Rosa damascena Mill. is cultivated for its high-value essential oil in different parts of the world. The flower yield and the composition of essential oil of R. damascena are strongly affected by a number of factors. Nevertheless, the interactive effects of foliar application of plant nutrients and kinetin and its time of application on yield and secondary metabolites profile of R. damascena under acidic conditions are still unclear. Thus, a field experiment comprising two different times of spray and five foliar spray treatments was conducted to test the hypothesis that flowering behavior and secondary metabolites profile can be modified through proper nutrient supply at right time. The foliar spray at flower bud appearance stage (S2) significantly (P ≤ 0.05) increased flower yield by about 10.0 % compared with the foliar application at axillary bud development stage (S1) during both years, regardless of plant nutrients. Among the foliar spray treatments, kinetin at 0.20 g L?1 registered about 23–39 % higher flower yield compared with the water spray control; however, remained statistically at par (P ≤ 0.05) with Ca(NO3)2 at 4.06 g L?1. Moreover, the percentage of major fragrance-bearing compounds of essential oil (β-citronellol + nerol, linalool, E-geraniol, and Z-citral) was marginally increased with Ca(NO3)2 compared with kinetin treatment. However, the percentages of major hydrocarbons, nonadecane and heneicosane, were noticeably increased when kinetin was applied at S1. Foliar application of kinetin and Ca(NO3)2 might be done to improve flower yield and essential oil content in R. damascena flowers.  相似文献

In vitro culture and genetic transformation of black gram are difficult due to its recalcitrant nature. Establishment of gene transfer procedure is a prerequisite to develop transgenic plants of black gram in a shorter period. Therefore, genetic transformation was performed to optimize the factors influencing transformation efficiency through Agrobacterium tumefaciens-mediated in planta transformation using EHA 105 strain harbouring reporter gene, bar, and selectable marker, gfp-gus, in sprouted half-seed explants of black gram. Several parameters, such as co-cultivation, acetosyringone concentration, exposure time to sonication, and vacuum infiltration influencing in planta transformation, have been evaluated in this study. The half-seed explants when sonicated for 3 min and vacuum infiltered for 2 min at 100 mm of Hg in the presence of A. tumefaciens (pCAMBIA1304 bar) suspensions and incubated for 3 days co-cultivation in MS medium with 100 µM acetosyringone showed maximum transformation efficiency (46 %). The putative transformants were selected by inoculating co-cultivated seeds in BASTA® (4 mg l?1) containing MS medium followed by BASTA® foliar spray on 15-day-old black gram plants (35 mg l?1) in green house, and the transgene integration was confirmed by biochemical assay (GUS), Polymerase chain reaction, Dot-blot, and Southern hybridisation analyses.  相似文献

Stress-induced methylglyoxal (MG) functions as a toxic molecule, inhibiting plant physiological processes such as photosynthesis and antioxidant defense systems. In the present study, an attempt was made to investigate the MG detoxification through glutathione metabolism in indica rice [Oryza sativa L. ssp. indica cv. Pathumthani 1] under salt stress by exogenous foliar application of paclobutrazol (PBZ). Fourteen-day-old rice seedlings were pretreated with 15 mg L?1 PBZ foliar spray. After 7 days, rice seedlings were subsequently exposed to 0 (control) or 150 mM NaCl (salt stress) for 12 days. Prolonged salt stress enhanced the production of MG molecules and the oxidation of proteins, leading to decreased activity of glyoxalase enzymes, glyoxalase I (Gly I) and glyoxalase II (Gly II). Consequently, the decreased glyoxalase activities were also associated with a decline in reduced glutathione (GSH) content and glutathione reductase (GR) activity. PBZ pretreatment of rice seedlings under salt stress significantly lowered MG production and protein oxidation, and increased the activities of both Gly I and Gly II. PBZ also increased GSH content and GR activity along with the up-regulation of glyoxalase enzymes, under salt stress. In summary, salinity induced a high level of MG and the associated oxidative damage, while PBZ application reduced the MG toxicity by up-regulating glyoxalase and glutathione defense system in rice seedlings.  相似文献

Pistachio is a tree of the arid and semi-arid regions where salinity and boron (B) toxicity can be major environmental stresses. In this study, individual and combined effects of different concentrations of NaCl (0, 800, 1600, 2400 and 3200 mg kg?1 soil) and B (0, 2.5, 5.0, 10.0 and 20.0 mg kg?1 soil) were studied on growth, gas-exchange and mineral composition of pistachio seedlings for a duration of 120 days. Excess amounts of salinity (> 1600 mg NaCl kg?1 soil) and B (20.0 mg kg?1 soil) significantly reduced the plant growth and CO2 assimilation, which was associated with accumulation of Na, Cl and B in leaves. There was also a decline in cell membrane stability index (MSI). Reduced stomatal conductance (g s) was the primary cause of inhibition of photosynthesis rate (P N) under mild to moderate salinity. However, under severe salt stress and B toxicity, non-stomatal effects contributed to the inhibition of CO2 assimilation in addition to the decline in g s value. Application of 5.0–10.0 mg B kg?1 soil significantly improved the plant growth and P N and also recovered the MSI as countermeasures against salt stress. These observations were related to the role of B in cell membrane structure and functioning which reduced the concentration of toxic ions in the leaves. However, cell membrane damages and chlorophyll loss in plants affected by salt were observed to be exacerbated when excess amounts of B were present. In conclusion, the results revealed that optimizing the B nutrition can improve the performance of pistachio seedlings under salt stress, and NaCl also showed a mitigating effect on B toxicity if its concentration in the soil is kept under the plant salt tolerance threshold.  相似文献

An efficient transformation system for high-throughput functional genomic studies of kiwifruit has been developed to overcome the problem of necrosis in Actinidia arguta explants. The system uses Agrobacterium tumefaciens strain EHA105 harbouring the binary vector pART27-10 to inoculate leaf strips. The vector contains neomycin phosphotransferase (nptII) and β-glucuronidase (GUS) (uidA) genes. A range of light intensities and different strengths of Murashige and Skoog (MS) basal salt media was used to overcome the problem of browning and/or necrosis of explants and calli. Callus browning was significantly reduced, resulting in regenerated adventitious shoots when the MS basal salt concentration in the culture medium was reduced to half-strength at low light intensity (3.4 μmol m?2 s?1) conditions. Inoculated leaf strips produced putative transformed shoots of Actinidia arguta on half-MS basal salt medium supplemented with 3.0 mg l?1 zeatin, 0.5 mg l?1 6-benzyladenine, 0.05 mg l?1 naphthalene acetic acid, 150 mg l?1 kanamycin and 300 mg l?1 Timentin®. All regenerated plantlets were deemed putative transgenic by histochemical GUS assay and polymerase chain-reaction analysis.  相似文献

Although gibberellins (GAs) have been shown to induce development of the physiological disorder blossom-end rot (BER) in tomato fruit (Solanum lycopersicum), the mechanisms involved remain largely unexplored. BER is believed to result from calcium (Ca) deficiency, but the relationship between Ca content and BER incidence is not strong. Our objectives were to better understand how GAs and a GA biosynthesis inhibitor affect BER development in tomato fruit. Tomato plants of two BER-susceptible cultivars, ‘Ace 55 (Vf)’ and ‘AB2,’ were grown in a greenhouse environment and subjected to Ca-deficiency conditions. Plants were treated weekly during fruit growth and development with 300 mg L?1 GA4+7, 300 mg L?1 prohexadione-calcium (Apogee®, a GA biosynthesis inhibitor), or water beginning 1 day after flower pollination. GA4+7 treatment induced an increase in BER incidence in both cultivars up to 100%, whereas ‘Ace 55 (Vf)’ and ‘AB2’ plants treated with Apogee did not show BER incidence. The number of functional xylem vessels was higher in the placental and pericarp tissue of tomato fruit treated with Apogee at the early stages of fruit growth. Treatment with Apogee also increased fruit pericarp Ca concentration. GA4+7 treatment enhanced the expression of the putative CAX and Ca-ATPase genes, that code for proteins involved in Ca movement into storage organelles. The lowest water-soluble apoplastic Ca concentration and the highest membrane leakage values were observed in the pericarp of GA4+7-treated fruit. These results suggest that GAs consistently reduced fruit Ca uptake and water-soluble apoplastic Ca concentration, leading to leakier plasma membranes and an increase in BER development in fruit tissue of both tomato cultivars.  相似文献

Abelmoschus manihot, an ornamental plant, was examined for phytoremediation purposes in accordance with the ability to accumulate cadmium and physiological mechanisms of cadmium tolerance. A net photosynthetic rate (A N) glasshouse experiment for 60 days was conducted to investigate the influence of different cadmium amounts (0–100 mg kg?1) on the growth, biomass, photosynthetic performance, reactive oxygen species (ROS) production, antioxidative enzyme activities, Cd uptake and accumulation of A. manihot. Exposure to cadmium enhanced plant growth even at 100 mg kg?1, without showing symptoms of visible damage. The cadmium concentration of shoots (stems or leaves) and roots was more than the critical value of 100 mg kg?1 and reached 126.17, 185.26 and 210.24 mg kg?1, respectively. BCF values of A. manihot plants exceeded the reference value 1.0 for all the Cd treatments, and TF values were greater than 1 at 15–60 mg kg?1 Cd treatment. The results also showed that cadmium concentrations of 60 mg kg?1 or less induced a significant enhancement in plant net photosynthetic rate (A N), stomatal conductance (G s), transpiration rate (T r), photosynthetic pigments and F v/F m. These parameters were slightly decreased at the higher concentration (100 mg kg?1). The ROS production (O2 ?, H2O2) and antioxidative response including SOD, CAT and POD were significantly enhanced by increasing cadmium. These results suggest that A. manihot can be considered as a Cd-hyperaccumulator and the hormetic effects may be taken into consideration in remediation of Cd contamination soil.  相似文献

To determine the effects of vermicompost leachate (VCL) on resistance to salt stress in plants, young tomato seedlings (Solanum lycopersicum, cv. Ailsa Craig) were exposed to salinity (150 mM NaCl addition to nutrient solution) for 7 days after or during 6 mL L??1 VCL application. Salt stress significantly decreased leaf fresh and dry weights, reduced leaf water content, significantly increased root and leaf Na+ concentrations, and decreased K+ concentrations. Salt stress decreased stomatal conductance (gs), net photosynthesis (A), instantaneous transpiration (E), maximal efficiency of PSII photochemistry in the dark-adapted state (Fv/Fm), photochemical quenching (qP), and actual PSII photochemical efficiency (ΦPSII). VCL applied during salt stress increased leaf fresh weight and gs, but did not reduce leaf osmotic potential, despite increased proline content in salt-treated plants. VCL reduced Na+ concentrations in leaves (by 21.4%), but increased them in roots (by 16.9%). VCL pre-treatment followed by salt stress was more efficient than VCL concomitant to salt stress, since VCL pre-treatment provided the greatest osmotic adjustment recorded, with maintenance of net photosynthesis and K+/Na+ ratios following salt stress. VCL pre-treatment also led to the highest proline content in leaves (50 µmol g??1 FW) and the highest sugar content in roots (9.2 µmol g??1 FW). Fluorescence-related parameters confirmed that VCL pre-treatment of salt-stressed plants showed higher PSII stability and efficiency compared to plants under concomitant VCL and salt stress. Therefore, VCL represents an efficient protective agent for improvement of salt-stress resistance in tomato.  相似文献

Shoot tip explants of Phyllanthus amarus were cocultivated with Agrobacterium tumefaciens strain LBA 4404 carrying plasmid pCAMBIA 2301 harbouring genes coding for betaglucuronidase (gus), kanamycin (kan), and neomycin phosphotransferase II (nptII) along with a gene coding for Linum usitatissimum PINORESINOL LARICIRESINOL REDUCTASE (Lu-PLR). Transformed shoot tip explants were maintained in a Murashige and Skoog (MS) medium containing TDZ 1.54 mg l?1, kan 50 mg l?1 and cephotaxime 62.5 mg l?1. The optimum medium for regeneration of multiple shoots was MS supplemented with TDZ 1.54 mg l?1, kan 50 mg l?1. Efficient and effective rooting of plantlets was achieved by culturing the in vitro regenerated shoots on liquid ½ MS medium containing 0.7 mg l?1 indole 3-butyric acid (IBA) and 5 mg l?1 kan. Rooted plants were acclimatized in the mixtures of vermiculite and soil. The transformation of kan-resistant plantlets regenerated from shoot-tip explants was confirmed by GUS and polymerase chain reaction (PCR) analysis. Southern blot and reverse transcribed PCR (RT-PCR) analysis confirmed successful integration and expression of Lu-PLR gene. Quantitative analysis of phyllanthin performed on transgenic and wild plants using high-performance liquid chromatography (HPLC) revealed that transgenic lines contained higher phyllanthin content (0.3–0.81% w/w) than wild plants (0.09% w/w). The highest yield of phyllanthin was detected in transgenic lines was up to 1.16, 1.22 and 1.23 folds higher than that of wild plant. This report highlights the transgenic approach to enhance the contents of phyllanthin and hypophyllanthin.  相似文献

It was aimed to investigate the ameliorative effect of exogenously applied 24-epibrassinolide (EBR) on some key growth parameters and mineral elements in two salt-stressed maize (PR 32T83 and PR 34N24) cultivars. A factorial experiment was designed with two electrical permeability (EC) levels (1.1 and 8.0 dS/m) and two levels (1.5 and 2.0 µM) of EBR supplied as a seed treatment, foliar spray, or both in combination. The foliar application of EBR was done once a week during the experiment. After 42 days of these treatments, the plants were harvested to assess growth, water relations, and oxidative and antioxidative systems. Salt stress markedly reduced plant fresh and dry weights, maximum fluorescence yield of PS-II, chlorophyll contents, leaf water potential, and leaf K and Ca, but it increased membrane permeability, the activities of superoxide dismutase (SOD; EC 1.15.1.1), peroxidase (POD; EC. 1.11.1.7), and catalase (CAT; EC. 1.11.1.6) enzymes, and the contents of proline and glycine betaine, leaf sap osmotic pressure, lipid peroxidation, hydrogen peroxide, and leaf Na and Cl. However, both seed treatment and foliar application of EBR to the maize plants exposed to saline conditions enhanced key growth attributes, water relations, and the activities of various antioxidant enzymes as well as the levels of proline, but they reduced electrolyte leakage, and H2O2 and MDA contents. Saline stress reduced leaf N, Ca2+, K+, and P contents as compared to those in the non-stressed plants. Both seed treatment and foliar application of EBR reduced Na+ and Cl? concentrations, but increased those of N, Ca2+, K+, and P. Foliar application of EBR was more effective in increasing nutrient levels of plants grown at the high saline regime compared to the seed treatment of EBR. The study clearly indicates that both seed treatment and foliar application of EBR at the rate of 2.0 µM can overcome the detrimental effect of salinity stress on maize growth, which was found to be significantly linked to reduced concentrations of Na, Cl, MDA, and H2O2 as well as EL and increased activities of key antioxidant enzymes in the maize plants.  相似文献

An in vitro organogenesis protocol for Carissa carandas L. was developed using an auxin transport inhibitor (quercetin) and silver nitrate (AgNO3), an inhibitor of ethylene action, in association with cytokinins in the culture medium. This protocol produced the maximum number of shoots from aseptic seedling-derived shoot apex explants of C. carandas. The highest rate of shoot multiplication was recorded on MS medium containing 2.0 mg L?1 6-benzylaminopurine; 0.5 mg L?1 kinetin, and 0.75 mg L?1 quercetin at after 4 wk of culture. Similar results were obtained when MS medium fortified with 2.0 mg L?1 BAP, 0.5 mg L?1 kinetin, and 1.5 mg L?1 AgNO3 was used. However, successful rooting was achieved on quarter strength MS medium with 0.5 mg L?1 indole-3-acetic acid. In this study, an inhibitor of auxin transport and ethylene action maximized shoot multiplication in medium fortified with cytokinins. The established rapid micropropagation method could be used to conserve elite genotypes of C. carandas.  相似文献

The effects of a bioinsecticide formulation based on extract of Annona squamosa L. (Annonaceae) containing 10,000 mg L?1 of acetogenin annonin as the main active ingredient were investigated against three primary pest species of stored grains in Brazil [maize weevil Sitophilus zeamais Motschulsky (Coleoptera: Curculionidae), Mexican bean weevil Zabrotes subfasciatus (Boheman) (Coleoptera: Chrysomelidae: Bruchinae), and cowpea weevil Callosobruchus maculatus (Fabricius) (Coleoptera: Chrysomelidae: Bruchinae)] by means of residual contact bioassays. In a concentration-dependent manner, the annonin-based commercial bioinsecticide caused significant adult mortality of C. maculatus (LC50 = 6890 μL kg?1), S. zeamais (LC50 = 2781 μL kg?1), and Z. subfasciatus (LC50 = 2120 μL kg?1) after 120 h of residual contact exposure. In addition to acute toxicity, the tested bioinsecticide also promoted a significant reduction of the number of eggs laid by females of C. maculatus (EC50 = 5949.7 μL kg?1) and Z. subfasciatus (EC50 = 552.7 μL kg?1). Moreover, the bioinsecticide significantly reduced the number of emerged insects (F1 generation) of C. maculatus (EC50 = 2763.0 μL kg?1), S. zeamais (EC50 = 1380.8 μL kg?1), and Z. subfasciatus (EC50 = 561.5 μL kg?1). The bioinsecticide also reduced the percentage of damaged grains for the three pest species studied, and its grain-protectant properties are comparable to or superior in efficacy in relation to a diatomaceous earth-based insecticide (Insecto® at 1000 mg kg?1) used as a positive control. Thus, this standardized formulation has promising bioactivity against stored insect species and can be a useful component for IPM of stored grains in Brazil and elsewhere.  相似文献

The experiments were conducted to examine the effects of seed priming in solutions (100, 150 and 200 mg L?1) of different synthetic auxins, i.e., 2,4-dichlorophenoxyacetic acid (2,4-D), 2,4,5-trichlorophenoxyacetic acid (2,4,5-T), α-naphthaleneacetic acid (NAA) on growth, grain yield, gaseous exchange characteristics, ionic and hormonal concentrations in two spring wheat (Triticum aestivum L.) cultivars MH-97 (salt intolerant) and Inqlab-91 (salt tolerant). The primed (soaked for 12 h) and non-primed seeds were sown in Petri plates in a growth room as well as in a field treated with 150 mM NaCl. Generally, all synthetic auxins did not increase germination percentage and rate in both cultivars when compared with hydropriming (control), and even decreased these attributes when applied at higher concentrations (200 mg L?1). Nonetheless, under salt stress, NAA (150 mg L?1) was most effective in increasing seedling shoot dry weight, fertile tillers per plant, number of grains per ear and grain yield in both cultivars. The plants raised from seed treated with NAA (150 mg L?1) had lower shoot [Na+] in the salt intolerant cultivar. Moreover, NAA treatment improved root [Ca2+] in both cultivars. Priming agents affected leaf free indole-3-acetic acid (IAA) and indole-3-butyric acid (IBA) concentrations differently in both cultivars. Treatment with NAA (150 mg L?1) lowered leaf free abscisic acid (ABA) and putrescine (Put) concentrations and raised salicylic acid (SA) and spermidine (Spd) concentrations in the salt intolerant cultivar. In conclusion, pre-treatment with NAA (150 mg L?1) showed consistent promotive effects on growth and grain yield in the two cultivars, which were partially attributed to the beneficial effects of NAA-priming on ionic and hormonal homeostasis under salt stress.  相似文献

Ascophyllum nodosum, a brown macroalga, is the most widely used seaweed in agriculture. We report a rapid method for the detection of cytokinin-like activity in plants treated with a commercial A. nodosum liquid concentrate (Stimplex®) using a transgenic line of Arabidopsis carrying the ARR5 promoter fused to ß-glucuronidase (GUS) reporter gene. Based on GUS activity assay, an increase in cytokinin-like activity was detected in plants grown in vitro treated with 3 mL L?1 Stimplex®, whereas foliar spray treatments showed similar cytokinin-like activity at a concentration of 5 mL L?1. Histochemical staining showed Stimplex®-induced GUS activity in leaf as well as in the root tissues. Taken together, our results suggest that Stimplex® contains compounds that may elicit endogenous cytokinin-like activity. Furthermore, it is shown that this bioassay can be used for rapid screening of extracts that can stimulate cytokinin-like activities using Arabidopsis AAR5::GUS reporter transgenic plants.  相似文献

Acetaldehyde strongly binds to the wine preservative SO2 and, on average, causes 50–70 mg l?1 of bound SO2 in red and white wines, respectively. Therefore, a reduction of bound and total SO2 concentrations necessitates knowledge of the factors that affect final acetaldehyde concentrations in wines. This study provides a comprehensive analysis of the acetaldehyde production and degradation kinetics of 26 yeast strains of oenological relevance during alcoholic fermentation in must under controlled anaerobic conditions. Saccharomyces cerevisiae and non-Saccharomyces strains displayed similar metabolic kinetics where acetaldehyde reached an initial peak value at the beginning of fermentations followed by partial reutilization. Quantitatively, the range of values obtained for non-Saccharomyces strains greatly exceeded the variability among the S. cerevisiae strains tested. Non-Saccharomyces strains of the species C. vini, H. anomala, H. uvarum, and M. pulcherrima led to low acetaldehyde residues (<10 mg l?1), while C. stellata, Z. bailii, and, especially, a S. pombe strain led to large residues (24–48 mg l?1). Acetaldehyde residues in S. cerevisiae cultures were intermediate and less dispersed (14–34 mg l?1). Addition of SO2 to Chardonnay must triggered significant increases in acetaldehyde formation and residual acetaldehyde. On average, 0.33 mg of residual acetaldehyde remained per mg of SO2 added to must, corresponding to an increase of 0.47 mg of bound SO2 per mg of SO2 added. This research demonstrates that certain non-Saccharomyces strains display acetaldehyde kinetics that would be suitable to reduce residual acetaldehyde, and hence, bound-SO2 levels in grape wines. The acetaldehyde formation potential may be included as strain selection argument in view of reducing preservative SO2 concentrations.  相似文献

Phycobiliproteins, light-harvesting pigments found in cyanobacteria and in some eukaryotic algae, have numerous commercial applications in food, cosmetic, and pharmaceutical industries. Colorant production from cyanobacteria offers advantages over their production from higher plants, as cyanobacteria have fast growth rate and high photosynthetic efficiency and require less space. In this study, three cyanobacteria strains were studied for phycobiliprotein production and the influence of sodium nitrate, potassium nitrate and ammonium chloride on the growth and phycobiliprotein composition of the strains were evaluated. In the batch culture period of 12 days, Phormidium sp. and Pseudoscillatoria sp. were able to utilize all tested nitrogen sources; however, ammonium chloride was the best nitrogen source for both strains to achieve maximum growth rate μ?=?0.284?±?0.03 and μ?=?0.274?±?0.13 day?1, chlorophyll a 16.2?±? 0.5 and 12.2?±? 0.2 mg L?1, and phycobiliprotein contents 19.38?±?0.09 and 19.99?±?0.14% of dry weight, whereas, for Arthrospira platensis, the highest growth rate of μ?=?0.304?±?0.0 day?1, chlorophyll a 19.1?±?0.5 mg L?1, and phycobiliprotein content of 22.27?±?0.21% of dry weight were achieved with sodium nitrate. The phycocyanin from the lyophilized cyanobacterial biomass was extracted using calcium chloride and food grade purity (A620/A280 ratio >?0.7) was achieved. Furthermore, phycocyanin was purified using two-step chromatographic method and the analytical grade purity (A620/A280 ratio >?4) was attained. SDS-PAGE demonstrated the purity and presence of two bands corresponding to α- and β-subunits of the C-phycocyanin. The results showed that Phormidium sp. and Pseudoscillatoria sp. could be good candidates for phycocyanin production.  相似文献

Lisianthus (Eustoma grandiflorum) is a cut or ornamental flower that is popular all over the world. This ornamental crop, however, lacks an effective weed control method due to its susceptibility to herbicide. In this study, transgenic plants of a lisianthus cultivar were produced using Agrobacterium-mediated delivery of the plasmid pCAMBIA3300, which carried the bialaphos resistance (bar) gene under driven by the CaMV 35S promoter. The transgenic calli were derived from wounded edges of the leaves grown on a shoot regeneration medium containing 100 mg l?1 cefotaxime and 2 mg l?1 glufosinate ammonium for 4 weeks. The callus that was detached from the wounded edge of the leaf was transferred to the shoot regeneration medium with 100 mg l?1 cefotaxime and 5 mg l?1 glufosinate ammonium for 4 weeks for shoot regeneration. The bar gene integration and expression in the transgenic plants were confirmed by Southern and Northern blot analyses, respectively. Subsequently, the transgenic lines were assessed in vitro and under greenhouse conditions for their resistance to the commercial herbicide Basta®, which contains glufosinate ammonium as the active component. Six transgenic lines showed high percentages (67–80%) of survival in vitro under the selection condition with glufosinate ammonium (up to 216 mg l?1). Under greenhouse conditions, the plants from these six lines remained healthy and exhibited a normal phenotype after spraying with glufosinate ammonium (up to 1,350 mg l?1). This is the first paper to provide a detailed survey of transgenic lisianthus expressing the bar gene and exhibiting herbicide-resistance under greenhouse conditions.  相似文献

Two ornamental plants of Althaea rosea Cav. and Malva crispa L. were exposed to various concentrations of lead (Pb) (0, 50, 100, 200 and 500 mg·kg?1) for 70 days to evaluate the accumulating potential and the tolerance characteristics. The results showed that both plant species grown normally under Pb stress, and A. rosea had a higher tolerance than M. crispa, while M. crispa had a higher ability in Pb accumulation than A. rosea. Besides, lower Pb concentration (50 mg·kg?1) stimulated the shoot biomass in both plant species. Pb accumulation in plants was consistent with the increase of Pb levels, and the main accumulation sites were the roots and the older leaves. In addition, the photosynthetic pigments content and chlorophyll fluorescence parameters were influenced by Pb stress. In such case, both of the plants could improve the activities of antioxidant enzymes of superoxide dismutase (SOD), peroxidase (POD), catalase (CAT) and ascorbate peroxidase (APX), and the contents of the total soluble sugar and soluble protein, which reached the highest value at Pb 100 mg·kg?1, as well as the accumulation of the total thiols (T-SH) and non-protein thiols (NP-SH) to adapt to Pb stress. Thus, it provides the theoretical basis and possibility for ornamental plants of A. rosea and M. crispa in phytoremediation of Pb contaminated areas.  相似文献

A complete protocol for the in vitro induction of Eclipta alba tetraploids has been optimized to enhance the wedelolactone content, an anti-cancerous compound. The effects of different concentrations of colchicine (0, 0.01, 0.05, 0.1, 0.2 and 0.3%; w/v) along with treatment durations (12, 24, 36 and 48 h) were investigated on shoot tip (ST) and nodal segment (NS). The treated explants were then incubated on Murashige and Skoog (MS) medium having 1.5 mg L?1 N6-benzylaminopurine and 0.5 mg L?1 α-napthalene acetic acid for shoot regeneration and afterward root was induced on 1.0 mg L?1 indole-3-acetic acid enriched ½MS medium. The tetraploids of E. alba were proficiently induced by the treatment of 0.1% colchicine for 24 h. The highest tetraploid induction efficiency was obtained from ST (30.56%) in comparison to the NS (22.22%). Analysis by spectrophotometry and flow cytometry showed that colchicine treated plants contained higher quantity of DNA than diploid plants. Cytological studies demonstrated doubled the chromosome number in tetraploids (2n?=?4x?=?44) than diploids (2n?=?2x?=?22). The ploidy level enhancement lead to alteration of other traits, like increased plant height, stem diameter, leaf size, stomatal size and chlorophyll content. As determined through high performance thin-layer chromatography, the ultimate achievement of this technique is the higher accumulation of wedelolactone in tetraploid plants (300.32 µg g?1 dry weight) in evaluation to in vitro diploid (131.31 µg g?1 dry weight) and in vivo diploid mother plants (93.26 µg g?1 dry weight), thus improving the pharmaceutical value of E. alba.  相似文献
