Carbohydrates were located on the surface of Phytomonas davidi using ultrastructural cytochemistry, and agglutination induced by lectins which bind to residues of mannose, N-acetylglucosamine, galactose, N-acetylgalactosamine, fucose and sialic acid. The surface charge of the cells was analysed by the binding of cationic particles (colloidal iron and cationized ferritin) to the cell surface and by cell electrophoretic mobility (EPM). Based on observations of binding of cationic particles to the cell surface; a decrease in the binding of these particles to the cell surface; a decrease in the mean EPM of the cells after their incubation in the presence of neuraminidase; and detection of N-acetylneuraminic acid by paper and gas-liquid chromatography, it was concluded that sialic acid residues are exposed on the surface of P. davidi. These residues may be glycolipids or are masked on the cell surface since only after brief trypsinization were the cells agglutinated by the lectin from Limulus polyphemus.  相似文献

The surface anionic groups of untreated or dimethyl sulfoxide (DMSO)-treated Herpetomonas samuelpessoai cells were analyzed by cell electrophoresis, ultrastructural cytochemistry, and identification of sialic acids using thin-layer chromatography. Differentiation of H. samuelpessoai induced by DMSO treatment caused a significant increase in the net negative surface charge. In flagellates exposed to DMSO, more cationized ferritin, colloidal iron hydroxide, and sendai virus particles bound to the cell surface. Treatment of both untreated and DMSO-treated flagellates with neuraminidase decreased markedly the EPM of cells to the cathodic pole. These findings suggest that sialic acid residues are the major anionogenic groups exposed on the surface of H. samuelpessoai. Thin-layer chromatography showed that N-acetyl and N,O-diacylneuraminic acids, in equal proportions, were present in H. samuelpessoai. However, N-acetylneuraminic acid predominates in DMSO-treated cells.  相似文献

 Strong anionic sites, as recognized by deposition of cationic colloidal iron even at pH 1.5, were distributed on the free surfaces of the mesothelia of the mouse pleura, pericardium, and peritoneum. Methylation inhibited colloidal iron staining on the surface, and successive saponification restored it. Digestion with neuraminidase or hydrolysis of sialic acid with H2SO4 erased the colloidal iron staining. Lectin Limax flavus agglutinin (LFA), which is specific for sialic acid, labeled the free surface of the mesothelium. All these findings strongly suggested that the surface substance contained sialic acid. Moreover, prior treatment with LFA inhibited the mesothelial surface stain with colloidal iron. In transmission electron microscopy, the colloidal iron (pH 7.3)-stained substance took the shape of fine strands of 50–300 nm in length. These characteristics of the substance on the mesothelial surface correspond well with biochemical properties of membrane-associated sialomucin, whose strong and abundant negative charges produce repulsive forces between facing serosal surfaces. Zusammenfassung Saure Mucopolysaccharide und sialinsäurehaltige Glycoproteine wurden elektronenmikroskopisch-histoohemisch mit der kolloidalen Eisenreaktion in der Niere dargestellt. Alle Zellmembranen in Rinde und Mark lagern an der Oberfläche positive kolloidale Eisenteilchen an. Die Dicke der Eisenschicht ist bei den Zellen unterschiedlich und variiert auch zwischen luminaler, lateraler und basaler Seite einer Zelle. Bei Tubuli und Capillaren besteht eine Parallele zwischen der Dicke des Außenblattes der Plasmalemm-Unit membrane und der Dicke der kolloidalen Eisenschicht. Auf Grund biochemischer Analysen von Zellmembranen und negativer histochemischer Eisenreaktion nach Sialidasebehandlung ist anzunehmen, daß diese kolloidale Eisenschicht sialinsäurehaltigen Glycoproteinen entspricht. Dieser Glycoproteinbesatz ist ein integrierender Bestandteil der Zellmembran.In mehreren Zellen war das endoplasmatische Retikulum eisenpositiv. Auch hier wurde schon biochemisch Sialinsäure ermittelt.Die positive kolloidale Eisenreaktion im extrazellulären Raum beruht in der Nierenpapille auf dem Vorkommen von sauren Mucopolysacchariden und Sialinsäure und in der Lamina rara interna und externa der glomerulären Basalmembran auf dem Sialinsäuregehalt.
Localization of acid mucopolysaccharides and sialic acid containing glycoproteins in the renal cortex and inner medulla by a combined electron microscopic histochemical method
Summary Acid mucopolysaccharides and sialic acid containing glycoproteins are localized in the kidney by the colloidal iron method. All cell membranes in cortex and medulla are coated by positive colloidal iron particles. This iron-coat variies in thickness at different cells and at the luminal, lateral and basal surface of one cell, too. Tubules and capillaries show a parallelism between the thickness of the outer leaflet of the plasmalemm unit-membrane and the thickness of the colloidal iron coat. Biochemical analyses of cell membranes and the negative result of the colloidal iron reaction after sialidase-treatment allow to conclude, that this colloidal iron coat represents sialic acid containing glycoproteins. This glycoprotein-coat is part of the whole cell membrane.In some cells the endoplasmatic reticulum reacts with colloidal iron. Here, too, sialic acid was found by biochemical analysis.The positive colloidal iron reaction in the extracellular space depends on acid mucopolysaccharides and sialic acid. In the lamina rara interna and externa of the glomerular basement membrane the positive colloidal iron reaction correlates to the sialic acid content.

Differentiation of H. muscarum muscarum induced by propranolol treatment caused a significant increase in the net negative surface charge. Binding of cationized ferritin (CF) and colloidal iron hydroxide particles was observed at the cell surface of both untreated and propranolol-treated cells. In cells incubated in the presence of the drug the CF particles were distributed in all membrane regions. However, there were small areas where the particles were absent. In H. muscarum muscarum exposed to propranolol the density of residues of sialic acid per cell was higher, and the agglutinating activity with Sendai virus was more intense. However, the pattern of sialic acid, characterized by the presence of N-acetylneuraminic acid derivative, was not modified upon cell interaction with the drug. Treatment of both control and propranolol-treated protozoa with neuraminidase significantly reduced the surface charge. These findings suggest that sialic acid residues are the major anionogenic groups exposed on the surface of H. muscarum muscarum .  相似文献

Summary The surface coat of the electrocyte of the main electric organ of Electrophorus electricus was studied using cytochemical methods (periodic acid-silver methenamine, periodic acid-chromic acid-silver methenamine, periodic acid-thiosemicarbazide-silver proteinate, Concanavalin A — horseradish peroxidase, ruthenium red, Alcian-blue lanthanum nitrate, colloidal iron hydroxide and cationized ferritin). The surface of the electrocyte presents perpendicularly oriented tubular invaginations of the cell membrane. The fibrous coat 50–100 nm thick, penetrates into the lumen of the invaginations. It is also observed in the synaptic clefts existent in the posterior face of the electrocyte. The coating of the surface membrane gives a positive reaction with all techniques used. Binding of colloidal iron hydroxide particles was observed only in the outer layer of the coat. With the Alcian-blue lanthanum nitrate technique, microtubules were observed in the cytoplasm of the electrocyte.The results indicate that the surface coat of the electrocyte contains mucopolysaccharides, glycoproteins, acid mucopolysaccharides and anionic sites detected at low (colloidal iron hydroxyde) and neutral (cationized ferritin) pH.This work has been supported by Conselho Nacional de Desenvolvimento Cientifico e Tecnológico (CNPq), Conselho de Ensino e Pesquisa da UFRJ (CEPG) and Banco Nacional de Desenvolvimento Econômico  相似文献

The concanavalin A agglutination patterns, sialyl transferase activity and sialic acid content were studied for cultured lymphoblastoid cell lines possessing either T or B surface markers. Concanavalin A caused marked agglutination of the two B cell lines studied, Raji and SB, while the two T cell lines, HSB-2 and CCRF-CEM, failed to agglutinate with this lectin. The surface sialyl transferase activity of the two B lines was significantly higher than on the two T lines studied. In contrast, the total cellular sialic acid content or the surface sialic acid that was released from the T and B cell lines by neuraminidase was not significantly different. This study indicates that T and B lymphoblastoid cells possess different levels of surface located sialyl transferase activity and display different agglutination patterns with Con A. Perhaps these assays can be of value in differentiating various classes of neoplastic lymphoid cells.  相似文献

The surface charge of Tritrichomonas foetus was evaluated by means of the binding of colloidal iron hydroxide particles at pH 1.8 and cationized ferritin particles at pH 7.2 to the cell surface, as visualized by electron microscopy and by direct measurements of the electrophoretic mobility (EPM), of cells suspended in solutions of different ionic strength and pH. At pH 7.2, T. foetus has a negative surface charge with a mean EPM of ?1.03 μmμs?1μV?1μcm. At lower pH, there is a decrease in the negative surface charge with an isoelectric point at pH 1.2. At higher pH (> 9.0), there is an increase in the surface charge reaching an EPM of ?2.5 μmμs?1μV?1μcm. These results indicate that the surface of T. foetus contains both negatively and positively charged dissociating groups. Binding of colloidal iron hydroxide and cationized ferritin particles throughout the cell surface of the protozoon was observed. Treatment of T. foetus with neuraminidase or trypsin reduced significantly the EPM of the cells. Enzyme-treated cells recovered their normal EPM when incubated for 6 h in fresh culture medium by a process that is inhibited by puromycin.  相似文献

The distribution of the negative charge on the surface of acrosome-reacted and intact spermatozoa of the starfish, Asterina pectinifera , was studied with the charged colloidal iron hydroxide (CIH) labelling technique. All parts of its surface were shown to have an equal negative charge. Influenza virus brought about a type of sperm agglutination in which cells lay parallel to each other. The even distribution of anions on the sperm surface may be due to the existence of sialic acid all over the sperm surface.  相似文献

The surface anionic groups of Entamoeba invadens were analysed by cell electrophoresis, by ultrastructural cytochemistry, and by identification of sialic acids using paper and gas-liquid chromatography. Binding of colloidal iron hydroxide (CIH) and of cationized ferritin (CF) particles at pH 1.8 and 7.2, respectively, was observed on the cell surface. E. invadens has a highly negative surface charge (-0.96 microns s-1 V-1 cm). Treatment of the cells with trypsin and neuraminidase significantly reduced the electrophoretic mobility by 24% and 40%, respectively. Treatment of the amoebae with neuraminidase also markedly decreased the binding of CIH to the cell surface. This finding suggests that sialic acid residues are the major anionogenic groups exposed on the surface of E. invadens. Paper and gas-liquid chromatography showed that N-acetylneuraminic acid was the only derivative characterized in E. invadens.  相似文献

Wild type Crithidia fasciculata and three drug-resistant mutant strains that have shown “flagellar adherence” were studied as to their ability to agglutinate with lectins specific for receptor molecules containing N-acetyl glucosamine, N-acetyl galactosamine, galactose, mannose-like residues, fucose, and sialic acid. Escherichia coli with mannose-sensitive fimbriae was also used as an agglutination probe. The presence of D-GalNAc, D-Gal, and mannose-like residues was detected in the wild strain. Generally, in the mutants, residues of these sugar units were present in greater concentrations when compared to the wild type strain. β-Galactosidase treatment showed that β-D-Galp units are exposed on the cell membrane. All types of cell agglutination including flagellum-flagellum (F-F), flagellum-soma (F-S), and soma-soma (S-S) were observed when lectins were used; however, with E. coli only the F-F type of cell agglutination was observed with the wild type strain and the TFRR1 mutant. All types of agglutination were observed with the other two mutants.  相似文献

The glycopeptides produced by B16 mouse melanoma cells grown in the presence of [3H]glucosamine were isolated and fractionated into two classes (I and II) with cetyl pyridinium chloride. The class I glycopeptides were of higher molecular weight and of higher negative charge (sialic acid content) than those in class II. Class I glycopeptides contained N-acetyl neuraminic acid, galactose and N-acetylgalactosamine and on treatment with alkaline-borohydride were degraded to apparently tri- and tetrasaccharides. The presence of this mucin-type glycoprotein on the cell surface was detected by mild trypsinization of intact cells.  相似文献

Using lectin and colloidal iron (CI) stainings in combination with neuraminidase digestion, glycoconjugates on the surface of erythropoietic cells of the yolk sac and liver in fetal mice were examined. Fetal hepatic macrophages were capable of distinguishing between phagocytozed and non-phagocytozed erythroid elements as described in our previous study. Marked differences between these two elements could be ultrahistochemically detected on their cell surface. The phagocytozed elements, such as nuclei expelled from erythroblasts and degenerating primitive erythroblasts, faintly bound neuraminidase-sensitive CI, and neuraminidase digestion imparted a weak peanut agglutinin (PNA) binding. In contrast, erythroblasts at various maturation stages, erythrocytes and normal primitive erythroblasts heavily bound neuraminidase-sensitive CI, and neuraminidase digestion imparted a moderate PNA binding. No differences in binding of either concanavalin agglutinin,Ricinus communis agglutinin-I or PNA were noted between phagocytozed and non-phagocytozed erythroid elements. Desialylation appears to be one of the most important signs for the recognition mechanism of fetal macrophage phagocytosis. During maturation of hepatic erythroblasts, sialic acid changes its affinity forLimax flavus agglutinin from strong to weak, and soybean agglutinin binding sites disappear at the basophilic erythroblast stage. Glycoconjugates on polychromatophilic erythroblasts acquire similar compositions to those of erythrocytes.  相似文献

ABSTRACT. Drugs that interact with microtubules (colchicine and vinblastine) and microfilaments (cytochalasin B) partially inhibited cell growth and motility of Tritrichomonas foetus. Parasites incubated with these substances became rounded and cell division was blocked. Neither colchicine nor vinblastine disrupted the microtubules that form the peltar-axostylar system. Any one of these drugs interfered with the net negative surface charge of T. foetus as evaluated by determination of the cellular electrophoretic mobility (EPM). The decrease in the EPM of cytochalasin B-treated cells was caused by dimethylsulfoxide, which was used as solvent. Untreated cells as well as cytochalasin B-treated cells showed a uniform distribution of anionic sites on the plasma membrane as seen with cationized ferritin particles. In cells treated with colchicine or vinblastine the anionic sites were distributed in patches. These results are discussed in terms of participation of labile cytoplasmic microtubules and microfilaments in the control of the distribution of anionic sitecontaining macromolecules located on the cell surface of T. foetus.  相似文献

Cell surface changes on the trophectoderm of the mouse blastocyst have been followed in the periimplantation period using electronhistochemical techniques. Examination of the ability of the trophectoderm to bind positively charged colloidal iron particles before and after enzyme treatment has shown that sialic acid-containing glycoproteins make a considerable contribution to the negative charge on the blastocyst surface. At implantation these membrane components are lost or undergo modification independently of direct maternal influence as indicated by a marked decline in colloidal iron binding at this time, both in vivo and in vitro. The findings are discussed in relation to other surface changes on the blastocyst and to the initiation of implantation.  相似文献

Neuraminidases (sialidases) catalyse the removal of terminal sialic acid from glycoconjugates. Bacterial pathogens often utilize neuraminidases to scavenge host sialic acid, which can be utilized either as a nutrient or as a decorating molecule to disguise themselves from host immune attacks. Herein, a putative neuraminidase (TDE0471) was identified in Treponema denticola, an oral spirochaete associated with human periodontitis. TDE0471 is a cell surface‐exposed exo‐neuraminidase that removes sialic acid from human serum proteins; it is required for T. denticola to grow in a medium that mimics gingival crevice fluid, suggesting that the spirochaete may use sialic acid as a nutrient in vivo. TDE0471 protects T. denticola from serum killing by preventing the deposition of membrane attack complexes on the bacterial cell surface. Animal studies revealed that a TDE0471‐deficient mutant is less virulent than its parental wild‐type strain in BALB/C mice. However, it causes a level of tissue damage similar to the wild type in complement‐deficient B6.129S4‐C3tm1Crr/J mice albeit the damage caused by both bacterial strains is more severe in these transgenic mice. Based on these results, we propose that T. denticola has evolved a strategy to scavenge host sialic acid using its neuraminidase, which allows the spirochaete to acquire nutrients and evade complement killing.  相似文献

Surface carbohydrates of drug-resistant and drug-susceptible strains of Trichomonas vaginalis were analysed using lectins. The presence of D-GalNAc, D-Gal and mannose-like residues was detected in T. vaginalis. Marked differences in exposed surface carbohydrates were documented, e.g. wheat germ agglutinin (WGA) selectively agglutinated the drug-susceptible strain whereas drug-resistant parasites reacted preferentially with concanavalin A (Con A). In drug-resistant, but not in drug-susceptible strains, trypsinization induced the appearance of soybean agglutinin. Binding studies using fluorescein-labelled WGA and Con A essentially confirmed the agglutination experiments. Both the intense cell agglutination and the fluorescent WGA-binding displayed by a drug-susceptible strain, were completely nullified by neuraminidase treatment, suggesting the presence of an exposed sialic acid moiety on the T. vaginalis surface.  相似文献

The etiologic agent of Chagas’ disease, Trypanosoma cruzi, is widely distributed in South America, affecting millions of people with thousands of deaths every year. Adherence of the infectious trypomastigote to host cells is mediated by sialic acid. T. cruzi cannot synthesize sialic acids on their own but cleave them from the host cells and link them to glycans on the surface of the parasites using the trans-sialidase, a GPI-anchored enzyme. The infectivity of the protozoan parasites strongly depends on the activity of this enzyme. In this report, we investigated whether the transfer of sialic acids from the host to the parasites can be attenuated using novel sialic acid precursors. The cell line 86-HG-39 was infected with T. cruzi and treated with defined N-acylmannosamine analogues bearing an elongated N-acyl side-chain. By treatment of these cells the number of T.cruzi infected cell was reduced up to 60%. We also showed that the activity of the bacterial sialidase C was reduced with N-glycan substrates with elongated N-acyl side chains of the terminal sialic acids. The affinity of this sialidase decreased with the length of the N-acyl side-chain. The data presented suggest that N-acyl modified sialic acid precursors can change the transfer of sialic acids leading to modification of infection. Since the chemotherapy of this disease is inefficient and afflicted by side effects, the need of effective drugs is lasting. These findings propose a new path to prevent the dissemination of T. cruzi in the human hosts. These compounds or further modified analogues might be a basis for the search of new agents against Chagas’ disease.  相似文献

Summary The surface charge of eosinophils, isolated from the peritoneal exudate of rats by the use of a Metrizamide gradient, was analysed by ultrastructural cytochemistry and cellular electrophoretic mobility. Binding of colloidal iron hydroxide and of cationized ferritin particles at pH 1.8 and 7.2 respectively, was observed on the surface of the eosinophils. An electrophoretic mobility of –1.08 and –1.39 m·s–1·V–1·cm was determined for living and glutaraldehyde-fixed eosinophils, respectively. Treatment of the cells with neuraminidase reduced the electrophoretic mobility to –0.64 m·s–1·V–1·cm (glutaraldehyde-fixed), reduced significantly and abolished completely the binding of both colloidal iron hydroxide and cationized ferritin particles to the surface of the cells. These results indicate that sialic acid exists on the surface of eosinophils, where it accounts for part of the negative surface charge.  相似文献
