Polysaccharide was synthesized by Aureobasidium pullulans (or Pullularia pullulans) 2552 in a sucrose medium. The field apparent viscosity of the culture medium from shake flask experiments rose to 24,500 cP and then dropped toward its initial value as the fermentation progressed. The magnitude of the maximum apparent viscosity depended on the initial pH of the fermentation broth. The inoculum age influenced the cultivation period before which the maximum viscosity was reached. Rheograms of the fermentation broths showed a change in viscosity behavior from Newtonian to pseudoplastic, and then toward Newtonian characteristics during the fermentation. The calculated non-Newtonian index was found to be a sensitive factor for the indication of the non-Newtonian behavior. Such behavior could not be detected from rheograms. Viscosity profiles of polysaccharide isolated from various stages of the fermentation showed a change from Newtonian to pseudoplastic behavior depending on the concentration (0–2%) of polysaccharide.  相似文献

Quaternized celluloses (QCs) were homogeneously synthesized by reacting cellulose with 3-chloro-2-hydroxypropyltrimethylammonium chloride (CHPTAC) in NaOH/urea aqueous solutions. The structure and solution properties of the QCs were characterized by using elemental analysis, FTIR, (13)C NMR, SEC-LLS, viscometer, and zeta-potential measurement. The results revealed that water-soluble QCs, with a degree of substitution (DS) value of 0.20-0.63, could be obtained by adjusting the molar ratio of CHPTAC to anhydroglucose unit (AGU) of cellulose and the reaction time. The QC solutions in water displayed a typical polyelectrolyte behavior, and the intrinsic viscosity ([eta]) value determined from the Fuoss-Strauss method increased with increasing DS value. Moreover, two QC samples (DS = 0.46 and 0.63) were selected and studied as gene carriers. The results of gel retardation assay suggested that QCs could condense DNA efficiently. QCs displayed relatively lower cytotoxicity as compared with PEI, and QC/DNA complexes exhibited effective transfection compared to the naked DNA in 293T cells. The quaternized cellulose derivatives prepared in NaOH/urea aqueous solutions could be considered as promising nonviral gene carriers.  相似文献

Information about the thermal degradation of the polysaccharides sodium alginate, carrageenan and carboxymethyl cellulose has been obtained from the time dependence of the viscosity at high temperatures measured using a slit viscometer. The viscosity is related to the molecular weight using previously-published relations between the zero shear specific viscosity and the coil overlap parameter in conjunction with the appropriate Mark-Houwink equation. It is found that alginate is much less stable than carboxymethyl cellulose and carrageenan. Activation energies for depolymerisation obtained from Arrhenius plots in the presence of oxygen ranged from 50 kJ/mol for alginate to 105 kJ/mol for κ-carrageenan.  相似文献

Oxalic acid and hydrogen peroxide have been suggested to be essential in the degradation of wood carbohydrates by brown-rot fungi. The production of oxalic acid, hydrogen peroxide and endo-β-1,4-glucanase activity by the brown-rot fungus Poria placenta was studied on crystalline cellulose, amorphous cellulose and glucose media. Oxalic acid and hydrogen peroxide by P. placenta were clearly produced on culture media containing either crystalline or amorphous cellulose. Oxalic acid and hydrogen peroxide were formed simultaneously and highest amounts of oxalic acid (1.0 g l−1) and hydrogen peroxide (39.5 μM) were obtained on amorphous cellulose after 3 weeks cultivation. On glucose medium the amounts were low. The endoglucanase activity was observed to increase during the cultivation and was most pronounced on glucose medium and thus indicated the constitutive characteristics of the brown-rot cellulases.  相似文献

裂褶菌纤维二糖脱氢酶(cellobiose dehydrogenase,CDH)可以提高纤维素酶对纤维素的降解。以纤维二糖为电子供体,CDH作用于羧甲基纤维可降低其溶液的粘度,作用纤维素CF11和磷酸膨胀纤维素,分别使其悬浊液的浊度提高7%和14.4%。CDH与纤维二糖水解酶或切纤维素酶在降解棉花纤维素时没有表现出协同作用。但若棉花事先在纤维二糖存在下用CDH预处理,则变得易于被水解。  相似文献

The apparent viscosity of baker's yeast suspensions was employed to control the cell concentration of the mixing of baker's yeast and distilled water. The apparent viscosity of baker's yeast was found to be a function of cell concentration and independent of the flow rate through the rotational viscometer. The viscometer and mixing unit were found to have first order dynamics. The open loop dynamics of the control system were nonlinear but could be linearized for small perturbations. The control system was employed to successfully control the cell concentration but exhibited nonlinear behavior.  相似文献

The viscosity–temperature behavior of N4 virus solution was investigated by using a Zimm type viscometer. The role of ionic strength (LiBr) and pH in the viscosity–temperature behavior was determined. Supporting measurements of the sedimentation coefficient, infectivity, and optical density are presented. The viscosity–temperature behavior generally includes a region of low viscosity, characteristic of the intact virus, followed, upon increasing temperature, by a rather abrupt increase of viscosity which corresponds to the ejection of native DNA from the protein shell, presumably through a suitable opening in the capside. The viscosity remains constant upon further increase of temperature until a second abrupt increase is observed which is shown to correspond to the degradation of the empty protein shell (ghost). Finally, at a still higher temperature, denaturation of DNA occurs. This sequence of events can be altered by changes in pH and ionic strength. Increasing LiBr concentration reduces the temperature of ghost degradation and raises the temperature of DNA ejection and DNA de naturation. It is suggested that polyelectrolyte effects play a controlling role in the ejection of native DNA and that the conformation of the latter within the protein shell is stabilized by a decrease of the electrostatic free energy due to the swamping effect of the co-ions.  相似文献

The formation and location of endo-1,4-beta-glucanases and beta-glucosidases were studied in cultures of Cellulomonas uda grown on microcrystalline cellulose, carboxymethyl cellulose, printed newspaper, and some mono- or disaccharides. Endo-1,4-Glucanases were found to be extracellular, but a very small amount of cell-bound endo-1,4-beta-glucanase was considered to be the basal endoglucanase level of the cells. The formation of extracellular endo-1,4-beta-glucanases was induced by cellobiose and repressed by glucose. Extracellular endoglucanase activity was inhibited by cellobiose but not by glucose. beta-Glucosidases, on the other hand, were formed constitutively and found to be cell bound. beta-Glucosidase activity was inhibited noncompetitively by glucose. Some characteristics such as the optimal pH for and the thermostability of the endoglucanases and beta-glucosidases and the end products of cellulose degradation were determined.  相似文献

The formation of cellulases by Trichoderma viride in a medium containing cellulose as a sole source of carbon depends on the oxygen transfer rate (OSR); the OSR, on the other hand, depends on the concentration of cellulose in the medium because the concentration of cellulose strongly affects the viscosity of the medium. In the work presented here, the dependence has been determined for the oxygen transfer rate on geometric relations and viscosity in cellulose-containing media during cultivation in shaken flasks, and the oxygen transfer rate on N(Re') N(G') and N(a) during cultivation in a laboratory fermentor of 3000-mL volume. Two cellulosic materials have been compared with a different effect on viscosity: Microcrystalline beach cellulose and fibrous cellulose. It has been found that, in an applicable range of concentration, microcrystalline cellulose does not affect the oxygen transfer rate (at concentrations up to 3%). Fibrous cellulose increases the OSR during cultivation in shake flasks but decreases it during civilization in fermentors. On the basis of these results, the optimization has been carried out on the cultivation conditions in fermentors.  相似文献

This article presents a review of the enzyme mechanisms involved in degradation of cellulose by the white-rot fungus Sporotrichum poulverulentum. The hydrolytic enzymes involved include: (1) five endo-1,4-β-glucanases; (2) one exo-1,4-β-glucanase, and (3) one or several 1,4-β-glucosidases. A recently discovered oxidative enzyme of importance in in vitro cellulose degradation seems to be a cellobiose oxidase. An oxidoreductase, cellobiose:quinone oxidoreductase, is of importance both in cellulose and in lignin degradation. Regulatory mechanisms of the extracellular enzyme activities, such as monosugar levels causing catabolite repression of the endoglucanases, have also been investigated. The enzymes used by S. pulverulentum in cellulose hydrolysis are compared to those used by Trichoderma viride. Very similar types of enzymes are used in both cases. However, no oxidative enzyme has so far been found to be involved in extracellular cellulose degradation in the case of T. viride. Recommendations for further research are given.  相似文献

The substrate specificities of three cellulases and a beta-glucosidase purified from Thermoascus aurantiacus were examined. All three cellulases partially degraded native cellulose. Cellulase I, but not cellulase II and cellulase III, readily hydrolyzed the mixed beta-1,3; beta-1,6-polysaccharides such as carboxymethyl-pachyman, yeast glucan and laminarin. Both cellulase I and the beta-glucosidase degraded xylan, and it is proposed that the xylanase activity is an inherent feature of these two enzymes. Lichenin (beta-1,4; beta-1,3) was degraded by all three cellulases. Cellulase II cannot degrade carboxymethyl-cellulose, and with filter paper as substrate the end product was cellobiose, which indicates that cellulase II is an exo-beta-1,4-glucan cellobiosylhydrolase. Degradation of cellulose (filter paper) can be catalysed independently by each of the three cellulases; there was no synergistic effect between any of the cellulases, and cellobiose was the principal product of degradation. The mode of action of one cellulase (cellulase III) was examined by using reduced cellulodextrins. The central linkages of the cellulodextrins were the preferred points of cleavage, which, with the rapid decrease in viscosity of carboxymethyl-cellulose, confirmed that cellulase III was an endocellulase. The rate of hydrolysis increased with chain length of the reduced cellulodextrins, and these kinetic data indicated that the specificity region of cellulase III was five or six glucose units in length.  相似文献

The rheological properties of the culture broths of some plant cells (Cudrania tricuspidata, Vinca rosea, and Agrostemma githago) at high density (10-18 g dry wt/L) were measured, and oxygen transfer in the broths in various bioreactors was investigated. The rheological properties of the broths were dependent on the size, specific gravity, and concentration of the cell aggregates contained in the broths. The broths were non-Newtonian and pseudoplastic fluids. The flow behavior index n was fairly constant (0.53) and the consistency index K varied in proportion to the sixth-to-seventh power of the cell mass concentration M. The apparent viscosity mu(a) of the broths was in proportion to the 6.5th power of M. The oxygen transfer in the broths was discussed on the basis of the results obtained for suspensions of granulated agars (agar concentration, 5.8%) in water, which were similar to the broths in rheological properties. The volumetric oxygen transfer coefficient k(L)a in the broths was dependent on mu(a)(k(L)a proportional, variant mu(a) (-m)) and decreased greatly at a certain apparent viscosity, mu(ac). The values of m and mu(ac) were closely related to the aeration-agitation mechanisms of the bioreactors. The values of mu(ac) in aeration-agitation type bioreactors was larger than that in aeration-type bioreactors, whereas for m, the reverse was true.  相似文献

Culture media and fermentation conditions for cultivation of an endophytic fungus Phomopsis sp. Hant25 were investigated in order to improve the yield of mycoepoxydiene, a novel fungal metabolite having potent cytotoxic activity against many cancer cell lines. Mycoepoxydiene accumulated in the culture broth during the stationary phase of fungal growth. Modified M1D medium was superior to malt Czapek, and Czapek yeast autolysate broths in supporting mycoepoxydiene production. Pellet growth was the morphological form that favored biosynthesis of mycoepoxydiene. This could be achieved by incubating the culture statically for 6 days before shaking at 120 rpm. Incorporation of a cellulose paper disc into the culture flask promoted fungal growth at the liquid surface, which accelerated mycoepoxydiene production and maximized the final yield to a level of 354 mg l−1, though fungal attachment to the solid support was not required. Since the peak concentration of mycoepoxydiene in the culture broth was followed by a steeply declining phase, the harvest time had to be precisely determined for maximum product yield. Understanding the factor(s) involved in rapid degradation of mycoepoxydiene could lead to improved final yields.  相似文献

β-1,4-glucosaminoglucan (GG) was prepared from the sheath of a sulfur-oxidizing bacterium Thiothrix nivea. Recently, GG was found to be adsorbed by cellulose (paper) and is therefore potentially applicable as an aminating agent for cellulose. We attempted to increase the yield of GG using a fed-batch cultivation method. Furthermore, the behavior of GG molecules in water was theoretically and experimentally investigated. NMR analysis in combination with molecular dynamics calculation suggested that GG molecules tend to form soluble aggregates in water. It was experimentally revealed that the self-aggregation is enhanced by the addition of NaCl and reduced temperature. Adsorption of GG onto cellulose via hydrogen bonding was confirmed by molecular dynamics simulation. Adsorption was also promoted in the presence of NaCl but was inhibited by a reduction in temperature. Only 11% of the amino groups in the GG-treated paper was reactive, suggesting that GG molecules adsorbed by the paper were forming aggregates.  相似文献

Measurement of blood viscosity using mass-detecting sensor   总被引:1,自引:0,他引:1  
A newly designed mass-detecting capillary viscometer is extended to measure the viscosity of whole blood over a range of shear rates without the use of anticoagulants in a clinical setting. In the present study as proof of principle, a single measurement of liquid-mass variation with time replaces the flow rate and pressure drop measurements that are usually required for the operation of a capillary tube viscometer. Using a load cell and capillary, we measured the change of mass flowing through capillary tube with respect to the time, m(t), from which viscosity and shear rate were mathematically calculated. For water and adulterated bloods, excellent agreement was found between the results from the mass-detecting capillary viscometer and those from a commercially available rotating viscometer. Also, the mass-detecting capillary viscometer measured the viscosity of unadulterated whole blood without heparin or EDTA. This new method overcomes the drawbacks of conventional viscometers in the measurement of the whole blood viscosity. First, the mass-detecting capillary viscometer can accurately and consistently measure the unadulterated blood viscosity over a range of shear rates in less than 2 min without any anticoagulants. Second, this design provides simplicity (i.e. ease of operation, no moving parts, and disposable) and low cost.  相似文献

This study investigated the influence of the degree of polymerization (DP) of cellulose materials (microcrystalline cellulose [MCC]) on some powder properties and the compression behavior of these materials. The DP was determined by measurements of viscosity (H). The weight average of molecular weight and the weight average of the different DPs were investigated after MCC was modified to cellulose tricarbanilate by light scattering measurements. The DP showed a remarkable influence on the physicochemical properties of the cellulose materials and, consequently, on the behavior of these materials during compression. MCC types with a high DP value showed greater water absorption than the types with a low DP value. No relevant relationship between the crystallinity index and the DP could be observed. DP 190 showed lower compactibility and compressibility parameters than DP 244 and 299. No significant differences could be observed between DP 244 and 299 when the same particle size fraction was compressed. Furthermore, the compressibility was increased by increasing the DP.  相似文献
