Pure cultures of fifty-two species of plant pathogenic and saprophytic fungi isolated from orchard (cock's-foot) grass seed (Dactylis glomerata L.) were incubated either in total darkness or exposed to a diurnal cycle of near-ultraviolet (NUV) radiation (12 h NUV/12 h darkness). Twenty-four species sporulated only after exposure to NUV including seven species of Drechslera, five species of Fusarium, as well as species olAscochyta, Photna, Septoria, Pyrenochaeta, Rhynchophoma and Stagonospora; six species sporulated moderately in darkness but more profusely following exposure to NUV; twenty species sporulated whether they were irradiated or not; and only two species failed to sporulate. To assess the pathogenic fungal microflora of orchard grass seed accurately, seeds should be incubated under a daily regime that includes NUV to induce sporulation.  相似文献

Two-step degradation of pyrene by white-rot fungi and soil microorganisms   总被引:1,自引:0,他引:1  
  The effect of soil microorganisms on mineralization of 14C-labelled pyrene by white-rot fungi in solid-state fermentation was investigated. Two strains of white-rot fungi, Dichomitus squalens and a Pleurotus sp., were tested. The fungi were incubated on milled wheat straw contaminated with [14C]pyrene for 15 weeks. CO2 and 14CO2 liberated from the cultures were determined weekly. To study the effect of soil microorganisms on respiration and [14C]pyrene mineralization in different periods of fungal development, the fungal substrate was covered with soil at different times of incubation (after 0, 1, 3, 5, 7, 9 or 11 weeks). The two fungi showed contrasting ecological behaviour in competition with the soil microflora. Pleurotus sp. was highly resistant to microbial attack and had the ability to penetrate the soil. D. squalens was less competitive and did not colonize the soil. The resistance of the fungus was dependent on the duration of fungal preincubation. Mineralization of [14C]pyrene by mixed cultures of D. squalens and soil microorganisms was higher than by the fungus or the soil microflora alone when soil was added after 3 weeks of incubation or later. With Pleurotus sp., the mineralization of [14C]pyrene was enhanced by the soil microflora irrespective of the time of soil application. With D. squalens, which in pure culture mineralized less [14C]pyrene than did Pleurotus sp., the increase of [14C]pyrene mineralization caused by soil application was higher than with Pleurotus sp. Two bacterial isolates from parathion-amended flooded soil, Pseudomonas sp. and Bacillus sp., were examined for their ability to decompose nitrophenols. Uniformly labelled 14C-p-nitrophenol was metabolized by both bacteria, 14CO2 and nitrite being end products. A substantial portion (23% for Pseudomonas sp. and 80% for Bacillus sp.) of radioactivity applied as p-nitrophenol was accounted for as 14CO2 at the end of a 72-h period; 8 to 16% remained in the water phase after solvent extraction. Pseudomonas sp. produced nitrite also from 2,4-dinitrophenol, but only after a lag, and not from o- and m-nitrophenols. Interestingly, m-nitrophenol, known for its resistance to biodegradation because of meta substitution, was decomposed by Bacillus sp., resulting in the formation of nitrite and phenol; o-nitrophenol and 2,4-dinitrophenol resisted degradation by this bacterium.  相似文献

A procedure that consumes less screening time was developed for screening chickpea rhizosphere-competent bacteria for suppression of the chickpea pathogenic fungi Fusarium oxysporum f. sp. ciceri, Rhizoctonia bataticola and Pythium sp. Of the 478 bacteria obtained by random selection of the predominant, morphologically distinct colonies, 386 strains that effectively colonize chickpea roots could be divided broadly into three different groups. The first group consisted of 44 good chickpea rhizosphere colonizers with 107 to 108 colony-forming units (CFU)/g root; the second group consisted of 253 medium chickpea rhizosphere colonizers with 104 to 106 CFU/g root; and the third group consisted of 89 poor chickpea rhizosphere colonizers with 100 (nondetectable) to 103 CFU/g root. Forty-four Rifr strains from the first group of good chickpea rhizosphere colonizers were further screened for their in vitro biocontrol activity against F. oxysporum f. sp. ciceri, R. bataticola, and Pythium sp. One bacterial strain was selected for further work because of its unique ability to inhibit all three fungi and its good chickpea rhizosphere colonization ability. This is the first report of a single biocontrol bacterium active against three most devastating pathogenic fungi of chickpea. In a greenhouse test, chickpea seed bacterization with P. fluorescens NBRI1303 increased the germination of seedlings by 25%, reduced the number of diseased plants by 45%, compared with nonbacterized controls. Increases in seedling dry weight, shoot length, and root length ranged from 16% to 18%. Significant growth increases in shoot length, dry weight, and grain yield, averaging 11.59%, 17.58%, and 22.61% respectively above untreated controls, were attained in field trials in Agra and Jhansi. A rifampicin-resistant mutant P. fluorescens NBRI1303R of the P. fluorescens NBRI1303, used to monitor chickpea root colonization, confirmed the rapid and aggressive colonization by the bacterium, making it a potential biocontrol agent against chickpea phytopathogenic fungi. The results, demonstrating an increase in the efficiency of screening and detection of plant beneficial strains, should greatly benefit future studies. Seed treatment of Phaseolus mungo Roxb. with non-volatile metabolites of dominant seed fungi such as Chaetomium indicum Corda and Curvularia lunata (Walker) Boedijn and the antibiotic substances: chetomin, curvularin and brefeldin reduced the percent incidence of microfungi and bacteria but induced the Streptomyces sp. The bacterial microflora of nine varieties of witloof chicory (Cichorium intybus L. var.foliosum Hegi) seeds was studied. The 184 isolates were characterized by protein profiles determined by SDS-protein polyacrylamide gel electrophoresis of the total cell proteins. Isolates with identical protein profiles were grouped into one fingerprint type. Sixty-seven fingerprint types were distinguished. Two quantitatively major fingerprint types,Erwinia herbicola and an arthrobacter, represented 52% of the total number of isolates and were found on different chicory varieties. The latter organism was inhibited at seed germination. Other isolates, i.e.,Xanthomonas maltophilia, Pseudomonas paucimobilis, Agrobacterium radiobacter, Pseudomonas syringae, and a fluorescentPseudomonas, were only occasionally found. A minority were gram-positive isolates, i.e.,Bacillus sp.,Streptomyces sp., and coryneforms. In vitro activity of the isolates was tested against five fungi. Priming is a technique used to improve seedling establishment of direct-seeded crops such as onion and carrot, resulting in a quick and uniform emergence. This work investigated the application of four selected beneficial microorganisms (Pseudomonas chlororaphis MA342, Pseudomonas fluorescens CHA0, Clonostachys rosea IK726d11 and Trichoderma harzianum T22) to onion and carrot seed during drum priming, and their subsequent survival and establishment in the rhizosphere once the seed was planted. Different application rates of fungi (7 log10 cfu g−1 dry seed) and bacteria (6 log10 cfu g−1 dry seed) were required on onion to achieve the end target of 5 log10 cfu g−1 dry seed, whereas a lower rate (5 log10 cfu g−1 dry seed for both bacteria and fungi) was successful on carrot. Microorganism-treated seed was planted in soil in the glasshouse and root and rhizosphere soil samples were taken at 2, 4 and 8 weeks post-planting. All seed-applied microorganisms were recovered throughout the experiment, although differences in the survival patterns were seen. The bacterial isolates declined in number over time, with P. fluorescens CHA0 showing better overall survival than P. chlororaphis MA342, particularly on the roots and in the rhizosphere soil of carrot. In contrast to the bacteria, the fungal isolate C. rosea IK726d11 showed good survival on both onion and carrot, and increased significantly in number throughout the 8-week period. Trichoderma harzianum T22 remained relatively constant in number throughout the experiment, but showed better survival on carrot than onion roots. Newly-emerged ears of barley soon became colonised by micro-organisms with numbers increasing to 5·8 × 105 viable propagules/g dry weight by harvest. Bacteria were numerically predominant throughout. Yeasts and yeast-like fungi were also numerous during the early stages of grain development but were exceeded by propagules of filamentous fungi during ripening, with Cladosporium spp., Alternaria alternata, Epicoccum purpurascens and Verticillium lecanii most abundant. Numbers of most micro-organisms were greatest in a wet season but Alternaria was equally abundant in hot dry weather. Application of fungicides at or after anthesis modified the grain microflora. Captafol was most effective overall in decreasing numbers of fungi. However, benomyl decreased Cladosporium inoculum more than catpafol but was ineffective against Alternaria which became more numerous on benomyl-treated grain than on untreated. Yield increases of up to 4% were obtained by late fungicide treatment, least in a hot, dry year. Germination was also increased by some treatments by up to 5%.  相似文献

The bacterial flora on the heads of four different witloof chicory varieties was examined. The 590 isolates were characterized by their SDS-PAGE protein profiles; they revealed 149 different protein fingerprint types. The fluorescentPseudomonas fingerprint type CH001 was abundantly found on all heads examined. Fourteen other fingerprint types occurred in high densities more than twice. Among these, the following were identified: fluorescentPseudomonas, nonfluorescentPseudomonas sp.,Erwinia herbicola, Erwinia sp., andFlavobacterium sp. The majority of the fingerprint types (90%) was found only once. It was also our objective to isolate bacteria applicable in the biological control of chicory phytopathogens. Isolates of all fingerprint types were tested for in vitro antagonistic activity and for possible deleterious effect on plant growth. FluorescentPseudomonas andSerratia liquefaciens isolates were antagonistic against fungi. Among the 161 fluorescentPseudomonas strains, five were able to produce disease symptoms on chicory leaves upon inoculation. Comparison of the results of this study with those obtained in two previous analyses revealed that the leaf microflora showed some similarities with the bacterial flora of chicory roots. The chicory seed microflora differed from that of both leaves and roots.  相似文献

Five caesalpinioid legumes, Afzelia africana, Afzelia bella, Anthonotha macrophylla, Cryptosepalum tetraphylum and Paramacrolobium coeruleum, and one Euphorbiaceae species, Uapaca somon, with a considerable range in seed sizes, exhibited different responses to inoculation by four species of ectomycorrhizal (ECM) fungi, Scleroderma dictyosporum, S. verrucosum, Pisolithus sp. and one thelephoroid sp. in greenhouse conditions. Thelephoroid sp. efficiently colonized seedlings of all of the five caesalpinioid legumes except U. somon, but provided no more growth benefit than the other fungi. Thelephoroid sp. and S. dictyosporum colonized seedlings of U. somon poorly, but stimulated plant growth more than the other fungi. The relative mycorrhizal dependency (RMD) values of the caesalpinioid legumes were never higher than 50%, whilst U. somon had RMD values ranging from 84.6 to 88.6%, irrespective of the fungal species. The RMD values were negatively related to seed mass for all plant species. Potassium concentrations in leaves were more closely related than phosphorus to the stimulation of seedling biomass production by the ECM fungi. Our data support the hypothesis that African caesalpinioid legumes and euphorbe tree species with smaller seeds show higher RMD values than those with the larger seeds.  相似文献

A range of seed and flower characters was examined in 37 species of the New Zealand genus Celmisia (Asteraceae) to determine whether there were any relationships between reproductive features and the geographic or altitudinal ranges of the species. Using published sources and herbarium material, flower attractiveness to pollinators was measured in terms of capitulum size and ray/disc length ratio. Dispersibility was measured in terms of seed dry weight, seed terminal velocity, pappus/seed length ratio, and scape/leaf length ratio. Altitude (minimum and maximum) and geographic ranges (number of 1o latitude x 1o longitude squares occupied) were obtained from published sources. A multivariate principal components analysis using 12 morphological characters revealed the presence of five groups of species differing in capitulum size, seed weight, pappus length, number of pappus bristles, leaf length and scape length. Linear regressions showed that altitude was significantly and negatively related to seed dry weight and capitulum size, and positively to scape/leaf length ratio. Geographic range is negatively related to capitulum size, and positively to ray/disc ratio and altitudinal range. Amongst the seed features measured, seed weight was the best predictor of terminal velocity (an inverse measure of dispersability), but lighter seeds also had a higher pappus/seed length ratio. The frequency distribution of the geographic ranges of the species is highly skewed, with many local species (86% occurring in 15 or fewer one‐degree squares) and very few common ones. Two species (C. gracilenta and C. graminifolia) are notably more common than all the others. A comparison of these species with the others shows that, on average, they have lighter seeds with more bristles, taller scapes and higher ray/disc ratios, but have smaller capitula. The low geographic abundance of the rare species could not readily be attributed to any specific reproductive feature, possibly because of the multiple types of rarity represented within the genus.  相似文献

We examined the role ofarbuscular mycorrhizal fungi (AMF) in thebioprotection of the sand dune grass Leymus arenarius against soil fungi andnematodes. Six soil fungi (Fusariumnivale, Fusarium sp., Cladosporiumherbarum, Cladosporium sp., Phomasp., Sporothrix sp.) and four species ofnematodes (Pratylenchoidesmagnicauda, Paratylenchusmicrodorus, Rotylenchus goodeyi, Merlinius joctus) were isolated from a coastalsand dune in Iceland where a population of L. arenarius was declining in vigour. Acommercial AMF inoculum (Microbio Ltd. England)containing Glomus caledonium, G.fasciculatum, and G. mossae was used.Seedlings of L. arenarius were grownunder controlled conditions in sterile sand andsubjected to the following treatments: (1)control, (2) + AMF, (3) + AMF + soil fungi, (4)+ AMF + nematodes, (5) + AMF + nematodes + soilfungi, (6) + soil fungi, (7) + soil fungi +nematodes, (8) + nematodes. Mycorrhizal plantshad significantly the highest root dry weightof all treatments. Mycorrhizal plants hadsignificantly higher leaf dry weight thanplants in other treatments, with the exceptionof AMF inoculated plants exposed to nematodes. Mycorrhizal plants exposed to soil fungi andnematodes had significantly higher growthparameters except total number of leaves thanAMF inoculated plants exposed to both soilfungi and nematodes. Mycorrhizal plantssubjected to a dual application of soil fungiand nematodes did not vary significantly in anygrowth parameters from plants without AMF thatwere exposed to a dual application of soilfungi and nematodes. This suggests asynergistic effect of soil fungi and nematodesthat break down the protection of AMF againstpathogens. 2017年8月对拉萨河流域23个采样点的底栖动物群落和环境因子特征进行了调查,采集到大型底栖动物41属(种),其中水生昆虫35属(种),占85.4%;软体动物2属(种),占4.9%;环节动物3属(种),占7.3%;线形动物1属(种),占2.4%。各点位平均物种数为8种,优势种组成差别较大,优势种主要有四节蜉属(Baetis spp.)、大蚊属(Tipula sp.)、纹石蛾属(Hydopsychesp.)、短石蛾属(Brachycentrus sp.)、水丝蚓属(Limnodrilus sp.)、摇蚊属(Chironomus sp.)等,各采样点底栖动物平均密度为362只/m2。运用生物多样性指数分析底栖动物的群落结构,Marglef丰富度指数、香农威纳指数和均匀度指数平均值分别为1.18、1.32和0.49,生物多样性指数水平较低。采用均匀度指数法和底栖动物BI指数法进行了水质评价,两种评价结果显示分别有30.3%和56.5%的采样点水质为中污染至重污染状态。典范对应分析结果表明:影响拉萨河流域底栖动物群落的主要环境因子是海拔、化学需氧量、硫化物、p H值、总磷和总氮。  相似文献

Biomass and lipid productivity, lipid content, and quantitative and qualitative lipid composition are critical parameters in selecting microalgal species for commercial scale‐up production. This study compares lipid content and composition, and lipid and biomass productivity during logarithmic, late logarithmic, and stationary phase of Nannochloropsis sp., Isochrysis sp., Tetraselmis sp., and Rhodomonas sp. grown in L1‐, f/2‐, and K‐medium. Of the tested species, Tetraselmis sp. exhibited a lipid productivity of 3.9–4.8 g m?2 day?1 in any media type, with comparable lipid productivity by Nannochloropsis sp. and Isochrysis sp. when grown in L1‐medium. The dry biomass productivity of Tetraselmis sp. (33.1–45.0 g m?2 day?1) exceeded that of the other species by a factor 2–10. Of the organisms studied, Tetraselmis sp. had the best dry biomass and/or lipid production profile in large‐scale cultures. The present study provides a practical benchmark, which allows comparison of microalgal production systems with different footprints, as well as terrestrial systems. Biotechnol. Mechanically isolated hyphal walls of the rhipidiacean fungi Mindeniella spinospora Kanouse and Araiospora sp. (Oomycetes) were examined by biochemical, cytochemical and x-ray diffraction analyses. In both fungi, the most abundant wall constituents were 1 → 3- and 1 → 6-linked β-glucans accounting for 91% of wall dry weight in M. spinospora and 87% in Araiospora sp. In addition, hyphal walls of M. spinospora contained 1.7% mannose, 4.3% protein, 2.0% ash and 1.0% lipid. The quantities of these components in Araiospora sp. were 1.9%, 1.8%, 1.5%, and 1.3%, respectively. Both species had cellulose contents ranging from one-fifth to one-fourth of wall dry weight and chitin was apparently absent. In general, wall composition of both fungi is quite similar to that of the related species Sapromyces elongatus, lending support to the assertion that a biochemical dichotomy exists with respect to hyphal wall composition between Rhipidiaceae and Leptomitaceae, the two families comprising the order Leptomitales.  相似文献

Microbial growth was studied in six damp buildings. Mesophilic fungi, especially Penicillium spp., yeasts, and species of Cladosporium and Aspergillus, occurred most abundantly on building constructions. Thermophilic fungi and mesophilic actinomycetes were occasionally found. A toxigenic fungus, Stachybotrys sp., was also detected on cellulose-based materials. In a cytotoxicity test, 23% of samples were positive. Spore counts varied considerably on materials, but no correlation between counts and the substrate or its water activity (aw) was observed. In experiments a rapid increase in CO2 production and spore propagule count was observed in all materials incubated at a relative humidity (RH) (RH=0·01*water activity) of 96–98°. Some differences were noted between materials in CO2 evolved, but not in propagule counts.  相似文献

High incidence of diseases associated with the use of seeds saved from previous harvests as desire of maintaining local varieties with special attributes is of increased concern in wheat industry worldwide. Prevalent of seed-borne fungi in farmer-saved seeds and seed dressing fungicides to prevent infection from seeds to seedlings was studied in Northern Tanzania. One hundred and thirty five untreated farmer-saved seed lots were collected randomly from farmers. Alternaria alternata, Bipolaris sorokiniana, Drechslera tritici, Fusarium graminearum, Fusarium moniliforme, Aspergillus flavus, Cladosporium sphaerospermum, Epicoccum purpurascens, Pyricularia oryzae and Penicillium corylophilum were fungi isolated in farmer-saved seeds. Mean seed infection was 29% causing average grain yield loss of 1.2 mt/ha?1. Seed dressing with Metalaxy plus (Methyl carboxaitide), Mancozeb (Manganase-zinc salt) and Baytan (Chlorophenoxy ethanol) increased seed germination by 14, 13 and 17%, respectively, and grain yield by 28, 20 and 18%, respectively. Laguna Grande, Monagas State, Venezuela, is a shallow, V-shaped lake created by the confluence of two rivers. Montrichardia arborescens (L.) Schott. dominated wetlands cover most of the north and south arms and the littoral zone of the main body of the lake. The vegetation and seed banks of Montrichardia wetland sites were sampled in the north arm, south arm and main body five times from the end of the dry season in 1991 to beginning of the rainy season in 1992. The composition of the vegetation was similar and changed very little at all three sites during the course of the study. These wetlands had 53 species. Besides M. arborescens, other common species were Hamelia patens Jacq., Mikania cordifolia (L.) Wild., Sarcostemma clausum (Jacq.) Roem. & Schult., and Vitis caribaea L. In both the vegetation and seed banks, species richness was highest during the dry season. Altogether, the seed banks contained the seeds of 61 species of which 35 were also found in the vegetation. Seeds of three tree species were found in the seed banks that did not grow anywhere in the lake. In the seed bank, seeds of Cyperus odoratus L., Eleocharis interstincta (Vahl.) R&S, Ludwigia hyssopifolia (G. Don.) Excell, L. lithospermifolia (Mich.) Hara, Polygonum acuminatum H.B.K., and Sacciolepis striata (L.) Nash were the most abundant. Mean total seed density over the entire study was 6,500, 3,800, and 6,000 seeds/m2 in the north arm, south arm, and main basin, respectively. Seed production and dispersal occur primarily during the dry season, and the highest seed densities at all sites were found in the dry season when there was no or little standing water. The lowest seed densities at all sites were found during the rainy season during which seed densities declined over 80% at the north and south arm sites. In the main body of the lake, however, seed densities during the rainy season, although lower than during the dry season, actually increased significantly from 3,600 seeds/m2 in August 1991 to 6,000 seeds/m2 in October 1991. A significant decrease in seed density in either the north or south arms or both and a significant increase in the main body site during the rainy season occurred for 5 of the 8 species whose seeds were the most abundant, for all life-form guilds, except hydrophytes and for the entire seed bank. Secondary dispersal by water currents during the rainy season appears to be transporting seeds from the north and south arms into the seed bank of the main body of the lake.  相似文献

Sixteen species of fungi were isolated from the pericarp, endosperm, scutellum, and embryo of low (<75% germination), moderate (75–89%), and high vigor (>89%) seedlots of supersweet sweet corn (sh2) hybrids over two years. Most frequently isolated species werePenicillium oxalicum, Rhizopus arrhizus (14.5%), andRhizopus spp (17.4%).Fusarium moniliforme, a common inhabitant in field or dent corn, was isolated from only 2.4% of the samples. The low mean isolation values may be the result of poor conditions for infection or the data may reflect the methods used including sterilization techniques or random tissue selection.Aspergillus niger, F. moniliforme, andP. oxalicum, were isolated at a significantly greater level than other fungi from the high vigor hybrids at 0.89, 0.46 m and 4.46 respectively compared to 0.04, 0, and 1.82 for the low vigor hybrids. When Total Fungi were compared mean isolations were significantly greater from the high vigor hybrids at 11.96, the moderate 8.79, or low 4.86 vigor groups. When data from seed sources for all vigor groups were compared, significantly greater mean isolations were obtained from Illinois Foundation Seed hybrids forCladosporium sp,R. arrhizus, andRhizopus spp., but greater rates were obtained forFusarium oxysporum from the Asgrow hybrids. Isolation frequencies for the 16 species were not significantly different between the seed tissue types from any of the hybrids evaluated during this investigation. Results from this study showed that there is a diverse group of fungi present within thesh2 seed and seed treatments must be developed which will minimize seed rot and seedling blight from both internal seedborne and external pathogens.  相似文献

The effects of climatic factors on the growth, reproductive development and seed yield of cowpea (cv. K 2809) were investigated in controlled environment cabinets. Plants were grown to maturity in eight environments comprising all combinations of two day lengths (11 h 40 min and 13 h 20 min), two day (27 and 33 oC) and two night (19 and 24 oC) temperatures. The plants were nodulated (Rhizobium strain CB 756) and received 197 ppm N throughout growth. Treatments changed the time to the appearance of first flowers by a maximum of 6 days but the later-flowering plants more than doubled their dry weight during this period, so that effects on plant form and, ultimately, seed production were considerable. Warm nights (24 oC) not only hastened the onset of flowering but also enhanced dry matter production during the pre-flowering period; they did not extend the total growing period. Warm days (33 oC) did not enhance dry matter production but shortened the duration of the growing period by an average of 21 days (20%). Variation in final seed yield was mainly due to differences in the number of pods borne on branches. Warm days markedly decreased the number of pods per plant (an overall average reduction of 49%) as did warm nights in conjunction with the long (13 h 20) daylength. The number of seeds per pod was effected only by daylength (8.3 and 7.6 seeds in the long and short daylengths, respectively). Mean seed weight was decreased by 19% in warm as compared to cool nights but was increased by 18% in warm as compared to cool days. These responses are compared with those obtained with soyabean cv. TK5 in a previous experiment and are shown, in general, to be similar.  相似文献
