A series of conditional mutants of the algal, biflagellate Chlamydomonas reinhardtii with temperature-sensitive defects in flagellar assembly and function were isolated. The genetics and phenotypes of 21 mutants displaying a rapid alteration in flagellar function upon shift from the permissive (20 degrees C) to the restrictive (32 degrees C) temperatures are described. These mutants designated as "drop-down" or dd-mutants have been placed in four categories on the basis of their defective phenotypes: (a) dd-assembly mutants - the preformed flagella are resorbed at 32 degrees C and reassembly of flagella is inhibited; (b) dd-fragile flagella mutants - the flagella are lost by detachment at 32 degrees C, but can be reassembled; (c) dd-motility mutants - the flagella are retained at 32 degrees C, but are functionally defective; (d) dd-lethal mutants - display combined defects in flagellar function and cell growth. Tetrad analysis of the mutants back-crossed to wild-type, recombination analysis of intermutant crosses, and complementation tests in the construction of heterozygous diploid strains indicate that at least 14 nuclear genetic loci are represented among 21 mutants. The availability of temperature-sensitive mutations affecting the assembly and function of the flagellum suggests that the morphogenesis of this complex eukaryotic organelle is amenable to genetic dissection.  相似文献

Autolytic enzymes were found to be required for flagellar morphogenesis in Bacillus subtilis 168 and Bacillus licheniformis 6346. Two previously characterized, poorly lytic, chain-forming mutants of B. subtilis 168, strains FJ3 (temperature conditional) and FJ6, each 90 to 95% deficient in the production of N-acetylmuramyl-L-alanine amidase and endo-beta-N-acetylglucosaminidase, were observed to be nonmotile at 35 degrees C in a variety of liquid and semisolid meida. In contrast, cells of the isogenic wild-type strain were motile and fully separated. Electron microscopy revealed the complete absence of flagella on the mutant cells. Similar observations were made with another poorly lytic strain of B. subtilis 168 (Nil5) and with two poorly lytic, phosphoglucomutase-deficient mutants of B. licheniformis 6346 (MH-3, MH-5). In minimal media lacking galactose (restrictive conditions), the B. licheniformis mutants failed to form flagella, or had serious abnormalities in flagellar morphogenesis and motility. Under permissive conditions, mutants FJ3 (grown at 17 degrees C) and MH-5 (grown with addend galactose) showed increased autolytic activities, grew in the dechained form, and regained their capacities to synthesize functional flagella. Examination of several classes of spontaneous revertants derived from the various mutant strains further demonstrated a close relationship between autolysin acttivity and flagellation in the two Bacillus spp.  相似文献

During spermiogenesis, haploid round spermatids undergo dramatic cell differentiation and morphogenesis to give rise to mature spermatozoa for fertilization, including nuclear elongation, chromatin remodeling, acrosome formation, and development of flagella. The molecular mechanisms underlining these fundamental processes remain poorly understood. Here, we report that MNS1, a coiled-coil protein of unknown function, is essential for spermiogenesis. We find that MNS1 is expressed in the germ cells in the testes and localizes to sperm flagella in a detergent-resistant manner, indicating that it is an integral component of flagella. MNS1-deficient males are sterile, as they exhibit a sharp reduction in sperm production and the remnant sperm are immotile with abnormal short tails. In MNS1-deficient sperm flagella, the characteristic arrangement of "9+2" microtubules and outer dense fibers are completely disrupted. In addition, MNS1-deficient mice display situs inversus and hydrocephalus. MNS1-deficient tracheal motile cilia lack some outer dynein arms in the axoneme. Moreover, MNS1 monomers interact with each other and are able to form polymers in cultured somatic cells. These results demonstrate that MNS1 is essential for spermiogenesis, the assembly of sperm flagella, and motile ciliary functions.  相似文献

Pili, along with the flagellum and DNA bacteriophage receptors, are structural markers for polar morphogenesis in Caulobacter crescentus. Pili act as primary receptors for a number of small, C. crescentus-specific DNA and RNA bacteriophages, and the timing of pilus-dependent adsorption of bacteriophage phiCb5 in synchronized cell populations has led to the general conclusion that pili are formed coordinately with the flagellum and other polar surface structures in the predivisional cell. The use of rotary platinum shadow casting and electron microscopy as a direct assay for formation of flagella and pili in synchronous cell cultures now shows, however, that when expressed as fractions of the swarmer cell cycle, flagella are assembled on the predivisional cells at approximately 0.8 and that pili are assembled on the new swarmer cells at approximately 0.1 of the next cell cycle. Adsorption of pilus-specific bacteriophage phiCb5 prevented the loss of pili from swarmer cells during development, which suggests that these structures are retracted at the time of stalk formation. Examination of temperature-sensitive cell division mutants showed that the assembly of pili depends on completion of cell separation. These results indicate that the stage-specific events required for polar morphogenesis in C. crescentus occur sequentially, rather than coordinately in the cell cycle, and that the timing of these events reflects the order of underlying cell cycle steps.  相似文献

Flagella are sophisticated organelles found in many eukaryotic microbes where they perform functions related to motility, signal detection, or cell morphogenesis. In many cases, several flagella are present per cell, and these can have a different composition, length, age, or function, raising the question of how this is managed. When the flagella are equivalent and constructed simultaneously such as in Chlamydomonas or Naegleria, we propose an equal access model where molecular components have free access to each organelle. By contrast, Trypanosoma and Leishmania contain temporally distinct organelles and elongate a new flagellum whilst maintaining the existing one. The equal access model could function providing that the mature flagellum is “locked” so that it can no longer be elongated or shortened. Alternatively, access of flagellar components could be restricted at the level of the basal body, the transition zone, or the loading on intraflagellar transport trains. In organisms that contains flagella of different age and composition such as Giardia, a temporal dimension is necessary, with the production of protein components of flagella spreading over one or more cell cycles. In the future, deciphering the molecular mechanisms involved in these processes should reveal new insights in flagellum assembly and function.  相似文献

The behaviour of a pool of flagellar precursors, assayed by the ability of cells to regenerate flagella in the absence of de novo protein synthesis, has been examined during organelle morphogenesis in the biflagellate alga Chlamydomonas. The results demonstrate that flagellar elongation can continue even when this pool is apparently empty and suggest that 2 sources of precursors are available to the regenerating flagella: those pre-existing in the cellular pool and those synthesized de novo. Further evidence for this was obtained by subjecting regenerating cells to pulses of cycloheximide. Cells exposed to this drug during the first 60 min post deflagellation formed only half-length (5-mum) flagella, whereas a pulse administered after this point allowed the formation of longer flagella and suggested that some de novo protein synthesis was required for the formation of full-length flagella, although it was not a prerequisite for the initiation of regeneration. In addition, it was found that, subsequent to the removal of the cycloheximide, flagellar regeneration did not recommence immediately, but was delayed for a period of approximately 45 min, irrespective of length of flagella formed prior to drug inhibition. The nature of this cycloheximide-induced delay is unclear and certain alternatives, based on the exhaustion of structural/regulatory components are considered. Although it is not possible to distinguish between these alternatives, tubulin is not the limiting component, since a pool of this protein is present when flagellar elongation is prevented by cycloheximide.  相似文献

Abstract: Using an experimental approach similar to that used for Euglena flagella, we found that flagella and flagellar membrane preparations (isoagglutinins) of the unicellular green algae Chlamydomonas moewusii and C. reinhardtii , but not cells without flagella, bind radiolabelled riboflavin with high affinity and specificity. In addition, flagella and isoagglutinins contain high amounts of methanol-extractable flavins. These results indicate an abundance of proteins with high affinity for riboflavin in the flagella. Since sexual adhesiveness of gametic flagella in C. moewusii is controlled by light, the possibility is discussed that flavoproteins in the flagella are involved in this reaction. Action spectra exhibit maxima at 450 and 600 nm, suggesting–at least for the 450 nm band–a typical blue-light receptor.  相似文献

Summary— The morphogenesis of basal bodies is described in the phytoflagellate Polytoma papillatum. The observations are based on the analysis of ultrathin serial sections through the flagellar apparatus of interphase, mitotic, and postmitotic cells using transmission electron microscopy. Formation of new basal bodies starts in prometaphase. Individual A-subfibres develop orthogonally to the long axis of mature basal bodies. The microtubules assemble at the surface of an annulus of amorphous material. By telophase, a complete cylinder of A-subfibres with a length of approximately 300 nm has formed. Although the proximal ends of these new probasal bodies are detached from the mature basal bodies, prominent reorientation of the probasal bodies does not occur. They remain with their proximal ends in the vicinity of mature basal bodies. In daughter cells with probasal bodies around 400 nm long, the assembly of microtubular triplets is initiated. B- and C-subfibres first show up distal from the mature basal bodies and may elongate towards them. Thus, A-subfibres on the one side and B- and C-subfibres on the other appear to growt with opposite polarity. If A-subfibres grow at their plus ends, B- and C-subfibres elongate at their minus ends. The latter is unusual in comparison with individual cytoplasmic and spindle microtubules. Possible the presence of a lateral template in the form of the A-subfibres is responsible for the deviating growth characteristics of the incomplete B- and C-subfibres. In interphase cells, the mature basal bodies extend into long flagella. The new basal bodies remain devoid of flagella and are less than 85 nm long. Thus, they have shortened relative to their precursors in mitotic and postmitotic cells. At the onset of a new division cycle, the flagellate basal badies shed their flagella. The breaking point is at the triplet-doublet transition of the flagellum.  相似文献

A stalk, a single falgellum, several pili, and deoxyribonucleic acid (DNA) phage receptors are polar surface structures expressed at a defined time in the Caulobacter crescentus cell cycle. When mutants were isolated as DNA phage phiCbK-resistant or ribonucleic acid (RNA) phage phiCp2-resistant, as well as nonmotile, strains, 5 out of 30 such mutant isolates were found not to possess stalks, but did possess inactive flagella. These stalkless mutants were resistant simultaneously to both DNA and RNA phages and did not possess pili and DNA pendent stalkless mutants. All motile revertants simultaneously regained the capacity to form stalks and susceptibility to DNA and RNA phages. It is suggested that a single mutation pleiotropically affects stalk formation, flagella motility, and coordinate polar morphogenesis of pili and DNA phage receptors. The stalkless mutants grew at a generation time similar to that of the wild-type strain at 30 degrees C. Cell size and morphology of a stalkless mutant, C. crescentus CB13 pdr-819, were also similar to those of the wild-type strain, except for the absence of a stalk. In addition, the CB13 pdr-819 predivisional cells were partitioned into smaller and larger portions, indicating asymmetrical cell division, as in the wild-type strain. From these results, it is suggested that swarmer cells undergo transition to cells of a stalked-cell nature without stalk formation and that the cell cycle of the stalkless mutant proceeds in an ordered sequence similar to that defining the wild-type cell cycle.  相似文献

Summary Amoebo-flagellate cells develop upon spore germination in the protostelidProtosporangium articulatum. The germling may emerge flagellate or as an amoeba. In either case the cell undergoes mitosis within an hour of germination. The spindle is open and centric, and typically has several pairs of kinetosomes at the poles. During telophase, the kinetosomes are found at the surface of the cell and flagella and flagellar rootlets begin to develop. Some flagella remain in close association with the nucleus, the nucleus-associated flagella; others are located away from the nucleus, the supernumerary flagella. The flagellar apparatus is identical for both nucleus-associated flagella and supernumerary flagella. However, only the nucleus-associated flagella are able to generate the jerking, helical swim typical of amoebo-flagellates with a swarm cell-like morphology.  相似文献

Helicobacter pylori colonizes the human stomach and can cause gastroduodenal disease. Flagellar motility is regarded as a major factor in the colonizing ability of H. pylori. The functional roles of flagellar structural proteins other than FlaA, FlaB, and FlgE are not well understood. The fliD operon of H. pylori consists of flaG, fliD, and fliS genes, in the order stated, under the control of a sigma(28)-dependent promoter. In an effort to elucidate the function of the FliD protein, a hook-associated protein 2 homologue, in flagellar morphogenesis and motility, the fliD gene (2,058 bp) was cloned and isogenic mutants were constructed by disruption of the fliD gene with a kanamycin resistance cassette and electroporation-mediated allelic-exchange mutagenesis. In the fliD mutant, morphologically abnormal flagellar appendages in which very little filament elongation was apparent were observed. The fliD mutant strain was completely nonmotile, indicating that these abnormal flagella were functionally defective. Furthermore, the isogenic fliD mutant of H. pylori SS1, a mouse-adapted strain, was not able to colonize the gastric mucosae of host mice. These results suggest that H. pylori FliD is an essential element in the assembly of the functional flagella that are required for colonization of the gastric mucosa.  相似文献

During intraflagellar transport (IFT), the regulation of motor proteins, the loading and unloading of cargo and the turnover of flagellar proteins all occur at the flagellar tip. To begin an analysis of the protein composition of the flagellar tip, we used difference gel electrophoresis to compare long versus short (i.e., regenerating) flagella. The concentration of tip proteins should be higher relative to that of tubulin (which is constant per unit length of the flagellum) in short compared with long flagella. One protein we have identified is the cobalamin-independent form of methionine synthase (MetE). Antibodies to MetE label flagella in a punctate pattern reminiscent of IFT particle staining, and immunoblot analysis reveals that the amount of MetE in flagella is low in full-length flagella, increased in regenerating flagella, and highest in resorbing flagella. Four methylated proteins have been identified in resorbing flagella, using antibodies specific for asymmetrically dimethylated arginine residues. These proteins are found almost exclusively in the axonemal fraction, and the methylated forms of these proteins are essentially absent in full-length and regenerating flagella. Because most cells resorb cilia/flagella before cell division, these data indicate a link between flagellar protein methylation and progression through the cell cycle.  相似文献

We have found that several kinds of helical flagella from Salmonella and Escherichia become straight in the presence of 0·5 m-citric acid at pH values below 4·0, while the straight flagella from a mutant Salmonella (SJ814) are transformed into a helical shape under the same conditions. These transformations are reversible and transitional.Current models of bacterial flagella (Calladine, 1976,1978; Kamiya, 1976) predict that the family of distinct wave-forms should include two types of straight flagella, which have either an extreme right-handed twist (about 7 ° at the surface of the flagellum) or an extreme left-handed twist (2 ° to 3 °). As the inclination of the near-longitudinal rows of subunits in the Salmonella SJ814 flagellum (O'Brien &; Bennett, 1972) agrees closely with the degree of twisting predicted for the right-handed type, this flagellum has been considered to be the right-handed type. We have determined that the basic (1-start) helix in flagella is right-handed, using the method of Finch (1972). This fact, together with the selection rule (O'Brien &; Bennett, 1972), strongly suggests that the near-longitudinal rows in an SJ814 flagellum are right-handed, in agreement with the prediction. However, our optical diffraction and X-ray diffraction studies have revealed that the near-longitudinal rows of subunits in the citric acid-induced straight flagella and in the straight flagella from a mutant E. coli (Kondoh &; Yanagida, 1975) tilt at an angle of 2 ° to 3 ° with respect to the flagellar axis. This inclination is probably left-handed. Thus the predicted presence of the two types of straight flagella seems to be proved.  相似文献

ATP and ADP are known to play inhibitory and activating roles, respectively, in the regulation of dynein motile activity of flagella. To elucidate how these nucleotide functions are related to the regulation of normal flagellar beating, we examined their effects on the motility of reactivated sea urchin sperm flagella at low pH. At pH 7.0-7.2 which is lower than the physiological pH of 8, about 90% of reactivated flagella were motionless at 1 mM ATP, while about 60% were motile at 0.02 mM ATP. The motionless flagella at 1 mM ATP maintained a single large bend or an S-shaped bend, indicating formation of dynein crossbridges in the axoneme. The ATP-dependent inhibition of flagellar movement was released by ADP, and was absent in outer arm-depleted flagella. Similar inhibition was also observed at 0.02 mM ATP when demembranated flagella were reactivated in the presence of Li+ or pretreated with protein phosphatase 1 (PP1). ADP also released this type of ATP-inhibition. In PP1-pretreated axonemes the binding of a fluorescent analogue of ADP to dynein decreased. Under elastase-treatment at pH 8.0, the beating of demembranated flagella at 1 mM ATP and 0.02 mM ATP lasted for approximately 100 and 45 s, respectively. The duration of beating at 0.02 mM ATP was prolonged by Li+, and that at 1 mM ATP was shortened by removal of outer arms. These results indicate that the regulation of on/off switching of dynein motile activity of flagella involves ATP-induced inhibition and ADP-induced activation, probably through phosphorylation/dephosphorylation of outer arm-linked protein(s).  相似文献

Flagellar gene networks are fascinating, owing to their complexity - they usually coordinate the expression of more than 40 genes - and particular wiring that elicits temporal expression coupled to organelle morphogenesis. Moreover, many of the lessons learned from flagellar regulation are generally applicable to type III secretion systems. Our understanding of flagellar networks is rapidly expanding to include diverse organisms, as well as deepening to enable the development of predictive wiring diagrams. Numerous regulators control the regulation of flagella, and one of the next challenges in the field is to integrate flagellar gene control into master blueprints of global gene expression.  相似文献

在完成小绒泡菌Physaum pusillum生活史的同时,观察了小绒泡菌P. pusillum生活史不同阶段显微结构的形成。生活史研究结果表明:小绒泡菌P. pusillum的孢子萌发方式为裂式,黏变形体具两根鞭毛,较短一根不易观察到,原质团类型为显型原质团,子实体形态建成过程约需12-13h。显微结构观察结果表明:石灰结和孢丝结构在原质团隆起约6h后开始形成,孢子在原质团隆起约9h后开始形成,石灰结和孢丝先于孢子形成。  相似文献

Video cameras with contrast and black level controls can yield polarized light and differential interference contrast microscope images with unprecedented image quality, resolution, and recording speed. The theoretical basis and practical aspects of video polarization and differential interference contrast microscopy are discussed and several applications in cell biology are illustrated. These include: birefringence of cortical structures and beating cilia in Stentor, birefringence of rotating flagella on a single bacterium, growth and morphogenesis of echinoderm skeletal spicules in culture, ciliary and electrical activity in a balancing organ of a nudibranch snail, and acrosomal reaction in activated sperm.  相似文献

The ultrastructure of the vas deferens, testes, spermatogenesis and spermatozoa of Gyrocotyle urna and G. parvispinosa is described. The vas deferens is ciliated and syncytial. Within the testes primary spermatocytes arise from the primary spermatogonia by incomplete mitotic divisions; the primary spermatocytes undergo two meiotic divisions leading to spermatids. In early spermatids microtubules are formed at the cell periphery. Later the spermatozoal cytoplasm (the ‘middle-piece’) grows out and the two spermatozoal flagella with their typical 9 + ‘1’ axonemes are formed. During ciliogenesis the flagella are at an angle of about 60° to the axis of the middle-piece. The flagella are inserted into basal bodies terminating in striated rootlets. Subsequently, the nucleus and isolated mitochondria migrate into the central axis. The angle between the flagella and the axis decreases; the flagella are incorporated to form the spermatozoon. In mature spermatozoa no basal body or rootlet elements were found. The phylogeny of parasitic Platyhelminthes is discussed with respect to the evolution of spermatozoa. The reduction of the acrosinoid granules which are found in spermatozoa of free-living Platyhelminthes and the incorporation of the spermatozoal flagella into the sperm body constitute autapomorphies of the Neodermata (the parasitic Platyhelminthes). Included in the Cestoda because of several common derived characters, Amphilinidea and Gyrocotylidea are the only cestodes with spermatozoa containing mitochondria. Their absence in Cestoidea—all taxa with a six-hooked larva and other characteristics—is an autapomorphy of this group.  相似文献
