A yeast strain AEH was isolated from oil contaminated soil and identified by analysis of 18S and 26S ribosomal DNA sequences asPichia anomala. Strain AEH was capable of degrading naphthalene, phenanthrene and chrysene, singly, and benzo(a)pyrene in combination. The yeast degraded 5.36 mg naphthalene l?1 within 2 days, and 5.04 mg phenanthrene l?1 and 1.54 mg chrysene 1?1 within 10 days. When a mixture of the four polycyclic aromatic hydrocarbons (PAHs) was treated at a concentration between 2.98 mg l?1 and 6.89 mg l?1, degradation rates were delayed for naphthalene and phenanthrene (3.79 mg l?1 and, 4.20 mg l?1 within 10 days, respectively), but enhanced for chrysene and benzo(a)pyrene (3.37 mg l?1 and, 1.91 mg l?1 within 10 days, respectively). In a binary system, all of the other 3 PAHs could be utilized as the carbon source for the cometabolic degradation of benzo(a)pyrene with naphthale ne as the best one.  相似文献

A detailed analytical study using combined normal phase high pressure liquid chromatography (HPLC), gas chromatography (GC) and gas chromatography/mass spectrometry (GC/MS) of Polynuclear Aromatic Hydrocarbons (PAHs) in fish from the Red Sea was undertaken. This investigation involves a preliminary assessment of the sixteen parent compounds issued by the U.S. Environmental Protection Agency(EPA). The study revealed measurable levels of Σ PAHs (the sum of three to five or six ring parent compounds) (49.2 ng g−1 dry weight) and total PAHs (all PAH detected) (422.1 ng g−1 dry weight) in edible muscle of fishes collected from the Red Sea. These concentrations are within the range of values reported for other comparable regions of the world. Mean concentrations for individual parent PAH in fish muscles were; naphthalene 19.5, biphenyl 4.6, acenaphthylene 1.0, acenaphthene 1.2, fluorene 5.5, phenanthrene 14.0, anthracene 0.8, fluoranthene 1.5, pyrene 1.8, benz(a)anthracene 0.4, chrysene 1.9, benzo(b)fluoranthene 0.5, benzo(k)fluoranthene 0.5, benzo(e)pyrene 0.9, benzo(a)pyrene 0.5, perylene 0.2, and indeno(1,2,3-cd)pyrene 0.1 ng g−1 dry weight respectively. The Red Sea fish extracts exhibit the low molecular weight aromatics as well as the discernible alkyl-substituted species of naphthalene, fluorene, phenanthrene and dibenzothiophene. Thus, it was suggested that the most probable source of PAHs is oil contamination originating from spillages and/or heavy ship traffic. It was concluded that the presence of PAHs in the fish muscles is not responsible for the reported fish kill phenomenon. However, the high concentrations of carcinogenic chrysene encountered in these fishes should be considered seriously as it is hazardous to human health. Based on fish consumption by Yemeni‘s population it was calculated that the daily intake of total carcinogens were 0.15 μg/person/day. A method was developed for simultaneous quantification of urinary 1- and 2-naphthols, 3- and 9-phenanthrols and 1-pyrenol using gas chromatography with mass spectrometry (GC-MS). This method was applied to urine samples from coke oven workers (n =28) and controls (n =22) from Northern China. Geometric mean levels of urinary 1-naphthol (58.8 μg l?1), 2-naphthol (34.1 μg l?1), 3-phenanthrol (7.35 μg l?1), 9-phenanthrol (1.28 μg l?1) and 1-pyrenol (25.4 μg l?1) were significantly higher among coke oven workers than controls. All the substances tested were highest among top-of-oven workers, who had 15-fold higher 1-naphthol, eight-fold higher 2-naphthol and 20-fold higher 1-pyrenol levels compared with controls. Using multiple linear regression models, 72.5% of the variation in 1- and 2-naphthol and 82.8% of the variation in 1-pyrenol were explained by the concentration of naphthalene or pyrene in the urine, the work category and the smoking intensity. Cigarette consumption significantly contributed to levels of urinary 1-pyrenol and naphthols, particularly 2-naphthol. A negative relationship between work category and the ratio of naphthols/1-pyrenol was observed among smokers. Our results suggest that urinary naphthols and phenanthrols reflect polycyclic aromatic hydrocarbon (PAH) exposure as well as the widely used 1-pyrenol, and that interactions between cigarette smoking and PAH exposure result in different patterns of metabolism for individual PAHs.  相似文献

Polycyclic aromatic hydrocarbons (PAHs) are ubiquitous pollutants of the environment. But is their microbial degradation equally wide in distribution? We estimated the PAH degradation capacity of 13 soils ranging from pristine locations (total PAHs ≈ 0.1 mg kg?1) to heavily polluted industrial sites (total PAHs ≈ 400 mg kg?1). The size of the pyrene- and phenanthrene-degrading bacterial populations was determined by most probable number (MPN) enumeration. Densities of phenanthrene degraders reflected previous PAH exposure, whereas pyrene degraders were detected only in the most polluted soils. The potentials for phenanthrene and pyrene degradation were measured as the mineralization of 14C-labeled spikes. The time to 10% mineralization of added 14C phenanthrene and 14C pyrene was inversely correlated with the PAH content of the soils. Substantial 14C phenanthrene mineralization in all soils tested, including seven unpolluted soils, demonstrated that phenanthrene is not a suitable model compound for predicting PAH degradation in soils. 14C pyrene was mineralized by all Danish soil samples tested, regardless of whether they were from contaminated sites or not, suggesting that in industrialized areas the background level of pyrene is sufficient to maintain pyrene degradation traits in the gene pool of soil microorganisms. In contrast, two pristine forest soils from northern Norway and Ghana mineralized little 14C pyrene within the 140-day test period. Mineralization of phenanthrene and pyrene by all Danish soils suggests that soil microbial communities of inhabited areas possess a sufficiently high PAH degradation capacity to question the value of bioaugmentation with specific PAH degraders for bioremediation.  相似文献

Pyrene and fluoranthene, when supplied as the sole carbon source, were not degraded by Burkholderia sp. VUN10013. However, when added in a mixture with phenanthrene, both pyrene and fluoranthene were degraded in liquid broth and soil. The amounts of pyrene and fluoranthene in liquid media (initial concentrations of 50 mg l−1 each) decreased to 42.1% and 41.1%, respectively, after 21 days. The amounts of pyrene and fluoranthene in soil (initial concentrations of 75 mg kg−1 dry soil each) decreased to 25.8% and 12.1%, respectively, after 60 days. None of the high molecular weight (HMW) polycylic aromatic hydrocarbons (PAHs) tested adversely affected phenanthrene degradation by this bacterial strain and the amount of phenanthrene decreased rapidly within 3 and 15 days of incubation in liquid broth and soil, respectively. Anthracene also stimulated the degradation of pyrene or fluoranthene by Burkholderia sp. VUN10013, but to a lesser extent than phenanthrene. The extent of anthracene degradation decreased in the presence of these HMW PAHs.  相似文献

Copper (Cu) is one of the most commonly reported metal pollutants in African water bodies, but there are few studies on African freshwater fish species of copper accumulation and copper toxicity. Adult O. mossambicus were exposed to 0mg l?1 (control) and 0.75mg l?1 Cu for 96h and 0 (control), 0.11, 0.29 and 0.47mg l?1 copper for 64 days. Samples of liver and gills were collected after 96h, and after 1, 32 and 64 days, respectively. There were significant differences in the mean Cu accumulation values in the liver and gills between the control and the Cu-exposed fish after the 96-h exposure. In fish exposed to 0.11 and 0.29mg l?1 Cu for 64 days there was an increase in copper level in the tissues. In fish exposed to 0.47mg l?1 Cu the concentration in the gill and liver tissue did not increase between Day 1 and Day 32. At this time, Cu accumulation in the liver was higher than for fish exposed to 0.11 and 0.29mg l?1 Cu for 64 days. Exposure to approximately 0.47mg l?1 Cu for more than 32 days induced mortality.  相似文献

Alkalinity stress is common in cultured aquatic animals and considered to be one of the major stress factors for fishes when they are transferred to saline‐alkali waters. To evaluate potential effects of alkalinity on the developmental biology of Oryzias latipes, fertilized eggs, larvae and breeding fish were exposed to different carbonate alkalinity concentrations of 1.5–64.5 meq l?1, for 9, 120, and 60 days, respectively. The mortality of embryos significantly increased when exposed to the high concentrations (16.5–64.5 meq l?1). Although more than 50% of survived embryos hatched in 16.5 and 31.4 meq l?1 concentrations of carbonate alkalinity, most were not able to swim up after hatching. Morphological abnormalities such as coagulated embryos, halted embryo development, and hatching failure were observed at stages 15, 29–33 and 38 in high concentrations (31.4, 64.5 meq l?1). Almost all larvae in 16.5 and 31.4 meq l?1 treatments died 70 d post‐hatch. Growth of juveniles exposed to carbonate alkalinity of 5.3 and 8.8 meq l?1 was not significantly different at 70 d and 120 d post‐hatch. The number of eggs released by breeders, the fertilization rate and the hatching rate of eggs were significantly lower in the 31.4 meq l?1 treatment than in other treatments. Although medaka are capable of surviving in high alkalinities (31.4, 64.5 meq l?1) for an extended period of time, these conditions are stressful to the fish, especially at the embryonic and reproductive stages.  相似文献

An efficient protocol for direct somatic embryogenesis in Capparis decidua has been developed. Mature zygotic embryos cultured in Murashige and Skoog (MS) liquid medium with 2,4-0 (0.1 mg l?1) and BA (0.5 mg l?1) produced somatic embryos directly without an intervening callus phase from the subepidermal cells. Treatment with ABA promoted maturation of somatic embryos and BA (1 mg l?1)promoted germination. One zygotic embryo produced approximately 230 somatic embryos within 14 to 15 weeks. Embryos germinated in eight weeks and acclimatized plants were transferred to pots.  相似文献

Plants were obtained via somatic embryogenesis in callus derived from in vitro raised leaf and petiole explants of Aconitum heterophyllum Wall. Callus was induced on a Murashige-Skoog medium supplemented with either 2,4-dichlorophenoxy acetic acid (2,4-d 1 mg l-1) and kinetin (KN 0.5 mg l-1) with coconut water (CW 10% v/v) or naphthalene acetic acid (NAA 5 mg l-1) and benzylaminopurine (BAP 1 mg l-1). Somatic embryos appeared after 2–3 months or 2 subculture passages when 2,4-d or NAA induced source of the callus was transferred to a MS medium containing BAP (1 mg l-1) and NAA (0.1 mg l-1). For successful plantlet formation, the somatic embryos were transferred to a medium containing 1/4 strength MS nutrient with indole-3-butyric acid (IBA 1 mg l-1). Alternatively, the somatic embryos were dipped in a concentrated solution of IBA for 5 min and placed on a hormone free medium. A newly isolated Rhodococcus sp. strain p52 could aerobically utilize dibenzofuran as the sole source of carbon and energy, and completely remove dibenzofuran at 500 mg?l?1 within 48 h. The strain metabolizes dibenzofuran by initial angular dioxygenation to yield 2,2′,3-trihydroxybiphenyl. Strain p52 could also remove 70 % of 100 mg?l?1 2-chlorodibenzofuran within 96 h and could metabolize a variety of aromatic compounds, namely dibenzo-p-dioxin, 2,8-dichlorodibenzofuran, dibenzothiophene, biphenyl, naphthalene, fluorene, phenanthrene, anthracene, carbazole, indole, xanthene, phenoxathiin, xanthone, and 9-fluorenone. Two distinct gene clusters encoding angular dioxygenases (DbfA and DfdA) were amplified and sequenced. The dbfA and dfdA gene clusters are located on two circular plasmids, pDF01 and pDF02, respectively. Both plasmids are self-transmissible; that is, they can transfer to the Gram-positive bacterium Bacillus cereus by conjugation.  相似文献

Saffron calli were induced from ovary explants on Murashige-Skoog (MS) medium supplemented with beyzyladenine (BA) and naphthalene acetic acid (NAA) as growth factors. MS medium with 5 mg l?1 BA and 10 mg l?1 NAA was selected for calli induction and undifferentiated calli growth, while MS medium with 1 mg l?1 BA and 1 mg l?1 NAA was the most appropriate for stigma differentiation. On this medium, stigma-like structures measuring 0.5–1.5 cm were obtained. Initially they were colourless, but yellow pigmentation, due to the presence of crocin, progressively increased with calli growth. Extracts of stigma-like structures were analysed by HPLC and the presence of saffron secondary metabolites was demonstrated. In addition, calli also showed yellow pigmentation.  相似文献

Biological treatment methods are effective at destroying polycyclic aromatic hydrocarbons (PAHs), and some of the highest rates of PAH degradation have been achieved using two-phase-partitioning bioreactors (TPPBs). TPPBs consist of a cell-containing aqueous phase and a biocompatible and immiscible organic phase that partitions toxic and/or recalcitrant substrates to the cells based on their metabolic demand and on maintaining the thermodynamic equilibrium of the system. In this study, the degradation of a 5-component mixture of high and low molecular weight PAHs by a defined microbial consortium of Sphingomonas aromaticivorans B0695 and Sphingomonas paucimobilis EPA505 in a TPPB was examined. The extremely low aqueous solubilities of the high molecular weight (HMW) PAHs significantly reduce their bioavailability to cells, not only in the environment, but in TPPBs as well. That is, in the two-phase system, the originally selected solvent, dodecane, was found to sequester the HMW PAHs from the cells in the aqueous phase due to the inherent high solubility of the hydrophobic compounds in this solvent. To circumvent this limitation, the initial PAH concentrations in dodecane were increased to sufficient levels in the aqueous phase to support degradation: LMW PAHs (naphthalene, phenanthrene) and fluoranthene were degraded completely in 8 h, while the HMW PAHs, pyrene and benzo[a]pyrene, were degraded by 64% and 11%, at rates of 42.9 mg l−1 d−1 and 7.5 mg l−1 d−1, respectively. Silicone oil has superior PAH partitioning abilities compared to dodecane for the HMW PAHs, and was used to improve the extent of degradation for the PAH mixture. Although silicone oil increased the bioavailability of the HMW PAHs and greater extents of biodegradation were observed, the rates of degradation were lower than that obtained in the TPPB employing dodecane.  相似文献

The impact of culture conditions and addition of antioxidants to media on microspore embryogenesis in rapeseed (Brassica napus cv. ‘PF704’) was investigated. Different concentrations of ascorbic acid (0, 5, 10, 20, 50, 100, and 200 mg l?1) and alpha (α)-tocopherol (0, 5, 10, 20, 50, 100, and 200 mg l?1) were evaluated along with two temperature pretreatments (18 d at 30°C; 2 d at 32.5°C followed by 16 d at 30°C). In addition, combinations of reduced glutathione (0, 10, 50, and 100 mg l?1) and ascorbic acid (5 and 10 mg l?1) were tested. Microspore embryogenesis was significantly enhanced using 10 mg l?1 ascorbic acid (334 embryos per Petri dish) compared with untreated cultures (184 embryos per Petri dish) at 30°C. α-Tocopherol (5 and 10 mg l?1) enhanced (312 and 314 embryos per Petri dish, respectively) microspore embryogenesis relative to untreated cultures (213 embryos per Petri dish) at 30°C, although there were no significant differences among cultures treated with 5–50 mg l?1 α-tocopherol. When 50 mg l?1 α-tocopherol was combined with 5 or 10 mg l?1 ascorbic acid, embryogenesis was significantly enhanced (308 and 328 embryos per Petri dish, respectively) relative to other ascorbic acid levels. Moreover, 10 mg l?1 of reduced glutathione and 5 mg l?l ascorbic acid enhanced microspore embryogenesis (335 embryos per Petri dish) compared to cultures without reduced glutathione (275 embryos per Petri dish). Microspore embryogenesis could be improved by adding ascorbic acid, α-tocopherol, and reduced glutathione when the appropriate combination and temperature pretreatment were selected.  相似文献

Nevirapine (NVP) is one of the HIV antiretrovirals detected recurrently in African surface waters. Liver side effects in humans were reported. Because the effects on fish are unknown, this study investigated the potential effects of NVP on selected biometric indices and liver histology of Oreochromis mossambicus (Mozambique tilapia) in a chronic exposure. Adult fish were exposed to two concentrations of NVP (1.48 and 3.74 µg l?1) in a static renewal system under controlled conditions for 30 days. At the end of the exposure, each fish was weighed, the total length recorded, and blood collected from the caudal vein. A standard necropsy was done, selected organs were sampled and weighed, and biometric indices calculated. Liver tissue was processed for histological assessment. Results showed significant differences (p < 0.001) in hepatosomatic indices between the exposed fish and the control, as well as in spleen indices of the fish exposed to the higher NVP concentration (p = 0.008). Liver tissue of exposed fish showed significant histological changes (p < 0.001), including hepatocyte apoptosis, vacuolation, and a mild fibrosis around some of the veins and bile ducts. Nevirapine in African surface waters could have long-term negative effects on the health of fish.  相似文献

This study presents data collected over a 6 year period on the effects of extremely low‐frequency magnetic fields (MFs) (1·4–1·6 µT, 500 Hz and 1·4–1·6 µT, 72·5 Hz) and MFs in combination with other environmental stressors (elevated temperature, 0·01 mg l?1 trichlorfon, 0·01 mg l?1 copper sulphate pentahydrate) on roach Rutilus rutilus embryos. Effects were studied during different stages of early development. Rutilus rutilus were raised in ponds for 4 months after exposure to MFs. The mass, standard length (LS) and morphological characteristics of underyearlings which were exposed as embryos were recorded. An increase in embryo mortality and a decrease in LS and mass indices in underyearlings were noted after they had been exposed to a combination of MFs and different adverse environmental factors. In addition, exposure to MFs led to changes in the total number of vertebrae and the number of seismosensory system openings in the mandibular bones of underyearlings. MFs of different frequency caused both increases (500 Hz) and decreases (72·5 Hz) in morphological diversity. The stressors used in this study, however, did not increase the fluctuating asymmetry of bilateral morphological characteristics. The possible microevolutionary effects of exposure to MFs alone and in combination with other adverse environmental factors upon natural fish populations are discussed.  相似文献

The ability of immature, mature and endosperm-supported mature embryos of diploid and tetraploid winter ryes (Secale cereale L) was tested to compare the callus induction and plant regeneration. Immature embryos were obtained from field grown rye. Immature embryos were aseptically excised and placed, with the scutellum upwards, on the callus culture medium consisted of Murashige and Skoog (MS) mineral salts supplemented with 2 mg l?1 2,4-dichlorophenoxyacetic acid (2,4-D). Mature embryos were aseptically excised the imbibed seeds and placed, scutellum up, on MS medium supplement with 2 mg l?1 2,4-D. Endosperm-supported mature embryos were moved slightly (not set free) in the imbibed mature seeds. The seeds with moved embryos were placed furrow downwards in dishes containing 8 mg l?1 2,4-D for callus induction. The developed calli and regenerated plant were maintained on hormone free MS medium. Comparison of the responses of the three explants used indicated that endosperm-supported mature embryo was the most useful explant for plant regeneration in both diploid and tetraplold ryes. This is the first report of winter ryes plants having been regenerated from endosperm-supported mature embryos.  相似文献

Menthol is known for its analgesic properties, but relatively little information is available on its potential as an anaesthetic on fish. The purpose of this study was to assess anaesthetic and sedative effectiveness of menthol and its safety in rainbow trout (Oncorhynchus mykiss). Juvenile rainbow trout 180 ± 28 g (mean ± SD) within a range of 152–208 g fish?1 were individually exposed to menthol concentrations between 10 and 150 mg l?1 and observed for behavioural responses, induction time to anaesthesia and recovery time. Menthol concentrations of 40–150 mg l?1 induced anaesthesia with varying exposure times. There was an exponential relationship (p < 0.001) between induction time and menthol concentration. Menthol concentrations of 80–150 mg l?1 induced anaesthesia within three minutes of exposure and fish recovered within three minutes. Induction and recovery data showed that 80 mg l?1 was most suitable for anaesthesia in juveniles of this species. Concentrations of 10–20 mg l?1 had sedative effects. Menthol stock solutions prepared using ethanol and acetone and storage time of stock solutions at room temperature for up to 48 h showed no significant differences in anaesthetic efficiency. When exposure time to menthol was kept constant at three minutes, 22% of fish had temporary cessation of gill ventilation. These fish had longer recovery times than those that did not show that response. Menthol was an effective anaesthetic and could be tested as a sedative for trout.  相似文献

The efficacy of clove oil as an anaesthetic and its effects on blood parameters in Nile perch Lates niloticus were evaluated in 2010. Clove oil concentrations of 49.3, 73.9 and 98.5?mg l?1 induced anaesthesia in <3?min, while the average recovery time from anaesthesia was 11?min 22 s. The optimal oil clove oil concentration was 49.3?mg l?1, inducing anaesthesia in 4?min 33 s, with recovery in 3?min 31 s. No stress response was elicited. Clove oil at a concentration of 24.6?mg l?1 was an effective sedative, whereas a concentration of 49.3?mg l?1 was sufficient for measuring fish and stripping gametes. A concentration of 73.9?mg l?1 induced anaesthesia within 4?min and fish recovered in 10?min. Therefore, clove oil was an effective anaesthetic and sedative for the handling of Nile perch within a mass range of 0.4–12?kg fish?1.  相似文献

A partitioning bioreactor, consisting of an aqueous phase containing Sphingomonas aromaticivorans and an immiscible organic phase (dodecane), loaded with naphthalene and phenanthrene, was operated at two scales, 5 l and 150 l. Complete degradation of 15 g and 300 g, respectively, of these polyaromatic hydrocarbon (PAH) mixtures was achieved in 21 h in both cases resulting in a volumetric PAH degradation rate of 238 mg l–1 h–1 based on reactor aqueous volumes.  相似文献

Embryogenic avocado cultures derived from ‘Hass’ protoplasts were genetically transformed with the plant defensin gene (pdf1.2) driven by the CaMV 35S promoter in pGPTV with uidA as a reporter gene and bar, the gene for resistance to phosphinothricin, the active ingredient of the herbicide Finale® (Basta) (Bayer Environmental Science, Research Triangle Park, Durham, NC ). Transformation was mediated by Agrobacterium tumefaciens strain EHA105. Transformed cultures were selected in the presence of 3.0 mg l?1 phosphinothricin in liquid maintenance medium for 3–4 mo. Liquid maintenance medium consisted of modified MS medium containing (per liter) 12 mg NH4NO3 and 30.3 mg KNO3 and supplemented with 0.1 mg l?1 thiamine HCl, 100 mg l?1 myo-inositol, 30 g l?1 sucrose, 3.0 mg l?1 phosphinothricin, and 0.41 μM picloram. Somatic embryo development from transformed cultures was initiated on MS medium supplemented with 45 g l?1 sucrose, 4 mg l?1 thiamine HCl, 100 mg l?1 myo-inositol, 10% (v/v) filter-sterilized coconut water, 3.0 mg l?1 phosphinothricin, and 6.0 g l?1 gellan gum. Limited plant recovery occurred from somatic embryos on semi-solid MS medium supplemented with 3.0 mg l?1 phosphinothricin, 4.44 μM 6-benzylaminopurine (BA), and 2.89 μM GA3; transformed shoots were micrografted on in vitro-grown seedling rootstocks. Approximately 1 yr after acclimatization in the greenhouse, transformed shoots were air-layered to recover transformed roots. Genetic transformation of embryogenic cultures, somatic embryos, and regenerated plants was confirmed by polymerase chain reaction (PCR), Southern blot hybridization, the XGLUC reaction for uidA, and application of the herbicide Finale® to regenerated plants.  相似文献
