Dairy propionibacteria are essential starters for Emmental cheese manufacture. The behavior of three commercial strains of Propionibacterium freudenreichii subsp. shermanii (P.f. 1, P.f. 2 and P.f 3) were studied in a liquid medium under air and N2 atmosphere using an on‐line pressure measurement technique. Growth kinetics and metabolite production were characterized under conditions usually reported as “optimal conditions” (pH 6.5, NaCl 0 %, temperature 30 °C) and also evaluated under “stressful conditions” (pH 5.2, NaCl 2 %, temperature 20 °C) simulating the cheese ripening conditions. In both cases, the effects of oxygen on growth were strain‐dependent. Under “stressful conditions”, two of the three strains were inhibited by oxygen under conditions of air atmosphere, while all three strains grew under conditions of N2 atmosphere. In the latter case, the duration of the lag phase and the maximum rate of pressure variation were significantly different, however, no significant differences were found between the strains with regard to the total fermentation time. Under “optimal conditions” metabolite production was strain‐dependent. In an air atmosphere, all strains produced more acetate and CO2 and less propionate than in a nitrogen atmosphere.  相似文献

The conservation of food products within a controlled atmosphere is efficient in packaging. To extend the cold storage of raw milk, the effects of five gas atmospheres enriched with carbon dioxide and nitrogen were investigated. Treated and control milk were stored at 7 °C for 10 days and analyzed for microbial counts, pH, proteolysis and lipolysis. The addition of CO2, N2, or their mixture had a significant inhibitory effect on psychrotrophic growth. The generation times of these microorganisms were significantly longer in treated milk, particularly for yeasts where they amounted to 16.63 h. The maximum inhibition was observed when a gas mixture of 50 % CO2 and 50 % N2 was used. As a result, psychrotrophic growth was affected to 98 % whereas this inhibition did not exceed 78 % when CO2 and 41 % N2 were applied. Milk treatment under the conditions of 50 % CO2 and 50 % N2 gave significantly lower counts for all groups of psychrotrophs being more efficient against Enterobacteriaceae with 99.5 % of inhibition. Storage of raw milk under the tested atmospheres had a different effect on extracellular enzyme productions. Significant decreases in protease and lipase activities were observed during the storage at 7 °C. These enzyme activities were not detectable with pure CO2 and a 50 % CO2 and 50 % N2 mixture. N2 has shown to be the less efficient treatment against lipases (65 %) and proteases (95 %). With regard to growth, the course of the pH and the protease and lipase activities, the tested gas mixture of 50 % CO2 and 50 % N2 was more suitable for extending the shelf life of raw milk.  相似文献

Summary The relationships between root exudation, root disease and anaerobic root stresses were investigated. Sand culture and mist chamber studies demonstrated that low O2 and high CO2 reduced plant growth and increased the exudation of ethanol, amino acids, and sugars by pea roots. The relative loss of ethanol by roots was much greater in treatments with atmospheres of N2 containing 30% CO2 than in treatments of air containing 30% CO2 or N2. Ethanol was not detected in the nutrient solution of aerated plant roots. Atmospheres of N2 plus 30% CO2 caused 500% greater mycelial growth ofFusarium solani f. sp.pisi and 400% more disease of inoculated pea roots. Relative losses of four amino acids and four sugars were much greater in atmospheres of N2 plus 30% CO2 than in N2 or air.  相似文献

From estuarine mud a rod-shaped, motile, gram-negative, anaerobic bacterium was isolated (strain asp 66). Asp 66 fermented several substrates including glucose, fructose, malate, fumarate, citrate and aspartate. Fermentation products were acetate, propionate and presumably CO2. Hydrogen was never formed nor utilized. Succinate conversion to propionate was catalyzed by cell suspensions but did not support growth. Asp 66 did not require vitamins and grew well in mineral media with a fermentable substrate. The pH range for growth was from 6.5 to 8.5. Temperature optimum was 27 to 30°C. The strain was able to fix N2 as evidenced by its growth with N2 as sole nitrogen source and its ability to reduce acetylene to ethylene. Cell-free extracts of cultures grown under air without shaking contained cytochrome(s) with absorption peaks at 523 nm and at 553 nm. The G+C content of the DNA was 60.8+-1 mol%. A fermentation process to increase type capsular polysaccharide production by different serotypes of Streptococcus agalactiae (group B Streptococcus) was established. As sialic acid is an integral component of the polysaccharide, its synthesis was used to monitor polysaccharide, its synthesis was used to monitor polysaccharide production. Culture conditions, examined both on laboratory and pilot-plant scales, allowed optimal bacterial growth and high polysaccharide production in a medium composed of ultrafiltered Todd Hewitt broth supplemented with 2% (w/v) glucose and 1.5% (w/v) Na2HPO4, at a constant pH of 7.2. Aims: The objective was to study the growth of Pseudomonas in a food product (tofu) where it typically occurs as a spoilage organism, and when this product is stored under modified atmosphere. Methods and Results: A Pseudomonas strain was isolated from the endogenous microflora of tofu. Tofu was inoculated with the strain, packaged in different gas conditions (air, 100% N2, 30% CO2/70% N2 or 100% CO2) and stored under refrigerated conditions. Microbial loads and the headspace gas composition were monitored during storage. Conclusions: The strain was capable of growing in atmospheres containing no or limited amounts of oxygen and increased amounts of carbon dioxide. Even when 100% CO2 was used, growth could not be inhibited completely. Significance and Impact of Study: In contrast to the general characteristics of the genus Pseudomonas (strictly aerobic, highly sensitive to CO2), it should not be expected in the food industry that removing oxygen from the food package and increasing the carbon dioxide content, combined with cold storage, will easily avoid spoilage by Pseudomonas species. Guarantee of hygienic standards and combination of strategies with other microbial growth inhibiting measures should be implemented.  相似文献

The role of carbon dioxide in glucose metabolism of Bacteroides fragilis   总被引:2,自引:0,他引:2  
The effect of CO2 concentration on growth and glucose fermentation of Bacteroides fragilis was studied in a defined mineral medium. Batch culture experiments were done in closed tubes containing CO2 concentrations ranging from 10% to 100% (with appropriate amounts of bicarbonate added to maintain the pH at 6.7). These experiments revealed that CO2 had no influence on growth rate or cell yield when the CO2 concentration was above 30% CO2 (minimum available CO2–HCO 3 - , 25.5 mM), whereas a slight decrease in these parameters was observed at 20% and 10% CO2 (available CO2–HCO 3 - , 17 and 8.5 mM, respectively). If CO2–HCO 3 - concentrations were below 10 mM, the lag phase lengthened and a decrease in maximal growth rate and cell yield were observed. The amount of acetate made decreased, while d-lactate concentration increased. A net production of CO2 allowed growth under conditions of extremely low concentrations of added CO2.When B. fragilis was grown in continuous culture with 100% CO2 or 100% N2, the dilution rate influenced the concentrations of acetate, succinate, propionate, d-lactate, l-malate and formate formed. Decreasing the dilution rate favored propionate and acetate production under both conditions. When the organism was grown with 100% N2, the amount of propionate formed was greater than the amount of succinate formed at all dilution rates. Except at slow dilution rates the reverse was true when 100% CO2 was used. B. fragilis was unable to grow at dilution rates faster than 0.154 h-1 when grown with 100% N2; the Y glc max was 67.9 g DW cells/mol glucose and m s was 0.064 mmol glucose/g DW·h. If the gas atmosphere was 100% CO2 the organism was washed out of the culture when the dilution rate exceeded 0.38 h-1; the Y glc max was 59.4 g DW cells/mol glucose and m s was 0.094 mmol glucose/g DW·h.Measurement of the phosphoenolpyruvate (PEP) carboxykinase (E.C. 4.1.1.49) with whole, permeabilized cells of B. fragilis showed an increase of specific enzyme activity with decreasing CO2 concentrations. The mechanisms used by B. fragilis to adjust to low levels of CO2 are discussed.  相似文献

The photosynthetic CO2 fixation by spinach leaf (Spinacia oleracea L. var. Kyoho) protoplasts was inhibited by substituting an atmosphere of N2 with one of either air (21% O2) or 100% O2. The inhibitory effect of 100% O2 was greater than that of air. The mode of inhibition by 100% O2 and air was competitive with respect to CO2; Ki(O2) value was 0.32 mM at pH 7 and 0.28 mM at pH 8.5 The labeling patterns of compounds in protoplasts exposed to 14CO2 in light after transferring them from N2 to O2 atmospheres were examined. There was no detectable 14CO2 incorporation into glycolate under anaerobic and O2 atmospheres; a more marked labeling of glycine occurred under an oxidative environment compared to that under the anaerobic condition, presumably because of a rapid transformation of glycolate to glycine in the protoplasts.  相似文献

Physiological responses of Opuntia ficus-indica to growth temperature   总被引:2,自引:0,他引:2  
The influences of various day/night air temperatures on net CO2 uptake and nocturnal acid accumulation were determined for Opuntia ficus-indica, complementing previous studies on the water relations and responses to photosynthetically active radiation (PAR) for this widely cultivated cactus. As for other Crassulacean acid metabolism (CAM) plants, net nocturnal CO2 uptake had a relatively low optimal temperature, ranging from 11°C for plants grown at day/night air temperatures of 10°C/0°C to 23°C at 45°C/35°C. Stomatal opening, which occurred essentially only at night and was measured by changes in water vapor conductance, progressively decreased as the measurement temperature was raised. The CO2 residual conductance, which describes chlorenchyma properties, had a temperature optimum a few degrees higher than the optimum for net CO2 uptake at all growth temperatures. Nocturnal CO2 uptake and acid accumulation summed over the whole night were maximal for growth temperatures near 25°C/15°C, CO2 uptake decreasing more rapidly than acid accumulation as the growth temperature was raised. At day/night air temperatures that led to substantial nocturnal acid accumulation (25°C/15°C.). 90% saturation of acid accumulation required a higher total daily PAR than at non-optimal growth temperatures (10°C/0°C and 35°C/25°C). Also, the optimal temperature of net CO2 uptake shifted downward when the plants were under drought conditions at all three growth temperatures tested, possibly reflecting an increased fractional importance of respiration at the higher temperatures during drought. Thus, water status, ambient PAR, and growth temperatures must all be considered when predicting the temperature response of gas exchange for O. ficus-indica and presumably for other CAM plants.  相似文献

A novel marine ice-nucleating bacterium, KUIN-5, was isolated from a marine algae, Monostroma latissum. Strain KUIN-5 was identified as a Pseudomonas sp. from its characteristics and taxonomies; the optimum temperature and pH for its growth were 25°C and 6.0, respectively. When strain KUIN-5 was aerobically cultured in Carlucci-Pramer medium (pH 6.0) for 50 h at 25°C, the highest ice-nucleating activity of the cells among the media for marine bacteria was obtained, and the ice-nucleating temperature, T50, was indicated to be ? 3.2°C. Also, the optimum concentration of NaCl for the growth in this medium, which was prepared with distilled water instead of seawater, was 2.0% (w/v) and then the ice-nucleating activity was inversely proportional to the NaCl concentration. Moreover, when strain KUIN-5 was cultured in Davis medium under optimum conditions, it produced insoluble polysaccharide (IPS) in the culture. The maximum amount of IPS production by strain KUIN-5 was 84.5 mg/ml of medium under optimum conditions. Therefore, this IPS was isolated and could be identified as cellulose, based on TLC or HPLC of the acid hydrolysate, and GC-MS of the acetylated polyalcohol prepared by periodate oxidation and Smith degradation of this polysaccharide. This is the first report of cellulose production by a marine ice-nucleating bacterium.  相似文献

The interactive effects of increased temperature and CO2 enrichment on the growth of 2‐year‐old saplings of Quercus myrsinaefolia, an evergreen broad‐leaved oak, were studied throughout an entire year in the vicinity of their northernmost distribution. Saplings were grown under different conditions in two chambers: (1) a temperature gradient chamber at ambient temperature, 3 and 5 °C warmer conditions with an ambient CO2 concentration, and (2) in a CO2 temperature gradient chamber at 3 °C warmer conditions with 1·5 times the normal CO2 concentration, and 5 °C warmer conditions with doubled CO2 concentration. The 3 and 5 °C warmer conditions enhanced the relative growth rate during almost the entire year, producing 53 and 47% increases in annual biomass production, 27 and 44% enhancement of root growth during shoot dormancy and 3 and 5 week prolongation of the shoot growing period, respectively. However, a daily mean air temperature exceeding 30 °C under the 5 °C warmer condition caused a marked reduction in net assimilation rate (NAR) from July to September. The CO2 enrichment further enhanced the positive effects of warming in spring and the resulting increases in NAR almost completely compensated for the negative effect of warming during summer. From autumn to winter, attenuation of the effects of CO2 was compensated by the increased sink strength produced by the warming. The annual biomass production was more than doubled by the combination of temperature elevation and CO2 enrichment.  相似文献

An extremely halophilic Chromohalobacter sp. TVSP101 was isolated from solar salterns and screened for the production of extracellular halothermophilic protease. Identification of the bacterium was done based upon biochemical tests and the 16S rRNA sequence. The partially purified enzyme displayed maximum activity at pH 8 and required 4.5 M of NaCl for optimum proteolytic activity. In addition, this enzyme was thermophilic and active in broad range of temperature 60–80°C with 80°C as optimum. The Chromohalobacter sp. required 4 M NaCl for its optimum growth and protease secretion and no growth was observed below 1 M of NaCl. The initial pH of the medium for growth and enzyme production was in the range 7.0–8.0 with optimum at pH 7.2. Various cations at 1 mM concentration in the growth medium had no significant effect in enhancing the growth and enzyme production but 0.5 M MgCl2 concentration enhanced enzyme production. Casein or skim milk powder 1% (w/v) along with 1% peptone proved to be the best nitrogen sources for maximum biomass and enzyme production. The carbon sources glucose and glycerol repressed the protease secretion. Immobilization of whole cells in absence of NaCl proved to be useful for continuous production of halophilic protease.  相似文献

Abstract Some characteristics of photosynthetic inorganic carbon uptake by Palmaria palmata, a marine red macroalga, have been measured under physiological conditions in artificial seawater. The apparent affinity of thallus for CO2 [K1/2(CO2)] at pH 8.0 and 15°C was 21.4±3.0mmol m?3 CO2 under air, and 25.7±70mmol m?3 CO2 under N2. The corresponding values of Vmax were 2.98 ± 0.42 and 3.65±0.87 mmol O2 evolved g Chr?1 s?l. The apparent Km(CO2) of isolated ribulose bisphosphate carboxylase was determined at pH 8.0 and 30 °C to be 30.2 mmol m?3 CO2, and the corresponding value of Vmax was 19.67 μniol CO2 g protein?1 s?1. The CO2 compensation points of the thallus were measured in artificial seawater at pH 8.0 under air and N2, using a gas-chromatographic method. The values were relatively low, rising from 10 cm3 m?3 at 15°C, to 35 cm3 m?3 at 25°C, but were not affected by the O2 concentration. The lack of an effect of O2 on photosynthesis and on compensation point indicates that there is little photorespiratory CO2 loss in this macroalga. The high affinity of the thallus for CO2, and the low CO2 compensation concentrations, are consistent with the occurrence of bicarbonate uptake in this alga.  相似文献

Although carbon dioxide (CO2) is known to inhibit growth of most bacteria, very little is known about the cellular response. The food-borne pathogen Listeria monocytogenes is characterized by its ability to grow in high CO2 concentrations at refrigeration temperatures. We examined the listerial responses of different strains to growth in air, 100% N2, and 100% CO2. The CO2-induced changes in membrane lipid fatty acid composition and expression of selected genes were strain dependent. The acid-tolerant L. monocytogenes LO28 responded in the same manner to CO2 as to other anaerobic, slightly acidic environments (100% N2, pH 5.7). An increase in the expression of the genes encoding glutamate decarboxylase (essential for survival in strong acid) as well as an increased amount of branched-chain fatty acids in the membrane was observed in both atmospheres. In contrast, the acid-sensitive L. monocytogenes strain EGD responded differently to CO2 and N2 at the same pH. In a separate experiment with L. monocytogenes 412, an increased isocitrate dehydrogenase activity level was observed for cells grown in CO2-containing atmospheres. Together, our findings demonstrate that the CO2-response is a partly strain-dependent complex mechanism. The possible links between the CO2-dependent changes in isocitrate dehydrogenase activity, glutamate metabolism and branched fatty acid biosynthesis are discussed.  相似文献

Aims: The objective of the current study was to examine the interactions between Pseudomonas putida and Escherichia coli O157:H7 in coculture studies on fish‐burgers packed in air and under different modified atmospheres (30 : 40 : 30 O2 : CO2 : N2, 5 : 95 O2 : CO2 and 50 : 50 O2 : CO2), throughout the storage at 8°C. Methods and Results: The lag‐exponential model was applied to describe the microbial growth. To give a quantitative measure of the occurring microbial interactions, two simple parameters were developed: the combined interaction index (CII) and the partial interaction index (PII). Under air, the interaction was significant (P < 0·05) only within the exponential growth phase (CII, 1·72), whereas under the modified atmospheres, the interactions were highly significant (P < 0·001) and occurred both in the exponential and in the stationary phase (CII ranged from 0·33 to 1·18). PII values for E.  coli O157:H7 were lower than those calculated for Ps. putida. Conclusions: The interactions occurring into the system affected both E. coli O157:H7 and pseudomonads subpopulations. The packaging atmosphere resulted in a key element. Significance and Impact of the Study: The article provides some useful information on the interactions occurring between E. coli O157:H7 and Ps. putida on fish‐burgers. The proposed index describes successfully the competitive growth of both micro‐organisms, giving also a quantitative measure of a qualitative phenomenon.  相似文献

The combined effects of carbon dioxide (CO2) enrichment and water deficits on nodulation and N2 fixation were analysed in soybean [Glycine max (L.) Merr.]. Two short-term experiments were conducted in greenhouses with plants subjected to soil drying, while exposed to CO2 atmospheres of either 360 or 700 μmol CO2 mol–1. Under drought-stressed conditions, elevated [CO2] resulted in a delay in the decrease in N2 fixation rates associated with drying of the soil used in these experiments. The elevated [CO2] also allowed the plants under drought to sustain significant increases in nodule number and mass relative to those under ambient [CO2]. The total non-structural carbohydrate (TNC) concentration was lower in the shoots of the plants exposed to drought; however, plants under elevated CO2 had much higher TNC levels than those under ambient CO2. For both [CO2] treatments, drought stress induced a substantial accumulation of TNC in the nodules that paralleled N2 fixation decline, which indicates that nodule activity under drought may not be carbon limited. Under drought stress, ureide concentration increased in all plant tissues. However, exposure to elevated [CO2] resulted in substantially less drought-induced ureide accumulation in leaf and petiole tissues. A strong negative correlation was found between ureide accumulation and TNC levels in the leaves. This relationship, together with the large effect of elevated [CO2] on the decrease of ureide accumulation in the leaves, indicated the importance of ureide breakdown in the response of N2 fixation to drought and of feedback inhibition by ureides on nodule activity. It is concluded that an important effect of CO2 enrichment on soybean under drought conditions is an enhancement of photoassimilation, an increased partitioning of carbon to nodules and a decrease of leaf ureide levels, which is associated with sustained nodule growth and N2 rates under soil water deficits. We suggest that future [CO2] increases are likely to benefit soybean production by increasing the drought tolerance of N2 fixation.  相似文献

An on-line technique, based on measuring the increase in pressure due to CO2 release in a closed air-tight reactor, was used to evaluate the fermentation of lactate by propionibacteria. The method was applied to batch cultures of Propionibacterium shermanii grown in yeast extract/sodium lactate medium containing lactate as a carbon source under micro-aerophilic conditions. Gas pressure evolution was compared both with substrate consumption and metabolites production and with acidification and growth. Linear relationships were found between gas pressure variation, lactate consumption and propionate and acetate production. The technique also enabled the evaluation of total CO2 produced, by taking account of pressure, oxygen and pH measurements. These results tend to show that this simple and rapid method could be useful to monitor propionic acid bacteria growth.  相似文献

The effect of varied anaerobic atmospheres on the metabolism of sweet potato (Ipomoea batatas [L.] Lam.) roots was studied. The internal gas atmospheres of storage roots changed rapidly when the roots were submerged under water. O2 and N2 gases disappeared quickly and were replaced by CO2. There were no appreciable differences in gas composition among the four cultivars that were studied. Under different anaerobic conditions, ethanol concentration in the roots was highest in a CO2 environment, followed by submergence and a N2 environment in all the cultivars except one. A positive relationship was found between ethanol production and pyruvate decarboxylase activity from both 100% CO2-treated and 100% N2-treated roots. CO2 atmospheres also resulted in higher pyruvate decarboxylase activity than did N2 atmospheres. Concentrations of CO2 were higher within anaerobic roots than those in the ambient anaerobic atmosphere. The level of pyruvate decarboxylase and ethanol in anaerobic roots was proportional to the ambient CO2 concentration. The measurable activity of pyruvate decarboxylase that was present in the roots was about 100 times less than that of alcohol dehydrogenase. Considering these observations, it is suggested that the rate-limiting enzyme for ethanol biosynthesis in sweet potato storage roots under anoxia is likely to be pyruvate decarboxylase rather than alcohol dehydrogenase.  相似文献

A novel thermophilic, anaerobic, rod-shaped bacterium strain, designated Buff, was isolated from buffalo-dung samples collected from a buffalo-farm located in Caserta (Campania, south of Italy). Strain Buff was Gram-positive, motile and no spore-forming. The growth temperature range was 40–65°C with an optimum at 60°C, while pH growth range at 60°C was 5.5–8.0 with an optimum at about pH 6.5. NaCl growth concentration ranged from 0 to 2.0% with an optimum at 0.5% (w/v); no growth was observed with the presence of NaCl 3.0% (w/v). The strain produced ethanol, acetate, lactate, H2, H2S and CO2 by glucose fermentation. The DNA G + C content was 34.4 mol%. As determined by 16S rRNA sequence analysis, this organism belonged to the genus Thermoanaerobacterium. On the basis of the physiological and molecular properties, we propose for strain Buff the new species designation Thermoanaerobacterium thermostercus sp. nov. This novel organism represents the first species of the genus Thermoanaerobacterium isolated from buffalo-dung. The type strain is Buff (=DSM 22141 = ATCC BAA-1776).  相似文献

A novel coccoid-shaped, hyperthermophilic, heterotrophic member of the archaea was isolated from a shallow marine hydrothermal system at Vulcano Island, Italy. The isolate grew between 56 and 90° C with an optimum around 85° C. The pH range for growth was 6.5 to 10.5, with an optimum around 9.0. Polysulfide and elemental sulfur were reduced to H2S. Sulfur stimulated the growth rate. The isolate fermented yeast extract, peptone, meat extract, tryptone, and casein. Isovalerate, isobutyrate, propionate, acetate, CO2, NH3, and H2S (in the presence of S°) were detected as end products. Growth was not inhibited by H2. Based on DNA-DNA hybridization and 16S rRNA partial sequences, the new isolate represents a new species of Thermococcus, which we named Thermococcus alcaliphilus.
