Adult cabbage maggots ( Delia radicum L.) were exposed to dry conidia of isolates of several hyphomycetous fungi by placing them in a centrifuge tube containing conidia, then releasing them into small screened plastic cages. Mortality was assessed after 48, 120 and 160 h. A Beauveria bassiana isolate (P89 from Musca domestica ) caused the highest mortality after 48 h, resulting in 100% mortality and 100% infection. Isolate L90 ( B. bassiana ) and one Metarhizium anisopliae isolate (ARSEF 2521) also caused fatal infection in more than 50% of the flies. To investigate exchange of inoculum, flies were placed in a small container with a dry powder formulation containing B. bassiana (Mycotrol ® ) on the bottom. The flies were removed to small screened cages containing untreated flies. This experiment confirmed the ability of flies to pass inoculum to other flies. In a similar experiment, one treated fly was placed in each cage with one untreated fly. When each fly died, one untreated fly was added to each cage after the dead fly was removed. This study showed that fly to fly transfer of fatal doses of inoculum was possible for a series of at least six flies. When female flies were exposed to the inoculum, then transferred to small cages containing males and an oviposition substrate, no eggs were laid. Further studies are being conducted to develop a system where flies attracted to a trap will be inoculated with the fungus and spread it to a field population.  相似文献

Abstract. 1. The efficiency of D.austrinus as a biological control agent depends on successful colonization of the cactus weed Opuntia aurantiaca .
2. Unlike other coccoid insects, the female crawlers (first-instar larvae) of cochineal insects develop long wax fiaments on the dorsal surface of the body that reduce their terminal velocity in air and enhance wind dispersal.
3. Dactylopiid males are dispersed as winged adults and male crawlers of D.austrinus have fewer and shorter filaments than the females and they are blown a shorter distance in a wind tunnel.
4. Vertical and horizontal wind dispersal of D.austrinus from O.aurantiaca in the field is usually very limited, but was greatly improved when crawlers were launched from an elevated 'tower'.
5. Preparatory to dispersal, female crawlers climb to the top of the host plant and 'take-off' from there. This behaviour is most marked in crawlers 2–3 days old at which stage the wax filaments are best developed: the behaviour is never manifest following artificial 'dispersal'.
6. Final settling on cladodes of the host plant is a function of the age of the female crawlers and of the condition of the cladodes.
7. When given a choice, approximately equal numbers of crawlers settled on cladodes that had received high and low fertilizer treatments. More crawlers settled on cladodes that had been kept in the shade than on those kept in the sun, and on basal rather than terminal cladodes.
8. The highest percentage of crawlers settled on cladodes that were 20% dehydrated. Settling success was much lower on more desiccated cladodes.  相似文献

Here we report a case of cutaneous alternariosis in a 74-year-old man treated by corticotherapy for myasthenia, and presenting with papular, crusted lesions on the left elbow and the right knee. Histological examination of the biopsy specimens showed fungal hyphae associated with round-shaped cells which were highly suggestive of alternariosis. Mycological culture allowed the isolation of a dematiaceous fungus which was identified as a member of the Alternaria infectoria species-group. This was confirmed by PCR amplification and sequencing of the internal transcribed spacer domain of the gene encoding nuclear ribosomal DNA and of the mitochondrial small subunit ribosomal DNA domain. The fungus was therefore referred to the Scientific Institute of Public Health where it was identified as Alternaria infectoria, on the basis of its very small 1 or 2-celled conidia often arranged in long chains and presenting with very long secondary conidiophores. Corticotherapy was stopped and a local antifungal treatment with ketoconazole was initiated, allowing the stabilisation of the cutaneous lesions within 2 months.  相似文献

The susceptibility of different geographical strains of Phlebotomus papatasi to a cytoplasmic polyhedrosis virus (CPV) was determined experimentally by feeding polyhedra to larvae. Of the Indian P. papatasi, 15.6% became infected, whereas Egyptian P. papatasi were mostly refractory. Infection rates were not augmented in colony flies from the Jordan Valley, 23.8% of which were naturally infected with CPV. The infectivity of Serratia marcescens and Beauvaria bassiana to P. papatasi were determined experimentally. A suspension of B. bassiana spores or S. marcescens bacteria, ingested by P. papatasi in sucrose solution, did not significantly augment mortality rates or reduce the number of eggs oviposited. However, B. bassiana spores smeared on a filter paper constituting 1 or 5% of the surface area available to flies induced 100% mortality of P. papatasi on days 5 and 4, respectively. Mortality in Lutzomyia longipalpis reached 100% on day 4. There were markedly lower mortality rates in the control groups and more eggs were produced by these females (P. papatasi: control = 48.5; experimental = 0.9-1.6 eggs/female; L. longipalpis; control = 17.1; experimental = 0 eggs/female). From wild-caught Colombian Lutzomyia spp., a nonfluorescent pseudomonas, an Entomophthorales fungus, and a Trypanosomatid protozoon (probably Leptomonas) were isolated in culture media. Gregarines (Ascogregarina saraviae) and nematodes (Tylenchida and Spirurida) were also recorded. In laboratory-reared flies, an ectoparasitic fungus was associated with high mortality rates of first instar Lutzomyia spp. larvae. Opportunistic ectoparasitic aggregates of bacteria, yeast, and fungi on the tarsi of colonized L. longipalpis and P. papatasi hindered their mobility and were associated with reduced colony vigor. Aspergillus flavus, B. bassiana, and S. marcescens were isolated from laboratory-bred P. papatasi adults.  相似文献

Successful control of greenhouse whitefly may be achieved by complementary activity of the parasitoidEncarsia formosa and the fungusAschersonia aleyrodis. One way to obtain an additive mortality effect of both entomopathogen and parasitoid would be achieved by the selection of healthy hosts by the parasitoid and rejection of fungus-infected hosts. Third and fourth instar larvae ofTrialeurodes vaporariorum which had been treated with a spore suspension ofA. aleyrodis 0, 4, 7, 10 or 14 days beforehand, were presented to female parasitoids. The parasitoids adopted the oviposition posture on untreated hosts as well as on treated hosts, irrespective of the different stages of infection in the hosts. However, significantly more hosts were parasitized byE. formosa in the control treatment than in the fungal treatment. The parasitoids offered treated hosts, showed rejection behaviour after probing on hosts showing detectable signs of infection (containing hyphal bodies or mycelium in the haemolymph). For instance, when hosts were offered seven days after spore treatment, the parasitoids showed an oviposition posture on a total of 83 (95.4%) out of 87 infected larvae, but laid only 4 eggs (4.6%). In contrast, on 48 (94.1%) out of 51 noninfected (or showing no detectable signs of infection) hosts an oviposition posture was adopted and 40 eggs (78.4%) were found after dissection. When infected hosts were encountered the oviposition posture lasted less than 1′40″ while rejection of non-infected hosts occurred after more than 1′40″. Other experiments were carried out offering treated hosts for 24 h to the parasitoids. The hosts were dissected afterwards. Again, significantly more eggs were laid in the non-infected hosts. When hosts were parasitized shortly after fungal spore treatment they were colonized by the fungus and the parasitoids did not develop. Transmission of the entomopathogen after probing infected hosts was observed to a limited extent. In conclusion,A. aleyrodis andE. formosa can be used together in a glasshouse situation. The parasitoid will be most effective when introduced more than seven days after application ofA. aleyrodis, because from that time onwards it is able to detect and reject fungus-infected hosts.  相似文献

To complement existing information on the mortality of larvae of the wasp Cotesia plutellae attacking moth caterpillars infected with Bacillus thuringiensis (Bt) we tested the direct and indirect effects of the bacterium on adult wasp longevity and oviposition behaviour. In one experiment with female parasitoids, mean longevity (SEM) was not significantly different between females exposed to Bt (1.98 +/- 0.08 days) and those not exposed (2.18 +/- 0.13 days). In a second experiment with both males and females, Bt treatment did not significantly effect either male or female parasitoids exposed to Bt. To observe the possible effects of Bt on oviposition behaviour of C. plutellae each of 10 females were given five larvae that had been treated with Bt and five untreated larvae at the same time. All parasitoids were observed to make oviposition attempts in both untreated and treated larvae. Upon dissection of the host larvae, one or more C. plutellae eggs were found in each of the larvae in which a parasitoid attempted oviposition. There was no effect of Bt treatment on parasitoid oviposition. The mean number of ovipositions in treated larvae (4.3 +/- 0.3) was not significantly different from untreated larvae (4.7 +/- 0.2).  相似文献

The entomopathogenic fungus Metarhizium anisopliae var. acridum is being developed for use as a mycoinsecticide against locusts and grasshoppers in Africa. In addition to causing significant mortality, studies have shown the pathogen to cause reductions in feeding of a number of target species during the disease incubation period. The present study extends this work by demonstrating significant reductions in feeding following infection in another important pest species, the brown locust Locustana pardalina (Walker). Insects collected from the field following a spray application and subsequently maintained in a field laboratory showed a significant reduction in per capita feeding, as indicated by faecal production, as early as 2 days after treatment; this was before any pathogen-induced mortality. This per capita reduction in feeding by infected individuals contributed to a total reduction in feeding of 65% relative to controls. In addition to this laboratory based assessment, experiments were conducted to examine the effects of infection on mortality, feeding and fecundity in the field. Mortality rates of infected individuals in the field were significantly slower than those in the laboratory (average survival time of approximately 39 days compared with 6 days), although total mortality and percentage mycosis were equivalent. Treated individuals showed a significant reduction in body fat accumulation at sexual maturity compared with controls. This may reduce dispersal ability and total reproductive fitness. However, treated individuals exhibited a reduced pre-oviposition period resulting in them laying more eggs than untreated individuals within the first weeks following fledging. Thus, no significant reductions in fecundity were recorded by the end of the assessment period when mortality of treated individuals exceeded 90%. Given that many untreated locusts were still alive and had the potential to continue oviposition for several more weeks, however, it is likely that differences in fecundity would have become apparent if the assessment had continued. These results provide important insights for evaluating overall impacts of spray applications of the mycoinsecticide.  相似文献

Larvae of the cedar web-spinning sawfly, Cephalcia tannourinensis Chevin (Hymenoptera: Pamphiliidae), infected with a white fungus were collected from the Tannourine-Hadath El-Jebbeh cedar forest. Macro- and micro-morphological data based on the examination of colonies, conidiophores, and conidial shape of the fungus suggested a Beauveria species. Sequence analysis of the internal transcribed spacer regions of the isolated fungus showed that it is most closely related to isolates of B. bassiana Clade C. The present study showed that the isolated B. bassiana is a naturally occurring entomopathogenic fungus parasitizing the larvae of C. tannourinensis in Lebanon. Laboratory bioassays showed that B. bassiana caused high mortality of eggs and larvae. The infected eggs turned brownish in color, while larvae of the first instar ceased feeding and showed immobility and rigidity within 5 days and before sporulating conidial mat appeared on their cuticle. Second and third larval instars took longer time to show fungal sporulation: mortalities ranged between 85 and 100% within 7 days when treated with different conidial concentrations. The efficacy of control of C. tannourinensis using B. bassiana was higher or equal to the reference insect growth regulator, diflubenzuron, suggesting the possibility of its success as a biological control agent.  相似文献

The effect of iprodione on the seed-borne phase of Alternaria brassicicola   总被引:2,自引:0,他引:2  
Alternaria brassicicola infection of Brassica oleracea seeds was effectively controlled by a dust application of iprodione (Rovral 50% w.P.). At 2.5 g a.i./kg the seed-borne fungus was usually eliminated from samples with up to 61.5% affected seeds (35.5% internally diseased) but higher levels of infection required increased doses for complete eradication of the fungus. The germination of healthy seeds, including samples from 7–yr-old stocks, on filter paper was unaffected by the treatment. However, the germination of diseased samples, particularly those internally infected with A. brassicicola, was improved. More seedlings emerged from iprodione treated than from untreated seeds in glasshouse soil but the differences were not significant. The application of gamma-hexachlorocyclohexane to iprodione treated seeds sown in soil did not adversely affect subsequent emergence or disease control. Disease control was maintained and germination was not affected by the treatment when treated infected seeds were stored for 2 yr at 10 °C, 50% r.h. In a field trial iprodione seed treatment reduced seedling infection in a cabbage crop grown from naturally diseased seeds (100% contaminated, 45.5% internally infected) from 5.6 to 0.04%.  相似文献

Filtrates of three isolates of the nematophagous fungus Verticillium leptobactrum were evaluated for their nematicidal activity against the root-knot nematode Meloidogyne incognita. The filtrates inhibited egg hatching, with maximum toxicity observed for isolate HR21 at 50% (v:v) dilution, after 7 days exposure. Filtrates also inactivated second-stage juveniles (J2) at 10-50% dilutions. A scanning electron microscopy study of treated eggs showed severe alterations caused by the filtrate of isolate HR43 on M. incognita eggs, which appeared collapsed and not viable, suggesting the production of chitin-degrading enzymes or other active compounds.  相似文献

Studies on the tolerance of pink hibiscus mealybug, Maconellicoccus hirsutus (Green), to ionizing irradiation were undertaken to determine the dose needed to disinfest commodities of this pest. Overall, radiotolerance of M. hirsutus was found to increase with maturity. Target doses of 50 Gy reduced eclosion of eggs to <50%, but doses as great as 750 Gy did not eliminate hatching during the study. At 100 Gy, M. hirsutus eggs, crawlers, and nymphs were controlled, because progeny were not produced despite crawlers and nymphs living for much longer periods than unexposed individuals. Fecundity of treated crawlers and nymphs was greatly impacted by treatment of 100 Gy; crawlers developing into adults produced no eggs, and 10 adults of 3,983 treated nymphs (0.25%) produced 309 eggs. Few adult females exposed as nymphs deposited eggs because male nymphs died during development, which left the females unfertilized. By comparison, 89% of female nymphs treated at 100 Gy and mated as adults with nonirradiated males produced a total of 1,447 eggs (19 eggs per female). Evidence from this study suggests M. hirsutus reproduces sexually, not parthenogenetically. Adults, the most resistant stage, exposed to target doses of 100 Gy produced eggs that were 1.2% viable, from which a small portion of individuals successfully completed development and produced progeny. A target dose of 250 Gy was sufficient to control adult M. hirsutus because, at that dose, none of the eggs produced by 3,093 irradiated adults eclosed. The minimum dose needed to ensure quarantine security is between 100 and 250 Gy.  相似文献

The causative agent of alveolar hydatidosis in humans, the fox tapeworm Echinococcus multilocularis, is extending its geographical range in Europe and has been found in domestic cats in some areas. A dermally applied cestocidal treatment for domestic cats has been developed and the efficacy of this treatment is reported. Thirty purpose-bred cats were experimentally infected each with 10000 protoscoleces of Echinococcus multilocularis. Ten days later one group of ten cats was treated with Droncit(R) Spot-on (Praziquantel) 4% w/v dermally in one place on the dorsal aspect of the neck at a dose of 8 mg/kg. Eleven days later (21 days p.i.) a second group of ten cats was also treated with Droncit(R) Spot-on the same way. One group of ten cats was left untreated as controls. Twenty three days after infection the cats were examined for the presence of E. multilocularis tapeworms. No E. multilocularis were recovered from any of the cats in either of the treated groups. Echinococcus multilocularis were recovered from eight of the ten cats left untreated as controls. The worm burdens in the untreated cats were 0, 0, 5, 15, 75, 110, 220, 815, 2635, and 3045 worms per cat. The worms ranged in development from the three to four segment stage. Many of the E. multilocularis with four segments contained unshelled eggs in the terminal segment. This study indicates that Droncit(R) Spot-on (Praziquantel) 4% w/v applied dermally at 8 mg/kg is highly effective in removing E. multilocularis from the small intestine of cats infected with immature and mature (prepatent) infections of E. multilocularis. In the cats with the mature infections all tapeworms were absent from the small intestine within 2 days of treatment.  相似文献

Lewia hordeicola with Alternaria anamorph was isolated from barley grains in Norway. The fungus is homothallic. It produces fertile ascomata on synthetic nutrient agar (SNA) after long incubation at 4 C in the dark. On PCA its anamorph resembles members of the A. infectoria species group. On SNA L. hordeicola differs from the latter in the shape and size of ascospores, the conidial sporulation patterns, and the shape, size, septation and roughness of conidia. Aims:  A possibility of using cycloheximide tolerance and/or sensitivity as an additional diagnostic tool for distinguishing morphologically related species within common small-spored Alternaria has been tested during this study.
Methods and Results:  A total of 33 strains from four Alternaria species-groups, namely Alternaria alternata , Alternaria arborescens , Alternaria infectoria and Alternaria tenuissima were tested for their growth response to 100 μg m−1 cycloheximide in potato carrot agar. All A. infectoria strains were completely inhibited, showing no growth at all even after prolonged incubation. In contrast, all other strains representing the remnant three species exhibited a high resistance to this antibiotic.
Conclusions:  Cycloheximide sensitivity represents a further important physiological character for distinguishing A. infectoria from the three similar species.
Significance and Impact of the Study:  The relevance of these findings corresponds with the potential ability of the Alternaria species produce mycotoxins. Cycloheximide may be in future used in the design of selective media for the isolation of some potentially toxigenic food-borne Alternaria species such as A. alternata , A. tenuissima and/or A. arborescens , for example in screening cereals for toxigenic Alternaria spp. and for their direct separation from nontoxigenic representatives of A. infectoria species-group.  相似文献

A fungus, identified as Alternaria alternata, was isolated from dying or dead aphids and proved to be pathogenic. It was isolated from different parts of Greece from aphid specimens on cultivated plants, ornamentals and weeds. In the laboratory, disease development started with the germination of spores on the insect integument and the subsequent growth of mycelium. The fungus formed apical and intercalary, globose or lobate appressoria which were firmly attached onto the host exoskeleton and facilitated entrance of the mycelium into the insect body. Under favorable conditions of temperature (15–35 °C) and relative humidity (100%), infected aphids died in 2–4 days. A characteristic brown discoloration accompanied the death of the insects. Both mycelial growth and sporulation were profuse on dead specimens. The pathogen infected all 26 aphid species tested but was unable to infect other insects (Drosophila melanogaster and Ceratitis capitata) or aphid host plants. There were significant differences in mortality rate among aphid species only during the first two days after inoculation. It is suggested that A. alternata may be a good candidate to be exploited for the biological control of aphids.  相似文献

Conidia ofBeauveria bassiana were evaluated for activity against adults ofTribolium castaneum, under laboratory conditions closely resembling the stored-grain conditions. Three doses of the fungus were applied in the treatments: 0.1 g, 0.5 g and 1 g per 20 insects/dose/flask. Treated insects and untreated were incubated in broken wheat. There were 5 replications for each test. A total of 1600 treated and control insects used were kept in a climatic chamber (70±5% and 27±2°C) for 45 days. Observations were performed daily and the recording of infected cadavers began when the percentage of mortality exceeded 50%. The evaluation of the efficiency of each dose ofB. bassiana was made 14, 21, 28 and 45 days after inoculation. Mortality of adults that were exposed to the beetles treated with 0.5 g and 1 g of conidia ofB. bassiana / 20 adults was 87 and 85% respectively, within 21 days of exposure. Onset of mortality was further delayed at lower dosage of the fungus; 55% after 21 days. After 28 days there were no difference in mortality between the 0.5 g and 1 g dosages ofB. bassiana. The most efficient treatment was 0.5 g/20 insects, which resulted in a control higher than 50% 14 days post treatment.  相似文献

A recently introduced fungus gnat, Bradysia agrestis, has caused serious problems in Korean propagation houses where vegetable seedlings are produced for transplant into the fields. Although chemical insecticides are available against this pest, alternate control measures are needed. A Korean isolate of the entomopathogenic nematode, Steinernema carpocapsae Pocheon strain, was tested against this insect in the laboratory and propagation house. In the laboratory, S. carpocapsae affected oviposition, with the untreated females laying an average of 121±25 eggs, whereas the treated females averaged 7±2 eggs. The infectivity of S. carpocapsae to the fungus gnat was affected by the developmental stage and temperature, with highest mortality observed with the third and fourth instars and pupal stage. Nematode mortality in the second instar fungus gnat ranged between 23 and 35%, but showed no significant differences among the temperatures tested. The egg and first instar were not infected by the nematode. In nematode dispersal studies, adult female fungus gnats alone dispersed S. carpocapsae from the nematode-treated area to the control area at a higher rate than male and female gnats or male gnats alone. In the propagation house experiments with watermelon seedlings, no significant difference was observed in fungus gnat larval reduction at S. carpocapsae concentrations of 5, 10, or 20 infective juveniles (IJs)/g of soil at 7, 14, and 21 days after treatment. In comparison with the control, the S. carpocapsae treatments significantly reduced B. agrestis larval numbers. When the watermelon seed was treated with S. carpocapsae at sowing, the larval density of B. agrestis was significantly reduced, that is, the number of B. agrestis larvae ranged from four to eight and from five to eight in the nematode-treated plots compared with 26 and 30 in the control plots on the 17th and 34th day post-treatment, respectively. In the chemical insecticide treatments, diflubenzuron and chlorpyrifos were significantly more effective than S. carpocapsae and diazinon in reducing larval populations of the fungus gnat. Our data show that, although some of the chemical insecticides were more effective than S. carpocapsae Pocheon strain, the nematode was still an effective tool for management of the fungus gnat larvae and in protecting seedlings from damage in propagation houses.  相似文献

The potential of a strain of Beauveria bassiana (Ascomycota: Clavicipitaceae) obtained from a naturally infected Rhynchophorus ferrugineus (Coleoptera: Curculionidae) pupa as a biological control agent against this weevil was evaluated both in the laboratory and in semi-field assays. Laboratory results indicate that this strain of B. bassiana can infect eggs, larvae and adults of R. ferrugineus (LC50 from 6.3 × 107 to 3.0 × 109 conidia per ml). However, mortality was not the only indicator of treatment efficacy because adults of either sex inoculated with the fungus efficiently transmitted the disease to untreated adults of the opposite sex, with male-to-female and female-to-male rates of transmission of 55% and 60%, respectively. In addition, treatment with B. bassiana significantly reduced fecundity (up to 62.6%) and egg hatching (32.8%) in pairing combinations with fungus-challenged males, females or both sexes. Likewise, 30-35% increase in larval mortality was observed in larvae obtained from eggs from fungus-challenged females or from untreated females coupled with inoculated males, resulting in an overall 78% progeny reduction. Semi-field preventive assays on potted 5-year old Phoenix canariensis palms, with efficacies up to 85.7%, confirmed the potential of this strain as a biological control agent against R. ferrugineus.  相似文献

Bee colonies in southern France were treated with conidia (asexual spores) from two strains of Beauveria bassiana, an entomopathogenic fungus. One strain was commercial (GHA) and the other had been isolated from Varroa mites in the region (Bb05002). Objectives were to evaluate treatment effect on colony weight, adult bee mass, capped brood, and on Varroa fall onto sticky boards. Treatments included conidia formulated with either carnauba or candelilla wax powder, candelilla wax powder alone, or control; in two treatment groups formulation was applied a second time after one week. Treatment did not affect colony health. Colonies treated twice with Bb05002 conidia and carnauba wax powder had significantly higher mite fall compared to colonies treated with blank candelilla wax powder. The proportion of fallen mites that were infected in both conidia treatments was higher than controls for 18 days after the second treatment. The number of fungal propagules on the bees themselves remained elevated for about 14 days after the second treatment. These results were compared to published results from previous experiments with regard to infection duration.  相似文献

Three approaches were used to investigate effects of host plant epicuticular waxes on oviposition site selection by Plutella xylostella (L.). In the first approach, oviposition on canola (Brassica napus L.) that had epicuticular wax reduced by application of a carbamate herbicide (S-ethyl dipropylthiocarbamate) was compared with oviposition on untreated control plants. A second approach compared oviposition on sibling strains of B. napus with different wax blooms (glossy and waxy), and a third approach compared oviposition by P. xylostella on parafilm that had been applied to glossy and waxy B. napus strains for transfer of leaf components. Significantly more eggs were deposited on herbicide-treated plants (with reduced epicuticular wax) than on untreated controls. Similarly, more eggs were deposited on glossy than on waxy sibling strains of B. napus. In parafilm assays significantly more eggs were deposited on treated than on untreated parafilm. Several mechanisms could explain the differences in attractiveness of surfaces with varying wax content as oviposition sites for P. xylostella, including visual, chemical, and tactile differences between substrates.
