The green seaweed Ulva has been shown to detect signal molecules produced by bacteria. Biofilms that release N-acylhomoserine lactones (AHLs) attract zoospores--the motile reproductive stages of Ulva. The evidence for AHL involvement is based on several independent lines of evidence, including the observation that zoospores are attracted to wild-type bacteria that produce AHLs but are not attracted to mutants that do not produce signal molecules. Synthetic AHL also attracts zoospores and the attraction is lost in the presence of autoinducer inactivation (AiiA) protein. The mechanism of attraction is not chemotactic but involves chemokinesis. When zoospores detect AHLs, the swimming rate is reduced and this results in accumulation of cells at the source of the AHL. It has been demonstrated that the detection of AHLs results in calcium influx into the zoospore. This is the first example of a calcium signalling event in a eukaryote in response to bacterial quorum sensing molecules. The role of AHLs in the ecology of Ulva is discussed. It is probable that AHLs act as cues for the settlement of zoospores, rather than being directly involved as a signalling mechanism.  相似文献

The strigolactones are internal and rhizosphere signalling molecules in plants that are biosynthesised through carotenoid cleavage. They are secreted by host roots into the rhizosphere where they signal host-presence to the symbiotic arbuscular mycrorrhizal (AM) fungi and the parasitic plants of the Orobanche, Phelipanche and Striga genera. The seeds of these parasitic plants germinate after perceiving these signalling molecules. After attachment to the host root, the parasite negatively affects the host plant by withdrawing water, nutrients and assimilates through a direct connection with the host xylem. In many areas of the world these parasites are a threat to agriculture but so far very limited success has been achieved to minimize losses due to these parasitic weeds. Considering the carotenoid origin of the strigolactones, in the present study we investigated the possibilities to reduce strigolactone production in the roots of plants by blocking carotenoid biosynthesis using carotenoid inhibitors. Hereto the carotenoid inhibitors fluridone, norflurazon, clomazone and amitrole were applied to rice either through irrigation or through foliar spray. Irrigation application of all carotenoid inhibitors and spray application of amitrole significantly decreased strigolactone production, Striga hermonthica germination and Striga infection, also in concentrations too low to affect growth and development of the host plant. Hence, we demonstrate that the application of carotenoid inhibitors to plants can affect S. hermonthica germination and attachment indirectly by reducing the strigolactone concentration in the rhizosphere. This finding is useful for further studies on the relevance of the strigolactones in rhizosphere signalling. Since these inhibitors are available and accessible, they may represent an efficient technology for farmers, including poor subsistence farmers in the African continent, to control these harmful parasitic weeds.  相似文献

Breeding for resistance to Striga in maize (Zea mays), with paucity of donor source and known mechanisms of resistance, has been challenging. Here, post-attachment development of S. hermonthica was monitored on two maize inbreds selected for field resistance and susceptibility reactions to Striga at the International Institute of Tropical Agriculture. Haustorial invasion of the parasite into roots of these inbreds was examined histologically. Morphological differences were observed between roots of the susceptible and the resistant inbreds. The resistant maize had fewer Striga attachments, delayed parasitic development and higher mortality of attached parasites compared with the susceptible inbred. Striga on the susceptible inbred usually penetrated the xylem and showed substantial internal haustorial development. Haustorial ingress on the resistant inbred was often stopped at the endodermis. Parasites able to reach resistant host xylem vessels showed diminished haustorial development relative to those invading susceptible roots. These results suggest that the resistant inbred expresses a developmental barrier and incompatible response against Striga parasitism.  相似文献

The rpoS gene which encodes a stationary phase sigma factor has been identified and characterised from the rhizosphere-colonising plant growth-promoting Pseudomonas putida strain WCS358. The predicted protein sequence has extensive homologies with the RpoS proteins form other bacteria, in particular with the RpoS sigma factors of the fluorescent pseudomonads. A genomic transposon insertion in the rpoS gene was constructed, these mutants were analysed for their ability to produce siderophore (iron-transport agent) and the autoinducer quorum-sensing molecules called homoserine lactones (AHL). It was determined that RpoS was not involved in the regulation of siderophore and AHL production, synthesis of these molecules is important for gene expression at stationary phase. P. putida WCS358 produces at least three different AHL molecules.  相似文献

The phytochemical study of M. polymnioides led to the isolation of two sesquiterpene lactones namely: 11alphaH-dihydrozaluzanin E and 1beta-hydroxy-4-oxo-11betaH-4-noreudesman-6,12-olide. Their structures were determined by spectroscopic methods. The relative stereochemistry was established by a combination of coupling constant analysis, NOESY correlations and molecular modeling. Three related known sesquiterpene lactones were also identified, and these data were used for chemotaxonomical purposes.  相似文献

The parasitic weed Striga causes devastating losses in cereal yields in sub-Saharan Africa. The parasite lifecycle is intimately linked with its host via a complex interchange of signals. Understanding the molecular basis of these interactions and of host resistance to Striga is essential for the identification of genes for improving crop yield via biotechnological or marker assisted breeding strategies. Cloning and sequencing of ESTs from the 'model' parasite Triphysaria versicolor is facilitating the identification of parasitism genes. The identification of resistance to Striga in sorghum and rice germplasm is allowing molecular dissection of these traits using genomic platforms and quantitative trait loci (QTL) analysis. QTL underlying different resistance phenotypes have been identified and the use of advanced backcross populations is allowing the exploitation of sources of resistance in wild relatives of cereals.  相似文献

Simple (non‐tandem) electrospray mass spectrometry (ESMS) can detect acyl homoserine lactones (AHL) in bacteriological media in picomole amounts. The chemical reactivity of AHLs and their extraction behaviour into ethyl acetate, coupled with detection in the ESMS, has shown that these lactones can be detected as the protonated pseudomolecular ions themselves as well as solvent and ammonium adducts, and as dimers. The Striga, particularly S. he rmonthica, problem has become a major threat to food security, exacerbating hunger and poverty in many African countries. A number of Striga control strategies have been proposed and tested during the past decade, however, further research efforts are still needed to provide sustainable and effective solutions to the Striga problem. In this paper, we provide an update on the recent progress and the approaches used in Striga management, and highlight emerging opportunities for developing new technologies to control this enigmatic parasite.

A germination stimulant, alectrol, for the seeds of the angiospermous root parasites Alectra vogelii Benth. and Striga gesnerioides (Willd.) Vatke has been isolated from root exudates of its genuine host plant Vigna unguiculata Walp cv Saunders Upright. Its spectroscopic data lead to a chemical structure closely related to (+)-strigol. From Picris kamtschatica two new guaianolide glycosides, including a beta-allopyranosyl analogue of ixerin F, were isolated, in addition to ten known sesquiterpene lactones and six phenolic compounds. This is the first time a sesquiterpene lactone alloside has been described from plants. The compounds were characterized based on mass, 1D and 2D NMR spectral data.  相似文献

The cell-to-cell communication of microorganisms is known to be via exertion of certain chemical compounds (signal molecules) and is referred to as quorum sensing (QS). QS phenomenon is widespread in microbial communities. Several Gram-positive and Gram-negative bacteria and fungi use lactone-containing compounds (e.g. acyl-homoserine lactones (AHLs), γ-heptalactone, butyrolactone-I) as signalling molecules. The ability of microorganisms to metabolise these compounds and the mechanisms they employ for this purpose are not clearly understood. Many studies, however, have focused on identifying AHL and other lactone-degrading enzymes produced by bacteria and fungi. Various strains that are able to utilise these signalling molecules as carbon and energy sources have also been isolated. In addition, several reports have provided evidence on the involvement of lactones and lactone-degrading enzymes in numerous biological functions. These studies, although focused on processes other than metabolism of lactone signalling molecules, still provide insights into further understanding of the mechanisms employed by various microorganisms to metabolise the QS compounds. In this review, we consider conceivable microbial strategies to metabolise AHL and other lactone-containing signalling molecules such as γ-heptalactones.  相似文献

This protocol describes a procedure for the synthesis of a 29-membered macrolactone. The facile mixed-anhydride method is very effective for the preparation of carboxylic esters and lactones using substituted benzoic anhydrides by the promotion of Lewis acid or basic catalysts under mild reaction conditions. Owing to the reaction rapidly proceeding to produce the monomeric compounds with high chemoselectivity, the protocol is quite powerful for the synthesis of not only the giant-sized lactones but also the highly strained cyclic compounds such as medium-sized lactones. The remarkable efficiency of the lactonizations promoted by the substituted benzoic anhydrides has been already shown in the synthesis of many natural complex molecules. It takes approximately 19 h to complete the protocol: 0.5 h to set up the reaction, 13.5 h for the reaction and 5 h for isolation and purification.  相似文献

Kava-kava is a traditional beverage of the South Pacific islanders and has had centuries of use without major side effects. Standardised extracts of kava-kava produced in Europe have led to many serious health problems and even to death. The extraction process (aqueous vs. acetone in the two types of preparations) is responsible for the difference in toxicity as extraction of glutathione in addition to the kava lactones is important to provide protection against hepatotoxicity. The Michael reaction between glutathione and kava lactones, resulting in opening of the lactone ring, reduces the side effects of the kava kava extracts. This protective activity was demonstrated using Acanthamoebae castellanii in which 100% cell death occurred with 100 mg ml(-1) kava lactones alone, and 40% cell death with a mixture of 100 mg ml (-1)glutathione and 100 mg ml (-1) kava lactones. A comparison of kava lactone toxicity with other pharmaceutical products is discussed and recommendations made for safe usage of kava-kava products  相似文献

An ethanolic extract of the aerial parts of Matricaria Chamomilla L. collected at Oued Tonga near El-Kala is eastern Argelia yielded two new substances named matricolone and chamolol, together with four known compounds. The structure of the new compounds were determined by chemical transformations and NMR spectroscopy with 1H-1H (COSY and ROESY), 1H-13C (one bond and long range correlations) two dimensional experiments. The results obtained indicate the presence of sesquiterpene lactones of the eudesmanolide, germacranolide and guaianolide types. These products can be considered to be of great interest from a chemotaxonomical point of view for this wild-growing plant, because these types of lactones have not been found together in other Matricaria species.  相似文献

Investigations were carried out at the Gezira Research Farm in the Anglo-Egyptian Sudan. The greater majority of the seeds of Striga hermonthica can only be germinated by excretions from roots of certain plants not all of which can act as hosts for this parasite. Unless it becomes attached to a host plant, the Striga seedling dies. The growing of those plants whose roots can stimulate the germination of Striga seed but cannot be parasitized by it, may be a means of ridding infested land of this parasite. Green ovaries picked from flowering plants produce viable seeds if left to dry. Sorghum is sown in the field during the rainy season, viz. June to October: the earlier the sowing date within this period the greater is the Striga attack. In the field Striga seeds are distributed in the soil to a depth of at least 15 in. When Striga seeds are evenly distributed through the soil, the number of Striga seedlings attached to a sorghum root is proportional to the root's development. Size of sorghum seed has no effect on the root size of a sorghum plant and consequently no effect on the degree of parasitism. The effect of severe Striga attack on the sorghum plant is to produce a reduction of about 60% in leaf and root weight. No reduction of Striga attack is obtained when various micro-elements are coated on sorghum seeds before sowing. Field and laboratory experiments show that light irrigation of the sorghum crop during the normal sowing period increases the Striga attack and heavy irrigation decreases it. This result was not obtained in laboratory experiments when sorghum was sown out of season. Striga attack is lessened when conditions favouring growth of the sorghum crop are improved.  相似文献

The herbivory activity of the bordered patch larvae (Chlosyne lacinia, Lepidoptera) on leaves of a Brazilian population of Tithonia diversifolia and the antifeedant potential of its leaf rinse extract were investigated. The caterpillars fed only on the adaxial face, where the density of glandular trichomes is very low, and avoided the abaxial face, which contains high levels of trichomes. Deterrent activity against the larvae was observed in leaf discs treated with leaf rinse extract at concentrations of 1-5% of fresh leaf weight. High-performance liquid chromatography (HPLC) analysis indicated that sesquiterpene lactones are the main constituents of the glandular trichomes. Dichloromethane rinse extracts of the leaves and inflorescences were chemically investigated, and 16 compounds were isolated and identified: 14 sesquiterpene lactones, a flavonoid and a diterpenoid. In this study, five sesquiterpene lactones are described for the first time in the genus, including two lactones, one of which has an unusual seco-guaianolide skeleton. Our findings indicate that the caterpillars avoid the sesquiterpene-lactone-rich glandular trichomes, and provide evidence for the antifeedant activity of the dichloromethane leaf rinse extract. In addition, a study of the seasonal variation of the main constituents from the leaf surface throughout a year demonstrated that a very low qualitative but a very high quantitative variation occurs. The highest level of the main metabolite tagitinin C was observed between September and October and the lowest was from March to June, the later corresponding to the period of highest infestation by the larvae.  相似文献

Experiments were conducted in a Striga-sick field to study the effect of soil temperature, moisture and nitrogen on Striga parasitism on sorghum. Striga seeds contained in nylon bags and buried at 2 cm in the soil, were exposed to different temperature and moisture treatments. Clear polythene, hay mulch and bare soil treatments were used to vary soil temperature. These treatments gave mean maximum temperatures of 60°, 48° and 37°C, respectively at 2-cm soil depth. Irrigation levels of 0, 30 and 60 mm were applied to change soil moisture. Striga seed germination, viability and emergence were studied. After 34 days of preconditioning, the exhumed Striga seeds from polythene-covered plots (solarized plots) did not germinate or retain viability when these seeds were exposed to sorghum root exudate. However, seeds similarly buried under hay mulch or bare soil, with mean maximum soil temperatures of 48° and 37°C, respectively, had similar germination and viability percentages. Of these 75% germinated and 85% of them were viable, regardless of the temperature treatment. Although seeds stored at high temperature and humidity (solarization) were killed, more Striga plants emerged under the polythene treatment compared to hay mulch and bare soil treatments. The observed Striga plants in the polythene mulch treatment were, therefore, assumed to have come from deeper layers where solarization was not effective. Irrigation treatments did not have significant effects on Striga seed germination and viability, but a slightly higher number of plants emerged at 60-mm irrigation level than at 30-mm and 0-mm. Striga emergence, on the other hand, was directly related to the rate of N application. Nitrogen rates of 0, 25, 50 and 100 kg ha–1 resulted in the emergence of 11, 34, 38 and 40 Striga plants per plot, respectively. Despite the high infestation at high N levels, sorghum plants did not show a loss of vigor. Nitrogen application, therefore, does not reduce Striga incidence, but seems to neutralize the harmful effects of Striga without reducing the extent of parasitism.  相似文献

The seeds of parasitic plants of the genera Striga and Orobanche will only germinate after induction by a chemical signal exuded from the roots of their host. Up to now, several of these germination stimulants have been isolated and identified in the root exudates of a series of host plants of both Orobanche and Striga spp. In most cases, the compounds were shown to be isoprenoid and belong to one chemical class, collectively called the strigolactones, and suggested by many authors to be sesquiterpene lactones. However, this classification was never proven; hence, the biosynthetic pathways of the germination stimulants are unknown. We have used carotenoid mutants of maize (Zea mays) and inhibitors of isoprenoid pathways on maize, cowpea (Vigna unguiculata), and sorghum (Sorghum bicolor) and assessed the effects on the root exudate-induced germination of Striga hermonthica and Orobanche crenata. Here, we show that for these three host and two parasitic plant species, the strigolactone germination stimulants are derived from the carotenoid pathway. Furthermore, we hypothesize how the germination stimulants are formed. We also discuss this finding as an explanation for some phenomena that have been observed for the host-parasitic plant interaction, such as the effect of mycorrhiza on S. hermonthica infestation.  相似文献

Acylated homoserine lactones (AHLs) are self-generated signal molecules that mediate population density-dependent gene expression (quorum sensing) in a variety of Gram-negative bacteria. These signal molecules diffuse from bacterial cells and accumulate in the medium as a function of cell growth. In selected foods AHLs contribute to product spoilage. As different bacterial species produce AHL analogs that differ in length of the N-acyl chain, ranging from 4 to 14 carbons and in the substitution at the C-3 position of the side chain (i.e., oxo or hydroxyl group), the suitability and applicability of a gas chromatography-mass spectrometry direct method for characterizing trace amounts of AHLs was evaluated using N-heptanoyl-homoserine lactone as internal standard. Crude cell-free supernatants of bacterial cultures of Aeromonas hydrophila, Aeromonas salmonicida, Pseudomonas aeruginosa, Pseudomonas fluorescens, Yersinia enterocolitica, and Serratia liquefaciens were screened for AHL production in selected ion monitoring mode, using the prominent fragment at m/z 143. The observed profiles of distinguishable N-acyl-homoserine lactones occurring in bacterial extracts were compared and discussed. The presence of a labile 3-oxo-hexanoylhomoserine lactone was evidenced but serious difficulties arose in estimating its concentration as thermal degradation occurs during the gas chromatographic separation. Oxidized halogen antimicrobials, such as hypochlorous and hypobromous acids, have been used extensively for microbial control in industrial systems. Recent discoveries have shown that acylated homoserine lactone cell-to-cell signaling molecules are important for biofilm formation in Pseudomonas aeruginosa, suggesting that biofouling can be controlled by interfering with bacterial cell-to-cell communication. This study was conducted to investigate the potential for oxidized halogens to react with acylated homoserine lactone-based signaling molecules. Acylated homoserine lactones containing a 3-oxo group were found to rapidly react with oxidized halogens, while acylated homoserine lactones lacking the 3-oxo functionality did not react. The Chromobacterium violaceum CV026 bioassay was used to determine the effects of such reactions on acylated homoserine lactone activity. The results demonstrated that 3-oxo acyl homoserine lactone activity was rapidly lost upon exposure to oxidized halogens; however, acylated homoserine lactones lacking the 3-oxo group retained activity. Experiments with the marine alga Laminaria digitata demonstrated that natural haloperoxidase systems are capable of mediating the deactivation of acylated homoserine lactones. This may illustrate a natural defense mechanism to prevent biofouling on the surface of this marine alga. The Chromobacterium violaceum activity assay illustrates that reactions between 3-oxo acylated homoserine lactone molecules and oxidized halogens do occur despite the presence of biofilm components at much greater concentrations. This work suggests that oxidized halogens may control biofilm not only via a cidal mechanism, but also by possibly interfering with 3-oxo acylated homoserine lactone-based cell signaling.  相似文献
