The ubiquitous occurrence of series of biosynthetically related plant secondary compounds within individual species has given rise to the suggestion that such multiplicity is adaptive; one possible mechanism that would serve to maintain such within-plant diversity is analog synergism. In a series of experiments, we provide evidence that synergism may account for the presence of multiple structurally related furanocoumarins in apiaceous plants. The black swallowtail, Papilio polyxenes, feeds exclusively on plant species containing furanocoumarins. Growth of larvae fed parsley leaves treated with both xanthotoxin and angelicin, two furanocoumarins that co-occur widely in swallowtail hostplants, was significantly slower than that of larvae fed leaves with an equimolar concentration of either xanthotoxin or angelicin. A multivariate combination of growth, food consumption and frass excretion differed significantly between larvae fed leaves treated with both xanthotoxin and angelicin and larvae fed leaves treated with angelicin alone. In addition, we measured rates of in vitro cytochrome P450-mediated metabolism of three furanocoumarins — bergapten, xanthotoxin, and angelicin. While bergapten and xanthotoxin, both linear furanocoumarins, were metabolized at similar rates (8.07 and 9.86 nmoles/min/g fw caterpillar, respectively), angelicin, an angular furanocoumarin, was metabolized more slowly (2.76 nmoles/min/g fw caterpillar). When all three furanocoumarins were assayed together, overall rates of metabolism were significantly reduced, suggesting substrate inhibition. Thus, the pattern of growth of larvae is consistent with the pattern of in vitro metabolism and is evidence in support of analog synergism. In a separate experiment, metabolism of xanthotoxin and angelicin individually and together were compared in six maternal families. Again, angelicin was metabolized more slowly than xanthotoxin and each furanocoumarin inhibited metabolism of the other. That significant family effects were found for rates of metabolism and for the ratio of moles of angelicin metabolized for each mole of xanthotoxin metabolized raises the possibility that genetic variation exists for the rate and specificity of metabolism and suggests that insect herbivores may be able to adapt to analog synergism.  相似文献

The sequence of steps versiconal hemiacetal acetate (VHA) to versiconal (VL) catalyzed by an esterase and VL to versicolorin B (VB) catalyzed by VL cyclase has been previously demonstrated in cell-free systems fromAspergillus parasiticus. VHA esterase and VL cyclase activities were estimated by determining the amounts of VL and VB after incubation of VHA in cell-free extracts from mycelia that were either synthesizing or not synthesizing aflatoxins. VHA esterase activity but not VL cyclase activity was present in extracts from cells grown in a nonaflatoxin-producing medium. VHA esterase activity was present in extracts from mycelia grown in aflatoxin-producing medium harvested after one to six days of incubation. VL cyclase activity was absent at one day, low at two days, maximal at three to five days, and lower at six days. VL cyclase activity appears in the later part of the growth period which is also the period of aflatoxin biosynthesis. This supports a role for VL cyclase in the aflatoxin biosynthetic pathway.  相似文献

Aspergillus niger or Aspergillus tamarii when grown as mixed cultures with toxigenic A. flavus inhibits biosynthesis of aflatoxin by A. flavus, owing primarily to its ability to produce inhibitors of aflatoxin biosynthesis and to their ability to degrade aflatoxin. Gluconic acid partly prevents aflatoxin production. The other factors such as changes in pH of the medium and the effect on the growth of A. flavus have no role in imparting capabilities to these cultures to inhibit aflatoxin production by A. flavus.  相似文献

Picea glehnii seedlings are affected by damping-off fungi in nurseries. The aims of this study were (1) to isolate fungi grown in the seedling rhizosphere in forest soil of P. glehnii, (2) to select fungi that produce antifungal compounds against Pythium vexans, and (3) to examine whether or not selected fungi can protect seedlings from P. vexans. Penicillium frequentans from Picea glehnii seedling roots produced antibiotic penicillic acid. Penicillic acid did not cause significant phytotoxicity to the seedlings. Penicillium frequentans increased the average percentage of surviving seedlings when inoculated together with Pythium vexans, but the increase was not significant. Vigorous mycelial growth of P. frequentans around seedling roots seems to be one of the mechanisms for protection, but the amount of penicillic acid was too low to show antifungal activity in the seedling rhizosphere.  相似文献

In recent yearsAspergillus flavus and aflatoxin production have been noted on several occasions in grain preserved with formic acid. Samples of mouldy barley treated with formic acid and stored in an open bin were investigated for the presence of fungi. In the lower part of the bin there was a clear dominance ofFusarium sporotrichioides, and deoxynivalenol and neosolaniol were detected.A. flavus andA. fumigatus were also present.Paecilomyces variotii occurred, almost as a pure culture, in the upper part of the bin, but no patulin was found. Cultivation of four fungal isolates from these genera on laboratory substrates containing formic acid showedP. variotii to be the most tolerant to formic acid, withstanding 150 mM, but still without patulin production.F. sporotrichioides andA. fumigatus tolerated only 6 mM formic acid. The growth ofA. flavus was reduced and atypical at 60 mM formic acid. Pretreatment ofA. flavus spores with formic acid increased aflatoxin production about 800 times.  相似文献

Ammi majus L. (Apiaceae), an important medicinal herb, constitutes the principal commercial source of xanthotoxin that is commonly used in leucoderma. Since the seed set and germination is poor, the present investigation was undertaken to propagate A. majus through tissue culture and monitor the yield of xanthotoxin (furanocoumarin) in the resultant callus and plantlets. The callus obtained from the cotyledonary leaves on MS medium supplemented with indoleacetic acid (IAA) + kinetin (Kn) + casein hydrolysate (CH), differentiated shoot buds on a medium additionally enriched with adenine. Plantlets 3.5 cm tall, resulted upon transfer of shoots to MS medium with indolebutyric acid (ISA) and glutamine. These plantlets flowered in vitro. The yield of xanthotoxin detected in regenerating cultures is more than hitherto reported from any other tissue including the seeds. lsozymes profile for leucineaminopeptidase (LAP), esterase and peroxidase could be used as reliable markers for the characterization of various stages of growth of callus and regenerants. The present report would prove advantageous in multiplication of Ammi majus and also in inducing enhanced levels of xanthotoxin.  相似文献

Previous studies have shown that two separate loci for 5S DNA sequences may exist within a species. Two size classes have been tentatively assigned in the ranges 327–468 bp (chromosome 1), and 469–500 bp (chromosome 5) and the entire data-base was subjected to various numerical taxonomic analyses. The results confirm the existence of two lineages of 5S DNA sequences represented by the two size classes.Hordeum vulgare is separate from the species known also asCritesion and, together withDasypyrum, occupy an intermediate position between the two size classes. The 5S DNA units fromTriticum spp. that have also been namedAegilops spp. in other classifications appear as a distinct group withinTriticum spp. at both loci. The consensus 5S DNA sequence fromPsathyrostachys is remote fromHordeum s.l. in contrast to opinions based on morphology. Other aspects are detailed and discussed, including the inadequacy of the computing methods used as well as the need for more data in view of the large amount of homoplasy. The merit of using the spacer for phylogenetic inference is also discussed.  相似文献

Ruta graveolens L. shoots cultured in stationary liquid phase produced furanocoumarins: psoralen, bergapten, xanthotoxin, isopimpinellin and imperatorin at the amount totalling almost 1 g/100 g dry wt of the shoots. The dominating metabolites were therapeutically important compounds: xanthotoxin – 0.33 g/100 g dry wt and bergapten – 0.32 g/100 g dry wt. Maximum contents of the majority of the compounds were observed on 28th day of culture.  相似文献

Combining different chromatography systems, unusual nonprotein amino acids were isolated and unequivocally identified from a small amount (less than 100 g fresh weight) ofAmanita gymnopus fruit body. Without obtaining crystals of these amino acids, on the basis of1H-NMR determination, high resolution mass spectrometry, chlorine analysis and oxidation with L-amino acid oxidase, one of them proved to be a new chloroamino acid, (2S)-2-amino-5-chloro-4-hydroxy-5-hexenoic acid (G2). The other three were (2S)-2-amino-5-hexenoic acid (G1), (2S)-2-amino-4,5-hexadienoic acid (G3) and (2S)-2-amino-5-hexynoic acid (G4). Amino acid (G1) was also encountered for the first time in natural products. Amino acid (G3) has been reported from several kinds of fungi belonging toAmanita, subgenusLepidella. Sialyltrisaccharides based on β-galactosyldisaccharides were synthesized using β-galactosidase andtrans-sialidase in one pot. Using β-galactosidase fromBacillus circulans andtrans-sialidase fromTrypanosoma cruzi simultaneously, 6 mM sialyltrisaccharides composed of about 95% NeuAcα(2,3)Galβ(1,4)GlcNAc and 5% NeuAcα(2,3)Galβ(1,6)GlcNAc were produced from a reaction mixture containing 25 mM 0-nitrophenyl-β-D-galactopyranoside, 100 mM N-acety lglucosamine and 10 mM p-nitrophenyl-α-D-N-acetylneuraminic acid. One beauty of this reaction was that a secondary hydrolysis of the disaccharide intermediate occurring between the activated galactopyranoside and N-acetylglucosamine was prevented. Using β-galactosidase fromEscherichia coli and the sametrans-sialidase, 15 mM sialyltrisaccharides composed of about 90% NeuAcα(2,3)Galβ(1,6)GlcNAc and 10% NeuAcα(2,3)Galβ (1,4)GlcNAc were produced from a reaction mixture containing 400 mM galactose, 800 mM N-acetylglucosamine and 20 mMp-nitrophenyl-α-D-N-acetylneuraminic acid. In this study, the reverse-galactosylation reaction between galactose and N-acetylglucosamine was dominant since the disaccharide intermediate mainly resulted in the sialylated product.  相似文献

Iturin A, a peptidolipid produced byBacillus subtilis, inhibits growth of a large number of fungi. In this study, the effects of iturin A were evaluated on nine isolates ofA. flavus and seven isolates ofA. parasiticus in liquid shake culture. The mycelial dry weight of theA. flavus isolates was not significantly influenced by iturin A, however, there was a significant reduction in mycelial dry weight for two of theA. parasiticus isolates. Aflatoxin production was significantly reduced in five of theA. flavus isolates and three of the six aflatoxigenicA. parasiticus isolates. For the other seven isolates, aflatoxin levels were either unchanged or significantly increased in the presence of iturin A. These results indicate that iturin A does not consistently reduce growth or aflatoxin production of these fungi in pure culture.  相似文献

A study has been carried out in Argentina on samples of corn genotypes from a breeding station as well as in commercially available corn meal. All samples were analyzed for fungal infection and aflatoxin B1.Mycological analysis of corn genotypes showed the presence of three principal genera of filamentous fungi Fusarium (100%), Penicillium (67%) and Aspergillus (60%). In the genus Fusarium three species were identified, F. moniliforme (42%), F. nygamai (56%) andF. proliferatum (1.8%). Eight species ofPenicillium were identified, the predominant species isolated were P. minioluteum, P. funiculosum and P. variabile. In the genus ranked third in isolation frequency, two species were identified, A. flavus and A. parasiticus, the percentage of infection was 78% and 21%, respectively. Only one corn genotype was contaminated with aflatoxin B1 at a level of 5 ppb. The cornmeal samples showed great differences in fungal contamination, the values ranging from 1 × 101 to 7 × 105 cfu g–1. Fusarium (68%), Aspergillus (35%) and Penicillium (21%) were the most frequent genera isolated. Among the genus, Aspergillus, A. parasiticus (38%) was the most frequent species isolated. All the samples of corn meal were negative to aflatoxin B1. These results indicate a low degree of human exposure to aflatoxins in Argentina through the ingestion of maize or corn meal.This revised version was published online in October 2005 with corrections to the Cover Date.  相似文献

A detailed karyotype analysis was made on the somatic complement ofPicea rubens andP. glauca. B-chromosomes were observed in someP. glauca populations. The karyotypes are generally asymmetrical with most of the chromosomes having median to median-submedian centromeres.Picea glauca chromosomes 2, 3, 7, and 8 have secondary constriction on their short arm and chromosome 10 has a secondary constriction on the long arm. Chromosome 3 was the most easily identifiable, as it has two secondary constrictions located on the short arm. InP. rubens, all the chromosomes but chromosomes 8 and 9 have one to four distinctive secondary constrictions. In general, the diagrammatic comparisons show a high degree of similarity amongP. mariana, P. rubens, andP. glauca. GenomicP. mariana probe strongly hybridized to dots of genomic DNA fromP. rubens andP. glauca indicating that there is a high sequence homology among these three species. The synchronizing agent, hydroxyurea was used at different concentrations to enhance the mitotic index of cell suspensions derived from embryogenic cultures. Hydroxyurea at 1.25 mM increased significantly the mitotic index. An increase of hydroxyurea from 1.25 mM to 5 mM and 10 mM resulted in a steady decrease of mitotic index.  相似文献

Sufficient supply of potassium (K) can alleviate the adverse effects of excess sodium (Na) on plant growth. However, it remains unclear if such a beneficial function is related to regulation of root growth and/or expression of K/Na transporters. Herein we report the responses of a rice cultivar, which was pretreated with normal nutrient solution for 1 month, to three levels of Na (0, 25, and 100 mM) without or with supply of K for 9 days. High Na (100 mM) significantly decreased plant growth, root activity, and total K uptake, and increased biomass ratio of roots to shoots. Short-term removal of K supply (9 days) did not affect root morphology and biomass ratio of roots to shoots, but decreased root activity of seedlings grown in high Na solution. K deficiency increased uptake of Na and transport of K from roots to shoots. Moreover, expression of OsHAK1, a putative K transporter gene, was upregulated by low Na (25 mM) and downregulated by high Na (100 mM) in roots. In leaves, its expression was suppressed by the Na treatments when K supply was maintained. Expression of OsHKT2;1, which encodes a protein that acts mainly as a Na transporter, was downregulated by high Na, but was enhanced by K deficiency both in roots and leaves. Expression of five other putative K/Na transporter or Na+/H+ genes, OsHKT1;1, OsHKT1;2, OsHKT2;3, OsNHX1, and OsSOS1, was not affected by the treatments. The results suggest that OsHAK1 and OsHKT2;1 were involved in the interactive effects of K and Na on their uptake and distribution in rice. Of 13 compounds tested, 11 inhibited malic enzyme activity in Mucor circinelloides, to some degree, at 5 mM. Four of these inhibitors (tartronic acid, morin, catechin and 2,5-dihydroxybenzoic acid) were studied further. Tartronic acid, morin and catechin were competitive inhibitors of malic enzyme (with respect to malate), with apparent Ki values of 0.04 mM, 5 μM and 0.6 mM, respectively. 2,5-Dihydroxybenzoic acid was a non-competitive inhibitor, with respect to malate, and had an apparent Ki value of 0.8 mM. Morin and tartronic acid did not inhibit any other NADPH-generating enzyme studied, although both inhibited malate dehydrogenase. The inhibitory actions of catechin and 2,5-dihydroxybenzoic acid were less specific. All four compounds inhibited malic enzyme, to some extent, when included in the culture medium. This inhibition was not as great as in vitro however and was insufficient to have an effect on lipid metabolism in M. circinelloides. We studied the tolerance of ectomycorrhizal (ECM) fungi to sodium chloride (NaCl) to find the best fungus to aid growth of Pinus thunbergii. Four ECM fungi, Cenococcum geophilum, Pisolithus tinctorius, Rhizopogon rubescens, and Suillus luteus, were grown in liquid MMN media with five different concentrations of NaCl for 30 days, and their mycelial weights were determined. Mycelial weights of P. tinctorius and R. rubescens were not significantly different between 0 mM and 200 mM, whereas those of C. geophilum and S. luteus decreased with increasing NaCl concentration, indicating that the former two species were more tolerant to higher NaCl concentrations than the latter species. We further studied the intraspecific differences in NaCl tolerance of nine P. tinctorius isolates. They were grown on MMN agar media with six different concentrations of NaCl for 21 days, and their radial growth was measured. In total, the hyphal growth at 25 mM NaCl was significantly higher than those at the other NaCl concentrations, and EC50 values were confirmed at between 50 mM and 200 mM. Among the isolates, Pt03 and Pt21 showed measurable growth at 200 mM; the growth of Pt03 was not significantly different between 0 mM and 200 mM. The results indicate that there are intraspecific variations in NaCl tolerance of Pisolithus species.  相似文献

Preharvest seed infection byAspergillus flavus and aflatoxin contamination in selected groundnut genotypes (fourA. flavus-resistant and fourA. flavus-susceptible) were examined in different soil types at several locations in India in 1985–1990. Undamaged mature pods were sampled at harvest and seed examined forA. flavus infection and aflatoxin content in two or more trials at ICRISAT Center on light sandy soils and red sandy loam soils (Alfisols), and on Vertisols, at Anantapur on light sandy soils, and at Dharwad and Parbhani on Vertisols. Rainy season trials (1985–1989) were all rainfed. Post-rainy season trials were irrigated; late-season drought stress (90 days after sowing (DAS) until harvest at 125 DAS) was imposed in the 1987/88 and 1989/90 seasons.A. flavus infection and aflatoxin contamination levels were much lower in seed of all genotypes from Vertisols than in seed from Alfisols across locations and seasons. Vertisols also had significantly lower populations ofA. flavus than Alfisols. There were no marked differences between light sandy soils and red sandy loam soils (Alfisols) in respect of seed infection byA. flavus and aflatoxin contamination. Significant interactions between genotypes and soil types were evident, especially in theA. flavus-susceptible genotypes. Irrespective of soil types,A. flavus-resistant genotypes showed lower levels of seed infection byA. flavus and other fungi than didA. flavus-susceptible genotypes. 筛选黄曲霉毒素生物防控菌,为黄曲霉毒素的生物防控提供支持。以花生原产地土壤为材料,采用牛津杯法筛选所需菌株。对筛选出的拮抗菌株进行抑制产毒曲霉菌株的生长、产孢、降解黄曲霉毒素实验。筛选出2株黄曲霉毒素生防细菌,编号21-1-2、17-3,经鉴定,拮抗菌21-1-2为枯草芽胞杆菌,拮抗菌17-3为地衣芽胞杆菌。分别对拮抗菌对曲霉孢子萌发的抑制、抑制黄曲霉的生长和菌丝延长以及减少黄曲霉毒素的产生、对黄曲霉毒素的分解作用等几个方面进行研究,结果表明,拮抗菌可以明显抑制产毒曲霉孢子的萌发、生长、菌丝的延长,减少黄曲霉毒素的产生以及分解黄曲霉毒素。  相似文献

Helminth parasites were identified in a natural population of primates at Polonnaruwa, Sri Lanka. Five fatally wounded or recently deceased toque macaques (Macaca sinica) and three langurs (Presbytis senex andP. entellus) were autopsied. The following nematodes were found:Oesophagostomum aculeatum (Chabertiidae),Streptopharagus pigmentata (Spirocercidae),Physaloptera sp. (Spiruridae),Enterobius vermicularis (Oxyuridae), andTrichuris trichiura (Trichuridae). One cestode,Hymenolepis was also recovered fromP. entellus. Among fresh faecal samples of 210M. sinica, worm eggs ofOesophagostomum andStrongyloides were most abundant, followed byTrichostrongylus. Two enzymes containing thiosulfate sulfur transferase activity were purified fromChlorobium vibrioforme f.thiosulfatophilum by ion exchange chromatography, gel filtration and isoelectrofocusing. Enzyme I is a basic protein with an isoelectric point at pH 9.2 and has a molecular weight of 39,000. TheK m-values for thiosulfate and cyanide of the purified basic protein were 0.25 mM (thiosulfate) and 5 mM (cyanide). Enzyme II is an acidic protein. The enzyme has an isoelectric point at pH 4.6–4.7 and a molecular weight of 34,000. TheK m-values of the acidic protein were found to be 5 mM for thiosulfate and 125 mM for cyanide.In addition to thiosulfate sulfur transferase activity, cellfree extracts ofChlorobium vibrioforme f.thiosulfatophilum also contained low thiosulfate oxidase activity and negligible thiosulfate reductase activity.
