The mature nodules ofPhaseolus aureus Roxb. var.mungo possessed, in comparison with young and old nodules, higher activities of nitrogenase (N2ase), and indol-3-ylacetic acid (IAA) metabolic enzymes like IAA oxidase, methylene oxindole reductase and peroxidase; higher levels of IAA-like, gibberellic acid-like (GA), and cytokinin-like (CK) substances and tryptophan, and lower level of phenol. The abscisic acid-like (ABA) substance level was higher in the old nodules. The N2ase activity in the mature nodules changed parallelly with IAA and OK. butoppositely with GA and ABA. The changes in tryptophan level, IAA oxidizing enzymes, and phenol metabolism controlled the IAA level in the nodules. The nodules developed similarly throughout the year, but they had variable hormone levels in different seasons. This indicated that the formation and growth of the nodules was controlled not only by the nodular hormones.  相似文献

The mature nodules ofLens esculenta Moench. contained higher levels of indolyl acetic acid (IAA), cytokinins (CK), gibberellic acid (GA)-like substances and more active in nitrogenase (N2-ase) activity than young or old ones. Synthesis of IAA and its metabolism was found to be controlled by tryptophan (tryp) and phenol metabolism, respectively, in nodules of different ages. An abscisic acid (ABA)-like substance being a ‘late growth phase’ hormone, was highest in old nodules. IAA and CK were highest in winter when N2-ase activity was also highest but then GA and ABA were low. The IAA metabolic pattern of both roots and nodules was regulated by phenols. The hormones hardly changed seasonally in the roots and showed higher activities of IAA oxidase, MeOx reductase, peroxidase and polyphenol oxidase than nodules. The nodular anabolic changes are more pronounced in winter as lentil is a winter crop. The size of nodules at a particular age was the same in different seasons, even though their hormone content varied with the season showing that the nodular hormones were not solely utilized for nodule development and growth.  相似文献

沙生柽柳扦插生根过程插穗相关理化特征分析   总被引:1,自引:0,他引:1  
选取沙生柽柳半木质化枝条进行苗床扦插,通过实验测定插穗生根过程中内源激素(IAA、GA3、ZR、ABA)含量、可溶性营养物质(糖、蛋白质)含量及相关氧化酶(PPO、POD、SOD、IAAO)活性的动态变化特征,探讨沙生柽柳插穗扦插生根机理。结果表明:(1)沙生柽柳插穗内源激素含量随生根进程而发生变化,其中,IAA含量在扦插35d最大,并出现较大的波动变化;ZR含量在扦插55d前后变化明显,呈现低水平向高水平转化趋势;ABA、GA3含量依次呈先升高后降低再升高的变化过程,并在扦插15d和55d(80d)呈现变化的峰值和谷值。(2)沙生柽柳扦插生根与相关氧化酶活性密切相关,其中,POD、IAAO活性在插穗扦插35d后长时间保持较高水平,直至插穗生根后POD活性明显降低,IAAO活性有所增加;PPO、SOD活性则在插穗扦插15d保持较高活性,且PPO活性的变化均匀,SOD活性的高低交替变化明显。(3)在沙生柽柳扦插生根期间,插穗可溶性糖含量呈现生根前消耗减少与生根后积累增加两大变化过程,可溶性蛋白质含量表现为扦插后逐步积累增加的变化趋势。研究表明,高水平的IAA、ZR和低水平的GA3、ABA共同调控着沙生柽柳插穗生根;IAA能够通过促进插穗POD、PPO、IAAO活性变化来影响生根,较高的POD、IAAO活性可调节插穗IAA水平,高水平的PPO活性则催化插穗IAA-酚酸复合物的形成,进而诱导插穗生根。  相似文献

Summary The content of endogenous gibberellin (GA)-like substances of roots and root nodules of SOya, and GA production byRhizobium japonicum cultures, were investigated by a combined thin layer chromatographic (TLC)-dwarf pea epicotyl bioassay technique. GAs were more concentrated in root nodules than in the roots, totalling 1.34 and 0.16 nM GA3 equivalents g−1 dry wt. respectively. GA production byR. japonicum cultures was demonstrated (1.00 nM GA3 equivalentsl −1) and comparison of the GA components of plant and bacterial culture medium extracts, suggested that rhizobial GA production may contribute to the nodule GA content. Cis-trans abscisic acid (ABA) was identified in root and nodule extracts by TLC-gas liquid chromatography (GLC), and amounted to 0.18 and 2.21 nM g−1 dry wt. respectively, whereas 0.30 and 4.63 nM ABA equivalents g−1 dry wt. were detected by a TLC-wheat embryo bioassay technique. 为探索黄花倒水莲春梢生理生化特性的差异以及不同内源激素的变化规律,该文对黄花倒水莲春梢的生长动态进行监测,采用间接酶联免疫吸附法(ELISA)测定脱落酸(ABA)、生长素(IAA)、赤霉素(GA)、乙烯(ETH)和玉米素核苷(ZR)五种内源激素含量的动态变化,并对两者间的相关性进行分析。结果表明:(1)黄花倒水莲春梢生长发育过程可分为快速增长期(0~12 d)、生长转折期(16~20 d)和缓慢增长期(24~32 d)三个阶段。(2)内源激素ABA、GA、ETH和ZR含量在缓慢增长期显著高于快速增长期和生长转折期,IAA含量各时期差异较小。(3)春梢长、底部叶长和叶宽在快速增长期与ABA、GA、ETH和ZR含量呈负相关,且与ZR含量具有一定显著性,与IAA含量呈正相关;生长转折期,各指标与GA、ETH和ZR含量呈正相关,与GA含量具有一定显著性,与ABA含量呈负相关;缓慢增长期,各指标与五种内源激素含量均呈正相关,与IAA和ZR含量具有一定显著性。该研究结果为生产上利用外源激素调控黄花倒水莲春梢抽出以及生长提供了理论基础。  相似文献

Effects of plant hormones indole-3-yl-acetic acid (IAA), gibberellic acid (GA), benzylaminopurine (BAP), abscisic acid (ABA) and ethrel (ETH) in 5 M concentration on gas exchange, ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO, EC 4.1.1.39) activity, pigment content and yield in cotton (Gossypium hirsutum L. cv. H-777) under drought were studied. At reproductive stage (55 – 60 d after sowing) these hormones were sprayed on shoots one day prior to stress imposition by withholding irrigation. The soil moisture of control plants was kept at field capacity. Net photosynthetic rate (PN), stomatal conductance (gs), transpiration rate (E), carboxylation efficiency (CE), water use efficiency (WUE), RuBPCO activity, boll number per plant, seed number per plant and lint mass per plant significantly decreased at drought while chlorophyll (Chl) b content and flower number per plant increased. ABA and ETH significantly reduced gas exchange parameters, Chl a and Chl b content. Detrimental drought effect on PN, gs, E, CE, RuBPCO and lint mass per plant was significantly alleviated by BAP and also its effect on seed number and lint mass per plant was significantly alleviated with the ABA treatment.  相似文献

Ethylene, indol-3-acetic acid (IAA), gibberellin-like substances (GAs) and abscisic acid (ABA) were analysed in extracts from normal, seed-containing and parthenocarpic tomato fruits throughout fruit development. Parthenocarpic fruit growth was induced with an auxin (4-CPA), morphactin (CME) or gibberellic acid (GA3) and compared with that of pollinated control fruits. Fruit growth was only affected by the treatment with GA3, decreasing size and fresh weight by 60%. The peak sequence of hormones during fruit development was ethylene-GAs-IAA-ABA. Seeded fruits contained the highest levels of IAA and ABA but the lowest levels of GAs. Also, in seeded fruits, a high proportion of IAA and ABA was found in the seeds whereas this was not the case for GAs.Hormone levels of tomato fruits may be successfully, easily and reproducibly altered by inducing parthenocarpic fruit growth and thus eliminating development of seeds which are a major source of hormone synthesis. In spite of markedly changed hormone levels, there was no obvious relationship between fruit growth and extractable hormones per se. However, the results indicate that a high ratio of GAs: auxins is unfavourable for growth of tomato fruits.  相似文献

The data obtained characterize the changes in the contents of endogenous phytohormones (IAA, cytokinins, GA, and ABA) in germinating pollen grains and growing pollen tubes of a self-compatible clone of petunia (sPetunia hybrida L.) within an 8-h period under in vitro conditions. The hydration and initiation of germination of pollen grains brought the ABA content down to a zero level, while the levels of GA, IAA, and cytokinins increased 1.5–2-fold. Later, in the growing pollen tubes, the GA content increased twofold, while the levels of IAA and cytokinins decreased. The exogenous ABA and GA3 considerably promoted pollen germination and pollen tube growth; however, only the treatment with GA3 produced the maximum length of pollen tubes. The exogenous IAA promoted and the exogenous cytokinins hindered the growth of pollen tubes. The membrane potential, as assessed with a potential-sensitive dye diS-C3-(5), considerably increased in the pollen grains treated with ABA and benzyladenine, whereas IAA and GA3 did not practically affect it. The authors conclude that the mature pollen grains contain the complete set of hormones essential for pollen germination and pollen tube growth. 采用开顶箱系统,研究了银杏叶片内源激素脱落酸(ABA)、吲哚乙酸(IAA)、玉米素核苷(ZR)和赤霉素(GA3)对大气CO2浓度升高(环境CO2浓度+350 μmol·mol-1,EC)的响应.结果表明,EC处理能使ABA含量降低,与对照(CK)相比, ABA含量最大降低63.0%(处理后120 d).EC处理使叶片IAA和ZR含量增加,而且随着处理时间的延长,差异均达显著水平;IAA含量在处理后100 d为CK的2倍,ZR含量在处理后80 d时为CK的2.5倍.EC处理使叶片GA3峰值提前出现. (IAA+GA3+ZR)/ABA比值随着银杏的生长逐渐降低,在处理后期(处理后40~120 d)明显高于CK,表明大气CO2浓度升高可促进银杏的生长发育.  相似文献

Accumulation of protein and starch in grain is a key process determining grain yield and quality in wheat. Under drought or waterlogging, endogenous plant hormone levels will change and may have an impact on the yield and quality of wheat. In a greenhouse experiment, four winter wheat (Triticum aestivum L.) varieties differing in grain protein content, Heimai 76, Wanmai 38, Yangmai 10 and Yangmai 9, were subjected to drought (SRWC = 4550%, DR), waterlogging (WL) and moderate water supply (SRWC = 7580%, CK), beginning from 4 days post-anthesis (DPA) to maturity. On the 10 (grain enlargement stage) and 20 (grain filling stage) DPA, endogenous abscisic acid (ABA), gibberellins (GA1+3), indole-3-acetic acid (IAA) and zeatin riboside (ZR) were determined in sink and source organs of wheat plants by enzyme linked immunosorbent assay (ELISA). The patterns of hormonal changes were similar in four varieties. The ABA levels were much higher under DR and WL than under CK. Compared with CK, GA1+3 levels in whole-plant under DR and WL changed a little at 10 DPA, but markedly decreased under DR and WL at 20 DPA. Changes of endogenous IAA level under DR and WL exhibited a complicated pattern, depending on organs and growth stages. Particularly at the 20 DPA, the mean levels of IAA in roots, leaves and grains decreased significantly under DR and WL. In comparison with CK, ZR levels in all organs significantly decreased under DR and WL at both stages. The correlation analyses between yields and contents of starch and protein in grains and levels and ratios of four hormones in source and sink organs indicated that the changes in yield and content of grain starch and protein under DR and WL were associated with the reduced IAA, ZR and GA1+3 levels and elevated ABA level in plants, especially in grains. It was proposed that the changed levels of endogenous hormones under post-anthesis DR and WL might indirectly affect protein and starch accumulation in grains by influencing the regulatory enzymes and processes.  相似文献

In the present report, we described the effects of indole-3-acetic acid (IAA), zeatin (ZT), gibberellin (GA3), and abscisic acid (ABA) on in vitro pollen germination and pollen tube growth in Torenia fournieri L. The results showed that IAA and GA3 stimulated in vitro pollen tube growth, ABA inhibited pollen tube growth, and ZT had no significant effect on the process. The stimulating effect of exogenous IAA was particularly distinct, and led to synchronous growth of straighter and more slender pollen tubes compared with the controls. However, no significant changes were found in the germination of the treated pollen. The auxin efflux inhibitor, 10 μM 1-N-naphthylphthalamic acid (NPA), was also found to stimulate pollen tube growth. We measured the content of hormones (free IAA, ZT, GA3, and ABA) in the stigmas and styles before and after pollination. The hormone contents of stigmas measured 0.5 h after pollination (0.5 HAP) showed that ABA content decreased, whereas the content of IAA, ZT, or GA3 did not change significantly. The hormone level in pollinated styles (4 HAP) when pollen tubes had grown into the middle part of style was characterized by an increase in free IAA and GA3 and a decrease in ABA, which was in agreement with the results that IAA and GA3 promoted but ABA inhibited pollen tube growth in vitro. Furthermore, the change of IAA level in styles was most notable, which was accordant to the fact that auxin stimulated significantly pollen tube growth in vitro. Using immunoenzyme and immunogold labeling techniques and an anti-IAA monoclonal antibody, we confirmed that free IAA was present throughout style tissues, and distributed in the nucleus and cytoplasm of style cells. All these results suggested that hormones, especially IAA, play important roles in pollen tube growth of T. fournieri. 以紫斑牡丹种子为试验材料,研究不同浓度的赤霉素(GA_3)处理对种子生根以及生根过程中营养物质、酶活性和内源激素水平变化的影响,为探讨紫斑牡丹种子萌发机制提供依据。结果表明:(1)GA_3处理能够促进种子生根,并以300 mg/L GA_3处理对种子生根效果最好,与对照相比可提前14.67 d生根,生根率可达71.00%。(2)与对照相比,GA_3处理可以在0~15 d时促进种子淀粉水解和可溶性糖的积累,并加速可溶性蛋白的消耗,在0~30 d促进过氧化物酶(POD)活性的提高,从而促进种子萌发生根。(3)在种子沙藏生根过程中,种子脱落酸(ABA)含量呈下降趋势,赤霉素(GA)、玉米素核苷(ZR)和吲哚乙酸(IAA)含量均表现出先上升后下降的趋势,与对照相比,GA_3处理可使种子GA、ZR和IAA的含量在沙藏前期明显上升,以解除种子休眠。研究发现,外源GA_3处理可以调控紫斑牡丹种子内源激素含量和POD活性的变化,促进营养物质转化,从而提前解除种子休眠使其萌发。  相似文献

The leaves of 30-d-old plants of Brassica juncea Czern & Coss cv. Varuna were sprayed with 10–6 M aqueous solutions of indole-3-yl-acetic acid (IAA), gibberellic acid (GA3), kinetin (KIN), and abscisic acid (ABA) or 10–8 M of 28-homobrassinolide (HBR). All the phytohormones, except ABA, improved the vegetative growth and seed yield at harvest, compared with those sprayed with deionised water (control). HBR was most prominent in its effect, generating 32, 30, 36, 70, 25, and 29 % higher values for dry mass, chlorophyll content, carbonic anhydrase (E.C. 4.2.1.1) activity, and net photosynthetic rate in 60-d-old plants, pods per plant, and seed yield at harvest, over the control, respectively. The order of response to various hormones was HBR > GA3 > IAA > KIN > control > ABA.  相似文献

Earlier, we reported that mutation in the Male Sterile33 (MS33) locus in Arabidopsis thaliana causes inhibition of stamen filament growth and a defect in the maturation of pollen grains [Fei and Sawhney (1999) Physiol Plant 105:165–170; Fei and Sawhney (2001) Can J Bot 79:118–129]. Here we report that the ms33 mutant has other pleiotropic effects, including aberrant growth of all floral organs and a delay in seed germination and in flowering time. These defects could be partially or completely restored by low temperature or by exogenous gibberellin A4 (GA4), which in all cases was more effective than GA3 Analysis of endogenous GAs showed that in wild type (WT) mature flowers GA4 was the major GA, and that relative to WT the ms33 flowers had low levels of the growth active GAs, GA1 and GA4, and very reduced levels of GA9, GA24 and GA15, precursors of GA4. This suggests that mutation in the MS33 gene may suppress the GA biosynthetic pathway that leads to GA4 via GA9 and the early 13-H C20 GAs. WT flowers also possessed a much higher level of indole-3-acetic acid (IAA), and a lower level of abscisic acid (ABA), relative to ms33 flowers. Low temperature induced partial restoration of male fertility in the ms33 flowers and this was associated with partial increase in GA4. In contrast, in WT flowers GA1 and GA4 were very much reduced by low temperature. Low temperature also had little effect on IAA or ABA levels of ms33 flowers, but did reduce (>2-fold) IAA levels in WT flowers. The double mutants, ms33 aba1-1 (an ABA-deficient mutant), and ms33 spy-3 (a GA signal transduction mutant) had flower phenotypes similar to ms33. 牡丹品种朱砂垒(PaeoniasuffruticosaAndr.cv.Zhushalei)在冬季室内催花过程中7种内源激素含量变化不同。玉米素核苷(Z ZR)、生长素(IAA)和赤霉素(GA3)的含量在花生长发育过程中处于较高水平;而脱落酸(ABA)、异戊烯基腺苷(IP IPA)、二氢玉米素核苷(DHZ DHZR)、赤霉素(GA4)的含量低于上述3种内源激素。激素平衡方面,GAs/ABA、CTKs/ABA、IAA/ABA处于较高水平,变化幅度较大。在催花过程中,内源激素以及其平衡影响牡丹花的生长发育。本研究结果对牡丹花期调控提供理论依据。  相似文献

关于中国沙棘克隆生长调节研究目前局限于外在机制,旨在探讨其克隆生长对灌水强度的响应规律及其激素调控的内在机制。结果表明:随着灌水强度的增大,分株生长、克隆繁殖、克隆扩散能力先升后降,IAA(吲哚-3-乙酸)、ZR(玉米素核苷)、GA_3(赤霉酸)含量及其与ABA(脱落酸)的比值先升后降而ABA含量先降后升。同时,分株生长、克隆繁殖、克隆扩散能力与IAA、ZR、GA_3含量以及IAA/ABA、ZR/ABA、GA_3/ABA呈极显著或显著正相关,与ABA含量呈极显著负相关。灌水强度过小或过大,IAA、ZR、GA_3含量以及IAA/ABA、ZR/ABA、GA_3/ABA低而ABA含量高,克隆生长潜力受到抑制,种群以分株小、数量少(分布稀疏)、扩散(水平根延伸和分枝)能力弱为特征,克隆生长格局倾向于"游击型"、种群早衰概率高;灌水强度适宜,IAA、ZR、GA_3含量以及IAA/ABA、ZR/ABA、GA_3/ABA高而ABA含量低,克隆生长潜力得以充分发挥,种群以分株大、数量多(分布密集)、扩散能力强为特征,克隆生长格局倾向于"聚集型"、种群稳定性高。随着灌水强度过小-适宜-过大的连续变化,中国沙棘通过改变激素状况调控克隆生长,从而形成与灌水强度相适应的克隆生长格局连续体"游击型-聚集型-游击型",种群稳定性呈"低-高-低"的连续变化过程。由此可见:灌水强度诱导内源激素发生改变,激素特征调控克隆生长格局,克隆生长格局决定种群稳定性。  相似文献

Similar ranges of gibberellins (GAs) were detected by high-performance liquid chromatography (HPLC)-immunoassay procedures in ten cultures of wild-type and mutant strains of Rhizobium phaseoli. The major GAs excreted into the culture medium were GA1 and GA4. These identifications were confirmed by combined gas chromatographymass spectrometry. The HPLC-immunoassays also detected smaller amounts of GA9- as well as GA20-like compounds, the latter being present in some but not all cultures. In addition to GAs, all strains excreted indole-3-acetic acid (IAA) but there was no obvious relationship between the amounts of GA and IAA that accumulated. The Rhizobium strains studied included nod and fix mutants, making it unlikely that the IAA- and GA-biosynthesis genes are closely linked to the genes for nodulation and nitrogen fixation.The HPLC-immunoassay analyses showed also that nodules and non-nodulated roots of Phaseolus vulgaris L. contained similar spectra of GAs to R. phaseoli culture media. During the entire period of internode growth of Merremia emarginata contents of gibberellic acid (GA3), phenyl-acetic acid (PAA), indole-3-acetic acid (IAA, free and conjugated) and abscisic acid (ABA, free and conjugated) were estimated by ELISA using polyclonal antibodies raised against each hormones. At the time of internode elongation free auxin content was low and increased with the decrease in the rate of elongation. In contrast, conjugated IAA showed declining trend where free IAA content was remarkably high, suggesting thereby that conjugated IAA might have mobilized during the later phase of internode development. The endogenous GA3 contents were high as compared to other hormones; however, no significant role of GA3 was discernible in elongation growth. Conjugated ABA contents remained very low during the elongation growth and increased thereafter.  相似文献

The root nodules of Melilotus alba DESR ., a fodder legume, contained high amounts of IAA. A tryptophan pool present in the nodule might serve as a source of IAA production. Presence of IAA oxidase and peroxidase in the nodules indicated the metabolism of IAA, at least in part, in the nodules. The Rhizobium species isolated from the root nodules produced a high amount of IAA (190 μg/ml) from L-tryptophan supplemented basal medium. IAA production and microbial growth were coincident. The production of IAA by the Rhizobium sp. was increased by 315% when the medium was supplemented with lactose (1%), NiCl2 (10 μg/ml), cetyl pyridinium chloride (0.5 μg/ml) and glutamic acid (0.4%), in addition to L-tryptophan (3 mg/ml). The possible role of the rhizobial production of IAA on the rhizobia-legume symbiosis is discussed.  相似文献
