Restriction fragment length polymorphisms (RFLPs) were examined in 12 winter strains of the marine diatom Skeletonema costatum (Grev.) Cleve using homologous chloroplast gene probes. The winter strains included eight different allozyme genotypes exhibiting physiological differences. These 12 winter strains were representative of the least diverse genetic group present in Narragansett Bay populations. Five chloroplast DNA probes and four different restriction enzymes were used to analyze the 12 Narragansett Bay strains and a reference strain “Skel.” A total of 46 restriction fragments were identified. All 12 of the winter strains had identical patterns. Strain Skel exhibited two RFLPs in comparison to the Narragansett Bay strains. Calculated diversity within the winter strain group was 0.0 and 0.85 for the chloroplast DNA and allozyme data, respectively. The chloroplast DNA polymorphisms revealed by this study are expected to represent a minimum level of the chloroplast DNA diversity present in Narragansett Bay seasonal populations.  相似文献

Abstract In reciprocal transplant experiments, Bertness and Gaines (1993) found that Semibalanus balanoides juveniles that had settled in an upper Narragansett Bay estuary survived better in that estuary that did juveniles from coastal localities. The observed pattern of survivorship led to the claim that local adaptation may result from a combination of limited gene flow between and strong selection within these habitats. Here we test the hypothesis that limited gene flow has led to habitat‐specific population differentiation using sequence and restriction fragment length polymorphism analyses of the mitochondrial DNA D‐loop region of S. balanoides. Samples were analyzed from replicated coastal and estuary localities in both Narragansett Bay, Rhode Island, and Damariscotta River, Maine. The patterns of FST indicate that gene flow between coast and estuary is extensive (Nm > 100) and is not lower in the estuary with lower flushing rates (Narragansett Bay). Given the high estimate of genetic exchange, adaptations for unpredictable environments seem more likely than local adaptation in this species because loci that respond to selection in one generation are essentially homogenized by the next seasons' settlement. Nevertheless, these estimates of neutral gene flow can help identify the strength of selection necessary for local adaptation to accumulate in Semibalanus.  相似文献

The brown tide picoalga Aureococcus anophagefferens Hargraves et Sieburth was present in approximately equal numbers in 12 large scale (13,000 L) mesocosms at the start of a nutrient addition experiment in June 1985. Increases in abundance in untreated systems mimicked the pattern of bloom development in Narragansett Bay, Rhode Island, the seawater source for the experiment. Aureococcus increased to maximal values of 2.6 × 109 cells. L?1 and persisted at high numbers (108 cells·L?1) for 7–8 weeks. In nutrient addition tanks, the picoalgae bloomed briefly (1–3 weeks) but rapidly declined to the usual level (~107 cells·L?1) for eukaryotic algae in Narragansett Bay. The decline in picoalgae abundance was followed by an increase in total diatoms in all nutrient treated tanks. Mean picoalgae abundance in the mesocosms and the bay was significantly (P < 0.05) and inversely correlated (r =–0.93) with mean concentration of dissolved inorganic nitrogen. The persistence of the brown tide species in control mesocosms and Narragansett Bay appears related to its ability to grow at very low concentrations of dissolved inorganic nitrogen, levels previously shown to limit diatom growth.  相似文献

Nutrient enrichment experiments were conducted with a Narragansett Bay clone of Rhizosolenia fragilissima to examine the potential influence of nutrients in regulating its seasonal cycle in this embayment, and their contribution to its coastal tendencies. Growth rates were measured at 18 C and 1000 ft-c continuous illumination in surface waters enriched in 15 different ways. Narragansett Bay was sampled in March, May, September, and December. Six stations on a transect from this Bay to the Sargasso Sea were sampled during late summer. The data and our previous autecological observations are consistent with the idea that the annual cycle of this species in Narragansett Bay is associated with temperature, chemical “water quality,” and unknown factors. Its absence during late fall and winter may reflect low temperatures and a trace metal (Co, Mo) inadequacy. In early spring, low temperature appears to be the limiting factor, whereas in late spring and early summer trace metals again appear to prevent active growth. A late summer-early autumn bloom occurs periodically during optimal temperature conditions; this can terminate independently of grazing pressure, and in spite of seemingly adequate light-temperature-salinity and phosphate conditions.  相似文献

The seasonal dynamics of the diatom genus Chaetoceros Ehrenbergwere investigated in Narragansett Bay, Rhode Island. Whole watersamples were collected weekly from July 1981 to October 1982at four locations in the lower Bay and were preserved, concentratedby settling and enumerated. Chaetoceros were most abundant inlate winter–early spring; a second peak occurred in thefall. As in many other temperate neritic areas, C. debilis,C. compressus and C. didymus were major components of die Chaetocerospopulation and could be found throughout the year. Species compositionand dynamics were quite similar at the Jerusalem, GSO Pier andWickford Harbor stations, but were markedly different in PettaquamscuttRiver, a reduced salinity tidal estuary.  相似文献

Summary Sedimentation of particulate matter measured at a depth of 100 m varied from 0.2 to 6.0 mg dry weight m-2 d-1 over approximately monthly intervals between September 1986 and June 1987 on the continental shelf of the western Arctic Ocean north of the Canadian Arctic Islands. Detritus deposited during December and January, the period of maximum sedimentation, contained relatively low amounts of organic carbon and nitrogen (4.0% and 0.5% of dry weight, respectively) with a carbon: nitrogen ratio > 7. Maximum rates of organic carbon and nitrogen sedimentation (0.60 and 0.12 mg m-2 d-1, respectively) during February were associated with debris enriched with organic matter (organic carbon and nitrogen content of 17.0% and 3.4% of dry weight, C:N=5). Dry weight of macrozooplankton, predominantly Calanus hyperboreus and Metridia longa, separated from preserved trap contents, equalled or exceeded by up to an order-of-magnitude the weight of sedimented debris. Lipid content of CV and adult female stages of C. hyperboreus decreased from 46%–64% of dry weight in September to 20%–30% in January. Females of C. hyperboreus with lower lipid levels (7% of dry weight) between April and June were probably in a post-reproductive condition.  相似文献

The ctenophore Mnemiopsis leidyi is at the northern extreme of its geographic range in Narragansett Bay, an estuary on the northeast coast of the United States. Blooms have typically been observed in late summer and fall according to records from 1950 to 1979. We document an expansion of the seasonal range of this important planktonic predator to include springtime blooms during the 1980s and 1990s. This shift to an earlier seasonal maximum is associated with increasing water temperature in Narragansett Bay. Temperatures in spring have risen, on average, 2 °C from 1950 to 1999 with warm years being associated with the positive phase of the North Atlantic Oscillation. During 1999, M. leidyi appeared earlier in spring and was more abundant than during any previous year for which records are available. Changes in the seasonal pattern and abundance of this predator are likely to have important effects on planktonic ecosystem dynamics of Narragansett Bay. These include reduction of zooplankton abundance in spring followed by increases in size and frequency of summer phytoplankton blooms. Earlier blooms of M. leidyi may also reduce survival of eggs and larvae of fish because, as in 1999, they coincide with the period of peak spawning.  相似文献

During ongoing DNA barcode (COI‐5P) surveys of the macroalgal flora along the northwest Atlantic coast, we discovered a population of Ceramium secundatum in Narragansett Bay, Rhode Island, USA. This species is regarded as common and widespread in the northeast Atlantic, ranging from Norway to Morocco, but until now has not been reported from the western Atlantic. Several lines of evidence suggest that C. secundatum may be introduced to Narragansett Bay: (1) despite extensive collecting, specimens have only been obtained from a limited geographic range in the northwest Atlantic; (2) three other nonindigenous seaweed species are reportedly introduced in this region, which is thought to be a consequence of shipping; and (3) this species is introduced to South Africa and New Zealand. To investigate this suspected introduction, we applied population genetic analyses (using the cox2‐3 spacer) to compare the Narragansett Bay C. secundatum population to native populations in the Republic of Ireland and the United Kingdom. Collectively, analyses of biogeographical and molecular data indicate that C. secundatum is likely introduced to Narragansett Bay. The implications of this discovery are discussed.  相似文献

Within the framework of an investigation into the carrying capacity of the Bay of Marennes-Oléron (France) for bivalve culture, thein situ uptake of suspended particulate material by oysters (Crassostrea gigas) and mussels (Mytilus edulis) was determined in experiments with benthic ecosystem tunnels. Very high fluctuations in seston quantity and quality were observed within and between tidal cycles. The percentage of organic carbon was inversely related to seston quantity at low concentrations. Organic carbon was diluted by resuspension of material rich in inorganic matter. At high seston concentrations a constant level of about 2% organic carbon was found. The C/N ratio was relatively constant throughout the seasons and fairly low (6.5 to 8.4). Owing to the presence of bivalves large fluxes of suspended particulate material were observed in thein situ measurements. Selective retention of organic carbon or nitrogen could not be demonstrated. Clearance rates based on chlorophyll uptake were within a normal range compared to other studies. A large contribution to the food of the bivalves seemed to be formed by resuspended microphytobenthos. Judged by the low C/N ratio, the food was of good quality. Although its quantity was variable by dilution, it may support largely the carrying capacity of the Bay of Marennes-Oléron for the cultivation of bivalves. Thirty-nine subgeneric taxa of macroalgae have been collected from 83 sites in Georgian Bay, the North Channel and their drainage basin. There were 15 species in the Bay and Channel and 32 species in streams, rivers and impoundments in the basin. Only 8 of the Bay and Channel species were also found in the watershed. Cladophora glomerata was the most important species in Georgian Bay and the North Channel, having an estimated 640 × 103 m2 cover and 19 × 104 kg fresh weight standing crop. However, this species was largely concentrated on the southwestern shorelines of these water bodies. Its distribution along the northeastern shoreline appears to be limited by total ion and phosphorus levels. Chara globularis/vulgaris was the subdominant taxon in Georgian Bay and the North Channel with an estimated 70 × 103 m2 cover and 15 × 103 kg fresh weight biomass. This species was more widely distributed than C. glomerata. No other taxon contributed significantly to the standing crop including the frequently occurring Ulothrix zonata, Zygnema spp. and Spirogyra spp. The maximum benthic macroalgal biomass was estimated to be approximately 10% of the phytoplankton biomass.  相似文献

The influence of 116 combinations of temperature (2, 7, 12, 16 C), salinity (5–35‰ at 5‰ intervals) and light (5 levels) on the mean daily cell division rate ( K ) of the Narragansett Bay clone of Detonula confervacea was examined following appropriate preconditioning. Growth did not occur at 16 C, but was excellent (K = 1.2–1.5) under certain combinations of light and salinity at 2, 7, and 12 C, being somewhat better at the 2 highest temperature levels. At 32%, and 1100–1200 ft-c, K increased approximately 2.5 fold from 0.6 to 1.5 between 2 and 12 C. A light-temperature relationship was found which had the general trend of an increased optimal light intensity with increasing temperature. Within the optimal salinity range of 15–30‰, the optimal light intensity was 200–600 ft-c at 2 C, 600–1200 ft-c at 7 C, and 1200–1800 ft-c at 12 C. The light-temperature relationship was most pronounced at 2 and 12 C. At 2 C, K decreased with increasing light intensity, but was independent of this factor at higher temperatures. The optimal salinity range of 15–30‰ was independent of temperature negligible growth occurred at 5‰. In situ and in vitro responses of Detonula confervacea to salinity were in general agreement but its pronounced cryophilic preference in nature (usually reaching maximum abundance below 1 C) contrasts with its excellent growth at 12 C in culture. The experiments suggest that termination of the bloom of Detonula confervacea in Narragansett Bay and elsewhere is not solely temperature-dependent. Temperature does not satisfactorily account for its apparent exclusion from waters contiguous to Narragansett Bay and from other more northerly portions of the northeastern coast of the U.S, or, together with light, for its equally surprising apparent unimportance in Norwegian coastal waters.  相似文献

Meiofaunal prominence and benthic seasonality in a coastal marine ecosystem   总被引:13,自引:0,他引:13
Summary The muds of a shallow (7 m) site in Narragansett Bay, Rhode Island contained higher abundances of meiofauna (averaging 17×106 individuals per m2 and ash free dry weight of 2.9 g/m2 during a 3 year period) than have been found in any other sediment. The majority of sublittoral muds, worldwide, have been reported to contain about 106 individuals per m2. This difference is attributed primarily to differences in sampling techniques and laboratory processing.Extremely high meiofaunal abundances may have also occurred because Narragansett Bay sediments were a foodrich environment. While the quantity of organic deposition in the bay is not unusually high for coastal waters, this input, primarily composed of diatom detritus, may contain an unusually high proportion of labile organics. Furthermore, meiofauna could have thrived because of spatial segregation of meiofauna and macrofauna. While meiofauna were concentrated at the sediment-water interface, most macrofauna were subsurface deposit feeders. Macrofaunal competition with, and ingestion of meiofauna may thus have been minimized. The seasonal cycles of meiofauna and macrofauna were similar. Highest abundances and biomass were observed in May and June and lowest values in the late summer and fall. Springtime increases of meiofaunal abundance were observed in all depth horizons, to 10 cm. We hypothesize that phytoplankton detritus accumulated in the sediment during the winter and early spring, and that the benthos responded to this store of food when temperatures rose rapidly in the late spring. By late summer, the stored detritus was exhausted and the benthos declined.  相似文献

Narragansett Bay is a relatively well-mixed, high salinity coastal embayment and estuary complex in southern New England (USA). Much of the shoreline is urban and the watershed is densely developed. We have combined our data on C, N, and P inputs to this system, on C, N, and P accumulation in the sediments, and on denitrification with extensive work by others to develop approximate annual mass balances for these elements. The results show that primary production within the bay is the major source of organic carbon (4 times greater than other sources), that land drainage and upstream sewage and fertilizer are the major sources of N, and that landward flowing bottom water from offshore may be a major source of dissolved inorganic phosphorus. Most of the nutrients entering the bay arrive in dissolved inorganic form, though DON is a significant component of the N carried by the rivers. About 40% of the DIN in the rivers is in the form of ammonia. Sedimentation rates are low in most of Narragansett Bay, and it appears that less than 20% of the total annual input of each of these elements is retained within the system. A very small amount of C, N, and P is removed in fisheries landings, denitrification in the sediments removes perhaps 10–25% of the N input, and most of the carbon fixed in the system is respired within it. Stoichiometric calculations suggest that some 10–20% of the organic matter formed in the bay is exported to offshore and that Narragansett Bay is an autotrophic system. Most of the N and P that enters the bay is, however, exported to offshore waters in dissolved inorganic form. This assessment of the overall biogeochemical behavior of C, N, and P in the bay is consistent with more rigorously constrained mass balances obtained using large living models or mesocosms of the bay at the Marine Ecosystem Research Laboratory (MERL).  相似文献

Clones of Skeletonema costatum (Grev.) Cl. isolated from Narragansett Bay, R.I., during different seasons were grouped according to their electrophoretic banding patterns. The growth rates, pg chlorophyll · cell?1, carbon uptake · cell?1· h?1, and carbon uptake · pg chl?1· h?1 were measured at 20°C, in a 14:10 h L:D cycle at 180 μE · m?2· s?1. Statistically significant sources of variation were found among groups of clones in growth rate, pg chl · cell?1, and carbon uptake · pg chl?1· h?1. It was concluded that there is a significant relationship between the physiological characteristics of clones isolated from populations in different seasons and patterns of genetic variation inferred from the electrophoretic studies. However, genetic diversity detected by banding patterns tends to underestimate the total genetic diversity in natural populations. The groups of clones most common in summer bloom populations had significantly higher growth rates, lower values of pg chl · cell?1, and higher rates of carbon uptake · pg chl?1· h?1 at 20°C than did the group of clones most common in winter bloom populations. However, differences among groups in these parameters at 20°C alone cannot account for the seasonal cycling of genetically variable populations of Skeletonema in Narragansett Bay. The range of growth rates among clones of this species is 0.1–5.0 divisions · d?1 under a single set of temperature and light conditions. Chlorophyll concentrations range from 0.2–1.7 pg chl · cell?1 and carbon uptake · pg chl?1· h?1 varies by a factor of 7 among clones. The range of physiological variation in this species means that it is difficult to use laboratory studies of single clones to analyze the responses of natural populations of Skeletonema.

The effects of glutamine-based dipeptides, glutamine and casein hydrolysate, as well as the deletion of organic nitrogen, were investigated during white spruce [Picea glauca (Moench) Voss] somatic embryogenesis. There were no differences in the fresh weight increase of the tissue masses grown on initiation medium with different combinations of organic nitrogen. This was also the case for subsequent growth on kinetin medium, except that glutamine alone produced a significantly lower fresh weight increase than the other organic nitrogen combinations. Without organic (i.e. with only inorganic) nitrogen in the medium, the fresh weight increase was significantly less than with organic nitrogen on both initiation and kinetin medium. No differences were found between the dry/fresh weight ratios obtained with the various nitrogen treatments. The number of mature embryos produced per gram fresh weight when cultured in the absence of organic nitrogen was significantly higher than that obtained in its presence. There were no differences in the total number of mature embryos produced in cultures grown with various organic nitrogen combinations or without organic nitrogen. There were large clone differences with respect to the number of mature somatic embryos per gram tissue and the total number of somatic embryos produced. Hence, nitrogen type influences culture growth rate but not the number of mature somatic embryos produced. The latter was clone dependent.Abbreviations BA 6-benzylaminopurine - CH casein hydrolysate  相似文献

In South Africa, gracilarioid red algae have been collected as wash-up to be dried and sold for agar extraction for at least 50 years. Despite much research, there is currently no commercial mariculture of the algae locally although this has been carried out in neighboring Namibia for a number of years. The industry is traditionally confined to Saldanha Bay on the west coast, although small wash-ups of Gracilariopsis longissima have been collected in nearby St Helena Bay. In Saldanha Bay, wash-ups of Gracilaria. gracilis have been very sporadic over the last few decades, with human alteration of the bay configuration possibly responsible for an initial major decline in 1974. This unpredictability in the amounts of wash-up has made the industry unstable and increasingly unprofitable. We compiled the results of previous surveys (some unpublished) of gracilariod populations in St Helena Bay and the Saldanha-Langebaan sytem, and re-surveyed these populations to examine long-term fluctuations. In Saldanha Bay and Langebaan Lagoon, standing stock of G. gracilis was estimated at 538 tons fresh weight and 71 tons fresh weight respectively. Less than 4 tons of gracilarioids are estimated to remain in St Helena Bay. We discuss the fluctuations in biomass and distribution of these South African gracilarioid populations.  相似文献

The vernal occurrence of toxic dinoflagellates in the Alexandrium tamarense/Alexandrium fundyense species complex in an enclosed embayment of Narragansett Bay (Wickford Cove, Rhode Island) was documented during 2005 and 2009–2012. This is the first report of regular appearance of the Alexandrium fundyense/Alexandrium tamarense species complex in Narragansett Bay. Thecal plate analysis of clonal isolates using SEM revealed cells morphologically consistent with both Alexandrium tamarense Lebour (Balech) and Alexandrium fundyense Balech. Additionally, molecular analyses confirmed that the partial sequences for 18S through the D1–D2 region of 28S were consistent with the identity of the two Alexandrium species. Toxin analyses revealed the presence of a suite of toxins (C1/2, B1 (GTX-5), STX, GTX-2/3. Neo, and GTX-1/4) in both Alexandrium tamarense (6.31 fmol cell−1 STX equiv.) and Alexandrium fundyense (9.56 fmol cell−1 STX equiv.) isolated from Wickford Cove; the toxicity of a Narragansett Bay Alexandrium peruvianum isolate (1.79 fmol cell−1 STX equiv.) was also determined. Combined Alexandrium tamarense/Alexandrium fundyense abundance in Wickford Cove reached a peak abundance of 1280 cells L−1 (May of 2010), with the combined abundance routinely exceeding levels leading to shellfishing closures in other systems. The toxic Alexandrium tamarense/Alexandrium fundyense species complex appears to be a regular component of the lower Narragansett Bay phytoplankton community, either newly emergent or previously overlooked by extant monitoring programs.  相似文献

Macroalgal bloom‐forming species occur in coastal systems worldwide. However, due to overlapping morphologies in some taxa, accurate taxonomic assessment and classification of these species can be quite challenging. We investigated the molecular and morphological characteristics of 153 specimens of bloom‐forming Ulva located in and around Narragansett Bay, RI, USA. We analyzed sequences of the nuclear internal transcribed spacer 1 region (ITS1) and the chloroplast‐encoded rbcL; based on the ITS1 data, we grouped the specimens into nine operational taxonomic units (OTUs). Eight of these OTUs have been previously reported to exist, while one is novel. Of the eight OTUs, all shared sequence identity with previously published sequences or differed by less than 1.5% sequence divergence for two molecular markers. Previously, 10 species names were reported for Ulva in Rhode Island (one blade and nine tube‐forming species) based upon morphological classification alone. Of our nine OTUs, three contained blade‐forming specimens (U. lactuca, U. compressa, U. rigida), one OTU had a blade with a tubular stipe, and six contained unbranched and/or branched tubular morphologies (one of these six, U. compressa, had both a blade and a tube morphology). While the three blade‐forming OTUs in Narragansett Bay can frequently be distinguished by careful observations of morphological characteristics, and spatial/temporal distribution, it is much more difficult to distinguish among the tube‐forming specimens based upon morphology or distribution alone. Our data support the molecular species concept for Ulva, and indicate that molecular‐based classifications of Ulva species are critical for proper species identification, and subsequent ecological assessment or mitigation of Ulva blooms.  相似文献

Low molecular weight organic acids (LMWOAs) derived from root exudates, decomposing organic matter, and other sources are important ligands. The species of these LMWOAs in the Tsuga rhizosphere soil (TRS), and Yushania rhizosphere soil (YRS), and bulk soil (BS) from an alpine forest region were identified. LMWOA and organic functional groups were used to those fresh twigs and leaves, litters, and roots as comparison. The objectives of this study were to (i) develop a method that could be used to determine LMWOAs in soil solution by gas chromatography (GC), (ii) assess methods for processing LMWOAs in soil samples, and (iii) determine the relative proportions of organic carbon functional groups in the TRS, YRS and BS, and fresh plant materials with13C nuclear magnetic resonance (13C NMR) analysis. The proportion of organic acid contents followed the order of YRS > TRS > BS, and also showed significant differences (P < 0.05) from GC analysis. The amounts of malonic, fumaric and succinic acids in the YRS samples were greater than in the TRS and BS. Samples analyzed after 1 month of deep freeze storage (–24°C) showed no signs of decomposition. The proportion of organic functional groups in the rhizosphere and bulk soils quantified by 13C NMR analyses followed the general order: alkyl-C > O-alkyl-C > N-alkyl-C > acetal-C > aromatic-C > carboxylic-C > phenolic-C.  相似文献
