The staphylinid beetle, Oligota pygmaea (Solier) is an important predator of the red spider mite, Oligonychus coffeae (Nietner) infesting tea. Biology, life table and predatory efficiency of O. pygmaea were studied under laboratory conditions. Duration of developmental stages of O. pygmaea was 3.2, 5.7 and 12.5 d for eggs, larvae and pupae, respectively with an average of 23.0 d from egg to adult emergence. After a mean preovipostion period of 2.9 d, each female laid an average of 400.5 eggs in its life span. Adult O. pygmaea lived for an average of 54.1 d. Adult females lived for a longer period of 58.8 d compared to the longevity of 49.4 d of adult male. Studies revealed that its life table characterized by an intrinsic rate of natural population increase (r) of 0.118 d, net reproductive rates (Ro) of 243.693 eggs/female, gross reproduction rate (Σmx) of 245.313 eggs/female, generation time (T) of 46.575 d, doubling time (DT) of 5.874 d and finite rate of increase (λ) of 1.125 d. Seasonal abundance of O. pygmaea and its prey, O. coffeae was monitored by sampling 25 tea leaves randomly from each experimental block grown under the prevailing field conditions. O. pygmaea showed a typical pattern of population dynamics with a peak during January to March and low incidence during June to September. Peak in the population of O. pygmaea coincided with the abundance of O. coffeae in the tea fields. Weather factors such as high temperature, low relative humidity and low sunshine hours adversely affected the populations of O. pygmaea. The first to third instar larvae of O. pygmaea consumed 31.0–133.2 eggs of mites per day. Third instar larva of O. pygmaea consumed an average of 133.2 eggs, 46.4 hexapod larvae, 39.6 nymphs and 11.4 adults per day. The cells of the red microalga Porphyridium UTEX 637 are encapsulated within a sulfated polysaccharide whose external part (i.e., the soluble fraction) dissolves into the medium. It is thought that the main function of the polysaccharide is to protect the algal cells from the extreme environmental conditions, such as drought and high light, prevailing in their native sea-sand habitat. In this study, we evaluated the antioxidant properties of the water-soluble polysaccharide of Porphyridium sp. by determining the ability of a polysaccharide solution to inhibit: (1) autooxidation of linoleic acid, as determined by the standard thiobarbituric acid (TBA) and ferrous oxidation (FOX) assays; and (2) oxidative damage to 3T3 cells as determined by the dichlorofluorescein (DCFH) assay. In all three assays, the polysaccharide inhibited oxidative damage in a dose-dependent manner. Antioxidant activity was also exhibited by fractions of the polysaccharide obtained by sonication followed by separation on a reverse-phase HPLC with a C8 semi-preparative column. It is suggested that the antioxidant activity of the sulfated polysaccharide protects the alga against reactive oxygen species produced under high solar irradiation, possibly by scavenging the free radicals produced in the cell under stress conditions and transporting them from the cell to the medium.  相似文献

The antimicrobial activity of the dichloromethane extract from aerial parts of Leuzea carthamoides DC. was tested in vitro against 19 Staphylococcus aureus strains (ATCC 25923, CNCTC Mau 43/60, clinical isolates). The extract was fractionated by column chromatography on silica gel into six fractions (petroleum ether, toluene, dichloromethane, ethyl acetate, methanol and water). The minimum inhibitory concentrations (MICs) of the fractions ranged from 64 to 1024 μg/mL. An ethyl acetate fraction (EA 1) with the widest range of activity inhibited all of the strains with MIC in the range 128–512 μg/mL. This fraction exhibited potent activity against strains which showed associated resistance to oxacillin, ciprofloxacin and erythromycin.  相似文献

基于2008—2016年青海海北站9年净初级生产力及气候因子监测数据,分析了青藏高原高寒小嵩草草甸和高寒金露梅灌丛两种植被净初级生产力年际动态,并探讨了气候因子对其影响及其不同土层深度根系周转值特征。结果表明:(1)年际尺度上,小嵩草草甸地上净初级生产力表现为显著增加趋势,增幅为7.02 g m~(-2) a~(-1),而金露梅灌丛地上净初级生产力相对较为稳定;对于其地下净初级生产力和总生产力,小嵩草草甸和金露梅灌丛均表现为增加趋势(P0.05),9年间小嵩草草甸地上、地下和总净初级生产力平均值分别为(217.55±9.95)、(1882.75±161.33) g m~(-2) a~(-1)和(2100.30±163.38) g m~(-2) a~(-1),金露梅灌丛地上、地下和总净初级生产力9年间平均值分别为(256.27±11.4)、(1614.31±173.03) g m~(-2) a~(-1)和(1870.58±177.93) g m~(-2) a~(-1)。(2)不同植被类型地上净初级生产力对气候因素响应不同,金露梅灌丛地上净初级生产力主要受温度影响,而温度对小嵩草草甸地上净初级生产力无显著影响。此外,降水不是限制高寒生态系统草地地上净初级生产力主要因子,相比于降水影响,高寒生态系统地上净初级生产力更受温度调控。(3)年均温和年降水对金露梅灌丛和小嵩草草甸地下净初级生产力均无显著影响(P0.05),表明高寒生态系统,其地下生产力受外界气候条件变化影响微弱,是一个稳定的碳库。(4)两种植被类型其根系周转值均随着土壤深度的增加呈逐渐增加趋势,且高寒灌丛根系周转值明显高于高寒草甸根系周转值。研究表明,在全球气候变暖背景下将会增加金露梅灌丛地上净初级生产力,而对小嵩草草甸地上净初级生产力无显著影响。  相似文献

Purpose of the present study was to evaluate antioxidant, antibacterial, antifungal, and antiviral activities of the petroleum ether, chloroform, ethyl acetate and methanol extracts as well as the alkaloid fraction of Lycopodium clavatum L. (LC) from Lycopodiaceae growing in Turkey. Antioxidant activity of the LC extracts was evaluated by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging method at 0.2 mg/ml using microplate-reader assay. Antiviral assessment of LC extracts was evaluated towards the DNA virus Herpes simplex (HSV) and the RNA virus Parainfluenza (PI-3) using Madin-Darby Bovine Kidney (MDBK) and Vero cell lines. Antibacterial and antifungal activities of the extracts were tested against standard and isolated strains of the following bacteria; Escherichia coli, Pseudomonas aeruginosa, Proteus mirabilis, Acinobacter baumannii, Klebsiella pneumoniae, Staphylococcus aureus, Bacillus subtilis as well as the fungi; Candida albicans and C. parapsilosis. All of the extracts possessed noteworthy activity against ATCC strain of S. aureus (4 μg/ml), while the LC extracts showed reasonable antifungal effect. On the other hand, we found that only the chloroform extract was active against HSV (16–8 μg/ml), while petroleum ether and alkaloid extracts inhibited potently PI-3 (16–4 μg/ml and 32–4 μg/ml, respectively). However, all of the extracts had insignificant antiradical effect on DPPH. In addition, we also analyzed the content of the alkaloid fraction of the plant by capillary gas chromatography-mass spectrometry (GC-MS) and identified lycopodine as the major alkaloid.  相似文献

According to Traditional Chinese Medicine, Alzheimer's disease (AD) is regarded as senile dementia, and the etiopathogenesis lies in kidney deficiency during aging. Dipsacus asper Wall (DAW), a well-known traditional Chinese medicine for enhancing kidney activity, may possess the therapeutic effects against AD. Our objectives were to investigate the protective effects of DAW against the amyloid-β peptide (Aβ)-induced cytotoxicity and explore its major active components. Injury of PC 12 cells mediated by Aβ25–35 was adopted to assess the cytoprotective effects of DAW aqueous extract and various fractions. Salvianolic acid B, a polyphenol compound isolated from Salvia miltiorrhiza, was employed as a positive control agent due to its markedly protective effect against neurotoxicity of amyloid β. Five chemical fractions (i.e. alkaloids, essential oil, saponins, iridoid glucoside and polysaccharides) were prepared for activity test and analyzed by HPLC for active components identification. In addition, Akebia saponin D (the most important compound in DAW saponins) and hederagenin (the mother nucleus of akebia saponin D) were prepared for testing of their activity. DAW water extract, saponins fraction and akebia saponin D had the neuroprotective capacity to antagonize Aβ25–35-induced cytotoxicity in PC 12 cells. In contrast, other fractions and hederagenin had no cytoprotective action. This research suggests that DAW may represent a potential treatment strategy for AD and akebia saponin D is one of its active components.  相似文献

Antioxidant, antidiabetes, and anticancer activities of fractions (water, hexane, EtOAc, and BuOH) from methanolic (MeOH) extract of Picrasma quassioides were evaluated in vitro. Among the four fractions, BuOH fraction showed cytotoxic activity on the human stomach carcinoma cells (NCI-N87) without cytotoxic effect on the human normal kidney cells (293) by MTT assay. Considering these results, we adapted BuOH fraction of MeOH extract to the assessment of apoptosis by flow cytometry and mRNA expression levels of established apoptotic-related genes on NCI-N87 cells. The experiments showed that BuOH fraction could induce apoptosis on NCI-N87 cells significantly. Take together these results, P. quassioides showed that it could be used for potential pharmaceutical products.  相似文献

In this work, two biosurfactant-producing strains, Bacillus subtilis and Bacillus licheniformis, have been characterized. Both strains were able to grow at high salinity conditions and produce biosurfactants up to 10% NaCl. Both extracted-enriched biosurfactants showed good surface tension reduction of water, from 72 to 26–30 mN/m, low critical micelle concentration, and high resistance to pH and salinity. The potential of the two lipopeptide biosurfactants at inhibiting biofilm adhesion of pathogenic bacteria was demonstrated by using the MBEC device. The two biosurfactants showed interesting specific anti-adhesion activity being able to inhibit selectively biofilm formation of two pathogenic strains. In particular, Escherichia coli CFT073 and Staphylococcus aureus ATCC 29213 biofilm formation was decreased of 97% and 90%, respectively. The V9T14 biosurfactant active on the Gram-negative strain was ineffective against the Gram-positive and the opposite for the V19T21. This activity was observed either by coating the polystyrene surface or by adding the biosurfactant to the inoculum. Two fractions from each purified biosurfactant, obtained by flash chromatography, fractions (I) and (II), showed that fraction (II), belonging to fengycin-like family, was responsible for the anti-adhesion activity against biofilm of both strains.  相似文献

Scavenger enzyme activities in subcellular fractions under polyethylene glycol (PEG)-induced water stress in white clover (Trifolium repens L.) were studied. Water stress decreased ascorbic acid (AA) content and catalase (CAT) activity and increased the contents of hydrogen peroxide (H2O2), thiobarbituric acid reactive substances (TBARS) (measure of lipid peroxidation), and activities of superoxide dismutase (SOD), its various isozymes, ascorbate peroxidase (APOX), and glutathione reductase (GR) in cellular cytosol, chloroplasts, mitochondria, and peroxisomes of Trifolium repens leaves. In both the PEG-treated plants and the control, chloroplastic fractions showed the highest total SOD, APOX, and GR activities, followed by mitochondrial fractions in the case of total SOD and GR activities, whereas cytosolic fractions had the second greatest APOX activity. However, CAT activity was the highest in peroxisomes, followed by the cytosol, mitochondria, and chloroplasts in decreasing order. Although Mn-SOD activity was highest in mitochondrial fractions, residual activity was also observed in cytosolic fractions. Cu/Zn-SOD and Fe-SOD were observed in all subcellular fractions; however, the activities were the highest in chloroplastic fractions for both isoforms. Total Cu/Zn-SOD activity, the sum of activities observed in all fractions, was higher than other SOD isoforms. These results suggest that cytosolic and chloroplastic APOX, chloroplastic and mitochondrial GR, mitochondrial Mn-SOD, cytosolic and chloroplastic Cu/Zn-SOD, and chloroplastic Fe-SOD are the major scavenger enzymes, whereas cellular CAT may play a minor role in scavenging of O2 and H2O2 produced under PEG-induced water stress in Trifolium repens.  相似文献

The Chilean annual,Microseris pygmaea, has differentiated in distinct coastal and inland series of populations after long-distance dispersal from western North America. Two plants from the most diverse biotypes were crossed, a large F2 was raised and analysed for segregation of 30 phenotypic characters. Segregation of molecular markers (47 RAPDs, 1 RFLP, 2 isozymes) was determined in a subpopulation of 45 plants which include all extremes for the phenotypic characters. 32 marker/character cosegregations were significant at the 1% level in t-tests between dominant and homozygous recessive marker genotypes. Considering linkage among markers and pleiotropy of certain marker loci, the number of independent quantitative trait loci (QTLs) is reduced to about 18. Interactions among 2 or 3 QTLs affecting one character have been characterized. The phenotypic differentiation ofM. pygmaea during its evolution from a single founder individual begins to be understood at the level of single-gene mutants.  相似文献

The reduction of inner (adaxial) pollen sacs (microsporangia, MS) as a diagnostic character for the three asteracean species, Microseris bigelovii, Microseris elegans and Microseris pygmaea, was analysed in an interspecific cross between Microseris douglasii and Microseris bigelovii with 4 MS and 2 MS, respectively, using the average number of MS per plant as a quantitative character. A previous QTL (Quantitative Trait Locus) analysis had revealed one major QTL (3B) and three modifier QTLs (3A, 4A, 7A) with epistatic effects only on the homozygous recessive 2 MS genotype of QTL 3B. Here we performed a bulked segregant analysis on four 2 MS and four 4 MS DNA-bulks with 407 EcoRI/MseI AFLP-primer combinations each. In this way additional AFLP markers were mapped close to QTL 3B and QTL 3A. Three of them were converted to SCAR (Sequence Characterized Amplified region) markers. All markers were tested in natural populations of the disporangiate (2 MS) species M. bigelovii, M. elegans and M. pygmaea, and in different populations of tetrasporangiate (4 MS) M. douglasii. The marker distribution suggests that locus 3B mutated in a progenitor of the disporangiate species. QTL 3A has evolved in the 2 MS background of the major gene in the disporangiate species. Since M. pygmaea and M. bigelovii are the sister group to M. elegans, the 4 MS genotype for (markers of) QTL 3A in M. pygmaea populations is most likely due to a back mutation to the 4 MS state and could explain the slight instability of the 2 MS phenotype in this species.Communicated by O. Two small cationic peptide fractions (5 kDa) were isolated from dry and germinated seeds of wheat, named WAP and GWAP, respectively. The antifungal and antibacterial activities of the peptides were analyzed using disk diffusion and turbidity measurement assays. The peptides in vitro exhibited effective antifungal activity against four plant pathogenic fungi at minimum concentration of 15 g(protein) cm–3. Their antimicrobial activity was negatively affected by the presence of 5 mM CaCl2. The peptides were less effective against Gram-negative bacterium Erwinia carotovora subsp. carotovora, but they demonstrated inhibitory activity against Gram-positive bacterium Clavibacter michiganensis subsp. sepedonicus. The antimicrobial activity of GWAP was more effective than WAP.  相似文献

The genome sequence of Streptomyces coelicolor A3(2) contains 51 putative lipase and esterase genes mostly of unknown function. The gene estB (locus SCO 6966) was expressed as a His-tagged protein in E. coli. Esterase B was active at low temperatures exerting its maximum activity at 30°C and retaining more than 25% of its activity at 4°C. The optimum pH was 8–8.5. The enzyme was active against short synthetic p-nitrophenylesters (C2–C10) with maximum activity towards the acetate ester (C2). The esterase was tested on 13 series of racemic esters of potential interest for the synthesis of chiral pharmaceutical compounds. 4 of the series were substrates and a modest degree of enantioselectivity was observed (enantiomeric ratios of 1.1–1.9).  相似文献

The toxicity of para-menthane-3,8-diol (PMD), the main arthropod-repellent compound in the oil of the lemon eucalyptus, Corymbia citriodora, was evaluated against nymphs of Ixodes ricinus using five methods (A–E) of a contact toxicity bioassay. Mortality rates were estimated by recording numbers of dead nymphs at 30 min intervals during the first 5 h after the start of exposure and at longer intervals thereafter. The mortality rate increased with increasing concentration of PMD and duration of exposure with a distinct effect after 3.5 h. From the results obtained by methods A, C and E, the LC50 range was 0.035–0.037 mg PMD/cm2 and the LC95 range was 0.095–0.097 mg PMD/cm2 at 4 h of exposure; the LT50 range was 2.1–2.8 h and the LT95 range was 3.9–4.2 h at 0.1 mg PMD/cm2. To determine the duration of toxic activity of PMD, different concentrations (0.002, 0.01, 0.1 mg PMD/cm2) were tested and mortality was recorded at each concentration after 1 h; thereafter new ticks were tested. This test revealed that the lethal activity of PMD remained for 24 h but appeared absent after 48 h. The overall results show that PMD is toxic to nymphs of I. ricinus and may be useful for tick control.  相似文献

The work was conducted with the purpose to evaluate antioxidant activity of Parmelia saxatilis (PS) by different analytical methods. Water and methanol were used as solvents and antioxidative effects were measured by a ferric thiocyanate method (FTC) and thiobarbituric acid test (TBA). The antioxidant activity increased with the increasing amount of extracts (from 50 to 250 μg) added to linoleic acid emulsion. The methanol extract of PS exhibited high antioxidative activity that was not significantly (P < 0.05) different from α-tocopherol, while aqueous extracts of PS showed low antioxidative activity. Similar trends of antioxidant activity were observed using either the FTC or TBA methods. Antioxidant activity, reducing power, free radical scavenging (DPPH·), superoxide anion radical scavenging, metal chelating and hydrogen peroxide scavenging activities of PS extracts showed dose dependence and increased with concentration of PS extract. The results obtained in the present study indicate that the PS might be a potential source of natural antioxidant.  相似文献

We have been attempting for some time to discover a compound evidencing antibacterial activity against methicillin-resistant Staphylococcus aureus (MRSA). The dieckol isolated from Ecklonia stolonifera has been shown to exhibit antibacterial activity against methicillin-susceptible S. aureus (MSSA) and MRSA. The minimum inhibitory concentrations (MICs) of dieckol were determined in a range of 32 to 64 μg/mL against standard MSSA and MRSA strains. Furthermore, dieckol clearly reversed the high-level ampicillin and penicillin resistance of MRSA. The MICs of ampicillin against two standard strains of MRSA were dramatically reduced from 512 to 0.5 μg/mL in combination with 1/4 MIC of dieckol (16 μg/mL). The fractional inhibitory concentration (FIC) indices of ampicillin and penicillin were measured from 0.066 to 0.266 in combination with 8 or 16 μg/mL of dieckol against all tested MRSA strains, thereby suggesting that dieckol-ampicillin or dieckol-penicillin combinations exert a synergistic effect against MRSA. The results of this study indicate that dieckol, administered in combination with β-lactams, may prove effective in the treatment of MRSA infections. Our finding may also contribute to the development of an alternative phytotherapeutic anti-MRSA agent.  相似文献

Oligosaccharides were prepared through mild hydrochloric acid hydrolysis of kappa-carrageenan from Kappaphycus striatum to compare the antitumor activity with carrageenan polysaccharides. Oligosaccharide fractions were isolated by gel permeation chromatography and the structure of fraction 1 (F1) was studied by using negative-ion electrospray ionization-mass spectrometry (ESI-MS), and 1H and 13C-NMR spectrometry. The in vitro antitumor effects in three human neoplastic cell lines (KB, BGC, and Hela) of polysaccharides and F1 were investigated. The bioassay results showed that F1 exhibited relatively higher antitumor activity against the three cancer cells than polysaccharides.  相似文献

Resin glycosides are secondary metabolites exclusive to the convolvulaceous plants. In this study, crypthophilic acids A–C (13), the first resin glycosides occurring in another family (Scrophulariaceae), and the other constituents of Scrophularia cryptophila were examined for in vitro antiprotozoal and antimycobacterial potentials. Except for crypthophilic acid B (2), all tested compounds exhibited growth-inhibitory effect against Trypanosoma brucei rhodesiense, with l-tryptophan (6) and buddlejasaponin III (7) being the most potent ones (IC50's 4.1 and 9.7 μg/ml). In contrast, the activity towards Trypanosoma cruzi was poor, and only crypthophilic acid C (3), 6 and 7 were trypanocidal at concentrations above 40 μg/ml. With the exception of 2 and 6, all compounds were active against Leishmania donovani. Harpagide (4) and 3 emerged as the best leishmanicidal agents (IC50's 2.0 and 5.8 μg/ml). Only compounds 3, 6 and 7 showed antimalarial activity against Plasmodium falciparum with IC50 values of 4.2, 16.6 and 22.4 μg/ml. Overall the best and broadest spectrum activity was presented by compounds 3 and 7, as they inhibited all four parasitic protozoa. None of the isolates had significant activity against Mycobacterium tuberculosis (MICs >100 μg/ml) or were toxic towards mammalian (L6) cells. This is the first report of antiprotozoal activity for natural resin glycosides, as well as for harpagide (4), acetylharpagide (5), tryptophan (6) and buddlejasaponin III (7).  相似文献

Protease inhibitors present in seeds of legumes possess strong inhibitory activity against trypsin and confer resistance against pests. In the present investigation, trypsin inhibitor activity was found in the seed flour extracts of all the eight selected varieties of mungbean under study which was further confirmed by dot blot analysis. All the varieties showed inhibitory activity in vitro against the gut protease of Helicoverpa armigera (HGP). Trypsin inhibitor was purified from mungbean seeds to near homogeneity with 58.1-fold and 22.8% recovery using heat denaturation, NH4(SO4)2 fractionation, ion-exchange chromatography on DEAE-Sephadex A-25 and gel filtration through Sephadex G-75. The molecular mass of the inhibitor was 47 kDa as determined by gel filtration and SDS-PAGE. The inhibitor retained 90% or more activity between pH 4 and 10, however, it was nearly inactive at extreme pH values. The inhibitor was stable up to 80°C but thereafter, the activity decreased gradually retaining nearly 30% of activity when heated at 100°C for 20 min. The inhibitor activity was undetectable at 121°C. Insect bioassay experiment using purified mungbean trypsin inhibitor showed a marked decline in survival (%) of larvae with increase in inhibitor concentration. The larval growth was also extended by the trypsin inhibitor. This study signifies the insecticidal potential of mungbean trypsin inhibitor which might be exploited for raising transgenic plants.  相似文献
