Summary The production of gamma-linolenic acid (GLA) by Mucor circinelloides CBS 203.28 and M. rouxii CBS 416.77 in fed-batch cultures operated in pH-stat mode with acetic acid as carbon substrate and titrant compared favourably with the performance of M. circinelloides in batch culture on glucose. On acetic acid M. circinelloides accumulated up to 39.8 mg GLA/g biomass, with a crude oil content of 28% containing 91% neutral lipids. The GLA content of the neutral lipid fraction was 15.6%.  相似文献

Lipid analyses were performed on 28 strains of various species of the genus Mucor. In shake flasks with glucose as carbon source, the gamma-linolenic acid (GLA) content in the neutral lipid (NL) fraction of some Mucor species was up to 38 mg GLA/g dry biomass. Some Mucor species produced more than 20% (w/w) stearic acid (18:0) and more than 60% of their NL content as symmetrical triacylglycerols (SUS-TAGs) which corresponded to those of cocoa butter. Three Mucor species were evaluated in terms of the production of SUS-TAGs and GLA in pH-stat, fed-batch cultures in an air-lift fermenter with acetic acid as titrant and carbon source. Mucor circinelloides f. circinelloides CBS 108.16 accumulated 27% 18:0 in the NL fraction, which constituted approximately 40% of the dry biomass. The effect of different initial acetic acid concentrations on the growth of and lipid and gamma-linolenic acid (GLA) production byMucor circinelloides CBS 203.28 was determined in a 14 litre stirred tank reactor operated in a fedbatch, pH-stat mode with acetic acid as carbon source and pH titrant. Increased acetic acid concentrations in the culture resulted in a significant increase in the crude oil content of the biomass. By contrast, all the other parameters such as the biomass concentration, GLA and oil yield on acetic acid, the GLA content of the biomass and oil, the growth rate and volumetric rate of GLA production decreased with an increase in acetic acid concentration. The best results were obtained with acetic acid at 2 g/1, which gave 39.8 mg GLA/g biomass and 15.6% GLA in the neutral lipid fraction, amounting to 340 mg GLA/1 culture. A decrease in the glyco- and phospho-lipid fractions during the cultivation coincided with an increase in the neutral lipid fraction. The GLA content of the biomass remained within rather narrow limits of 3.5% to 4% of the biomass, irrespective of the oil content of the biomass. The fatty acid profile was not greatly affected by the acetic acid concentration. The hyphae of the fungus were characterized by the accumulation of large intracellular oil droplets and some septa delimited the hyphae.  相似文献

The Fischer-Tropsch reaction water, which contains C2 to C5 monocarboxylic acids, generated as a co-product of the Sasol industrial oil-from-coal process, constitutes a potential cheap carbon substrate for the production of gamma-linolenic acid (GLA) by selced Mucor species. Three strains of Mucor were each grown in an air-lift reactor operated in a fed-batch, pH-stat mode under N-limitation with a mixture of C2 to C5 monocarboxylic acids as both pH titrant and carbon source. The production of GLA from this substrate was evaluated. Growth typically resulted in the rapid assimilation of acetic, n-butyric and n-valeric acids. Although propionic, iso-butyric and iso-valeric acids were assimilated to varying degrees, these acids accumulated in the culture. Mucor circinelloides CBS 203.28 gave the best results in that it assimilated 36% to 100% of each acid, had a biomass yield coefficient of 0.3 (calculated on acids utilized), and contained 28% crude oil, 84% of which comprised neutral lipids with a GLA content of 14.4%, giving 33 mg GLA/g biomass. GLA accumulation coincided with a decrease in the stearic-acid content of the neutral-lipid fraction. The results were comparable with previous results obtained with acetic acid and glucose as sole carbon sources, demonstrating the feasibility of producing GLA from the above mixture of organic acids.  相似文献

Summary The production of -linolenic acid (GLA) and lipid was studied in Mucor rouxii CBS 416.77. In a fed-batch culture productivities of 39.4 mg/l per hour for GLA and 99 mg/l per hour for the total amount of lipid were determined at 18 h of cultivation. At this point the highest value of GLA in lipid (39.7%, w/w) was also reached. Production of GLA was also studied in a series of continuous cultures. It was observed that, in addition to growth rate, the nitrogen concentration of the input medium was of great importance for high productivities. The highest productivity values for GLA (37 mg/l per hour) and for lipid (95 mg/l per hour) were reached at a dilution rate of 0.10 h-1 with a concentration of 4.5g/l NH4Cl in the input medium.  相似文献

The oleaginous fungus, Mucor circinelloides, is one of few fungi that produce high amounts of γ-linolenic acid (GLA); however, it usually only produces <25% lipid. Nevertheless, a new strain (WJ11) isolated in this laboratory can produce lipid up to 36% (w/w) cell dry weight (CDW). We have investigated the potential mechanism of high lipid accumulation in M. circinelloides WJ11 by comparative biochemical analysis with a low lipid-producing strain, M. circinelloides CBS 277.49, which accumulates less than 15% (w/w) lipid. M. circinelloides WJ11 produced more cell mass than that of strain CBS 277.49, although with slower glucose consumption. In the lipid accumulation phase, activities of glucose-6-phosphate dehydrogenase and 6-phosphogluconate dehydrogenase in strain WJ11 were greater than in CBS 277.49 by 46% and 17%, respectively, and therefore may provide more NADPH for fatty acid biosynthesis. The activities of NAD+:isocitrate dehydrogenase and NADP+:isocitrate dehydrogenase, however, were 43% and 54%, respectively, lower in WJ11 than in CBS 277.49 and may retard the tricarboxylic acid cycle and thereby provide more substrate for ATP:citrate lyase (ACL) to produce acetyl-CoA. Also, the activities of ACL and fatty acid synthase in the high lipid-producing strain, WJ11, were 25% and 56%, respectively, greater than in strain CBS 277.49. These enzymes may therefore cooperatively regulate the fatty acid biosynthesis in these two strains.  相似文献

The fungus gnat Bradysia impatiens (Johannsen) is a worldwide pest inhabiting organic-rich environments, including mushroom-cultivation substrates, which leads to massive production losses of edible mushrooms. To promote a non-insecticidal pest control strategy, we evaluated adult behavioural response towards two saprophytic fungal strains Aspergillus flavus Link and Mucor circinelloides Tiegh., isolated from B. impatiens-inhabiting substrates and six edible mushrooms species. Our results indicated that (a) B. impatiens was most attracted to the oyster mushroom Pleurotus ostreatus among all six edible mushrooms; (b) males and females demonstrated a significant attraction for M. circinelloides over A. flavus and P. ostreatus; (c) adults demonstrated dynamic attraction tendencies that varied with the culture periods of A. flavus and M. circinelloides, wherein females represented most (mean = 78.6%) of the attracted individuals; and (d) larvae that fed on mycelial cultures of M. circinelloides could complete their entire life cycles, but larvae could not survive on a non-fungal or A. flavus diet. These results demonstrate the potential for the development of M. circinelloides in a push-pull strategy to biocontrol this pest in edible mushroom cultivation.  相似文献

Fifteen strains of filamentous fungi from theCulture Collection of Fungi (Charles University, Prague) were tested for their lipid production, fatty acid composition with emphasis on accumulation of oligounsaturated fatty acids. All cultures contained palmitic (16:0), palmitoleic (16:1), stearic (18:0), oleic (18:1), linoleic (18:2) and γ-linolenic (18:3) acid (GLA). The mycelium ofCunninghamella elegans, Rhizopus arrhizus, Mortierella parvispora, M. elongata andM. alpina contained arachidonic acid (ARA) in the range of 2.3–33.5% of the total fatty acids. The strains used in our experiment were capable to accumulate a relatively high amount of intracellular lipid (9.6–20.1% in dry biomass). The highest content of GLA (22.3 mg/g) was found inMucor circinelloides. The strain ofM. alpina containing 47.1 mg/g of ARA could be considered as the best producer of ARA.  相似文献

Four Mucor strains were tested for their ability to grow on four cereal substrates and enriched them with gamma-linolenic acid (GLA) and β-carotene. M. circinelloides CCF-2617 as the best producer accumulated of both GLA and β-carotene in high amounts during utilization of rye bran/spent malt grains (3:1). The first growth phase was characterized by rapid GLA biosynthesis, while distinct β-carotene formation was found in the stationary fungal growth. Therefore various cultivation conditions were tested in order to optimize the yield of either GLA or β-carotene. The fungus grown on cereal substrate supplemented with glucose produced maximal 8.5 mg β-carotene and 12.1 g GLA in 1 kg fermented substrate, respectively. On the other hand, the highest amount of GLA in the fermented substrate (24.2 g/kg) was achieved when 30% of sunflower oil was employed to the substrate. Interestingly, β-carotene biosynthesis was completely inhibited when either whey or linseed oil were added to the substrate.  相似文献

White rot fungi Fomes lividus and Trametes versicolor, isolated from the Western Ghats region of Tamil Nadu, India, were used to treat pulp and paper industry effluents on a laboratory scale and in a pilot scale. On the laboratory scale a maximum decolourization of 63.9% was achieved by T. versicolor on the fourth day. Inorganic chloride at a concentration of 765 mg/l, which corresponded to 227% of that in the untreated effluent, was liberated by F. lividus on the 10th day. The chemical oxygen demand (COD) was also reduced to 1984 mg/l (59.3%) by each of the two fungi. On the pilot scale, a maximum decolourization of 68% was obtained with the 6-day incubation by T. versicolor, inorganic chloride 475 mg/l (103%) was liberated on the seventh day by T. versicolor, and the COD was reduced to 1984 mg/l corresponding to 59.32% by F. lividus. These results suggested that F. lividus seems to be another candidate efficient for dechlorination of wastewater.  相似文献

Ceramic industry wastewaters not only contain high suspended and total solids but also significant amounts of dissolved organics resulting in high BOD or COD loads. Suspended solids can be removed from the wastewater by chemical precipitation. However, dissolved BOD/COD compounds can only be removed by biological or chemical oxidation. Effluent wastewater from chemical sedimentation stage of EGE CERAMIC industry was characterized and subjected to biological treatment in a laboratory scale activated sludge unit. Experiments were conducted at different hydraulic and solids retention times. The best results were obtained with Šc=20 h of hydraulic and Šc=20 days of solids retention times (sludge age) resulting in effluent COD concentration of 40 mg/l from a feed wastewater of 720 mg/l COD content. The suspended solids content of the activated sludge effluent was approximately 52 mg/l.  相似文献

For better operational control of the completely mixed oxygen activated sludge process (CMOAS), a study concerning the kinetics, performance, and operational stability of the Ramanathan-Gaudy model was conducted. Short-term experiments were conducted at various dilution rates (1/9, 1/6, 1/3, 1/1.5, and 1/1.0 hr?1) by using two recycle solids concentration values (5000 and 10,000 mg/liter). The influent substrate was an actual industrial organic wastewater (soft drink waste) and its concentration was maintained at 1000 mg/liter COD. The hydraulic recycle ratio, α, was maintained at 0.30. It was found that for CMOAS system with constant recycle cell concentration, a “steady state” with respect to reactor biological solids and effluent COD at different dilution rates could be attained. No appreciable dilute-out of reactor biological solids and substrate was observed up to the dilution rate of 1 hr?1 for both systems of different XR (5000 and 10,000 mg/liter). For the system of XR = 5000 mg/liter, except the dilution rate of hr?1, the effluent filtrate COD was lower than 100 mg/liter, the aerator biological solids concentration was about 1550 mg/liter, and the COD removal efficiency was higher than 90% for all dilution rates. For the system of XR = 10,000 mg/liter, the effluent filtrate COD was lower than 71 mg/liter, the aerator biological solids concentration was about 2750 mg/liter, and the COD removal efficiency was higher than 90% throughout all the dilution rates selection in the present study. The value of the Sludge Volume Index (SVI) was the range of 37.0 to 58.5 and provided good settleability of sludge. The sludge yield was 0.53 for the system of XR = 5000 mg/liter and 0.57 for the system of XR = 10,000 mg/liter. The carbohydrate and the protein content of the cells were 10.1–21.6% and 35.6–50.6%, respectively. For predicting the reactor biological solid and effluent COD of the CMOAS system by using the Ramanathan-Gaudy model, two sets of values for the biological kinetic constants should be considered since it provided the best fit of predicted values of the observed values. In the present study, μm = 0.4 hr?1, ks = 92 mg/liter for 1/3 ? D ? 1, and μm = 0.05 hr?1, ks = 11.1 mg/liter for 1/9 ? D < 1/3 were used to calculate the predicted values of reactor biological solid and effluent filtrate COD.  相似文献

Summary An anaerobic hybrid reactor was used in the anaerobic treatment of an acidic petrochemical effluent. An organic loading rate of 20.04 kg COD/(m3d) at a HRT of 17 hours was obtained with a volatile fatty acid removal of 91%, and COD removal of 84%. A final reactor effluent containing 44 mg/l ammonia nitrogen and 12.3 mg/l PO4-P was produced.  相似文献

The upflow aerated biofilter with polyurethane foam cubes as the supporting medium was used for the investigation of nitrification studies on municipal sewage (secondary treated as well as untreated domestic sewage). In case of secondary treated sewage effluent, a synthetic composition of NH4 +-N and COD of each 50?mg/l was studied for a HRT variation of 24, 12, 8 and 6 hours. The ammonium removal efficiencies were found to be in the range of 98 to 100% with the steady-state effluent concentrations of NH4 +-N and NO2 ?-N in the range of 1–4 mg/l and 0.1–0.2?mg/l respectively. In case of domestic sewage system, nitrification studies along with suspended solids removal study was carried-out on untreated sewage for a HRT variation of 24, 12 and 6 hours. The ammonium removal efficiencies of 100% were observed for all the three HRT values at very high COD/NH4 +-N ratio of 15. The suspended solids removal efficiencies of 95 to 98% were observed with the average effluent suspended solids concentration of 5.9 to 15.9?mg/l. The experiments were conducted in non-backwash conditions of the biofilter. The study has revealed the best use of the upflow biofilter system for nitrification applications and suspended solids removal.  相似文献

γ-Linolenic acid (GLA; C18:3 Δ6,9,12) is a nutritionally important fatty acid (FA) playing a vital role in biological structures and cellular functions, which is not produced in oil seed crops. Many oil seed plants, however, produce significant quantities of linoleic acid, a FA that could be converted into GLA by the enzyme Δ6 desaturase, if it is present. As a first step to produce GLA in oil seed crops, we isolated a cDNA encoding the Δ6-FA desaturase from filamentous fungus Mucor circinelloides M29. Expression of this gene in transgenic tobacco resulted in the accumulation of GLA to the levels of 23.1% of the total FA. The results suggested that it is feasible to introduce the M. circinelloides Δ6 desaturase gene into conventional oil crop to produce a large amount of GLA for functional foods and pharmaceutical products. Fishmeal wastewater, a seafood processing waste, was utilized for production of lactic acid and fungal biomass by Rhizopus oryzae AS 3.254 with the addition of sugars. The 30 g/l exogenous glucose in fishmeal wastewater was superior to starch in view of productivities of lactic acid and fungal biomass, and COD reduction. Fishmeal wastewater can be a replacement for peptone which was the most suitable nitrogen source for lactic acid production among the tested organic or inorganic nitrogen sources. Exogenous NaCl (12 g/l) completely inhibited the production of lactic acid and fungal growth. In the medium of COD 5,000 mg/l fishmeal wastewater with the addition of 30 g/l glucose, the maximum productivity of lactic acid was 0.723 g/l h corresponding to productivity of fungal biomass 0.0925 g/l h, COD reduction 84.9% and total nitrogen removal 50.3% at a fermentation time of 30 h.  相似文献

In this work, the ability of Pleurotus spp.:P. sajor-caju; P. platypus and P. citrinopileatus to treat pulp and paper mill effluent on a laboratory and pilot scale were studied. On the laboratory scale treatment, P. sajor-caju decolorized the effluent by 66.7% on day 6 of incubation. Inorganic chloride liberated by P. sajor-caju was 230.9% (814.0 mg/dl) and the COD was reduced by 61.3% (1302.0 mg/dl) on day 10 of treatment. In the pilot scale treatment maximum decolorization was obtained by P. sajor-caju (60.1%) on day 6 of the incubation. Inorganic chloride content was increased by 524.0 mg/dl (113.0%) and the COD was reduced by 1442.0 mg/dl (57.2%) by P. sajor-caju on day 7 of incubation. These results revealed that the treatment of pulp and paper mill effluent by P. sajor-caju proved as better candidate for the purpose than P. platypus and P. citrinopileatus.  相似文献

Due to increasing demand for natural sources of both polyunsaturated fatty acids (PUFAs) and beta-carotene, 28 Zygomycetes fungal soil isolates were screened for their potential to synthesize these biologically active compounds. Although all fungi produced C18 PUFAs, only nine strains also formed beta-carotene. Although Actinomucor elegans CCF 3218 was the best producer of gamma-linolenic acid (GLA) (251 mg/L), Umbelopsis isabellina CCF 2412 was found to be the most valuable fungus because of the dual production of GLA (217 mg/L) and beta-carotene (40.7 mg/L). The calculated ratio of formed PUFAs provided new insight into activities of individual fatty acid desaturases involved in biosynthetic pathways for various types of PUFAs. The maximal activity of delta-9 desaturase was accompanied by high accumulation of storage lipids in fungal cells. On the other hand, maximal activity of delta-15 desaturase was found in strains synthesizing low amounts of oleic acid due to diminished delta-9 desaturase. Activities of delta-6 desaturase showed competition for fatty acids engaged in n3, n6, and n9 biosynthetic pathways. Such knowledge about fatty acid desaturase activities provides new challenges for the regulation of biotechnological production of PUFAs by Zygomycetes fungi.  相似文献

Summary A high-strength baker's yeast effluent was anaerobically treated using a hybrid digester under mesophilic conditions. The digester was subjected to a substrate COD concentration of 21 767 mg/I at three different HRTs. At HRTs of 3.0, 2.0 and 1.0 d, the digester reduced the substrate COD by 76, 61 and 33%, respectively. Although the best COD removal was obtained at an OLR of 7.30 kg COD/m3.d, the highest COD removal rate (6.51 kg COD/M3-d) was found at 10.65 kg COD/m3.d at an HRT of 2.0 d. The low methane yield and VFA accumulation found in the digester effluent, indicated inhibition on methanogenic level and this was considered to be the rate-limiting step during the anaerobic treatment process. The overall efficiency of the digester indicated that this digester design and support medium was suitable for the treatment of a high-strength, sulfate-rich baker's yeast effluent.  相似文献

The lipid accumulation, fatty acid composition and γ-linolenic acid (GLA) production by 28 strains belonging to Mucorales were investigated. The lipid content varied from 5 to 30% on dry biomass and the percentage of γ-linolenic acid in total intracellular lipid was in a range from 2.5 to 15.4% (w/w). The best yield of γ-linolenic acid (expressed as mg GLA per 1 g biomass) was found for Mucor mucedo CCF – 1384 (28.4) and Cunninghamella echinulata CCF – 103 (25.1).  相似文献
