The effects of ultraviolet radiation on the gravitactic orientation of the freshwater flagellate,Euglena gracilis, were determined by a real time image analysis system. Both artificial UV radiation and solar radiation in a temperature-controlled growth chamber were employed. Histograms of gravitaxis showed that the degree of orientation decreased with increasing exposure time; this can be quantified using the Rayleigh test and upper quadrant summation. The effects of artificial UV radiation on the orientation are considerably stronger than those of solar radiation, probably because the radiation source emits higher fluence rates below 300 nm than found in solar radiation. The effects of monochromatic ultraviolet radiation on motility have been determined, and an action spectrum has been calculated.  相似文献

The effects of solar radiation on motility, photoorientation and pigmentation have been studied in a freshwater Cryptomonas species. The diaphototactic orientation performed by the cells is impaired within about 90 min of solar radiation. Likewise, the percentage of motile cells within the population and the average velocity of the swimming cells decreases within about the same exposure time. This effect is not due to a thermal stress but rather seems to be caused by the solar UV-B component, since decreasing short wavelength UV radiation by means of an artificial ozone filter or UV cut-off filters increased the tolerated exposure time. Solar radiation also bleached the photosynthetic pigments of the cells as shown by absorption difference spectra.  相似文献

The effects of solar radiation and artificial UV irradiation on motility and pigmentation were studied in the flagellate system Cyanophora paradoxa. Both percentage of motile cells and average velocity decreased drastically after a solar exposure of a few hours. This effect was not due to an overheating since the cells were exposed under temperaturecontrolled conditions. Partial reduction of the UV-B radiation by cut-off filters or by insertion of an artificial ozone layer increased the tolerated exposure times. Artificial UV radiation also induced the same effects. Under both solar and artificial UV irradiation the photosynthetic pigments within the cyanelles were bleached also within short exposure times. Kinetics of pigment destruction showed that the accessory phycobilins are lost with a half life of 1.3 h while the chlorophylls had a half life of 33 h and a carotenoid with an absorption maximum at 480 nm of 17.3 h.  相似文献

UV-B radiation is absorbed effectively by nucleic acids and other sensitive targets, potentially causing harmful photochemical effects. Protection against UV-B radiation may be afforded by flavonoids and other phenolics, which absorb strongly in the UV region, but little is known about the role played by other compounds, such as terpenes. Grindelia chiloensis, native of Patagonia (Argentina), can accumulate as much as 25% resin (terpenes) in its leaves. The present investigation was carried out to test the effect of solar UV-B radiation on the allocation of photoassimilates to biomass and terpenes. Exposure to UV-B radiation reduced whole plant biomass, plant height and leaf area, and increased leaf thickness and resin accumulation in Grindelia chiloensis. Higher absorbance was found for refined resin in the UV-B waveband from plants grown under solar UV-B radiation than plants without UV-B radiation. These chemical and structural changes could protect the plant from UV radiation, and may help elucidate the importance of epicuticular resins for a species as G. chiloensis native to an environment with maximum daily integrated values of solar UV-B irradiance.  相似文献

The effect of solar UV radiation on the physiology of the intertidal green macroalga Ulva lactuca L. was investigated. A natural Ulva community at the shore of Helgoland was covered with screening foils, excluding UV-B or UV-B + UV-A from the solar spectrum. In the sampled material, changes in the activity and concentration of ribulose-1,5-bisphosphate carboxylase-oxygenase (Rubisco), and the concentration of photosynthetic and xanthophyll cycle pigments were determined. Exclusion of UV radiation from the natural solar spectrum resulted in an elevated overall activity of Rubisco, related to an increase in its cellular concentration. Among the photosynthetic pigments, lutein concentration was substantially elevated under UV exclusion. In addition, marked UV effects on the xanthophyll cycle were found: exclusion of solar UV radiation (and particularly UV-B) resulted in an increased ratio of zeaxanthin concentration to the total xanthophyll content, indicating adverse effects of UV-B on the efficiency of photoprotection under high irradiances of photosynthetically active radiation. The results confirm a marked impact of present UV-B levels on macroalgal physiology under field conditions.  相似文献

At Helgoland, in the North Sea, growth of the high sublittoral brown macroalga Dictyota dichotoma (Hudson) Lamoroux was examined in October (the time of tetraspore release) in an outdoor tank by exposing 2-day-old germlings to four solar radiation treatments achieved with different filter materials or an additional artificial light source: photosynthetically active radiation (PAR; 395–700 nm), PAR plus ultraviolet (UV)-A (320–700 nm), full solar spectrum, or solar spectrum plus artificial UV radiation (UVR). Based on length measurements over a period of 3 weeks, the growth rate in germlings strongly decreased in conditions with UVR compared to PAR: by 14% under PAR+UV-A, by 31% under the full solar spectrum and by 65% with additional UVR. Although growth rates of germlings under UVR were reduced mainly in the first week, the plants did not regain the size of the untreated plants even after 9 weeks. Regardless of the exposure, no defects in morphology or anatomy including the exposed apical meristem were detected, except for a reduction in cell division rates perhaps due to additional cost for photoprotective or repair mechanisms. Depending on the actual position of D. dichotoma plants in the natural habitat, individuals in high positions receive substantial amounts of the more harmful UV-B while those lower down might only receive UV-A during part of the day, thus the effect of UV-B on the growth of D. dichotoma will depend on its position in the field. The effects of tidal variation of the light climate and the implications of our results for the zonation of D. dichotoma are discussed. Lanzhou city is located in north central China near inner Mongolia. The solar UV-B background radiation in this area is occasionally extremely high (8 microW/cm2). Such high background solar UV-B radiation could be attributed to the sporadic depletion of the ozone layer in the stratosphere. The excessive UV-B radiation is a potential hazard in the environment. This prompted the present study on the effect of UV-B radiation on the cytogenetic damage to pollen mother cells of the plant Tradescantia. The Tradescantia-micronucleus (Trad-MCN) test was used to determine the genotoxicity of UV-B radiation. In addition to the usual 10 h of solar emission of UV light a series of increasing dosages (2, 4, 6, 8 h) of artificial UV-B radiation was applied to Tradescantia (clone 3) plant cuttings. Inflorescences of the treated and control plants were fixed and used for preparation of microslides. Micronuclei frequencies were observed in the early tetrads to show the degree of genotoxicity. Results of two repeated experiments show a dose-related increase of MCN frequencies under normal sunshine days. In the third experiment conducted under a cloudy and rainy day and an extraordinary high solar UV-B background, the MCN frequencies were markedly higher than that of the negative control but did not show the clear dose response to the treatment as in the first two experiments. The Trad-MCN test has successfully detected the effect of artificial UV-B radiation over the solar UV-B background radiation.  相似文献

The physiological effects unique to solar ultraviolet (UV)-B exposure (280-315 nm) are difficult to accurately replicate in the laboratory. This study evaluates the effectiveness of the sodium urate anion in a liquid filter that yields a spectrum nearly indistinguishable from the solar UV-B spectrum while filtering the emissions of widely used UV-B lamps. The photochemical properties and stability of this filter are examined and weighed against a typical spectrum of ground-level solar UV-B radiation. To test the effectiveness of this filter, light-saturated photosynthetic oxygen evolution rates were measured following exposure to UV-B filtered either by this urate filter or the widely used cellulose acetate (CA) filter. The ubiquitous marine Chlorophyte alga Dunaliella tertiolecta was tested under identical UV-B flux densities coupled with ecologically realistic fluxes of UV-A and visible radiation for 6 and 12 h exposures. These results indicate that the urate-filtered UV-B radiation yields minor photosynthetic inhibition when compared with exposures lacking in UV-B. This is in agreement with published experiments using solar radiation. In sharp contrast, radiation filtered by CA filters produced large inhibition of photosynthesis.  相似文献

The effects of UV-B radiation on the heterotrophic nanoflagellate Bodo saltans (Kinetoplastida) were examined under controlled conditions with artificial UV sources and also under natural solar radiation in an oligotrophic lake. In both types of experiments, the characteristic elongated cell morphology of this flagellate changed into a spherical one. This effect was due to UV-B but also to UV-A radiation, and after 4 h of exposure at 0.5 m of depth, 99% (UV-B plus UV-A plus photosynthetically active radiation) and 69% of the cells (UV-A plus photosynthetically active radiation) were spherical. At 6 m of depth where only 10% of the UV-B (305 nm) at the surface was measured, no significant effect was observed. The spherical cells were nonmotile, but before the morphological change took place, the swimming speed was ca. 3.5 times lower in the plus-UV-B treatment. The negative relation between the abundance of spherical cells and the average ingestion of fluorescently labeled bacteria per cell indicates that these cells are not able to feed upon bacteria. In bacterivory experiments lasting for 6 h, the total number of grazed bacteria was up to 70% lower in the plus-UV-B treatment than in the control without UV-B. This resulted in a positive feedback between UV-B and bacterial growth. The high sensitivity of B. saltans to solar UV-B and UV-A radiation strongly reduces its ability to live near the surface at times of high UV radiation.  相似文献

During the last few decades many experiments have been performed to evaluate the responses of plants to enhanced solar UV-B radiation (280–320 nm) that may occur because of stratospheric ozone depletion; most of them were performed in controlled environment conditions where plants were exposed to low photosynthetically active radiation (PAR) levels and high UV-B irradiance. Since environmental radiative regimes can play a role in the response of plants to UV-B enhancement, it appears doubtful whether it is valid to extrapolate the results from these experiments to plants grown in natural conditions. The objective of this work was to evaluate the effects on physiology and morphology of a bean (Phaseolus vulgaris L.) cultivar Nano Bobis, exposed to supplemental UV radiation in the open-air. UV-B radiation was supplied by fluorescent lamps to simulate a 20% stratospheric ozone reduction. Three groups of plants were grown: control (no supplemental UV), UV-A treatment (supplementation in the UV-A band) and UV-B treatment (supplemental UV-B and UV-A radiation). Each group was replicated three times. After 33 days of treatment plants grown under UV-B treatment had lower biomass, leaf area and reduced leaf elongation compared to UV-A treatment. No significant differences were detected in photosynthetic parameters, photosynthetic pigments and UV-B absorbing compounds among the three groups of plants. However, plants exposed to UV-A treatment showed a sort of 'stimulation' of their growth when compared to the control. The results of this experiment showed that plants may be sensitive to UV-A radiation, thus it is difficult to evaluate the specific effects of UV-B (280–320 nm) radiation from fluorescent lamps and it is important to choose the appropriate control. Environmental conditions strongly affect plant response to UV radiation so further field studies are necessary to assess the interaction between UV-B exposure and meteorological variability.  相似文献

Abstract: The effects of solar ultraviolet radiation (UV) on carbon uptake, oxygen evolution and motility of marine phytoplankton were investigated in coastal waters at Kristineberg Marine Research Station on the west coast of Sweden (58° 30'N, 11° 30'E). The mean irradiances at noon above the water surface during the investigation period were: photosynthetic active radiation (PAR, 400–700 nm) 1670 μmol m−2 s−1; ultraviolet-A radiation (UV-A, 320–400 nm) 35.9 W m−2 and ultraviolet-B radiation (UV-B, 280–320 nm) 1.7 W m−2. UV-B radiation was much more attenuated with depth in the water column than were PAR and UV-A radiation. UV-B radiation could not be detected at depths greater than 100–150 cm. Inhibition of carbon uptake by UV-A and UV-B in natural phytoplankton populations was greatest at 50 cm depth and the effects of UV-B were greater than those of UV-A. At depths greater than 50 cm there was almost no effect of ultraviolet radiation on carbon uptake. PAR, UV-A and UV-B decreased oxygen evolution by the dinoflagellate Prorocentrum minimum . Inhibition of oxygen evolution was greater after 4 h than 2 h but it was not possible to distinguish the negative effects of the different light regimes. The motility of P. minimum was not affected by PAR, UV-A and UV-B. The importance of exposure of phytoplankton to different light regimes before being exposed to natural solar radiation is discussed.  相似文献

Abstract: The effects of solar radiation on photosynthetic oxygen production and pulse amplitude modulated (PAM) fluorescence were measured in the marine brown macroalga Padina pavonia harvested from different depths from the Greek coast near Korinth. In fluence rate-response curves the light compensation point for photosynthetic oxygen production increased and the saturation level decreased with increasing exposure time to solar radiation. Cutting off the UV-B wavelength range (280–315 nm) from solar radiation reduced the inhibition of photosynthesis, and the organisms were less affected when all of the UV radiation was filtered out. Algae collected from 7 m depth were much more prone to photoinhibition than those harvested from rock pools exposed to unfiltered solar radiation. During continuous exposure to solar radiation, rock pool algae showed photoinhibition after longer periods of time than specimens from 7 m or from dark adapted habitats. When subjected to unfiltered solar radiation the ratio of the variable fluorescence to the maximal fluorescence     (Fv = Fm− Fo) rapidly declined with increasing exposure time. However, again algae from 7 m depth were more prone to photoinhibition than rock pool algae. The differences between the two ecological strains were less obvious when UV-B or total UV was removed from solar radiation. Only in the latter case a complete recovery was observed after 2 h while, when exposed to unifiltered sunlight, only the rock pool algae recovered completely within that time.  相似文献

Continuous depletion of the stratospheric ozone layer has resulted in an increase in solar ultraviolet-B (UV-B; 280–315 nm) radiation reaching the Earth's surface. The consequences for aquatic phototrophic organisms of this small change in the solar spectrum are currently uncertain. UV radiation has been shown to adversely affect a number of photochemical and photobiological processes in a wide variety of aquatic organisms, such as cyanobacteria, phytoplankton and macroalgae. However, a number of photosynthetic organisms counteract the damaging effects of UV-B by synthesizing UV protective compounds such as mycosporine-like amino acids (MAAs) and the cyanobacterial sheath pigment, scytonemin. The aim of this contribution is to discuss the responses of algae and cyanobacteria to solar UV-B radiation and the role of photoprotective compounds in mitigating UV-B damage.  相似文献

由大气臭氧层减薄导致的UV-B辐射变化将直接影响到凋落物的分解。目前,有关UV-B辐射影响木本植物凋落物分解的研究还很少,在国内还没有开展。采用分解袋法开展了马尾松凋落叶在自然环境和UV-B辐射滤减两种辐射环境下的分解试验。结果表明:在UV-B辐射滤减环境下的马尾松凋落叶年分解速率比对照环境减慢了47.74%。UV-B辐射极显著(p<0.01)地加快了马尾松凋落叶的分解速率,促进了凋落叶中碳、磷、钾的释放和木质素的降解,对氮的释放无明显影响。研究结果意味着UV-B辐射将加快马尾松林的营养循环速度,降低马尾松林凋落物层的碳储量。  相似文献

Several unicellular algae were exposed to artificial UV-B (280–320 nm) radiation after adaptation to high (43 W m−2) and low (19 W m−2) visible light. UV-B radiation had different effects on rates of photosynthesis, motility and absorption spectra for these species. Photosynthesis of Euglena gracilis and the diatom Phaeodactylum tricomution was more sensitive to UV-B inhibition than that of the dinoflagellates Heterocapsa triquetra and Prorocentrum minimum . Not only UV-B radiation but also high visible light had a photoinhibitory effect on photosynthesis in all four organisms. The effect on photosynthesis was observed both on the quantum yield and on the light saturation rate of photosynthesis. The dinoflagellates, in contrast to E. gracilis and P. tricorntum , absorbed strongly in the UV region (334 nm) and their absorption peaks increased after growth under high visible light or with or without UV-B radiation for one week. The swimming speed of H. triquetra decreased more after low visible light and UV-B radiation compared to high visible light and UV-B radiation. The negative effects of UV-B radiation on P. minimum and E. gracilis were most pronounced after high visible light.  相似文献

The loss of stratospheric ozone and the accompanying increase in solar UV flux have led to concerns regarding decreases in global microbial productivity. Central to understanding this process is determining the types and amounts of DNA damage in microbes caused by solar UV irradiation. While UV irradiation of dormant Bacillus subtilis endospores results mainly in formation of the "spore photoproduct" 5-thyminyl-5,6-dihydrothymine, genetic evidence indicates that an additional DNA photoproduct(s) may be formed in spores exposed to solar UV-B and UV-A radiation (Y. Xue and W. L. Nicholson, Appl. Environ. Microbiol. 62:2221-2227, 1996). We examined the occurrence of double-strand breaks, single-strand breaks, cyclobutane pyrimidine dimers, and apurinic-apyrimidinic sites in spore DNA under several UV irradiation conditions by using enzymatic probes and neutral or alkaline agarose gel electrophoresis. DNA from spores irradiated with artificial 254-nm UV-C radiation accumulated single-strand breaks, double-strand breaks, and cyclobutane pyrimidine dimers, while DNA from spores exposed to artificial UV-B radiation (wavelengths, 290 to 310 nm) accumulated only cyclobutane pyrimidine dimers. DNA from spores exposed to full-spectrum sunlight (UV-B and UV-A radiation) accumulated single-strand breaks, double-strand breaks, and cyclobutane pyrimidine dimers, whereas DNA from spores exposed to sunlight from which the UV-B component had been removed with a filter ("UV-A sunlight") accumulated only single-strand breaks and double-strand breaks. Apurinic-apyrimidinic sites were not detected in spore DNA under any of the irradiation conditions used. Our data indicate that there is a complex spectrum of UV photoproducts in DNA of bacterial spores exposed to solar UV irradiation in the environment.  相似文献

Besides playing an essential role in plant photosynthesis, solar radiation is also involved in many other important biological processes. In particular, it has been demonstrated that ultraviolet (UV) solar radiation plays a relevant role in grapevines (Vitis vinifera) in the production of certain important chemical compounds directly responsible for yield and wine quality. Moreover, the exposure to UV-B radiation (280–320 nm) can affect plant–disease interaction by influencing the behaviour of both pathogen and host. The main objective of this research was to characterise the solar radiative regime of a vineyard, in terms of photosynthetically active radiation (PAR) and UV components. In this analysis, solar spectral UV irradiance components, broadband UV (280–400 nm), spectral UV-B and UV-A (320–400 nm), the biological effective UVBE, as well as the PAR (400–700 nm) component, were all considered. The diurnal patterns of these quantities and the UV-B/PAR and UV-B/UV-A ratios were analysed to investigate the effect of row orientation of the vineyard in combination with solar azimuth and elevation angles. The distribution of PAR and UV irradiance at various heights of the vertical sides of the rows was also studied. The results showed that the highest portion of plants received higher levels of daily radiation, especially the UV-B component. Row orientation of the vines had a pronounced effect on the global PAR received by the two sides of the rows and, to a lesser extent, UV-A and UV-B. When only the diffused component was considered, this geometrical effect was greatly attenuated. UV-B/PAR and UV-A/PAR ratios were also affected, with potential consequences on physiological processes. Because of the high diffusive capacity of the UV-B radiation, the UV-B/PAR ratio was significantly lower on the plant portions exposed to full sunlight than on those in the shade.  相似文献

We tested the effects of solar radiation, and UV-B in particular, on the growth of Antarctic terrestrial fungi. The growth responses to solar radiation of five fungi, Geomyces pannorum, Phoma herbarum, Pythium sp., Verticillium sp., and Mortierella parvispora, each isolated from Antarctic terrestrial habitats, were examined on an agar medium in the natural Antarctic environment. A 3-h exposure to solar radiation of >287 nm reduced the hyphal extension rates of all species relative to controls kept in the dark. Pythium sp. cultures exposed to solar radiation for 1.5 h on five consecutive days were most sensitive to radiation of >287 nm, but radiation of >313 nm also inhibited growth to a lesser extent. Radiation of >400 nm had no effect on hyphal growth relative to controls kept in the dark. Short-wave solar UV-B radiation of between 287 and 305 nm inhibited the growth of Pythium sp. hyphae on and below the surface of the agar medium after 24 h, but radiation of > or =345 nm only reduced the growth of surface hyphae. Similar detrimental effects of UV-B on surface and, to a lesser extent, submerged hyphae of all five fungi were shown in the laboratory by using artificial UV-B from fluorescent lamps. A comparison of growth responses to solar radiation and temperature showed that the species that were most resistant to UV radiation grew fastest at higher temperatures. These data suggest that solar UV-B reduces the growth of fungi on the soil surface in the Antarctic terrestrial environment.  相似文献

Plant responses to solar UV radiation are numerous and have often been considered from a perspective of negative outcomes for plant productivity. In this study, we used two experimental approaches consisting of: (1) field-based spectrally modifying filters in addition to (2) controlled indoor exposure to UV-B, to examine the effects of UV radiation on growth and photosynthetic performance of lettuce (Lactuca sativa L.) seedlings. Various aspects of growth were affected in plants grown under a UV-inclusive environment compared to a UV-depleted environment, including reductions in leaf expansion, increases in leaf thickness and the rate of net photosynthesis. After transplantation to a uniform field environment, lettuce plants initially propagated under the UV-inclusive environment exhibited higher harvestable yields than those from a UV-depleted environment. In controlled conditions, photosynthetic rates were higher in plants grown in the presence of UV-B radiation, and relative growth of plants pre-acclimatized to UV-B was also increased, in addition to higher maximum photochemical efficiency of photosystem II (PSII) (F(v) /F(m) ) following subsequent exposure to high photosynthetically active radiation (PAR) and temperature stress. Our findings are discussed within the context of sustainability in agriculture and the paradigm shift in photobiology which such beneficial responses to UV radiation could represent.  相似文献
