The effect of Chromium VI on leaf water potential (w), solutepotential (a), turgor potential (p) and relative water content(RWC) of primary and first trifoliatc leaves of Phaseolus vulgarisL. was studied under normal growth conditions and during anartificially induced water stress period in order to establishthe possible influence of this heavy metal on the water stressresistance of plants. Plants were grown on perlite with nutrientsolution containing 0, 1•0, 2•5, 5•0 or 10•0µg cm–3 Cr as Na2Cr2O7.2H2O. The effect of Cr onwater relations was highly concentration dependent, and primaryand first trifoliate leaves were affected differently. The growthreducing concentrations of Cr (2•5, 5•0 and 10•0µg cm–3) generally decreased s and w and increasedp in primary leaves. The 1•0 µg cm–3 Cr treatmentdid not affect growth, but altered water relations substantially:in primary leaves w and p were increased and s decreased, whilein trifoliate leaves the effect was the opposite. All Cr treatedplants resisted water stress for longer than control plants.The higher water stress resistance may be due to the lower sand to the increased cell wall elasticity observed in Cr VItreated plants. The difference in electrical potential between the xylem exudateof isolated roots of 1-week-old maize seedlings and the culturesolution surrounding the roots has been determined togetherwith the concentrations of potassium, calcium, chloride, andsulphate in the xylem sap. The difference in electro-chemicalpotential (µ) for these ions has been calculated fromthe measurements. The effects on µ of varying the saltstatus of the roots, the composition of the culture solutionand of 2-4-dinitrophenol have been examined. µ for potassiumand chloride was always positive, implying that movement ofthese ions to the xylem sap was under metabolic constraint.However, pretreatment of the maize seedlings with potassiumchloride and increasing levels of dinitrophenol in the culturesolution over the exudation period caused little or no significantchange in µ for potassium although the rate of movementof ions to the xylem was substantially reduced. For calcium,µ was negative with roots in dilute culture solutionsin the absence of dinitrophenol, implying that calcium couldenter the xylem by passive diffusion. Addition of dinitrophenolchanged the sign of µ and thus brought about an apparentlyactive transport of calcium.Only in the absence of competinganions was the rate of entry of chloride significantly correlatedwith µ, and no such relationship was found for potassiumor calcium individually. These results encourage doubt as towhether measurements of µ provide a valid basis for decidingto what extent the movement of individual ions depends on specific‘active’ or ‘passive’ transport processes.Thedifference in electrical potential appears to be a characteristicof the living root and to depend on the total concentrationof ions in the external solution and on their rate of transferinto the xylem sap.  相似文献

Treatments were applied to vary C and N availability in Alnusglutinosa L. and plant growth, nodule activity (including acetylenereduction) and amino acid composition of the xylem sap weremeasured. Removing the buds, a sink for N, caused a decreasein nodule activity. Flushing root systems daily with 100% O2destroyed nitrogenase activity and substantially decreased theamount of citrulline in the xylem sap. The amino acid compositionof xylem saps also altered according to the mode of N nutrition.In plants fed , xylem sap composition was similar to N2-fixing plants, however, when plants were fed, citrulline content increased. The assimilation and subsequent distribution of nitrate wasfollowed in an experiment in which labelled 15 was added to the base of plant pots. After 12 h7% of root N was from applied 15 and this increased to 75% at 7 d; substantial enrichment ofN from 15 also occurred in stems, buds and leaves. After 7 d, 3.5% of nodule N was from15, consistent with some N being supplied by recycling of shoot N. Xylem saps were alsocollected and after 12 h, glutamate and aspartate were enrichedwith 15N to 53% and 37% increasing after 7 d to 80% and 49%,respectively. Citrulline content of the xylem sap increasedfrom 3 to 9 µmol cm–3 following addition of 15 and at 7 d 80% of the N in the citrullinehad been derived from 15N. It is hypothesized that the growthand activity of A. glutinosa root nodules is sensitive to theN status of the plant and that the level of citrulline (or otheramino acids) returning to the nodules may feed back to regulatenodule growth and activity. Smith, J. R. 1987. Potassium transport across the membranesof Chara. II. 42K fluxes and the electrical current as a functionof membrane voltage.—J. exp. Bot. 38: 752–777. The current required to clamp the trans-membrane voltage ofinternodal cells of Chara australis at different levels wasmeasured simultaneously with either the 42K influx or efflux.Examination of the voltage-dependence of the ratio of the electricalcurrent to the unidirectional tracer fluxes yielded no evidenceof any amplification of the electrical driving force on theK+ ions. There was thus no evidence for the interaction of K+ions with themselves or any other species during their passageacross the membrane. These measurements allow the determinationof , the fraction of the electrical current carried by K+ ions.When the external [K+] = 10 mol m–3, the average valueof was 0?85 for Vm > –125 mV and 07?5 for Vm <–150 mV. When the external [K+] = 0?1 mol m–3, was 0?6 for Vm < –80 mV and 0?1 for Vm > –250mV. It was also found that the conductance associated with K+transport was inhibited by hyperpolarization. Shelp, B. J. 1987. The composition of phloem exudate and xylemsap from broccoli (Brassica oleracea var. italica) suppliedwith NH+4, NO3 or NH4NO3.—J. exp. Bot. 38: 1619–1636. The detailed composition of xylem sap and exudate from stemincisions of attached inflorescences of broccoli (Brassica oleraceavar. italica) was compared in plants supplied with NH+4, NO3or NH4NO3. A phloem origin for the exudate was suggested fromthe high levels of sugars (71–133 mg cm-3), amino acids(8·1-26·7 mg cm3) and K. (2·3–3·8mg cm3), the low levels of NO3 and Ca, the high C: N (w/w) ratios(8·3–33), and the alkaline pH (7·2–7·3).In contrast, the xylem sap was mildly acidic (pH 5·6–6·0),and possessed lower levels of all organic and inorganic solutesbut NO3 and Ca, and lower ratios of K: Ca, Mg: Ca and C: N (0·6–4·4). Glutamine was the predominant o-phthalaldehyde-reactive aminocompound in both transport fluids with the next most abundantamino acids dependent on sap type and N-form. Together witharginine, -aminobutyric acid, which was found only in the xylemstream, was enhanced by NH+4compared to NO3 -nutrition suggestingthat glutamate metabolism was stimulated in the roots. Underlimiting N the amino acid concentrations in the transport fluidswere greater with NH+4 than with NO3. NO3 reduction occurredin both the root and shoot with the latter site predominatingover the entire N range (0-300 mol m3). Even though the compositionof nitrogenous solutes in the xylem was dependent on cultivarand N source, the composition of the phloem streams supplyingthe developing inflorescence was relatively unaffected. The data on the element composition of organs and phloem sapare interpreted to suggest that, in spite of the restrictedmobility of some elements such as B and Mn, a significant proportionof their total supply to developing sinks is carried in thephloem stream. Plants of the C3 species Phaseolus vulgaris and Xanthium strumariumand of the C4 salt-sensitive Zea mays and the C4 halophyte Atriplexhalimus were grown with and without NaCl salt-stress at normal(340 µl I–1) and at high (2500 µl I–1)ambient CO2. In all four species growth (dry weight increment)was enhanced by CO2 supplementation. The relative response wasgreater in the salinized than in the control plants. Charge balance and intracellular pH regulation were studiedin Phaseolus vulgaris grown in water culture on or as N source, or nodulated and in N-free medium. Seedlings and 36-d-old plants were analysed for total P, C, organic N and S and ash alkalinity;xylem sap was analysed for mineral ion content and amino acids,amides and dicarboxylates. Both water uptake and H+ or baseexcreted during water culture were measured. It was shown that in -grown plants, H+ excretedwas related directly to the uptake and assimilation of ; there was no cation uptake associated with netH+ exchange. All shoot N was supplied as organic N, mainly glutamine,so that shoot pH perturbation would be limited to uronate productionin situ. This could be regulated by both shoot reduction and the transport of OH-generating carboxylatesin the xylem. In plants on , the great majority of reduction occurred in the root and most of the OHproduced was converted to -COO, transported up the xylemand stored, so that the leaves in particular had a high ashalkalinity (20-fold that in the roots). Some OH, however,was excreted. Nodulated plants had a low N content, compared with the others,indicating a low rate of N2-fixation for the earlier periodof water culture. At harvest, however, the xylem sap organicN content was comparable to that of plants. H+ excretion was in excess of that required for N assimilation.It was concluded that in these plants some net H+/cation exchangehad occurred which may have been a response to low nutrient(i.e. nitrogen) status. Plants of two genotypes of Lolium perenne L. cv. S23 and a L.perenne ? L. multiflorum Lam. hybrid cv. Augusta were grownin flowing solution culture. N was suppled in one treatmentat 10 mmol m–3 NO–3 throughout (HN), and in another(LN) the N supply was terminated after 10 d for 11 d. When was re-supplied both LN and HN plants were leftentire or defoliated. The two genotypes showed similar responsesto all treatments. The concentration of N in shoot dry matterdeclined from 4.4% to 2.0% and in the root from 2.8% to 1.0%over the 11 d of N deprivation, with 95% of the initially present being assimilated during this period. LN plantsassimilated 10% more of their total uptake than did HN plants. The in vitro nitrate reductase activity(NRA) was 10- to 50-fold higher in the youngest fully-expandedleaves than in roots and declined in the leaves during N deprivation.Between 2–6 d after defoliation, there was a large increasein NRA in leaves of HN (but not LN) plants. After defoliationof HN plants, net uptake from 10 mmol m–3 declined to negligible levels within 15 h, but in defoliatedLN plants it increased to levels similar to those of entireHN plants (15–20 µmol h–1 g–1 fr. wt.root) within 8 h. When was re-supplied to entire LN plants, uptake of increased to levels similar to those of entire HN plants within 2.3 h, butdid not markedly exceed that of HN plants for at least 10 h.Net uptake of by LN plants during depletion of stirred static nutrient solutions containing 1.0 mol m–3 lagged behind that by HN plants by 70–100 min, but the maximum unit absorption rate was similar for LNand HN plants (5–7 µmol h–1 g–1 fr.wt. root). The nature of the short-term demand for uptake following recovery from the stresses of defoliation andN starvation is discussed. This study evaluated the possible role of hydrogen peroxide(H2O2) in the acclimation of a Mediterranean shrub, Cistus albidusL., to summer drought growing under Mediterranean field conditions.For this purpose, changes in H2O2 concentrations and localizationthroughout a year were analysed. H2O2 changes in response toenvironmental conditions in parallel with changes in abscisicacid (ABA) and oxidative stress markers, together with ligninaccumulation, xylem and sclerenchyma differentiation, and leafarea were also investigated. During the summer drought, leafH2O2 concentrations increased 11-fold, reaching values of 10µmol g–1 dry weight (DW). This increase occurredmainly in mesophyll cell walls, xylem vessels, and sclerenchymacells in the differentiation stage. An increase in ABA levelspreceded that of H2O2, but both peaked at the same time in conditionsof prolonged stress. C. albidus plants tolerated high concentrationsof H2O2 because of its localization in the apoplast of mesophyllcells, xylem vessels, and in differentiating sclerenchyma cells.The increase in ABA, and consequently of H2O2, in plants subjectedto drought stress might induce a 3.5-fold increase in ascorbicacid (AA), which maintained and even decreased its oxidativestatus, thus protecting plants from oxidative damage. Afterrecovery from drought following late-summer and autumn rainfall,a decrease in ABA, H2O2, and AA to their basal levels (60 pmolg–1 DW, 1 µmol g–1 DW, and 20 µmol g–1DW) was observed. This work aimed to study the impacts of acquisition and assimilationof various nitrogen sources, i.e. NO3, NH4+ or NH4NO3,in combination with gaseous NH3 on plant growth and acid-basebalance in higher plants. Plants of C3 Triticum aestivum L.and C4 Zea mays L. grown with shoots in ambient air in hydroponicculture solutions with 2 mol m–3 of nitrogen source asNO3, NH4+ or NH4NO3 for 21 d and 18 d, respectively,had their shoots exposed either to 320 µg m–3 NH3or to ambient air for 7 d. Variations in plant growth (leaves,stubble and roots), and OH and H+ extrusions as wellas the relative increases in nitrogen, carbon and carboxylatewere determined. These data were computed as H+/N, H+/C, (C-A)/N,and (C-A)/C to analyse influences of different nitrogen sourceson acid-base balance in C3 Triticum aestivum and C4 Zea maysplants. Root growth in dry weight gain was significantly reduced bytreatment with 320 µg m–3 NH3 in Triticum aestivumand Zea mays growing with different N-forms, whereas leaf growthwas not significantly affected by NH3. In comparison with C3Triticum aestivum, non-fumigated C4 Zea mays had low ratiosof OH/N in NO3–3-grown plants and of H+/N in NH4+- and NH 4NO3-grown plants. Utilization of NH3 from the atmospherereduced both the OHN ratios in NO3 -grown plantsand the H+/N ratio in NH4+ - and NH4NO3 -grown plants of bothspecies. Furthermore, Zea mays had higher ratios of (C-A)/Nin NH4+ - and NH4NO3-grown plants than Triticum aestivum. Thismeans that C4 Zea mays had synthesized more organic anion perunit increase in organic N than C3 Triticum aestivum plants.Within both species, different nitrogen sources altered theratios of (C-A)/N in the order: NH4NO3>NH4+>NO3.Fumigation with NH3 increased organic acid synthesis in NO3- and NH4+ - grown plants of Triticum aestivum, whereas it decreasedorganic acid synthesis in Zea mays plants under the same conditions.Furthermore, these differences in acid-base regulation betweenC3 Triticum aestivum and C4 Zea mays plants growing with differentnitrogen sources are discussed. Nitrate Uptake and Assimilation following Nitrate Deprivation   总被引:2,自引:0,他引:2  
Upon first exposure to , the uptake and reduction capacities of dark-grown corn (Zea maysL.) roots are initially low, but increase markedly within 6h. The development of the accelerated uptake rate appears to be substrate ‘induced’ as is reductase (NR), the first enzyme in the assimilatory pathway. However, the ‘induction’of uptake is independent of NR induction. The effect of deprivation was studied to determine the role of endogenous on subsequent uptake and reduction. Corn roots were ‘induced’ for 24 h in 0–5 mol m–3 nutrient solution and then exposed for 0 to 32 h to -free nutrient solution. Uptake and reduction of were determined periodically by exposing sets of roots to a1 h pulse of 0.5 mol m–3 . Neither uptake (4.57 µmol root–1 h–1)nor the percentage of absorbed reduced (27%) was changed significantly (P 0.05) by exogenous deprivation. However, the estimated ‘induced’ componentof uptake decreased significantly (50% after 32 h). Concurrently, the ‘non-induced’ basal componentof uptake increased. Previously accumulated decreased from 23 to 4.5 µmol root–1 after 32 h of exogenous deprivation. Nearly equivalent quantities of endogenous were used for translocation and reduction during deprivation. During each 1 h pulse, the amounts of translocation and net efflux of to the uptake solution were similar. Net efflux of was strongly correlated (r = 0.991) to the amount of endogenous . The remaining endogenous and its rate of utilization were apparently sufficient to minimize a rapid declineor complete loss in both the ‘induced’ uptake state and the rate of in vivo assimilation. Effects of light flux density (LFD) during growth and uptakeassay on induction of transport system and kinetics of transport were studied using the Azolla pinnata-Anabaena azollae association (Azolla). Theinduction and uptake kinetics of the transport system were determined using an automated system that measuredthe NO3 concentration in the growth medium as a function oftime, using an on-line high performance liquid chromatograph(HPLC) with a UV-VIS detector. Full induction of the transport system required about 1.5 to 2.0 h and occurred without any apparent lag phase,regardless of the LFD provided. The level of induction of transport of Azolla grown at 600 µmol m–2s–1 LFD was higher than for that grown at 100 µmolm–2 s–1. Similarly, 600 µmol m–1 s–1LFD during the assay resulted in a higher level of inductionthan did 100 umol m–2 s–1. An increase in the LFDeither during the growth or the assay period increased the uptake rate; however, an increase in LFD duringthe latter period had greater effect. Azolla grown and assayedat 600 umol m–2 s–1 had the highest uptake rate. The uptake rate at 50 cm3 m–3ambient CO2 concentration was initially higher than at 305 cm3m–3, but the uptake rate decreased rapidly with time andeventually dropped below that at 305 cm3 m–3 CO2. Thesedata suggest that the energy required for transport in Azolla may bypass the photosynthetic CO2 fixationand carbon-cycling. Charge balance and intracellular pH regulation were studiedin Ricinus communis grown in water culture with 1–0 molm–3 or l-0 mol m –3 as sole N source. Seedling and 70-d-old plant parts were analysed for total P, C, organic N and S, and ash alkalinity;xylem and phloem saps were analysed for mineral ion content,and amino acids, amides, and dicarboxylates. Excreted H+ andbase were also measured. It was shown that in -N plants, H + produced and excreted directly bythe roots accounted for all net —COOH produced in theplant, but not for cation uptake by net H+ exchange. IntracellularpH perturbation in the shoot was regulated partly by reduction and partly by the transportof OH- -generating dicarboxylates in the xylem. Phloem sap hadthe capacity to transport organic N and carboxylates excessto shoot requirements back to the root. In -N plants, 60% of total reduction occurred in the root, and 70% of all OH- produced by root and reduction was excreted directly as base. Very little —COOfrom root reduction was stored in the root: most was transported to the shoot as xylem dicarboxylateand stored in the shoot. Of the OH- produced from shoot and reduction, 40% was stored as shoot carboxylate: the phloem saphad the capacity to transport the rest back to the roots whereit was excreted as base. Four species of seagrasses, Halophila stipulacea, Thalassodendronciliatum, Halodule uninervis, and Syringodium isoetifolium,were investigated for their ability to utilize and CO2 as exogenous carbon sources for photosynthesis. Ratesof photosynthesis were measured as rates of O2 evolution ina closed system in which the pH was continuously controlled.A computer program was written to calculate the concentrationsof different carbon species as a function of pH and other specifiedexperimental conditions. Bicarbonate as well as CO2 were readily assimilated by all fourseagrass species. Saturating concentrations of , at saturating light intensities, were 0.5–1.8 mM dependingon the species. Rates of photosynthesis under such conditionswere 0.1–0.55 µmol O2 min–1 mg–1 chlorophyll.At saturating CO2 concentrations, i.e. 0.5–1.3 mM, ratesof photosynthesis were 0.22–1.4 µmol CO2 min–1mg–1 chlorophyll. Photosynthetic rates in each specieswere considerably higher when CO2 rather than was supplied at saturating concentrations. The concentration of in natural seawater was found to be saturating, and that of CO2 insufficient forconsiderable photosynthetic rates in these plants under thegiven conditions It was thus concluded that is the major carbon source for photosynthesis in seagrasses.  相似文献

The euryhaline charophyte Lamprothamnium papulosum (Wallr.)J. Gr. was adapted to media with decreasing salinities rangingfrom 550 to 0 mosmol kg–1. Vegetative plants grown inmedia with osmotic pressures (0) in the range of 550 to 130mosmol kg–1 maintained a constant turgor pressure () at309 + 7 mosmol kg–1. The ions K+, Na+ and Cl–, werethe predominant solutes in the vacuole. Changes in their concentrationsaccount for the variation in internal osmotic pressure (1) with,0. The divalent ions Mg2+, Ca2+ and were also present in significant amounts, but their concentrationsdid not alter with changes in, 0. In cells subjected to hypo-osmotic shock the regulation of was incomplete. The turgor pressure increased from 302 to 383mosmol kg–1. The first rapid response to the sudden decreasein 0 was a loss of K+ and Cl. In contrast to the decreasein ionic concentrations an accumulation of sucrose occurredwhich could account for the increase of . The increase in sucroseconcentration started 24 to 48 h after the downshock and reachedits highest value after 3 to 4 weeks. The sucrose concentrationin the vacuole was up to 320 mol m–3. During this timethe ionic content continued to decrease but did not counterbalancethe sucrose concentration sufficiently to regain the original. High sucrose levels accompanied by an enhanced were also observedduring the period of fructification (sexual reproduction: formationof antheridia and oogonia) in Lamprothamnium kept under conditionsof constant salinity. It is concluded that high sucrose content and elevated arecharacteristic of sexual reproduction in this charophyte. Previous single-cell studies on the upper epidermis of barleyleaves have shown that cells differ systematically in theirsolute concentrations depending on their location relative tostomatal pores and veins and that during NaCl stress, gradientsin osmotic pressure () develop (Fricke et al., 1995, 1996; Hinde,1994). The objective of the present study was to address thequestion to which degree these intercellular differences insolute concentrations and it are associated with intercellulardifferences in turgor or water potential (). Epidermal cellsanalysed were located at various positions within the ridgeregions overlying large lateral or intermediate veins, in thetrough regions between those veins or in between stomata (i.e.interstomatal cells). Turgor pressure of cells was measuredusing a cell pressure probe, and of extracted cell sap wasdetermined by picolitre osmometry. For both large and intermediatelateral veins, there were no systematic differences in turgorbetween cells located at the base, mid or top of ridges, regardlessof whether plants were analysed at low or high PAR (10 or 300–400µmol photons m–2 s–1). However, turgor withina ridge region was not necessarily uniform, but could vary byup to 0.14 MPa (1.4 bar) between adjacent cells. In 60 out of63 plants, turgor of ridge cells was either slightly or significantlyhigher than turgor of trough (lowest turgor) or interstomatalcells (intermediate turgor). The significance and magnitudeof turgor differences was higher in plants analysed under highPAR or local air flow than in plants analysed under low PAR.The largest (up to 0.41 MPa) and consistently significant differencesin turgor were found in plants treated for 3–9 d priorto analysis with 100 mM NaCl. For both NaCl-treated and non-treated(control) plants, differences in turgor between cell types weremainly due to differences in since differences in were negligible(0.01–0.04 MPa). Epidermal cell , in NaCl-treated plantswas about 0.38 MPa more negative than in control plants dueto higher . Turgor pressures were similar. Following a suddenchange in rooting-medium or air humidity, turgor of both ridgeand trough cells responded within seconds and followed the sametime-course of relaxation. Plants of four cultivars of Phaseolus vulgaris L. differingin drought resistance were grown in pots under greenhouse conditionsand prior to flowering water was withheld from the pots untilthe mid-day transpiration rate reached values below 1.0 µgH2O cm–2 s–1 (designated the ‘drought’stage). At this point leaves were harvested on 3 or 4 occasionsover 24 h to determine the abscisic acid (ABA) concentration,total water potential (), solute potential (1) and turgor potential(p). Results showed that values of , 1, and p differed between cultivarswhen they reached the ‘drought’ stage. The stomatalsensitivity to changes in and p, was as follows: Michoacán12A3 > Negro 150 Cacahuate 72 > Flor de Mayo. These datacorrelated well with the pattern of drought resistance reportedfor the cultivars. ABA accumulation at the ‘drought’ stage differedbetween cultivars at each sampling time, but overall differencesin ABA level between cultivars were not significant. ABA levelsdid not, therefore, correlate with the drought resistance propertiesreported for the cultivars. Evaluation of the 15N naturalabundance method and xylem sap analysis for assessing N2 fixationof understorey legumes in jarrah (Eucalyptus marginata Donnex Sm.) forest in S.W. Australia.—J. exp. Bot 38: 1446–1458. Nodulated seedlings of Acacia pulchella, A. alata and A. extensawere grown in glasshouse sand culture under a range of levels(0–16 mol m3) of nitrate, supplied as 15NO3, or as unenrichedlaboratory grade nitrate (15N value 5·5%o). Nitrate at8·0 mol m 3 or above was highly inhibitory to growthof all species. Using 15N dilution analysis of the 15N enrichedcultures to measure symbiotic dependency, it was shown that15N values of the parallel unenriched cultures increased innear linear fashion from close to zero in fully symbiotic plantsto values close to that of the supplied NO3 in plants experiencingnitrate levels (4·0 mol m3 or above) inhibiting N2 fixationby over 90%. Xylem sap analyses (0·4 mol m3 NO3 treatments)showed asparagine as the major nitrogenous solute, relativelylittle spill-over of free nitrate, and no evidence of majorshifts in balance of amino compounds with increasing dependenceon nitrate. This essentially invalidated use of the techniqueas a field assay for N2 fixation by the species. 15N values for total N of soil sampled at 64 widely distributedsites in jarrah forest ranged from – 2·15 to +5·4(mean +2·1). Comparable values for soil mineral N (NH+4and NO3) were +0·3 to + 14·2 (mean +5·1).15N values of the total plant N of the legumes and of non-N2-fixingreference species were also highly variable between sites, withlittle evidence of reference plant N accurately reflecting the15N abundance of soil nitrogen, or of visibly well nodulatedlegume components showing consistently lower 15N values thantheir companion reference plants. At one site it was possibleto compare 15N values of first season seedling legumes withpreviously published estimates of their progressive N2 fixationusing C2H2 reduction assays. It was concluded that heterogeneity in 15N discrimination ofsoil within the ecosystem precluded effective use of the 15Nnatural abundance technique for assessing legume N2 fixation. Thomas, H. 1987. Physiological responses to drought of Loliumperenne L.: Measurement of, and genetic variation in, waterpotential, solute potential, elasticity and cell hydration.—J.exp. Bot. 38: 115–125. Clonally-replicated genotypes of Loiium perenne L. were grownin a controlled environment. Leaf water potential (w) osmoticpotential (s), turgor potential (p = ws), elasticity(E), leaf hydration (g water per g dry matter, H) and numberof green leaves per tiller (NGL) were measured before and duringa 42 d drought treatment. A simplified method of estimating E (at w < 1?0 MPa) usingonly six measurements was developed to permit a measurementrate of 8 leaves per hour. Measurement errors in all characterswere 3% or less. During drought, w and s (at w = 0?5 MPa) decreased significantly,p and E increased significantly, and H decreased slightly. Plantsize during drought was negatively correlated with s, and Hand positively correlated with p, osmotic adjustment, E andNGL. Measurements made on the genotypes before draughting didnot give a reliable indication of their physiological conditionafter adaptation to drought. Genetically controlled variation (‘broad sense heritability’)of drought-adapted plants for E was 15%, w 23%, s, 34%, p, 35%,H 34% and NGL 64%. The possibilities for, and effectivenessof, divergent selection of genotypes with high and low expressionof the characters are discussed. The fibre length L and average helical angle of microfibrilorientation in fibres from successive growth rings in both normaland ‘rubbery’ wood of stems of apple var. Lord Lambournehave been measured. It has been shown that the normal relationship,L = A+B cot , does not hold for ‘rubbery wood’.There is a tendency for the helix to remain flat, the angle fairly constant independently of fibre length. This, togetherwith abnormalities in the lignin chemistry, causes the rubberywood to have low tensile strength and high extensibility.  相似文献
