为了探讨氟化物在家蚕Bombyx mori体内的代谢途径, 以家蚕耐氟品种T6和氟化物敏感品种734为研究材料, 在5龄幼虫1-7 d内分别添食经50, 100, 200和400 mg/kg NaF溶液浸泡后的新鲜桑叶, 检测家蚕中肠羧酸酯酶(CarE)和全酯酶活性的变化。结果表明: 734添氟组的CarE活性是对照组的1.21~1.98倍, 而T6添氟组约是对照组的0.72~1.10倍。734和T6添氟组的全酯酶活性数值变化规律与其各自对照组相似, 且2品种之间的酶活性数值很相近。2品种在相同浓度下, 不同天数之间的全酯酶活性差异均显著(P<0.05)。推测氟化物对敏感家蚕中肠CarE有促进作用, 对耐氟家蚕中肠CarE有抑制作用, 但是对全酯酶活性影响不大。  相似文献

为了探讨氟化物对家蚕代谢机制的影响,以家蚕耐氟品种T6和氟化物敏感品种734为研究对象,从5龄起蚕开始分别添食50、100、200、400mg/kg NaF溶液浸泡后的新鲜桑叶,检测家蚕血液中羧酸酯酶(CarE),全酯酶活性的变化。结果表明,734、T6添氟组的CarE活性分别是对照组的73%—88%和72%—81%,734两个低浓度添氟组的CarE活性与对照组和两个高浓度添氟组的差异极显著(P<0.01),T6各处理组之间的差异不显著。734、T6添氟组的全酯酶活性分别是对照组的89%—97%和73%—92%,734各处理组之间的差异不显著,T6对照组的全酯酶活性仅与最高浓度添氟组差异极显著(P<0.01)。说明氟化物对家蚕血液CarE和全酯酶活性具有一定的抑制作用。  相似文献

【目的】研究氟化物对家蚕Bombyx mori肠道内留存产酶菌的影响, 有助于了解家蚕耐氟和氟敏品种的耐氟力差异。【方法】分别给家蚕耐氟品种T6和氟敏品种734添食NaF溶液浸泡后的新鲜桑叶, 至5龄第3天取材。采用筛选培养基筛选产蛋白酶、 纤维素酶、 脂肪酶、 淀粉酶的菌株, 并结合16S rDNA系统发育关系, 对菌株进行分类鉴定。【结果】家蚕肠道内产消化酶细菌有芽孢杆菌属Bacillus、 葡萄球菌属Staphylococcus、 肠杆菌属Enterobacter和不动杆菌属Acinetobacter和微小杆菌属Exiguobaterium, 其中芽孢杆菌Bacillus sp.、 葡萄球菌Staphylococcus sp. 和不动杆菌Acinetobacter sp.细菌可同时产4种酶。氟中毒后T6肠道内产酶菌由5种减少到4种, 734肠道内产酶菌由2种减少到1种。【结论】家蚕肠道内留存的产酶菌与家蚕自身的耐氟能力相关。  相似文献

摘要:【目的】探讨氟化物对家蚕肠道微生物菌群的影响,开发具有潜在应用价值的益生菌以提高家蚕对氟化物的抗性。【方法】PCR扩增家蚕肠道内细菌16S rRNA基因并构建克隆文库;用核糖体DNA限制性分析(Amplified ribosomal DNA restriction analysis,ARDRA)方法对克隆子进行分型。从家蚕T6和734肠道样品中共获得14种分类操作单元(Operational taxonomic unit,OTUs),以16S rRNA基因为基础构建系统发育树进行分析。再经巢式PCR-DGGE技术检测家蚕肠道内优势菌群的变化。【结果】结果表明:家蚕氟中毒后肠道内肠球菌属Enterococcus和芽孢杆菌属Bacillus细菌菌群减少,而葡萄球菌属Staphylococcus的细菌增多。【结论】氟化物能通过改变家蚕肠道内细菌的多样性和比例,从而破坏家蚕肠道微生物菌群平衡,且对家蚕734的影响作用大于T6。  相似文献

随着有机氟化物在各领域的广泛应用,含氟有机废水处理面临巨大挑战。活性污泥作为有机废水处理的核心技术之一,微生物在其中发挥着极其重要的作用。本综述首先聚焦在活性污泥微生物群落多样性、组成、结构和功能及其与含氟废水类型、处理工艺和处理效率之间的关系,进而讨论了功能微生物降解/转化有机氟化物的途径和作用机制,最后展望了结合分离培养降解有机氟化物的关键微生物,以及微生物组学技术解析活性污泥微生物群落构建、互作、代谢等核心问题,以提高对含氟有机废水微生物降解机理的认识,优化含氟有机废水处理工艺。  相似文献

口腔微生物是人体微生物组的重要组成部分,其群落组成丰富且独特。现有研究显示,口腔微生物与龋病、牙周炎等口腔健康问题有直接的联系,因而具有重要的研究价值。随着高通量测序技术的发展,人们对口腔中未培养微生物多样性的认识不断加深,这进一步催生对微生物分离培养技术需求的增加。为此,本文将围绕口腔未培养微生物及其分离培养策略的研究进展,首先介绍口腔中未培养微生物的研究现状;其次分析口腔微生物分离培养中可能的限制因素;最后综述微生物分离培养技术发展及其在口腔未培养微生物研究中的应用。全文旨在为口腔未培养微生物的分离培养提供思路和技术参考。  相似文献

Tooth enamel is a hybrid organic–inorganic bionanocomposite comprised predominantly of enamel rods. Understanding the effects of anti-caries treatment on the biomechanical properties of these rods is essential in developing effective caries prevention strategies. Calcium fluoride-like deposits play an important role in caries prevention and their nanotribological properties have a direct effect upon their long-term effectiveness. Accordingly, this study utilizes a variety of techniques, namely nanoindentation, nanoscratch tests, nanowear tests and atomic force microscopy (AFM), to characterize the mechanical and tribological properties of single enamel rods before and after topical fluoride application. The results show that the CaF2-like deposits formed on the enamel surface following fluoride application increase the coefficient of friction of the enamel rods, but decrease their critical load and nanohardness. As a result, the nanowear depth of the treated enamel surface is around six times higher than that of the native enamel surface under an applied load of 300 μN. Following the removal of the surface deposits, however, the modulus of elasticity and wear depth of the underlying enamel surface are found to be similar to those of the original enamel surface. However, a notable increase in the surface roughness is observed.  相似文献

Dental caries remains one of the most common global chronic diseases caused by Streptococcus mutans,which is prevalent all over the world.The caries prevalence of children aged between 5-6 years old in China is still in very high rate.A potent and effective anti-caries vaccine has long been expected for caries prevention but no vaccines have been brought to market till now mainly due to the low ability to induce and maintain protective antibody in oral fluids.This review will give a brief historical retrospect on study of dental caries and pathogenesis,effective targets for anti-caries vaccines,oral immune system and immunization against dental caries.Then,salivary IgA antibodies and the protective responses are discussed in the context of the ontogeny of mucosal immunity to indigenous oral streptococcal.The methods and advancement for induction of specific anticaries salivary sIgA antibodies and enhancement of specific anti-caries salivary sIgA antibodies by intranasal immunization with a safe effective mucosal adjuvant are described.The progress in the enhancement of salivary sIgA antibodies and anticaries protection by intranasal immunization with flagellin-PAc fusion protein will be highlighted.Finally,some of the main strategies that have been used for successful mucosal vaccination of caries vaccine are reviewed,followed by discussion of the mucosal adjuvant choice for achieving protective immunity at oral mucosal membranes for development of a nasal-spray or nasal-drop anti-caries vaccine for human.  相似文献

The aim of the study was to evaluate the amount of fluoride remaining in the oral cavity of children after brushing with fluoride gel (1.25% F). The study involved six groups of 7-year-old and six groups of 11-year-old children. The procedure was carried out according to the manufacturer’s recommendations. Fluoride concentrations were determined using ion-selective fluoride electrode. No statistically significant difference was found between the amount of fluorides that remained in the oral cavity of younger and older age group (1.2 and 1.3 mg, respectively; p > 0.05). The amount of fluorides swallowed during the procedure in both age groups proves to be within acceptable limit, as far as risk of acute poisoning symptoms is concerned. The individual daily fluoride exposure during the day of procedure seems to be twice as high compared to average fluoride intake from diet and dentifrice, and it does not exceed Tolerable Upper Intake Level for children more than 8. In younger children, it seems justifiable to reduce the amount of the preparation applied on a toothbrush, especially when daily use of the gel is recommended.  相似文献

Saliva is a critical and useful biological fluid necessary for good health and for the appropriate execution of mouth activities. Orthodontic biomaterials have a complex relationship with many components, including the oral environment. Treatment with fixed orthodontic appliances may causes dental caries. As a result, it is necessary to comprehend how orthodontic therapy and various fluoride regimens affect the chances of developing dental cavities as well as individual risk factors. Usage of fluoride will tend to reduce the caries in the patients diagnosed with the fixed orthodontic treatment. The aim of this study was to screen the biochemical parameter of the fluoride levels in the patients undergone and completed the treatment of orthodontic fixed appliances. In this study, 35 patients have been visited on day 1 as well as day 35 and categorized as T0 and T1 groups. Saliva samples were collected and fluoride levels were measured between T0 and T1 groups. Using the fluoride kit with the spectrophotometer, fluoride levels were measured. The results confirmed similar fluoride levels between T0 (26.11 ± 4.86) and T1 (27.71 ± 4.40) groups. There was no significant association observed in this study (p = 0.56). Fluoride might have no role in the patients undergoing orthodontic treatment.  相似文献

One of the major effects of fluoride on oral bacteria is a reduction in acid tolerance, and presumably also in cariogenicity. The reduction appears to involve transport of protons across the cell membrane by the weak acid HF to dissipate the pH gradient, and also direct inhibition of the F1F0, proton-translocating ATPases of the organisms, especially for Streptococcus mutans. This direct inhibition by fluoride was found to be dependent on aluminum. The dependence on aluminum was indicated by the protection against fluoride inhibition afforded by the Al-chelator deferoxamine and by loss of protection after addition of umolar levels of Al3+, which were not inhibitory for the enzyme in the absence of fluoride. The F1 form of the enzyme dissociated from the cell membrane previously had been found to be resistant to fluoride in comparison with the F1F0 membrane-associated form. However, this difference appeared to depend on less aluminum in the F1 preparation in that the sensitivity of the F1 enzyme to fluoride could be increased by addition of umolar levels of Al3+. The effects of Al on fluoride inhibition were apparent when enzyme activity was assayed in terms of phosphate release from ATP or with an ATP-regenerating system containing phosphoenolpyruvate, pyruvate kinase, NADH and lactic dehydrogenase. Also, Be2+ but not other metal cations, e.g. Co2+, Fe2+, Fe3+, Mn2, Sn2+, and Zn2+, served to sensitize the enzyme to fluoride inhibition. The differences in sensitivities of enzymes isolated from various oral bacteria found previously appeared also to be related to differences in levels of Al. Even the fluoride-resistant enzyme of isolated membranes of Lactobacillus casei ATCC 4646 could be rendered fluoride-sensitive through addition of Al3+. Thus, the F1F0 ATPases of oral bacteria were similar to E1E2 ATPases of eukaryotes in being inhibited by Al-F complexes, and the inhibition presumably involved formation of ADP-Al-F inf3 sup- complexes during catalysis at the active sites of the enzymes.  相似文献

BackgroundSuccessful oral health interventions must be based on the specific needs of the population that they serve. Evaluation of habits related to dental caries development and estimation of fluoride exposure in a target group of young patients helps to plan effective and safe caries prevention strategies.ObjectivesThe study aimed to evaluate factors affecting dental caries experience and sources of fluoride exposure in preschool children living in two areas: with optimal and low natural content of fluoride in drinking water.Materials and methodsThe study included a group of 73 children of both sexes aged 4–7 years attending two kindergartens in Środa Wielkopolska and Turek (Wielkopolska Voivodeship, Poland), where the content of fluoride in drinking water according to data obtained in the sanitary station ranged from 0.68 to 0.74 mg/L (optimal concentration of fluoride) and from 0.19 to 0.30 mg/L (low concentration of fluoride), respectively. Parents of patients completed a survey about diet, hygiene, and dental care, taking into account the child's fluoride exposure. The calibrated dentist assessed the oral health condition using a mirror, a CPI probe, and a headlamp. Oral hygiene was recorded using the Silness and Löe plaque index, caries experience by calculating the numbers of decayed, missing, and filled primary, and permanent teeth (dmf and DMF, respectively) while caries frequency by calculating the percentage of children with caries experience above 0. In order to assess the fluoride concentrations in urine and drinking water, parents were asked to provide a urine sample collected on fasting and a tap water sample. Fluoride concentrations were assessed using a 09–37 (MARAT) fluoride ion-selective electrode and a RAE 111 silver-chloride reference electrode. Statistical analysis was conducted using the data analysis software system Statistica (version 12, StatSoft, Inc. 2014), assuming a statistical significance level p < 0.05.ResultsNo statistically significant differences were found between caries indices of the examined children in each kindergarten (p > 0.05). Urinary fluoride levels were higher in children who tended to swallow toothpaste or used fluoride rinses and positively correlated with fluoride concentrations in the drinking water. Dental caries experience in the examined children depended on the effectiveness and frequency of oral hygiene procedures and dietary habits.ConclusionsThe strategy aimed at improving the oral health of the examined group of children should include accomplishing oral hygiene, promoting a non-cariogenic diet, and, finally, controlling fluoride exposure from at-home fluoride products. Caries prevention program ought to be adjusted to individual characteristics of each child, taking into consideration oral hygiene practices, dietary habits and total fluoride intake.  相似文献

A fluoride-resistant Streptococcus mutans isolate NCH105 was characterized and compared with wild-type strain UA130. The growth and lactic acid production of strain NCH105 were found to be unaffected by the presence of fluoride at the initial medium pH values of 6.5 and 6. In addition, NCH105 was found to be capable of lowering the pH of the medium to approximately 5.5, which is the critical level where enamel demineralization begins in vivo. Lactic acid production, glucose uptake at pH 6.5 and 6, as well as ATPase activity at pH 5 were found to be unaffected by fluoride. Finally, strain NCH105 is capable of binding as well as the wild-type bacterium to artificial tooth pellicles. These results are unusual when compared with previously isolated fluoride-resistant mutants and suggest that NCH105 may have the ability to colonize the tooth surface and initiate dental caries.  相似文献

The key role of osteoblasts in skeletal fluorosis makes the exploration of the possible mechanisms of the fluoride-induced oxidative stress of osteoblasts of great importance. In this article, the in vitro effects of fluoride on the oxidative stress of osteoblasts are presented. To study the inhibitory effect of baicalein on the oxidative stress of osteoblasts, the antioxidant activity of baicalein was evaluated for osteoblasts exposed to fluoride. Calvarial osteoblasts were prepared and respectively treated with α-MEM (5% calf serum) containing 0.5, 1.0, 2.0, 4.0, 8.0, 12.0, and 20.0 mg/L fluoride for 48 h. Baicalein (10 μmol/L) was added to the cells for the same period of time as that of the fluoride treatment. Low concentrations of fluoride (0.5–2 mg F-/L) stimulated the mitochondrial activity of osteoblasts and produced significant reaction to the oxidative stress, whereas high concentrations of fluoride (≽12 mg F-/L) inhibited cell proliferation and the activity of antioxidant enzymes. This suggests that the oxidative stress induced by low concentrations of fluoride might mediate or participate in the process of fluoride inducing the proliferation of osteoblasts. The viability of osteoblasts in the high concentrations of fluoride with the addition of 10 μmol/L baicalein (≽12 mg /L) was higher than those of the same level of fluoride-treated groups without the addition of baicalein. The protective role of baicalein is obvious as an inhibitor of lipid peroxidation against the damage induced by the high concentration of fluoride.  相似文献

In previous epidemiological studies, it has been suggested that geochemical factors besides fluoride may affect the prevalences of dental diseases. Our objective in this study was to determine whether the prevalences of periodontal diseases, coronal caries, and root caries for prehistoric inhabitants vary between geochemical regions of the state of Missouri. Burial sites were located on unique maps that depict geochemical variation among regions of the state. Data on dental caries and alveolar bone loss were gathered from 179 of the best preserved skeletal remains of the Late Woodland (A.D. 400-900) and Mississippian (A.D. 900-1700) periods. Mean caries scores and proportions of individuals with caries were calculated for these geochemical regions. Average alveolar bone loss was regressed on age for the individuals of these regions. Significant differences in caries and bone loss were found between several regions. These differences cannot be adequately explained by fluoride concentrations or by diet. It is suggested that geochemical factors, in addition to fluoride, may have affected the prevalences of dental diseases.  相似文献

In general, an antimicrobial test for screening anti-caries natural extracts was performed by measuring the minimum bactericidal concentration (MBC) against the type strains of mutans streptococci. However, it is unclear if the antimicrobial efficiency of natural extracts on the type strains of mutans streptococci is the same on the clinical strains. In this study, we introduced a bacterial model system for the screening of anti-caries and determining the optimal concentration of them to develop oral hygiene products for Korean populations.  相似文献
