Aluminum in lake water and in the organs of the fish Tribolodon hakonensis was investigated in Lake Usoriko (pH 3.6), Lake Inawashiroko (pH 5.0), and the Tenryu River (pH 7.7). The concentration of total soluble aluminum in the water was 0.51 mg l−1 in Usoriko, 0.05 mg l−1 in Inawashiroko, and less than 0.01 mg l−1 in the Tenryu. The chemical forms of soluble aluminum in the acid water were characterized as Al3+, AlL2+, and AlL≦1+. More than 90% of soluble aluminum in the water of Usoriko was Al3+, whereas AlL2+ was dominant in the water of Inawashiroko. The aluminum concentration in the organs of T. hakonensis in Usoriko was 42 μg g−1 wet weight in gills, 4.2 μg g−1 in muscle, 6.9 μg g−1 in bone, 12.7 μg g−1 in liver, 6.0 μg g−1 in kidney, and 6.0 μg g−1 in intestine, indicating accumulation of aluminum in the gills. The aluminum concentration in the organs of T. hakonensis living in Inawashiroko was approximately the same, in spite of the difference in water chemistry of the two acid lakes, especially for pH and aluminum. This suggests that aluminum accumulation might be controlled in the fish living in the acid lakes. In contrast, the aluminum concentration in the gills of T. hakonensis from the Tenryu was 2 μg g−1. The properties and activity of brown adipose tissue have been investigated in suckling, pre-obese, obob mice in order to determine whether decreased thermogenesis in the tissue precedes the development of obesity in this mutant. At 14 days of age there was no difference between the obob and normal animals in the total amount of interscapular brown adipose tissue, and the DNA content, protein content, and cytochrome oxidase activity of the tissue were similar in the two groups of mice. Respiration rates of brown adipose tissue mitochondria in the presence of albumin were, however, greater in the normal than the obob animals, although after the addition of GDP to recouple the mitochondria there was no difference between the two groups. The mitochondrial membrane potential, measured with [3H]methyltriphenylphosphonium, was less affected by exogenous GDP in obob mice than in normal animals. GDP binding to brown adipose tissue mitochondria, an index of the proton conductance pathway, was much greater in normal than in obob mice at both 10 and 14 days of age; the decreased GDP binding in the mutant animals was found to result from a reduction in the number of binding sites. It is concluded that brown adipose tissue mitochondria of pre-obese obob mice are more tightly coupled than those of normal siblings, and that the activity of the ‘thermogenic’ proton conductance pathway is lower in the mutant animals. A decrease in thermogenesis in brown adipose tissue is therefore an early event in the development of the obob mouse and precedes the appearance of obesity.  相似文献

A comparative study has been made of the mucogenic epidermis of the common carp, Cyprinus carpio var. communis, and the three Indian major carps, Catla catla, Labeo rohita and Cirrhina mrigala: on the basis of epidermis structural organization, these species are easily differentiated. The epithelial cells in the superficial layer, as in most fishes, show secretory activity, evidenced by positive histochemical reactions, which is high in C. carpio var. communis, moderate in C. catla and low in L. rohita and C. mrigala. The epithelial cells in the underlying two or three layers also give positive reactions, though their intensity is relatively weak. The mucous cells in C. carpio var. communis are distributed in large numbers arranged in several superimposed layers in the outer regions of the epidermis, whereas in C. catla they are fewer in number and are widely separated in the surface layers as well as in the deeper layers of the epidermis; in both species the mucous cells appear rounded, large, and open on the surface by wide pores. In contrast, in L. rohita and C. mrigala the mucous cells are smaller, restricted mainly to the superficial layer, close together in a single row, and open on the surface by narrow pores. The overall density of mucous cells in L. rohita and C. mrigala, as in C. catla, is much lower than in C. carpio var. communis. In the epidermis of C. carpio var. communis there are a large number of mucous cells, and the few club cells are restricted to the deeper layers. In contrast, in the epidermis of the three Indian major carp the overall density of the mucous cells is much lower and the club cells are very numerous. It is suggested that the high density of club cells compensates an overall low density of mucous cells as an adaptation for an effective defence mechanism. Increased mucus production in the epidermis of C. carpio var. communis, as evidenced by a large number of mucous cells in outer regions and high secretory activity of superficial layer epithelial cells, is associated with increased precipitation of mud held in suspension, needed as an adaptation to the species’peculiar bottom-scooping habits. The levels of purine and pyrimidine nucleotides in suspensioncultures of Catharanthus roseus were determined 24 h after stationary-phasecells were transferred to fresh complete (‘+Pi’)or phosphate-deficient (‘–Pi’) Murashige-Skoogmedium. The levels of ATP, GTP, UTP and CTP were from approx.3 to 5-fold greater in the cells grown in ‘+Pi’medium than in the cells grown in ‘–Pi’ medium.The levels of almost all other nucleotides were slightly higherin the cells in ‘+Pi’ medium. The rates of de novoand salvage biosynthesis of purine and pyrimidine nucleotideswere estimated from the rates of incorporation of radioactivityfrom [14C]formate, [2–14C]glycine, NaH14CO3, [6–14C]orotate,[8–14C]adenine, [8–14C]adenosine, [2–14C]uraciland [2–14C]uridine. The results indicated that the activityof both the de novo and the salvage pathway was higher in thecells in ‘+Pi’ medium than in the cells in ‘–Pi’medium. The rate of degradation estimated from the rate of releaseof 14CO2 from labelled purines and pyrimidines indicated thatdegradation of uridine was significantly reduced in the cellsin ‘+Pi’ medium, but no significant difference wasfound in the degradation of adenine, adenosine and uracil. Thepossible role of Pi in the control of the biosynthesis of nucleotidesand in the degradation of uridine is discussed. We estimated in rats the expression of early gene c-fos (marker of neuronal activation) and NADPH-diaphorase activity (NO-synthase marker) in the limbic structures of the basal forebrain and in the hypothalamus. Estimations were performed in the norm, in the state of starvation, and after realization of long-lasting (repeated 4 to 12 times per minute for 30 min) motivated stereotyped food-procuring forelimb movements. In food-deprived animals, a significantly greater (Р < 0.05), as compared with the control, number of Fos-immunoreactive (Fos-ir) and NADPH-diaphorase-reactive (NADPH-dr) neurons was observed in limbic structures, namely in the medial septum (MS), nuclei of the vertical and horizontal branches of the diagonal fascia (VDB and НDB), magnocellular preoptic nucleus (MCPO), complex of the substantia innominata−basal nucleus of Meynert of the pallidum, SI-GP(B), as well as in the laterodorsal tegmental nucleus (LDTg), medial part of the pallidum (MGP), paraventricular and lateral nuclei of the hypothalamus (Pa and LH), and islands of Calleja (ICj and ICjM). In the limbic structures and pontine nuclei of rats of the experimental group (that performed operant movements), greater mean densities of labeled neurons were found in the succession LDTg < SI < MCPO < GP(B) < MS < VDB < HDB. The maximum mean density of Fos-ir neurons (13.8 ± 0.9 labeled nuclei within 200 × 200 μm2 area) was found in the HDB. In the hypothalamic nuclei of starving rats, c-fos expression was two times higher than that in the control. After realization of operant movements, the intensity of expression in the LH was somewhat smaller, while in the Ра it was higher. The maximum density of NADPH-dr neurons was observed in the Pa (303.4 ± 18.7 cells), in the ICj and ICjM (287 ± 11.6 and 260 ± 8.7 neurons, respectively), and in the MGP (93 ± 6.7 labeled cells). When analyzing the distribution of labeled neurons in experimental rats, we found high densities of double-labeled cells (Fos + NADPH-d positivity) in the Pa, MGP, ICj, and ICjM. Such specificity of changes in the c-fos expression and NADPH-d reactivity in the hypothalamus correlates, perhaps, with the formation of motivation signals related to a delay in food accessibility and supply of food. Modifications of neuronal activity in limbic structures reflect involvement of the latter in the formation of motor programs for food-procuring movements and their realization. The physiological response of leaves developed in low light (L) on Fagus crenata seedlings exposed to different levels of high light (H: high light, M: medium light) was studied. Measurements were conducted on potted seedlings in the F. crenata forest understory. The seedlings with leaves developed in L were transferred to H (L–H) and M (L–M) in summer. On exposure to high light, the photochemical efficiency of dark-adapted PSII (Fv/Fm) immediately decreased and was followed by a subsequent recovery in both L–H and L–M leaves. The mean value of Fv/Fm in L–H leaves was lower than that in L–M leaves through experiments, indicating that the degree of photoinhibition in L–H leaves was greater than that in L–M leaves. About 1 month after transfer, 37% and 5% of leaves had fallen in L–H and L–M seedlings, respectively. This result also indicated the greater photoinhibition in L–H leaves. Moreover, the photosynthetic capacity (PNmax) of L–H leaves decreased. In contrast, the PNmax of L–M leaves increased, although the PNmax was lower than that of M control leaves. An increase in the xanthophyll cycle pool (VAZ), indicating an increase of the photoprotective function, was found in both L–H and L–M leaves. Especially, the VAZ pool in L–M leaves was higher than that in M leaves by the end of experiments. L–M leaves may avoid photoinhibition effectively by the decrease in excess light with the increase of the PNmax or VAZ pool, compared to L–H leaves. Thus, the physiological acclimation on exposure to high light depended on the degree of high light. To achieve successful photosynthetic acclimation with slight photoinhibition, the variation of light intensity before and after exposure to high light would be an important factor because of the difference in excess light.  相似文献

研究了北京市区沿城乡梯度带河流底栖硅藻的空间分布及其季节变化特征。通过在2014年的春季、夏季和秋季在山区对照溪流、城市上游河道和城市下游河道3个样区,共23个样点进行了底栖硅藻样品采集。分析表明,主要优势种的空间差异明显。在对照溪流,主要优势种为极小曲丝藻(Achnanthidium minutissimum);在城市上游河道,主要优势种分别为短文假十字脆杆藻(Pseudostaurosira brevistriata)和连结十字脆杆藻腹面变种(Staurosira construens var.venter);在城市下游河道,主要优势种为谷皮菱形藻(Nitzschia palea)。然而,3个样区内的主要优势种相对丰度季节变化较小(P0.05)。其中,A.minutissimum及P.brevistriata的季节变化特征均为春季秋季夏季,而S.construens var.venter及N.palea的季节变化特征均为春季夏季秋季。结果发现,北京市城乡梯度带的河流硅藻种类组成差异较大,但主要优势属种季节变化相对较小,表明硅藻种类能较好地反映城乡梯度带的河流环境变化状况,可用于北京市河流水质生物监测。  相似文献

Zusammenfassung Die Sippen desArabis hirsuta-Aggregats wachsen in Österreich im wesentlichen in folgender Verbreitung und in den folgenden Vegetationseinheiten. VonA. allionii gibt es einige gesicherte Fundorte in der subalpinen Stufe der Silikat-Gebirge im salzburgisch-steirisch-kärntnerischen Grenzbereich, und zwar in Hochstaudenfluren und Silikatschutthalden.A. planisiliqua ist von mehreren über den größten — insbesondere den östlichen — Teil Österreichs zerstreuten Fundpunkten aus±tiefen (planaren) Lagen bekannt (für Steiermark, Tirol und Vorarlberg nicht sicher nachgewiesen); sie bevorzugt Feuchtwiesen, Au- und Bruchwälder, aber auch Ruderalstandorte, alle über feuchten bis frischen Böden.A. sagittata kommt im wesentlichen am Alpenrand, im außeralpinen Anteil (besonders im Osten) und im Süden Österreichs vor, für Nordtirol und Vorarlberg ist sie nicht nachgewiesen; sie wächst auf meist±warmen, trockenen, kalkreichen Standorten in der collinen und montanen Stufe in primären bis vom Menschen mäßig (seltener stark) beeinflußten Biozönosen, vor allem in Trocken- und Steppenrasengesellschaften (auch als Pionier) sowie in wärmeliebenden anspruchsvollen Eichenmischwäldern und Gebüschen und in Föhrenwäldern.A. hirsuta s. str. ist in Österreich allgemein von der planaren bis in die subalpine Stufe verbreitet und hat die weiteste ökologische Amplitude, die von warmen, trockenen Standorten (Pionier auf Felsen und Schutt, insbesondere aber in Trockenrasen) bis zu Kultur- und Moorwiesen einerseits, zu Gebüschen, Föhren- und Fichtenwäldern sowie subalpinen Latschengebüschen u. dgl. anderseits reicht;A. hirsuta s. str. wächst mit einer gewissen Vorliebe in gestörten und sekundären Gesellschaften. — Die zum Vergleich untersuchteA. corymbiflora ist nicht nur über die alpine und subalpine sondern auch über die montane Stufe der österreichischen Kalk- und Zentralalpen auf Karbonat-Urwiesen sowie Gesteinsschuttfluren einerseits und Halbtrockenrasen anderseits verbreitet.Die Sippen desA. hirsuta-Aggregats sind ± sympatrisch differenziert, und zwar sind sie durch ökologische Divergenz (auf diploider Stufe) bzw. durch Allopolyploidisierung (A. hirsuta s. str.) und durch die fakultative Autogamie aller voneinander isoliert; Hybridisierung ist selten. Die Differenzierungsmuster der Sippen beruhen nicht zuletzt auch auf deren Kurzlebigkeit sowie den hohen Samenansätzen und Keimprozentsätzen einerseits, auf dem Fehlen wirksamer Verbreitungseinrichtungen und der geringen strukturellen Differenzierung der Chromosomen anderseits. Die Populationen sind meist groß, ± kontinuierlich und individuenreich. Das Rekombinationssystem aller Sippen ist folglich mäßig eingeschränkt.
Summary Distribution, sites and associates of the taxa ofArabis hirsuta agg. in Austria have been investigated with the following results.A. allionii is growing only in few localities in the subalpine tier of silicate mountains in central Austria in tall herb communities and silicate screes.A. planisiliqua is known from a number of places scattered throughout the greater part of Austria, especially from the lowland of eastern Austria, growing in wet meadows, fenwoods, ruderal sites, etc.A. sagittata occurs principally in the eastern and southern parts of Austria in warm, dry, calcareous habitats in the colline and montane tier, in the main in natural (sometimes also secondary) xerophilous grasslands or as a pioneer in rock-sites, as well as in oak woods, scrubs, and in pine woods.A. hirsuta s. str. is to be found in all parts of Austria from the lowland up to the subalpine zone; it has the greatest ecological amplitude, occurring in warm, dry habitats (as a pioneer plant on rocks, screes, in natural and secondary xerophilous grasslands), in ± cultivated dry and wet meadows, in scrubs, in pine and spruce woods, etc., often preferring disturbed or ± artificial plant communities. —A. corymbiflora is growing not only in plant associations of the subalpine and alpine zone of nearly all parts of the Austrian Alps but often in the Mesobromion of the montane tier of the Alps too.The differentiation of the taxa of the aggregate is ± sympatric, on the diploid level they are isolated by ecological divergence,A. hirsuta s. str. is isolated by allopolyploidy, and all species are facultative selffertilizers; thus hybridization is weak. Furthermore the differentiation patterns of the taxa are based on the one hand upon the relatively short generations as well as the abundant setting of seed normally germinating to a large extent, and on the other hand upon the lack of special dispersal mechanisms and the weak structural differentiation of the chromosomes. The populations are usually large and ± continous, containing many individuals. The recombination systems of all taxa are moderately restricted.

We usedfluorescent-labeled microspheres in pentobarbital-anesthetized dogs tostudy the effects of unilateral alveolar hypoxia on the pulmonary bloodflow distribution. The left lung was ventilated with inspiredO2 fraction of 1.0, 0.09, or 0.03 in random order; the right lung was ventilated with inspiredO2 fraction of 1.0. The lungs wereremoved, cleared of blood, dried at total lung capacity, then cubed toobtain ~1,500 small pieces of lung (~1.7 cm3). The coefficient ofvariation of flow increased (P < 0.001) in the hypoxic lung but was unchanged in the hyperoxic lung.Most (70-80%) variance in flow in the hyperoxic lung wasattributable to structure, in contrast to only 30-40% of thevariance in flow in the hypoxic lung(P < 0.001). When adjusted for thechange in total flow to each lung, 90-95% of the variance in thehyperoxic lung was attributable to structure compared with 70-80%in the hypoxic lung (P < 0.001). Thehilar-to-peripheral gradient, adjusted for change in total flow,decreased in the hypoxic lung (P = 0.005) but did not change in the hyperoxic lung. We conclude thathypoxic vasoconstriction alters the regional distribution of flow inthe hypoxic, but not in the hyperoxic, lung.

Fish parasite communities have strong interactions with the environment, the host fish, and the aquatic invertebrate communities. Thus, factors directly involving their different life cycles, such as different host populations and environments, as well as seasonal fluctuations in water levels can cause different responses. The main factors structuring parasite communities may be the hydrodynamic variations, together with seasonal variations in the availability of infectious stages of parasites in the environment. The purpose of this study was therefore to investigate the influence of the rainy and dry seasons on parasite infracommunities in fish species in the Amazon River System of Brazil. Between October 2009 and April 2011, specimens of Colossoma macropomum and Colossoma macropomum × Piaractus brachypomus hybrids (tambatinga) were examined for the presence of parasites. The parasite communities were similar in these two species that both showed aggregate dispersion, although the hybrid specimens were less parasitized. For both hosts, the Brillouin diversity, species richness, evenness and Berger‐Parker dominance indices were similar in the rainy season and dry season, except that the Berger‐Parker dominance index for the C. macropomum × P. brachypomus hybrid was higher during the dry season. Ichthyophthirius multifiliis was the dominant parasite, followed by Piscinoodinium pillulare, in both hosts. However, C. macropomum had a higher prevalence and abundance in the dry season, while in the C. macropomum × P. brachypomus hybrid, only the I. multifiliis prevalence increased. The abundance of monogenean species (Anacanthorus spathulatus, Linguadactyloides brinkmanni, Mymarothecium and Notozothecium janauachensis) was higher during the dry season in the gills of C. macropomum. Despite overdispersion of the lernaeid Perulernaea gamitanae, a seasonal pattern was seen only in the C. macropomum × P. brachypomus hybrid, with higher prevalence in the rainy season. The low prevalence and abundance of the ectoparasites Tetrahymena sp., Trichodina sp. and Braga patagonica, as well as of the endoparasites Procamallalus (Spirocamallanus) inopinatus and Neoechinorhynchus buttnerae, were not affected by seasonality. This information is applicable in farming practice for these economically important fish, as it indicates the best time for prophylactic management and treatment against parasites, in order to prevent economic losses in fish farms.  相似文献

以江西九连山国家自然保护区常绿阔叶林为对象,研究林分不同层次优势种受灾程度,分析物种海拔、坡度对受灾程度的影响。结果表明:乔木上层和中层树木以断稍为主,乔木下层树木以断稍和腰折为主,小树和幼树以压弯为主。在平均受损指数MDI(Mean Damage Index)方面,乔木上层米槠MDI值最高,拟赤杨最低; 乔木中层鸭公树MDI值最高,浙江新木姜子最低; 乔木下层米槠MDI值最高,浙江新木姜子最低; 小树米槠MDI值最高,细枝柃最低; 幼树二列叶柃MDI值最高,狗骨柴最低。海拔对不同林分层次树木的部分受损指标有显著影响(P<0.05); 整体上,不同林分层次树木的受损指标在700~760 m海拔生境高于640~700 m海拔生境。不同坡度生境下树木的受损指标均无显著差异(P>0.05)。  相似文献

An association between sperm DNA integrity and fertility was recently shown for frozen–thawed Norwegian Red (NRF) bull semen diluted in skimmed milk egg yolk (SMEY). In general the fertility of NRF cattle is high, however, in comparison with NRF semen in SMEY, NRF semen diluted in Tris EY based extenders has shown reduced fertility. The aim of the present study was to do a split-sample comparison of sperm DNA integrity of NRF bull semen (n = 20) in SMEY and Triladyl® (Tris EY based) during routine cryopreservation procedure and during in vitro incubation of frozen–thawed semen in modified synthetic oviduct fluid (mSOF). In contrast to the high fertility of NRF cattle, Holstein cattle are experiencing a marked decline in fertility. Therefore, the present study also aimed to compare sperm DNA integrity of NRF (n = 20) and Holstein (n = 20) semen diluted in Triladyl® during in vitro incubation. The sperm DNA integrity was measured by susceptibility to in situ acid induced denaturation by the Sperm chromatin structure assay (SCSA). Compared to initial values of frozen neat semen, an increase in DNA damage was observed after dilution and cooling (5 °C) and after freezing–thawing of NRF semen in SMEY, but only after freezing–thawing for NRF semen diluted in Triladyl®. Sperm DNA damage of NRF semen increased during in vitro incubation in mSOF; the increase in percentage of spermatozoa with DNA damage was more prominent in SMEY than in Triladyl®, while the degree of damage was higher in Triladyl®, throughout the incubation period. However, while the correlation between DNA damage and sperm survival was negative in SMEY throughout the incubation period, a positive correlation was observed in Triladyl® after 9 h of incubation, indicating a higher presence of DNA damage in the live sperm population. In comparison with Holstein spermatozoa, the sperm DNA integrity of NRF semen reflected a better ability to withstand alterations induced during in vitro incubation in mSOF. In conclusion, sperm DNA integrity of NRF bull semen was altered during the cryopreservation procedure and in vitro incubation in mSOF. Dilution in Triladyl® maintained bull sperm DNA integrity better than dilution in SMEY. Furthermore, alterations in Holstein sperm DNA integrity was more pronounced during in vitro incubation in mSOF compared to NRF bull spermatozoa.  相似文献

The aim of this work was to determine the protective effects of intraperitoneally administrated vitamin E and Se on total lipid, total cholesterol, and fatty acid composition of rat liver and muscle tissues. Total lipid content of muscle tissue in Se and combination groups decreased as compared to the control group. However, the level of total lipid in the liver tissues was seen to decrease only in the combination group (P < 0.05). While the amount of total cholesterol in liver tissue was lower (P < 0.05) in the vitamin E and combination groups, the amount of total cholesterol in muscle tissue decreased (P < 0.05) in the combination group. The amount of linoleic acid in muscle tissue slightly decreased (P < 0.05), whereas the eicosenoic and eicosatrienoic acid amounts significantly increased (P < 0.01, P < 0.001) in the vitamin E group as compared to the control group. The amounts of most fatty acid decreased (P < 0.05) in the combination group. The proportions of eicosenoic, eicosatrienoic, and total polyunsaturated fatty acid (PUFA) within the total fatty acid were higher (P < 0.05) in vitamin E group, whereas these fatty acids proportions were lower (P < 0.05) in the Se group. Although the proportions of palmitic, linolenic, and total saturated fatty acids were low (P < 0.05), oleic and total unsaturated fatty acid proportions were higher (P < 0.05) in the combination group than in the control group. The amount of palmitic acid and total saturated fatty acid in liver tissue decreased (P < 0.01 and P < 0.05, respectively) in the vitamin E and combination groups. However, the amount of linoleic acid only decreased (P < 0.05) in the combination group. The amount of PUFA was slightly higher (P < 0.05) in vitamin E. The proportions of stearic acid and linoleic acid decreased (P < 0.05) both in the Se and combination groups. However, the proportions of eicosatrienoic, ω 6, and PUFA were slightly higher (P < 0.05) in the vitamin E group, but total saturated fatty acid proportion significantly decreased (P < 0.01) in both the vitamin E and combination groups. In conclusion, the level of total lipid and cholesterol in muscle and liver tissues were reduced by administrating vitamin E and Se together. Additionally, the fatty acid synthesis in the muscle and liver tissues was decreased by this process. However, it was observed that the protective effect of intraperitoneally administrated vitamin E was higher than Se on fatty acid composition in muscle and liver tissues. J. Cell. Suckling mammals, including mice, differ from adults in the abundant expression of enzymes that synthesize arginine from citrulline in their enterocytes. To investigate the importance of the small-intestinal arginine synthesis for whole-body arginine production in suckling mice, we floxed exon 13 of the argininosuccinate synthetase (Ass) gene, which codes for a key enzyme in arginine biosynthesis, and specifically and completely ablated Ass in enterocytes by crossing Ass fl and Villin-Cre mice. Unexpectedly, Ass fl/fl /VilCre tg/- mice showed no developmental impairments. Amino-acid fluxes across the intestine, liver, and kidneys were calculated after determining the blood flow in the portal vein, and hepatic and renal arteries (86%, 14%, and 33%, respectively, of the transhepatic blood flow in 14-day-old mice). Relative to control mice, citrulline production in the splanchnic region of Ass fl/fl /VilCre tg/- mice doubled, while arginine production was abolished. Furthermore, the net production of arginine and most other amino acids in the liver of suckling control mice declined to naught or even changed to consumption in Ass fl/fl /VilCre tg/- mice, and had, thus, become remarkably similar to that of post-weaning wild-type mice, which no longer express arginine-biosynthesizing enzymes in their small intestine. The adaptive changes in liver function were accompanied by an increased expression of genes involved in arginine metabolism (Asl, Got1, Gpt2, Glud1, Arg1, and Arg2) and transport (Slc25a13, Slc25a15, and Slc3a2), whereas no such changes were found in the intestine. Our findings suggest that the genetic premature deletion of arginine synthesis in enterocytes causes a premature induction of the post-weaning pattern of amino-acid metabolism in the liver.  相似文献

The genetic mechanisms underlying fruit development have been identified in Arabidopsis and have been comparatively studied in tomato as a representative of fleshy fruits. However, comparative expression and functional analyses on the bHLH genes downstream the genetic network, ALCATRAZ (ALC) and SPATULA (SPT), which are involved in the formation of the dehiscence zone in Arabidopsis, have not been functionally studied in the Solanaceae. Here, we perform detailed expression and functional studies of ALC/SPT homologs in Nicotiana obtusifolia with capsules, and in Capsicum annuum and Solanum lycopersicum with berries. In Solanaceae, ALC and SPT genes are expressed in leaves, and all floral organs, especially in petal margins, stamens and carpels; however, their expression changes during fruit maturation according to the fruit type. Functional analyses show that downregulation of ALC/SPT genes does not have an effect on gynoecium patterning; however, they have acquired opposite roles in petal expansion and have been co‐opted in leaf pigmentation in Solanaceae. In addition, ALC/SPT genes repress lignification in time and space during fruit development in Solanaceae. Altogether, some roles of ALC and SPT genes are different between Brassicaceae and Solanaceae; while the paralogs have undergone some subfunctionalization in the former they are mostly redundant in the latter.  相似文献

Dinitrogen fixation in white clover (Trifolium repens L.) grown in pure stand and mixture with perennial ryegrass (Lolium perenne L.) was determined in the field using 15N isotope dilution and harvest of the shoots. The apparent transfer of clover N to perennial ryegrass was simultaneously assessed. The soil was labelled either by immobilizing 15N in organic matter prior to establishment of the sward or by using the conventional labelling procedure in which 15N fertilizer is added after sward establishment. Immobilization of 15N in the soil organic matter has not previously been used in studies of N2 fixation in grass/clover pastures. However, this approach was a successful means of labelling, since the 15N enrichment only declined at a very slow rate during the experiment. After the second production year only 10–16% of the applied 15N was recovered in the harvested herbage. The two labelling methods gave, nonetheless, a similar estimate of the percentage of clover N derived from N2 fixation. In pure stand clover, 75–94% of the N was derived from N2 fixation and in the mixture 85–97%. The dry matter yield of the clover in mixture as percentage of total dry matter yield was relatively high and increased from 59% in the first to 65% in the second production year. The average daily N2 fixation rate in the mixture-grown clover varied from less than 0.5 kg N ha−1 day−1 in autumn to more than 2.6 kg N ha−1 day−1 in June. For clover in pure stand the average N2 fixation rate was greater and varied between 0.5 and 3.3 kg N ha−1 day−1, but with the same seasonal pattern as for clover in mixture. The amount of N fixed in the mixture was 23, 187 and 177 kg N ha−1 in the seeding, first and second production year, respectively, whereas pure stand clover fixed 28, 262 and 211 kg N ha−1 in the three years. The apparent transfer of clover N to grass was negligible in the seeding year, but clover N deposited in the rhizosphere or released by turnover of stolons, roots and nodules, contributed 19 and 28 kg N ha−1 to the grass in the first and second production year, respectively. The distribution patterns of flavonoids hyperoside, isoquercitrin, quercitrin, quercetin, I3,II8-biapigenin and naphtodianthrones hypericin and pseudohypericin were studied in reproductive structures during ontogenetic phase of flowering in Hypericum maculatum Crantz. Considerable differences in the content of these secondary metabolites, in the particular flower parts were found. The content of all the metabolites studied is stable during the whole period of flowering in green flower parts (sepals). In petals, stamens and pistils their content undergoes considerable change associated with the biological functions of particular metabolites. The most conspicuous changes during ontogenetic phase of flowering were the decrease of hyperoside and isoquercitrin content in petals (average content in buds 1.589 mg g−1 dry weight, average content in overblown flowers 0.891 mg g−1 dry weight), the decrease of the I3,II8-biapigenin content in stamens (in buds 1.189 mg g−1 dry weight, in overblown flowers 0.319 mg g−1 dry weight), and the increase of hypericin and pseudohypericin content in both petals (total average content of hypericins in the buds 0.547 mg g−1 dry weight; in overblown flowers 0.792 mg g−1 dry weight) and stamens (in buds 0.189 mg g−1 dry weight; in overblown flowers 0.431 mg g−1 dry weight). Hypericins are absent in the pistil. The flavonoids hyperoside and isoquercitrin, the content of which decreased during ontogenetic phase of flowering, reach the highest contents in the pistil.  相似文献

Net Ca2+ and Mg2+ absorption rates were measured in vivo from buffer solutions placed in the washed reticulo-rumen, isolated in situ in 30 conscious, trained sheep. An increase in concentration of short chain fatty acids (SCFA) in the buffer, over the range 0–50 mM, was shown to stimulate the net rates of absorption of Ca2+ and Mg2+ ions from the rumen. Similarly, the results of in vitro experiments, carried out with ovine rumen epithelium mounted in short-circuited Ussing chambers, showed that the absence of SCFA from the chamber fluid resulted in a reduction in Jnet Ca2+ caused by reduced flux of Ca2+ ions in the mucosal to serosal direction (Jms Ca2+). The addition of 1 mM acetazolamide, an inhibitor of carbonic anhydrase, to the ruminal buffer used in the in vivo experiments led to significant reductions in the net absorption rates of Ca2+and Mg2+ ions in the presence of SCFA (50 mmol l−1) but not in the absence of SCFA. However, in the in vitro experiments, the addition of 60 μM ethoxyzolamide had no significant effect on Jnet Ca2+. A reduction in pH of the intraruminal buffer in vivo from 6.8 to 5.4 led to significant increases in the net absorption rates of Ca2+and Mg2+ ions, an effect which was duplicated for Ca2+ in preliminary in vitro experiments in which the pH of the mucosal buffer was reduced from 7.4 to 5.4. This stimulatory effect was confined to Jms Ca2+ and Jnet Ca2+. Ussing chambers were also used to demonstrate that Jnet Ca2+ was reduced by a high transmural potential difference (PD), caused by voltage clamping, independently of the mucosal K+ concentration. Both unidirectional Ca2+ fluxes consisted of a PD-dependent and a K+-insensitive PD-independent component. The latter may be represented by a Ca2+/2H+ antiporter. It is postulated that SCFA, and to a lesser extent H2CO3, can stimulate Jms Ca2+ by activation of an apical Ca2+/2H+ antiporter through the provision of protons within the ruminal epithelial cell. A mild reduction in ruminal pH may also lead to a similar stimulation of this putative electroneutral exchange. Accepted: 26 July 2000  相似文献
