Seed germination in Talinum triangulare as affected by photoperiod,with or without previous incubation in the dark in water at25 or 4 °C, was studied. The time course and quantity ofseed germination in photoperiods of 1 h and above were similarwith or without dark pretreatment, but the time to half maximumgermination was reduced from 12 days in non-dark pretreatedseeds to 4 days in seeds given 20 days in the dark at 25°C.A photoperiod of 0·25 h gave a lower rate and total germinationthan photoperiods of 1 h and above. Un-pretreated seeds required17 cycles of 24 h photoperiod for maximum germination as comparedwith 7 or less cycles if the seeds received more than 10 daysdark pretreatment at 25 °C. Both the rate and total germinationin light increased as the length of dark pretreatment at 25°C was increased from zero to 30 days. Incubation of theseeds in water in the dark at 4 °C for 5 to 30 days priorto illumination at 21 °C, reduced both the rate and quantityof seed germination in light as compared with those similarlyincubated in the dark at 25 °C. However, previous incubationin the dark for 30 days at 4 °C partially substituted forthe light requirement. Supplementing pollen suspension cultures with CO2 (3–5per cent) caused a marked increase in germination and tube growthin vitro in Brassica campestris L. cv. toria. A weakening ofself-incompatibility by increased CO2 levels from 3–5per cent was observed. The percentage of pollen tubes whichpenetrated the cuticle layer of stigmatic papilla cells in self-pollinatedpistils was high when CO2 level was 5 per cent. Phosphoenolpyruvate (PEP) carboxylase activity was greater in the pollengerminated in 4 per cent CO2 as compared to air (0.03 per cent).A possible role of CO2 for self-recognition and control of pollentube growth is proposed, proposed. Seeds of Araucaria hunsteinii K. Schum. dried quicker at 29°Cthan at 19°C and quicker with the seed-coat removed thanwhen intact; seeds enclosed in polyethylene bags increased inmoisture content. At 15°C, seeds in a flow of air driedquicker than seeds in a box with silica gel, which in turn driedquicker than seeds in a box with no desiccant. No loss of germinationability occurred on drying fresh seed from 53 to about 32 percent moisture content (fresh weight basis); during further desiccationthe percentage germination was related to percentage moisturecontent in the form of a sigmoid curve, culminating in a completefailure to germinate at approximately 14 per cent moisture content.A consistent relationship was observed for all treatments andthe mean critical moisture content for seed death (failure togerminate) was near 23 per cent. Excised embryos grew on 1 percent agar but died if previously subjected to 14 h of desiccationat 15°C. In contrast, no relationship was found between germination andmoisture content of A. cunninghamii D. Don on desiccation from21 to 7 per cent moisture content. Possible causes for the observeddifference in response to desiccation are discussed and methodsfor seed storage are considered. Seeds of two lettuce cultivars (Lactuca sativa L., cv. Meikoninginand cv. Grand Rapids) were hermetically stored with constantmoisture contents ranging between 3.6 and 17.9 per cent (freshweight basis) at constant temperatures ranging between 5 and75 °C. The decline with time in percentage germination andpercentage normal seedlings was determined for each storagetreatment. The data were fitted to an equation which containsthe constants: K1, the probit of the initial percentage germinationor normal seedlings; KE, a species constant; CW, the constantof a logarithmic moisture term; CH, the constant of a lineartemperature term and CQ, the constant of a quadratic temperatureterm. Regression analysis of data from storage periods up to5.5 years at temperatures of 5–75 °C and seed moisturecontents of 3.6–13.6 per cent yielded the following values:KE= 8.218, CW=4.797±0.163, CH=0.0489±0.0050 andCQ=0.000365±0.000056. Although this equation consistentlyprovided a better fit, simplified equations, assuming eithera log-linear relationship between seed longevity and temperature,or a log-linear relationship between seed longevity and bothmoisture content and temperature, accounted for more than 94per cent of the variation at the restricted temperature rangeof 5–40 °C. Longevity of the same seed lots at sub-zero temperatures (–5,–10 and –20 °C) was studied in separate tests.Freezing damage, resulting in abnormal seedlings in the germinationtest, occurred at –20 °C when the moisture contentof the seeds exceeded 12 per cent. No decline in percentagenormal seedlings was observed after a storage period of 18 monthsor longer at –20 °C, provided the seed moisture contentdid not exceed 9.5 per cent. For seeds stored at –5 and–10 °C with 9.6–12.5 per cent moisture content,the observed rate of decline of percentage normal seedlingswas adequately predicted by the viability equation, using theabove values for the constants. This suggests that for low moisturecontents the viability equation can be applied to estimate longevityat sub-zero temperatures. Simulated swards of each of two selection lines of Lolium perennecv. S23 with ‘fast’ and ‘slow’ ratesof ‘mature tissue’ respiration were establishedin growth rooms at 20/15 °C day/night temperatures and studiedover four successive regrowth periods of 46, 30, 26 and 53 daysduration. The ‘slow’ line outyielded the ‘fast’,both in harvestable shoot (above a 5 cm cut) and in root andstubble. Its advantage increased over successive regrowth periodsto 23 per cent (total biomass). Gas analysis measurements onthe entire communities (including roots), during the final regrowthperiod, showed that the ‘slow’ line had a 22–34per cent lower rate of dark respiration per unit dry weight.This enabled it to maintain its greater mass of tissue for thesame cost in terms of CO2 efflux per unit ground area. Halfthe extra dry weight produced by the ‘slow’ line,relative to the ‘fast’, could be attributed to itsmore economic use of carbon. The rest could be traced to a 25per cent greater tiller number which enabled the ‘slow’line to expand leaf area faster (though not at a greater rateper tiller), intercept more light and fix more carbon, earlyin the regrowth period. Over 50 per cent germination has been obtained from Elaeis guineensisform pisifera seeds stored in unaerated sterile distilled waterfor 6 months. The moisture levels of the seeds and excised embryoswere of the same order (20–30 and 60–70 per cent,respectively) as those of fully imbibed fresh seeds. The implicationsof an apparent lower oxygen requirement by seeds stored underwater as against germinating seeds are discussed in the contextof the successful storage. Damage and degradation of cellular proteins is observed duringage-induced seed deterioration. L-Isoaspartyl protein methyltransferase(EC 2.1.1.77 [EC] ) is an enzyme hypothesized to play a role in limitingand repairing age-induced damage to proteins. Tomato (Lycopersiconesculentum Mill. ‘New Yorker’) seeds were assayedfor changes in L-isoaspartyl methyl-transferase activity duringaccelerated ageing and after osmotic priming. Accelerated ageingof seeds for 1–4 d at 45C and 100% relative humidityreduced germination from 94% to 71%, increased the mean timeof germination (MTG) from 2.4 to 5.8 d, and was accompaniedby a correlative decrease in L-isoaspartyl methyltransferaseactivity (r2=0.90). Aged and untreated seeds were primed for7 d at 20C in darkness using aerated solutions of 3% KNO3 orpolyethylene glycol 8000 (PEG) with equivalent osmotic potential(–1.25 MPa). Priming with KNO3 decreased the MTG, butdid not improve germination percentage for untreated seeds.Priming did not affect L-isoaspartyl methyltransferase activityin untreated seeds, but restored activity in aged seeds primedin KNO3 to levels near that of untreated seeds. Priming withPEG did not effectively improve the MTG or increase L-isoaspartylmethyltransferase activity. During germination, L-isoaspartylmethyltransferase activity remained constant for 48 h post-imbibitionand then declined, suggesting that the enzyme was developmentallyregulated and inactivated or degraded as radicle emergence occurred. Ethylene (10 µ1–1) caused about one-third of highlydark-dormant seeds of common purslane (Portulaca oleracea L.)to germinate in the dark. Attempts were made to increase germinationin the dark with nitrate and ethylene combinations. When applieddirectly to the seeds, KNO3 did not stimulate germination andKNO3 plus ethylene did not increase germination above that ofethylene alone. Pre-incubation of seeds in KNO3 for 4 to 7 dbefore the ethylene applications significantly increased germination.The effects of the KNO3 pre-incubation were additive at eachof four ethylene concentrations (0.1–100 µ11–1).Potassium nitrate was effective only when ethylene followedthe KNO3 pre-incubation period. Potassium nitrite stimulatedabout 25 per cent of the seeds to germinate without a pre-incubationperiod and without ethylene. Also, ethylene plus KNO2 enhancedgermination above that achieved by either stimulus alone. Silvernitrate did not block the ethylene promotion of germination,but reversed the typical ethylene inhibition of seedling growthfollowing germination. The results support the views that nitrateexerted its effect via conversion to nitrite within the seedand that the rate of nitrate conversion may be a limiting factorin the dark germination of common purslane seeds. Predominantly linear relations between germination rate andtemperature (over specific ranges) have hitherto been foundamong the main crop plants. Given linearity, accumulated temperatureabove a base (Tb) is a reliable predictor of germination time.In contrast, this paper shows marked non-linearities in theresponse of germination to temperature for four oilseed rapecultivars, two ‘double high’ (Martina and Askari)and ‘two double low’ (Rocket and Express). Linearregressions of 1/time to germination of given percentiles indicateTb is about 3 C for all cultivars. However, consistent deviationsfrom linearity occur especially at low temperature, such thatgermination time at 5C is up to twice as fast as predictedfrom a linear model. Moreover, two of the cultivars show anincreasing proportion of seeds not germinating as temperatureis lowered. These seeds were still viable as demonstrated bytheir ability to germinate when transferred to warmer temperature.The consequence of non-linearity was to increase the spreadof time over which seeds germinated at low temperature. Thispaper defines the effects and indicates the margin of errorlikely to follow from applying linear rate/temperature modelsto whole populations. The findings have implications for thepropensity of different oilseed rape cultivars to become volunteerweeds and feral populations. A 10-min irradiation with red light (R) of ‘Grand Rapids’lettuce (Lactuca sativa L.) seeds increased respiratory rates(QO2 and QCO2) over those of far red (FR)-irradiated seeds within2 to 3 hr. Differences in respiration between R- and FR-irradiatedseeds became more pronounced with time. The respiration, indarkness at 25?C, of seeds air dried after germination promotingimpregnation treatments with 1.0 mM GA3+0.5 mM kinetin in acetonewas stimulated slightly by the 6th hour after planting and markedlyby the 12th and 24th hour. Results with seeds in GA3+kinetindissolved in water were similar. ABA, which inhibits germinationand seedling growth, prevented stimulation in respiration oflight-induced seeds. ABA also repressed respiration when usedwith GA3+kinetin. The germination and growth retardant, isopropylN-(3-chlorophenyl) carbamate (CIPC), did not affect the respirationof either R- or FR-treated seeds. The effects of red light, far-red light, Gibberellin A3, andethephon were studied on the germination of lettuce seeds cv.Grand Rapids harvested at different stages of development. Seeds did not become capable of germination until 8 days afteranthesis. Red light promoted seed germination from the age of8–9 days following anthesis up to the newly mature stage.Ten or 11 days following anthesis, a large percentage of seedsbecame capable of germination in the dark and therefore couldbe considered not dormant. They were affected by far-red light,but less so than the mature seeds. The effect of light on the germination of developing seeds appearedto be similar to the known light effect on mature lettuce seedgermination. The rapid and uniform establishment of soya bean [Glycine max(L.) Merr.] stands is conducive to higher yields. This studywas undertaken to determine the effects of cultivar, temperature,and seed size on the rate of germination and emergence. No cultivar effect on the germination rate was observed. However,in an emergence study from a sand-soil-peat mixture, cultivardifferences in emergence rates were noted(‘Chippewa 64’> ‘Wayne’ > ‘Amsoy 71’). In anotheremergence study (sand media) the cvs ‘Calland’ and‘Williams’ emerged faster than the cv. 'Wayne or‘Wells’. Time required for 50 per cent germination decreased (18.8–4.0days) as the temperature increased from 10 to 30 °C (5 °Cincrements). Emergence (50 per cent) from a sand-soil-peat mixturewas more rapid (19.8–6.3 days) as the simulated plantingdate (growth chamber set to simulate field temperatures) wasdelayed from 16 April to 15 June with an intermediate date of16 May. In addition, time required for 50 per cent emergence of thecultivars from sand decreased (793–76 h) as the temperaturewas increased from 10 to 30 °C with no decrease from 30to 35 °C. Seed size effects were apparent, with the very small seed germinatingslower than the three larger seed sizes. In the emergence studieswith both the sand and sand-soil-peat mixture there was a generaltrend toward more rapid emergence with the smaller seeds. However,the absolute differences were small. Significant cultivar x temperature interactions were observedfor the germination and emergence rates. In most cases the cultivarsmerged in terms of germination and emergence rates at temperaturesbetween 10 and 20 °C and at the higher temperatures thecultivar rankings were different from those observed at temperaturesbelow the merging point. Plants of the biennial Arctium tomentosum were grown from seedto seed-set in an open field under three different treatments:control plants receiving full light intensity, plants with aleaf area reduced by 45 per cent, and shaded plants receivingonly 20 per cent of natural illumination. At various stagesof development the youngest fully expanded leaf of one plantin each treatment was exposed to 14CO2 for half an hour. Subsequentdistribution of labelled assimilates in various plant partswas determined after eight hours. In the first year, the mostdominant sink was the tap root irrespective of variation inassimilate supply. During the production of new vegetative growthin the second season, a larger amount of radioactive photosynthatewas recovered from above ground parts, especially during formationof lateral branches. Seed filling consumed 80–90 per centof labelled carbon exported from the exposed leaf. In the secondyear, the most pronounced difference between treatments wasin the degree of apical dominance, being highest in shaded plantsand lowest in the plants with cut leaves. Results from 14C experimentsagreed fairly well with a ‘partitioning coefficient’derived from a growth analysis of plants grown independentlyunder the same experimental conditions. Under submerged conditions the germination of maize seeds isinhibited. Under normal conditions, i.e. on moist filter paper,more than 80 per cent of the seeds can germinate whereas lessthan 24 per cent of the seeds germinate under submerged conditions.The inhibition of germination under submerged conditions canbe fully overcome by the application of an appropriate concentrationof GA2, and KN. The effect of the ‘leafless’ mutation (in whichtendrils replace leaflets and the stipules are reduced to avestigial form) upon foliage area, photosynthetic net CO2 uptakepotential, dry matter production and seed yield in Pisum sativumwas studied by comparing two near-isogenic lines of genotypeafafstst and ++++. The mutation is of potential agronomic valuein that it offers improved lodging resistance, crop drying andharvester throughput. In the conventional phenotype the total foliage area of themain axis attained a plateau (456 cm2) at day 56 from seedlingemergence, whereas corresponding values for the ‘leafless’mutant showed a total area of 208 cm2 at day 68 with no indicationof a plateau. The agronomic consequence of this is discussed.During the vegetative phase of the plant the maximum CO2 uptakepotential in the fully expanded conventional leaf was 8·5mg CO2 leaf–1 h–1 and in the ‘leafless’mutant this value was 7·0 mg CO2 leaf–1 h–1.For most ‘leaves’ of the latter phenotype this valuewas between 30 and 60 per cent less than for their conventionalcounterpart. There was a consistently higher photosyntheticpotential per unit area in tendrils of the ‘leafless’mutant than in leaflets of the conventional phenotype. The respectivemean specific values for the two phenotypes were 53 and 37 mgCO2 dm–2 h–1. The problem of obtaining a meaningfulsurface area value for tendrils is discussed and the cylindricalnature of tendrils is taken into account. The ‘leafless’ mutant consistently accumulated 50per cent less dry matter than did conventional plants in theperiod from seedling emergence to anthesis and yield of maturedry seed per plant showed a reduction of 50 per cent both inseed number and total seed weight. The implications for future breeding and selection programmesaimed at haulm reduction are discussed in relation to evaluatingthe ability of the background genotype to produce adequate tendrilsin the presence of afafstst. Positive linear relationships were shown between constant temperaturesand the rates of progress of germination to different percentiles,G, for single populations of each of five genotypes of chickpea(Cicer anetinum L.). The base temperature, Tb, at which therate of germination is zero, was 0·0°C for all germinationpercentiles of all genotypes. The optimum temperature, To(G),at which rate of germination is most rapid, varied between thefive genotypes and also between percentiles within at leastone population. Over the sub-optimal temperature range, i.e.from Tb to To(G), the distribution of thermal times within eachpopulation was normal. Consequently a single equation was appliedto describe the influence of sub-optimal temperatures on rateof germination of all seeds within each population of each genotype.The precision with which optimum temperature, Tb(G), could bedefined varied between populations. In each of three genotypesthere was a negative linear relationship between temperatureabove Tb(G) and rate of germination. For all seeds within anyof these three populations thermal time at supra-optimal temperatureswas constant. Variation in the time taken to germinate at supra-optimaltemperatures was a consequence of normal variation in the ceilingtemperature, To(G)—the temperature at or above which rateof progress to germination percentile G is zero. A new approachto defining the response of seed germination rate to temperatureis proposed for use in germplasm screening programmes. In two populations final percentage germination was influencedby temperature. The optimum constant temperature for maximumfinal germination was between 10°C and 15°C in thesepopulations; approximately 15°C cooler than the optimumtemperature for rate of germination. The bases of differences in germination rates (GRg = inverseof time to germination [tg] of percentage g) among three cold/salt-toleranttomato (Lycopersicon esculentum Mill.) accessions (PI 341988,PI 120256, and PI 174263) and one cold/salt-sensitive tomatocultivar (T5) were investigated. The effects of seed priming(6 d imbibition in aerated –1.2 MPa polyethylene glycolsolution at 20 ?C followed by redrying) and of removing theendosperm/testa cap covering the radicle on the temperaturesensitivity of GRg, and the interaction of these treatmentswith genotypes, were also examined. GRg decreased linearly withdecreasing temperature for all genotypes and seed treatments.The minimum or base temperatures for germination (Tb) variedby 1 ?C among the tomato lines, so genotypic differences inGRg were due to differing thermal time requirements for germination.The mean thermal time requirement for germination of T5 seeds was 22% and 19% greater than that of PI 341988 andPI 120256 seeds, respectively, but only 9% greater than thatof PI 174263 seeds. Seed priming did not lower Tb of any genotype,but significantly reduced by 24, 49, 41, and 49% in T5, PI 341988, PI 120256, and PI 174263, respectively,indicating that priming increased the rate at which the seedsprogressed towards germination when T>Tb, but did not lowerthe minimum temperature at which germination could occur. Primingincreased the GRg of T5 seeds to equal or exceed those of control(non-primed) seeds of the cold/salt-tolerant genotypes at anyT>Tb, but the PI lines exhibited an even greater responseto priming. Times to germination within each seed lot were normallydistributed on a logarithmic scale. Priming increased the variancein tg within a seed lot when compared to control seeds. However,the variation in thermal time for germination between the 10thand 90th percentiles of the seed population (T(10–90))was relatively unaffected by priming due to the reduction in in primed seeds. Removing the endosperm cap and testa opposite the radicle tip decreased almost 6-fold and and reduced Tb by 5 ?C in T5 and PI 341988,implicating processes in the endosperm/testa as the limitingfactors in germination at suboptimal temperatures. In this paper we report for the first time the occurrence ofan inducible weak CAM in leaves of Talinwn triangulare (Jacq.)Willd. This plant is a terrestrial perennial deciduous herbwith woody stems and succulent leaves which grows under fullexposure and in the shade in northern Venezuela. Plants grownin a greenhouse (‘sun’ plants) and a growth cabinet(‘shade’ plants) with daily irrigation showed CO2uptake only during the daytime (maximum rate, 4?0 µmolm–2 s–1) and a small acid accumulation during thenight (6?0 µmol H+g–1 FW). Twenty-four hours aftercessation of irrigation, no CO2 exchange was observed duringpart of the night. Dark fixation reached a maximum (1?0 µmolCO2 m–2 s–1, 100 µmol H+ g–1 FW) onday 9 of drought. By day 30 almost no gas exchange was observed,while acid accumulation was still 10 µmol H+ g–1FW. Rewatering reverted the pattern of CO2 exchange to thatof a C3 plant within 24 h. Daytime and night-time phosphoenolpyruvatecarboxylase activity increased up to 100% (shade) and 62% (sun)of control values after 10 and 15 d of drought, respectively.Light compensation point and saturating irradiance were similarin well-watered sun and shade plants, values being characteristicof sun plants. CAM seems to be important for the tolerance ofplants of this species to moderately prolonged (up to 2 months)periods of drought in conditions of full exposure as well asshade, and also for regaining high photosynthetic rates shortlyafter irrigation. Changes in Lipoxygenase Components of Rice Seedlings during Germination   总被引:1,自引:0,他引:1  
Changes in lipoxygenase (LOX) activity were followed duringthe germination of rice seeds. The enzyme activity of 3-day-oldseedlings was 20 times higher than that of ungerminated seeds.Sixty per cent of the increased activity was found in shoots.The increase in LOX activity was mainly due to an increase inlipoxygenase-2 (LOX-2), a minor component in ungerminated seeds;this increase was inhibited by cycloheximide. LOX-2 was isolatedfrom the 3-day-old seedlings and compared for its enzymologicalproperties with rice lipoxygenase-3 (LOX-3), a major componentin ungerminated seeds. Both LOX-2 and LOX-3 were stable at pH5 to 8, but LOX-2 was more heatstable than LOX-3. Apparent Kmvalues of LOX-2 and LOX-3 for linoleic acid were 170 and 59µM, and those for linolenic acid were 5,300 and 88 µM,respectively. Both LOXs were inhibited by some metal ions andantioxidants. Previous work on chlorophyll-deficiency mutations in pea andbarley has shown that a significant increase in mutations isinduced by storing seeds under various conditions which leadto losses of viability to about 50 per cent. The work here showsthat a detectable increase in mutation frequency is also associatedwith much smaller losses of viability. Pea seeds were storedat 35 °C and 16.5 per cent moisture content for 40 and 57d when viability fell from 99 to 93 and 82 per cent, respectively.At the same time mutation frequency (percentage of seeds containingrecessive point mutations) increased from 1.62 per cent in thecontrol treatment to about 3 to 4 per cent. Barley seeds at15.5 per cent moisture content were stored at 50 °C for42 and 54 h, and at 35 °C for 28 and 39 d. During theseageing treatments viability fell from 98 to 75, 26, 93 and 48per cent respectively and the mutation frequency increased fromzero to between about 0.3 to 0.9 per cent. In both species theinduction of mutation by ageing treatments was significant butthe differences between the various ageing treatments were not.It is concluded that there is probably no safe threshold lossof viability which completely avoids mutation, and these resultssupport the view that for genetic conservation seeds shouldbe stored under conditions which minimise loss of viability.
