The horizontal and vertical distributions of two species of endemic, burrowing mud crabs Helice crassa Dana 1851 (Grapsidae) and Macrophthalmus hirtipes (Jacquinot 1853) (Ocypodidae) are described for the Avon-Heathcote Estuary (43°33′S: 172°44′E), Christchurch, New Zealand. Substratum preference is shown to be the most important factor influencing mud crab distribution, but lack of tolerance to salinities below 4‰ is also a significant factor preventing M. hirtipes from occurring at points close to freshwater input. Both species had similar sediment organic content and particle size requirements. Helice crassa was concentrated in well-drained, compacted sediments above mid-tide level, whilst Macrophthalmus hirtipeswas found in waterlogged areas below mid-tide level. This vertical separation is shown not to be caused by differential desiccation tolerances, but by feeding and burrowing adaptations related to these different substrata.  相似文献

Two intertidal snails, Littorina saxatilis (Olivi, 1972) (upper eulittoral fringe/maritime zone) and Littorina obtusata (Linnaeus, 1758) (lower eulittoral) were collected from a boulder shore on Nobska Point, Cape Cod, Massachusetts, in July and acclimated for 15–20 days at 4 ° or 21 °C. Oxygen consumption rate (Vo2) was determined for 11–15 subsamples of individuals at 4 °, 11 ° and 21 °C with silver/platinum oxygen electrodes. Multiple factor analysis of variance (MFANOVA) of lo10 transformed values of whole animal Vo2 with log10 dry tissue weight (DTW) as a covariant revealed that increased test temperature induced a significant increase in Vo2 in both species (P<0.00001). In contrast, MFANOVA revealed that temperature acclimation did not affect Vo2 in either L. saxatilis (P= 0.35) or L. obtusata (P= 0.095). Thus, neither species displayed a capacity for the typical metabolic temperature compensation marked by an increase in Vo2 at any one test temperature in individuals acclimated to a lower temperature that is characteristic of most ectothermic animals. Lack of capacity for metabolic temperature acclimation has also been reported in other littorinid snail species, and may be characteristic of the group as a whole. Lack of capacity for respiratory temperature acclimation in these two species and other littorinids may reflect the extensive semi-diurnal temperature variation that they are exposed to in their eulittoral and eulittoral fringe/maritime zone habitats. In these habitats, any metabolic benefits derived from longer-term temperature compensation of metabolic rates are negated by extreme daily temperature fluctuations. Instead, littorinid species appear to have evolved mechanisms for immediate metabolic regulation which, in L. saxatilis and L. obtusata and other littorinids, appear to centre on a unique ability for near instantaneous suppression of metabolic rate and entrance into short-term metabolic diapause at temperatures above 20–35 °C, making typical seasonal respiratory compensation mechanisms characteristic of most ectotherms of little adaptive value to littorinid species.  相似文献

Oxygen consumption rates of nauplii of the brine shrimp Artemia franciscana Kellogg 1906 were determined over a range of salinities from 10 to 110 ppm, in temperatures from 0 to 30°C, using a multi-factorial design. The oxygen micro-sensors employed have a fast response time and are capable of accurately measuring oxygen concentrations at temperatures well below 0°C. Oxygen uptake rate ranged from 0.03 to 0.66 μmol O2 mg−1 h−1 and was sensitive to changes in both salinity and temperature. Temperature was the dominant factor affecting oxygen consumption rates, which showed a significant increase with increasing temperature. A slight decrease was measured in oxygen consumption with increasing salinity related to differential solubility of oxygen in waters of different salinities. Thermal sensitivity of oxygen consumption determined from calculations of Q 10, indicated physiological adaptation of Artemia nauplii to the ranges of temperatures tested. The vertical distribution of the tubificid worm Rhyacodrilus hiemalis Ohtaka, the numerically dominant species of oligochaete in the littoral of Lake Biwa, was studied with special reference to seasonal vertical migration in the lake sediment. Monthly collections of lake sediment cores were made using PVC tubes. Core sections of sample sediments ranged from 76 to 117 cm. The vertical distribution of the worms showed no diurnal variation; therefore diel vertical migration was not evident. Seasonal downward migration started in April, and upward migration started in October. From December to March, almost all worms remained in the near-surface sediment layer (surface to 30 cm deep), while from July to September almost all worms remained deeper than 30 cm. However, few individuals migrated deeper than 90 cm. No discontinuous layers were found in grain size composition, water content, loss on ignition, particulate carbon, nitrogen or phosphorus. In deep sediment there was no free oxygen, as evidenced by negative ORP values. For 4 months in summer, R. hiemalis aestivated, probably utilizing anaerobic respiration. It appeared that R. hiemalis moved deeper in the sediment in response to sediment temperature, because sediment temperatures in the deep layers seemed to converge at around 20–21 °C in the summer months. The life history traits of seasonal vertical migration and summer aestivation perhaps arose as an adaptation to the climatic conditions accompanying the geographical origin of R. hiemalis, and they also serve to minimize predation risk during summer when most invertebrate predatory fishes are active.  相似文献

This is the first report describing the purification and enzymatic properties of a native invertase (β-D-fructosidase) in Thermotogales. The invertase of the hydrogen-producing thermophilic bacterium Thermotoga neapolitana DSM 4359 (hereby named Tni) was a monomer of about 47 kDa having an amino acid sequence quite different from other invertases studied up to now. Its properties and substrates specificity let us classify this protein as a solute-binding protein with invertase activity. Tni was specific for the fructose moiety and the enzyme released fructose from sucrose and raffinose and the fructose polymer inulin was hydrolyzed in an endo-type fashion. Tni had an optimum temperature of 85°C at pH 6.0. At temperatures of 80–85°C, the enzyme retained at least 50% of its initial activity during a 6 h preincubation period. Tni had a K m and k cat /K m values (at 85°C and pH 6.0) of about 14 mM and 5.2 × 108 M−1 s−1, respectively. Dedicated to the memory of Prof. R. A. The vertical distribution of meiobenthic copepods was investigated within muddy sediments of a eutrophic lagoon (fish ponds of Arcachon Bay, France). The aim of the study was to determine if in muddy sediments, as previously established in sandy sediments, meiobenthic copepods migrate vertically according to the seasons or diel periods. Two experimental approaches were used, viz: a three-season comparison was made of the diel vertical distribution of the harpacticoid Canuella perplexa T. & A. Scott (1893) and secondly the depth distribution of a meiobenthic copepod assemblage was followed for a 24 h period, in shallow water subtidal locations. The harpacticoid C. perplexa vertically migrated through the top three centimeters of the sediment, showing diel and seasonal variations in depth distribution. The differential vertical distributions shown by the dominant meiobenthic populations suggest that emergence into the water column may mainly concern surface dwelling copepods. The physical and biological factors affecting seasonal and diel changes in the copepod assemblage of the fish ponds are discussed.  相似文献

Sensitivity to cold and freezing differs between populations within two species of live oaks (Quercus section Virentes Nixon) corresponding to the climates from which they originate. Two populations of Quercus virginiana (originating from North Carolina and north central Florida) and two populations of the sister species, Q. oleoides, (originating from Belize and Costa Rica) were grown under controlled climate regimes simulating tropical and temperate conditions. Three experiments were conducted in order to test for differentiation in cold and freezing tolerance between the two species and between the two populations within each species. In the first experiment, divergences in response to cold were tested for by examining photosystem II (PS II) photosynthetic yield (ΔF/F m′) and non-photochemical quenching (NPQ) of plants in both growing conditions after short-term exposure to three temperatures (6, 15 and 30°C) under moderate light (400 μmol m−2 s−1). Without cold acclimation (tropical treatment), the North Carolina population showed the highest photosynthetic yield in response to chilling temperatures (6°C). Both ecotypes of both species showed maximum ΔF/F m′ and minimum NPQ at their daytime growth temperatures (30°C and 15°C for the tropical and temperate treatments, respectively). Under the temperate treatment where plants were allowed to acclimate to cold, the Q. virginiana populations showed greater NPQ under chilling temperatures than Q. oleoides populations, suggesting enhanced mechanisms of photoprotective energy dissipation in the more temperate species. In the second and third experiments, inter- and intra-specific differentiation in response to freezing was tested for by examining dark-adapted F v/F m before and after overnight freezing cycles. Without cold acclimation, the extent of post-freezing declines in F v/F m were dependent on the minimum freezing temperature (0, −2, −5 or −10°C) for both populations in both species. The most marked declines in F v/F m occurred after freezing at −10°C, measured 24 h after freezing. These declines were continuous and irreversible over the time period. The North Carolina population, however, which represents the northern range limit of Q. virginiana, showed significantly less decline in F v/F m than the north central Florida population, which in turn showed a lower decline in Fv/F m than the two Q. oleoides populations from Belize and Costa Rica. In contrast, after exposure to three months of chilling temperatures (temperate treatment), the two Q. virginiana populations showed no decline in F v/F m after freezing at −10°C, while the two Q. oleoides populations showed declines in F v/F m reaching 0.2 and 0.1 for Costa Rica and Belize, respectively. Under warm growth conditions, the two species showed different F 0 dynamics directly after freezing. The two Q. oleoides populations showed an initial rise in F 0 30 min after freezing, followed by a subsequent decrease, while the Q. virginiana populations showed a continuous decrease in F 0 after freezing. The North Carolina population of Q. virginiana showed a tendency toward deciduousness in response to winter temperatures, dropping 58% of its leaves over the three month winter period compared to only 6% in the tropical treatment. In contrast, the Florida population dropped 38% of its leaves during winter. The two populations of the tropical Q. oleoides showed no change in leaf drop during the 3-months winter (10% and 12%) relative to their leaf drop over the same timecourse in the tropical treatment. These results indicate important ecotypic differences in sensitivity to freezing and cold stress between the two populations of Q. virginiana as well as between the two species, corresponding to their climates of origin.  相似文献

Nitrogenous excretion in two snails, Littorina saxatilis (high intertidal) and L. obtusata (low intertidal) was studied in relation to temperature acclimation (at 4° and 21°C), including total N excretion rates, the fraction of urea in N excretion, corresponding O:N ratios and the partitioning of deaminated protein between catabolic and anabolic processes at 4°, 11° and 21°C. Aggregate N excretion rates in both species showed no significant compensatory adjustments following acclimation. Total weight specific N excretion rates at 21°C were higher in standard 3 mg L. saxatilis (739 ng N mg−1 h−1) than standard 5 mg L. obtusata (257 ng N mg−1 h−1) for snails acclimated to 21°C. Comparisons of Q10 values of total weight specific N excretion to Q10 values for weight specific oxygen consumption ({xxV}O2) between 4° to 11 °C and 11° to 21°C indicated that, while total rates of catabolic metabolism ({xxV}O2) and protein deamination in L. obtusata were essentially parallel, the relationship between N excretion and {xxV}O2 in L. saxatilis revealed the partitioning of a larger share of deaminated protein carbon into anabolism at 4° and 21°C than at 11°C. Urea N accounted for a larger share of aggregate N excreted in L. saxatilis than in L. obtusata, but in both species urea N is a greater proportion of total N excreted when acclimated at 4°C (urea N: ammonia N ratio range: 1 to 2.15) than in snails acclimated to 21°C (urea N: ammonia N ratio range: 0.46 to 1.39). Molar O:N ratios indicate that the proportion of metabolism supported by protein catabolism is greater in L. saxatilis (O:N range: 2.5–8.4) than in L. obtusata (O:N range: 7.3–13.0). In both species, regardless of acclimation temperature, the O:N ratios are generally lowest (high protein catabolism) at 4°C and highest at 21°C.  相似文献

A multilayer sediment-water exchange model was used to evaluate the importance of bioturbation in the profundal sediments of L. Esrom. The temporal variation of the vertical distribution of sedimentary phosphorus fractions was modelled with an objective function of 1.50. Deviations between measured and simulated values occurred in the spring, where the measured pool of sedimentary phosphorus sharply declined in the surface sediments. The application of a model for the activity ofChironomus anthracinus based on biomass, oxygen consumption and temperature improved the model in the spring period. The downwards transport of easy-degradable surface sediments reduced the average release of sedimentary phosphorus from 12 mg P · m−2 · day−1 to 11 mg P · M−2 · day−1. The introduction of a similar model for the other important burrowing species in L. Esrom,Potamothrix hammoniensis, lowered the objective function to 1.37 and increased the average release to 12.5 mg P · m−2 · day−1. Summary While on land and recirculating branchial water the Australian semaphore crab Heloecius cordiformis (Decapoda: Ocypodidae), a semi-terrestrial airbreathing mangrove crab, sequentially depresses and elevates its carapace in a regular pump-like manner. The functional role of these carapace movements in aerial oxygen consumption is investigated. Carapace immobilisation (reversible and non-injurious) did not appear to affect branchial water circulation. In dry crabs (branchial water removed) carapace immobilisation had no effect on the rate of oxygen consumption (VO2), heart rate or whole-body lactate (WBL) levels. In wet crabs (with branchial water) carapace immobilisation caused VO2 to drop by 38% from 81 to 46 l O2 · g-1 · h-1, heart rate to decline by 32%, from 2.5 to 1.7 Hz, and WBL levels to increase over 2.5-fold, from 0.27 to 0.67 mg · g-1, after 3 h of carapace immobilisation. The (VO2) of carapace-immobilised crabs with branchial water was similar to lung-occluded crabs with branchial water. Severe hypoxia induced physiological responses similar to those of carapace-immobilised crabs with branchial water. After 3 h of severe hypoxia, heart rate had declined by 80%, from 2.2 to 0.43 Hz, and the incidence of carapace pumping slowed by 85%, from 2.4 to 0.37 cycles · min-1. It is concluded that in the absence of carapace movements branchial water in some way inteferes with lung ventilation. Under normal circumstances water circulation and lung ventilation are mutually exclusive processes (due to their singular dependence on the scaphognathites), yet in Heloecius these processes must be carried out simultaneously. The chlorophyll fluorescence (F) temperature curves in a linear time-temperature heating/cooling regime were used to study heat-induced irreversible F changes in primary green leaves of spring barley (Hordeum vulgare L. cv. Akcent). The leaf segments were heated in a stirred water bath at heating rates of 0.0083, 0.0166, 0.0333, and 0.0500 °C s−1 from room temperature up to maximal temperature T m and then linearly cooled to 35 °C at the same rate. The F intensity was measured by a pulse-modulated technique. The results support the existence of the two critical temperatures of irreversible F changes postulated earlier, at 45–48 and 53–55 °C. The critical temperatures are slightly dependent on the heating rate. Two types of parameters were used to characterize the irreversibility of the F changes: the coefficient of irreversibility μ defined as the ratio of F intensity at 35 °C at the starting/ending parts of the cycle and the slopes of tangents of linear parts of the F temperature curve. The dependence of μ on T m revealed a maximum, which moved from 54 to 61 °C with the increasing heating/cooling rate v from 0.0083 to 0.0500 °C s−1, showing two basic phases of the irreversible changes. The Arrhenius and Eyring approaches were applied to calculate the activation energies of the initial increase in μ. The values varied between 30 and 50 kJ mol−1 and decreased slightly with the increasing heating rate.  相似文献

The aim of this study was to examine the impact of bioturbation by the Manila clam, Ruditapes philippinarum, on sediment stability. A laboratory benthic annular flume system (AFS) was deployed to evaluate the relationship between sediment stability of a subtidal mudflat and density of the infaunal clam under the influence of different current velocities. There was a significant correlation between mean erosion rate and current velocities in all treatments with clams (p < 0.001). There was also a significant correlation between mean erosion rate and R. philippinarum density (p < 0.001), reflecting bioturbation-enhanced sediment erosion. The effects of clam density on sediment erodability were more marked at the lower current velocities. In the control, the critical erosion velocity (Ūcrit) was about 32 cm s−1. With increasing R. philippinarum density, Ūcrit decreased down to the minimum value of about 20 cm s−1 at a density of 206 clams m−2. This study demonstrated that the burrowing activity of R. philippinarum reduces sediment stability, particularly at relatively low current velocities (25 cm s−1) and at densities below those found in the clam cultivation areas within the Sacca di Goro lagoon.  相似文献

The effects of doubled ambient [CO2] and different temperature levels on young Pinus sylvestris growing in phytotron chambers were studied. Five chambers were supplied with ~380 (‘ambient air’) and five with ~700 μmol mol−1 CO2 (‘elevated [CO2]’). Temperature levels in the chambers ranged in increment steps of 2°C from −4°C to +4°C relative to the long-term monthly (day and night) average air temperature levels in Berlin–Dahlem. Substrate was medium fertile; soil moisture and air humidity were kept constant. After three vegetation periods twigs and stems were harvested, weighed, homogenized, and analyzed chemically. There was no significant temperature effect on wood mass accumulation, clearest positive [CO2] effect occurred in the youngest twigs. In total, wood mass increased by 28.5% at doubled ambient [CO2]. N-contents (percentage) decreased at elevated [CO2] in the uppermost stem sections and not in twig wood causing wider C/N ratios in total. In response to elevated temperature, N-contents decreased slightly in twigs (~0.3%). Traces of free glucose, fructose and sucrose, which decreased from the top to the bottom, were found in stem wood, in contrast to traces of starch that increased from the top to the bottom. In response to elevated [CO2] only a little more (0.05%) was accumulated in the top shoot and in tendency; glucose, fructose, and sucrose contents were lower at the bottom of stems as compared to the control. There was no obvious response of these non-structural carbohydrates to elevated temperature except for starch that decreased to half of the content from the lowest to the highest temperature level. Among the hemicellulose compounds, rhamnose and arabinose declined from the top shoot to the bottom of stem, whereas 4-O-methyl-d-glucuronic-acid, mannose, and xylose increased. Contents (percentage) of galactose remained approximately stable along the stem. The clearest positive effect of elevated [CO2] along the whole stem was found for mannose with differences of 0.6–0.3%. In contrast to rhamnose and arabinose that showed a negative response to elevated [CO2], mannose was reduced towards the uppermost stem sections. The 4-O-methyl-d-glucuronic-acid was slightly lowered at the bottom, and galactose and xylose showed no [CO2] response. The only hemicellulose compound which reacted to temperature elevation was galactose. It increased slightly (~0.1% per 1°C). Cellulose and lignin (Klason) behaved oppositely: cellulose increased and lignin decreased from the top to the bottom. These structural components behaved reversely also in response to elevated [CO2]. In stem parts above the bottom section, cellulose content was slightly higher at elevated [CO2], and lignin content was slightly lower at the bottom. Lignin reacted to temperature elevation by a very slight increase on the average (~0.1% per one 1°C). Cellulose, however, decreased by ~0.2% per 1°C temperature elevation. The importance of persistent sinks of carbon in woody plant parts is discussed in respect to the greenhouse effect.  相似文献

The intensity of infection of the mussel Mytilus galloprovincialis Lamarck in the Black Sea by the turbellarian Urastoma cyprinae (Graff), which lives on its gills, was found to be higher in larger hosts, reaching a maximum in mussels of 50–70 mm length. Greater numbers occured in mussels inhabiting a silty bottom than in cultivated mussels suspended above the bottom. Over the period 1982–1987, U. cyprinae was most numerous in winter and especially so in years that were colder. The spionid polychaete Polydora ciliata (Johnston) also infects M. galloprovincialis, burrowing into the shell. Young spionids of up to 1 mm length occured in mussels with a shell length of 35 mm. Numbers of this commensal reached a maximum in mussels of intermediate size.  相似文献

The aim of this study was to develop a methicillin-resistant Staphylococcus aureus (MRSA) detection method based on the melting temperature analysis profiling of S. aureus clinical isolates from three different hospitals in Malaysia. Simplex and duplex real-time PCR assay was used for the simultaneous detection of nuc (species-specific) and mecA (methicillin-resistance) genes in a single SYBR Green I real-time PCR tube assay. Evaluations were based on the melting temperature (T m) analysis of the amplicons using 23 S. aureus clinical isolates including three ATCC S. aureus standard strains. Real-time PCR amplification products with melting peaks at 78.39 ± 0.4°C and 74.41 ± 0.6°C were detected for nuc and mecA genes, respectively. Each real-time PCR assay was completed within two hours. This rapid genotypic method is useful for the detection of resistant determinant (mecA) and identification of S. aureus (nuc) clinical isolates, thus benefiting patient therapy in hospitals.  相似文献

The aim of this study was to examine the variation in body surface temperature of grey seal (Halichoerus grypus) pups throughout lactation in response to different environmental conditions. Radiative surface temperatures (T r, °C) of pups were measured on the Isle of May (56°11′N, 02°33′W), southeast Scotland from 29 October to 25 November 2003. Records were obtained from a total of 60 pups (32 female and 28 male) from three different pupping sites during early and late lactation. Pups were sheltered from high wind speeds but air temperature, humidity and solar radiation at pupping sites were similar to general meteorological conditions. The mean T r of all pups was 15.8°C (range 7.7–29.7°C) at an average air temperature of 10.2°C (range 6.5–13.8°C). There was no difference in the mean T r of pups between early and late lactation. However, the T r varied between different regions of the body with hind flippers on average 2–6°C warmer than all other areas measured. There was no difference in mean T r of male and female pups and pup body mass did not account for the variation in T r during early or late lactation. Throughout the day there was an increase in the T r of pups and this explained 20–28% of the variation in T r depending on stage of lactation. There was no difference in the mean T r of pups between pupping sites or associated with different substrate types. Wind speed and substrate temperature had no effect on the T r of pups. However, solar radiation, air temperature and relative humidity accounted for 48% of the variation in mean T r of pups during early lactation. During late lactation air temperature and solar radiation alone accounted for 43% of the variation in T r. These results indicate that environmental conditions explain only some of the variation in T r of grey seal pups in natural conditions. Differences in T r however indicate that the cost of thermoregulation for pups will vary throughout lactation. Further studies examining intrinsic factors such as blubber thickness and activity levels are necessary before developing reliable biophysical models for grey seals.  相似文献

The sand-bubbler crab Dotilla fenestrata (Hilgendorf, 1869) is the most common and abundant brachyuran in the intertidal area of many East African sandy shores. Monthly sampling surveys were performed from March 2001 to February 2002 at Praia do Triunfo, Southern Mozambique. At each sampling survey, 15 stations (0.25 m2) were sampled to a depth sediment of 30 cm. Crabs were identified, sexed, and measured for carapace width (mm). A total of 2456 crabs were collected of which 1247 were males (mean ± sd: 5.36 ± 0.04 mm), 938 non-ovigerous females (5.03 ± 0.03 mm) and 271 ovigerous females (5.15 ± 0.06 mm). The overall size frequency distribution showed a unimodal pattern suggesting a stable population with continuous recruitment. A clear sexual dimorphism was observed, with males being larger than ovigerous females, which in turn, were larger than non-ovigerous females. The overall sex ratio did not differ from 1:1. The annual reproductive cycle of Dotilla fenestrata was continuous and ovigerous females were recorded throughout the year, with peaks of occurrence during the warmer months (February–August). Despite of this continuous cycle, few juveniles were observed during the study period. These observations and the maximum size attained by adults supports the hypothesis that for this burrowing crab, recruitment may occur in different habitat than that occupied by adults. However, the studied area is favourable for growth, since the analysed population attains sexual maturity in smaller sizes than that from close areas.  相似文献

Carbon isotopic composition of soils subjected to C3–C4 vegetation change can be used to estimate C turnover in bulk soil and in soil organic matter (SOM) pools with fast and intermediate turnover rates. We hypothesized that the biological availability of SOM pools is inversely proportional to their thermal stability, so that thermogravimetry can be used to separate SOM pools with contrasting turnover rates. Soil samples from a field plot cultivated for 10.5 years with the perennial C4 plant Miscanthus×gigantheus were analyzed by thermogravimetry coupled with differential scanning calorimetry (DSC). Three SOM fractions were distinguished according to the differential weight losses and exothermic or endothermic reactions measured by DSC. The δ13C and δ15N values of these three fractions obtained by gradual soil heating were measured by IRMS. The weight losses up to 190 °C mainly reflected water evaporation because no significant C and N losses were detected and δ13C and δ15N values of the residual SOM remained unchanged. The δ13C values (−16.4‰) of SOM fraction decomposed between 190 and 390 °C (containing 79% of total soil C) were slightly closer to that of the Miscanthus plant tissues (δ13C = −11.8‰) compared to the δ13C values (−16.8‰) of SOM fraction decomposed above 390 °C containing the residual 21% of SOM. Thus, the C turnover in the thermally labile fraction was faster than that in thermally stable fractions, but the differences were not very strong. Therefore, in this first study combining TG-DSC with isotopic analysis, we conclude that the thermal stability of SOM was not very strongly related to biological availability of SOM fractions. In contrast to δ13C, the δ15N values strongly differed between SOM fractions, suggesting that N turnover in the soil was different from C turnover. More detailed fractionation of SOM by thermal analysis with subsequent isotopic analysis may improve the resolution for δ13C.  相似文献

Seasonal changes in abundance and distribution pattern of soil micro-arthropods were studied in connection with a few environmental factors in a Japanese cedar (Cryptomeria japonica D. Don) plantation. The soil arthropods were sampled from three different depths at intervals of two months for two years. Of the collected animals (total 51000–155000 m−2), Collembola (20000–76000 m−2), oribatid mites (19000–55000 m−2) and carnivorous mites (6200–21000 m−2) were the numerically dominant animal groups. Low seasonal variations in abundance indicated their seasonal stability in population levels. The trends in seasonal fluctuation were similar among these groups and between the two years, showing bimodal pattern with little peaks in early summer and winter. The pattern of seasonal fluctuation in abundance of carnivorous mites (P d) was significantly synchronized with that in the total abundance of Collembola and oribatid mites (P τ). Thus, the number-ratios (P d/P τ) were fairly constant, ranging from 0.10 to 0.25. Seasonal changes in vertical distribution of the three animal groups showed a similar pattern for both years. The downward migrations were shown to be more affected by low temperatures in winter accompanied by snow coverage rather than by the desiccation of the surface soil in summer. All the three groups demonstrated as a whole slightly aggregated patterns of horizontal distribution throughout the two years. Temporal increases in the patchiness indices were observed from summer to autumn when the moisture content of the surface soil was low.  相似文献

Aspergillus versicolor grown on xylan or xylose produces two β-xylosidases with differences in biochemical properties and degree of glycosylation. We investigated the alterations in the biochemical properties of these β-xylosidases after deglycosylation with Endo-H or PNGase F. After deglycosylation, both enzymes migrated faster in PAGE or SDS-PAGE exhibiting the same Rf. Temperature optimum of xylan-induced and xylose-induced β-xylosidases was 45°C and 40°C, respectively, and 35°C after deglycosylation. The xylan-induced enzyme was more active at acidic pH. After deglycosylation, both enzymes had the same pH optimum of 6.0. Thermal resistance at 55°C showed half-life of 15 min and 9 min for xylose- and xylan-induced enzymes, respectively. After deglycosylation, both enzymes exhibited half-lives of 7.5 min. Native enzymes exhibited different responses to ions, while deglycosylated enzymes exhibited identical responses. Limited proteolysis yielded similar polypeptide profiles for the deglycosylated enzymes, suggesting a common polypeptide core with differential glycosylation apparently responsible for their biochemical and biophysical differences.  相似文献
