Excised root cultures of Atropa belladonna L. and A. belladonnavar. lutea Döll, established from liquid-grown seedlingsand from callus, when allowed to continue growth without subculturefor several weeks, spontaneously initiate shoot buds from smallnodules of callus which arise at the cut ends of the roots.The frequency and rapidity of formation of such buds is dependentupon the number of passages through which the roots have beensubcultured. The morphogenetic expression of callus cultures and of suspensioncultures derived from them is influenced not only by the timethat the root cultures have been maintained in culture but bythe composition of the medium used for callus initiation andsubsequent culture. The presence of elevated levels of ammoniumions in these media favours the development of incipient plantsrather than roots. Cultures have thus been obtained in whichthe predominant form of morphogenesis is embryogenesis (as establishedin a subsequent paper by Konar, Thomas, and Street, 1972).  相似文献

Reciprocal grafts involving leaf petioles and lamina of thejuvenile and mature phase of Hedera helix were prepared to determinethe relative importance of petiole and lamina on root initiationin leaf cuttings. The results indicated that root initiationwas mainly a function of the potential of cells in the petioleto respond in a specific morphogenetic pattern. Initially, rootinitiation was unaffected by the type of lamina. However, overtime, a factor translocated from the juvenile lamina promotedroot initiation in the mature petiole. This factor decreasedthe time taken for root initiation and increased the numberof roots per mature petiole. Summary Hypocotyls of detached stems standing in culture solution produced adventitious roots sooner than did petioles of detached primary leaves. An auxin, probably indol-3-ylacetic acid, appeared in the solutions before the hypocotyls or petioles produced roots. After attaining a maximum, the amounts of auxin in the solutions decreased as fewer roots were formed. Two cytokinins were found in the culture solutions; one had a similar Rf to zeatin, the other ran more slowly on chromatograms. The amounts of cytokinin in the solutions were associated with root formation. Stems soon died unless their hypocotyls formed roots, but the primary leaves survived without roots forming provided a callus formed on the petiole. Hence adventitious roots, or callus tissues, may have produced cytokinins that replaced those produced by the original roots, found in sap exuded from the stem stumps, and were essential for survival of the stems and leaves.  相似文献

Procedures are described for producing axillary shoots fromseedling apices and adventitious shoots from petioles and leaf-derivedcallus of sugar beet cultivars. The rate of adventitious shootregeneration from petioles was influenced by temperature, BAPconcentration of the medium, and the time in culture of theseedling apices from which the petioles were excised. Petiolesectioning confirmed that adventitious shoots originated inthe sub-epidermal parenchyma. Two distinct types of callus wereproduced from leaf explants, but only white friable callus wascapable of shoot development. This callus developed from browntissue and was composed of thin-walled cells with dense cytoplasmand prominent nuclei. Green compact callus with thick-walledlignified cells developed from green tissue, but did not produceshoots. Successful seed sterilization and shoot regenerationfrom petiole explants and callus was cultivar-dependent. Adventitiousshoots were rooted and successfully transplanted to pottingcompost under glasshouse conditions. Cotyledons of Sinapis alba cultured in the light on filter-papermoistened with water initiated root primordia within 2 daysafter detachment. These increased in size and penetrated thebase of the petiole after a further 3 days. Culture in sucrosesolution in the light inhibited root formation mainly by affectingthe extension phase when many primordia regressed resultingin a decline in the numbers of primordia per petiole. When cotyledonswere cultured in the dark no primordia were initiated in water,but sucrose promoted root formation. Levels of RNA rose to a peak value in the terminal petiole segmentat a time when primordia were being initiated in this region,protein reached a peak 2 days later. This pattern was much lessevident in the subterminal segments. It did not occur at allin cotyledons cultured in sucrose in the light where a smallinitial increase in RNA and protein was followed by a rapiddecline. The level of RNA and protein in the terminal segmentin light-cultured cotyledons in water was related to the numberof light periods given following detachment as was the percentageof cotyledons which formed roots. The effect of sucrose on RNAand protein content was less marked when cotyledons were transferredfrom sucrose to water instead of being maintained in sucrosethroughout. Sucrose clearly modifies the biochemical and structural changestaking place at the petiole base having particularly inhibitoryeffects on the extension phase of the primordia. The evidenceindicates that sucrose could be causing premature ageing ofcells forming the primordia.  相似文献

Summary Root formation was obtained on the petioles of detached leaves of several leguminous plants, particularly on the primary leaves of bean. Root formation is easily obtained in artificial light at a temperature of 22 to 24°C. In the greenhouse it is optimal in early spring and late autumn. During hot summer seasons no roots but callus was formed on the petioles. Root formation was inhibited when the pulvinus was left on the petiole. Nodulation of the rooted leaves is inhibited by combined nitrogen and high temperatures. The optimum light intensity for rooted leaves is low in comparison with that of intact plants. Far-red light reduces root-nodule formation; its inhibitory effect is partly eliminated by subsequent irradiation with red light.  相似文献

The CO2 exchange of fully expanded detached primary leaves ofdwarf bean (Phaseolus vulgaris) with roots on the petioles hasbeen measured. Rates of apparent photosynthesis and respirationincreased as roots grew, decreased when roots were removed,and increased again as root regenerated. Rates of photosynthesisof different leaves were highly correlated with the dry weightof root on their petioles. Photosynthesis and respiration weredecreased when root growth was restricted by kinetin, and wereincreased when root growth was stimulated by IAA. Photosynthesisof an attached leaf declined with time while that of a comparabledetached leaf increased. The results suggest that photosynthesisis correlated with the size of the roots that are a sink forphotosynthates.  相似文献

The involvement of polyamines during adventitious root formationwas evaluated using a de-bladed petiole rooting assay for theeasy-to-root juvenile and difficult-to-root mature phase ofEnglish ivy (Hedera helix L.). Auxin (NAA 0.1 mM) stimulatedroot formation in juvenile phase cuttings, but failed to promoterooting in the mature phase. The addition of putrescine, spermineor spennidine (1.0 mM) with or without NAA (0.1 mM) did notaffect the rooting response in either the juvenile or maturephase cuttings. There was a significant increase in endogenouslevels of putrescine and spermidine in NAA-treated cuttings,but the only significant difference between the root formingjuvenile and the non-root forming mature phase cuttings wasan increase in putrescine levels. In NAA-treated juvenile cuttings,the polyamine biosynthesis inhibitor DFMA (1.0 mM) promotedroot formation from 9.2 to 14.5 roots per cutting, while DFMO(1.0 mM) reduced root formation from 9.1 to 1.4 roots per cutting.The promotion of rooting by DFMA was completely reversed byputrescine (1.0 mM), but putrescine, spermine or spermidine(1.0 mM) could not reverse the inhibitory effect of DFMO. NeitherDFMA nor DFMO promoted root formation in mature phase cuttings.DFMA was also added to NAA-treated juvenile petioles at variousstages during the root formation process. DFMA promoted rootingwhen applied during the early stages of root induction (0–3d), but became inhibitory to root formation when applied duringthe organization (6–9 d) or root elongation stages (9–12d). Root cultures of Lotus corniculatus L. cv. Leo transformed withAgrobacterium rhizogenes (C58Cl-pRi15834) grew rapidly in liquidmedium when cultured in the dark and produced large numbersof shoots when illuminated. The shoots, which could be regeneratedto produce fertile plants, were maintained in liquid mediumas shoot-organ cultures The accumulation and cellular distribution of condensed tanninswas determined during the growth of these root and shoot organcultures and in primary callus from non-transformed explants.Root and shoot cultures predominantly accumulated insolublepolymeric tannins which yielded both cyanidin and delphinidinon hydrolysis at ratios equivalent to control plants. Methanol-solublevanillin-positive compounds were isolated but no free oligomericproanthocyanidins, monomeric flavans or dihydroflavonols weredetected in these extracts. Condensed tannin accumulation waslinearly related to root growth and had a similar spatial distributionin ‘tannin’ cells in roots and leaves as comparedto control plants. Tannin-containing cells were absent frommeristematic cells of the root tip and root/shoot interface.Primary callus cultures failed to accumulate condensed tanninson media containing auxins, and exogenously supplied auxinswere found to inhibit tannin accumulation by transformed rootand shoot cultures Key words: Lotus corniculatus, Agrobacterium rhizogenes, hairy roots, condensed tannins, shoot and root cultures.  相似文献

Koide, R. 1985. The nature and location of variable hydraulicresistance in Helianthus annuus L. (sunflower).—J. exp.Bot. 36: 1430–1440. Hydraulic resistances for whole sunflower plants (Helianthusannuus L.) and sunflower leaves, stems, petioles and roots weremeasured. Whole plant hydraulic resistance was shown to declinewith an increase in transpiration. Leaf hydraulic resistancewas shown, with one technique employing transpiring leaves,to vary with transpiration and with another technique, employingpressure-induced flow in leaves, to be constant over a widerange of transpiration. Stem and petiole hydraulic resistanceswere constant over a wide range of exudation. Pressure-inducedflow through root systems was shown to be an inappropriate methodfor characterizing their hydraulic properties because flow mayoccur through unnatural paths. The technique employing measuredtranspiration rates and water potentials of non-growing leavesand soil is suggested to be better. The radially-organized petiole of Phaseolus and the dorsiventralpetioles of Datura and Lycopersicon were used as stocks in budgrafting. Petiolar structure was examined after some weeks'growth of the scion, during which the grafted petiole came tofunction as a stem in supporting normal, vigorous shoot growthand was not abscised even at the end of the growing season.Below the graft union, reactivated petiolar cambium producedmassive amounts of secondary tissue with greatly enlarged vessels.Cambial activity was confined to the existing vascular bundlesexcept for the development of a little inter-fascicular cambiumin young, grafted Phaseolus petioles. The initiation and development of root primordia in detachedcotyledons of Sinapis alba (white mustard) and Raphanus sativus(radish) are studied, together with the inhibitory effects ofsucrose and mineral nutrients on these processes. Root primordium initiation on petioles of excised mustard cotyledonscultured in petridishes in water commenced after 3 days andwas completed after 5 days in culture, by which time a numberof the primordia had extended and emerged from the petiole.Both sucrose and mineral nutrient solution had an inhibitoryeffect which was most marked when the cotyledonswere culturedin the solution from the time of excision. The total numberof primordia initiated, their rate of development, and the finaltotal number of emerged roots were all reduced. The later thetime of transfer from water either to sucrose or to nutrient,the less marked the inhibition. Indeed, nutrient solution enhancedroot growth in mustard when cotyledons were transferred after5 days in water when root emergence had just commenced. The effects of sucrose and nutrients in relation to applicationbefore and after initial meristem formation has taken placeare discussed, together with the ways in which these two solutionsmay exert their effect on root initiation and development.  相似文献

Callus was induced from mature leaf lamina, petiole, stem, and main and branch root of Panax ginseng and maintained under different nutrient and light conditions. Heterogenous callus cultures differentiated organoids and/or embryoids. Embryoids that arose from callus cultures of leaf origin germinated into shoots. Summary Adventious root and shoot formation was obtained from cotyledon fragments of chestnut (Castanea sativa Mill.) and these processes followed two phases. In a first stage after detachment of the embryonic axis, the cotyledon fragments in culture formed a cotyledon petiole, which elongated for about 6d. Thereafter, root primordia arose at the tip of the cotyledon petioles, followed by normal root development. In some cases, the cotyledon, petioles showed adventitious shoot regeneration from a nodular structure previously formed at the end of the petioles. The presence or absence of growth regulators did not significantly influence root regeneration, whereas cytokinins stimulated shoot formtion. The processes of root and shoot differentiation were studied also at the histological level. Observation with a light microscope showed that the developing root apical meristems were connected with a vascular bundle of the cotyledon petiole. Similarly, shoot bud meristem connections were observed with vascular tissue inside the nodular structure.  相似文献

The effect of root temperature and form of inorganic nitrogensupply on in vitro nitrate reductase activity (NRA) was studiedin oilseed rape (Brassica napus L. cv. bien venu). Plants weregrown initially in flowing nutrient solution containing 10 µMNH4NO3 and then supplied with either nitrate or ammonium for15 d at root temperatures of 3, 7, 11 or 17 °C. Shoot temperatureregime was similar for all plants; 20/15 °C, day/night.Root NRA was highest when roots were grown at 3 and 7 °C.In laminae and petioles NRA was highest when roots were 11 or17 °C. The plants supplied with ammonium had much lowerlevels of NRA in roots after 5 d than the plants supplied onlywith nitrate. NRA in the laminae of plants supplied with ammoniumwas low relative to that in plants supplied with nitrate onlywhen root temperature was 11 or 17 °C. Values of the apparent activation energy (Ea) of NR, calculatedfrom the Arrhenius equation, in laminae and petioles were differentfrom roots suggesting difference in enzyme conformation. Evidencethat the temperature at which roots were growing affected Eawas equivocal. Transgenic hairy roots were induced from petiole and root segments of in vitro plant Aralia elata, a medicinal woody shrub, after co-cultivation with A. rhizogenes ATCC 15834. The percentage of putative hairy root induction from root segments was higher (26.7%) than petiole explants (10.0%). Hairy roots showed active production of lateral roots with vigorous elongation. Transgenic plants were regenerated from hairy roots via somatic embryogenesis. These plants had wrinkled leaves, short petioles and numerous lateral hairy roots. The RT-PCR analysis showed the expression of rol A, B, C, D, aux 1 and 2 genes differed between the transgenic lines. Endogenous IAA level was higher in transgenic than non-transgenic plants. Conclusively, transgenic hairy roots were developed for first time in A. elata and the transgenic hairy root lines showed distinct morphological growth pattern and gene expression.  相似文献

Rooting experiments carried out with isolated primary leavesof dwarf bean demonstrated the effect of GA on increasing theIAA level and IAA synthesis. Pretreatment of the lamina of isolatedleaves with GA, especially when tryptophane was added at thesame time, strikingly increased the rooting of petioles. Enhancedroot formation on the petiole of GA-pretreated leaves can beattributed to increase of IAA level by GA in the lamina, i.e.in IAA biosynthesis the utilization of tryptophane releasedfrom the proteins of isolated leaves is promoted by GA. Application of TIBA on the upper part of petiole suppressedthe stimulatory effect of GA on root initiation presumably becauseTIBA inhibits the transport of IAA into the petiole, which resultsin failure of rooting. Pretreatment of the leaves with cytokinins also results in failureof root production. These substances retard protein decompositionin isolated leaves so that the quantity of TPP available forauxin synthesis is less. Simultaneously, this treatment increasesthe cytokinin concentration in the leaf tissues so that theauxin/cytokinin ratio alters and mainly callus forms on thebase of the petioles and differentiation of root primordia cannottake place.  相似文献

Arabidopsis pumila is a type of cruciferous ephemeral plant, which in China mainly grows in the desert environments of northern Xinjiang. A. pumila not only has a short growth duration, but also has high photosynthetic efficiency, seed yield, salt tolerance, and drought resistance. It is an ideal species for the study of environmental adaptations in ephemeral plants. We induced callus tissue formation on the roots and hypocotyls of 8-day-old seedlings, and on the leaves and petioles of 4-week-old seedlings, and obtained multiple adventitious shoots on these tissues grown on Murashige and Skoog induction medium supplemented with 0.5 mg/L 6-Benzylaminopurine and 0.1 mg/L α-Naphthalene acetic acid. Young roots, hypocotyls, leaves, and petioles could all induce calluses, but the induction rate was highest on young roots. In addition, the leaves and petioles of 4-week-old seedlings were used as explants, the Δ1-pyrroline-5-carboxylic acid synthase gene 1 of A. pumila controlled by 35S promoter of cauliflower mosaic virus was used as target gene, and hygromycin B was used as screening antibiotic to explore Agrobacterium tumefaciens GV3101 mediated transformation. The results showed that the callus induction rate of petiole explants was the highest when they were treated with Agrobacterium suspension (OD600?=?0.6) for 10 min and thenco-cultured in dark for 2 days. The qRT-PCR results showed that the ApP5CS1.1 gene was overexpressed in the transgenic plants. These protocols provide working research methods for exploring the cellular level adaptative mechanisms of this species to desert environments.
