This paper considers the numerical approximation for the optimal supporting position and related optimal control of a catalytic reaction system with some control and state constraints, which is governed by a nonlinear partial differential equations with given initial and boundary conditions. By the Galerkin finite element method, the original problem is projected into a semi-discrete optimal control problem governed by a system of ordinary differential equations. Then the control parameterization method is applied to approximate the control and reduce the original system to an optimal parameter selection problem, in which both the position and related control are taken as decision variables to be optimized. This problem can be solved as a nonlinear optimization problem by a particle swarm optimization algorithm. The numerical simulations are given to illustrate the effectiveness of the proposed numerical approximation method.  相似文献

The determination of an optimum feeding profile of a fed-batch fermentation requires the solution of a singular optimum control problem, which is often complicated by changes in the process kinetics during the fermentation. The procedure of optimization may be sufficiently simple, if the feeding part of fermentation is carried out in the quasi-steady state. In this work an algorithm for operating a fed-batch fermentation using mentioned regime is offered. The algorithm supposes a periodical correction of the feeding strategy. Applying to fed-batch lysine fermentation demonstrate efficacy of this algorithm over frequently used strategies.  相似文献

The optimal substrate feeding policy for the fed batch fermentation which is governed by product and substrate inhibited kinetics is presented. The conjunction point between nonsingular and singular arcs and the feeding policy along the singular arc are derived analytically in terms of the concentrations of substrate and product and the liquid volume. Thus, it is possible to determine the feeding rate by monitoring the state variables (i.e., closed loop control). As a specific example, an optimization study of the fed batch fermentation for ethanol production by Saccharomyces cerevisiae is presented. It is shown that the optimal feeding patterns are heavily dependent upon the initial conditions. The point selectivity provides the guideline for predicting the optimal feeding patterns and explaining the results of rigorous mathematical analysis.  相似文献

The Briggs–Haldane standard quasi-steady state approximation and the resulting rate expressions for enzyme driven biochemical reactions provide crucial theoretical insight compared to the full set of equations describing the reactions, mainly because it reduces the number of variables and equations. When the enzyme is in excess of the substrate, a significant amount of substrate can be bound in intermediate complexes, so-called substrate sequestration. The standard quasi-steady state approximation is known to fail under such conditions, a main reason being that it neglects these intermediate complexes. Introducing total substrates, i.e., the sums of substrates and intermediate complexes, provides a similar reduction of the number of variables to consider but without neglecting the contribution from intermediate complexes. The present theoretical study illustrates the usefulness of such simplifications for the understanding of biochemical reaction schemes. We show how introducing the total substrates allows a simple analytical treatment of the relevance of significant enzyme concentrations for pseudo first-order kinetics and reconciles two proposed criteria for the validity of the pseudo first-order approximation. In addition, we show how the loss of zero-order ultrasensitivity in covalent modification cycles can be analyzed, in particular that approaches such as metabolic control analysis are immediately applicable to scenarios described by the total substrates with enzyme concentrations higher than or comparable to the substrate concentrations. A simple criterion which excludes the possibility of zero-order ultrasensitivity is presented.  相似文献

In this article, the occurrence of dead core in catalytic particles containing immobilized enzymes is analyzed for the Michaelis–Menten kinetics. An assessment of numerical methods is performed to solve the boundary value problem generated by the mathematical modeling of diffusion and reaction processes under steady state and isothermal conditions. Two classes of numerical methods were employed: shooting and collocation. The shooting method used the ode function from Scilab software. The collocation methods included: that implemented by the bvode function of Scilab, the orthogonal collocation, and the orthogonal collocation on finite elements. The methods were validated for simplified forms of the Michaelis–Menten equation (zero-order and first-order kinetics), for which analytical solutions are available. Among the methods covered in this article, the orthogonal collocation on finite elements proved to be the most robust and efficient method to solve the boundary value problem concerning Michaelis–Menten kinetics. For this enzyme kinetics, it was found that the dead core can occur when verified certain conditions of diffusion–reaction within the catalytic particle. The application of the concepts and methods presented in this study will allow for a more generalized analysis and more accurate designs of heterogeneous enzymatic reactors.  相似文献

Diffusion problem with variabale diffusion coefficient in a spherical biological system is investigated. Also included in this study is the biological reaction of the Michaelis-Menten type. The problem formulated consists of a highly nonlinear differential equation which, however, can be efficiently solved by the orthogonal collocation method on a digital computer. The effects of the dimensionless governing parameters on the transient and steady state concentration responses are parametrically examined for the diffusion system with and without biological reaction.  相似文献

This paper proposes a numerical approximation method for computational optimal control of a time fractional convection-diffusion-reaction system. The proposed method involves discretizing the spatial domain by finite element method, approximating the admissible controls by control parameterization, and then obtaining an optimal parameter selection problem which can be solved by numerical optimization algorithms such as sequential quadratic programming. Specifically, an implicit finite difference method is employed to solve the time fractional system, and the sensitivity method for gradient computation in integer order optimal control problems is adjusted to the fractional order case. Simulation results demonstrate the validity and accuracy of the proposed numerical approximation method.  相似文献

The aim of this study is the optimization of a product-driven self-cycling bioprocess and presentation of a way to determine the best possible decision variables out of a set of alternatives based on the designed model. Initially, a product-driven generalized kinetic model, which allows a flexible choice of the most appropriate kinetics is designed and analysed. The optimization problem is given as the bi-objective one, where maximization of biomass productivity and minimization of unproductive loss of substrate are the objective functions. Then, the Pareto fronts are calculated for exemplary kinetics. It is found that in the designed bioprocess, a decrease of emptying/refilling fraction and an increase of substrate feeding concentration cause an increase of the biomass productivity. An increase of emptying/refilling fraction and a decrease of substrate feeding concentration cause a decrease of unproductive loss of substrate. The preferred solutions are calculated using the minimum distance from an ideal solution method, while giving proposals of their modifications derived from a decision maker’s reactions to the generated solutions.  相似文献

The optimal temperature policy that maximizes the time-averaged productivity of a continuous immobilized enzyme packed bed reactor is determined. This optimization study takes into consideration the enzyme thermal deactivation with substrate protection during the reactor operation. The general case of reversible Michaelis-Menten kinetics under constant reactor feed flow rate is assumed. The corresponding nonlinear optimization problem is solved using the calculus of variations by applying the disjoint policy. This policy reduces the optimization problem into a differential-algebraic system, DAE. This DAE system defines completely the optimal temperature-time profiles. These profiles depend on the kinetic parameters, feed substrate concentration, operating period, and the residence time and are characterized by increasing form with time. Also, general analytical expressions for the slopes of the temperature and residual enzyme activity profiles are derived. An efficient solution algorithm is developed to solve the DAE system, which results into a one-dimensional optimization problem with simple bounds on the initial feed temperature. The enzymatic isomerization of glucose into fructose is selected as a case study. The computed productivities are very close to that obtained by numerical nonlinear optimization with simpler problem to solve. Moreover, the computed conversion profiles are almost constant over 90% of the operating periods, thus producing a homogeneous product.  相似文献

The objective of this contribution is the design of optimal feeding strategies for fed-batch bioprocesses, where complex dynamic models with input and state constraints are present. For the solution of this dynamic optimization problem a transformation to a finite dimensional optimization problem is made using piecewise linear control profiles. The optimization of these profiles is performed by a sequential approach, that includes an ODE solver for the solution of the model ODE's. Further an adaptive mesh selection algorithm was investigated for an appropriate discretization of the control profiles. The implementation of the resulting optimal feeding profiles is shown for a process example, namely the production of nikkomycin by Streptomyces tendae. This implementation uses a hierarchical process control framework, that consists of components for process monitoring, state estimation, and trajectory control.  相似文献

In enzymatic lysis of yeast for the recovery of intracellular proteins, the rupture of whole cells is caused by the action of a lytic system consisting primarily of protease and glucanase. A first-principles mechanism for the lytic reaction based on a two-layer model of the wall structure and a burst model for the disruption of cells is pre sented. The fed-batch model results in a dynamic optimization problem, with the enzymes, activities being the control variables. Orthogonal collocation is applied to discretize the state equations, and the resulting non linear program is solved using successive quadratic pro gramming to determine the enzyme and protease inhibitor add-in rates and pH control profiles that maximize the recovery of intracellular protein. Applying the proposed approach, optimal profiles were determined such that a significant increase of the production of proteins in a fed-batch reactor is realized. Also, the optimal control policies in a series of continuous-flow stirred tank reactors (CFSTRs) are determined.  相似文献

A new method for continuous cultivation of microbes, called adaptastat, is described. It involves automatic adaptation of microbial cultures to their maximum feeding rates and avoids substrate accumulation. The state of the culture is estimated at intervals by briefly (and markedly) decreasing the substrate feeding rate and measuring, via the dissolved oxygen response, the time taken to exhaust the residual substrate. The method has been exemplified by cultivating Escherichia coli on single carbon sources (glucose, acetate, succinate, and fully deuteriated medium based on deuteriated succinate) and also by simultaneous limitation of two feeding channels (succinate/acetate and glucose/ammonium chloride). Several possible applications of the adaptastat technology are presented. The method provides an efficient means of labelling microbial components and products with stable isotopes. In particular, adaptastat technology can be used to adapt disabled bacterial strains to the use of simple, inexpensive substrates. It can also be used more generally in the study of microbial cell cultures, for example for the determination of maximum specific growth rates and the stoichiometric ratio of utilisation of two nutrients in conditions of simultaneous limitation.  相似文献

In Part I(1) of this research a complex model was obtained for describing the ethanol fermentation in a cascade reactor. This complexity is due to both the nonlinearity and the large scale representation. Based on techniques of partitioning and relaxation, a decentralized successive approximation method is developed for static optimization. The influence of the way of fermentation during continuous culture in multistage fermentors is studied in the case of a double inhibition of cell growth and product formation by both substrate and final product. The optimal number of reactors is discussed with respect to the strength of the ethanol inhibition, while the interest of head feeding or distributed feeding is evaluated in relation to the strength of substrate inhibition.  相似文献

A simple, accurate model capable of predicting cell growth and methanol utilization during the mixed substrate fed-batch fermentation of MutS recombinant Pichia pastoris was developed and was used to design an exponential feeding strategy for mixed substrate fed-batch fermentation at a constant specific growth rate. Mixed substrate feeding has been shown to boost productivity in recombinant fed-batch culture of P. pastoris, while fixed growth rate exponential feeding during fed-batch culture is a useful tool in process optimization and control.  相似文献

A generic methodology for feeding strategy optimization is presented. This approach uses a genetic algorithm to search for optimal feeding profiles represented by means of artificial neural networks (ANN). Exemplified on a fed-batch hybridoma cell cultivation, the approach has proven to be able to cope with complex optimization tasks handling intricate constraints and objective functions. Furthermore, the performance of the method is compared with other previously reported standard techniques like: (1) optimal control theory, (2) first order conjugate gradient, (3) dynamical programming, (4) extended evolutionary strategies. The methodology presents no restrictions concerning the number or complexity of the state variables and therefore constitutes a remarkable alternative for process development and optimization. A simple structured mathematical model coupled with a methodology of state and parameter estimation is developed for lipase production by Candida rugosa in batch fermentation. The model describes the system according to the following qualitative observations and hypothesis: Lipase production is induced by extracellular oleic acid present in the medium. The acid is transported into the cell where it is consumed, transformed, and stored. Lipase is excreted to the medium where it is distributed between the available oil-water interphase and aqueous phase. Cell growth is modulated by the intracellular substrate concentration. Model parameters have been determined and the whole model validated against experiments not used in their determination. The estimation problem consists in the estimation of three state variables (biomass, intra- and extracellular substrate) and two kinetic parameters by using only the on-line measurement provided by exhaust gas analysis. The presented estimation strategy divides the complex problem into three subproblems that can be solved by stable algorithms. The estimation of biomass (X) and the specific growth rate (mu), is achieved by a recursive prediction error algorithm using the on-line measurement of the carbon dioxide evolution rate. mu is then used to perform an estimation of intracellular substrate and the other kinetic parameter related to substrate transport (A) by an adaptive observer. Extracellular substrate is then evaluated by means of the estimated values of intracellular substrate and biomass through the material balance of the reactor. This paper analyzes feeding strategies in a sequential batch reactor (SBR) with the objective of reaching a given (low) substrate level as quickly as possible for a given volume of water. Inside the SBR, several species compete for a single substrate, which leads to a minimal time control problem in which the control variable is the feeding rate. Following Gajardo et al. (2008) SIAM J Control Optim 47(6):2827–2856, we allow the control variable to be a bounded measurable function of time combined with possible impulses associated with instantaneous dilutions. For this problem, the extremal trajectories of the singular arc type are characterized as the strategies used to maintain the substrate at a constant level. Since this optimization problem is difficult to solve, this characterization provides a valuable tool for investigating the optimality of various feeding strategies. Our aim is thus to illustrate the use of this tool by proposing potential optimal feeding strategies, which may then be compared with other more intuitive strategies. This aim was accomplished via several numerical experiments in which two specific strategies are compared.  相似文献

The dynamics of pure and simple competition between two microbial species are examined for the case of interaction arising in a distributed and nonstagnant environment. The environment is modeled as a tubular reactor. It is shown that for relatively small values of the dispersion coefficient (i.e., for small, but nonzero, backmixing of the medium), the two competing populations can coexist in a stable steady state. It has been assumed that the species grow uninhibited and that if there are maintenance requirements they are satisfied from endogenous sources. From numerical studies it has been found that a necessary condition for coexistence is that the net specific growth rate curves of the two competitors cross each other at a positive value of the concentration of the rate-limiting substrate. The model equations have been numerically solved by using the methods of orthogonal and spline collocation.  相似文献

The biopharmaceutical industry continuously seeks to optimize the critical quality attributes to maintain the reliability and cost-effectiveness of its products. Such optimization demands a scalable and optimal control strategy to meet the process constraints and objectives. This work uses a model predictive controller (MPC) to compute an optimal feeding strategy leading to maximized cell growth and metabolite production in fed-batch cell culture processes. The lack of high-fidelity physics-based models and the high complexity of cell culture processes motivated us to use machine learning algorithms in the forecast model to aid our development. We took advantage of linear regression, the Gaussian process and neural network models in the MPC design to maximize the daily protein production for each batch. The control scheme of the cell culture process solves an optimization problem while maintaining all metabolites and cell culture process variables within the specification. The linear and nonlinear models are developed based on real cell culture process data, and the performance of the designed controllers is evaluated by running several real-time experiments.  相似文献

Reverse engineering is the problem of inferring the structure of a network of interactions between biological variables from a set of observations. In this paper, we propose an optimization algorithm, called MORE, for the reverse engineering of biological networks from time series data. The model inferred by MORE is a sparse system of nonlinear differential equations, complex enough to realistically describe the dynamics of a biological system. MORE tackles separately the discrete component of the problem, the determination of the biological network topology, and the continuous component of the problem, the strength of the interactions. This approach allows us both to enforce system sparsity, by globally constraining the number of edges, and to integrate a priori information about the structure of the underlying interaction network. Experimental results on simulated and real-world networks show that the mixed discrete/continuous optimization approach of MORE significantly outperforms standard continuous optimization and that MORE is competitive with the state of the art in terms of accuracy of the inferred networks.  相似文献
