In frog melanotrope cells, gamma-aminobutyric acid (GABA) induces a biphasic effect, i.e. a transient stimulation followed by a more sustained inhibition of alpha-MSH release, and both phases of the GABA effect are mediated by GABAA receptors. We have previously shown that the stimulatory phase evoked by GABAA receptor agonists can be accounted for by calcium entry. In the present study, we have investigated the involvement of the chloride flux on GABA-induced [Ca2+]c increase and alpha-MSH release. We show that GABA evokes a concentration-dependent [Ca2+]c rise through specific activation of the GABAA receptor. The GABA-induced [Ca2+]c increase results from opening of voltage-activated L- and N-type calcium channels, and sodium channels. Variations of the extracellular Cl- concentration revealed that GABA-induced [Ca2+]c rise and alpha-MSH release both depend on the Cl- flux direction and driving force. These observations suggest for the first time that GABA-gated Cl- efflux provokes an increase in [Ca2+]c increase that is responsible for hormone secretion.  相似文献

The effects of four tremorgenic and one nontremorgenic mycotoxins were studied on gamma-aminobutyric acid (GABAA) receptor binding and function in rat brain and on binding of a voltage-operated Cl- channel in Torpedo electric organ. None of the mycotoxins had significant effect on [3H]muscimol or [3H]flunitrazepam binding to the GABAA receptor. However, only the four tremorgenic mycotoxins inhibited GABA-induced 36Cl- influx and [35S] t-butylbicyclophosphorothionate [( 35S]TBPS) binding in rat brain membranes, while the nontremorgenic verruculotoxin had no effect. Inhibition of [35S]TBPS binding by paspalinine was non-competitive. This suggests that tremorgenic mycotoxins inhibit GABAA receptor function by binding close to the receptor's Cl- channel. On the voltage-operated Cl- channel, only high concentrations of verruculogen and verruculotoxin caused significant inhibition of the channel's binding of [35S]TBPS. The data suggest that the tremorgenic action of these mycotoxins may be due in part to their inhibition of GABAA receptor function.  相似文献

Presynaptic nerve terminals of inhibitory synapses in the dorsal horn of the spinal cord and brain stem can release both GABA and glycine, leading to coactivation of postsynaptic GABAA and glycine receptors. In the present study we have analyzed functional interactions between GABAA and glycine receptors in acutely dissociated neurons from rat sacral dorsal commissural nucleus. Although the application of GABA and glycine activates pharmacologically distinct receptors, the current induced by a simultaneous application of these two transmitters was less than the sum of currents induced by applying two transmitters separately. Sequential application of glycine and GABA revealed that the GABA-evoked current is more affected by glycine than glycine-evoked responses by GABA. Activation of glycine receptors decreased the amplitude and accelerated the rate of desensitization of GABA-induced currents. This asymmetric cross-inhibition is reversible, dependent on the agonist concentration applied, but independent of both membrane potential and intracellular calcium concentration or changes in the chloride equilibrium potential. During sequential applications, the asymmetric cross-inhibition was prevented by selective GABAA or glycine receptor antagonists, suggesting that occupation of binding sites did not suffice to induce glycine and GABAA receptors functional interaction, and receptor channel activation is required. Furthermore, inhibition of phosphatase 2B, but not phosphatase 1 or 2A, prevented GABAA receptor inhibition by glycine receptor activation, whereas inhibition of phosphorylation pathways rendered cross-talk irreversible. Taken together, our results demonstrated that there is an asymmetric cross-inhibition between glycine and GABAA receptors and that a selective modulation of the state of phosphorylation of GABAA receptor and/or mediator proteins underlies the asymmetry in the cross-inhibition.  相似文献

本文的目的是为了探讨γ-氨基丁酸是否可激发人精子顶体反应,受主其可能的作用方式。实验采用金霉素荧光染色法,胞内游离C62+测定和精子穿透去透明带仓鼠卵试验,分别评价GABA诱发人精子AR及其穿卵能力。结果表明;GABA可诱发人获能精子发生AR,且随精子获得进程而显著增加,并存在明显的量效关系,该作用可被P4加强。  相似文献

用微电极技术研究不同浓度吗啡对豚鼠右心室乳头肌动作电位的作用及作用机制。低浓度吗啡(0.2—1.6 umol/L)使动作电位时程(APD)和有效不应期(ERP)缩短,呈剂量依赖性。此作用可被1μmol/L 纳洛酮、酚妥拉明,四乙胺和氯化铯阻断,但不能被异搏定阻断。高浓度吗啡(15—120μmol/L)则使 APD 和 ERP 延长,呈剂量依赖性,这作用不被1.2μmol/L纳洛酮阻断,但可被10μmol/L 纳洛酮、酚妥拉明、四乙胺、氯化铯和异搏定阻断。这些实验提示低浓度和高浓度的吗啡可能作用于不同的阿片受体亚型,低浓度吗啡的作用可能与钾通道有关,高浓度吗啡的作用可能与钾通道、钙通道或钙激活的钾通道有关。阿片受体的作用与α受体存在密切关系。  相似文献

Different combinations of cloned rat brain subunit isoforms of the GABAA receptor channel were expressed in Xenopus oocytes. The voltage-clamp technique was then used to measure properties of the GABA-induced membrane currents and to study the effects of various modulators of the GABAA receptor channel (diazepam, DMCM, pentobarbital, and picrotoxin). This approach was used to obtain information on the minimal structural requirements for several functional properties of the ion channel. The combination alpha 5 beta 2 gamma 2 was identified as the minimal requirement reproducing consensus properties of the vertebrate GABAA receptor channel, including cooperativity of GABA-dependent channel gating with a Ka in the range of 10 microM, modulation by various drugs acting at the benzodiazepine binding site, picrotoxin sensitivity, and barbiturate effects.  相似文献

We have studied the effects of GABA on cytosolic free Ca2+ concentration ([Ca2+]i) as a means of investigating the role of GABA in adrenal catecholamine (CA) secretion. It was demonstrated that GABA caused an elevation of [Ca2+]i via the GABAA receptor in a concentration-dependent manner, which was well correlated with an increase of 45Ca uptake, an increase of CA release and a depolarization of chromaffin cells assessed with bis-oxonol fluorescence. Since the GABA-induced rise of [Ca2+]i was absolutely dependent on the presence of extracellular Ca2+ and partly sensitive to nifedipine, at least one entry route for Ca2+ facilitated by GABA via a voltage-sensitive Ca2+ channel was suggested. When extracellular Cl- was lowered, GABA-induced CA release, depolarization, and rise of [Ca2+]i were all markedly enhanced. It is possible that GABA plays a modulatory role in the regulation of adrenal CA secretion as a facilitatory modulator.  相似文献

The sodium channels of dissociated single ventricular cells of adult guinea pig heart were inactivated by partial depolarization in high K+ (25 mmol/L) Tyrode's solution and slow response action potential was elicited by intracellular stimulation. An obvious inhibition of the response was observed in the presence of 20 mumol/L sodium tanshinone II-A sulfonate (DS-201). In the concentration range from 1 mumol/L to 20 mumol/L, the inhibition effect of sodium tanshinone II-A sulfonate on the slow response action potentials enhanced by 0.28 mumol/L isoprenaline is concentration-dependent. Moreover, the inhibition effects of sodium tanshinone II-A sulfonate become stronger with the increase (in the range of 6.9 nmol/L to 0.55 mumol/L) of isoprenaline. The above-mentioned results suggest that sodium tanshinone II-A sulfonate may be a kind of effective calcium channel blocker. Under the effect of high concentration (50-100 mumol/L) of sodium tanshinone II-A sulfonate, the amplitude of fast response action potential of dissociated ventricular myocytes of adult guinea pig was decreased and the time to reach the peak was prolonged. All these results indicate that sodium channels were blocked to a certain extent by the high concentration of sodium tanshinone II-A sulfonate.  相似文献
