From 14 values reported for nine species of polychaetes, a highly significant (P < 0.0005) multiple correlation (r = 0.987) between the nitrogen content of food (N) and absorption efficiency (Ae) is observed and is adequately fitted by the polynomial cubic equation: Ae = 11.744−13.353 N+6.510 N2−0.458 N3.  相似文献

The tumor suppressor protein p53 is a key regulatory element in the cell and is regarded as the “guardian of the genome”. Much of the present knowledge of p53 function has come from studies of transgenic mice in which the p53 gene has undergone a targeted deletion. In order to provide additional insight into the impact on the cellular regulatory networks associated with the loss of this gene, microarray technology was utilized to assess gene expression in tissues from both the p53−/− and p53+/− mice. Six male mice from each genotype (p53+/+, p53+/−, and p53−/−) were humanely killed and the tissues processed for microarray analysis. The initial studies have been performed in the liver for which the Dunnett test revealed 1406 genes to be differentially expressed between p53+/+ and p53+/− or between p53+/+ and p53−/− at the level of p ≤ 0.05. Both genes with increased expression and decreased expression were identified in p53+/− and in p53−/− mice. Most notable in the gene list derived from the p53+/− mice was the significant reduction in p53 mRNA. In the p53−/− mice, not only was there reduced expression of the p53 genes on the array, but genes associated with DNA repair, apoptosis, and cell proliferation were differentially expressed, as expected. However, altered expression was noted for many genes in the Cdc42-GTPase pathways that influence cell proliferation. This may indicate that alternate pathways are brought into play in the unperturbed liver when loss or reduction in p53 levels occurs.  相似文献

The first step in the activation of the anti-retroviral nucleoside analogue azidothymidine (AZT) involves its conversion to a 5′-monophosphate. In this study, we have evaluated the role of cytosolic thymidine kinase (Tk), the major enzyme involved in phosphorylating thymidine and its analogues, in the nuclear DNA damage produced by AZT in neonatal mice. Tk+/+, Tk+/− and Tk−/− mice were treated intraperitoneally with 200 mg/kg/day of AZT on postnatal days 1 through 8, and micronuclei were measured in peripheral blood 24 h after the last dose. AZT treatment increased the micronucleus (MN) frequencies to similar extents in both the reticulocytes (RETs) and normochromatic erythrocytes (NCEs) of Tk+/+ and Tk+/− mice; AZT did not increase the frequency of micronucleated RETs (MN-RETs) or micronucleated NCEs (MN-NCEs) in Tk−/− mice. Unexpectedly, neonatal Tk−/− mice treated with the vehicle had significantly elevated MN frequencies for both RETs and NCEs relative to Tk+/+ and Tk+/− mice (e.g., 3.4% MN-RETs and 4.8% MN-NCEs in Tk−/− mice versus 0.7 and 0.6% MN-RETs and MN-NCEs in neonatal Tk+/+ mice). Additional assays performed on untreated Tk−/− mice showed that elevated spontaneous MN frequencies persisted until at least 20 weeks of age, which approaches the average lifespan of Tk−/− mice. These results indicate that metabolism by Tk is necessary for the genotoxicity of AZT in neonatal mice; however, the genotoxicity of AZT is not altered by reducing the Tk gene dose by half. The elevated spontaneous MN frequencies in Tk−/− mice suggest the presence of an endogenous genotoxic activity in these mice.  相似文献

To examine the effect of compound deficiencies in antioxidant defense, we have generated mice (Sod2+/−/Gpx1−/−) that are deficient in Mn superoxide dismutase (MnSOD) and glutathione peroxidase 1 (Gpx1) by breeding Sod2+/− and Gpx1−/− mice together. Although Sod2+/−/Gpx1−/− mice showed a 50% reduction in MnSOD and no detectable Gpx1 activity in either mitochondria or cytosol in all tissues, they were viable and appeared normal. Fibroblasts isolated from Sod2+/−/Gpx1−/− mice were more sensitive (4- to 6-fold) to oxidative stress (t-butyl hydroperoxide or γ irradiation) than fibroblasts from wild-type mice, and were twice as sensitive as cells from Sod2+/− or Gpx1−/− mice. Whole-animal studies demonstrated that survival of the Sod2+/−/Gpx1−/− mice in response to whole body γ irradiation or paraquat administration was also reduced compared with that of wild-type, Sod2+/−, or Gpx1−/− mice. Similarly, endogenous oxidative stress induced by cardiac ischemia/reperfusion injury led to greater apoptosis in heart tissue from the Sod2+/−/Gpx1−/− mice than in that from mice deficient in either MnSOD or Gpx1 alone. These data show that Sod2+/−/Gpx1−/− mice, deficient in two mitochondrial antioxidant enzymes, have significantly enhanced sensitivity to oxidative stress induced by exogenous insults and to endogenous oxidative stress compared with either wild-type mice or mice deficient in either MnSOD or Gpx1 alone.  相似文献

We have derived a comprehensive structure–activity relationship (SAR) picture for a new series of natural acetylcholinesterase inhibitors isolated from Sarcococca saligna. A set of 32 previously isolated and tested pregnane-type steroidal alkaloids inhibitors were investigated with respect to their IC50 values (pIC50) against the AChE enzyme in order to derive CoMFA models using atom-based alignment. A highly significant CoMFA model was obtained with r2 value of 0.974. The q2 (cross validation r2) value also confirms the statistical significance of our model.  相似文献

Smokers are exposed to a number of carcinogenic compounds including aromatic amines such as 4-aminobiphenyl. Antioxidants are thought to be involved in the defence against the damaging effect of such carcinogens. Recently it has been shown that plasma antioxidant status in smokers is diminished compared with non-smokers. In this study we investigated in 40 smokers whether the trolox equivalent antioxidant capacity (TEAC) in plasma could be quantitatively related to exposure to cigarette smoke. The biomarkers 4-aminobiphenylhaemoglobin (4-ABP-Hb) adduct and cotinine were determined as indices of cigarette smoke exposure. A correlation between 4-ABP-Hb adduct levels and plasma cotinine levels was found for the whole population studied, who smoked 4-70 cigarettes per day (n = 40, r2 = 0.12, p = 0.03). A significant inverse relationship was found between TEAC and 4-ABP-Hb levels (n = 40, r2 = 0.17, p = 0.008). Multiple regression analysis showed a strong relationship between 4-ABP-Hb levels and plasma TEAC and cotinine levels (n = 40, r2 =0.29, p = 0.002). These findings provide strong evidence that the 4- ABP-Hb adduct represents a valuable biomarker of (internal) exposure to tobacco smoke, and also that the formation of this marker is dependent on the plasma antioxidant status. The multiple regression analysis results show that the measure of effect (4-ABP-Hb adduct formation) is largely determined by dose (cotinine) and protection (TEAC).  相似文献

Juvenile English sole were exposed intramuscularly to nitrofurantoin (NF) and the levels of 8-hydroxy-2′deoxyguanosine (8-OH-dG) in liver, kidney and blood were determined using reversed-phase HPLC with electrochemical detection. Identification and quantitation of the 8-OH-dG in the samples was accomplished by comparison with standard 8-OH-dG, which was characterized by UV spectroscopy and fast-atom bombardment mass spectrometry. The levels of hepatic 8-OH-dG increased (r2 = 0.59, P = 0.015) with the dose of NF (0.10 – 10 mg NF/kg fish). In kidney and blood, however, the levels of 8-OH-dG were significantly higher than controls only at the highest dose tested. The level of binding in liver ranged from 0.37 to 0.76 fmol 8-OH-dG/μg DNA. The levels of hepatic 8-OH-dG reached a maximum (approx. 1 fmol 8-OH-dG/μg DNA) between 1 and 3 days after exposure, followed by a decrease to control levels (approx. 0.25 fmol 8-OH-dG/μg DNA) at 5 days post-exposure. These data demonstrate the first direct evidence for the formation of oxidized DNA bases resulting from the metabolism of a nitroaromatic compound by fish.  相似文献

It was our objective to develop a rapid, sensitive and specific assay to quantify the immunosuppressive macrolide 40-O-(2-hydroxyethyl)rapamycin (SDZ-RAD) in blood of transplant patients. SDZ-RAD was extracted from blood by solid–liquid extraction. SDZ-RAD and its internal standard 28,40-diacetyl rapamycin were quantified using HPLC–electrospray MS. The assay was linear from 0.1 to 100 μg/l (r2=0.99). The mean recovery was 83% for SDZ-RAD and 80.5% for the internal standard. The mean day-to-day precision was 8.0%. Extracted samples were stable at 20°C for at least 48 h and SDZ-RAD blood samples at −80°C for at least six months.  相似文献

(1) Exercise-induced hyperthermia is associated with a gradual slowing of the electroencephalogram (EEG), an increase in perceived exertion (RPE) and a lowering of the cerebral perfusion.

(2) During exercise EEG changes were linearly correlated to core temperature (r2=0.67; P<0.05) and RPE (r2 =0.54, P<0.05), but manipulation of cerebral perfusion by voluntary breathing efforts and by CO2 inhalation did not alter RPE or EEG.

(3) In conclusion EEG changes with hyperthermic exercise are not a simple effect of the reduced cerebral perfusion but may relate to the fatigue that arises concomitantly with the increases in core and brain temperatures.  相似文献

The effect of guanidinium chloride solutions on the circular dichroism of native (ZnZn-) and apophospholipase C (Bacillus cereus) indicated marked protein unfolding at denaturant concentrations of 1.4–1.8 M and 0.1–0.6 M, respectively. With the apoenzyme near u.V. region circular dichroism bands remained even after all ordered structure appeared to have been lost. Apophospholipase C bound two equivalents of Ni2+, Cd2+, Co2+, Mn2, Pb2+ or Cu2−, with only the latter metal causing marked changes either in circular dichroism or protein fluorescence relative to the native enzyme. Stability in guanidinium chloride for the metalloforms of phospholipase C decreased in the order: ZnZn->ZnCo->NiNi->CoCo->PbPb->CdCd->MnMn-apoenzyme.  相似文献

Twenty-one derivatives of taxchinin A (1) and brevifoliol (2) were synthesized and evaluated for cytotoxicity against human non-small lung cancer (A549) cell line. Nine derivatives showed potent activity with IC50 values from 0.48 to 6.22 μM. 5-Oxo-13-TBDMS-taxchinin A (11) and 5-oxo-13,15-epoxy-13-epi-taxchinin A (15) are the most potent derivatives, with IC50 at 0.48 and 0.75 μM, respectively. The structure–activity relationship (SAR) of these compounds established that exocyclic unsaturated ketone at ring C is the key structural element for the activity, while the ,β-unsaturated ketone positioned at ring A has no effect for the activity. The significant cytotoxicity of derivatives 11 and 15 may be due to the conformational change in the taxane rings. The 3D-QSAR study was conducted on this series of compounds, which provided optimal predictive comparative molecular field (CoMFA) model with cross-validated r2 (q2) value of 0.64.  相似文献

Swimming dynamics of the giant Australian cuttlefish, Sepia apama, were investigated using swimtunnel respirometry. Relationships between jet pressure, fin frequency, swimming speed and oxygen consumption were defined. Laboratory calibration of swimming parameters is necessary to allow estimates of swimming costs in the field.

Jet pressure was the best predictor of oxygen consumption with an averaged equation of MO2 = 722 (jet pressure) + 107 r2 = 0.51. Individually, fin frequency and jet pressure correlated highly to swimming speed, but due to the complicated usage of finning and jetting, the correlation between swimming speed and oxygen consumption was weaker. Cuttlefish were not optimal swimtunnel subjects and could not swim at high speeds for extended periods. At 15°C and a swimming speed of 0.06 m s-1, the gross cost of transport was calculated to be 10.1 kg-1 m -1, with a net cost of 4.1 kg-1 m-1.  相似文献

Hirudin N-terminal core domain residues 1–43 (r-Hir1–43) were prepared from limited proteolysis of recombinant hirudin by V8 Staphylococcal protease followed by purification with reversed-phase HPLC. r-Hir1-43 lacks the putative reactive site of hirudin (Lys47), but binds to thrombin (with Ki of 300 nM) and blocks the catalytic activity of the protease. The structural element which accounts for the thrombin inhibitory activity of r-Hir1–43 is analyzed in this report.  相似文献

A novel neurokinin-1 receptor antagonist, (±)-(1R*,3S*,4S*,5S*)-4-[(N-(2-methoxy-5-trifluoromethoxybenzyl)amino]-3-phenyl-2-aza-7-oxabicyclo[3.3.0]octane (1), was synthesized stereoselectively using Padwa’s intramolecular 1,3-dipolar cycloaddition methodology as the key step. Compound (±)-1 showed high affinity for the NK-1 receptors in human IM-9 cells with an IC50 value of 0.22 nM. A simple flow injection chemiluminescence method with synergistic enhancement has been investigated for the rapid and sensitive determination of azithromycin. The synergistic action was significant in the chemiluminescence system of luminol–hydrogen peroxide with azithromycin as an enhancer. The enhanced chemiluminescence intensity was linear with the concentration of azithromycin over the range from 0.1 pg mL−1 to 1.0 ng mL−1 (r2=0.9988) with a detection limit (3σ) of 0.04 pg mL−1. At a flow rate of 2.0 mL min−1, a complete analytical process could be performed within 0.5 min, including sampling and washing, with a relative standard deviation of less than 3.0%. The proposed method was applied successfully in the assay of azithromycin in pharmaceutical preparations, human urine and serum without any pre-treatment procedure.  相似文献

Comparative measurements of bacterial total counts and volumes of flow cytometry (FCM), transmission electron (TEM), and epifluorescence microscopy (EFM), were undertaken during a four week mesocosm experiment. Total counts of bacteria measured by TEM, EFM, and FCM were in the range of 1 · 106−6 cells ml−1, 1 · 106−3 · 1016 cells ml−1, and 5 · 105 cells ml−1 respectively. The mean volume of the bacterial community, measured by means of EFM and TEM, increased from 0.12–0.15 μm3 at the start of the experiment to 0.39–0.53 μm3 at the end. Generally, there was good agreement between the two methods and regression analyses gave r = 0.87 (p < < 0.01) for cell volume and r = 0.97 (p < < 0.01) for cell number. DAPI stained bacteria with volumes less than 0.2 μm3 were not detected by flow cytometry and these were generally an order of magnitude lower than counts made by TEM and EFM. For samples where the mean bacterial cell volume was longer than 0.3 μm3, all three methods were in agreement both with respect to counts and volume estimates.  相似文献

The viscous energy dissipation in a two generation model of the human bronchial tree is determined from inspiratory velocity and static pressure data obtained for large Reynolds numbers (104 < Re < 105). This dissipation is found to be an increasing function of both Re and distance downstream from the inlet of the model. The ratio of the dissipation in the model to the energy dissipation in an equivalent straight pipe system is determined. This ratio, Z*, for the model is compared to values in the literature for lower (laminar) Re. There is more dissipation in the branched model than in a straight pipe (Z* > 1) and turbulence keeps Z* at roughly a fixed value for large Reynolds numbers (104 < Re < 105). Z* values for curved pipes are also compared to the branching system values. It is found that the energy dissipation for the branched model behaves similarly to that in curved pipes.  相似文献

Qualitative estimates of the relative stability of hypothetical heterofullerenes C55Y5 (Y=Si, Ge, Sn, B, Al, N, P, SiH, GeH, SnH) and some η5-π-complexes LiC55Y5 were carried out by the MNDO method. Atoms Y (or groups XH) are assumed to substitute those C atoms in fullerene C60 which are located at the -positions of a separated pentagonal face (pent*) of this polyhedral molecule. It is shown that the spin densities in radicals C55Y5 (Y=SiH, GeH, SnH, B, Al, N, P) are localized on the separated pentagon atoms and the Li-pentagonal face (Li-pent*) bonds in η5-π-complexes of these radicals with the Li atom are considerably stronger than Li-pent* bonds in complexes [η5-π-LiC60]+ and [η5-π-LiC60] of unsubstituted C60. In addition, it is established that the Li-pent* bond energies in η5-π-complexes LiC55B5 and LiC55Al5 exceed the energy of the Li-pent* bond in the η5-π-complex LiC60H5 studied earlier. In contrast, the energies of similar bonds for Y=N, P are close to the energy of the Li-pent* bond in the η5-π-complex LiC60H5.  相似文献

Total syntheses of (±)-ovalicin, its C4(S*)-isomer 44, and C5-side chain intermediate 46 were accomplished via an intramolecular Heck reaction of (Z)-3-(tert-butyldimethylsilyloxy)-1-iodo-1,6-heptadiene and a catalytic amount of palladium acetate. Subsequent epoxidation, dihydroxylation, methylation, and oxidation led to (3S*,5R*,6R*)-5-methoxy-6-(tert-butyldimethylsilyloxy)-1-oxaspiro[2.5]octan-4-one (2), a reported intermediate. The addition of a side chain with cis-1-lithio-1,5-dimethyl-1,4-hexadiene (27) followed by oxidation afforded (±)-ovalicin. The functional group manipulation afforded a number of regio- and stereoisomers, which allow the synthesis of analogs for bioevaluation. The structure of 44 was firmly established via a single-crystal X-ray analysis. The stereochemistry at C4 generated from the addition reactions of alkenyllithium with ketones 2, 40, and 45 is dictated by C6-alkoxy functionality. Anti-trypanosomal activities of various ovalicin analogs and synthetic intermediates were evaluated, and C5-side chain analog, 46, shows the strongest activity. Compound 44 shows antiproliferative effect against HL-60 tumor cells in vitro. Compounds 46 and a precursor, (3S*,4R*,5R*,6R*)-5-methoxy-4-[(E)-(1′,5′-dimethylhexa-1′,4′-dienyl)]-6-(tert-butyldimethylsilyloxy)-1-oxaspiro[2.5]octan-4-ol (28), may be explored for the development of anti-parasitic drugs.  相似文献

Growth hormone (GH, 0.0025 and 0.025 I.U. ml−1) and γ-aminobutyric acid (GABA, 50 μg ml−1) enhance rotifer population growth in batch cultures. In order to further understand the mechanism of their actions, we conducted experiments culturing isolated females at low food and high free ammonia levels. At an optimum food level of 7×106 Nannochloropsis oculata cells ml−1 or at low free ammonia level of 2.4 μg ml−1, the F1 offspring of rotifers treated with GH at 0.0025 I.U. ml−1 had significantly higher population growth rate (r) and net reproduction rate (Ro), and shorter generation time than untreated rotifers. At a lower food level of 7×105 cells ml−1 or at high free ammonia level of 3.1 μg ml−1, rotifers treated with GABA at 50 μg ml−1 had significantly higher r and Ro, and shorter generation time. These results indicate that GABA is effective in enhancing rotifer reproduction when rotifers are cultured under stress whereas GH enhances rotifer reproduction when culture conditions are optimal. Significant effects were also observed in F1 and F2 generations which were not treated with hormones. These data may be useful for treating rotifer mass cultures to mitigate the effects of stress caused by high population densities.  相似文献
