Bioavailable N and P release rates by juveniles and adults of three Daphnia taxa (D. hyalina, D. galeata and its interspecific hybrids D. hyalina × galeata) were measured to assess the effect of weight and interspecific differences on these rates in Daphnia. Immobilized Scenedesmus obliquus cells were used to estimate the release rates. The specific release rate of N varied between 5.19–5.71 g N mg C-1 h-1 for juveniles and 3.00–3.42 g N mg C-1 h-1 for adults. P excretion rate ranged between 1.93–2.37 g P mg C-1 h-1 for juveniles and 1.00–1.24 g P mg C-1 h-1 for adults. Our results show that the taxonomic affiliation of Daphnia individuals did not affect their N and P release rates.  相似文献

This paper provides an approach for optimizing the cell density (Xc) and dilution rate (D) in a chemostat for a Pichia pastoris continuous fermentation for the extracellular production of a recombinant protein, interferon (INF-). The objective was to maximize the volumetric productivity (Q, mg INF- l–1 h–1), which was accomplished using response surface methodology (RSM) to model the response of Q as a function of Xc and D within the ranges 150 Xc 450 g cells (wet weight) l–1 and 0.1 mD0.9 m (m=0.0678 h–1, the maximum specific growth rate obtained from a fed-batch phase controlled with a methanol sensor). The methanol and medium feed rates that resulted in the desired Xc and D were determined based on the mass balance. From the RSM model, the optimal Xc and D were 328.9 g l–1 and 0.0333 h–1 for a maximum Q of 2.73 mg l–1 h–1. The model of specific production rate (, mg INF- g–1 cells h–1) was also established and showed the optimal Xc=287.7 g l–1 and D=0.0361 h–1 for the maximum (predicted to be 8.92×10–3 mg–1 g–1 h–1). The methanol specific consumption rate (, g methanol g–1 cells h–1) was calculated and shown to be independent of the cell density. The relationship between and (specific growth rate) was the same as that discovered from fed-batch fermentations of the same strain. The approach developed in this study is expected to be applicable to the optimization of continuous fermentations by other microorganisms.  相似文献

Summary From acetylene reduction assays over a 10-month period starting in April 1979, nodule activities averaged 18.78 (se 4.67) moles C2H4 g nodule dw–1 h–1 forAlnus rubra and 59.95 (se 12.14) moles C2H4 g nodule dw–1 h–1 forCytisus scorparius. Plant rates were 1.91 (se. 47) moles C2H4 plant–1 h–1 forA. rubra and 0.55 (se. 17) moles C2H4 plant–1 h–1 forC. Scoparius. Plant activity and total leaf N were strongly correlated with the dw of other plant parts, but nodule activity and percent leaf N were not. Plant and nodule activities were not associated with temperature, moisture stress, precipitation events or percent light for either species over the growing season nor for 54A. rubra sampled in mid-season 1979 on one replication. After 5 to 6 growing seasons, 14A. rubra on the same site ranged from 30 to 332 cm in height and showed strong correlation between nodule dw, leaf dw, plant size and total leaf N. Results from this study and others indicate logistic equations may be modified to predict the effect of adding a N2 fixing plant to a population of non N2 fixing trees.  相似文献

Summary The kinetics ofBordetella pertussis growth was studied in a glutamate-limited continuous culture. Growth kinetics corresponded to Monod's model. The saturation constant and maximum specific growth rate were estimated as well as the energetic parameters, theoretical yield of cells and maintenance coefficient. Release of pertussis toxin (PT) and lipopolysaccharide (LPS) were growth-associated. In addition, they showed a linear relationship between them. Denitrification processes were measured by the acetylene-blockage technique under changing flood conditions along the aquatic/terrestrial transition zone on the Amazon floodplain at Lago Camaleão, near Manaus, Brazil. In flooded sediments, denitrification was recorded after the amendment with NO 3 (100 mol liter–1) throughout the whole study period from August 1992 to February 1993. It ranged from 192.3 to 640.7 mol N m–2 h–1 in the 0- to 5-cm sediment layer. Without substrate amendment, denitrification was detected only during low water in November and December 1992, when it occurred at a rate of up to 12.2 mol N m–2 h–1 Higher rates of denitrification at an average rate of 73.3 mol N m–2 h–1 were measured in sediments from the shallow lake basin that were exposed to air at low water. N2O evolution was never detected in flooded sediments, but in exposed sediments, it was detected at an average rate of 28.3 mol N m–2 h–1 during the low-water period. The results indicate that under natural conditions there is denitrification and hence a loss in nitrogen from the Amazon floodplain to the atmosphere. Rates of denitrification in flooded sediments were one to two orders of magnitude smaller than in temperate regions. However, the nitrogen removal of exposed sediments exceeded that of undisturbed wetland soils of temperate regions, indicating a considerable impact of the flood pulse on the gaseous turnover of nitrogen in the Amazon floodplain.  相似文献

Thylakoids isolated from cells of the red alga Porphyridium cruentum exhibit an increased PS I activity on a chlorophyll basis with increasing growth irradiance, even though the stoichiometry of Photosystems I and II in such cells shows little change (Cunningham et al. (1989) Plant Physiol 91: 1179–1187). PS I activity was 26% greater in thylakoids of cells acclimated at 280 mol photons · m–2 · s–1 (VHL) than in cells acclimated at 10 mol photons · m–2 · s–1 (LL), indicating a change in the light absorbance capacity of PS I. Upon isolating PS I holocomplexes from VHL cells it was found that they contained 132±9 Chl/P700 while those obtained from LL cells had 165±4 Chl/P700. Examination of the polypeptide composition of PS I holocomplexes on SDS-PAGE showed a notable decrease of three polypeptides (19.5, 21.0 and 22 kDa) in VHL-complexes relative to LL-complexes. These polypeptides belong to a novel LHC I complex, recently discovered in red algae (Wolfe et al. (1994a) Nature 367: 566–568), that lacks Chl b and includes at least six different polypeptides. We suggest that the decrease in PS I Chl antenna size observed with increasing irradiance is attributable to changes occurring in the LHC I-antenna complex. Evidence for a Chl-binding antenna complex associated with PS II core complexes is lacking at this point. LHC II-type polypeptides were not observed in functionally active PS II preparations (Wolfe et al. Two strains of cultured tobacco cells (Nicotiana tabacum L. cv. Wisconsin 38) differing in their requirement for exogenous cytokinins (cytokinin-dependent and cytokinin-autonomous) were immobilized on polyphenylenoxide (Sorfix) activated with glutaraldehyde. Columns packed with immobilized cells were continually eluted with diluted Murashige and Skoog's medium lacking or supplemented with synthetic cytokinin (6-benzylaminopurine; BA). Purified samples of column eluates were fractionated by HPLC, andtrans-zeatin (t-Z) andtrans-zeatin riboside (t-ZR) content was estimated by enzyme immunoassay. Both cytokinin-autonomous and cytokinin-dependent tobacco cells produced and excretedt-Z and its riboside, and there were significant quantitative differences between the strains. The steady-state excretion rate oft-Z was 19.8 ng · g–1 dw · h–1 and 4 ng · g–1 dw · h–1, respectively, and that oft-ZR 4 ng · g–1 dw · h–1 and 1 ng · g–1 dw · h–1, respectively. Exposure of cytokinin-dependent cells to BA after 72 h of starving for this synthetic cytokinin caused temporary increase in excretion of both zeatin and its riboside. After the application of 5 M BA for 24 h, the excretion rate oft-ZR reached 5 ng · g–1 dw · h–1 (5-fold increase), and that oft-Z achieved 12 ng · g–1 dw · h–1 (3-fold increase). The elevation oft-Z excretion was delayed about 13 h compared witht-ZR excretion, which started increasing almost immediately after BA application. Two flight parameters (take-off and duration) and respiration level were measured, in two years in summer and early autumn, in dormant Coccinella septempunctata L. (Coleoptera: Coccinellidae) collected while hidden in grass tussocks in hibernation sites (HID) and in beetles collected on plants (PLA). The duration of tethered flight of HID beetles measured in the laboratory in late August and September 1995 (range of geometric means 190–440 s) was slightly longer than the flight of PLA beetles (80–310 s), both being much longer than trivial flight recorded in beetles foraging for prey during the breeding season (35 s). In general, the flight performance had a tendency to increase in September and to decrease in October.The oxygen consumption in HID beetles increased throughout September 1994 from 430 to 780 l g–1 h–1 and throughout October 1995 from 710 to 1060 l g–1 h–1. This increase is ascribed to a concomitant decrease in diapause intensity. A similar increase was observed also in PLA beetles in 1994 and oxygen consumption was always higher than in HID beetles, most probably due to feeding and digestion in PLA beetles.Laboratory feeding of HID beetles on aphids induced maturation of ovaries and increased oxygen uptake (from 680 to 1110 l g–1 h–1). Feeding on honey and pollen left their oxygen uptake unchanged. Effect of feeding on the flight parameters was mostly not significant. In agreement with its less suitable body shape and usually less distant dormancy sites, C. septempunctata was found a less apt flier than long-distance migrating coccinellid species.  相似文献

Feeding rates of Daphnia pulex fed a range of levels of the alga Chlamydomonas reinhardi of 15 °C are strongly density-dependent. At lower densities, Daphnia (30 1–1) fed at higher rates than crowded (270 1–1) Daphnia which manifest a relatively depressed saturation feeding response. At 30 individuals/liter, Daphnia consumed 8.5 – 15.7 × 104 cells d–1h–1 (on a volume basis, 12.1 – 22.2 × 106 m3), at 270 L–1 3.7 – 3.9 × 104 (5.2 – 5.5 = 106 m3 cells d–1h–1 when feeding on algae at 80 000 cells ml–1 (11.3 × 106 m3 ml–1). The feeding rate data best fit an Ivlev feeding function. An autoallelopath might be causing the repression. Water preconditioned with crowded Daphnia completely repressed feeding in uncrowded Daphnia after six hours.  相似文献

Degradation of polyaromatic hydrocarbons by Burkholderia cepacia 2A-12   总被引:1,自引:1,他引:0  
A new strain of bacterium degrading polyaromatic hydrocarbons (PAHs), Burkholderia cepacia 2A-12, was isolated from oil-contaminated soil. Of three PAHs, the isolated strain could utilize naphthalene (Nap) and phenanthrene (Phe) as a sole carbon source but not pyrene (Pyr). However, the strain could degrade Pyr when a cosubstrate such as yeast extract (YE) was supplemented. The PAH degradation rate of the strain was enhanced by the addition of other organic materials such as YE, peptone, glucose, and sucrose. YE was a particularly effective additive in stimulating cell growth as well as PAH degradation. When 1 g YE l–1, an optimum concentration, was supplemented into the basal salt medium (BSM) with 215 mg Phe l–1, the specific growth rate (0.30 h–1) and Phe-degrading rate (29.6 mol l–1 h–1) were enhanced approximately ten and three times more than those obtained in the BSM with 215 mg Phe l–1, respectively. Both cell growth and PAH degradation rates were increased with increasing Phe and Pyr concentrations, and B. cepacia 2A-12 had a tolerance against Phe and Pyr toxicity at the high concentration of 730–760 mg l–1. Through kinetic analysis, the maximum specific growth rate ( max) and PAH degrading rate ( max) for Phe were obtained as 0.39 h–1 and 300 mol l–1 h–1, respectively. Also, max and max for Pyr were 0.27 h–1 and 52 mol l–1 h–1, respectively. B. cepacia 2A-12 could simultaneously degrade crude oil as well as PAHs, indicating that this bacterium is very useful for the removal of oils and PAHs contaminants.  相似文献

Summary Measurements have been made of the rate of turnover of tritiated water in the eggs of the plaice,Pleuronectes platessa and of the permeability of the vitelline membrane. The rate constant of water exchange in body cavity eggs is 1.0 h–1, corresponding to a permeability of 0.086 3/2/sec. When shed into sea water the rate constant of exchange declines to 0.2 h–1 after 1 hour and to 0.02 h–1 after one day corresponding to a permeability of 0.0017 [3/2/sec. As the embryo develops the rate of turnover increases again. The buoyancy of the egg is dependent on its low salt content which counteracts the weight of the protein. During development the water loss by osmosis reduces the egg almost to neutral buoyancy.  相似文献

Concentrations of heavy metals commonly known to impact the environment and other related trace elements were quantified in the water bodies of nine Ethiopian rift-valley lakes and six rivers (their inflows) as well as in effluents from two factories. In about half of the samples the concentrations of As was 10–700 g l–1 and Se, ranged from 10 to 28 g l–1, were much higher than the maximum permissible level (MPL) according to international standards for drinking water. Mercury (Hg) was detected in four lakes and one river with high values ranging from 2 to 165 g l–1. Concentrations of Mo in three soda lakes were as high as 544–2590 g l–1. Iron (Fe) was found in high concentrations (567–4969 g l–1) in three lakes, which are known to be discolored from inorganic colloids. Levels of Cd, Pb, and Cr ranged between 5 and 9, 12 and 20 and 104 and 121 g l–1, respectively. The rest of the heavy metals analyzed, Ba, Cu, Mn, Ni and Zn, were either not detected in the samples or were found in much lower concentrations than the MPL for drinking water. Effluent from a tannery contained about 15, 141, 523, and 19 g l–1 of As, Cr, Fe, and Se, respectively, and effluent from a textile factory contained high concentrations of As (10.6), Hg (3.8) and Se (20) g l–1. Compared to more industrialized regions and other African lakes the concentrations of heavy metals in Ethiopian rift-valley lakes (with the exception of the soda lakes) and their inflows were low.  相似文献

Three-year-old spruce (Picea abies) saplings were planted and cultivated for 2 years in pots with 3 1 substrate, consisting of a homogenized mixture of sand, peat and forest soil with a high organic content (volume ratio 11.52). This substrate was amended with 10–180 mol Cd [kg soil dry weight (DW)]–1, 50–7500 mol Zn (kg soil DW)–1 (determined with 1 M ammonium acetate extracts) or combinations of both elements. Annual xylem growth rings in stems of plants treated with 50 mol Cd (kg soil DW)–1 or 7500 mol Zn (kg soil DW)–1 were significantly narrower than in control plants. Growth reductions were more pronounced in the second year of the experiment. The contents of Cd and Zn in stem wood and needles were positively correlated with the substrate concentrations. The Mg contents of the spruce needles were inversely correlated with soil concentrations of Cd and Zn. Root development was impeded at moderate concentrations of Cd (50 mol kg–1) or Zn (1000 mol kg–1) in the substrate. The adverse effects of potentially toxic trace elements, like Cd or Zn, on xylem growth of spruce plants are discussed with regard to possible growth reductions in forest trees under field conditions.  相似文献

We intended to evaluate the relative contribution of primary production versus allochthonous carbon in the production of bacterial biomass in a mesotrophic estuary. Different spatial and temporal ranges were observed in the values of bacterioplankton biomass (31–273 g C l–1) and production (0.1–16.0 g C l–1 h–1, 1.5–36.8 mg C m–2 h–1) as well as in phytoplankton abundance (50–1700 g C l–1) and primary production (0.1–512.9 g C l–1 h–1, 1.5–512.9 mg C m–2 h–1). Bacterial specific growth rate (0.10–1.68 d–1) during the year did not fluctuate as much as phytoplankton specific growth rate (0.02–0.74 d–1). Along the salinity gradient and towards the inner estuary, bacterio- and phytoplankton biomass and production increased steadily both in the warm and cold seasons. The maximum geographical increase observed in these variables was 12 times more for the bacterial community and 8 times more for the phytoplankton community. The warm to cold season ratios of the biological variables varied geographically and according to these variables. The increase at the warm season achieved its maximum in the biomass production, particularly in the marine zone and at high tide (20 and 112 times higher in bacterial and phytoplankton production, respectively). The seasonal variation in specific growth rate was most noticeable in phytoplankton, with seasonal ratios of 3–26. The bacterial community of the marine zone responded positively – generating seasonal ratios of 1–13 in bacterial specific growth rate – to the strong warm season increment in phytoplankton growth rate in this zone. In the brackish water zone where even during the warm season allochthonous carbon accounted for 41% (on average) of the bacterial carbon demand, the seasonal ratio of bacterial specific growth rate varied from about 1 to 2. During the warm season, an average of 21% of the primary production was potentially sufficient to support the whole bacterial production. During the cold months, however, the total primary production would be either required or even insufficient to support bacterial production. The estuary turned then into a mostly heterotrophic system. However, the calculated annual production of biomass by bacterio- and phytoplankton in the whole ecosystem showed that auto- and heterotrophic production was balanced in this estuary.  相似文献

Nitrate induction in spruce: an approach using compartmental analysis   总被引:6,自引:0,他引:6  
Using 13NO 3 -efflux analysis, the induction of nitrate uptake by externally supplied nitrate was monitored in roots of intact Picea glauca (Moench) Voss. seedlings over a 5-d period. In agreement with our earlier studies, efflux analysis revealed three compartments, which have been identified as surface adsorption, apparent free space, and cytoplasm. While induction of nitrate uptake was pronounced, NO 3 fluxes in induced plants were decidedly lower and the induction response was slower than in other species. Influx rose from 0.1 mol·g–1·h–1 (measured at 100 M [NO 3 o) in uninduced plants to a maximum of 0.5 mol·g–1h–1 after 3 d of exposure to 100 M [NO 3 o and declined to 0.3–0.4 mol·g–1h–1 at the end of the 5-d period. Efflux remained relatively constant around 0.02-0.04 mol·g–1h–1, but its percentage with respect to influx declined from initially high values (around 30%) to steady-state values of 4–7%. Cytoplasmic [NO 3 ] ranged from the low micromolar in uninduced plants to a maximum of 2 mM in plants fully induced at 100 M [NO 3 ]o. In-vivo root nitrate reductase activity (NRA) was measured over the same time period, and was found to follow a similar pattern of induction as influx. The maximum response in NRA slightly preceded that of influx. It increased from 25 nmol·g–1·h–1 without prior exposure to NO 3 to peak values around 150 nmol· g–1h–1 after 2 d of exposure to 100 M [NO 3 ]o. Subsequently, NRA declined by about 50%. In order to maintain axenic seedstock cultures axenically of thecommercially important red seaweed, Porphyra yezoensis, aprocedure was developed for axenic isolation and culture of conchocelis andmonospores. For axenic isolation of the conchocelis, contaminated microalgaewere most effectively removed by filtering contaminated samples through a100-m mesh after sonication. Removal of bacteria and otheralgaewas accomplished using a mixture of 5 agents (0.02% chitosan, 100 gml–1 GeO2, 10 gml–1 ampicillin, 40 gml–1 kanamycin and 200 gml–1 streptomycin). Axenic single colonies wereisolatedfrom a semi-solid medium prepared from 1% transfer gel. After collectingmonospores from the 40–50% density layer on a percoll-gradient, removalofbacteria and fungi from the monospores was accomplished using a mixture of 5antibiotics (3.5 g ml–1 nystatin, 2 mgml–1 ampicillin, 400 gml–1 kanamycin, 50 gml–1 neomycin and 800 gml–1 streptomycin). Severely yellowed ten-year-old spruce trees growing in the Vosges Mountains on an acidic soil were fertilised with Magnesium lime during the spring of 1990. The effects of this treatment were assessed 18 months later. A very significant improvement of the mineral status of the trees was detected, with increasing Mg contents in the needles, and as a consequence, reduced yellowing and improved chlorophyll content. Only slight differences with control trees were observed for height increase. Effects of this improved nutrition on photosynthesis were tested measuring net CO2 assimilation rates and chlorophyll a fluorescence. Light-saturated net assimilation rates of current-year needles were high, reaching 5.3 mol m–2 s–1 on a total needle area basis. The improvement in chlorophyll and Mg content had no significant effect on net assimilation rates or on any parameter describing photochemical functions of both current-and previous-year needles. Despite the strong inter-individual variability in needle chlorophyll and Mg contents (ranging from 0.2 to 0.8 mg g–1 fresh weight, and 0.05 to 0.5 mg g-1 dry weight respectively), photochemical efficiency of PS II under limiting irradiance only decreased significantly on older needles displaying Mg contents below 0.1 mg g–1. Thymidine and leucine incorporation into macromolecules of soil bacteria extracted by homogenization-centrifugation were measured after size-fractionation of the bacterial suspension through different sized filters (1.0, 0.8, 0.6, 0.4 m). The specific thymidine incorporation rate was highest for the unfiltered and 1.0 m filtered suspensions (approximately 10 × 10–21 mol thymidine bacteria–1 h–1), but decreased to 1.39 × 10–21 mol bacteria–1 h–1 for bacteria passing the 0.4 m filter. The proportion of culturable bacteria (percent colony forming units/acridine orange direct counts) also decreased with bacterial cell size from 5.0% for the unfiltered bacterial suspension to 0.8% in the 0.4 µm filtrate. A strong linear correlation (r 2 = 0.995) was found between the specific thymidine incorporation rate and the proportion of culturable bacteria. Leucine incorporation gave similar results to the thymidine incorporation. No effects of cell size on the degree of isotope dilution or unspecific labeling of other macromolecules were found either for the thymidine or the leucine incorporation technique. These data indicate that small bacteria, although more numerous than larger ones, not only constitute a smaller proportion of the soil bacterial biomass than larger bacteria, but also contribute to a lesser degree to carbon transformations in soil.  相似文献

In Taxus cuspidata callus, vanadyl sulfate (10 mg l–1) induced a high (146 g g–1 dry wt) production of 10-deacetylbaccatin III in comparison to 7 g g–1 dry wt of the control. The content of paclitaxel in this species increased from 16 g g–1 to 74 g g–1 dry wt when 20 mg phenylalanine l–1 was used. In T. media, p-aminobenzoic acid induced the highest content of 10-deacetylbaccatin III (481 g g–1 dry wt) versus 181 g g–1 in the control. Paclitaxel increased from 89 to 139 g g–1 dry wt after adding chitosan (20 mg l–1) to the cultures.  相似文献
