbiont of cnidarians, indicating a metabolic commonality in these phylogenetically discrete dinoflagellate symbionts with unrelated host organisms.  相似文献

Phaeodactylum tricornutum and Chaetoceros sp. (Badllariophyceae), Isochrysis galbana (clone T-Iso) and Pavlova lutheri (Prymnesiophyceae), Nannochloris atomus (Chlorophyceae), Tetraselmis sp. (Prasinophyceae), and Gymnodinum sp. (Dinophyceae) were cultured at different extents of nutrient-limited growth: 50 and 5% of μmax. The lipid content of the algae was in the range 8.3–29.5% of dry matter and was generally higher in the Prymnesiophyceae than in the Prasinophyceae and the Chlorophyceae. Increasing extent of phosphorus limitation resulted in increased lipid content in the Bacillariophyceae and Prymnesiophyceae and decreased lipid content in the green flagellates N. atomus and Tetraselmis sp. The fatty acid composition of the algae showed taxonomic conformity, especially for the Bacillariophyceae, where the major fatty adds were 14:0, 16:0, 16:1, and 20:5n-3. These fatty acids were dominant also in the Prymnesiophyceae together with 22:6n-3. An exception was I. galbana, in which 18:1 was the major monounsaturated fatty add and 20:5n-3 was absent. The fatty acids of N. atomus and Tetraselmis sp. varied somewhat, but 16:0, 16:1, 18:1, 18:3n-3, and 20:5n-3 were most abundant. Gymnodinum sp. contained mainly 16:0, 18:4n-3, 20: 5n-3, and 22:6n-3. An increased level of nutrient limitation (probably phosphorus) resulted in a higher relative content of 16:0 and 18:1 and a lower relative content of 18:4n-3, 20:5n-3, and 22:6n-3. The nutrient limitation probably reduced the synthesis of n-3 polyunsaturated fatty acids.  相似文献

The effect of sex, source of saturated fat (lard v. palm oil) and glycerol inclusion in the fattening diet on composition and fatty acid positional distribution in the triglyceride molecule was studied in pigs from 78 to 110 kg BW. Average daily gain and carcass characteristics, including ham and loin weight, were not affected by dietary treatment but sex affected backfat depth (P<0.01). A significant interaction between sex and glycerol inclusion was observed; dietary glycerol increased lean content in gilts but not in barrows (P<0.05 for the interaction). Individual and total saturated fatty acid (SFA) concentrations were greater in barrows than in gilts. In contrast, the concentration of total polyunsaturated fatty acids (PUFA) and of C18:2n-6, C18:3n-3, C20:3n-9 and C20:4n-6 in the intramuscular fat (IMF) was higher (P<0.05) in gilts than in barrows. Sex did not affect total monounsaturated fatty acids (MUFA) concentration in the IMF. The proportion of SFA in the subcutaneous fat (SF) was higher in barrows than in gilts (P<0.001). Within the individual SFA, sex affected only the concentrations of C14:0 and C16:0 (P<0.001). Dietary fat did not affect total SFA or PUFA concentrations of the IMF but the subcutaneous total MUFA concentration tended to be higher (P=0.079) in pigs fed lard than in pigs fed palm oil. Dietary glycerol increased total MUFA and C18:1n-9 concentration in the IMF and increased total MUFA and decreased C18:2n-6, C18:3n-3 and total PUFA concentrations in the SF. The data indicate that altering the fatty acid composition of the triglyceride molecule at the 2-position, by dietary intervention during the fattening phase, is very limited.  相似文献

Dietary fatty acid incorporation and changes in various lipid and phospholipid classes in the mussel Mytilus galloprovincialis subjected to three different dietary regimens were analysed and compared. Group A was unfed; group B received a diet consisting of 100% Thalassiosira weissflogii, exhibiting the typical fatty acid composition of diatoms, and group C received a diet consisting of 100% wheat germ conferring a 18:2:n-6 abundance. Biochemical analyses of diets and mussels were carried out at the beginning and at the end of the 30-day experimental period. Starvation and T. weissflogii based diet poorly affected mussel growth and fatty acid composition which remained unchanged. On the contrary, the wheat germ-based diet increased the condition index and deeply affected the fatty acid profile of all lipid and phospholipid classes. The high dietary 18:2n-6 level drastically reduced tissue content of 20:4n-6, 20:5n-3 and 22:6n-3. The biosynthesis of Non Methylene Interrupted (NMI) dienoic fatty acid appeared to be insensitive to the high input of 16:1n-7 and 18:1n-9 respectively from diet B and C, and to the PUFA shortage of diet C. Nevertheless the two NMI trienoic derivatives, 20:3Δ5,11,14 and 22:3Δ7,13 16, were found higher in C with respect to other groups, presumably due to the high 18:2n-6 content of this diet.  相似文献

Four species of red marine algae (Rhodophyceae), five species of brown marine algae (Pheophyceae) and two species of green marine algae (Chlorophyceae) were examined for the fatty acid composition of the three lipid groups separated by silica gel column chromatography (neutral lipids, glycolipids, phospholipids). The four red algae had high contents of 16:0 and C20-polyunsaturated fatty acids (PUFA), 20:5n-3 ranging from 18 to 49% of the total fatty acid content and 20:4n-6 from 1.4 to 22.5%, these fatty acids were evenly distributed in all lipid groups. The five brown algae had high contents of 18:1n-9, 18:2n-6 and 18:3n-3 but low content of 20:5n-3. No precise trend was detected for the distribution of these fatty acids in the three lipid groups. The two green algae had high contents of 16:0, 18:1n-7 and 18:3n-3 and a very low content of PUFA. They contained also large amounts of 16:4n-3 together with 16:2n-6 and 16:3n-3. While 16:2n-6 was mainly found in phospholipids, 16:4n-3 was mainly distributed in neutral lipids and glycolipids.Porphyra umbilicalis represents the richest source of 20:5n-3 whileUndaria pinnatifida can be selected when a balanced mixture of (n-6) and (n-3) PUFA is required.Author for correspondence  相似文献

The fatty acid compositions of neutral lipid, glycolipid and phospholipid fractions from ice algae sampled from the Barents Sea in spring and autumn were examined for seasonal differences. The ice-algal assemblages were dominated by diatoms. In spring, Nitzschia frigida was the most common species whereas resting stages of Thalassiosira bioculata and Actinocyclus cf curvatulus predominated in autumn. With the exception of one spring sample, neutral lipids predominated over glycolipids and phospholipids in all algal samples. The lipid fractions displayed characteristic fatty acid compositions. In the spring samples the major fatty acids of the neutral lipid fraction were 16:0, 16:1(n-7) and 20:5(n-3) whilst the glycolipid fraction was characterised by higher levels of 20:5(n-3) and C16 polyunsaturated fatty acids, particularly 16:4(n-1). Phospholipids contained higher levels of 22:6(n-3) than the other two lipid fractions although 20:5(n-3) was still the major polyunsaturated fatty acid. In the autumn samples, the neutral lipid fraction contained higher proportions of saturated fatty acids and 16:1(n-7) than the two polar lipid fractions and 22:6(n-3) was most abundant in phospholipids. As with the spring samples, 20:5(n-3) was the major polyunsaturated fatty acid in all lipid fractions of the autumn algae. Overall, the fatty acid compositions of the lipid fractions from spring and autumn algal samples were similar and are consistent with diatoms being the predominant group in the ice algae studied. The high level of neutral lipids observed in both spring and autumn samples suggests that the production of neutral lipids is characteristic of ice algae regardless of season. Nevertheless, some species-specific differences in lipid production may exist since the neutral lipid content of autumn samples containing mainly A. curvatulus was substantially higher than those in which T. bioculata predominated. The lipid content and composition of Nereis (Hediste) diversicolor O. F. Müller (Annelida, Polychaeta, Nereidae) a mud-dwelling, intertidal errant polychaete in the Tagus estuary (Portugal), were examined on the monthly basis by lipid extraction, TLC and capillary GC. In this estuary, N. diversicolor is by far the dominant species among polychaeta and the main food item in the natural diet of several flatfishes. The biochemical elucidation of its lipid structure and distribution throughout the year, described in this study, provides information not only about the physiological role of lipids in the animal under consideration but also about dietary fatty acid requirements of some flatfishes in the wild and under laboratory conditions.The total lipid content varied between a maximum of 19.3% lyophilized dry weight in February (4.4% fresh weight) and a minimum of 6.6% in August (1.9% fresh weight). The major lipid classes were triacylglycerol, phospholipid, free sterol, free fatty acid, sterol ester/wax ester and alkyldiacylglycerol.The fatty acid composition was rather unsaturated with a 1:2 mean ratio of n-3: n-6. The major fatty acids were C160:0, C18:1n-9, C18:2n-6, and C20:5n-3; there were smaller amounts of C180:0, C18:1n-11, C18:1n-7, C18:3n-3, C20:1, C20:2n-6, C20:4n-6, C22:2, C22:5n-3, and many other fatty acids were detected at trace levels. The unsaturation ranged from 36.9 mg/g dry weight in summer to 107.4 mg/g in winter. An accumulation of fatty acids from plant origin was evident, in particular linoleic acid (C18:2n-6), which was quantitatively one of the major fatty acids throughout the year.  相似文献

This study examines the composition of lipids, fatty acids, and fatty aldehydes in two marine bryozoan species, Berenicea meandrina and Dendrobeania flustroides, from the Sea of Okhotsk. The share of neutral lipids was up to 57.3% in D. flustroides and 54.9% in B. meandrina; the share of polar lipids was 33.2 and 40.4%, respectively. In all, 57 fatty acids (FA) and 9 aldehydes were identified in total lipids. The main FAs were 16:0, 18:0, 22:6n-3, and 20:5n-3. The content of branched saturated FA in bryozoans was on the average 6.4%. Three isomers of 16:1 (n-9, n-7, and n-5), five isomers of 18:1 (n-13, n-11, n-9, n-7, and n-5), four isomers of 20:1 (n-13, n-11, n-9, and n-7), as well as 22:1n-9 and 22:1n-13 were found; the presence of 7-methyl-6-hexadienoic acid (on the average, 3.0% of total FAs) was demonstrated. Non-methylene-inter-rupted FAs contributed 8.9 and 1.6% of the total FAs in D. flustroides and B. meandrina, respectively, and were identified as 20:2(5,11), 20:2(7,13), 20:3(5,11,14), 22:2(7,13), and 22:2(7,15). In B. meandrina, minor amounts of 24:0, 24:1, 25:0, 26:0, 24:4n-3, 26:3(5,9,19), and 28:3(5,9,19) were found, suggesting sponge biofouling on some bryozoan colonies. Aldehydes (branched saturated and unsaturated C16–19 homologues) did not exceed 10.3 and 1.9% of the total FAs in D. flustroides and B. meandrina, respectively. The presence of the FA markers that are characteristic of microalgae, protozoans, and detritus in bryozoan lipids agrees well with data on polytrophic feeding of these bryozoans.  相似文献

Twelve species of microalgae, isolated from north Australian marine, freshwater and hypersaline environments, were grown under controlled conditions of temperature, pH, photon flux density and salinity, and analysed for ash, total protein, water soluble carbohydrates, chlorophylla, total lipids, total fatty acids and fatty acid composition. Highest levels of the polyunsaturated fatty acid eicosapentaenoic acid [20:5(n-3)] were found in the marine diatoms.Nitzschia (frustulum) andN. closterium (23.1% and 15.2% of total fatty acids, respectively). None of the species studied had levels of docosahexaenoic acid [22:6(n-3)] greater than 1.1 % of total fatty acids. None of the chlorophyte species contained significant levels of either 20:5(n-3) or 22:6(n-3). The highest total fatty acid concentration of all species in the study was found in the freshwater chlorophyte speciesScenedesmus dimorphus (105 mg g–1 dry wt). The hypersaline speciesDunaliella salina had the highest total lipid content (28.1% dry wt), followed byN. closterium, N. (frustulum) andNavicula sp. (24.2–27.8% dry wt).Chlamydomonas sp. had the highest protein content (66.9% dry wt).N. Changes in the fatty acid composition of docosahexaenoic acid (DHA)-producing Schizochytrium limacinum SR21 were investigated. The addition of cyanocobalamin, which is an active component of vitamin B12, decreased the content of odd-chain fatty acids such as pentadecanoic acid (C15:0) and heptadecanoic acid (C17:0). Cyanocobalamin may upregulate the cobalamin-dependent methylmalonyl-CoA mutase, which converts propionic acid to succinic acid, thereby decreasing the content of odd-chain fatty acids. The addition of p-toluic acid resulted in a decrease in docosapentaenoic acid (DPA, 22:5n-6) content and an increase in eicosapentaenoic acid (EPA, 20:5n-3) content in a dose-dependent manner. Two additional peaks of fatty acids, characterized as Δ4,7,10,14-eicosatetraenoic acid (20:4n-7) and Δ4,7,10,14-docosatetraenoic acid (22:4n-9), were detected.  相似文献

Porphyridium cruentum was grown in 10 L batch culture at 18°C, pH 8.0 and 28‰ salinity. The cells were harvested in the stationary phase and the fatty acid composition analysed by GC and tocopherol content by HPLC. A total of 14 fatty acids were identified including saturated fatty acids (13:0, 14:0, 14:0 iso, 15:0, 16:0, 16:0iso) and monounsaturated fatty acids (MUFAs; 16:1(n-7), 18:1(n-7), 18:1(n-9). Polyunsaturated fatty acids (PUFAs) were the predominant fatty acids detected, reaching 43.7% of total fatty acids in the stationary phase of culture. Among the PUFAs, eicosapentaenoic acid (EPA, 20:5(n-3)) was dominant (25.4%), followed by 12.8% arachidonic acid (AA, 20:4(n-6)). α-Tocopherol and γ-tocopherol contents were 55.2 μg g−1 dry weight and 51.3 μg g−1 dry weight respectively.  相似文献

Several commercially available lipases have been evaluated with regard to their substrate specificity in the esterification of fatty acids having specific positions of cis double bonds, e.g. petroselinic acid (n-12 18:1), alpha-linolenic acid (n-3 18:3), gamma-linolenic acid (n-6 18:3), stearidonic acid (n-3 18:4), dihomogamma-linolenic acid (n-6 20:3), eicosapentaenoic acid (n-3 20:5) and docosahexaenoic acid (n-3 22:6), with n-butanol. A common feature of most lipases, e.g. those from Penicillium cyclopium, Candida cylindracea, Mucor miehei, Rhizopus arrhizus and Penicillium sp. is that fatty acids having the first double bond from the carboxyl end as a cis-4 (n-3 22:6), cis-6 (n-12 18:1, n-6 18:3, n-3 18:4) or a cis-8 (n-6 20:3) double bond are strongly discriminated against compared to the other fatty acids, such as myristic acid (14:0), the reference standard, and n-3 18:3. In the case of the lipase from porcine pancreas, however, the discrimination against the above fatty acids is not as strong as with the other lipases. In contrast, the lipase from Chromobacterium viscosum shows a preference for n-12 18:1, n-6 18:3 and n-3 18:4. The observed substrate specificities can be utilized for enrichment of particular fatty acids by lipase-catalysed kinetic resolution from fatty acid mixtures, derived from naturally occurring fats and other lipids.Dedicated to Prof. David A. Larvae of the Chilean–Peruvian scallop Argopecten purpuratus were fed a Dunaliella tertiolecta diet (Dun-diet) supplemented with lipid emulsions, rich in 20:5n-3 (EmEPA), 22:6n-3 (EmDHA) or in saturated fatty acids (EmCOCO). A mixed algal diet of Isochrysis galbana (T-iso) and Chaetoceros neogracile served as a positive control (St-diet). Lipid supplementation to the Dun-diet improved the larval growth and increased the percentage of eyed larvae significantly, compared to the non-supplemented Dun-diet because of the extra energy supplied and not because of its fatty acid composition. No significant differences were observed between supplementation with EmEPA, EmDHA or EmCOCO. A mixture of 20% EmEPA+20% EmDHA (40% EmHUFA) was more efficient in raising the total lipid content in larvae than 40% EmCOCO. Both emulsions increased the triacylglycerol content in larvae compared to the non-supplemented Dun-diet. The best result, however, was obtained with the St-diet, probably because of a more suitable 18:2n-6/18:3n-3 ratio and a higher level of arachidonic acid. A positive relationship between the 18:2n-6/18:3n-3 ratio and the larval performance was found. No significant difference was observed in post-settlement larval size or percentage of metamorphosed larvae between the St-diet and the St-diet supplemented with 20% EmHUFA or 20% EmCOCO. The metamorphosed larvae had a constant DHA/EPA ratio of 3.7, independent from the diet, which suggested a metabolic control of the two fatty acids and a species-dependency of the ratio.  相似文献

Changes in lipid components, particularly glycerophospholipids in the adductor muscle of giant ezo scallop during storage at −20°C, were investigated. During storage, the contents of total lipid (TL) and polar lipid (PL) decreased but that of non-polar lipid (NL) increased. Glycerophosphorylcholine (GPC) and glycerophosphorylethanolamine (GPE) decreased by 50 and 15% of the each initial content, while lyso-GPC and free fatty acids increased. The percentages of polyunsaturated fatty acids such as 20:5n-3 and 22:6n-3 in the TL and PL fractions decreased during storage, but those of the polyunsaturated fatty acids in the NL increased. In the alkenylacyl-GPE and diacyl-GPC, the percentages of relatively longer acids in the sn-1 positions of glycerol moieties decreased at higher rates than did the shorter chains, whereas the results for diacyl-GPE were opposite to those of alkenylacyl-GPE and diacyl-GPC. In the prominent fatty acids in the sn-2 positions of alkenylacyl-GPE and diacyl-GPC, the percentage of 22:6n-3 decreased from compared to the high level of 20:5n-3, while that of diacyl-GPE increased.  相似文献

Total and phospholipid fatty acid composition of fat body and Malpighian tubules from two larval stages, pupae and adults, of Zophobas atratus were analyzed. Saturated and unsaturated C16 and C18 fatty acids were major components and varied by life stage and tissue source. Eicosanoid-precursor fatty acids, including 20:3n-6, 20:4n-6 and 20:5n-3, were present in low quantities and varied by life stage and tissue source. 20:3n-6 was always present in the lowest proportions, indicating that eicosanoids derived from 20:4n-6 and 20:5n-3 (the 2- and 3-series) are likely to be of greater physiological significance in this insect. Fatty acid composition of Z. atratus fat body and Malpighian tubules was independent of diet, suggesting that this insect controls its fatty acid composition to meet the needs of individual tissues and ontogenetic constraints.  相似文献
