We have found an alfalfa cDNA clone that encodes an acidic leghemoglobin. To date, 14 alfalfa leghemoglobin clones have been identified. Five different leghemoglobin components have been biochemically defined on the basis of their pI. A higher-resolution comparison, provided by sequence data analysis, identifies six leghemoglobin classes. All 14 leghemoglobins are assigned to the six classes, which can be distributed among the five leghemoglobin components. The newly identified leghemoglobin is the only member of a sixth class of leghemoglobins, and it also is the only member of one of the acidic leghemoglobin components IV or V.  相似文献

The major barrier for xenotransplantation in humans is the presence of (1–3) Galactosyl epitopes (Gal) in xenogeneic tissue and the vast quantities of natural antibodies (Ab) produced by humans against this epitope. The binding of anti-Gal Ab to cells expressing Gal triggers a complement-mediated hyperacute rejection of target cells. The hyperacute rejection of whole cancer cells, modified to express Gal epitopes, could be exploited as a new cancer vaccine to treat human cancers. We tested this hypothesis in Galactosyltransferase knockout (GT KO) mice which, like humans, do not express Gal on their cell surfaces and can produce anti-Gal Ab. Forty-five percent of mice with preexisting anti-Gal Ab rejected Gal positive melanoma cells (B16Gal). These mice remained tumor-free for more than 90 days. The majority of control mice injected with B16Null, Gal negative cells succumbed to melanoma. The rejection of B16Gal induced strong long-lasting antitumor immunity against B16Null measured by the expansion of cytotoxic T lymphocytes. In addition, mice rejecting B16Gal were protected against melanoma since they survived a second rechallenge with B16Null. Protected mice developed antitumor immunity in the absence of autoimmune depigmentation (vitiligo). These results show that rejection of Gal positive melanoma cells can efficiently boost the immune response to other tumor associated antigens present in Gal negative melanoma cells. This study supports the concept of a novel anticancer vaccine to treat human malignancies.  相似文献

The absorption maximum of the far-red absorbing form of phytochrome in the difference spectrum for phototransformation (Pfr max) was investigated in vivo and in in vitro pellets from dark grown Hordeum vulgare L. primary leaves. Exposure of pellets in Honda medium from tissue pre-irradiated with red light to far red light gave a Pfr max of 734 nm, a slightly longer wavelength than was seen in vivo (730 nm). After incubation as the red absorbing form of phytochrome (Pr) for 2 h at 0° C irradiation with red light showed that Pfr max had shifted to shorter wavelength (716 nm) in Honda medium. Further incubation as Pfr for 2 h at 0° C and irradiation with far red light showed that Pfr max had shifted to longer wavelength (726 nm). Similar shifts were also seen in other media, although the peak positions were different. Phytochrome remained pelletable throughout these experiments and Pfr max is compared to that of soluble phytochrome in similar media. Hypothyroidism was induced in a group of male Fischer 344 rats by administration of 0.05% propylthiouracil (PTU) in the drinking water for 12 weeks. Control rats were not treated. Plasma levels of thyroid hormones indicated that PTU treatment had produced severe thyroid hormone deficiency. In PTU-treated rats compared to control rats, levels of total T3 and total T4 were reduced 54.5% and 53.7%; while levels of free T3 and free T4 were reduced 87.1% and 96.5%. Functional hypothyroidism was demonstrated by: (i) a 49.1% decrease in hepatic plasma membrane 1-adrenergic receptor binding, and (ii) a 11.2- fold increase in hepatic -glutamyltranspeptidase activity; relative to the expression of these parameters in control rats. Membranes were isolated from hippocampi of control, PTUinduced hypothyroid and thyroxine-replaced rats and specific adrenergic receptor binding determined by radioligand binding techniques. Hypothyrodism resulted in a shift in the balance of 1 and 2 adrenergic receptor binding by evoking: an increase in 1- adrenergic receptor binding to 1.57-fold of control levels; and, a decrease in 2-adrenergic receptor binding to 64% of control levels. Thyroid hormone replacement carried out in PTU-treated hypothyroid rats at 30 g/kg s.c. per day for the last 3 days of the 12 week PTU-treatment protocol, which reversed physical and functional hypothyroidism, reversed the observed changes in hippocampal adrenergic receptor binding, indicating them to be thyroid hormone, and not PTU, -dependent. This receptor shift evoked by hypothyroidism may, in part, explain the protective effect of hypothyroidism on ischemia-induced hippocampal damage by favoring inhibitory input and limiting excitotoxic input by catecholamines.  相似文献

Hybridization between taxonomically similar species is an often-overlooked mechanism limiting the recovery of threatened and endangered species. We present molecular genetic data for the first time demonstrating that Canada lynx and bobcats hybridize in the wild. We verify that two microsatellite loci Lc106 and Lc110 have non-overlapping allele ranges between Canada lynx and bobcats, and that three putative lynx from Minnesota contain DNA from both bobcats and lynx. Additionally, we use a published test for the 16S rRNA region of mitochondrial DNA (mtDNA) to determine the maternal species; all hybrids had lynx mothers. Fifteen per cent (3/20) of our putative lynx samples were hybrids, although these data are not from a representative sampling effort. Hybridization may be an under-appreciated factor limiting the distribution and recovery of lynx. The presence of hybrids is thus a new factor in the population management of both species with potential implications for hunting and trapping of bobcats.  相似文献

Summary ZO-1, a cytoplasmic plaque protein of tight junctions, exists in vivo as two major isoforms which are defined by the presence or absence of an 80 amino acid domain termed . The ZO-1 (+) isoform is expressed in most epithelial cells while ZO-1(–) isoform expression is restricted to endothelial cells and some highly specialized epithelial cells, suggesting that the isoforms serve different functions. We had previously demonstrated that both ZO-1 isoforms are expressed in A 431 cells and are tyrosine phosphorylated in response to epidermal-growth-factor treatment. In the present study, we found that the (-) isoform of ZO-1 was more tightly associated with the cytoskeleton than was the (+) isoform, based on extraction in nonionic detergent. In addition, the ZO-1(-) was preferentially tyrosine phosphorylated in response to epidermal-growth-factor treatment. However, both isoforms became more tightly associated with the cytoskeleton after A 431 cells were exposed to epidermal growth factor. Immunofluorescence analysis of A 431 cells with isoform-specific antibodies demonstrated that functional differences in ZO-1 isoform behavior were not due to differences in their subcellular locations. The coincident localization of these isoforms does not rule out different affinities for interacting proteins related to the presence or absence of the domain, and it is these interactions that are likely to explain functional differences between the isoforms.  相似文献

Summary The cytosolic chaperonin containing t-complex peptide-1 (CCT) is involved in the correct folding of newly synthetized actin and tubulin molecules. To get insight into potential additional functions of plant CCT, the localization of the subunit CCT was followed throughout cell cycle, cell elongation, and cell differentiation in the tobacco cell culture VBI-O with relation to the microtubular cytoskeleton by double-immunofluorescence and confocal microscopy. The CCT subunit was found to colocalize with sites of microtubule nucleation such as nuclear envelope and preprophase band. In addition, CCT was associated with tubulin in sites of elevated wall synthesis such as phragmoplast or along secondary-wall thickenings. CCT and its substrate tubulin were found to be soluble during periods of cytoskeletal dynamics, whereas sedimentable, vesicle-bound forms of CCT and tubulin prevailed during cell differentiation. The sedimentability of CCT was increased by calcium, whereas it was detached from microsomes by ATP. CCT can bind to both polymerized microtubules and tubulin dimers. Water is one of the key limiting factors for the survival and growth of plant species in arid and semi-arid steppe regions. Different plant functional groups (PFGs) based on life-forms differ in their strategies to cope with limited water availability. The foliar carbon isotope discrimination () value provides an integrated measurement of internal plant physiological and external environmental properties affecting photosynthetic gas exchange over the time interval when the carbon was fixed. In this study, we surveyed the composition and values of various life-forms (shrubs, sub-shrubs, perennial grasses, perennial forbs and annuals) in eight different plant communities along a soil moisture gradient in the Xilin River Basin, Inner Mongolia, China. Our results showed that: (1) life-forms occurred variously in eight steppe communities with different soil moisture status; (2) in wetter habitats, forbs were more abundant and accounted for the majority of aboveground biomass, whereas grasses became more important in drier habitats. Shrubs and sub-shrubs increased with decreasing soil water availability and their relative biomass rapidly increased in degraded steppe and sand dune communities. (3) The numerical order of the mean values of life-forms is as follows: perennial grasses (15.86) < shrubs (16.10) < perennial forbs (16.45)=annuals (16.41) < sub-shrubs (17.55), reflecting their differences in water use efficiencies. The significant differences in the values among these life-forms suggested that life-form-based PFGs not only represent a morphological classification of these plants, but could also represent a functional group integrating different physiological processes such as water use strategies, which may partially explain the differences in PFG composition and competitive ability of co-existing species along environmental gradients in the Xilin River Basin.  相似文献

The meaning of niche and competitive ability have long been surrounded by controversy. The reason for this stems from the obscure relationship that exists between these terms. This extends from the views of Darwin through Eltonian tradition to current views in which the meaning of competitive ability is implicitly infused into the paradigm of niche. Distinct operational definitions for niche and competitive ability are therefore established with special reference to plants. It is proposed that potential niche refer explicitly to a theoretical hyperspace of places where a species would leave descendents if all biotic interactions were precluded, and that competitive ability refer to the relative capacity to leave descendents in a particular place in the face of restrictions imposed by competitive interaction. This leads to a qualitative comprehensive theory for coexistence which may be extended to any type of biotic interaction. Niche and competitive ability are both determined by the biological attributes of a species and may be independently adjusted in a population by natural selection in contexts of competition. Species coexistence in nature may therefore be a consequence of alternative evolutionary mechanisms which may operate to various degrees in concert: (1) natural selection leading to niche differentiation; (2) an ongoing process of reciprocal selection (coevolution) which maintains an approximate balance in relative competitive abilities for contested resources.  相似文献

As part of an integrated forest vegetation and soil survey, tree species composition (DBH 10 cm) was recorded in 95 plots of 0.1 ha, distributed over the principal physiographic units in the middle Caquetá area, Colombian Amazonia. A total of 1077 tree species was found, classified into 271 genera and 60 families. Leguminosae and Sapotaceae show high familial importance values in all physiographic units. Lauraceae, Chrysobalanaceae, Moraceae, and Lecythidaceae are more important on well drained (flood plain or upland) soils, while Palmae, Guttiferae, Bombacaceae, and Apocynaceae are more important in swamps and on podzolised (white sand'rs) soils. Plots on well drained soils show a lower degree of dominance than plots in swamps or on podzolised (white sand) soils. The composition of the most dominant species in the plots changes continuously. Most species (59%) are only recorded in one plot. Individual plot pairs generally show a low overlap of about 2–5 tree species, resulting in Jaccard coefficients below 20%.complementary to a previous forest classification based on TWINSPAN analyses, detrended and canonical correspondence analyses were carried out, using CANOCO 3.1. Despite of a low amount of tree species variance explained (only 6.2% by the first two canonical axes), meaningful patterns of tree species composition were recognised. These are most strongly related to drainage, flooding, humus forms, and soil nutrient status. Forest types are well separated in the CCA ordination diagram. The most frequently found tree species are listed according to their preference with respect to drainage, flooding, and soil nutrient status.Tree species composition in the well drained upland forests was analysed separately. In view of the model explaining high NW Amazonian tree species diversity on the basis of dense community packing and high beta diversity along soil gradients, the canonical analysis here focused on the effect of soils. By means of partial canonical ordination it was found that patterns of tree species composition depended significantly on soil properties, even though the edaphic component explains only a small fraction of the tree species variance. The results show that the well drained uplands of the middle Caquetá area are covered by a complex of two intergrading tree species assemblages. The first assemblage (community of Goupia glabra-Clathrotropis macrocarpa) is associated to some-what less poor, clayey soils developed in Andean origin deposits or Tertiary sediments from the Pebas formation. The second assemblage (community of Swartzia schomburgkii-Clathrotropis macrocarpa) shows affinities to very poor, loamy soils developed in parent materials derived from the Guiana shield. This simple dichotomous pattern of geology, soils, and forest types is incompatible with concepts of high soil heterogeneity and associated beta diversity controlling tree species diversity in well drained uplands of NW Amazonia. The gradient length of tree species in the detrended correspondence analysis was low (3.7 SD), also suggesting a low beta diversity.  相似文献

Summary A new method for measuring structural change in sets of species which have been subjected to natural or experimental perturbation is developed and is shown to be superior to static diversity and evenness measures for this purpose. Three parameters, H, J, and X;} are shown to provide necessary and sufficient information on the severity of a perturbation as well as the uniformity of its effect on all species in the set. When positive and negative changes in species abundance are considered separately, the method is sensitive to compensatory changes which are not detected by static measures.The parameters are then calculated for some data sets on polluted and unpolluted fish communities in second and third order streams from the Clemons Fork watershed in eastern Kentucky. Results indicate that H, the diversity of change over two sampling seasons, is high for perturbed and unperturbed systems, but J the eveness of change is lower for the communities which were polluted in the second sampling season. Severe pollution results in the suppression of most major fish species, whereas more moderate pollution results in a large number of compensatory changes. The biological basis for such an outcome is discussed, and the notion of these three parameters as the vital signs of a healthy ecosystem is presented.  相似文献

Summary Two specific -N-acetylglucosaminyltransferases involved in the branching and elongation of mucin oligosaccharide chains, namely, a 1,6 N-acetylglucosaminylsaminyltransferase that transfers N-acetylglucosamine from UDP-N-acetylglucosamine to Gal3GalNAc-Mucin to yield Gal3(GlcNAc6)GalNAc-Mucin and a 3-N-acetylglucosaminyl transferase that transfers N-acetylglucosamine from UDP-N-acetylglucosamine to Gal3(GlcNAC6)GalNAc-mucin to yield GlcNAc3Gal3 (GlcNAc6)GalNAc-Mucin were purified from the microsomal fraction of swine trachea epithelium. The 1,6-N-acetylglucosaminyltransferase was purified about 21,800-fold by procedures which included affinity chromatography on DEAE columns containing bound asialo Cowper's gland mucin glycoprotein with Gal1,3GalNAc side chains. The apparent molecular weight estimated by gel filtration was found to be about 60 Kd. The purified enzyme showed a high specificity for Gal1,3GalNAc chains and the most active substrates were mucin glycoproteins containing these chains. The apparent Km of the 6-glucosaminyltrans-ferase for Cowper's gland mucin glycoprotein containing Gal1,3GalNAc chains was 0.53 µM; for UDP-N-acetylglucosamine, 12 µM; and for Gal 1,3GalNAc NO2ø, 4 mM. The activity of the 6-glucosaminyltransferase was dependent on the extent of glycosylation of the Gal3GalNAc chains in Cowper's gland mucin glycoprotein.The best substrate for the partially purified 3-Glucosaminyltransferase was Cowper's gland mucin glycoprotein containing Gal1,3(GlcNAc6)GalNAc side chains. This enzyme showed little or no activity with intact sialylated Cowper's gland mucin glycoprotein or derivatives of this glycoprotein containing GalNAc or Gal1,3GalNAc side chains.The radioactive oligosaccharides formed by these enzymes in large scale reaction mixtures were released from the mucin glycoproteins by treatment with alkaline borohydride, isolated by gel filtration on Bio-Gel P-6 and characterized by methylation analysis and sequential digestion with exoglycosidases. The oligosaccharide products formed by the 6- and 3-glucosaminyltransferases were shown to be Gal3(GlcNAC6) GalNAc and GlcNAc3 Gal3(GlcNAC6)GalNAc respectively.Taken collectively, these results demonstrate that swine trachea epithelium contains two specific N-acetylglucosaminyltransferases which catalyze the initial branching and elongation reactions involved in the synthesis of O-linked oligosaccharide chains in respiratory mucin glycoproteins. The first enzyme a 6-glucosaminyltransferase converts Gal3GalNAc chains in mucin glycoproteins to Gal3(GlcNAc6)GalNAc chains. This product is the substrate for a second 3-glucosaminyltransferase which converts the Gal3(GlcNAc6)GalNAc chains to GlcNAc3Gal(GlcNAc6)GalNAc chains in the glycoprotein. For parasites with a clearly defined life-cycle we give threshold quantities that determine the stability of the parasite-free steady state for autonomous and periodic deterministic systems formulated in terms of mean parasite burdens. We discuss the biological interpretations of the quantities, how to deal with heterogeneity in both parasite and host populations, how to incorporate the effects of periodic discontinuities, and the relation of the threshold quantities to the basic reproduction ratio R 0. Examples from the literature are given. The analysis of the periodic case extends easily to micro-parasitic systems.  相似文献

The seeds of 19 sunflower species were compared on the basis of their protein contents and the relative proportions of their protein fractions. The globulin content varied from 50% to about 70% and the albumin content from 18% to 35% according to the species. The level of intermediateMr polypeptides showed a great variability (9.6 to 24.3%). Comparative studies onMr polymorphism were carried out by means of sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) of non reduced and/or reduced samples using both mono- and bidimensional procedures. Polypeptide constituents of helianthinin were compared including both number and molecular size (cultivatedH. annuus was used as a standard). Studies focused on differences observed between the major two (Mr 38 000), (Mr 32 000) and (Mr 25 500), (21 000) polypeptides families constituting the main A, B, and C subunits. and polypeptides analyses permit to discriminate easilyH. petiolaris from the other species. Charge polymorphism was studied using isoelectric focusing (IEF) and IEF-PAGE in mono and bidimensional procedures in the presence or absence of 2-mercaptoethanol (2-ME). Only a specific 4 polypeptide enables an easy discrimination betweenH. petiolaris and all the other species. Detailed nomenclature of the , and , polypeptides constituting the different helianthinin globulin subunits is given via the results of pI andMr analyses. Monodimensional IEF patterns of the more basic albumins (pI > 8.0) appear to provide a more valuable approach to identifying specific protein markers.  相似文献

A recently described protocol for thein vitro production of ascomata was employed to determine the sexual incompatibility systems of five species ofCapronia. The formation of mature ascomata in isolates derived from single ascospores demonstrated thatC. epimyces, C. mansonii, andC. munkii n. sp. are homothallic. In contrast, fertile ascomata were observed only in mass-ascospore isolates and pairwise crosses between specific single-ascospore isolates inC. dactylotricha n. sp. andC. moravica. TheExophiala anamorphs ofC. dactylotricha andC. munkii are described and aPhialophora-like synanamorph is reported for the former species. Germinating ascospores ofC. munkii formed conidiogenous cells directly, while the ascospores of the remaining species germinated to produce germ tubes and hyphae. The application of the terms microcyclic conidiation to secondary conidium production and sclerotial body and stroma to the multicellular structures produced by species ofCapronia andExophiala are discussed.  相似文献

Ectomycorrhizal (ECM) fungi have previously been divided into two groups (early-stage and latestage) based on their position in ectomycorrhizal succession. This classification has a number of limitations, particularly in the lack of precision in the terminology adopted and its general applicability. Many species cannot be easily assigned to either stage, and the behaviour of some species is modified by environmental conditions. In addition, a number of different mechanisms of ECM succession have been proposed, including the influence of organic matter and root exudates. In this paper, it is suggested that the phenomenon of ECM succession can be explained simply by the relative ability of different fungi to colonize and spread from different sources of inoculum. Epidemiological characteristics, such as the ability to produce mycelial strands or to spread on root systems by secondary infection, may therefore provide a more appropriate basis for a functional classification of ECM fungi. The functionally important characteristics of ECM fungi are discussed.  相似文献

The objective of this study was to determine how adjustment in stomatal conductance (g s) and turgor loss point (tlp) between riparian (wet) and neighboring slope (dry) populations of Acer grandidentum Nutt. was associated with the susceptibility of root versus stem xylem to embolism. Over two summers of study (1993–1994), the slope site had substantially lower xylem pressures (px) and g s than the riparian site, particularly during the drought year of 1994. The tlp was also lower at the slope (-2.9±0.1 MPa; all errors 95% confidence limits) than at riparian sites (-1.9±0.2 MPa); but it did not drop in response to the 1994 drought. Stem xylem did not differ in vulnerability to embolism between sites. Although slope-site stems lost a greater percentage of hydraulic conductance to embolism than riparian stems during the 1994 drought (46±11% versus 27±3%), they still maintained a safety margin of at least 1.7 MPa between midday px and the critical pressure triggering catastrophic xylem embolism (pxCT). Root xylem was more susceptible to embolism than stem xylem, and there were significant differences between sites: riparian roots were completely cavitated at -1.75 MPa, compared with -2.75 MPa for slope roots. Vulnerability to embolism was related to pore sizes in intervessel pit membranes and bore no simple relationship to vessel diameter. Safety margins from pxCT averaged less than 0.6 MPa in roots at both the riparian and slope sites. Minimal safety margins at the slope site during the drought of 1994 may have led to the almost complete closure of stomata (g s=9±2 versus 79±15 mmol m-2 s-1 at riparian site) and made any further osmotic adjustment of tlp non-adaptive. Embolism in roots was at least partially reversed after fall rains. Although catastrophic embolism in roots may limit the minimum for gas exchange, partial (and reversible) root embolism may be adaptive in limiting water use as soil water is exhausted.  相似文献

The visual pigments and photoreceptor types in the retinas of three species of Pacific salmon (coho, chum, and chinook) were examined using microspectrophotometry and histological sections for light microscopy. All three species had four cone visual pigments with maximum absorbance in the UV (max: 357–382 nm), blue (max: 431–446 nm), green (max: 490–553 nm) and red (max: 548–607 nm) parts of the spectrum, and a rod visual pigment with max: 504–531 nm. The youngest fish (yolk-sac alevins) did not have blue visual pigment, but only UV pigment in the single cones. Older juveniles (smolts) had predominantly single cones with blue visual pigment. Coho and chinook smolts (>1 year old) switched from a vitamin A1- to a vitamin A2-dominated retina during the spring, while the retina of chum smolts and that of the younger alevin-to-parr coho did not. Adult spawners caught during the Fall had vitamin A2-dominated retinas. The central retina of all species had three types of double cones (large, medium and small). The small double cones were situated toward the ventral retina and had lower red visual pigment max than that of medium and large double cones, which were found more dorsally. The inheritance of host plant resistance and its effect on the relative infection efficiency for leaf blast was studied in the crosses IR36/CO39 (partially resistant × highly susceptible) and IR36/IR64 (both partially resistant). On the natural scale, gene action appeared multiplicative. After log transformation, additive effects described most of the genetic variation in the cross IR36/CO39, while additive and dominance effects were about equal in magnitude in the cross IR36/IR64. Dominance was towards increased resistance. No transgressive segregation occurred in the cross IR36/CO39. The number of genes that reduce lesion number was estimated to be zero in CO39 and five or more in IR36. The cross IR36/IR64 showed transgressive segregation in both directions, and IR36 and IR64 each contain at least one gene that is not present in the other cultivar. The heritabilities (narrow sense) in the F2 were low (range 0.06–0.16), while narrow sense heritabilities based on F3 lines were much higher (range 0.41–0.68). Lesion numbers in F3 lines were reasonably correlated with those in F5 progenies derived from the same F2 plant (r was±0.6 in both crosses). Partial resistance can be effectively improved by selecting the most resistant plants from the most resistant F3 lines.  相似文献

Summary The ratio of deuterium to hydrogen (expressed as D) in hydrogen released as water during the combustion of dried plant material was examined. The D value (metabolic hydrogen) determined on plant materials grown under controlled conditions is correlated with pathways of photosynthetic carbon metabolism. C3 plants show mean D values of-132 for shoots and -117 for roots; C4 plants show mean D values of -91 for shoots and-77 for roots and CAM plants a D value of-75 for roots and shoots. The difference between the D value of shoot material from C3 and C4 plants was confirmed in species growing under a range of glasshouse conditions. This difference in D value between C3 and C4 species does not appear to be due to differences in the D value (tissue water) in the plants as a result of physical fractionation of hydrogen isotopes during transpiration. In C3 and C4 plants the hydrogen isotope discrimination is in the same direction as the carbon isotope discrimination and factors contributing to the difference in D values are discussed. In CAM plants grown in the laboratory or collected from the field D values range from-75 to +50 and are correlated with 13C values. When deprived of water, the D value (metabolic hydrogen) in both soluble and insoluble material in leaves of Kalanchoe daigremontiana Hamet et Perr., becomes less negative. These changes may reflect the deuterium enrichment of tissue water during transpiration, or in field conditions, may reflect the different D value of available water in areas of increasing aridity. Whatever the origin of the variable D value in CAM plants, this parameter may be a useful index of the water relations of these plants under natural conditions.  相似文献
