Heterodera raskii n. sp. is described and illustrated from specimens collected from roots of bulb grass, Cyperus bulbosus, in Hyderabad, India. The new species belongs to the ''goettingiana'' group and differs from closely related H. cyperi by the elongate ovoid shaped cysts and females, greater fenestral length, width, vulval slit, and absence of egg sac. The stylet knob shape was round in second-stage juveniles and posteriorly sloping in females and males of H. raskii n. sp., while it was anteriorly directed in second-stage juveniles and spherical in females and males of H. cyperi.  相似文献

Afenestrata koreana n. sp. collected from roots of bamboo (Phyllostachys pubescens) in Gyeongnam Province in the southern part of the Korean peninsula is described and illustrated. Its primary differentiating characteristics are a globose to subspherical body in adults with a prominent neck and terminal cone, thick cuticle, terminal vulva, and deep vagina. Fenestra, bullae, and underbridge are absent. The anus is on the immediate posterior side of the cone. Superficial small tubercules cover all the terminal cone area. The new species differs markedly from the other two known species in the genus, specifically in having three incisures in the lateral field of juveniles and a shorter stylet length in juveniles and adults. Cactodera salina n. sp. (Heteroderinae) is described from roots of the estuary plant Salicornia bigelovii (Chenopodiaceae), in Puerto Pefiasco, Sonora, Mexico, at the northern tip of the Sea of Cortez. The halophyte host is grown experimentally for oilseed in plots flooded daily with seawater. Infected plants appear to be adversely affected by C. salina relative to plants in noninfested plots. Cactodera salina extends the morphological limits of the genus. Females and cysts have a very small or absent terminal cone and deep cuticular folds in a zigzag pattern more typical of Heterodera and Globodera than of Cactodera spp. Many Cactodera spp. have a tuberculate egg surface, whereas C. salina shares the character of a smooth egg with C. amaranthi, C. weissi, and C. acnidae. Only C. milleri and C. acnidae have larger cysts than C. salina. Face patterns of males and second-stage juveniles, as viewed with scanning electron microscopy, reveal the full complement of six lip sectors as in other Cactodera spp. Circumfenestrae of C. salina are typical for the genus.  相似文献

Body wall cuticle of adult females of eight genera within the Heteroderidae was examined by transmission electron microscopy for comparison with previously studied species within the family. Cuticle structure was used to test some current hypotheses of phylogeny of Heteroderidae and to evaluate intrageneric variability in cuticle layering. Verutus, Rhizonema, and Meloidodera possess striated cuticle surfaces and have the simplest layering, suggesting that striations have not necessarily arisen repeatedly in Heteroderidae through convergent or parallel evolution. Atalodera and Thecavermiculatus possess similar cuticles with derived characteristics, strengthening the hypothesis that the two genera are sister groups. Similarly, the cuticle of Cactodera resembles the specialized cuticle of Globodera and Punctodera in having a basal layer (D) and a surface layer infused with electron-dense substance. Heterodera betulae has a unique cuticle in which the thickest layer (C) is infiltrated with an electron-dense matrix. Little intrageneric difference was found between cuticles of two species of Meloidodera or between two species of Atalodera. However, Atalodera ucri has a basal layer (E) not found in other Heteroderidae. The most striking intrageneric variation in cuticle structure was observed between the thin three-layered cuticle of Sarisodera africana and the much thicker four-layered cuticle of Sarisodera hydrophila; results do not support monophyly of Sarisodera.  相似文献

Morphological comparisons with light microscopy and scanning electron microscopy were made among second-stage juveniles (J2) and males of several isolates of the three subspecies of the tobacco cyst nematode complex, Globodera tabacum sspp. tabacum, virginiae, and solanacearum. Observations focused on the anterior region, (including head shape, lip pattern, and stylet morphology) and the tail region (including tail shape in J2 and spicules in males). The three subspecies could not be separated on the basis of any of these characters.  相似文献

Gracilacus wuae n. sp. from soil associated with cow parsnip in Ontario, Canada is described and illustrated. Morphologically, females have a long stylet ranging from 80 to 93 µm long, the lip region not offset from the body contour, without lateral lips but with large and flat submedian lobes, the mouth opening slit-like elongated laterally and surrounded by lateral flaps, the excretory pore is anterior to the knobs of the stylet; males without stylet and the pharynx degenerated. The fourth-stage juveniles lack a stylet, the pharynx degenerated, and can be differentiated into preadult females and males based on the position of the genital primordia. The third-stage juveniles are similar to females but smaller. Phylogenetic studies using the rDNA small subunit 18S, large subunit 28S D2/D3, and internal transcribed spacer (ITS) sequences collectively provide evidence of a grouping with other Gracilacus and some species of Paratylenchus with stylet length of females longer than 41 µm deposited in GenBank.  相似文献

A root-knot nematode from Portugal, Meloidogyne lusitanica n. sp., is described and illustrated from specimens obtained from olive trees (Olea europaea L.). Females of the new species have a characteristic perineal pattern with medium to high trapezoidal dorsal arch with distinct punctuations in the tail terminus area. The excretory pore is located posterior to the stylet, about 1.5-2.5 stylet lengths from the anterior end. The stylet is 17.1 μm long with pear-shaped knobs. Males have a rounded, posteriorly sloping head cap and head region not annulated. The robust stylet, 24.5 μ long, has large, elongate knobs. Mean length of the second-stage juveniles is 449.5 μm, stylet length 14.2 μm, and tail length 44.1 μm. Scanning electron microscope observations provide further details of perineal patterns and head and stylet morphology of females, males, and second-stage juveniles. Meloidogyne lusitanica n. sp. did not reproduce on any of the differential hosts used to separate the four most common Meloidogyne species. The common name "olive root-knot nematode" is proposed for M. lusitanica n. sp.  相似文献

Absence of the phasmid was demonstrated with the transmission electron microscope in immature third-stage (M3) and fourth-stage (M4) males and mature fifth-stage males (M5) of Heterodera schachtii, M3 and M4 of Verutus volvingentis, and M5 of Cactodera eremica. This absence was supported by the lack of phasmid staining with Coomassie blue and cobalt sulfide. All phasmid structures, except the canal and ampulla, were absent in the postpenetration second-stage juvenile (J2) of H. schachtii. The prepenetration V. volvingentis J2 differs from H. schachtii by having only a canal remnant and no ampulla. This and parsimonious evidence suggest that these two types of phasmids probably evolved in parallel, although ampulla and receptor cavity shape are similar. Absence of the male phasmid throughout development might be associated with an amphimictic mode of reproduction. Phasmid function is discussed, and female pheromone reception ruled out. Variations in ampulla shape are evaluated as phylogenetic character states within the Heteroderinae and putative phylogenetic outgroup Hoplolaimidae.  相似文献

Meloidogyne hispanica n. sp. is described and illustrated from specimens obtained from peach rootstock, Prunus persica silvestris Batsch, from the Seville district of Spain. The perineal pattern of the female is oval shaped to rectangular with low dorsal arch and often widely spaced lateral lines with fringe-like striae. The stylet, 14.1 μm long, has broad, distinctly set off knobs. Males have a high, rounded head cap that slopes posteriorly. Labial disc and medial lips are fused to form elongate lip structures. The robust styler, 23.5 μm long, has large, rounded knobs that are slightly set off from the shaft. Mean second-stage juveniles length is 392.6 μm. The truncate head region is generally not annulated. The distinctly rounded and raised labial disc and the crescent-shaped medial lips form dumbbell-shaped lip structures. The stylet, 11.1 μm long, has rounded, posteriorly sloping knobs. The slender tail, 46.4 μm long, has large irregular-sized annules in the posterior region and ends in a bluntly rounded tip. Tomato was a good host; tobacco, pepper, and watermelon were poor hosts; cotton and peanut were nonhosts. Meloidogyne hispanica n. sp. reproduces by mitotic parthenogenesis and has a somatic chromosome number of 2n = 33-36. The esterase pattern is unique among Meloidogyne species.  相似文献

Merlinius paniculoides n. sp. is described from a bisexual Italian population found in northern Italy. This species has a coarsely annulated cuticle with approximately 30 longitudinal striae in addition to the six lateral field incisures; consequently the cuticle appears tessellate. The female tail is subcylindrical with a bluntly, rounded, non-annulated terminus. Merlinius paniculoides shares the tessellate cuticular ornamentation and the offset labial region with M. lenorus (Brown, 1956) Siddiqi, 1970 and M. tartuensis (Krall, 1959) Siddiqi, 1970. It differs from M. lenorus by the higher c and lower c'' ratios (c = 16-19 vs. 12-15 and c'' = 2.2-2,8 vs. 3.7) and greater number of longitudinal striae (28-32 vs. 24). It differs from M. tartuensis by a fewer number of annuti from stylet knob base to lip constriction (8-9 vs. 21), shorter stylet (19-21 vs. 24 μm), presence of males, and higher c'' value (c'' = 2.2-2.8 vs. 1.9).  相似文献

A new genus and species of cyst nematode, Vittatidera zeaphila, is described from Tennessee. The new genus is superficially similar to Cactodera but is distinguished from other cyst-forming taxa in having a persistent lateral field in females and cysts, persistent vulval lips covering a circumfenestrate vulva, and subventral gland nuclei of the female contained in a separate small lobe. Infective juveniles (J2) are distinguished from all previously described Cactodera spp. by the short stylet in the second-stage juvenile (14-17 μm); J2 of Cactodera spp. have stylets at least 18 μm long. The new species also is unusual in that the females produce large egg masses. Known hosts are corn and goosegrass. DNA analysis suggests that Vittatidera forms a separate group apart from other cyst-forming genera within Heteroderinae.  相似文献

Meloidogyne mayaguensis n. sp. is described and illustrated from specimens obtained from galled roots of eggplant, Solanum melongena L., from Puerto Rico. The perineal pattern of females is round to ovoid with fine, widely spaced striae. It has occasional breaks of striation laterally and a circular tail tip area lacking striae. The stylet, 15.8 μm long, has reniform knobs that merge gradually with the stylet shaft. Males have a high, rectangular, smooth head region, not set off from the body contour. The labial disc is continuous with the medial lips which do not slope posteriorly. The styler, 22.9 μm long, has large rounded backward sloping knobs; the shaft is of uneven diameter. Mean body length of second-stage juveniles is 453.6 μm. The truncate head region is not annulated, and the rounded, slightly raised labial disc and the crescentic medial lips form dumbbell-shaped lip structures. The stylet, 11.6 μm long, has rounded, posteriorly sloping knobs. The slender tail, 54.4 μm long, gradually tapers to a bluntly pointed tip. Tomato, tobacco, pepper, and watermelon are good hosts; cotton and peanut are not hosts. M. mayaguensis n. sp. reproduces by mitotic parthenogenesis and has a somatic chromosome number of 2n = 44-45. The enzyme patterns are unique among Meloidogyne species.  相似文献

A cyst nematode, Heterodera goldeni n. sp., is photographed and described from Qasabagrass roots (Panicum coloratum L.) in Alexandria, Egypt. It is characterized in having second-stage juveniles with body length of 546 µm (450-612), stylet length of 22.6 µm (22-23.5) with anchor-shaped knobs, lateral field with 3 lines, tail 60-75 µm, hyaline tail terminus 38.4 µm (33-43); cysts are lemon-shaped, dark to light brown with an extensive sub-crystalline layer covering the entire cyst, cuticular midbody pattern zig-zag, cysts ambifenestrate, well-developed underbridge with finger-like projections, bullae present, vulva slit measuring 44-48 µm long. Males are absent, and females have heavy punctations on the cuticle. Its relationship to H. graminophila described from Florida and Louisiana and H. leuceilyma described from Florida are discussed. The present known distribution is restricted to Alexandria, Egypt. Its economic importance in rangeland grasses and cultivated crops such as rice is not known.  相似文献

A morphometric evaluation of second-stage juveniles (J2), males, females, cysts, and eggs of several isolates of the tobacco cyst nematode (TCN) complex, Globodera tabacum tabacum (GTT), G. t. virginiae (GTV), and G. t. solanacearum (GTS) is presented. Morphometrics of eggs, J2, and males are considerably less variable than of females and cysts. No measurements of eggs and J2 are useful for identification of the three subspecies. Distance from the median bulb and excretory pore to the head end in J2 and males is quite stable. Stylet knob width of males is useful for identifying GTV isolates and tail length in separating males of GTT isolates from GTV and GTS. Body length/width (L/W) ratio of females and cysts discriminates GTT from GTV and GTS; stylet knob width is an auxiliary character for identifying GTV. This subspecies complex has a continuum of values for the other characters. Data suggest a close relationship between GTV and GTS, which also occur in close proximity in Virginia.  相似文献

Meloidogyne microcephala n. sp. is described and illustrated from specimens obtained from tobacco (Nicotiana tabacum L.) in Thailand. The female perineal pattern usually has a low dorsal arch, coarse striae, and a series of small cuticular flaps around the tail terminus. The stylet of the female is 14.4 μm long, with large, square to rectangular stylet knobs, The distinctive male head region is narrow, small, and truncate with a low, flattened head cap. The stylet length is 20.6 μm, and the knobs are small, angular, and set off from the shaft. Mean length of second-stage juveniles is 457.5 μm, and stylet length is 9.3 μm. The tail tip in the juveniles is set off from the rest of the tail as a small finger-like projection. M. microcephala reproduces by mitotic parthenogenesis, and has a chromosome number of 2n = 36.  相似文献

A new species of cyst nematode, Globodera ellingtonae, is described from soil collected from a field in Oregon. Second-stage juveniles (J2) of the species are characterized by body length of 365-515 μm, stylet length of 19-22.5 μm, basal knobs rounded posteriorly and pointed anteriorly, tail 39-55 μm, hyaline tail terminus 20-32.5 μm, and tail tapering uniformly but abruptly narrowing and constricted near the posterior third of the hyaline portion, ending with a peg-like, finely rounded to pointed terminus. Cysts are spherical to sub-spherical, dark to light brown and circumfenestrate and cyst wall pattern is ridge-like with heavy punctations. Males have a stylet length of 21-25 μm and spicule length of 30-37 μm with a pointed thorn-like tip. Females have a stylet length of 20-22.5 μm, one head annule and labial disc, heavy punctations on the cuticle, and short vulval slit 7.5-8 μm long. Morphologically this new, round-cyst species differs from the related species G. pallida, G. rostochiensis, G. tabacum complex and G. mexicana by its distinctive J2 tail, and by one or another of the following: shorter mean stylet length in J2, females and males; number of refractive bodies in the hyaline tail terminus of J2; cyst morphology including Granek’s ratio; number of cuticular ridges between the anus and vulva; and in the shape and length of spicules in males. Its relationship to these closely related species are discussed. Based upon analysis of ribosomal internal transcribed spacer (ITS) sequences, G. ellingtonae n. sp. is distinct from G. pallida, G. rostochiensis, G. tabacum and G. mexicana. Bayesian and Maximum Parsimony analysis of cloned ITS rRNA gene sequences indicated three clades, with intraspecific variability as high as 2.8%. In silico analysis revealed ITS restriction fragment length polymorphisms for enzymes Bsh 1236I, Hinf I, and Rsa I that overlap patterns for other Globodera species.  相似文献

Clones of two partially resistant and two susceptible white clover, Trifolium repens, genotypes were exposed to eggs of Heterodera trifolii and nematode development in stained roots measured at 2, 4, 7, 11, 18, 23, and 37 days after inoculation. The differences in development between nematode populations in resistant and susceptible genotypes showed that resistance operated after infection during feeding and development. At 7 days after inoculation, counts of second-stage juveniles did not differ between genotypes, whereas at 37 days more adults had developed in the susceptible than in the resistant genotypes. In a separate experiment, cysts hosted by susceptible genotypes were larger and contained more eggs than those on resistant genotypes so that the product of the values for cysts per plant and for eggs per cyst resulted in a more sensitive measure of resistance than from using cysts per plant alone.  相似文献

A new apparatus to release eggs from cysts of soybean cyst nematode (Heterodera glycines) is described and its efficiency evaluated. A rubber stopper was mounted on a bolt, and cysts were ground against a 60-mesh screen. Eggs and second-stage juveniles were washed into a series of screens nested underneath the apparatus. This method was fast and efficient, and had no ill effect on prepared inoculum.  相似文献
