Abstract The allocation of photosynthetically fixed carbon in the leaf blades and sheaths of Poa annua (a ruderal grass) and Poa x jemtlandica (a sub-arctic grass) was followed over a light-dark cycle. Labelling with 14Carbon and gas exchange measurements provide data for an eight-compartment model describing the partitioning of carbon between spatially and chemically separated pools and their rates of turnover. Soluble sugars and fructans were turned over rapidly in the leaf blades of both species. The flux of carbon through pools of storage carbohydrates was higher in the leaves of P. x jemtlandica than in P. annua. The exchange of carbon between pools was slower in the sheath than the blade. Carbohydrates stored in the sheath appeared to have no significant role in metabolism over the light-dark cycle studied here.  相似文献

In both reproductive and vegetative plants of Lolium temulentumL., the export of 14C-labelled assimilates from each healthyleaf on the main shoot to terminal meristem, stem, tillers,and roots was measured each time a new leaf was expanded, fora period of 5 to 6 weeks. Some labelled assimilates moved fromeach leaf on the main shoot to every meristem in the same shoot,as well as to the tops and roots of adjacent organically attachedtillers. The terminal meristem of the reproductive shoot, which includedthe developing inflorescence, received 70–80 per centof the carbon assimilated by the emerged portion of the growingleaf, 15–25 per cent of the carbon assimilated by thetwo youngest expanded leaves, and 5–10 per cent of thatfrom each of the older leaves. A similar pattern of carbon supplyto the terminal meristem was found in vegetative shoots, exceptthat older leaves on young vegetative shoots supplied even lessof their carbon to the terminal meristem. The general conclusionis that developing leaves at the tip of the shoot receive aboutthe same proportion of carbon from each leaf as does a developinginflorescence. Young expanded leaves provided most labelled assimilates forstem growth; during both reproductive and vegetative growth,expanded leaves increased their export of labelled carbon tostem, and exported less of their 14C to roots and sometimesto tillers. In these reproductive and vegetative shoots, grown in a constantexternal environment, the major changes in the pattern of distributionof labelled assimilates appeared to be the result of increasedmeristematic activity in stem internodes; the development ofan inflorescence had no obvious direct effect on the carboneconomy of shoots.  相似文献

We wished to determine the nature of differences in epidermalcell numbers and dimensions between leaves of different lengthin mutants of barley (Hordeum vulgare L. ‘Himalaya’).Three comparisons were made: leaf one (L1)vs. leaf four (L4);wild typevs. nine dwarf mutants and wild typevs. a slender mutant.L1 was shorter than L4, and for most lines this was associatedwith a change in epidermal cell number for the blade, and inboth cell number and length for the sheath. Compared to wildtype, the smaller leaves of dwarf plants generally had shorterand fewer cells in both blade and sheath. The blade of slenderplants was the same length (L1) or longer (L4) than wild type,while the sheath was longer than that of wild type for bothL1 and L4. Slender plants had longer but fewer cells than thewild type along the blade of L1, and shorter but more cellsfor the blade of L4. In the sheath, slender plants had longerand more (L1) or fewer (L4) cells than did the wild type. ForL1, variation in blade width amongst the barley lines was associatedwith a change in file width and file number. For L4, blade widthvaried only with file number, except for slender plants wherenarrow blades were associated with reduced file width. Hencethere was no consistent correlation between changes in cellsize or cell (or file) number with changes in leaf length orwidth. Differences depended on the leaf (L1vs. L4), leaf part(bladevs. sheath), and the nature of the mutation (dwarfvs.slender). Hordeum vulgare cv. California Mariout was established in sandculture at two different NaCl concentrations (0.5 mol m–3‘control’ and 100 mol m–3) in the presenceof 6.5 mol m–3 K +. Between 16 and 31 d after germination,before stem elongation started, xylem sap was collected by useof a pressure chamber. Collections were made at three differentsites on leaves 1 and 3: at the base of the sheath, at the baseof the blade, i.e. above the ligule, and at the tip of the blade.Phloem sap was collected from leaf 3 at similar sites throughaphid stylets. The concentrations of K +, Na+, Mg2+ and Ca2+were measured. Ion concentrations in xylem sap collected at the base of leaves1 and 3 were identical, indicating there was no preferentialdelivery of specific ions to older leaves. All ion concentrationsin the xylem decreased from the base of the leaf towards thetip; these gradients were remarkably steep for young leaves,indicating high rates of ion uptake from the xylem. The gradientsdecreased with leaf age, but did not disappear completely. In phloem sap, concentrations of K+ and total osmolality declinedslightly from the tip to the base of leaves of both controland salt-treated plants. By contrast, Na+ concentrations inphloem sap collected from salt-treated plants decreased drasticallyfrom 21 mol m–3 at the tip to 7.5 mol m–3 at thebase. Data of K/Na ratios in xylem and phloem sap were used to constructan empirical model of Na+ and K+ flows within xylem and phloemduring the life cycle of a leaf, indicating recirculation ofNa+ within the leaf. The carbon economy of a flowering tiller of Poa annua L. hasbeen examined over the period from inflorescence emergence tograin shedding. The total import of 14C by the inflorescencereached a maximum at late grain filling but the relative importof assimilate was greatest 14 days after its appearance andrepresented 20–25 per cent of that assimilated by theinflorescence itself. The inflorescence continued to be an importantassimilatory organ after grain ripening when it exported morethan 50 per cent of its assimilate to the stem, roots and othertillers. The patterns of distribution of assimilates from the youngestuppermost and the oldest green leaf of the reproductive tillerwere largely determined by the stage of development of the inflorescence.The youngest leaf mainly supported the inflorescence up to theend of the grain-filling stage but then supplied assimilatesbasally to the roots and adjacent tillers. The oldest greenleaf supported the growth of the stem and the inflorescenceup to anthesis but after this supplied assimilates mainly tothe roots and tillers. Removal of grains or the entire inflorescence only 1 h beforesupplying 14CO2 greatly reduced the rate of fixation of 14CO2and the export of radiocarbon, as well as changing the patternof distribution of assimilates within the plant. The significanceof these results is discussed and comparisons made with cerealsand perennial grasses.  相似文献

The relative assimilatory activity of the inflorescence, itsindividual components, and the leaves of flowering tillers ofPoa annua L. and Lolium perenneL. was determined over the periodfrom inflorescence emergence to seed shedding. The pattern of14CO2 fixation was similar for both species and the inflorescencewas by far the most important assimilatory organ of the reproductivetiller, particularly over the latter period of seed developmentas leaf senescence progressed. With the exception of the seedsall parts of the inflorescence showed significant assimilatoryactivity and the lemmas and paleas accounted for 40–50per cent of the total 14C fixed by the inflorescence in bothspecies. The importance of the grass inflorescence as a photosyntheticstructure is discussed in relation to similar studies on cereals. Assimilate distribution in leaves of Lolium temulentum was establishedby root absorption of [14C]sucrose and after exposure to 14CO2.Age determined the amount of carbon assimilated, with more labelbeing incorporated during expansion than at maturity. Duringsenescence 14C assimilation was much lower. Ethanol-solubleextracts from various tissues of root-labelled plants containedmost of the radioactivity chiefly in basic and acidic compounds.The neutral fraction was composed predominantly of sucrose. Sucrose was comparably labelled in leaves from plants fed equalamounts of either [14C]sucrose, glucose, or fructose and onlytraces of labelled monosaccharides appeared in extracts. Radioactive sucrose was translocated rapidly from mature leaveswhereas, in the expanding leaf, carbon incorporation was directedtowards growth and the greater proportion of label present atligule formation was in ethanol-insoluble material. Induced senescence, of a mature leaf fed during expansion, produceda rapid loss from the pool of insoluble 14C. This was accompaniedby a reduction in the contents of chlorophyll and soluble proteinand an accumulation of amino acids. The onset of senescencecaused changes in leaf sugar levels which were correlated withincreased rates of respiration.  相似文献

Pathways of Uptake and Accumulation of Sugars in Tomato Fruit   总被引:2,自引:0,他引:2  
The route of sucrose unloading from the conducting tissue, theregulation of sucrose hydrolysis and the uptake and subsequentmetabolism of sugars were investigated in the rapidly growingtomato fruit. During the first two weeks of fruit enlargement, the vacuoleaccounted for more than 85% of the protoplast volume and theintercellular space accounted for 20% of the fruit placentaltissue. The plasmodesmatal frequency was highest between phloemparenchyma cells and lowest between phloem sieve cells and phloemparenchyma. The total invertase activity was about 8 µmolglucose g–1 d. wt min–1 during the rapid growingperiod and increased six-fold at ripening. The wall-bound invertaseaccounted for less than 11% of the total activity. Invertaseactivity increased with increasing sucrose concentrations (upto 50 mM) in the incubation medium, but decreased at higherconcentrations. Sucrose synthase activity could only be detectedwhen fruit was older than 19 d. The uptake and metabolism of sugars by fruit cells were investigatedby incubation of fruit slices with 14C-sugars for 3 h. The uptakeof sucrose increased with the sucrose concentration up to 200mM. The rate of glucose uptake and its conversion to the ethanol-insolublefraction were higher than those of sucrose. The uptake of sucrosedid not compete with that of glucose or vice versa, providedthe osmotic potential of the incubation solution was maintainedconstant. The uptake of sucrose was not inhibited by metabolicinhibitors such as PCMBS, CCCP, sodium azide or vanadate. TheATPase activity in the fruit tissue was low. These findings did not identify conclusively the mode of sucroseunloading. However, the uptake of sugars by fruit cells is non-specificand does not appear to require a membrane carrier or plasmalemmaATPase to provide energy for sucrose uptake. The respiratory losses and the pattern of carbon supply froma leaf of unicuim barley were examined during a complete diurnalperiod using a steady state 14C-labelling technique. After a delay of c. 1 h a portion of the 14C exported from acontinuously assimilating leaf was lost in respiration in thelight. This respiratory loss amounted to c. 20% of the total14C fixed. A further 28% of the total 14C fixed was respiredduring the dark period. In the light, carbon was fixed at a rate of c. 8·9 mgC dm–2 h–1 and exported from the leaf at c. 5·3mg C dm–2 h–1. Dark export averaged c. 31% of theday-time rate. Carbohydrate was stored in the leaf during the day and was almostcompletely remobilized during the dark. Sucrose, the major reservecarbohydrate, was exported first whilst the starch level remainedconstant. After some 9 h of darkness, sucrose declined to alow level and starch remobilization began.  相似文献

Cellular plasmolysis with l M solutions of mannitol appearedto sever plasmodesmatal interconnections between all cells ofthe stems of Phaseolus vulgaris plants except the sieve element-companioncell (se—cc) complexes. Phloem loading and uptake of [14C]sucroseby the storage cells of the stems was unimpaired by cellularplasmolysis followed by rehydration of the stem tissues. Accumulationof phloem-transported 14C-photosynthates of the treated stemswas inhibited in summer-grown plants and unaffected in winter-grownplants indicating that phloem unloading follows a symplasticand a free-space route respectively depending on growth season.At a concentration that did not interfere with cellular metabolism,p-chloromercuribenzene sulphonic acid (PCMBS) applied to thestems blocked [14C]sucrose loading into the phloem and storagecells of the stem, but had no effect on the pool size of free-spacesugars. This latter response is consistent with a facilitatedmechanism of sugar unloading to the stem free-space. Accumulationof phloem-transported 14C-photosynthates was stimulated by PCMBSand this effect was most pronounced in winter-grown plants.Cellular plasmolysis followed by rehydration abolished the PCMBSaction on 14C-photosynthate accumulation. This effect is consistentwith a PCMBS induction of phloem unloading through the stemsymplast. It is proposed that phloem unloading in bean stemsmay follow either a free-space or symplastic route and thatthe latter route is entrained under sink-limited conditions. Changes in the time in sugar and starch content of the firstleaf sheath were compared among normal, d5, and GA1-treated(1µg/plant) d5 seedlings of Zea mays L., from which endospermshad been removed. The patterns of changes in the levels of thesecarbohydrates were more or less similar to one another amongthe three groups of seedlings. Generally, the levels decreasedtoward the time of maturation of the leaf sheath except forthe level of sucrose which showed a sharp increase followedby a decrease. There was an inverse relationship over time betweengrowth and levels of glucose and fructose. Although treatmentwith GA1 resulted in a decrease in sucrose content, it had practicallyno significant effect on the levels of the other sugars. Cane leaf discs secrete sucrose into 10M iodoacetate(IOA) (Schoolar and Edelman, 1970). This secretion was foundto occur from edges of leaf material cut at right angles tothe venation but the phloem did not appear to be involved inthe secretion process. Secretion seemed to be dependent on thecarbon flow within the sugar-cane leaf, i.e. from the mesophylland the bundle sheath cells, and the action of IOA could probablybe ascribed to a diversion of this flow.  相似文献

Fresh and dry weights and leaf size of Poa pratensis were reducedwhen treated with 6-azauracil (AzU), (2-chloroethyl)phosphonicacid (CEPA), or (2-chloroethyl)trimethylammonium chloride (CCC).AzU and CEPA inhibited epidermal cell division without inhibitingcell elongation, while CCC inhibited mainly cell elongationand cell division to a small extent. The ratio of blade lengthto sheath length and the blade length/width ratio were reduced,but leaf emergence and tillering were increased by AzU and CEPA.CCC affected only the latter three features. Like GA3, CEPAinduced stem formation, but internodes were shorter. GA3 was ineffective in preventing leaf-growth inhibition byAzU, which inhibited Ga3-induced cell elongation. The inhibitoryeffect of CEPA on leaf growth was apparently reversed by GA3,but this was due solely to increased cell elongation, the reductionin cell number being unaffected. Ga3 reversed the effect ofCCC on leaf length, as well as on cell size and number. Simultaneousapplication of the inhibitors produced a complex interactionin reducing leaf length and number and size of epidermis cells.It is postulated that AzU, CEPA, and CCC have different modesof action because they have specific effects on plant growthand different effects on GA3-induced cell elongation.  相似文献

Much of the work on the distribution of 14C-labelled assimilatesin tomato has been done in winter under low light intensities,and consequently the reported distribution patterns of 14C maynot be representative of plants growing in high light. Further,there are several somewhat conflicting reports on patterns ofdistribution of 14C-assimilates in young tomato plants. We soughtto clarify the situation by studying the distribution of 14C-assimilatesin tomato plants of various ages grown in summer when the lightintensity was high. In addition, the role of the stem as a storageorgan for carbon was assessed by (a) identifying the chemicalfractions in the stem internode below a fed leaf and monitoring14 C activity in these fractions over a period of 49 d, and(b) measuring concentrations of unlabelled carbohydrates inthe stem over the life of the plant. The patterns of distribution of 14C-assimilates we found fortomato grown under high light intensity confirmed some of thosedescribed for plants grown under low light, but export of 14Cby fed leaves was generally higher than reported for much ofthe earlier work. Lower leaves of young plants exported over50% of the 14C they fixed, although export fell sharply as theplants aged. Initially, the roots and apical tuft were strongsinks for assimilates, but they had declined in importance bythe time plants reached the nine-leaf stage. On the other hand,the stem became progressively more important as a sink for 14C-assimilates.Older, lower leaves exported more of their 14C-assimilates tothe upper part of the plant than to the roots, whereas youngleaves near the top of the plant exported more of their assimilatesto the roots. The stem internode immediately below a fed leafhad about twice the 14C activity of the internode above theleaf. Mature leaves above and below a fed leaf rarely importedmuch 14C, even when in the correct phyllotactic relationshipto the fed leaf. In the first 3 d after feeding leaf 5 of nine-leaf plants, theorganic and amino acid pools and the neutral fraction of theinternode below the fed leaf had most of the 14C activity, butby 49 d after feeding, the ethanolic-insoluble, starch and lipidfractions had most of the 14C activity. Glucose, fructose andsucrose were the main sugars in the stem. Although concentrationsof these sugars and starch declined in the stem as the plantsmatured, there was little evidence to indicate their use infruit production. Stems of plants defoliated at the 44-leafstage had lower concentrations of sugars and starch at maturity,and produced less fruit than the controls. Petiole heat-girdle treatments (followed by a 5 min 14CO2 assimilation)were performed on mature leaves of Vicia faba, in order to assesstheir effect on the partitioning of photo-assimilates to theminor vein phloem. Whole leaf autoradiographic evidence indicateda high leaf-to-leaf variation in the image intensity over theminor veins (relative to the mesophyll/epidermal background)in both control and heat-girdled groups of leaves. The averagedegree of minor vein labelling in heat-girdled leaves, however,was found to be significantly lower than that in controls. Comparativeassessment of vein labelling was based on microscopic densityreadings of silver grains over veinal and interveinal regionsin autoradiographic images. Investigations into the cause ofthis alteration in vein labelling indicated no involvement ofan inhibition of apoplasmic phloem loading, as both heat-girdledand control leaves of Vicia were shown to have comparable minorvein uptake of exogenously supplied 14C-sucrose. Heat-girdlingwas shown, however, to increase significantly the partitioningof recently fixed carbon into the insoluble (mainly starch)fraction relative to the ethanol-soluble fraction, within 12min of the treatment. We suggest that this carbon partitioningchange can primarily account for the change in vein labelling,since an increase in the insoluble fraction would result in(1) more 14C-activity remaining in the leaf mesophyll and (2)less 14C-activity going into the mesophyll export pool, andthus, less 14C-sucrose being transferred to the minor vein region.Additionally, although leaf export was completely halted inheat-girdled leaves, 14C-activity was found within the majorveins as far as the point of petiole heat-girdling (followinga 5 min assimilation and 4 h chase). Apparently, continued (butlimited) solution flow within the sieve elements is maintainedby transport pathway unloading within the treated leaves. Using Proctor barley grown in sand under controlled-environmentconditions it was shown that when application of nitrogen, asnitrate, was delayed beyond day 4, at which time the first leafwas beginning to unfold, absolute and relative growth-ratesof seedlings were reduced so that the young plants were significantlysmaller. Delay in nitrate application led to reduction in length,breadth, area, and dry weight of the first leaf, and also toa lower photosynthetic activity on day 8, as measured by infra-redgas analysis. Measurement of the uptake of 14CO2 by first leaves showed thatapplication of nitrate on days 2 or 4 led to high rates of fixationof carbon over the period days 8–12, whereas applicationon days 6 or 8 led to a substantially lower maximum rate offixation which was maintained for a shorter period. When nitratewas applied on day 8 total fixation of carbon over the perioddays 7–14 was only 60 per cent of that for leaves on plantsfor which nitrate was supplied on day 2. When amounts of carbon fixed were compared on a leaf dry-weightbasis, maximum values were found to be similar for all treatments,suggesting that the differences in fixation per leaf resultmainly from the effect of treatment on leaf area. For all treatmentsit was confirmed that a decline in photosynthetic activity occurredby day 14. This was not correlated with photosynthetic activityin the second leaf, nor with emergence of the third leaf. Analysis of different parts of the lamina of the first leafshowed all to be affected by the timing of the supply, bothin growth and in photosynthetic activity. Leaf dry weight didnot increase after day 8 for any treatment, yet when nitratewas applied on day 8 plants showed a fourfold increase in photosyntheticrate. The rate of carbon transport from an old tomato leaf (54 days),grown at 80 W m–2, was measured under light flux densitiesbetween 7 and 90 W m–2. Under low light, the rate of carbontransport over a 6 h period was about 1 mg C dm–2 h–1,well in excess of the concurrent photosynthetic rate. The lossfrom these leaves of 14C-leaf assimilate which was fixed beforethe experimental period amounted to 62 per cent of the totalinitial uptake and was higher than that from leaves with higherconcurrent photosynthetic rates. The higher loss of 14C fromleaves with low photosynthetic rates was due to a greater contributionof 14C from the starch and residue fractions. The rate of transportappeared to be determined by the concentration of the labilesucrose, not the total sucrose concentration. In comparisonwith young fully-expanded tomato leaves (Ho, 1976) the sizeof the labile sucrose pool appeared to decrease with age. Thephotosynthesistranslocation coefficient was low (k1k2=0•21)for an old tomato leaf. Based on these results a scheme of carbonpartitioning in relation to translocation is proposed. Silicon deposition in the roots, culm and leaf of canary grass(Phalaris canariensis L.) was investigated using light microscopy,scanning electron microscopy and electron probe microanalysis. In adventitious roots grown in solution silicon was concentratedin four endodermal walls. Silicon was not detected in the endodermisof aerial adventitious roots, but was present in the epidermisand outer cortical cell layers. Silicon deposition in the culm mainly took place in the epidermis,and particularly in epidermal papillae. The silica deposition pattern in the leaf was typical of thesub-group Festucoideae. The leaf blade showed deposits in costalprickle hairs and wavy rods, but few intercostal deposits. Inthe ligule deposition was confined to isolated groups of pricklehairs on the abaxial surface. The major sites of silica depositionin the leaf sheath were the stomatal subsidiary cells, papillaeand intercostal idioblasts. Prickle hairs were much less commonin the sheath than the blade, and costal wavy rods appearedto be absent in the sheath. Effects of the interaction between assimilate availability andsink demand on the metabolism of 14C assimilates in tomato leaveshave been examined in plants where the source—sink relationshipof assimilates was simplified to one leaf and one fruit truss. During experimentation the source leaf was exposed to either80 or 20 W m–2 (PAR), while the truss was either retainedor removed. Under these four source-sink conditions, a timecourse study was made on 14C assimilate distribution in thesource leaf over a period of 23 h after pulse feeding with 14CO2. While truss removal caused a temporary increase of 14C sucrosein leaves under both irradiances, the principal assimilatesaccumulated were starch and hexoses. Decreased 14C export followingtruss removal was observed within a day in well-illuminatedleaves but after 3 days in leaves under low light. The accumulationof 14C sucrose at the end of the light period was affected bytruss removal in high light leaves only 3 days later. These observations suggest that while the compartmentation ofnewly fixed assimilate was affected rapidly by the change ofsource—sink relationship, carbon export, as measured by14C loss, was affected only gradually. Glycinebetaine, proline, asparagine, sucrose, glucose, and dimethylsulphoniopropionate(DMSP) were the major organic solutes in Spartina alternifloraleaf blades. To investigate the physiological role(s) of thesesolutes, the effects of salinity, nitrogen, and sulphur treatmentson leaf blade solute levels were examined. Glycinebetaine wasthe major organic solute accumulated in leaf blades grown at500 mol m–3 NaCl, although asparagine and proline alsoaccumulated when the supply of nitrogen was sufficient. Thesesolutes may play a role in osmotic adjustment. In contrast,DMSP levels either did not change or were reduced in responseto the 500 mol m–3 NaCl treatment. Furthermore, elevatednitrogen supply decreased leaf blade DMSP levels, which wasopposite to the response of glycinebetaine, proline, and asparagine.A 1000-fold increase in external sulphate concentration hadno effect on the leaf blade levels of DMSP, glycinebetaine,proline, or asparagine. These findings suggest that the majorphysiological role of DMSP in S. alterniflora leaf blades isnot for osmotic adjustment, even under conditions of nitrogendeficit and excess sulphur. Instead, DMSP which was presentat 45—130 µmol g–1 dry weight, may play arole as a constitutive organic osmoticum.
