Cattle ticks, Boophilus annulatus (Say), previously reared only on cattle, were placed on 3 white-tailed deer, Odocoileus virginianus L. Ticks were maintained through successive generations solely on the same deer as they aged (3, 6, and 9 mo of age) and received repeated challenges (0, 1, and 2 previous challenges). Cattle were infested simultaneously to assess tick viability and provide a comparison of tick numbers, female weight, egg mass weight, and egg hatch. The initial infestation (3,000 larvae/animal) produced a mean of 12.7 and 506.7 females from deer and cattle, respectively. Ticks recovered from deer weighed less, laid smaller egg masses, and had lower egg hatchability than cattle-reared ticks. A second infestation (3,000 larvae/animal) produced a 6.3-fold reduction in tick numbers on deer (means = 2.0 females/deer), whereas the number on cattle increased (means = 578.0 females/calf). Ticks reared on the deer were again smaller, laid fewer eggs, and had lower egg hatch, although differences were not significant. A third infestation of deer (1,900 larvae/deer) produced only 1 engorged female tick and no viable eggs, thus eliminating the population of deer-reared ticks within 3 generations. Results of the study suggest that a population of B. annulatus will not be sustained indefinitely through time solely on deer; thus, efforts to reduce deer populations severely as a means of eradicating ticks are unnecessary.  相似文献

A penned study for obtaining definitive information on the status of white-tailed deer (Odocoileus virginianus) as a host for cattle feverticks (Boophilus microplus) was conducted on St. Croix of the U.S. Virgin Islands. Four generations of fever-ticks were propagated on one deer during a six month period. Nine wild white-tailed deer also were collected from four insular estates to evaluate the carrier status of these animals on an island where cattle fever-ticks are indigenous. Two deer were infested with B. microplus where contact with domestic livestock had not occurred for 20 years; five deer were free of B. microplus where a vigorous cattle dipping program had been practiced for three years; and, two deer were infested with B. microplus where contact with fever-tick infested cattle occurred at irregular intervals. It was concluded that white-tailed deer constitute a host species for B. microplus and must be considered in future fever tick eradication endeavors. This study also suggested that, through routine dipping of cattle, fever ticks may be eradicated from an area where cattle and deer cohabit the same premises.  相似文献

Tick resistance and blood composition were studied in British (1/2 Shorthorn x 1/2 Hereford) and zebu (1/2 Brahman x 1/2 British) calves from birth to 33 days of age in a tropical grazing area in which B. microplus is endemic. Calves of the 2 breeds were either naturally infested or were, in addition, artificially infested with 5,000 larvae at 2 and 9 days of age. Total numbers of mature female ticks carried from either type of infestation were significantly lower (P less than 0.01) on zebu than on British calves. In the artificially infested calves of both breeds, the total number of ticks maturing between 20 to 26 days of age was significantly higher (P less than 0.01) than the number maturing between 27 to 33 days of age emphasizing that a major component of resistance is acquired. However, in the naturally infested calves, breed differences in the numbers of ticks maturing during these 2 periods suggested the presence of genetic differences in innate resistance. Within breeds, total tick numbers carried during the study were negatively correlated with calf weight gain and with the concentrations of serum albumin, total protein, and cholesterol.  相似文献

Rapid and sensitive detection of resistance to insecticides in arthropods is needed. In the cattle tick, Boophilus microplus, resistance to a variety of acaricides is widespread. The most commonly used assay for resistance, the larval packet test, takes at least two, but generally six weeks for a one-host tick like B. microplus to complete and may take up to three months to complete for three-host ticks. Here we describe a test for resistance to organophosphate acaricides that can be used on larvae and adult ticks which takes less than 24 hours. The test measures the difference in acetylcholinesterase (AChE) activity in homogenates of ticks in the presence and absence of propoxur, a carbamate acaricide. We found clear discrimination of organophosphate-susceptible and organophosphate-resistant adults with 100 M propoxur. AChE from susceptible ticks had almost no activity at this concentration of propoxur whereas AChE from resistant ticks had 67% of its potential activity. AChE from heterozygote ticks could also be distinguished from AChE from homozygous-susceptible and homozygous-resistant ticks. This is the first biochemical test for resistance to an acaricide. Rapid, sensitive tests like ours will allow resistance to organophosphates to be detected soon after it develops in the field, thus, the spread of resistance might be slowed and the useful life of acaricides extended.  相似文献

Larvae of Boophilus microplus were labelled with [32P] and used to study the rejection of larvae from British breed cattle with different levels of resistance. On animals of high resistance the loss of larvae due to grooming ranged from 9-54% during the first 24 h of infestation, and more time was spent grooming by these animals. Most of the grooming activity could be attributed to the presence of tick larvae and the presence of older tick stages did not increase the percentage of larvae lost. Grooming was directed to attached larvae and these could be removed. Animals of low resistance did not lose a significant number of larvae as a result of grooming, but all previously infested hosts lost a proportion of the larvae (18-39%) which could not be accounted for by grooming. This loss was greater than the total loss of larvae (11%) on animals not previously exposed to B. microplus.  相似文献

Octopamine is a biogenic amine neurotransmitter of invertebrates that binds to a G-protein coupled receptor that has seven transmembrane domains. Formamidine pesticides like amitraz are highly specific agonists of the octopamine receptor. Amitraz is used extensively to control the cattle tick, Boophilus microplus, and many other ticks but now there are strains of ticks that are resistant to amitraz. We have isolated a cDNA from the cattle tick, B. microplus, that belongs to the biogenic amine family of receptors. The predicted amino acid sequence from this cDNA is most similar to octopamine receptors from insects. The nucleotide sequence of this gene from amitraz-resistant and amitraz-susceptible cattle ticks was identical. Thus, a point mutation/s did not confer resistance to amitraz in the strains we studied. Alternative explanations for resistance to amitraz in B. microplus are discussed.  相似文献

The efficacy of coumaphos, an organophosphorus acaricide, was tested on a strain of Boophilus microplus (Canestrini) that was 9.5 times more resistant to the chemical than a susceptible strain. Cattle infested with larval, nymphal, and adult ticks were dipped into a vat charged at concentrations of 0.031, 0.083, 0.160, and 0.279% (AI). Assessment of efficacy was based on surviving tick numbers, female weight, egg mass weight, egg hatchability, and index of reproduction. At 0.031% (AI), the level of control was lower than the other coumaphos treatments, and the weight, fecundity, and fertility of surviving females was similar to untreated females, indicating only minimal adverse effects on the ticks. At 0.160 and 0.279% (AI), no differences in control, female weight, egg mass weight, or percentage egg hatch were observed, but effects were greater than that of untreated ticks or ticks treated at 0.031% (AI). At 0.083% (AI), most biological parameters were intermediate, but there was no difference in control compared with the 2 higher concentrations. Thus, even with the increase in concentration of more than 3 times between treatments of 0.083 and 0.279% (AI), there was no substantial increase in control. The residual efficacy of all 4 treatments showed that none provided complete protection against larval reinfestation for even 1 wk after treatment. Therefore, the presence of organophosphorus-resistant ticks at U.S. ports-of-entry could jeopardize the success of the Boophilus eradication program.  相似文献

We describe the isolation and characterisation of two putatively new acetylcholinesterase genes from the African cattle ticks Boophilus decoloratus and Rhipicephalus appendiculatus. The nucleotide sequences of these genes had 93% homology to each other and 95% and 91% identity, respectively, to the acetylcholinesterase gene from an Australian strain of another cattle tick, Boophilus microplus. Translation of the nucleotide sequences revealed putative amino acids that are essential for acetylcholinesterase activity: the active site serine, and the histidine and glutamate residues that associate with this serine to form the catalytic triad. All known acetylcholinesterases have three sets of cysteines that form disulfide bonds; however, the acetylcholinesterase genes of these three species of ticks encode only two sets of cysteines. Acetylcholinesterases of B. microplus from South Africa, Zimbabwe, Kenya and Mexico had 98-99% identity with acetylcholinesterase from B. microplus from Australia, whereas acetylcholinesterase from B. microplus from Indonesia was identical to that from Australia. Preliminary phylogenetic analyses surprisingly indicate that the acetylcholinesterases of ticks are closer phylogenetically to acetylcholinesterases of vertebrates than they are to those of other arthropods.  相似文献

PCR and nested-PCR methods were used to assess the frequency of Babesia bovis and Babesia bigemina infection in Boophilus microplus engorged females and eggs and in cattle reared in an area with endemic babesiosis. Blood and the engorged female ticks were from 27 naturally infested calves and 25 crossbred cows. The frequency of both Babesia species was similar in calves and cows (P>0.05). Babesia bovis was detected in 23 (85.2%) calves and in 25 (100%) cows and B. bigemina was detected in 25 (92.6%) calves and in 21 (84%) cows. Mixed infections with the both Babesia species were identified in 42 animals, 21 in each age category. Of female ticks engorged on calves, 34.9% were negative and single species infection with B. bigemina (56.2%) was significantly more frequent (P<0.01) than with B. bovis (4.7%). Most of the females (60.8%) engorged on cows did not show Babesia spp. infection and the frequency of single B. bovis infection (17.6%) was similar (P>0.05) to the frequency of single B. bigemina infection (15.9%). Mixed Babesia infection was lower (P<0.01) than single species infection in female ticks engorged either in cows (5.7%) or in calves (4.3%). An egg sample from each female was analysed for the presence of Babesia species. Of the egg samples from female ticks infected with B. bovis, 26 (47.3%) were infected while from those from female ticks infected with B. bigemina 141 (76.6%) were infected (P<0.01). The results showed that although the frequency of both species of Babesia was similar in calves and cows, the infectivity of B. bigemina was higher to ticks fed on calves while to those ticks fed on cows the infectivity of both Babesia species was similar.  相似文献

In India, control of tick infestations is a major problem facing the owners of highly productive crossbred cattle. To develop suitable immunoprotective measures against the multitick infestations, specific problem encountered by the Indian farmers, an attempt has been made to identify candidate protein molecules in the larvae of Hyalomma anatolicum anatolicum and Boophilus microplus. Employing two steps affinity chromatography method, glycoproteins of 34 and 29 kDa were isolated from H. a. anatolicum and B. microplus, respectively. The additive effect of the isolated glycoproteins was tested by the immunisation of crossbred male calves at a dose of 200 microg/animal of each glycoprotein, three divided dosages. Following challenge infestations by the stages of two species of ticks, a significant rejection of (73.6%; P < 0.01) of larvae and adults (75.0%; P < 0.01) of H. a. anatolicum and adults (89.8%; P < 0.01) of B. microplus were noted. The direct effect of immunisation on the stages of the challenged ticks (%DT) was calculated as 69%, 52% against larvae and adults of H. a. anatolicum and 60% against adults of B. microplus. A direct corelation between protection and anti-glycoprotein antibody titre was detected. The effect of the isolated glycoproteins on limiting the tick infestation is discussed in detail.  相似文献

The objective of this study was to evaluate the inheritance mode of resistance to flumethrin in the Mexican Aldama Boophilus microplus strain. Two Mexican strains were used, the Chiapas susceptible (SS), and the Aldama flumethrin-resistant from Tamaulipas. Six steers weighing ca. 250 kg were randomly assigned for each of six crosses: the susceptible (SS), resistant (RR), and the F1 (RS, SR) reciprocal crosses and F2 (RS x RS and SR x SR). The reciprocal crosses were made to evaluate maternal and sex linkage effects. Bioassays tested resistant and susceptible larvae along with their hybrid F1 and F2 progeny against a series of concentrations of flumethrin (0, 0.0075, 0.00150, 0.00300, 0.00600 and 0.01200 microg/g). To test the single-gene hypothesis of resistance, a nonparametric line-cross test proposed by Collins was used. The bioassay data were subjected to probit analysis and the resistance factor and effective dominance obtained. Results of this study indicated that inheritance for flumethrin resistance in the Aldama strain was autosomal and controlled for more than one gene. The F1 and F2 larvae had similar lower resistant factor (RF 2.8-4.5) while the resistant Aldama strain was 21-fold higher (RF 81.8) than the mean of the F1 and F2. The extent of flumethrin resistance in the Aldama B. microplus strain depended upon the concentration of the pesticide used. Resistance was almost dominant at the lowest dose while almost completely recessive at the highest dose. Maternal effects were shown for egg-mass. These results shown here indicate more than one gene basis of flumethrin resistance in B. microplus ticks are present. Therefore it is necessary to locate and understand the major loci for elucidate the mechanism of resistance and improve the ability to track and delay the evolution of resistance.  相似文献

The effect of a decoction of pulverized aloe leaves (Aloe marlothii) mixed with tap water and administered orally through a gastric tube was determined against ticks on cattle. The decoction was administered to six randomly selected calves seven times at 3-day intervals while the remaining calves received an equivalent amount of tap water only. Five hours after first treatment the calves were infested with Boophilus decoloratus larvae hatched from one gram of eggs. Resultant engorged female ticks were collected, counted and weighed daily. Fertility estimates were determined for 30 engorged female ticks collected from each calf in both the treated and control groups of calves. The treatment had no significant effect on total or daily numbers of engorged females collected per group, nor on their biomasses. Fertility estimates showed the treatment group to yield a marginally higher, yet insignificant, egg laying response (ELR) and reproductive estimate (RE).  相似文献

The purpose of this study was to describe an unreported entomopathogenic fungus that naturally infects the cattle tick Rhipicephalus (Boophilus) microplus (Acari: Ixodidae). Engorged female ticks, showed symptoms of fungal infection after controlled tick infestation of cattle. Infected ticks developed a distinctive dark colour, a pale mould grew over the cuticle and the ticks eventually died covered with fungal conidiophores. The responsible fungus was isolated and cultured on mycological medium and submitted to microscopic morphology, biochemical phenotyping and 18S rRNA ribotyping analyses, which identified it as aflatoxin-producing Aspergillus flavus. Spores from the cultured fungus were experimentally sprayed over healthy engorged female ticks, obtaining an 80% prevalence of experimental infection of healthy ticks and their egg masses, the larval progeny after incubation under laboratory conditions was also infected. These results demonstrate that A. flavus is the causative agent of the natural fungal disease of the cattle tick R. microplus described here.  相似文献

A crude antigenic preparation of Babesia bigemina was used to develop an ELISA for the detection of IgM antibodies. Optimal dilutions of the antigen, using positive and negative reference sera, were determined by checkerboard titrations. Negative sera from cattle imported from tick-free areas, serum samples collected from infected B. bigemina cattle were used to validate the test. The specificity was 94% and sensitivity of the Elisa 87.5%. Sera from 385 cattle deriving from areas free from tick-borne diseases, which were submitted to a preimmunization process, were screened by this technique. The Elisa detected seroconversion on the 14th day post-inoculation in animals either infested with Boophilus microplus ticks (infected with B. bigemina), or inoculated with B. bigemina infected blood. Antibody titers decreased after day 33; however, all animals remained positive until the end of the experiment (124 days). The ELISA described may prove to be an appropriate serological test for the detection of IgM antibodies against B. bigemina.  相似文献

The cattle tick, Boophilus microplus, is a major pest of cattle in Australia, Central and South America, and parts of Africa and Asia. Control of ticks with organophosphates (OPs) and carbamates, which target acetylcholinesterases (AChE), led to evolution of resistance to these pesticides. Alleles at the locus studied here, AChE2, from OP-susceptible female ticks from Australia and Mexico differed at 46 of 1689 nucleotide positions (20 putative amino acid differences) whereas alleles from three strains of OP-resistant ticks from Australia differed with the allele from the Australian susceptible ticks at six to 13 nucleotide positions (three to six putative amino acid differences). However, the role, if any, of these polymorphisms in the OP-resistance phenotype is unknown. Certainly none of the polymorphisms correspond to sites in AChE that are involved in catalysis or binding of acetylcholine in other organisms. Both of the AChE loci of B. microplus, AChE1 and AChE2, are apparently expressed in synganglia; AChE1 is also expressed in salivary glands and ovaries, in OP-susceptible and OP-resistant ticks. This seems to contradict studies of enzyme kinetics, which indicated that only one form of AChE was present in the synganglia, the site of the action of OPs, in this species of tick.  相似文献

We used an expressed sequence tag approach to initiate a study of the genome of the southern cattle tick, Boophilus microplus. A normalized cDNA library was synthesized from pooled RNA purified from tick larvae which had been subjected to different treatments, including acaricide exposure, heat shock, cold shock, host odor, and infection with Babesia bovis. For the acaricide exposure experiments, we used several strains of ticks, which varied in their levels of susceptibility to pyrethroid, organophosphate and amitraz. We also included RNA purified from samples of eggs, nymphs and adult ticks and dissected tick organs. Plasmid DNA was prepared from 11,520 cDNA clones and both 5' and 3' sequencing performed on each clone. The sequence data was used to search public protein databases and a B. microplus gene index was constructed, consisting of 8270 unique sequences whose associated putative functional assignments, when available, can be viewed at the TIGR website (http://www.tigr.org/tdb/tgi). A number of novel sequences were identified which possessed significant sequence similarity to genes, which might be involved in resistance to acaricides.  相似文献

Friesian cattle were immunized with two inoculations of anti-tick Bm86 (Tick-GARD) vaccine and were challenged 30 or 90 d later with Boophilus annulatus larvae derived from 1.2 g of eggs. No nymphs or adult ticks were found on the immunized cattle during four weeks after challenge. Repeated infestations (2 to 4) with larvae on three other calves during a period of 160 and 390 d after the immunization did not result in development of nymphal and adult stages. In control, non-immunized cattle infested with corresponding batches of larvae 1380 to 4653 replete adult female ticks were collected. Larvae issued from Babesia bovis-infected female ticks transmitted the infection to Bm86-immunized cattle, but the progeny of B. bigemina-infected females did not. Since B. bigemina is transmitted exclusively by nymphal stages of Bo. annulatus these results support the observation that immunity induced by Bm86 affects the larval stage of this tick.  相似文献

A prevalence study was conducted to survey tick larvae populations in Puerto Rico (PR), compare the number of infested sites with Rhipicephalus (Boophilus) microplus larvae between the wet and dry season, and assess the associations of ecologic factors on the presence of R. microplus larvae. Ninety-six sites were selected using a GIS-based sampling method. Each site was sampled twice; the first sampling was performed during the dry season (March 4-18, 2007) and the second sampling during the wet season (August 13-26, 2007). Sites were sampled using a tick drag with a 1-m(2) white flannel cloth along a 50-m straight course. Only 2 tick species were identified. In the dry season, 15 sites (0.16, 95?% CI?=?0.09-0.24) were identified with R. microplus larvae (n?=?606) and 9 sites (0.09, 95?% CI?=?0.04-0.17) with Dermacentor (Anocentor) nitens larvae (n?=?779), whereas in the wet season 5 sites (0.05, 95?% CI?=?0.02-0.12) were identified with R. microplus (n?=?94), and 5 sites (0.05?%, 95?% CI = 0.02-0.12) with D. nitens (n?=?275). Difference in the number of infested sites with R. microplus was significant (P =?0.031) between the 2 seasons. Factors associated with the presence of R. microplus larvae in PR were wind speed of >4.0?km/h (OR?=?0.07, 95?% CI?=?0.01-0.63), more than 25?% bushes and shrubs on the site (OR?=?11, 95?% CI?=?1.6-71), and presence of cattle on the site (OR?=?26, 95?% CI?=?3.4-188).  相似文献

The histology of early feeding lesions of the cattle tick B. microplus has been studied using 32P labelled larvae to standardize the duration of attachment. Critical studies were made on 3-h lesions in six separate experiments on different groups of British breed animals. Each group consisted of three animals--one previously unexposed to ticks, one of high resistance and one of low resistance. The degree of mast cell disruption, eosinophil concentration and degranulation, and the extent of epidermal vesiculation were all significantly greater at the site of attachment on highly resistant hosts. In previously unexposed animals there was no mobilization of eosinophils nor mast cell breakdown and no epidermal vesiculation. Possible immune mechanisms producing mast cell disruption and the infiltration and concentration of eosinophils are suggested, and the effect of eosinophil degranulation on larval attachment and feeding is discussed.  相似文献

Boophilus ticks (n = 25,987) were collected from cattle at 30 communal dip tanks and five commercial farms in the Soutpansberg region, Limpopo Province, South Africa, between May 1999 and December 2001. Only 6.6% were Boophilus decoloratus, while 93.4% were Boophilus microplus, a tick that had not been reported from this area previously. B. microplus was the most common Boophilus tick on communal dip tank cattle, while B. decoloratus was more commonly found on commercial farms. Where the two species occurred together, B. microplus tended to displace B. decoloratus. The displacement was almost complete at the communal dip tanks, while on the commercial farms the population change was still in progress at the end of the survey. The present study demonstrated that a postulated reproductive interference was insufficient in preventing B. microplus from spreading when the climatic conditions were favourable to this species, as the displacement in most of the areas appeared to be rapid and complete.  相似文献
